背景技术
随着人类平均寿命的增加,如何长时间保持年轻比如何长时间生存更重要。随着人的年龄增长,老化是一个不可避免的过程。特别是,由于现代社会环境污染和压力越来越严重以及紫外线暴露的增加,老化会开始得比之前要早。
所有人体器官逐渐老化,且皮肤老化受外部环境的影响特别大。绝大多数早期的皮肤老化都始自皮肤干燥。当皮肤变得干燥时,它会失去弹性,且会变得容易产生皱纹。皮肤老化后,皮肤的再生需要很长时间,由于角质化皮肤会变得粗糙,因为胶原合成的减少产生皱纹。由于紫外线防御的减少产生雀斑、活斑等色素沉着,因为皮肤保护的减弱,皮肤变得易受药物或外部环境刺激的攻击。

在本发明的一个典型实施例中,所述皮肤护理组合物包括香豆雌酚、含有香豆雌酚的天然物质或其提取物。在本发明的另一个典型实施例中,含有香豆雌酚的天然物质可以是选自大豆、豌豆、绿豆和从这些豆发出的豆芽、红三叶草(苜蓿)、抱子甘蓝或类似物中的一种或多种豆。在本发明的另一个典型实施例中,含有香豆雌酚的天然物质可以是豆。

本领域的技术人员能确定所述活性成分的剂量。例如,该剂量可以是0.1-5000mg/kg/天,优选50-500mg/kg/天,但并不仅限于此,且该剂量可因治疗对象的年龄、生理条件、并发症等因素的不同而有变化。
将通过测试实施例对本发明的特征和作用进行详细描述。但是,以下测试实施例仅用作说明用途,并非为了限制本发明的范围。
[测试实施例1] 对UVB-诱导的活性氧自由基(ROS)的清除作用的评价
用2’,7’-二氯二氢荧光素二乙酯(DCFH-DA,Molecular Probes公司,俄勒冈州,美国)来评价香豆雌酚对UVB-诱导的活性氧自由基(ROS)的清除作用。DCFH-DA是一种脂溶性物质,已知作为细胞中ROS的检测剂。它能很容易被细胞吸收,并在细胞中酯酶的作用下,发生乙酸酯基团的水解,转化成2’,7’-二氯二氢荧光素。由于2’,7’-二氯二氢荧光素与细胞内的氧化剂反应后转化成发出荧光的2’,7’-二氯荧光素(DCF),所以可通过测定用DCFH-DA处理的细胞的荧光强度来评价细胞内ROS的数量。
将角蛋白细胞(从德国癌症研究中心的Fusenig博士处获得)接种到96孔黑色板(Greiner BioOne公司,北卡罗莱纳州,美国)上用于荧光测试,密度为每孔1x104个细胞,培养24小时。接着,用在新鲜的无FBS的培养基中的、稀释至最终浓度为0.01、0.1或1μM的香豆雌酚处理所述细胞。加入测试物质后,培养细胞24小时。在用HEPES缓冲盐溶液(HCSS)冲洗去除剩余的培养基之后,加入DCFH-DA和HCSS混合液(DCFH-DA:20μM,HCSS:100μL)。将所述细胞在37℃和5%CO2条件下,培养20分钟后,用HCSS冲洗细胞。接着,加入100μL 的HCSS,用30mJ/cm2的UVB辐照细胞,以诱导产生ROS。随后,用在HCSS中稀释至相同浓度的香豆雌酚,在37℃和5%CO2条件下,培养细胞3小时。香豆雌酚用Wallac VICTOR2荧光计(Wallac,土尔库,芬兰)在激发波长485nm和发射波长530nm的条件下,测定DCF的荧光强度。通过测定的荧光强度,计算相对于对照组的ROS清除作用(%)。结果见表1。

由表1可以看出,香豆雌酚具有清除UVB-诱导的ROS的作用,其作用随着香豆雌酚浓度的增加而增加。因此,含有香豆雌酚的组合物可提供抑制由UV引起的皮肤氧化,抑制光老化和美白皮肤的作用。
[测试实施例2] 对UV-诱导的MMP-1表达的抑制作用的评价
用人体成纤维细胞(CAS-C-004-5C,Invitrogen,加利福尼亚州,美国)来评价香豆雌酚对UV-诱导的MMP-1表达的抑制作用。将细胞接种到24孔板,密度为每孔1x104个细胞,在含有10%FBS的培养基中培养约24小时,直至约80%的铺满。用无FBS的培养基替代之前的培养基之后,培养细胞24小时。用PBS冲洗1次后,加入100μL PBS,用15 mJ/cm2的UVB辐照细胞。去除PBS后,用在新鲜的无FBS的培养基中的、稀释至最终浓度为0.01,0.1或1μM的香豆雌酚处理细胞。用测试物质处理48小时后,评价MMP-1的表达。从细胞培养物中收集上清液后,用酶联免疫吸附测定(ELISA)试剂盒(RPN 2610,Biotrak Amersham PharmaciaBiotech,英国)来评价MMP-1的表达。通过总蛋白量来测定MMP-1的数量。计算MMP-1的活性(%),结果见表2。

由表2可以看出,香豆雌酚具有抑制UV-诱导的MMP-1活性的能力,且该能力随着香豆雌酚浓度的增加而增强。因此,含有香豆雌酚的组合物,可通过抑制胶原蛋白的损坏,来改善皮肤弹性和皱纹。
[测试实施例3] 对I型pN-胶原蛋白合成的效果评价
用人体成纤维细胞(CAS-C-004-5C,Invitrogen,加利福尼亚州,美国)来评价香豆雌酚对I型pN-胶原蛋白的合成作用。将细胞接种到24孔板,密度为每孔1x104个细胞,在含有10%FBS的培养基中培养约24小时,直至约80%的铺满。用在新鲜的无FBS的培养基中的、稀释至最终浓度为0.01,0.1或1μM的香豆雌酚处理所述细胞。在用测试物质处理48小时之后,从细胞培养物中收集上清液,用IC型前胶原-缩氨酸(PIP)EIA试剂盒(MK 101,TakaraShuzo,京都,日本)来测定I型pN-胶原蛋白的合成数量。通过总蛋白量来测定I型pN-胶原蛋白的数量。计算I型pN-胶原蛋白的合成数量,结果见表3。

由表3可以看出,香豆雌酚促进了I型pN-胶原蛋白的合成。因此,含有香豆雌酚的组合物可改善皮肤弹性和皱纹。
[测试实施例4] 对皮肤细胞糖化的抑制作用的评价
用人体成纤维细胞(CAS-C-004-5C,Invitrogen,加利福尼亚州,美国)来评价香豆雌酚对皮肤细胞糖化的抑制作用。将细胞接种到6孔板,密度为每孔5x104个细胞,在含有10%FBS的培养基中培养约24小时,直至约80%的铺满。用PBS冲洗1次后,加入100μL PBS,用50mJ/cm2的UVB辐照细胞。去除PBS后,用在新鲜的无FBS的培养基中的、稀释至最终浓度为2.5μM的香豆雌酚处理所述细胞。用测试物质处理24小时后,用OxiselectTM高级糖化终产物(AGE)ELISA试剂盒来评价皮肤细胞的糖化。测定糖化终产物的浓度,如表4所示。

由表4可以看出,香豆雌酚抑制了皮肤细胞的糖化。因此,含有香豆雌酚的组合物可改善皮肤弹性和提供美白皮肤的作用。
[测试实施例5] ATP生成作用的评价
在对人体成纤维细胞(CAS-C-004-5C,Invitrogen,科罗拉多州,美国)进行测试实施例4的处理之后,用ATP测定试剂盒(Invitrogen,A22066)评价细胞中ATP的生成。测定的ATP的浓度如表5所示。

由表5可以看出,香豆雌酚促进了细胞中ATP的产生。因此,含有香豆雌酚的组合物可促进皮肤再生,通过促进皮肤细胞的能量代谢改善皮肤弹性和活化皮肤细胞。
[测试实施例6] 对黑色素生成的抑制作用的评价
将取自C57BL/6小鼠的小鼠黑素细胞(Mel-Ab细胞),在37℃和5%的CO2中,含有10%FBS、100nM 2-氧-十四烷酰佛波醇-13-乙酸酯(2-O-tetradecanoylphorbol-13-acetate)和1 nM霍乱毒素的Dulbecco改良的Eagles培养基(DMEM)中培养。用0.25%胰蛋白酶-EDTA分离培养后的Mel-Ab细胞,并在24-孔板上培养,每孔105个细胞。从第二天开始,连续3天加入0.01、0.1或1μM的香豆雌酚。接着,去除培养基后,用PBS冲洗,将细胞悬浮在1N的氢氧化钠中,测定400nm处的吸光度。用10μM的对苯二酚作为阳性对照组,已知其具有卓越的黑色素的生成抑制作用。根据测定的吸光度计算黑色素生成抑制作用(%),结果见表6。

由表6可以看出,香豆雌酚具有极佳的黑色素生成抑制作用。因此,含有香豆雌酚的组合物可具有卓越的皮肤美白作用。
[测试实施例7] 对前列腺素E2(PGE2)生成的抑制作用的评价
按如下方法测试香豆雌酚对前列腺素E2生成的抑制作用。将人体成纤维细胞(CAS-C-004-5C,Invitrogen,加利福尼亚州,美国)在CO2培养箱中培养24小时。用新鲜的无FBS的培养基替换培养基之后,培养2小时,然后用在新鲜的无FBS的培养基中的、稀释至最终浓度为0.01,0.1或1μM的香豆雌酚处理细胞。然后将细胞在并在37℃和5%(体积比)CO2培养箱中培养2小时。接着,用50μM的钙离子载体A23187和花生四烯酸处理细胞5分钟以刺激前列腺素E2,细胞裂解,用ELISA读数器测定450nm处的吸光度来定量前列腺素E2,结果见表7。计算相对于未经香豆雌酚处理组的抑制作用(%)。测定5次,结果取平均值。

由表7可知,香豆雌酚可抑制前列腺素E2的生成。因此,含有香豆雌酚的组合物具有消炎作用,可防止皮脂分泌、缓解皮肤问题、改善痤疮和皮肤过敏。
[测试实施例8] 对脂多糖类(LPS)诱导的环氧合酶-2(COX-2)生物合成的抑制作用的评价
用人体成纤维细胞(CAS-C-004-5C,Invitrogen,加利福尼亚州,美国)评价香豆雌酚对COX-2生物合成的抑制作用。将细胞接种到6孔板,密度为每孔5x104个细胞,在含有10%FBS的培养基中培养约24小时,直至约80%铺满(80% confluency)。用在新鲜的无FBS的培养基中的、稀释至最终浓度为0.01,0.1或1μM的香豆雌酚处理细胞,然后在37℃和5%(体积比)CO2培养箱中培养24小时。收获细胞,并用蛋白质印迹法对产生的COX-2进行定量。计算相对于对照组的COX-2表达水平(%),结果见表8。

由表8可知,香豆雌酚抑制了LPS诱导的COX-2的生物合成。因此,含有香豆雌酚的组合物可提供消炎作用,改善痤疮和皮肤过敏。
[测试实施例9] 对特应性皮炎的改善作用的评价
将诊断为具有过敏性症状(发痒、水疱或严重的干燥)或表现出遗传性过敏症状的10岁-50岁的30人分成2组,分别给予含有0.1%香豆雌酚的软膏或不含香豆雌酚的软膏(对照组)。要求他们在其过敏性症状的部位涂敷上述软膏连续12周,每天两次。12周后,通过调查问卷检验过敏性症状的改善。检查到特应性皮炎的典型症状包括“发痒”、“干燥”、“角质化”、“头皮屑”、“红斑”、“肿胀”、“皮肤裂”、“分泌物和湿疹”的改善。让受试者为症状的改善情况打分,取平均值来评价特应性皮炎的改善,结果见表9。

由表9可以看出,香豆雌酚和含有香豆雌酚的组合物具有改善特应性皮炎的作用。
[测试实施例10] 对皮肤保湿的改善作用的评价
为了评价皮肤保湿能力的改善,在恒温、恒湿(24℃,相对湿度40%)条件下,用皮肤水分测试仪测定测试实施例9中受试者的经皮水分损失,结果取平均值,见表10。

由表10可以看出,香豆雌酚能抑制经皮水分的损失,且随着时间的延长,抑制作用会增加。因此,含有香豆雌酚的组合物可提供皮肤保湿的作用。
下面将描述本发明组合物的剂型实施例,但是本发明的范围并不仅局限于以下实施例。
[剂型实施例1] 柔肤乳液
根据通常采用的方法,用表11中描述的成分制备柔肤乳液。

根据通常采用的方法,用表12中描述的成分制备滋养乳液。

根据通常采用的方法,用表13中描述的成分制备滋养霜。

[剂型实施例4] 片剂
将香豆雌酚(80mg)、维生素E(9mg)、维生素C(9mg)、低聚半乳糖(200mg)、乳糖(60mg)和麦芽糖(140mg)混合,用硫化干燥床造粒。加入糖酯(6mg)后,根据通常采用的方法,将所得的组合物(500mg)制备成片剂。
[剂型实施例5] 饮剂
将香豆雌酚(80mg)、维生素E(9mg)、维生素C(9mg)、葡萄糖(10g)、柠檬酸(0.6g)和寡糖糖浆(25g)混合。加入纯净水(300mL)后,将200mL所得混合物填充到瓶中。接着,在130℃消毒4-5秒便制成饮剂。
本领域的技术人员可以理解在本发明的范围内,可对本发明进行各种更改或修改。
Claims (3)
1.香豆雌酚或者包含香豆雌酚的豆提取物在制备用于缓解或改善皮肤过敏的皮肤护理化妆品组合物中的应用,
其中,所述皮肤过敏是由于皮肤的成纤维细胞中COX-2或前列腺素E2的增加引起的。
2.根据权利要求1所述的应用,其特征在于,所述组合物含有占所述组合物总重量的0.001-30wt%的香豆雌酚或含有香豆雌酚的豆提取物。
3.根据权利要求1或2所述的应用,其特征在于,所述组合物为化妆品组合物。
