技术领域
本发明涉及含有来源于植物、动物或矿物组份的保健品,特别涉及一种补血调经的中药保健品及其制备方法。
背景技术
女子以肝为先天,故妇科疾病多从肝论治。肝体阴而用阳,劳役过度、郁怒及经、孕、产、乳等无不耗伤阴血,以致肝失血养、气失疏泄,或气郁日久化热。月经病俗称月经不调,是指月经的周期或经量出现异常,包括月经先期、月经后期、月经先后无定期、经期延长、月经过多、月经过少。中医学认为月经的产生与调节受脏腑气血盛衰、经络通畅的直接影响,其中与肾的关系尤为密切,如《素问·上古天真论》中所说:“女子七岁肾气盛,齿更发长;二七而天癸至,任脉通,太冲脉盛,月事以时下,故有子。七七任脉虚,太冲脉衰少,天癸竭,地道不通,故形坏而无子也”。月经病的诊疗重在辨月经的周期、量、质、色以及伴随症状。月经紊乱是指与月经有关的多种疾病,包括月经的周期、经期、经量、经色、经质的改变以及痛经、闭经、经前期紧张综合症等伴随月经周期前后出现的某些症状为特征的多种病症的总称。月经紊乱可分为两类,一为无排卵型月经失调;另一为有排卵型月经失调。无排卵型月经失调因不排卵,故必然不育,有排卵型月经失调虽然有排卵但因黄体功能异常,故此可能不孕或易于孕早期流产,这两类月经失调经治疗后均可能恢复正常月经并怀孕。
痛经是妇女最常见的一种病,其发病原因多为经行之际,受各种致病因素的影响,使冲任气血运行不畅,严重者经血滞于胞宫、胞络,以致“不通则痛”;或气血虚弱、肾气亏损致子宫失于濡养,“不荣而痛”。病位在子宫、冲任,以“不通则痛”或“不荣则痛”为主要病机,临床分功能性(又称原发性)和器质性(又称继发性)。常表现出腹痛多发生在经潮前(1~2)d,行经第1d达高峰,可呈阵发性痉挛性或胀痛伴下坠感,严重者可放射到腰骶部、肛门、阴道、股内侧。甚至可见面色苍白、出冷汗、手足发凉等晕厥之象。也有少数于经血将净或经净后(1~2)d始觉腹痛或腰腹痛者。结合体格检查,一般不伴腹肌紧张或反跳痛,青春期或未孕年轻女性者多属功能性痛经;育龄期或反复有孕产史及宫腔手术史伴盆腔内有粘连、包块、结节或增厚者,则多属器质性如盆腔炎症、子宫内膜异位症等病所致。部分患者也可见子宫体极度屈曲或宫颈口狭窄。
《灵枢·经脉》指出:“肝足厥阴之经脉,起于大指丛毛之际......过阴器,抵小腹......是动则病腰痛不可以俯仰,丈夫瘣疝,妇人少腹痛,甚则嗌干,面尘脱色。”《素问·举痛论》也指出:“寒气客于脉中,则血泣脉急,故胸肋与少腹相引痛矣”。寒凝肝经,经脉拘急,气滞血瘀是引起子宫内膜异位症痛经的主要病因病机。临床表现如下腹拘急冷痛,畏寒肢冷,面色苍白。诸多温经散寒药如牡丹皮、吴茱萸、肉桂,皆入肝经而达到温经止痛之功效。厥阴之脉,厥阴之脉者,络阴器系于肝,肝失疏泄,气机郁滞所表现的证候。多因情志抑郁,或突然的精神刺激及其他病邪的侵扰而发病。。气聚血结,日久成啯瘕积聚。治宜疏肝解郁,必要时配以理气化痰,活血软坚等法。
对於女仕们月经不调,紊乱,痛经等诸症,均为肝失疏泄,气机郁结,则情志抑郁;久郁不解,失其柔顺舒畅之性,故急躁易怒。气郁生痰,痰随气升,搏结于咽则见梅核气,积聚于颈项则为瘿瘤,气病及血,气滞血瘀,冲任不调,故月经不调或经行腹痛气血虚弱。
研究结果认为活血化瘀类中医中药亦可能有效。但中医药制剂因人而异,服用使用不便,往往能很难对症用药,并且针对性不强,治疗效果一般;故寻找安全、有效、全面疏肝解郁,活血化瘀,调经、缓解痛经,补血养气的保健品,是摆在广大医药研究工作者面前的重要课题之一。
发明内容
本发明所要解决的技术问题在于,目前调经、缓解痛经的常规办法是口服维生素E和B1、或者服用乌鸡白凤丸、补血口服液等保健品,而中医药治疗保健效果因人而异,针对性差强人意,本发明在治疗时采用从肝论治,以辨证论治为基础,在应用活血化瘀药同时分别加疏肝、温肝、养肝、清肝等调肝药物,月经失调、经期腹痛、腰腿酸软、赤白带下、小腹虚冷、经来腹痛、寒凝血滞、容颜易老、久不成孕临床取得很好的疗效。

所述补血调经的中药保健品,其所述保健品剂型可以为:片剂、糖衣片剂、薄膜衣片剂、肠溶衣片剂、胶囊剂、硬胶囊剂、软胶囊剂、口服液、口含剂、颗粒剂、冲剂、蜜丸剂、散剂、丹剂、溶液剂、注射剂、硬膏剂或糖浆剂。
为解决上述技术问题,本发明还提供一种补血调经的中药保健品的制备方法,所述保健品为胶囊剂时,制备方法包括:
a.将所述原料放入粉碎机,粉碎后过筛;
b.将过筛后剩下的颗粒物放入乙醇中加热回流提取2次;
c.再取上述乙醇提取后的药渣,加水加热回流提取1次;
d.将上述两种提取液合并,减压回收乙醇并喷雾干燥,得药粉;混合过筛得到的细粉装入胶囊制为胶囊剂。
所述步骤a中,可以将所述原料放入粉碎机粉碎后过100目细筛,使能通过100目筛的细粉量在达到30%,收集通过120目筛的原料细粉备用;
所述步骤b中,可以将过筛后剩下的颗粒物放入乙醇中加热回流提取2次,每次1~2小时,将2次提取液合并静置;
所述步骤c中,可以将上述乙醇提取过的药渣加10倍量水加热1~2小时回流提取1次;将上述3次提取液合并静置;所述步骤d中,将上述提取液合并,减压浓缩相对密度为80℃时1.06的滤液,回收乙醇,将所得滤液调整比重到1.06,通入喷雾干燥机进行喷雾干燥,过喷雾干燥机喷头的压缩空气压力为4kg/m2,干燥室热气流温度是80℃,得原料药粉;将过筛后得到的原料细粉与乙醇提炼药粉混合装入胶囊。
为解决上述技术问题,本发明再提供一种补血调经的中药保健品的制备方法,所述制剂剂型为口服液剂的制备方法包括:
a、取原料放入乙醇中浸泡,热提取2次,成膏状,为组分1;
b、药渣水提取2次,浓缩过滤为膏状,为组分2;
c、将上述两种膏状浓缩成糊状,灭菌,加入糊精,然后真空冷冻密封。
所述步骤a中,可以取原料加入5-10倍量的乙醇中浸泡1-2小时,加热提取2次,每次1-2小时,去上清液,合并提取液,100-120目滤过,再经截流分子量为5000-10000的超滤柱超滤,超滤液减压浓缩相对密度为80℃时1.36的浸膏,加热浓缩至膏状,静置备用,成为组分1;所述步骤b中,可以将乙醇提取过的药渣放入10倍量水中,加热回流提取2次,每次提取时间为1~2小时,100-120目滤过,再经截流分子量为5000-10000的超滤柱超滤,超滤液减压浓缩相对密度为80℃时1.36的浸膏,加热浓缩至膏状,静置备用,成为组分2;所述步骤c中,可以将上述两种组分膏状浓缩成糊状,浓缩液用CO 60灭菌,辐照剂量8k,加入糊精,然后真空罐装密封即得,使用时直接饮用。
本发明对月经失调、经期腹痛、腰腿酸软、赤白带下、小腹虚冷、经来腹痛、寒凝血滞、容颜易老、久不成孕等,均有显著疗效。产后常服,更能培补天真,恢复原来健美身段。少女常服,可助长发育健全、滋补养颜,青春常驻。本发明应用活血化瘀药同时分别加疏肝、温肝、养肝、清肝等调肝药物,具有平衡女性激素分泌、保护卵巢、维持子宫肌弹性不易老化之独特功效。诚为女仕们滋补强身、延展青春、美丽之珍品也。
具体实施方式
清代何梦瑶《医碥》载:“百病生于郁,郁而不疏,则皆肝木之病矣。”女子生理性周期气血易波动,情志易于忧郁,情志郁结,肝气不舒,肝失疏泄,导致气机郁滞,气血互结,血运不畅,而冲任胞脉瘀阻发生痛经。子宫内膜异位症痛经的本质在于瘀血,而关键则是气。月经病的常见证型包括血虚证、血瘀证、气虚证、肾虚证、血热证、血寒证、气滞证等。
“气为血之帅,血为气之母”,气与血在生理上互根互用,病理上相互影响的关系,最终决定了疏肝解郁法在治疗月经紊乱、痛经的治疗中有着不可替代的作用。治当疏肝行气解郁,肝主疏泄功能正常,则气血调和,经行通畅,通则不痛。治宜疏肝解郁,必要时配以理气化痰,活血软坚等法。女子以肝为先天,故妇科疾病多从肝论治。肝体阴而用阳,劳役过度、郁怒及经、孕、产、乳等无不耗伤阴血,以致肝失血养、气失疏泄,或气郁日久化热。据此病机遣方,方中以白芍用其养血调经、平肝止痛、敛阴止汗之功,以解血虚、阴虚有热致月经不调,肝阴不足致头痛眩晕、肋胁疼痛以及阴虚盗汗等证;女贞子具有补肝肾阴、清热明目之效,除腰膝酸痛,阴虚内热,骨蒸劳热,头晕目眩等之痹;丹参清热燥湿,泻火解毒,退热除蒸,解阴虚致烘热汗出,五心烦热等证。丹参养血活血,制首乌补益肝肾,滋阴退热,熟地,牡丹皮提阳除烦安神之效,诸药配伍具有清热除烦,补肾益精、益气活血,壮骨补髓,消除健忘失眠、腰膝酸痛、头晕耳鸣、口干便艰;月经紊乱之功效。
益母草味辛苦、凉。活血、祛淤、调经、消水。治疗妇女月经不调,胎漏难产,胞衣不下,产后血晕,瘀血腹痛,崩中漏下,尿血、泻血,痈肿疮疡。益母草含益母草碱、延胡索酸等物质,有活血调经、降血压、利尿消肿等作用。
覆盆子性温;甘、酸;归肾、膀胱经。主治:益肾,固精,缩尿。用于肾虚遗尿,小便频数,阳痿早泄,遗精滑精。《本草经疏》:″覆盆子,其主益气者,言益精气也。肾藏精、肾纳气,精气充足,则身自轻,发不白也。苏恭主补虚续绝,强阴建阳,悦泽肌肤,安和脏腑。甄权主男子肾精虚竭,阴痿,女子食之有子。大明主安五脏,益颜色,养精气,长发,强志。皆取其益肾添精,甘酸收敛之义耳。″《药性论》载其:″主男子肾精虚竭,女子食之有子。主阴痿。″
中药何首乌有生首乌与制首乌之分,直接切片入药为生首乌,用黑豆煮汁拌蒸后晒干入药为制首乌。二者的功用有所不同:生首乌功能解毒、消痈、润肠通便,常用于治疗瘰疬疮痈、风疹瘙痒、肠燥便秘;制首乌功能补肝肾、益精血、乌须发、强筋骨,用于血虚萎黄、眩晕耳鸣、须发早白、腰膝酸软、肢体麻木、崩漏带下、久疟体虚等。本发明所采用的制首乌是补肾益肾的佳品。
女贞子用于肝肾阴虚的目暗不明,视力减退,须发早白,腰酸耳鸣及阴虚发热等。本品能补肝肾阴,但药力平和,须缓慢取效。治目暗不明,常配熟地,枸杞等同用,治须发早白,常配桑葚同用,阴虚发热,常配地骨皮。肝肾阴虚,头昏目眩、遗精耳鸣、腰膝酸软,须发早白,冠心病、高脂血症、高血压、慢性肝炎。对肝脏的保护作用研究认为,女贞子含有齐墩果酸,给大鼠皮下注射,可降低血清谷丙转氨酶,对四氯化碳诱发的肝损伤有明显保护作用,并能促进肝细胞再生.齐墩果酸亦可使肝内甘油三酯蓄积减少、肝细胞变性坏死明显减轻、糖原蓄积增加、血清γ球蛋白下降;超微结构可见肝细胞内线粒体肿胀与内质网囊泡变性减轻、肝组织间质炎症反应减弱。
白芍入肝、脾经,养血柔肝,缓中止痛,敛阴收汗。治胸腹胁肋疼痛,泻痢腹痛,自汗盗汗,阴虚发热,月经不调,崩漏,带下,妇人血闭不通,消瘀血,能蚀脓,益女子血。《日华子本草》:治风、补劳,主女人一切病,并产前后诸疾,通月水,退热,除烦,益气,天行热疾,瘟瘴,惊狂,妇人血运,及肠风,泻血,痔瘘。发背,疮疥,头痛,明目,目赤努肉。赤色者多补气,白者治血。
山茱萸,主治心下邪气寒热,温中,逐寒温痹,去三虫,久服轻身有强阴益精、安五脏、通九窍、止小便淋沥之功;久服明目、强力长年。山茱萸,又名山芋肉、药枣、实枣儿、枣皮、肉枣等,为我国常用名贵中药材,应用历史悠久。它以其补力平和、壮阳而不助火,滋阴而不腻膈,收敛而不留邪等特殊功效被历代医学所喜用。张仲景以山茱萸为君创制了“金匮肾气丸”。据化学分析,山茱萸含有生理活性较强的山茱萸甙、酒石酸、没食子酸、苹果酸、树酯、鞣质和多种维生素等有效成分,具有增强免疫、抗炎、抗菌等药理作用,是中医临床中常用的一味药。
丹参苦,微寒。归心、肝经。祛瘀止痛,活血通经,清心除烦。用于月经不调,经闭痛经,症瘕积聚,胸腹刺痛,热痹疼痛,疮疡肿痛,心烦不眠。用于胸肋胁痛,风湿痹痛,症瘕结块,疮疡肿痛,跌仆伤痛,月经不调,经闭痛经,产后瘀痛等。治疗胸肋疼痛、症瘕结块,以及月经不调、经闭经痛具有良效。.《纲目》载:丹参,按《妇人明理论》云,四物汤治妇人病,不问产前产后,经水多少,皆可通用,惟一味丹参散,主治与之相同。盖丹参能破宿血,补新血,安生胎,落死胎,止崩中滞下,调经脉。丹参主要成分丹参酮有扩张血管、改善循环、解除血管内凝血等作用,具有活血化痕之功效,并能促进肝脏血液循环,利于肝细胞再生。
香附辛微苦甘,平功能主治:理气解郁,调经止痛。用于肝郁气滞,胸、胁、脘腹胀痛,消化不良,月经不调,经闭痛经,寒疝腹痛,乳房胀痛。《纲目》载其:散时气寒疫,利三焦,解六郁,消饮食积聚,痰饮痞满,跗肿,腹胀,脚气,止心腹、肢体、头、目、齿、耳诸痛,痈疽疮疡,吐血,下血,尿血,妇人崩漏带下,月候不调,胎前产后百病。
当归,其味甘而重,故专能补血,其气轻而辛,故又能行血,补中有动,行中有补,诚血中之气药,亦血中之圣药。大约佐之以补则补,故能养营养血,补气生精,安五脏,强形体,益神志,凡有形虚损之病,无所不宜。佐之以攻则通,故能祛痛通便,利筋骨,治拘挛、瘫痪、燥、涩等证。营虚而表不解者,佐以柴、葛、麻、桂等剂,大能散表卫热,而表不敛者,佐以大黄之类,又能固表。惟其气辛而动,故欲其静者当避之,性滑善行,大便不固者当避之。凡阴中火盛者,当归能动血,亦非所宜,阴中阳虚者,当归能养血,乃不可少。
柴胡清虚热中药,用于感冒发热、寒热往来、疟疾、肝郁气滞、胸肋胀痛、脱肛、子宫脱落、月经不调。柴胡始载于《神农本草经》,列为上品。历代本草对柴胡的植物形态多有记述。《本草正》:″柴胡,用此者用其凉散,平肝之热。其性凉,故解寒热往来,肌表潮热,肝胆火炎,胸胁痛结,兼治疮疡,血室受热;其性散,故主伤寒邪热未解,温病热盛,少阳头痛,肝经郁证。
五灵脂为复齿鼯鼠的干燥粪便,可用于瘀血内阻、血不归经之出血,如妇女崩漏经多,色紫多块,少腹刺痛。既可单味炒研末,温酒送服,又可与其他药物配合使用。活血散瘀,炒炭止血。用于心腹淤血作痛,痛经,血瘀经闭,产后淤血腹痛;炒炭治崩漏下血;外用治跌打损伤,蛇、虫咬伤。五灵脂苦泄温通,“通利气脉”,“通则不痛。”故《本草经疏》谓之“血滞经脉,气不得行,攻刺疼痛等证,在所必用。”
干姜味辛,性热。归脾、胃、心、肺经。温中散寒,回阳通脉,燥湿消痰,温肺化饮。脘腹冷痛;呕吐;泄泻,亡阳厥逆;寒饮喘咳;寒湿痹痛。《别录》:“治寒冷腹痛,中恶、霍乱、胀满,风邪诸毒,皮肤间结气,止唾血。”
川芎辛,温。归肝、胆、心包经。功能活血行气,祛风止痛。用于月经不调,经闭痛经,症瘕腹痛,胸胁刺痛,跌扑肿痛,头痛,风湿痹痛。川穹性温,是妇科常用药品,有活血顺气的作用。
淫羊藿辛、甘、温,归肝、肾经。能补肾助阳,强筋健骨,祛风除湿,善治肾阳不足,阳痿遗精,遗尿尿频;风湿痹痛,骨痿瘫疾。《本草述》云“淫羊火,《本经》药主阴痿绝伤,《日华子》亦首言其疗男子绝阳,女子绝阴,则谓入命门、补真阳者是也。盖命门为肾中之真阳,即人身之元气也,其所谓绝阳绝阴,不本之元气,何以嘘之于既槁。所谓益气力,强志,并治冷气劳气,筋骨挛急等证,皆其助元气之故。老人昏耄,中年健忘,皆元阳衰败而不能上升者也。须知此味以降为升,其升。
党参为中国常用的传统补益药,古代以山西上党地区出产的党参为上品,具有补中益气,健脾益肺之功效。现代研究,党参含多种糖类、酚类、甾醇、挥发油、黄芩素葡萄糖甙、皂甙及微量生物碱,具有增强免疫力、扩张血管、降压、改善微循环、增强造血功能等作用。此外对化疗放疗引起的白细胞下降有提升作用。党参具有补中益气,健脾益肺的功效。用于脾肺虚弱,气短心悸,食少便溏,虚喘咳嗽,内热消渴等。《本草从新》记载:“补中益气、和脾胃、除烦渴。中气微弱,用以调补,甚为平妥。
鸡血藤性苦、甘,温。归肝、肾经,补血,活血,通络。用于月经不调,血虚萎黄,麻木瘫痪,风湿痹痛活血舒筋;养血调经,《本草纲目拾遗》载其能医手足麻木;肢体瘫痪;风湿痹痛;凌霄花,鸡血藤内服是治疗血淤不散,养血化淤活血的良药。
大枣:补中益气,养血安神。用于脾虚食少,乏力便溏,妇人脏躁。据国外的一项临床研究显示:连续吃大枣的病人,健康恢复比单纯吃维生素药剂快3倍以上。红枣所含有的环磷酸腺苷,是人体细胞能量代谢的必需成分,能够增强肌力、消除疲劳、扩张血管、增加心肌收缩力、改善心肌营养,对防治心血管系统疾病有良好的作用;中医中药理论认为,红枣具有补虚益气、养血安神、健脾和胃等作用,是脾胃虚弱、气血不足、倦怠无力、失眠多梦等患者良好的保健营养品。红枣对慢性肝炎、肝硬化、贫血、过敏性紫癜等病症有较好疗效;红枣含有三萜类化合物及环磷酸腺苷,有较强的抗癌、抗过敏作用。①《本经》:″主心腹邪气,安中养脾,助十二经。平胃气,通九窍,补少气、少津液,身中不足,大惊,四肢重,和百药。
茺蔚子中药名,由唇形科植物益母草的成熟果实风干后而成。具有活血调经、清肝明目的功能。炒后有活血调经之效。炒炒茺蔚子:炒后寒性减弱,并且质地酥脆,易于粉碎和煎出成分。长于活血调经。用于经闭,月经不调,痛经。《本草经疏》:茺蔚子,为妇人胎产调经之要药。此药补而能行,辛散而兼润者也。目者,肝之窍也,益肝行血,故明目益精。其气纯阳,辛走而不守,故除水气。肝脏有火则血逆,肝凉则降而顺矣。大热头痛心烦,皆血虚而热之候也,清肝散热和血,则头痛心烦俱解。
凌霄花一种传统中药材,具有行血去瘀、凉血祛风利湿,清热解毒,味辛酸,性微寒归肝、心包经、功能活血散瘀,凉血祛风。主治月经不调、经闭症瘕、产后乳肿、皮肤瘙痒。《本草衍义补遗》:凌霄花,治血中痛之要药也,且补阴捷甚,盖有守而独行,妇人方中多用何哉。《南宁市药物志》:有消炎解毒、止血止痛、排脓生肌之功,治红白痢。
黄芪具有补气固表、利水退肿、托毒排脓、生肌等功效。现代研究,黄芪含皂甙、蔗糖、多糖、多种氨基酸、叶酸及硒、锌、铜等多种微量元素。有增强机体免疫功能、保肝、利尿、抗衰老、抗应激、降压和较广泛的抗菌作用。但表实邪盛,气滞湿阻,食积停滞,痈疽初起或溃后热毒尚盛等实证。黄芪具有补气固表,利尿托毒,排脓,敛疮生肌的功效。用于气虚乏力,食少便溏,中气下陷,久泻脱肛,便血崩漏,表虚自汗,痈疽难溃,久溃不敛,血虚萎黄,内热消渴。《本经》记载:“主痈疽,久败疮,排脓止痛。补虚,小儿百病。”《日华子本草》记载:“助气壮筋骨,长肉补血。”
牡丹皮又名牡牡丹皮。《本草纲目》载其:“滋阴降火,解斑毒,利咽喉,通小便血滞。后人乃专以黄蘖治相火,不知牡丹皮之功更胜也。赤花者利,白花者补,人亦罕悟,宜分别之。”现代研究,所含牡丹酚及其以外的糖苷类成分均有抗炎作用;牡牡丹皮的甲醇提取物有抑制血小板作用;牡丹酚有镇静、降温、解热、镇痛、解痉等中枢抑制作用及抗动脉粥样硬化、利尿、抗溃疡等作用。
熟地又名熟地黄或伏地,属玄参科植物,是一种上好中药材,具有补血滋阴功效,可用于血虚萎黄,眩晕,心悸失眠,月经不调,崩漏等症,亦可用于肾阴不足的潮热骨蒸、盗汗、遗精、消渴等症,是虚证类非处方药药品六味地黄丸主要成分之一。本品质润入肾,善滋补肾阴,填精益髓,为补肾阴之要药。古人谓之“大补五脏真阴”,“大补真水”。常与山药、山茱萸等同用,治疗肝肾阴虚,腰膝酸软、遗精、盗汗、耳鸣、耳聋及消渴等,可补肝肾,益精髓,如六味地黄丸。
茯苓味甘、淡、性平,入药具有利水渗湿、益脾和胃、宁心安神之功用。现代医学研究:茯苓能增强机体免疫功能,茯苓多糖有明显的抗肿瘤及保肝脏作用。用于小便不利,水肿等症茯苓功能利水渗湿,而药性平和,利水而不伤正气,为利水渗湿要药。凡小便不利、水湿停滞的症候,不论偏于寒湿,或偏于湿热,或属于脾虚湿聚,均可配合应用。如偏于寒湿者,可与桂枝、白术等配伍;偏于湿热者,可与猪苓、泽泻等配伍;属于脾气虚者,可与党参、黄耆、白术等配伍;属虚寒者,还可配附子、白术等同用。
白术味苦、甘,温。归脾、胃经。主治健脾益气,燥湿利水,止汗,安胎。用于脾虚食少,腹胀泄泻,痰饮眩悸,水肿,自汗,胎动不安。白术对瘤细胞有细胞毒作用,能降低瘤细胞的增殖率,减低瘤组织的侵袭性,提高机体抗肿瘤反应的能力。白术挥发油对小鼠艾氏腹水癌、淋巴肉瘤腹水型、食管癌(Ecal09)、肉瘤180等有抑制作用。白术内酯B腹腔注射对小鼠肉瘤民。也有显著抑制作用。《药性论》载其:君,味甘,辛,无毒。能主大风痹,多年气痢,心腹胀痛,破消宿食,开胃,去痰涎,除寒热,止下泄。主面光悦,驻颜,去黑。治水肿胀满,吐呕逆,腹内冷痛,吐泻不住,及胃气虚冷痢。
本发明蜜炼丸剂的制作过程为:取党参1000g,淫羊藿1000g,鸡血藤1000g,炒茺蔚子1000g,凌霄花1000g,益母草1000g,白芍1000g,覆盆子1000g,大枣1000g,当归1000g,山茱萸1000g,黄芪1000g,川芎1000g,女贞子1000g,制首乌1000g,柴胡1000g,丹参1000g,五灵脂1000g,牡丹皮1000g,香附1000g,茯苓1000g,白术1000g,干姜1000g,熟地1000g,取所述原料药浸泡于70%-80%乙醇中,加热回流提取三次,时间1.5小时、1小时、1小时,合并提取液,滤过,滤液回收乙醇,浓缩至干,于60℃下真空干燥,取出,粉碎,过100目筛,细粉备用;加入蜂蜜制成蜜炼丸剂。
本发明胶囊剂的制作过程为:取党参1200g,淫羊藿1300g,鸡血藤1500g,炒茺蔚子1200g,凌霄花1200g,益母草1300g,白芍1400g,覆盆子1300g,大枣1500g,当归1300g,山茱萸1200g,黄芪1200g,川芎1300g,女贞子1400g,制首乌1400g,柴胡1200g,丹参1400g,五灵脂1400g,牡丹皮1300g,香附1300g,茯苓1200g,白术1300g,干姜1100g,熟地1300g,将所述原料放入粉碎机粉碎后过100目细筛,使能通过100目筛的细粉量在达到30%,收集通过120目筛的原料细粉备用;未过筛粗颗粒加入10倍量乙醇中加热提取2次,每次1~2小时,将2次提取液合并静置;将上述乙醇提取过的组份原料药材的药渣加10倍量水加热回流提取1次,每次1~2小时,将提取液合并静置。将上述两种提取液合并,减压回收乙醇并浓缩至药液浓度为0.6g生药/mL,抽滤后,滤液的相对密度约为20℃时1.08;减压至0.03-0.08MPa,温度保持在60-80℃,浓缩至相对密度为1.20,温度至60℃-70℃的浸膏,放入烘箱干燥得到组份药粉,加前述过筛得到的药粉,装入胶囊制为胶囊剂。每粒胶囊装0.3g。可以每12粒为一版。
本发明口服液剂的制作过程为:取党参1300g,淫羊藿1300g,鸡血藤1400g,炒茺蔚子1300g,凌霄花1200g,益母草1300g,白芍1400g,覆盆子1300g,大枣1200g,当归1300g,山茱萸1200g,黄芪1500g,川芎1300g,女贞子1500g,制首乌1300g,柴胡1200g,丹参1300g,五灵脂1200g,牡丹皮1300g,香附1300g,茯苓1200g,白术1300g,干姜1200g,熟地1400g,取原料药加入5-10倍量的乙醇浸泡1-2小时,加热提取2次,每次1-2小时,去上清液,合并提取液,100-120目滤过,再经截流分子量为5000-10000的超滤柱超滤,超滤液减压浓缩相对密度为80℃时1.38的浸膏,加热浓缩30分钟~45分钟,浓缩至浸膏状,静置备用,成为组分1;将水提取过的药渣放入10倍量水中,加热回流提取1次,提取时间为1~2小时,100-120目滤过,再经截流分子量为5000-10000的超滤柱超滤,超滤液减压浓缩相对密度为80℃时1.38的浸膏,加热浓缩30分钟~45分钟,浓缩至浸膏状,静置备用,成为组分2。将上述两种浸膏浓缩成糊状,浓缩液用CO 60灭菌,辐照剂量8k,加入糊精,然后真空罐装密封即得。
本发明片剂制备步骤为:取党参1500g,淫羊藿1200g,鸡血藤1400g,炒茺蔚子1300g,凌霄花1400g,益母草1400g,白芍1400g,覆盆子1400g,大枣1200g,当归1300g,山茱萸1200g,黄芪1500g,川芎1300g,女贞子1500g,制首乌1300g,柴胡1200g,丹参1300g,五灵脂1200g,牡丹皮1500g,香附1300g,茯苓1200g,白术1200g,干姜1100g,熟地1300g,药材加水浸泡30分钟至1小时,加热煮沸30~40分钟,过滤,滤液备用;滤渣加水第二次加热,煮沸30~40分钟,过滤,滤液备用;滤渣再加水,第三次加热煮沸30~40分钟,过滤;将三次滤液合在一起,加热浓缩至糊状药液,放入烘箱80度烘干至粉末状,加适量淀粉成型压片。
药理学毒性试验
急性毒性实验资料
为了全面反映本发明对小鼠短期内的毒性反应,采用小鼠最大给药量试验进行急性毒性试验研究。在本实验条件下,选用ICR品系清洁级小鼠,灌胃给予2.68g生药/ml本发明胶囊剂药液(最大给药浓度40ml/kg.次),连续观察7d,未见小鼠死亡和明显毒副作用,7d后处死动物尸体解剖,肉眼未见明显病理变化,对主要脏器无显著影响。实验结果表明,本发明小鼠的最大给药剂量为107.2g生药/kg,相当于临床拟用量的916倍,在此剂量下小鼠是安全的。
长期毒性试验:
1、实验方法
将豚鼠随机份成4组,每组15只。在豚鼠背部脊柱两侧将脱毛剂均匀涂上,使其脱毛范围约40平方厘米。洗净脱毛剂,观察24小时后每组豚鼠分别涂本发明内服中药制剂胶囊剂0.2、0.4和0.8ml,分别含生药92mg、184mg和368mg,另一组涂溶媒0.8ml每日二次,连续30天,观察豚鼠的一般情况,实验结束后处死动物进行血液学、血液生化及病理学检查。
2、结果
上述三组用药豚鼠躯干去毛区,未见局部皮肤有水肿、充血、红斑、出血点及溃疡。用药组与对照组动物毛发色泽、摄食、四肢活动等对照组无明显差异,血液生化检查,用药组与对照组均在正常范围。病理组织学检查,实验各组心、肝、肾及局部皮肤均未见明显病变。长期毒性试验(一个月)结果表示,整个给药期间,动物行为、运动、精神状态无异常表现,摄食、二便正常,体重增长率各组之间无显著差异,无死亡;尿八项、血常规及血清10项生化指标在各组之间均无统计学意义;各组心率、心电图均正常,各组动物各脏器外观均未发现肉眼可见的病理变化,主要脏器系数各组之间无显著性差异;病理组织学检查,结果各组动物心、肝、脾、肺、肾等17种脏器均未见由药物引起的中毒性病理变化。本发明制剂长期用药对局部皮肤及全身重要脏器均未见明显的毒性作用。试验结果表明:本发明制剂实验安全。
累计毒性实验:
本发明胶囊剂对小鼠按7.69、19.18和43.21g生药/kg连续用药15周(1.0ml/100g体重,每天2次)及停药3周后,结果表明:本发明对大鼠的毛发、行为、大小便、体重、脏器重量、血象、肝肾功能、血糖、血脂等指标均无明显影响,脏器肉眼没有发现异样变化和组织学检查结果表明,用药15周及停药3周后,大鼠各脏器均无明显改变。说明本发明胶囊剂对大鼠长期用药后毒性小,停药后也没有异样反应,应用安全。
药理学实验说明:
1.对去势大鼠雌激素水平的影响
方法:将60只大鼠分成两批供做实验,先随机抽取10只作为空白对照组;其余50只大鼠行双侧卵巢切除术,备用。取上述手术后3个月的大鼠,随机分为5组,再取一组空白对照组大鼠,共6组,分别为空白对照组,摘除卵巢模型组,阳性对照组(摘除卵巢三月后,给己烯雌酚0.02mg/kg+鱼肝油450u/kg+钙0.5g/kg混合液),本发明药9.2,4.6和2.3g/kg组,分组灌胃给药,每日一次,共3个月。给药结束后,腹主动脉取血制备血清,用试剂盒测定血清雌二醇,计算各组数据的均值和标准差,进行组间t测验。
结果:本发明9.2,4.6和2.3g/kg组及阳性对照组均能显著地升高去势大鼠血清雌二醇值,分别与模型组比较,差异均有非常显著的意义(P均<0.001)。模型组与正常对照组比较,血清雌二醇明显降低,差异有非常显著性意义(P均<0.001)。见表1。

2.促进去势大鼠子宫重量的影响
方法:造模,分组和给药均同试验1,将摘除卵巢三个月后给药3个月的大鼠处后,剖取子宫称重,计算各组子宫重量指数的均值和标准差,进行组间t测验。子宫重量指数=子宫重量/体重×100%。
结果:本发明9.2和4.6g/kg组及阳性对照组均能显著地增加去势大鼠子宫重量指数,各组分别与模型组比较,差异均有非常显著的意义(P均<0.001)。模型组与正常对照组比较,子宫重量指数明显降低,差异有非常显著性意义(P<0.001)。见表2。

经药效学实验表明:本发明药具有提高去势大鼠血清中雌二醇的含量,促进子宫发育,增加子宫重量作用。
临床资料:
1资料与方法
1.1一般资料选择2009-2012年我院收治的80例病例均为女性,年龄在18-62岁,平均41.4±6.5岁,病程在2-15年,平均3.5±0.7年。所有病例均符合《黄褐斑的临床诊断及疗效标准》和《中药新药临床研究指导原则》(1993年)制定的月经不调诊断标准。并排除雀斑、老年斑、瑞尔氏黑变病、阿狄森氏病以及避孕药等引起的黄褐斑,非黄褐斑引起的月经不调,有其它严重原发性疾病,准备妊娠或已经妊娠的妇女。通过计算机产生的随机数将患者分为常规治疗组(n=40)和本发明口服液治疗组(n=40)。两组在年龄、病程等方面具有均衡性。
1.2研究方法:常规治疗组外涂维甲酸霜(0.10%,重庆华邦制药有限公司生产),同时口服维生素C和E(200mg/粒×50粒,江苏江山制药有限公司),3次/d;本发明口服液组,方用:党参15g,淫羊藿12g,鸡血藤14g,炒茺蔚子13g,凌霄花14g,益母草13g,白芍14g,覆盆子11g,大枣12g,当归13g,山茱萸12g,黄芪15g,川芎13g,女贞子15g,制首乌13g,柴胡12g,丹参13g,五灵脂12g,牡丹皮15g,香附13g,茯苓12g,白术12g,干姜11g,熟地13g,制成口服液剂,1剂/d,分3次口服。用量可根据病例具体情况适当的加减。治疗3个月后观察各组黄褐斑颜色、面积和月经不调情况的变化,评价和比较各组的疗效。
1.3疗效标准月经总评分标准为四个等级:优秀记为1分,良记为2分,可记为3分,差记为5分。分数越高表明月经不协调,其标准参考1993年卫生部颁布的《中药新药临床研究指导原则(第1辑)》。痊愈:黄褐斑消失,月经周期、经质和经色不调恢复正常;显效:黄褐斑颜色明显减轻、面积显著减小,月经周期、经质和经色不调得到明显改善;有效:黄褐斑颜色减轻、面积减小,月经周期、经质和经色不调有所改善;无效:黄褐斑以及月经不调无改善。总有效=治愈+显效+有效。
1.4统计学处理计量数据显著性检验采用t检验,以均数±标准差(-x±s)表示,计数数据采用卡方卡方检验,设定P<0.05为差异具有统计学意义,所有计算分析采用SAS 9.0统计软件完成。
2结果
2.1两组临床疗效比较
本发明治疗组治愈率和总有效率分别达到了22.50%和92.50%,显著高于常规治疗组,而无效率显著低于常规治疗组(P<0.05)。两组的显效率和有效率差异无显著性(P>0.05)。两组组疗效比较结果见表1。

2.2桃红四物汤对黄褐斑面积的影响治疗前两组患者的黄褐斑面积无显著差异(P>0.05),在相应治疗后各自的黄褐斑面积较治疗前均显著缩小(t=4.38,7.7,P<0.05),本发明治疗组面积缩小较常规治疗组明显,两组组比较差异有显著性(P<0.05)。常规治疗组和本发明治疗组患者治疗前、后黄褐斑面积见表2.

2.3两组月经不调疗效比较治疗前两组患者的月经总评分无显著差异(P>0.05),在相应治疗后各自的月经总评分较治疗前均显著缩小(t=2.67,6.24,P<0.05),本发明治疗组与常规治疗组比较,两组治疗后月经总评分差异有显著性(P<0.05)。常规治疗组和本发明治疗组黄褐斑伴月经不调患者治疗前、后月经总评分见表3。

3讨论
月经病的治疗以调经为本,包括温、清、补、消、调冲任,同时应遵循寒者温之,热者清之,虚者补之,实者泻之的治疗原则,并注意主症及兼症、病变所在部位的不同,采用相应的调经法。本发明对月经失调、经期腹痛、腰腿酸软、赤白带下、小腹虚冷、经来腹痛、寒凝血滞、容颜易老、久不成孕等,均有显著疗效。产后常服,更能培补天真,恢复原来健美身段。少女常服,可助长发育健全、滋补养颜,青春常驻。本发明应用活血化瘀药同时分别加疏肝、温肝、养肝、清肝等调肝药物,具有平衡女性激素分泌、保护卵巢、维持子宫肌弹性不易老化之独特功效。诚为女仕们滋补强身、延展青春、美丽之珍品也

实施例1:贾某,女,32岁,公司职员。初诊时间:2009年5月21日。患者主诉:月经不调4年。现病史:月经周期约28-30天,经期4-5天。末次月经时间:2008年5月6日。此次月经色黑,量少,有血渣,无痛经,行经期间偶有腰酸,盗汗,纳可,眠安,大便干,1-2天一次。舌红少苔,脉细弱无力。辨证:阴血亏虚。治法:养血调经。处方:党参15g,淫羊藿12g,鸡血藤14g,炒茺蔚子13g,凌霄花14g,益母草13g,白芍14g,覆盆子11g,大枣12g,当归13g,山茱萸12g,黄芪15g,川芎13g,女贞子15g,制首乌13g,柴胡12g,丹参13g,五灵脂12g,牡丹皮15g,香附13g,茯苓12g,白术12g,干姜11g,熟地13g,制成口服液,每日三次。二诊时间:2009年8月20日。患者自述服上方3个月后,症状改善。末次月经时间:8月5日,已净,量较前次多,仍有血渣,行经期间偶觉腰酸,盗汗,便质软,日一行。脉弦细无力,舌红少苔,有齿痕。继续服用1个月,患者服药后随访,经期正常,1年无异常。
实施例2:成某,女,35岁。经行腹痛伴未避孕未孕6年余。痛经一般在经前1~2天到月经第2天明显,待月经量渐多,有大血块排出后疼痛缓解。月经30天一潮,量中等、色紫、有血块,伴有经前乳胀,舌红边有紫气,脉弦数。中医诊断为痛经,证属肝郁血瘀。治以疏肝解郁,活血化瘀,方用膈下逐瘀汤加减,但一直断断续续,效果不明显。改用本发明制剂,方用:党参15g,淫羊藿12g,鸡血藤14g,炒茺蔚子13g,凌霄花14g,益母草13g,白芍14g,覆盆子11g,大枣12g,当归13g,山茱萸12g,黄芪15g,川芎13g,女贞子15g,制首乌13g,柴胡12g,丹参13g,五灵脂12g,牡丹皮15g,香附13g,茯苓12g,白术12g,干姜11g,熟地13g,制成颗粒剂,每天三包冲服。嘱经前1周服,连服7天。患者连续3个周期经前1周服药,症状明显缓解,并成功受孕。
实施例3:杨某,女,患者某,女,34岁。初诊时间:2009年2月17日。主诉:月经不调10余年。现病史:月经周期20-25天,经期一般4-5天,末次月经时间:2009年2月15日。此次月经提前5天,有血块,经期5天,白带量多挟血丝。行经期间腰痛,双下肢冷痛。双侧颧部痤疮,经前期加重。疲劳乏力,纳少,眠安,二便调。舌淡苔微黄,脉弦细。辨证:肝气郁结,脾肾阳虚。治法:疏肝解郁,健脾温肾。处方:党参12g,淫羊藿13g,鸡血藤13g,炒茺蔚子13g,凌霄花14g,益母草13g,白芍14g,覆盆子11g,大枣12g,当归13g,山茱萸14g,黄芪15g,川芎13g,女贞子15g,制首乌13g,柴胡12g,丹参13g,五灵脂11g,牡丹皮12g,香附13g,茯苓12g,白术12g,干姜11g,熟地13g,7剂,制成口服液,日三次。二诊时间:2009年2月24日。患者自述服上方后症状减轻。现症大便不成形,日一行。腰腿酸软疼痛,畏寒。舌淡苔微黄腻,舌下青紫,脉弦细。继续服用1个礼拜。三诊时间:2009年3月2日。患者自述服上方后,症状减轻。现舌淡苔微黄,脉弦细。连服三月后复诊,自诉经行或经后腹痛消失,追踪随访3个月,未见复发。

4.根据权利要求1~3中任一项所述补血调经的中药保健品,其特征在于,所述保健品剂型为:糖衣片剂、薄膜衣片剂、肠溶衣片剂、硬胶囊剂、软胶囊剂、口含剂、颗粒剂、冲剂、蜜丸剂、散剂、注射剂、硬膏剂或糖浆剂。
5.一种如权利要求1~3中任一项所述补血调经的中药保健品的制备方法,其特征在于,所述保健品为胶囊剂,其制备方法包括:
a.将所述原料放入粉碎机,粉碎后过筛;
b.将过筛后剩下的颗粒物放入乙醇中加热回流提取2次;
c.再取上述乙醇提取后的药渣,加水加热回流提取1次;
d.将上述两种提取液合并,减压回收乙醇并喷雾干燥,得药粉;混合过筛得到的细粉装入胶囊制为胶囊剂。
6.根据权利要求5所述制备方法,其特征在于,所述步骤a中,将所述原料放入粉碎机粉碎后过100目细筛,使能通过100目筛的细粉量在达到30%,收集通过120目筛的原料细粉备用。
7.根据权利要求5所述制备方法,其特征在于,所述步骤b中,将过筛后剩下的颗粒物放入乙醇中加热回流提取2次,每次1~2小时,将2次提取液合并静置。
8.根据权利要求5所述制备方法,其特征在于,所述步骤c中,将上述乙醇提取过的药渣加10倍量水加热1~2小时回流提取1次;将上述3次提取液合并静置;所述步骤d中,将上述提取液合并,减压浓缩相对密度为80℃时1.06的滤液,回收乙醇,将所得滤液调整比重到1.06,通入喷雾干燥机进行喷雾干燥,过喷雾干燥机喷头的压缩空气压力为4kg/m2,干燥室热气流温度是80℃,得原料药粉;将过筛后得到的原料细粉与乙醇提炼药粉混合装入胶囊。
9.一种如权利要求1~8中任一项所述补血调经的中药保健品的制备方法,其特征在于,所述制剂剂型为口服液剂,其制备方法包括:
a、取原料放入乙醇中浸泡,热提取2次,成膏状,为组分1;
b、药渣水提取2次,浓缩过滤为膏状,为组分2;
c、将上述两种膏状浓缩成糊状,灭菌,加入糊精,然后真空冷冻密封。
10.根据权利要求9所述制备方法,其特征在于,所述步骤a中,取原料加入5-10倍量的乙醇中浸泡1-2小时,加热提取2次,每次1-2小时,去上清液,合并提取液,100-120目滤过,再经截流分子量为5000-10000的超滤柱超滤,超滤液减压浓缩相对密度为80℃时1.36的浸膏,加热浓缩至膏状,静置备用,成为组分1;所述步骤b中,将乙醇提取过的药渣放入10倍量水中,加热回流提取2次,每次提取时间为1~2小时,100-120目滤过,再经截流分子量为5000-10000的超滤柱超滤,超滤液减压浓缩相对密度为80℃时1.36的浸膏,加热浓缩至膏状,静置备用,成为组分2;所述步骤c中,将上述两种组分膏状浓缩成糊状,浓缩液用CO 60灭菌,辐照剂量8k,加入糊精,然后真空罐装密封即得,使用时直接饮用。
