Abstract
本发明提供了柴石退热颗粒的新用途,柴石退热颗粒由如下重量份数的原料制备得到:柴胡315g、黄芩315g、石膏630g、青蒿252g、板蓝根315g、金银花315g、大黄210g、蒲公英315g、知母210g、连翘210g,并公开了其制备方法。
Description
技术领域
本发明涉及中药技术领域,具体涉及柴石退热颗粒的提取方法和新用途。
背景技术
肝脏是合成血浆蛋白的主要场所,除全部的血清白蛋白外,还有部分球蛋白也在肝脏合成。肝硬化时,肝脏功能的障碍加上营养不良等因素导致白蛋白合成障碍,肾脏损害使白蛋白丢失和消耗增多,从而使血清白蛋白含量下降。因此提高肝脏蛋白质含量的药物尤为重要。
发明内容
发明目的:为了解决上述问题,本发明的目的在于提供柴石退热颗粒的提取方法和新用途。
技术方案:本发明的目的是通过如下的方案实现的:
柴石退热颗粒在制备提高肝脏蛋白质合成药物中的应用。
柴石退热颗粒由如下重量份数的原料制备得到:柴胡315g、黄芩315g、石膏630g、青蒿252g、板蓝根315g、金银花315g、大黄210g、蒲公英315g、知母210g、连翘210g。
柴石退热颗粒的制备方法为:
(1)将大黄热浸三次,热浸温度为85-90℃,每次加8倍水量,每次半小时,合并浸出液,滤液备用;
(2)将生石膏粉碎成粉与柴胡、黄芩、青蒿、板蓝根、金银花、大黄、蒲公英、知母、连翘加水煎煮两次,第一次煎煮加15倍水,煎煮1.5小时,第二次煎煮加12倍水,煎煮1小时,合并煎煮液,滤过备用;
(3)将上述的浸出液和煎煮液合并,采用真空浓缩至相对密度为1.17-1.18/35℃的清膏;
(4)采用转速为23000-25000r/min的高速离心机进行分离将清膏除杂,然后喷雾干燥成干粉,按8g/包进行包装即成。
本发明中原料均符合中国药典标准。
有益效果:柴石退热颗粒能提高肝脏蛋白质合成。

以下通过实施例形式,对本发明的上述内容再作进一步的详细说明,但不应将此理解为本发明上述主题的范围仅限于以下的实例,凡基于本发明上述内容所实现的技术均属于本发明的范围。
一、制备实施例
实施例1:称取柴胡315g、黄芩315g、石膏630g、青蒿252g、板蓝根315g、金银花315g、大黄210g、蒲公英315g、知母210g、连翘210g,将大黄热浸三次,热浸温度为85-90℃,每次加8倍水量,每次半小时,合并浸出液,滤液备用;将生石膏粉碎成粉与柴胡、黄芩、青蒿、板蓝根、金银花、大黄、蒲公英、知母、连翘加水煎煮两次,第一次煎煮加15倍水,煎煮1.5小时,第二次煎煮加12倍水,煎煮1小时,合并煎煮液,滤过备用;将上述的浸出液和煎煮液合并,采用真空浓缩至相对密度为1.17-1.18/35℃的清膏;采用转速为23000-25000r/min的高速离心机进行分离将清膏除杂,然后喷雾干燥成干粉,按8g/包进行包装即成。
对照实施例1:称取柴胡315g,加水煎煮两次,第一次煎煮加15倍水,煎煮1.5小时,第二次煎煮加12倍水,煎煮1小时,合并煎煮液,滤过备用;采用真空浓缩至相对密度为1.17-1.18/35℃的清膏;采用转速为23000-25000r/min的高速离心机进行分离将清膏除杂,然后喷雾干燥成干粉,按8g/包进行包装即成。
对照实施例2:称取黄芩315g,加水煎煮两次,第一次煎煮加15倍水,煎煮1.5小时,第二次煎煮加12倍水,煎煮1小时,合并煎煮液,滤过备用;采用真空浓缩至相对密度为1.17-1.18/35℃的清膏;采用转速为23000-25000r/min的高速离心机进行分离将清膏除杂,然后喷雾干燥成干粉,按8g/包进行包装即成。
上述喷雾干燥条件均为进风温度为110℃,出风温度为85℃,物料温度为85℃,雾化压力为0.3兆帕,喷雾速度为7.5ml/s。
二、效果实施例:柴石退热颗粒对大鼠肝脏蛋白质合成的研究
2.1实验动物:SD大鼠,南京医科大学实验动物中心,质量150g-180g。
2.2药品与试剂:柴石退热颗粒按上述制备实施例中实施例1方法制备得到,柴胡颗粒、黄芩颗粒分别按照对比实施例1和对比实施例2方法制备得到。总蛋白(TP)、白蛋白(ALB)、球蛋白(GLB)试剂盒购自南京建成生物试剂研究所。
2.3实验方法:
2.3.1分组及处置:50只大鼠适应性喂养1周后,按体重分层,随机分为5组,每组10只:空白组、实施例1高剂量组、实施例1低剂量组,对照实施例1组、对照实施例2组,均饲喂普通饲料,给予各组大鼠相应的药物干预,其中空白组按lmL/100g给予蒸馏水灌胃,按生药量计算,实施例1高剂量组给予20g/kg、实施例1低剂量组给予10g/kg,对照实施例1组给予20g/kg、对照实施例2组给予20g/kg,每日一次,直至10周结束,实验期间动物自由进食和进水,每周称取体重一次并观察动物进食、饮水、行为、精神状态、毛发及二便等情况。到期处死动物(处死前禁食12h),腹主动脉采血,取动物肝脏,称重。
2.3.2指标检测与观察:实验结束后处死各组大鼠(处死前禁食12h),腹主动脉采血,采用全自动生化分析仪测定总蛋白(TP)、白蛋白(ALB)、球蛋白(GLB),各项指标检测结果见表1。

2.3.3结果表明:本发明实施例1高剂量组、实施例1低剂量组能提高大鼠的血清总蛋白TP(P<0.05)、白蛋白和球蛋白(P<0.01)水平均较空白组明显增高,与对比实施例1和对比实施例2有显著性差异(P<0.05),说明用柴胡配伍黄芩、石膏、青蒿、板蓝根、金银花、大黄、蒲公英、知母、连翘,或者用黄芩配伍柴胡、石膏、青蒿、板蓝根、金银花、大黄、蒲公英、知母、连翘均取得意想不到的效果。
Claims (2)
1.柴石退热颗粒在提高肝脏蛋白质合成药物中的应用,其特征在于,柴石退热颗粒由如下重量份数的原料制备得到:柴胡315g、黄芩315g、石膏630g、青蒿252g、板蓝根315g、金银花315g、大黄210g、蒲公英315g、知母210g、连翘210g。
2.如权利要求1所述柴石退热颗粒在提高肝脏蛋白质合成药物中的应用,其特征在于,柴石退热颗粒的制备方法为:
(1)将大黄热浸三次,热浸温度为85-90℃,每次加8倍水量,每次半小时,合并浸出液,滤液备用;
(2)将生石膏粉碎成粉与柴胡、黄芩、青蒿、板蓝根、金银花、大黄、蒲公英、知母、连翘加水煎煮两次,第一次煎煮加15倍水,煎煮1.5小时,第二次煎煮加12倍水,煎煮1小时,合并煎煮液,滤过备用;
(3)将上述的浸出液和煎煮液合并,采用真空浓缩至相对密度为1.17-1.18/35℃的清膏;
(4)采用转速为23000-25000r/min的高速离心机进行分离将清膏除杂,然后喷雾干燥成干粉,按8g/包进行包装即成。
