Abstract

发明背景
皮肤的生长,修复和愈合是一个复杂的生物控制机制,其通过阳性和阴性信号起作用。这种信号在控制细胞增殖,分化和迁移水平起作用,并典型地由生长因子多肽介导。在此方面,重要的生长因子包括表皮生长因子(EGF)和胰岛素样生长因子(IGF-I和-II)。
已经报道人IGF-I发挥广泛的生物学活性,包括刺激细胞增殖,分化和迁移,保护蛋白质免于降解和细胞程序死亡,以及调节内分泌因子如生长激素。IGF-II与IGF-I具有相似性质,但表现为与癌发生及胎儿与胚胎发育更相关,IGF-I在出生以后的发育中具有更大的作用。
IGF-I和IGF-II均通过与I型IGF受体(IGFR)结合而发挥作用。这种相互作用所需的IGF的可利用性由胰岛素样生长因子结合蛋白(IGFBP 1-6)调节。IGFBP已知可阳性和阴性调节IGF功能,以及呈现IGF-非依赖性活性。

发明目的
本发明人令人惊奇地揭示了IGFBP能结合玻连蛋白,而且当IGF-I与IGFBP结合时,其可以结合玻连蛋白。
因此本发明的一个目的是提供一种分离的含有IGFBP和玻连蛋白的复合物。
发明概述
第一方面,本发明提供了一种分离的多肽复合物,其包含一种生长因子结合蛋白和玻连蛋白。
优选地,所述分离的多肽复合物还包含一种生长因子。
一种优选的生长因子是胰岛素样生长因子-I(IGF-I)。
其它生长因子包括表皮生长因子(EGF),成纤维细胞生长因子(FGF),碱性成纤维细胞生长因子(bFGF),骨桥蛋白,血小板反应蛋白-1,腱生蛋白-C,PAI-1,纤溶酶原,纤维蛋白原,纤维蛋白和运铁蛋白。
一种优选的生长因子结合蛋白是选自IGFBP1,IGFBP2,IGFBP3,IGFBP4,IGFBP5和IGFBP6的一种胰岛素样生长因子结合蛋白。
一种更优选的胰岛素样生长因子结合蛋白是IGFBP2,IGFBP3,IGFBP4或IGFBP5。
一种最优选的胰岛素样生长因子结合蛋白是IGFBP3或IGFBP5。
第二方面,本发明提供了一种分离的蛋白质复合物,其包含一种IGFBP-相关蛋白和玻连蛋白。

因此,本发明还提供了包含本发明分离的蛋白质复合物一种外科植入物或修复物。所述外科植入物或修复物可以用所述分离的蛋白质复合物包被、浸渍或以其它方式预处理。
根据本发明处理的动物可以是哺乳动物,优选是人,或可以是非哺乳动物脊椎动物,如鱼类,爬行动物或鸟,或分离自其中的细胞。
在本说明书中,除非特别指出,所用术语“包含”的含义是包含而不排除其它,所以所指出的整体或整体组可以包括一或多个其它非未指出的整体或整体组。

发明详述
本发明至少部分得自本发明人的发现,即IGF-I通过IGFBP与玻连蛋白之间的结合相互作用而结合玻连蛋白。另外,本发明阐述了变体IGF和IGFBP,其可以用于增加或减少IGF,IGFBP和玻连蛋白之间的结合。这些发现使本发明人得以在体外运用这些结合相互作用以操纵与细胞生长,增殖和迁移相关的偶然体内生物学事件。本发明因此在医学和兽医学领域中的医学治疗如创伤愈合,皮肤修复和保养,骨再生,动脉硬化和癌症治疗中具有用途。
本发明中,“分离的”是指从其天然环境中分离,或以其它方式进行人工操纵。分离的物质可以完全或基本没有在其天然状态中通常伴随的成分,或者可以操纵为加上其天然环境中通常伴随其的成分的人工状态。
“多肽”也意味着“蛋白质”,是指氨基酸聚合物。
包括生长因子,生长因子结合蛋白和玻连蛋白的蛋白质和肽,可以以天然的,化学合成的或重组合成形式分离。
“肽”是指具有不超过50个氨基酸的蛋白质。
“生物活性片段”是指蛋白质的片段、部分或区段,其呈现所述蛋白质的至少1%,优选至少10%,更优选至少25%,及最优选至少50%的生物学活性。
肽可以易于通过重组或化学合成方法合成。例如,可参考溶液合成或固相合成方法,例如Blaclcwell科学出版社出版的Nicholson编辑的题目为“合成疫苗”的出版物中由Atherton和Shephard所著第九章“肽合成”中所述方法。肽合成方法也见于Coligan等编辑的《蛋白质科学通用方法》第18章所述(John Wiley&Sons NY.1997),在此并入参考。
或者,肽可以通过用蛋白酶如endoLys-C,endoArg-C,endoGlu-C和葡萄球菌V8-蛋白酶消化本发明多肽而产生。消化的片段可以通过例如高效液相层析(HPLC)技术纯化。
在一个优选的形式中,本发明提供了一种分离的蛋白质复合物,其包含玻连蛋白,生长因子和生长因子结合蛋白。
“生长因子”是指一种分子,其刺激或促进机体或所述机体细胞生长。优选的生长因子是刺激细胞分化,尤其哺乳动物细胞分化的一种蛋白质或肽。
优选地,本发明分离的蛋白质复合物包含生长因子IGF-I。
分离的IGF和IGFBP多肽可从如GroPep(Adelaide,澳大利亚)商购,而VN多肽可从Promega公司(Madison WI,美国)商购。重组的IGF,IGFBP和VN易于由本领域技术人员生产,在下文详加阐述。
本领域技术人员还会意识到本发明还包括IGF的前体。pro-IGF-I蛋白的例子是已经除去信号肽但未由切割E-结构域而完全加工的IGF-I。IGF-I具有得自不同的mRNA剪接的三个不同E结构域,而且这些前体蛋白可以存在于本发明分离的蛋白质复合物中。

本领域技术人员还会意识到本发明分离的蛋白质复合物可以包括本领域熟知的“天然”,“变性”或“延伸”状态的玻连蛋白。
本发明还涵盖了包含nectinepsin的分离的生长因子复合物,nectinepsin是一种胞外基质蛋白,与VN在氨基酸水平示出60%同源性(Blanchert等,1996,生物化学杂志271 26220-26226)。
也应理解生长因子,生长因子结合蛋白和/或玻连蛋白的变体可以用于形成本发明分离的蛋白质复合物,并可以用于本发明方法中。
如本文所用,本发明的“变体”蛋白质,多肽和肽包括其中一或多个氨基酸由不同氨基酸置换的那些物质。
本领域熟知一些氨基酸可以用具有广泛相似性质的其它氨基酸置换,而不改变所述多肽的活性性质(保守取代)。
功能上的实质变化可以通过选择保守性较低的取代产生。其它置换是非保守取代而且相对较少可以被耐受。通常地,可能产生多肽性质最大变化的取代是那些其中(a)亲水性残基(例如Ser或Thr)由疏水性残基(例如Ala,Leu,Ile,Phe或Val)取代;(b)半胱氨酸或脯氨酸由任何其它残基取代;(c)具有阳电侧链的残基(例如Arg,His或Lys)由阴电残基(例如Glu或Asp)取代;或(d)具有大侧链的残基(例如Phe或Trp)由具有较小侧链的残基(例如Ala,Ser)或无侧链残基(例如Gly)取代。

本发明涵盖的其它多肽和肽变体包括但非限于侧链修饰,在肽,多肽或蛋白质合成期间掺入非天然氨基酸和/或其衍生物,及使用交联剂和对本发明多肽和肽变体具有构象限制的其它化合物。本发明涵盖的侧链修饰例如包括修饰氨基基团,如用乙酐酰化;用琥珀酐和四氢邻苯二甲酸酐酰化氨基基团;用甲基酰亚胺化物酰胺化;用氰酸盐氨甲酰化氨基基团;用5-磷酸吡哆醛吡哆化赖氨酸随后用NaBH4还原;通过与醛反应随后用NaBH4还原进行还原烷化;及用2,4,6-三硝基苯磺酸(TNBS)三硝基苄基化氨基基团。
羧基可以经过O-酰异脲形成随后衍生化而通过碳二亚胺活化而修饰,例如修饰为相应酰胺。
精氨酸残基的胍基可以通过与如2,3-丁二酮,苯甲酰甲醛和乙二醛等试剂形成杂环缩合产物而修饰。
修饰巯基可以通过如过甲酸氧化为胱氨酸;使用4-氯高汞苯磺酸,4-氯高汞苯甲酸,2-氯高汞-4-硝基酚,氯化苯汞和其它汞剂形成汞衍生物;与其它硫醇化合物形成混合的二硫化物;与马来酰亚胺,马来酐或其它取代的马来酰亚胺反应;用碘乙酸或碘乙酰胺进行羧甲基化;及用氰酸盐在碱性pH进行氨甲酰化。
修饰色氨酸残基可以例如通过用2-羟基-5-硝基苄基溴化物或磺酰卤化物烷化吲哚环,或用N-溴琥珀酰亚胺氧化进行。
修饰酪氨酸残基可以通过用四硝基甲烷硝化形成3-硝基酪氨酸衍生物而进行。
修饰组氨酸残基的咪唑环可以用焦碳酸二乙酯进行N末端carbethoxylation或用碘乙酸衍生物烷化进行。
在肽合成期间掺入非天然氨基酸和衍生物的实例包括但非限于使用4-氨基丁酸,6-氨基己酸,4-氨基-3-羟基-5-苯戊酸,4-氨基-3-羟基-6-甲基庚酸,叔丁基甘氨酸,正亮氨酸,正缬氨酸,苯基甘氨酸,鸟氨酸,肌氨酸,2-噻吩基丙氨酸和/或氨基酸的D-异构体。
修饰还包括O-和N-连接的糖基化变体和天然发生状态为糖基化的蛋白质的非糖基化形式。
关于这些变体,可以通过诱变多肽或诱变编码核酸而产生,如通过随机诱变或定点诱变。核酸诱变方法例如见于Ausubel等,分子生物学通用方法,第9章所述,在此并入参考。
本领域技术人员意识到,当有助于生物学活性的氨基酸残基的知识可以获得时,进行定点诱变是最佳的。在许多情况中,这个信息不能获得,或只能例如通过分子模拟近似值推断。
在这种情况中,进行随机诱变。随机诱变方法包括通过羟胺化学修饰蛋白质(Ruan等,1997,基因18835),在核酸中掺入dNTP类似物(Zaccolo等,1996,分子生物学杂志255589),及基于PCR的随机诱变如Stemmer,1994,美国科学院院报9110747或Shafikhani等1997,生物技术23304所述,在此均并入参考。也注意到基于PCR的随机诱变试剂盒可商购,如DiversifyTM试剂盒(Clontech)。
本发明还涵盖了生长因子变体如des(l-6)IGF-II和des(1-3)IGF-I形成本发明分离的蛋白质复合物的应用。
用作兴奋剂或拮抗剂的其它变体IGF和IGFBP在下文进一步阐述。
重组生长因子复合物
可以意识到分离的蛋白质复合物可以通过在本领域熟知的适当宿主细胞或无细胞表达系统中表达核酸而用重组生长因子,生长因子结合蛋白和/或玻连蛋白产生。
术语“核酸”是指单链或双链的mRNA,RNA,cRNA和DNA,所述DNA包含cDNA和基因组DNA。
“多核苷酸”是具有80或更多个连续核苷酸的核酸,而“寡核苷酸”是少于80个核苷酸的核酸。
“探针”可以是适当标记的单链或双链寡核苷酸或多核苷酸,以例如在Northern或Southern印迹中检测互补序列。
“引物”通常是单链的寡核苷酸,优选具有15-50个连续核苷酸,其能与互补核酸“模板”退火,并以模板依赖性方式通过DNA聚合酶(如Taq聚合酶)的作用而延伸。
本领域熟知编码IGF,EGF,IGFBP,ALS和VN的核酸,并且已供使用多年。然而,本领域技术人员可参考表1,其中列出了提供这些核酸序列的参考文献。表1中的所有参考文献均并入参考。
本发明的核酸可以根据以下程序制备:
(i)产生任选是简并的引物,其中每种引物均包含靶核酸的各自部分;及

重组蛋白可以通过本领域技术人员已知的任何适当方法制备。
例如,重组蛋白可以通过包括以下步骤的方法制备:
(i)制备一种表达构建体,其包含可操纵地连接于一或多个调节核苷酸序列的一种核酸;
(ii)用所述表达构建体转染或转化一种适当的宿主细胞;及
(iii)在所述宿主细胞中表达所述多肽。
为进行宿主细胞表达,所述重组核酸可操纵地连接于表达载体中的一或多个调节序列。
“表达载体”可以是自身复制染色体外载体如质粒,或整合入宿主基因组的载体。
“可操纵地连接”是指所述调节核苷酸序列相对于本发明重组核酸的定位能起始、调节或以其它方式控制转录。
调节核苷酸序列一般适于用于表达的宿主细胞。针对不同的宿主细胞,本领域已知多种类型的适当表达载体和适当调节序列。
典型地,所述一或多个调节核苷酸序列可以包括但非限于启动子序列,前导或信号序列,核糖体结合位点,转录起始和终止序列,翻译起始和终止序列,及增强子或激活子序列。
本发明涵盖本领域已知的组成型或可诱导启动子。所述启动子可以是天然发生的启动子,或组合一个以上启动子的杂合启动子。
在一个优选的实施方案中,所述表达载体包含一个选择标记基因,以选择转化的宿主细胞。本领域熟知选择标记基因并可根据所用宿主细胞而变化。
所述表达载体还可以包含一个融合配偶体(典型由所述表达载体提供),以便本发明的重组多肽与所述融合配偶体形成融合多肽而表达。融合配偶体的主要优点是其有助于鉴别和/或纯化所述融合多肽,并还增强蛋白质表达水平和整体产量。
为表达所述融合多肽,必需将本发明的核酸序列与所述表达载体连接,以便融合配偶体的翻译读框与本发明核苷酸序列一致。
融合配偶体的熟知实例包括但非限于谷胱甘肽S-转移酶(GST),人IgG的Fc部分,麦芽糖结合蛋白(MBP)和六组氨酸(HIS6),这些物质特别用于通过亲和层析分离所述融合多肽。为通过亲和层析纯化融合多肽,亲和层析的相关基质分别是谷胱甘肽,直链淀粉和镍或钴缀合的树脂。许多这种基质可以以试剂盒形式获得,如具有(HIS6)融合配偶体的QIA表达系统(Qiagen)和Pharmacia GST纯化系统。
本领域熟知的另一种融合配偶体是绿荧光蛋白(GFP)。这种融合配偶体作为荧光“标记”,可以使本发明的融合多肽通过荧光显微镜或流式细胞仪鉴别。所述GFP标记可用于评估本发明融合多肽的亚细胞定位,或分离表达本发明融合多肽的细胞。流式细胞术如荧光激活细胞分选(FACS)在后一应用中特别有用。
在一些情况中,所述融合配偶体还具有蛋白酶切割位点,如因子Xa或凝血酶,其使相关的蛋白酶部分消化本发明融合多肽,并从而从中释放本发明重组多肽。释放的多肽然后通过随后的层析分离可以自融合配偶体中分离。
本发明融合配偶体还包含“附加表位”,其通常是短肽序列,由此可获得特异抗体。可易于获得特异的单克隆抗体的附加表位的熟知实例包括c-myc,流感病毒血凝素和FLAG标记。

在另一个实施方案中,可以在相同细胞中进行生长因子,生长因子结合蛋白和玻连蛋白的重组表达,并从中形成复合物。
在这里,本发明多肽可以通过培养用所述表达构建体转化的宿主细胞产生,所述表达构建体包含编码本发明多肽或其类似物的核酸。
适于蛋白质表达的条件随着选择的表达载体和宿主细胞而变化。这易于由本领域技术人员通过常规试验而确定。
适于重组表达的宿主细胞包括细菌如大肠杆菌,梭菌,假单胞菌,酵母,植物细胞,昆虫细胞(如Sf9)和哺乳动物细胞如成纤维细胞和角质形成细胞。
优选的宿主细胞是人角质形成细胞。
本发明涵盖了诱导型和非诱导型表达载体。在适当转染的哺乳动物细胞中,已经设计了许多诱导和阻遏系统,包括金属硫蛋白(MT)诱导的及四环素抑制(tetR)系统,本发明涵盖每种系统。

模拟物,兴奋剂和拮抗剂
本发明涵盖了可以促进,阻止或破坏蛋白质复合物形成的制剂,所述蛋白质复合物包含生长因子,生长因子结合蛋白和玻连蛋白。这种制剂可以是一种模拟物。术语“模拟物”是指这样的分子,其被设计为模拟蛋白质或肽的特定功能区域,本领域熟知的术语“兴奋剂”,“类似物”和“拮抗剂”包括在其范围内。
关于负责IGF-IGFBP结合的IGF和IGFBP部分的阐述见于国际出版物WO00/23469所述。另外,已经产生选择性结合IGFBP-1或IGFBP-3的IGF-I的兴奋剂变体,如国际出版物WO00/40612所述。
因此本发明涵盖了可以被工程化以破坏或阻止IGFBP与VN之间形成多肽复合物的制剂。一个实例是一种肽,其通过模拟VN或IGFBP上的结合位点而竞争IGFBP与VN的结合。
如后文的更详细阐述,IGF-II是一种能抑制IGFBP与玻连蛋白之间结合的制剂。也提议IGF-II的C结构域中第34-40位RVSRRSR序列的V35或S36残基连同S39,可以突变为碱性残基,从而模拟IGFBP3的HBD(BXBBB,其中B是一个碱性氨基酸残基)。这样可以产生更能抑制IGFBP与玻连蛋白之间复合物的形成的制剂。
本发明涵盖的兴奋剂的一个实例是被工程化而包含一个IGFBP的肝素结合结构域(HBD)从而直接结合玻连蛋白的IGF-I。例如,IGF-I的C结构域中SSSRRAPQT序列可以工程化为具有BXBBB基序。一个优选的BXBBB基序是IGFBP-3的KGRKR序列(第228-232位残基)。
或者,可以将推定的IGF-II的玻连蛋白结合结构域RVSRRSR (第34-40位残基)导入IGF-I中。
本发明拮抗剂的一个实例是被工程化而突变HBD中的碱性残基(如前文所述)以降低或阻止IGFBP与玻连蛋白的结合的IGFBP,。
适当地,工程IGFBP能结合IGF-I。
优选地,工程IGFBP是IGFBP-3或IGFBP-5。
本发明还涵盖了IGFBP的类似物,其可以被工程化使所述类似物与VN之间形成复合物。适当地,所述类似物还可以结合IGF。这种类似物的潜在优点是其比IGFBP更易于合成或分离,具有特别需要的生物学半衰期并或许已被工程化而特异性结合IGF-I。
上述模拟物可以是具有所需生物学活性和半衰期的肽,多肽或其它有机分子,优选小有机分子。
计算机辅助结构数据库检索日益用作鉴别模拟物的程序。数据库检索方法原则上可以适用于鉴别模拟物,见于国际出版物WO94/18232(涉及产生HIV抗原模拟物),美国专利No.5,752,019和国际出版物WO 97/41526(涉及鉴别EPO模拟物)所述,在此均并入参考。
其它方法包括鉴别分子相互作用的各种生物学方法。这些方法可以根据所述候选分子是否影响IGF-IGFBP-VN复合物的形成而筛选候选分子。适当的方法如竞争性放射性配体结合分析(见Upton等,1999,前述相关方法),分析超速离心,微量量热法,表面等离子体共振和基于光学生物传感器的方法,由Coligan等编辑的《蛋白质科学》第20章所提供(John Wiley & Sons,1997),在此并入参考。
药物组合物
本发明包括以药物组合物形式的本发明蛋白质复合物的施用。本发明的药物组合物可以包括分离的蛋白质复合物,其包含变体IGF和/或IGFBP或如前述破坏或阻止所述复合物形成的制剂。
适当地,所述药物组合物包含一种可药用载体。药物组合物还可以包含如前述多肽变体,片段或模拟物。

可以使用任何适当途径为患者提供本发明组合物。例如可以应用经口服,直肠,非肠道,舌下,口腔,静脉内,关节内,肌内,皮内,皮下,吸入,眼内,腹膜内,脑室内,经皮等方法。例如为施用免疫原性组合物、疫苗和DNA疫苗合适的是肌内和皮下注射。
剂型包括片剂,分散液,悬浮液,注射液,溶液,糖浆,药片,胶囊,栓剂,气雾剂,经皮贴片等。这些剂型还可以包括特别为此设计的注射或植入控制释放装置,或经修改以此方式起作用的其它形式植入体。所述治疗剂的控制释放可以例如用疏水性聚合物包衣而实现,所述聚合物包括丙烯酸树脂,蜡,高级脂肪醇,聚乳酸和聚乙醇酸和某些纤维素衍生物如羟丙甲基纤维素。另外,所述控制释放可以通过使用其它聚合物基质,脂质体和/或微球实现。
适于口服或非肠道施用的本发明的药物组合物可以以分立单位存在,如胶囊,小袋或片剂,每剂均含有预定数量的一或多种本发明治疗剂,可以以粉末或颗粒或溶液或在水溶液中的悬浮液,非水溶液液体,水包油乳状液或油包水液态乳状液形式存在。
关于包含IGF和IGFBP的药物组合物,特别参考美国专利5,936,064和国际出版物W099/62536和W099/54359所述,在此并入参考。
本发明的药物组合物还可以包含表达载体,如病毒载体如痘苗,及在基因治疗中有用的病毒载体。后者包括腺病毒和腺病毒伴随病毒(AAV),如Braun-Falco等,1999,基因治疗6432所述,逆转录病毒和慢病毒载体,如Buchshacher等,2000,血液952499所述,及得自单纯疱疹病毒和巨细胞病毒的载体。用于内分泌基因治疗的病毒载体的综述见于Stone等,2000,内分泌学杂志164103所述。

治疗用途
本发明提供了使用本发明多肽复合物的治疗方法。这些方法特别针对于治疗性处理哺乳动物,尤其是人。
这种方法包括施用上述药物组合物,而且可以通过显微针注射入特异组织部位,如美国专利6,090,790所述,用于创伤,烧伤或溃疡的局部乳液,洗剂或密封敷料,如美国专利6,054,122所述,或释放所述组合物的植入体,如国际出版物W099/47070所述。
在这方面也可以应用基因治疗,如美国专利5,929,040和美国专利5,962,427所述方法。
还有可以遗传修饰皮肤细胞以产生皮肤替代品的方法,如通过遗传工程化所需生长因子的表达(Supp等,2000,J.Invest.Dermatol.1145)。关于这个领域的一个实例见于Bevan等,Biotechnol.Gent.Eng.Rev.16231所述。
本发明还涵盖了将转染的或转化的细胞“种植”给一种受体的方法,如国际出版物W099/11789所述。
这些方法可用于刺激细胞增殖,并从而促进或加速创伤和烧伤愈合,修复皮肤损害如溃疡,组织置换和移植如通过体外培养自体皮肤进行,内部器官如肾和肺的再上皮化,及修复损伤的神经组织。
皮肤置换治疗已为本领域所熟知,并可以利用共同培养的上皮/角质形成细胞系,如Kehe等,1999,Arch.Dermatol.Res.291600所述,或体外培养原代(通常是自体的)表皮、真皮和/或角质形成细胞。这些技术也可以利用工程生物材料及合成的聚合物“支架”。
关于这个领域的实例见于Terskikh & Vasiliev,1999,Int.Rev.Cytol.18841和Eaglestein&Falanga,1998,Cutis 621提供。
更特别地,在颅面手术中所用的置换口腔粘膜的产生见于Izumi等,2000,J.Dent.Res.79798所述。胎儿角质形成细胞和真皮成纤维细胞可以在体外扩展,以产生用于移植的皮肤,治疗皮肤损伤,如Fauza等,J.Pediatr.Surg.33357所述,同时得自在透明质酸衍生的生物材料上体外培养的真皮和表皮的皮肤替代品示出具有治疗烧伤的潜力(Zacchi等,1998,J.Biomed.Mater.Res.40187)。
上皮细胞疗法的另一方面涉及胃肠道上皮内膜的修复,以治疗或防止肠道功能衰退。
本发明还涵盖了聚合物支架,其促进置换皮肤工程,例如Sheridan等,2000,控制释放杂志14 91和Fauza等,1998,如前所述,本发明还包括作为将皮肤细胞输送至创伤和烧伤部位的制剂的微球(LaFrance & Armstrong,1999,Tissue Eng.5153)。
根据本发明可容易地利用上述方法,使用本发明分离的蛋白质复合物促进皮肤细胞增殖以进行组织置换和美容性皮肤处理。
关于骨再生,本发明提供了用本发明分离的蛋白质复合物包被、浸渍或以其它方式预处理的外科或修复植入体。
相反,通过阻止或破坏IGF-IGFBP-VN复合物形成而阻止或抑制细胞迁移,可以进行预防性或治疗性处理牛皮癣或恶性病如上皮细胞癌如乳癌。
本发明还涵盖了通过将本发明分离的蛋白质复合物给予干细胞或祖细胞而分化干细胞或祖细胞的方法。在此方法中也可以使用包含变体生长因子和IGFBP的分离的蛋白质复合物。
根据这种方法产生的分化的细胞可以用于如前述治疗方法中。
例如,据信平滑肌细胞是“间充质干细胞”,而且可以被驱动分化为成纤维细胞,基质细胞,内皮细胞,骨细胞或脂肪细胞。
为使本发明易于理解并实际应用,现在通过以下非限制性实施例阐述特别优选的实施方案。
在此所述的所有放射性配体竞争结合分析均基本如Upton等,1999,如前所述进行。
实施例1:确定胰岛素、pro-IGF-II和IGF-I与IGF-II竞争结合玻连蛋白的能力的竞争结合分析
由于IGF和胰岛素之间结构相似,因此检测胰岛素与玻连蛋白的结合。通过Upton等,1999(如前)所述进行交联试验表明胰岛素不太可能与IGF-II竞争结合玻连蛋白,用IGF-I出现相同情况。因此,检测高浓度的胰岛素以确定胰岛素是否能与放射标记的IGF-II竞争结合玻连蛋白。示于图1的结果表明与最初用IGF-I观测到的情况相同(Upton等,1999,如前),胰岛素几乎不与[125I]-IGF-II竞争结合玻连蛋白。
IGF-I作为IGF-II结合玻连蛋白的竞争剂的研究表明IGF-I可以竞争[125I]-IGF-II与玻连蛋白的结合(图2)。然而,在与放射标记的IGF-II竞争结合玻连蛋白中,IGF-I的效力明显低于IGF-II,要达到同样效力,需要IGF-I的浓度高于IGF-II的大约3000倍。
相似研究表明proIGF-II可以与[125I]-IGF-II竞争结合玻连蛋白(图3)。ProIGF-II在与[125I]-IGF-II竞争结合玻连蛋白中没有IGF-II那样的效力,IC50值分别为65.6nM和9.6nM。因此,在proIGF-II内E结构域的存在代表了改变与玻连蛋白结合的结构修饰。这可以是由于与IGF-II相比proIGF-II中额外的结构域的位阻现象,或者可以是由于proIGF-II分子结构改变从而改变了proIGF-II与玻连蛋白的亲和性。
实施例2:PAI-1和uPAR作为IGF-II与玻连蛋白结合的竞争剂的研究
对PAI-1和suPAR可能是与IGF-II竞争结合玻连蛋白的分子进行研究,因为已经报道这些蛋白质结合玻连蛋白(Declerck等,1988,生物化学杂志263 15454;Wei等,1994,生物化学杂志269 32380;Kanse等,1996,Exp.Cell Res.224 344)。在高达2000nM浓度,观测到PAI-1与[125I]-IGF-II竞争结合玻连蛋白IC50值为大约524nM(图4)。尽管从IC50值看这个浓度相对较高,但是发现这类浓度可与体内肿瘤相关(Grondahl-Hansen等,1993,癌症研究53 2513)。事实上,Kjoller等,1997,Exp.Cell Res.232 420,发现需要370nM的PAI-1才能在确定PAI-1抑制WISH细胞迁移能力的分析中达到半数最大效应。推测这种抑制通过PAI-1与整联蛋白和uPAR竞争结合玻连蛋白而产生。因此,在PAI-1,IGF-II和玻连蛋白之间观测到的相互作用确实可能具有一些体内功能结果,特别当有时在肿瘤中发现IGF-II和PAI-1水平是高度表达的时候。
尽管已知PAI-1结合玻连蛋白N末端生长调节素B结构域,仍需要高浓度PAI-1才能有效与IGF-II竞争结合玻连蛋白,提示这些研究未提供关于玻连蛋白上IGF-II结合位点的位置的明确信息。然而,所述结果可以以两种方式解释。第一种可能是在IGF-II和PAI-1之间观测到的竞争是由于在玻连蛋白生长调节素B结构域内一级高亲和性位点由于位阻现象的部分竞争所致。或者,所述竞争可以是由于IGF-II和PAI-1之间的直接竞争,结合玻连蛋白上PAI-1结合亲和性降低的位点。需要进一步的实验研究以阐明这个问题。
尽管本发明人不能示出可溶的suPAR与IGF-II竞争结合VN,但这可能是由于在国际运输后使用冻干的suPAR所致。没有迹象表明复水的suPAR是生物学活性的,尽管观测到[l25Il-suPAR不结合VN(数据未示出)也许支持用于这些研究的suPAR是失活的这种解释。
使用与鉴别玻连蛋白上PAI-1结合位点位于含有44个N末端氨基酸的一玻连蛋白片段内的相似方法(Deng等,1995,Thromb.Haemost.7466),可以确定在PAI-1和IGF-II之间结合玻连蛋白的竞争是否是任一这些可能性的结果。
以前已经示出可溶形式的尿激酶受体(suPAR)可以结合固定的玻连蛋白(Wei等,1994,sUP7-a)。然而,在此报道的研究中,甚至浓度高达300nM,在suPAR和[125I]-IGF-II之间也未观测到竞争结合玻连蛋白。
实施例3:在存在非糖基化重组IGFBP-3情况下,IGF-I和IGF-II与玻连蛋白的结合
为研究IGFBP是否能介导IGF与玻连蛋白结合,评定增加浓度的IGFBP-3在存在[125I]-IGF-I或[125I]-IGF-II的情况下的作用。结果示于图5。
在所测试浓度,10ng/孔的IGFBP-3导致最大量的[125I]-IGF-I与玻连蛋白包被孔结合,而30ng/孔的IGFBP-3导致最大量的[125I]-IGF-II与玻连蛋白包被孔结合。所述作用在[125I]-IGF-I的情况中最显而易见,因为低计数的[125I]-IGF-I结合玻连蛋白,甚至在最低浓度的IGFBP-3(3.7ng),这种结合也增加(图5A)。相反,[125I]-IGF-II未示出在玻连蛋白包被孔上获得的结合增加,直至浓度为11ng/100μL-33ng/100μL也未达到(图5B)。
用[125I]-IGF-I或[125I]-IGF-II预温育IGFBP-3,与未预温育的IGFBP-3和[125I]-IGF-I或[125I]-IGF-II相比,不改变与玻连蛋白的结合。
实施例4:在存在在哺乳动物细胞中产生的重组IGFBP的情况下,IGF-I与玻连蛋白的结合
如Upton等,1999所述(如前)进行结合分析,所述分析检测在存在IGFBP的情况下,[125I]-IGF-I结合VN包被平皿的能力,所述IGFBP与放射标记同时加入。数据示于图6。IGFBP-2,-4和-5的数量增加导致标记IGF-II与玻连蛋白包被孔结合增加。在测试的IGFBP最高量(5ng),针对IGFBP-2,4和5,标记的IGF-I的结合与对照孔相比提高大约2.8,3.8和8倍,所述对照孔中存在VN而不存在IGFBP。
存在0.05、0.2和0.5ng/孔的IGFBP-3也增加标记IGF-I与VN的结合,而2和5ng/孔的IGFBP-3表现为抑制放射标记的IGF-I的结合。测试的所有浓度的IGFBP-1和IGFBP-6也均是抑制性的。
这些结果表明与IGF-II情况不同,观测到IGF-I与VN的最小直接结合。然而,IGFBP、尤其IGFBP-2,-3,-4和-5的存在,增强IGF结合VN包被孔,提示在这种情况中IGFBP介导IGF-I与VN的结合。另外,数据提示IGFBP具有既增强又抑制IGF-I与VN结合的潜力,这取决于a)存在何种IGFBP及b)IGFBP存在量。
实施例5:在存在IGFBP-3变体的情况下,标记IGF-II与VN的结合
如上所述进行结合分析,所述分析检测在存在IGFBP-3变体的情况下,[125I]-IGF-I结合VN包被平皿的能力,所述变体中推定的“肝素结合结构域”被突变(IGFBP-3HBD)及其中糖基化位点被突变(Non-gly IGFBP-3,即非糖基化IGFBP-3)。数据示于图7。
这些结合分析中使用的IGFBP-3糖基化突变体中的三个潜在的O-糖基化位点Asn89,Asn109,Asnl72突变为Ala。
IGFBP-3HBD突变体不增强[125I]-IGF-I与VN包被平皿的结合,提示IGFBP-3的肝素结合结构域参与IGFBP-3与VN的结合。其它研究已经鉴别推定的“肝素结合结构域”之外的氨基酸残基与IGF-I结合IGFBP-3相关,因此很可能这个结果表示IGFBP-3与VN结合降低,而不是标记的IGF-II与IGFBP-3结合降低(Imai等,2000,生物化学杂志275:18188-18194)。
令人感兴趣地,非糖基化IGFBP-3突变体明显增强[125I]-IGF-I与VN包被平皿的结合。在存在2ng突变体IGFBP-3的情况下,标记的IGF-I与VN包被孔的结合惊人地提高20倍,这是令人感兴趣的并提示IGFBP-3的糖基化抑制a)IGF-I与IGFBP-3或b)IGFBP-3与VN的相互作用。或者,这两个相互作用均可以通过存在碳水化合物而阻碍。
非糖基化IGFBP-3在体内是否功能相关还未确定。然而,这个发现提示结合VN的非糖基化IGFBP-3可以是将IGF-I输送至需要IGF-I以加强细胞功能之处的一种有效方式,如在刺激细胞增殖中。或者,结合VN的非糖基化IGFBP-3可以提供一种机制,在不需要细胞增殖的情况下隔绝过量IGF-I,如在过表达IGF的肿瘤中。
实施例6:确定在存在非糖基化重组IGFBP-3的情况下,IGF变体与标记的IGF竞争结合VN的能力的竞争结合分析
在竞争分析中使用图5所述产生放射标记的IGF最大结合的IGFBP3浓度,以确定在存在IGFBP3情况下,浓度增加的IGF和desIGF是否可以与放射标记的IGF竞争结合玻连蛋白。结果表明高浓度的IGF-I,IGF-II,des(1-6)IGF-II及或许des(1-3)IGF-I(未示出),在存在10ng的IGFBP3情况下,可以与[125I]-IGF-II竞争结合玻连蛋白(图8)。这些实验结果表明在存在30ng的IGFBP3情况下,只有IGF-I、IGF-1I和des(1-6)IGF-II可以与IGF-II竞争结合玻连蛋白包被孔。
实施例7:包含IGF-II和玻连蛋白的分离的蛋白质复合物对细胞增殖的刺激
在这个研究中使用将IGF-II与VN预先结合的策略,以尝试更真实反映体内胞外环境。大多数细胞培养方法均在溶液相中加入外源底物;从而所述细胞持续暴露于处理之下。组织中细胞不会遇到体内这种“持续溶液相”环境。因此适于这个研究的方法是将IGF与VN预先结合,更真实反映体内条件。
在IGF与VN在培养皿中预先结合之后,将细胞种植于孔中,并通过制定的方法检测与VN复合的IGF刺激蛋白质合成的能力(Francis等,1986,生物化学杂志233:207-213)。蛋白质合成增加与细胞数增加相关,因此是细胞增殖的反映。应答以相对于对照孔(无VN或IGF)的百分比表示,通过测定在48小时后[3H]-亮氨酸掺入新合成的蛋白质的情况表示。
参考图9中数据,当3、10、30、100、300和1000ng的IGF-II与所述孔在没有VN情况下预先结合时,分别观测到产生超出对照孔(-VN,-IGF)8%、12%、10%、16%、24%和43%的应答。另外,这些相同剂量的预先结合VN包被孔的IGF-II刺激[3H]-亮氨酸掺入蛋白质的作用分别为19%、29%、39%、51%、70%和101%。针对3、10、30、100、300和1000ng的IGF-II,组合只用VN(12%)获得的应答和只用IGF-II获得的应答(参考上文),分别产生20%、24%、22%、28%、36%和55%的预计加合作用。这些数值明显不同于当IGF-II以除了两个最低测试量IGF-II(3和10ng)之外所有剂量与VN预先结合时观测到的实际作用(p<0.05)。因此,预先结合VN的IGF-II在蛋白质合成中刺激协同作用,比计算的加合作用高5-46%。这些应答可能是IGF-II与VN直接结合的结果,也可能是IGF-II与VN通过IGFBP间接结合产生的。HaCAT角质形成细胞产生大量IGFBP-3(Wraight等,1994,J.Invest.Dermatol 103:627-631)。
实施例8:在存在在哺乳动物细胞中产生的重组IGFBP情况下,标记的IGF-II与VN的结合
检测[125I]-IGF-II在存在IGFBP的情况下结合VN包被的平皿的能力的结合分析如Upton等,1999(如前)所述进行,所述IGFBP与放射标记同时加入。这些研究中使用的是在哺乳动物中产生的糖基化IGFBP。如图10所示,增加量的IGFBP-1,-3和-6导致标记的IGF-II与玻连蛋白包被孔的结合以剂量依赖性方式降低。IGFBP2还表现为竞争标记IGF-II与VN的结合,但效力比IGFBP-1,-3或-6低。另一方面,IGFBP-4对IGF-II与VN的结合的作用很小,而IGFBP-5表现为少量增强IGF-II结合。IGFBP-3的抑制作用可以得自IGFBP3与IGF-II竞争结合VN上相同结合区域。或者,或另外,IGFBP-3以及IGFBP-1和IGFBP-6的抑制作用,可以得自IGF-II与VN的亲和性比IGF-II与这些IGFBP的亲和性低,因此这些IGFBP隔绝IGF-II并且所述复合物不结合VN。
实施例9:包含IGF-I,IGFBP-5和玻连蛋白的分离的蛋白质复合物对细胞增殖的刺激
参考表2,IGF-I与IGFBP-5和玻连蛋白在所有测试浓度均刺激角质形成细胞增殖(通过3H-亮氨酸掺入新合成的蛋白质中测定),在最高测试浓度观测到协同作用。
本发明人提示分离的蛋白质复合物对细胞增殖的作用可能在比表2所述相对低量更高浓度的IGFBP-5存在下更强。
实施例10:IGF和IGFBP变体的工程化
IGFBP-3中在与ECM结合中重要的并已经阐明是推定的“肝素结合结构域”的氨基酸残基是在第228-232位的KGRKR残基。在IGFBP-5的相应区域发现相似残基。在此报道的结合研究中使用的IGFBP-3HBD突变体是残基KGRKR变化为MDGEA,其基于在IGFBP-I中相应位置中发现的氨基酸。这些变化导致蛋白质这个部分的电荷逆转。这个突变体仍然高亲和性结合IGF-I和IGF-II,但很少结合酸不稳定亚单位及细胞表面(Firth等,1998,生物化学杂志2732631-2638)。
不同类别蛋白质中肝素结合基序最初由Cardin等,1989,动脉硬化921-32阐述。
人IGF-II的C结构域含有许多阳性电荷的氨基酸残基,尤其第34-40位含有氨基酸RVSRRSR。这些阳性电荷的氨基酸在介导IGFBP与细胞表面和VN结合中起重要作用,IGF-II中的这些氨基酸在IGF-II与VN的直接结合中也起重要作用。不管怎样,通过产生缺失V35或S36及S39的一种IGF-II突变体而导入与在IGFBP-3中发现的序列相似的“肝素结合基序”是一个相对简便的方法。介导IGF-II与VN结合的正电荷残基的重要性,还通过本发明人揭示了鸡IGF-II突变体(desR40)-IGF-II与VN的结合降低这个证据得以进一步例证。
另一方面,IGF-I的C结构域在IGF-II蛋白质上述相应区域中含有相对非极性氨基酸链。这可以解释为什么IGF-I不直接结合VN。另外,也不结合VN(或IGFBP)的胰岛素不具有相应的C结构域,因为其在成熟蛋白质中被切除。

实施例11:分离的蛋白质复合物,细胞增殖和存活
Bcl-2转录是细胞存活途径的一个关键因子,其在通过α-β3整联蛋白附着于VN的细胞中增加(Matter & Ruoslahti,2001,生物化学杂志276 27757-27763)。另外,IGF-I通过以AKT依赖性方式增加bcl-2转录而保护细胞免于程序死亡(Pugazhenthi等,1999,生物化学杂志274 27529-35)。IGF受体与α-β3整联蛋白物理缔合,对细胞生长具有协同作用(Schneller等,1997,EMBO杂志165600-5607)。因此本发明分离的蛋白质复合物可以提供一个整合胞外点,以起始由整联蛋白和生长因子受体介导的细胞存活信号。
已经表明IGFBP-5在前列腺癌细胞中加强IGF-I的抗细胞程序死亡和促进有丝分裂作用(Miyake等,2000,内分泌学141 2257-2265)。另外,VN在体内由神经胶质瘤细胞的合成及在结肠直肠腺癌中的合成与肿瘤等级相关(Uhm等,1999,临床癌症研究5 1587-1594;Tomasini-Johansson等,1994,Exp Cell Res.214 303-312;Gladson等,1995,细胞科学杂志108 947-56;Gladson&Cheresh,1991,临床研究杂志88 1924-32)。
因此,根据本发明,提示在体内形成的IGF:IGFBP:VN复合物可以促进肿瘤细胞存活和转移。本发明因此涵盖了破坏体内复合物形成的治疗剂。
已经报道VN的肝素结合结构域抑制纤连蛋白基质装配(Hocking等,1999,生物化学杂志274 2725727264)。纤连蛋白沉积降低与肿瘤细胞入侵相关,因为细胞迁移速度降低与聚合的纤连蛋白水平提高相关(Morla等,1994,自然367 193-196)。因此,结合VN的肝素结合结构域的IGF:IGFBP复合物可以阻碍纤连蛋白基质装配并促进肿瘤侵入局部结缔组织。因此本发明涵盖了破坏这些体内复合物以降低肿瘤侵染的治疗剂。
分离的蛋白质复合物与创伤愈合
可以使用相反的论据支持所述复合物在需要细胞迁移如创伤修复的情况中应用。IGF系统在创伤愈合中起重要作用,而且IGF-I和IGFBP-3均以显著浓度存在于创伤处体液中(Skottner等,1990,ActaScand.Suppl.367 63-66;Clark R(ed)1996,创伤修复的分子和细胞生物学,pp 3-50,Plenum出版社,纽约;Robertson等,1996,内分泌学137 2774-2784;Vogt等,1998,生长激素IGF研究,8 Suppl B:107-9)。
已经表明IGFBP降低IGF从创伤处清除速度(Robertson等,1999,.Am J Physiol.276E663-71)。IGFBP-3:IGF-I复合物在体外结合纤维蛋白凝块,引起这种情况也在体内发生使IGF-I在创伤部位积聚的推测(Campbell等,1999,生物化学杂志274 30215-30221)。相似地,玻连蛋白结合纤维蛋白(Podor等,2000,生物化学杂志27519788-19794)。还注意到玻连蛋白-裸鼠表现为创伤处纤维蛋白溶解增强及微血管发生减少(Jang等,2000,外科学127696-704)。
根据本发明,提示结合IGFBP的IGF可以结合VN,其接着与纤维蛋白凝块结合,因此在创伤部位提供了IGF的集聚。因此本发明分离的蛋白质复合物可以施用于创伤处以促进修复过程。
本发明涵盖的创伤愈合的一个特殊方面涉及糖尿病足部溃疡的愈合。创伤愈合在糖尿病患者中很慢。生长因子影响愈合过程,而且尤其IGF已经示出刺激角质形成细胞增殖。然而,对糖尿病患者皮肤和足部溃疡组织的分析表明:与非糖尿病患者组织切片相对比,在基底层和成纤维细胞中没有IGF-I表达(Blakytny等,2000,病理学杂志190 589-594)。本发明分离的蛋白质复合物可以用于将IGF-I输送于这类创伤处。
分离的蛋白质复合物与骨工程
IGFBP-5通过一种非依赖于IGF受体的机制促进标记的IGF-I与骨的结合,(Mohan等,1995,生物化学杂志270 2042420431),并增强IGF刺激的成骨细胞功能(Andress,1995,生物化学杂志27028289-28296)。
在许多研究中已经示出植入材料羟磷灰石是高度生物学相容的,及与现有骨良好整合。近来的迹象表明羟磷灰石从血清中比其它常规使用的植入材料如钛和不锈钢中吸收更多的VN。VN的吸收通过成骨细胞前体细胞的更强结合而实现(Kilpadi等,2001,J.Biomed.Mater.Res.57 258-267)。
最近检测了VN对纳米相氧化铝/常规氧化铝的作用,发现VN增强成骨细胞附着(Webster等,2001,Tissue Eng 7291-301)。
本发明人提示本发明分离的蛋白质复合物可用于包被这些原料,并从而在整形外科应用如髋关节置换中促进骨细胞附着,生长及整合。
VN在兔骨细胞培养物中增强IGF-I刺激的破骨细胞再吸收和蛋白酶活性(Rousselle等,2001,组织学和组织病理学16727-734),IGFBP-5增强IGF刺激的成骨细胞有丝分裂(Andress&Birnbaum,1992,生物化学杂志267 22467-22472)。
本发明人提示由于骨细胞产生的主要IGFBP是IGFBP-5,因此通过VN引起的IGF作用增强很可能也涉及IGFBP-5。
分离的蛋白质复合物和治疗动脉硬化
IGF-I已经预先包含于试验性动脉硬化损伤的研究中。另外,α-β3抑制剂已经表明降低动脉硬化损伤,这与IGF-I介导的信号化抑制相关(Nichols等,1999,Circ.Res.851040-1045)。
鉴于:
(i)IGFBP-5和VN由动脉平滑肌细胞合成和分泌,并存在于血管壁中;
(ii)IGF:IGFBP-5:VN复合物促进血管平滑肌细胞有丝分裂和迁移(Nam&Clemmons,2000,生长激素IGF研究10:A23);
本发明人提示IGF:IGFBP-5:VN复合物参与动脉硬化损伤的形成。因此破坏所述复合物形成的治疗剂可以有效治疗动脉硬化。
