Abstract
一种山药配方颗粒，包括山药饮片3000～5000重量份、糊精或乳糖或麦芽糊精或它们的组合物250～640重量份和硬脂酸镁0.5～1.5重量份。其制备方法是以山药药材为原料，经清洗切片后，用温水浸泡润透，再采用动态循环提取方法提取，提取液合并，浓缩，喷雾干燥，得到山药提取物，加入辅料混合均匀，制粒，制得山药配方颗粒，山药配方颗粒每克相当于山药饮片3～5克。本发明的山药配方颗粒服用方便、质量可控、易保存、便携带。其制备方法先用合适温度的水浸泡饮片，能有效的防止饮片表层糊化，提高水分渗透药材效果，再采用低温强制动态循环提取技术，提高了药材有效成分的溶出效率。
Description
技术领域
本发明涉及一种山药配方颗粒及其制备方法，属于中药配方颗粒制备技术领域。
背景技术
山药为薯蓣科( Dioscor eaceae ) 植物薯蓣( Dioscorea opposita Thunb. ) 的块茎。山药性平，味甘, 具有补脾养胃，生津益肺，补肾涩精的功能，不仅是健脾补气的良药, 也是传统的保健食品。山药是最早被列入本草的中药之一,《神农本草经》中列为上品。其用法很多, 可单味应用, 也可与其他补品或药物配合，有煎剂、散剂、成药、药粥及菜肴等，能健脾、补肺、固肾、益精，用于治疗脾胃虚弱、食少倦怠、小儿营养不良虚劳咳嗽、糖尿病等多种疾病。近年研究表明, 山药不仅能健脾胃,益肺肾,还具有降糖、抗衰老、调节免疫、抗突变、降血脂等新功能。山药主要成分有淀粉、多糖、蛋白质等成分，其中山药多糖是目前公认的主要有效成分。
中药配方颗粒是以中药饮片为原料，利用现代生产技术，按照一定生产工艺制成仅供临床灵活调剂使用的颗粒剂。它是以中药汤剂的临床使用为基础，在中医理论指导下使用，但与传统的中药汤剂相比，它具有服用方便、易储藏、便携带、质量可控等特点。山药配方颗粒属于中药配方颗粒的一种，是以山药药材为原料制成的颗粒剂，可根据医生处方灵活调配。山药在医生处方中大量使用，在中药成药中也经常使用。按照传统提取煎煮方法，是将山药切片后直接投入提取罐中，加冷水浸泡后煎煮提取。由于山药饮片在提取罐中堆积在一起，采用冷水浸泡，由于山药片较厚，且含有大量的淀粉等物质，冷水无法迅速和充分的渗透进饮片内，渗透效果差；冷水浸泡后煎煮，由于升温迅速，热水未能及时和完全渗入山药饮片内部，表面就会因温度过高而迅速糊化，阻止了水分的进一步渗入，影响提取效果；而浸泡时采用温度较高水，易致药材表面糊化，也不利于水分渗透；如山药切片太薄，则煎煮中易碎，由于其含有大量的淀粉，加热中易糊化焦化，在生产中极易堵塞管道无法过滤药液；因此，传统的提取方法在煎煮中其往往不能煎煮透心，有效成分得不到提取完全，药材提取效率低，药材资源浪费大，并且由于提取温度较高，提取液易糊化焦化，严重影响产品的质量问题。目前，也还未见相关山药配方颗粒低温动态循环提取方法的文献报道。
发明内容
本发明提供了一种山药配方颗粒及其制备方法，该方法提取效率高，能源消耗低，操作简便，易于控制，提取液不会出现糊化、焦化的现象，可在大生产中广泛使用。
本发明山药配方颗粒包括以下重量份数的原料：
山药饮片 3000～5000份；
糊精或乳糖或麦芽糊精或它们的组合物 250～640份；
硬脂酸镁0.5～1.5份。
以上所述的山药配方颗粒的制备方法，是取山药药材，经清洗炮制后切片，切片厚度2～10mm；在6～10小时内投入提取罐吊篮中，加入5～10倍的山药重量的40～60℃水浸泡，浸泡时间为1～2小时；在90℃～97℃常压下用动态循环设备进行动态循环提取方法，提取时间2～4小时，滤过，滤液浓缩，加辅料糊精或乳糖或麦芽糊精或它们的组合物，混合，干燥，得到山药提取物，粉碎，再加硬脂酸镁，制粒，制得山药配方颗粒。
以上所述的动态循环提取方法是提取过程中，不断的把提取罐内的液体用泵通过管道从底部抽到打回提取罐上部进行动态循环流动。
以上所述的动态循环设备包括提取罐、循环泵和循环管，提取罐顶部有罐盖，罐体内部安装有吊篮，罐体底部有下降管，下降管接循环泵，罐体上部安装油气上升管，通到油水分离器，分离出挥发油以后蒸馏水从回流管返回罐体内。分离出的挥发油含皂甙、精氨酸、尿囊素、胆硷等，可以作为美容产品。
本发明的提取原理是：在提取过程中，不断的把提取罐内的液体从提取罐底部，用泵通过管道从提取罐的上部从新打回提取罐，通过液体的循环流动，增强固-液两相之间的接触，增强有效成分从药材内部溶出。为了避免药材堵塞提取罐底部管道，提取罐设计成罐中罐的方式，即提取罐中有3个吊篮，药材装于吊篮中，再将吊篮放置于提取罐中，而非药材直接放入提取罐。
本发明得到的产品为山药配方颗粒，其味甘、性平，归脾、肺、肾经，具有补脾养胃、生津益肺、补肾涩精等功能，用于脾虚食少、久泻不止、肺虚喘咳、肾虚遗精、带下、尿频、虚热消渴等。麸炒山药补脾健胃，用于脾虚食少，泄泻便溏，白带过多。
本发明的优点：
1、将山药制成服用方便、质量可控、易保存、便携带、可按医生处方灵活调配的中药配方颗粒。
2、为解决山药提取过程中减少淀粉溶出及糊化，本发明是把山药切成2～10mm的厚片，而不是粉碎成粒度很小的颗粒，避免提取过程中山药表面发生糊化而导致水分无法向内部渗透的现象，首先用温水浸泡山药，使水分充分渗透到药材内部，然后才在高温下提取。本发明山药提取物的制备过程是在90～97℃下进行的，相比于传统的提取工艺，温度较低；为增强有效成分的溶出，采用动态循环提取技术。动态循环提取技术，即在提取过程中，不断的把提取罐内的液体从提取罐底部，用泵通过管道从提取罐的上部重新打回提取罐，通过液体的强制循环流动，增强固-液两相之间的接触，增强有效成分从药材内部溶出。为避免药材堵塞提取罐底部管道，提取罐设计成罐中罐的方式，即，提取罐中有3个吊篮，药材是装在吊篮中，而不是直接放入提取罐。这样通过控制水的温度既不会使饮片表面糊化又能提高水分渗透力，可使水分有效快速进入饮片中，提高有效成分的溶出效率。
3、由于在动态循环提取过程中，中药材的浸泡提取是溶剂进入药材，将有效成分从固相转移到液相的过程，通过在增加药材比表面积的情况下，利用机械手段，采用强制循环方式，以增加固-液相接触；药材与溶剂之间始终保持相对浓度差，可提高药材中的溶质向溶剂的溶出效率。
4、低温动态循环提取和传统提取比较，动态循环提取具有物料受热均匀，温度低、时间短、节能、得膏率高、有效组分基本不变、可自动控制等优点，而传统提取工艺的煎煮温度高、时间长、耗能大、有效成分损失较多。因此，本发明采用强制由下至上强制动态循环提取方法，山药的提取效率高，操作简便，易控制，可适合推广于大生产中。其动态循环提取工作原理见附图。

本发明所述的动态循环设备包括提取罐2、循环泵8和循环管9，提取罐2顶部有罐盖1，罐体2内部安装有吊篮6，罐体底部有下降管7，下降管7接循环泵8，罐体上部安装油气上升管3，通到油水分离器4，分离出挥发油以后蒸馏水从回流管5返回罐体2内。
具体实施方式
实施例1
取山药药材300kg，经清洗炮制后切片，切片厚度2～4mm；投入提取罐吊篮中，加入山药重量10倍的水，在40℃下浸泡2小时；然后采用动态循环提取技术，在常压90℃下提取4小时，过滤，滤液浓缩，加辅料糊精25kg，混合均匀，喷雾干燥，粉碎，再加入硬脂酸镁0.1kg,制粒，制得山药配方颗粒100kg。
实施例2
取山药药材500kg，经清洗炮制后切片，切片厚度3～6mm；投入提取罐吊篮中，加入山药重量5倍的水，在50℃下浸泡1.5小时；然后采用动态循环提取技术，在常压95℃下提取3小时，过滤，滤液浓缩，加辅料麦芽糊精56kg,混合均匀，喷雾干燥，粉碎，再加入硬脂酸镁0.05kg，制粒，制得山药配方颗粒100kg。
实施例3
取山药药材400kg，经清洗炮制后切片，切片厚度6～8mm；投入提取罐吊篮中，加入山药重量8倍的水，在60℃下浸泡1小时；然后采用动态循环提取技术，在常压97℃下提取2小时，过滤，滤液浓缩，喷雾干燥，加辅料乳糖38kg,混合均匀，喷雾干燥，粉碎，再加入硬脂酸镁0.07kg，制粒，制得山药配方颗粒100kg。
实施例4
取山药药材300kg，经清洗炮制后切片，切片厚度7～10mm；投入提取罐吊篮中，加入山药重量10倍的水，在60℃下浸泡1.5小时；然后采用动态循环提取技术，在常压90℃下提取4小时，过滤，滤液浓缩，加辅料麦芽糊精29kg、糊精35kg,混合均匀，喷雾干燥，粉碎，再加入硬脂酸镁0.15kg，制粒，制得山药配方颗粒100kg。
实施例5
取山药药材500kg，经清洗炮制后切片，切片厚度2～5mm；投入提取罐吊篮中，加入山药重量8倍的水，在60℃下浸泡1小时；然后采用动态循环提取技术，在常压95℃下提取3小时，过滤，滤液浓缩，加辅料乳糖15kg、糊精29kg,混合均匀，喷雾干燥，粉碎，再加入硬脂酸镁0.1kg，制粒，制得山药配方颗粒100kg。

1.一种山药配方颗粒，其特征在于：包括以下重量份数的原料：
山药饮片 3000～5000份；
糊精或乳糖或麦芽糊精或它们的组合物 250～640份；
硬脂酸镁0.5～1.5份。
2.如权利要求1所述的山药配方颗粒的制备方法，其特征在于：取山药药材，经清洗炮制后切片，切片厚度2～10mm；在6～10小时内投入提取罐吊篮中，加入5～10倍的山药重量的40～60℃水浸泡，浸泡时间为1～2小时；在90℃～97℃常压下用动态循环设备进行动态循环提取方法，提取时间2～4小时，滤过，滤液浓缩，加辅料糊精或乳糖或麦芽糊精或它们的组合物，混合，干燥，得到山药提取物，粉碎，再加硬脂酸镁，制粒，制得山药配方颗粒。
3.如权利要求2所述的山药配方颗粒的制备方法，其特征在于：所述的动态循环提取方法是提取过程中，不断的把提取罐内的液体用泵通过管道从底部抽到打回提取罐上部进行动态循环流动。
4.如权利要求2所述的山药配方颗粒的制备方法，其特征在于：所述的动态循环设备包括提取罐、循环泵和循环管，提取罐顶部有罐盖，罐体内部安装有吊篮，罐体底部有下降管，下降管接循环泵，罐体上部安装油气上升管，通到油水分离器，分离出挥发油以后蒸馏水从回流管返回罐体内。
