本发明涉及阿胶在制备改善发质的药物中的应用
背景技术
目前改善发质及生发的药物很多,大多认为病因是头发根部出现了问题,许多防脱发的药物也都是直接在头部用药。但效果往往很难持续。
中医理论认为“发为血之余”,也就是说毛发的生长与质地是靠肝血和肾精的滋养,发质枯黄,毛囊萎缩,头发脱落,易断这些外在表现,根源在毛发缺乏肝血和肾精的滋养有关。因此有必要寻找一种既能补肝血,又能补肾精,还能为毛发的生长代谢提供必需的营养物质的药物。
发明内容
本发明的目的在于提供阿胶在制备改善发质的药物中的应用,特别是阿胶在制备生发的药物中的应用以及阿胶在制备提升皮肤毛囊数量的药物中的应用。
本发明阿胶阿胶为马科动物驴EquusasinusL.的干燥皮或鲜皮经煎煮、浓缩制成的固体胶。制法为:将驴皮浸泡去毛,切块洗净,分次水煎,滤过,合并滤液,浓缩(可分别加入适量的黄酒、冰糖及豆油)至稠膏状,冷凝,切块,晾干,即得。
本发明通过小鼠活体试验,进一步说明了阿胶在制备改善发质的药物中的应用,特别是阿胶在制备生发的药物中的应用以及阿胶在制备提升皮肤毛囊数量的药物中的应用。
本发明的优点在于:
1、本发明提供了阿胶新的医疗用途,开拓了新的应用领域;
2、阿胶对毛发的滋养作用明显,主要表现为:阿胶可以增加环磷酰胺致小鼠贫血模型体内的白细胞数量和种类;氨基酸含量测定结果显示有提高动物毛发中氨基酸含量的作用;毛发生长长度及毛囊数量结果显示有促进毛发生长及提升毛囊数量的作用。
3、阿胶原料来源于食材,服用安全,使用历史悠久,适合于不同年龄阶段的需要改善发质的患者服用。

具体实施方式
以下通过实施例进一步说明本发明,但不用来限制本发明。
实施例1阿胶的制备
将侵泡处理后的驴皮切块,置蒸汽化皮机内,第一次加1.5倍水,加热使蒸汽化皮机内温度达117℃-119℃之间,保持120分钟后放出胶汁,重复化皮3次,将三次胶汁过滤、离心后蒸发。将蒸发后胶液置夹层锅内继续浓缩(可分别加适量黄酒、冰糖及豆油)至稠膏状,冷凝,切块,晾干,即得。
以下通过药理实验,进一步说明本发明。
试验例1
1材料与动物
1.1材料:东阿阿胶(山东东阿阿胶股份有限公司提供,批号:1211037);注射用环磷酰胺(山西普德药业股份有限公司提供,批号:04110603);松香、石蜡和生理盐水(山东齐都药业有限公司提供,批号:2011112603)。
1.2实验动物120只SPF级C57BL/6雌性小鼠,15~20g(北京华阜康生物科技股份有限公司提供,生产许可证号:SCXK(京)2009-0004);小鼠饲料(北京华阜康生物科技股份有限公司提供,生产许可证号:SCXK(京)2009-0008)。
1.3环境条件山东省实验动物中心屏障环境动物室,使用许可证号:SYXK(鲁)20090014,室温20~25℃,相对湿度50~70%。
2方法
2.1试剂配制将东阿阿胶制成粉末后,用适量蒸馏水溶解,配制成剂量为135mg/mL,67.5mg/mL,33.75mg/mL的溶液,分别标记为高、中、低剂量溶液。
2.2实验动物预处理将松香和石蜡按1:1比例混合溶解,涂抹至小鼠背部皮肤(2×3cm2),待其凝固后拔除涂抹区域皮肤表面的毛发。
2.3试验分组经脱毛24h后,将75只脱毛区域光滑无皮肤破损小鼠随机分为5组,分别记为高、中、低剂量组,阴性对照和模型对照组,15只/组。
2.4建立模型阿胶试验组和模型对照组,按250mg/100g·bw的剂量一次性腹腔注射环磷酰胺,给药量为0.2mL/10g·bw;阴性对照组,按0.2mL/10g·bw给药量一次性腹腔注射生理盐水,建立试验模型。
2.5给药各试验组经口灌胃对应剂量的阿胶溶液,每日灌胃量0.2mL/10g·bw;阴性对照组和模型对照组经口灌胃蒸馏水,每日灌胃量0.2mL/10g·bw。各组均连续给药20d。
2.6试验步骤对C57BL/6小鼠进行预处理24h后,挑选75只脱毛区域光滑无皮肤破损小鼠分组,建立小鼠贫血模型,灌胃给药,如2.5中所述。分别在给药4d、12d、20d时,采集各组小鼠血液和脱毛区域的皮肤组织,进行血液学检测和组织病理学检查。在给药后20d,收集各组小鼠的毛发样本,借助电子显微镜进行形态学观察,并检测各组动物剩余新生毛发的氨基酸含量。
3结果
3.1血液学项目白细胞、红细胞、血红蛋白、红细胞压积、血小板。
3.2毛发检测指标长度、形态、氨基酸。
3.3组织病理检查皮肤完整性,充血、水肿、炎细胞浸润程度;进行毛囊计数并统计。
3.4血液学指标检查结果
小鼠血液学指标检查结果统计表显示,试验组及阴性对照组白细胞及其分类数量大于模型对照。各剂量组间存在显著及极显著差异(*P<0.05,**P<0.01)。结果见表1,表2和表3。

3.5毛发中氨基酸含量测定
从表4中可以看出,高、中剂量组动物毛发中氨基酸含量数值均大于模型对照。

3.6小鼠毛发生长状况
从小鼠毛发长度统计结果(结果见表5)和毛发生长照片(见图1-图3)中可以看出,20d时处死小鼠的毛发长度及浓密程度均优于模型对照但不及阴性对照组。试验组与阴性对照组动物毛发长度无显著差异(P>0.05),模型对照与阴性对照组动物毛发长度差异及显著(P<0.01)。

3.7电镜结果
电镜观察结果显示,模型对照动物新生毛发出现竹节样改变,阴性对照组动物毛发生长情况正常,试验组动物给药后的结果出现组间差异,各组间毛发直径差异明显,阴性对照组毛发生长情况良好,试验组间毛发直径高剂量组明显大于中、低剂量组,试验组毛发直径明显大于模型对照。
3.8病理检查结果
从表6和图4中可以看出,各剂量组动物病理观察未见充血、水肿及炎细胞浸润。试验组动物毛囊数量统计结果大于模型对照但小于阴性对照组。试验组动物毛囊数量与阴性对照组动物毛囊数量差异显著(P<0.05),模型对照动物毛囊数量与阴性对照组动物毛囊数量差异极显著(P<0.01)。

4结论
试验过程中,动物毛发生长情况在各组之间出现明显差异,试验组及模型对照动物皮肤在4d时变黑,阴性对照组动物于4d时皮肤颜色仍为粉色,试验组中高剂量组动物20d时毛发浓密程度明显优于低剂量组及对照组,但不及阴性对照组。
上述试验表明:阿胶可以增加环磷酰胺致小鼠贫血模型体内的白细胞数量和种类;氨基酸含量测定结果显示有提高动物毛发中氨基酸含量的作用;毛发生长长度及毛囊数量结果显示有促进毛发生长及提升毛囊数量的作用。
综上,通过整体试验结果分析,阿胶可以通过多方面综合作用,促进贫血小鼠的毛发生长及改善毛发质量的作用。
虽然,上文中已经通过一般性说明、具体实施方案以及活性实验等对本发明作了详尽的描述,但在本发明基础上,可以对之作一些修改或改进,这对本领域技术人员而言是显而易见的。因此,在不偏离本发明精神的基础上所做的这些修改或改进,均属于本发明要求保护的范围。

1.阿胶在制备提升皮肤毛囊数量的药物中的应用,所述阿胶为马科动物驴EquusasinusL.的干燥皮或鲜皮经煎煮、浓缩制成的固体胶。
