Description
技术领域
本发明提供了包含抗炎化合物的共混物的组合物。该组合物可用于(例如)局部施用至皮肤。
背景技术
抗炎化合物是已知的。本发明人已认识到,存在对识别提供改善性能的抗炎化合物的新组合的需要。
本发明人已发现,称为NFκB抑制剂的抗炎化合物尽管能够降低通过NFκB-通道的免疫应答,但有时促进炎性细胞因子IL-1的释放。本发明人还发现,这些抗炎化合物具有降低通过利用T-细胞受体(TCR)激活剂植物血细胞凝集素刺激的人淋巴细胞产生的细胞因子的强效能力,尤其是具有约70μg/ml或更低的IC50的那些,能够降低免疫应答,但有时趋于促进炎性细胞因子IL-8的释放。
因此,目前已经发现的是,NFκB抑制剂与具有低IC50(尤其是约70μg/ml或更低的IC50)的抗炎化合物的组合提供如下组合物,所述组合物令人吃惊地降低从皮肤和其他组织释放炎性细胞因子IL-1和IL-8,同时还通常提供皮肤和其他组织中的期望的NFκB抑制。
发明内容
在一个方面,本发明提供了包含NFκB抑制剂和抗炎化合物的组合物。当根据本文所述的抗炎检测分析法进行测试时,该抗炎化合物并非为NFκB抑制剂并且具有约70μg/ml或更低的IC50。
根据另一个方面,本发明提供了处理皮肤老化迹象的方法,该方法包括给需要这种处理的皮肤局部施用包含NFκB抑制剂和抗炎化合物的组合物。当根据抗炎检测分析法进行测试时,该抗炎化合物并非为NFκB抑制剂并且具有约70μg/ml或更低的IC50。

所述组合物包含NFκB抑制剂和抗炎化合物,所述抗炎化合物并非为NFκB抑制剂而且具有低IC50。这两种成分均为皮肤和其他组织提供抗炎有益效果,并且这些抗炎有益效果为互补的。本发明人已吃惊地发现,与NFκB抑制剂混合的具有约70μg/ml或更低的IC50的抗炎化合物抑制IL-1和IL-8细胞因子的增加(所述增加可在其他情况下因使用单独化合物而产生)。
在本发明的一个实施例中,当通过示于实例V中的IL-1α检测分析法测量时,NFκB抑制剂与具有约70μg/ml或更低的IC50的抗炎化合物的组合使得所产生的细胞因子IL-1α降低至少约35%。
在本发明的另一个实施例中,当通过示于实例VIII的IL-8检测分析法测量时,NFκB抑制剂与具有约70μg/ml或更低的IC50的抗炎化合物的组合使得所产生的细胞因子IL-8降低至少约80%。
因此,本发明还提供通过将NFκB抑制剂和下述抗炎化合物的组合局部施用至皮肤或其他组织来使该皮肤或其他组织中所产生的细胞因子IL-1α降低至少约35%(如通过IL-1α检测分析法测得)的方法,所述抗炎化合物并非为NFκB抑制剂并且具有约70μg/ml或更低的IC50。如在下文进一步所述,可以局部用组合物的形式来施用NFκB抑制剂和抗炎化合物的组合。
另外提供了通过将NFκB抑制剂和下述抗炎化合物的组合局部施用至皮肤或其他组织来使该皮肤或其他组织中所产生的细胞因子IL-8降低至少约80%(如通过IL-8检测分析法测得)的方法,所述抗炎化合物并非为NFκB抑制剂并且具有约70μg/ml或更低的IC50。如在下文进一步所述,可以局部用组合物的形式来施用NFκB抑制剂和抗炎化合物的组合。
NFκB 抑制剂
如本文所用,“NFκB抑制剂”意指可抑制细胞转录因子核转录因子k-B(NFκB)的化合物。在一个实施例中,NFκB抑制剂在根据如下文定义的“NFκB抑制测试”进行测试时,在以优选每毫升1微克至每毫升约100微克的浓度测试时具有至少约30%、优选至少约50%、更优选至少约70%、最优选至少约90%的NFκB抑制百分比。也就是说,化合物以在每毫升1微克至每毫升100微克范围内的至少一种浓度下显示出所列出的NFκB抑制百分比。该化合物不必在每毫升1微克至每毫升100微克的所有浓度下提供所列出的NFκB抑制百分比,而是在该范围内的至少一个浓度下提供所列出的NFκB抑制百分比。
在一个优选的实施例中,当以每毫升10微克的浓度测试时,NFκB抑制剂具有至少约35%、优选至少约55%、更优选至少约70%、最优选至少约90%的NFκB抑制百分比。
“NFκB抑制测试”以如下方式进行。使用FB293细胞,其为含有NF-κB的报告基因的稳定转染的人上皮细胞系。它们可从(如)Panomics(Fremont,CA)获得。将FB293以5×104个细胞/mL的密度接种于合适的培养基中,例如补充有10%胎牛血清(Invitrogen,San Diego,CA)的达尔伯克氏改良伊格尔培养基(DMEM)中。在存在测试样品的情况下,用100ng/mL的肿瘤坏死因子α(TNFα,可得自Sigma-Aldrich(St Louis,MO))刺激该FB293细胞。单独地,测试其中未施用测试样品的对照样品。在37℃下使用5%的CO2孵育24小时之后,利用40μl的报告裂解缓冲液(Promega,Madison,WI)来裂解细胞。将20μL的裂解产物等分试样用虫荧光素酶测定试剂盒(Promega)进行测定,并在Lmax发光计(Molecular Devices,Sunnyvale,CA)中计数10秒,数据以相对光单位/秒表示。测试样品的NFκB抑制百分比计算如下:

一种或多种NFκB抑制剂可以任何合适的量存在于本发明组合物中,例如约0.01重量%至约10重量%、优选约0.1%至约20%、更优选约0.1%至约5%、甚至更优选约0.2%至约2%。
在一个实施例中,NFκB抑制剂选自如下化合物:取代的间苯二酚、(E)-3-(4-甲基苯基磺酰基)-2-丙烯腈(如“Bay 11-7082”,可从Sigma-Aldrich(St.Louis,Missouri)商购获得)、四氢姜黄素(如四氢姜黄素CG,可得自Sabinsa Corporation(Piscataway,NJ))、毛泡桐木的提取物以及它们的组合。
在一个优选的实施例中,NFκB抑制剂是取代的间苯二酚。间苯二酚是具有如下结构的二羟基酚化合物(即1,3二羟基苯):
如本文所用,“取代的间苯二酚”意指在2、4、5或6位中具有至少一个取代基间苯二酚。因而,取代的间苯二酚可具有少至一个且多至四个的取代基。取代的间苯二酚的位置1和3具有-OH羟基,如上所示。
高度优选的是,取代的间苯二酚的所有取代基无苯基(-C6H5芳族)部分。在某些实施例中,所有取代基无(具有或不具有杂原子的)芳族部分。
在另一个实施例中,优选的是取代的间苯二酚的所有取代基无酮官能团(与另外两个碳原子键合的羰基)。
在某些优选的实施例中,取代的间苯二酚的所有取代基既无苯基官能团也无酮官能团。
在某些优选的实施例中,取代的间苯二酚包含至少一个具有5至11个碳原子、优选5至10个碳原子、更优选5至9个碳原子、最优选5至8个碳原子的取代基。在某些其他实施例中,至少一个取代基包含烷基,例如具有上述碳原子数目的烷基。该烷基优选是不饱和的。
在某些实施例中,间苯二酚的4位被取代,在某些实施例中,仅4位被取代。在另一个实施例中,4位置被烷基基团取代。在某些优选的实施例中,取代的间苯二酚包含单个4位取代基,该取代基包含烷基。在某些其他的优选实施例中,取代的间苯二酚包含单个4位取代基,该取代基由与苯环直接键合的烷基组成。
特别合适的取代的间苯二酚包括4-己基间苯二酚和4-辛基间苯二酚,特别是4-己基间苯二酚。4-己基间苯二酚和4-辛基间苯二酚的结构在下面示出:
4-己基间苯二酚可以“SYNOVEA HR”从Sytheon(Lincoln Park,NJ)商购获得。4-辛基间苯二酚可从City Chemical LLC(West Haven,Connecticut)商购获得。
在某些实施例中,取代的间苯二酚在2、4、5或6位具有至少两个取代基。这种取代基可任选连接而形成环,例如任选包含杂原子例如硫或氧的环状脂族烃。例如连接的取代基可包含5至10碳原子,如8至10碳原子,并任选包括1至3杂原子。具有连接2位和3位的环状脂族取代基的合适取代的间苯二酚的例子包括玉米赤霉酮和β-玉米赤霉醇:

在某些其他实施例中,取代的间苯二酚具有含卤和/或含亚硝基的取代基。合适的例子含有与苯环直接键合的-Cl或-N=O。这些取代基可存在于(例如)2和4位、2和6位或4和6位上。二卤取代的间苯二酚的例子为2,6-二氯间苯二酚。二亚硝基取代的间苯二酚的例子为A2,4-二亚硝基间苯二酚:

取代的间苯二酚可通过本领域已知的方法制备,例如使用美国专利No.4,337,370中描述的技术制备,将该专利的内容以引用的方式并入本文。
取代的间苯二酚可具有任何合适的分子量。在某些实施例中,取代的间苯二酚的分子量在约175至约300之间的范围内。
泡桐为亚洲原产的一个植物属,其已逐渐传播至欧洲和美国。在日本,泡桐称为日本泡桐,其专指一个物种:毛泡桐(也称为“公主树”)。通常使用的其他名字为“皇后树”、“毛地黄树”、“皇家树”、“泡桐”(中国)和“Odong-Namoo”(韩国)。学名为“毛泡桐”,其具有报告于各种文献中的多个同义词,即“锈毛泡桐”、“兴山桐”和“毛泡桐”。毛泡桐属于“泡桐”科(有时称为“玄参科”)。美国农业部(plants.USDA.gov)植物数据库通过独特符号“PATO2”来鉴别公主树以及为同义名的毛泡桐和锈毛泡桐。
可使用任何合适的毛泡桐木的提取物。一般而言,毛泡桐树的木材包括来自树杆、树枝或这二者的组合的木材。毛泡桐木的合适提取物可源于木屑、木粉和/或小切屑等等。
取代的间苯二酚在组合物中以安全有效量存在,以组合物的重量计,以例如约0.01%至约10%、优选约0.1%至约5%、更优选约0.2%至约2%、甚至更优选约0.5%至约1.5%的量存在。
抗炎化合物
本发明的组合物还包含并非为NFκB抑制剂的抗炎化合物。具体地讲,如通过NFκB抑制测试测得,当以至多约每毫升100微克的浓度测试时,抗炎化合物具有低于约30%、优选低于约20%、更优选低于约10%的NFκB抑制百分比。即,化合物在至多约每毫升100微克的所有浓度下均显示出低于约30%、优选低于约20%、更优选低于约10%的NFκB抑制百分比。
在一个优选的实施例中,当以至多约每毫升10微克的浓度测试时,抗炎化合物具有低于约30%、优选低于约20%、更优选低于约10%的NFκB抑制百分比。
对于本文所示的抗炎分析检测法中的白细胞介素2,抗炎化合物具有约70μg/ml或更低的IC50(化合物实现50%的发炎抑制的浓度)。在一个优选的实施例中,抗炎化合物的IC50为约50μg/ml或更低、优选约40μg/ml或更低、更优选约30μg/ml或更低。
抗炎检测分析法评估试剂降低通过利用T-细胞受体(TCR)激活剂植物血细胞凝集素(PHA)刺激的人淋巴细胞产生的细胞因子的能力,并且按下述方式来进行。通过白细胞单采术从健康成年男性采集人白细胞,然后在无血清淋巴细胞生长培养基(ExVivo-15,Biowhittaker,Walkersville,Md.)中调整为1×106细胞/mL的密度。按照所公开的方法(Hamamoto Y.等人,Exp Dermatol 2:231-235,1993),在存在或不存在测试样品的情况下利用10μg/mL的PHA来刺激PBL。在37℃下使用5%的CO2孵育48小时之后,去除上清液,并且使用市售的多重细胞因子检测套件来评估细胞因子含量。

在某些优选实施例中,抗炎化合物选自:黄柏外皮提取物(PCE)、甘菊(小白菊)、银杏(银杏属)、黄栌(黄栌属)、枸杞子(枸杞)、奶蓟提取物(水飞蓟属)、苋菜油(繁穗苋)、石榴(石榴科)、巴拉圭茶(巴拉圭茶叶提取物)、百合花提取物(百合)、橄榄叶提取物(橄榄)和根皮素(苹果提取物)。
在某些更优选的实施例中,抗炎化合物选自:甘菊(小白菊)、枸杞子(枸杞)、奶蓟提取物(水飞蓟属)、苋菜油(繁穗苋)、石榴(石榴科)、巴拉圭茶(巴拉圭茶叶提取物)、百合花提取物(百合)、橄榄叶提取物(橄榄)和根皮素(苹果提取物)。
“甘菊的提取物”意指(例如)可根据名称为“PARTHENOLIDEFREE BIOACTIVE INGREDIENTS FROM FEVERFEW(TANACETUMPARTHENIUM)AND PROCES SES FOR THEIR PRODUCTION”(得自甘菊(小白菊)的不含小白菊内酯的生物活性成分及其制备方法)的美国专利申请公开No.2007/0196523中设置的细节制备的植株“小白菊”的提取物。一种尤其合适的甘菊提取物可以约20%的活性甘菊从IntegratedBotanical Technologies(Ossining,NY)商购获得。
本发明的组合物可包含美容上有效量的一种或多种抗炎化合物。该组合物优选包含基于活性物质计的约0.1%至约10%、更优选约0.5%至约5%的第二抗炎化合物。
在本发明组合物中,NFκB抑制剂与抗炎化合物的重量比可为变化的。例如,NFκB抑制剂和抗炎化合物可以约0.001至约100、优选约0.01至约10、更优选约0.1至约10的重量浓度比(重量浓度比通过用NFκB抑制剂的重量浓度除以抗炎化合物的重量浓度来确定)存在。
局部用组合物
本发明的组合物可局部施用至人皮肤或毛发。除了第一和第二抗炎化合物,该组合物还可包含美容上可接受的局部用载体,该局部用载体可为组合物的约50重量%至约99.99重量%(如,组合物的约80重量%至约99重量%)。在本发明的一个优选的实施例中,美容上可接受的局部用载体包括水。
可将组合物制备成多种产品类型,包括但不限于洗剂、霜剂、凝胶剂、棒剂、喷雾剂、油膏剂、清洁液体洗剂和固体皂剂、洗发剂和毛发调理剂、毛发固定剂、糊剂、泡沫剂、粉末剂、摩丝、剃须膏、擦拭物、贴剂、水凝胶、成膜产品、面膜和皮肤膜剂、膜剂和化妆品如粉底以及睫毛膏。这些产品类型可含有几种类型的美容上可接受的局部用载体,包括(但不限于)溶液、悬浮液、乳状液(如微乳液和纳米乳液)、凝胶、固体和脂质体。下面是这类载体的非限制性例子。其他的载体可由本领域普通技术人员配制。
可用于本发明的组合物能配制成溶液。溶液通常包含水性溶剂或有机溶剂(例如,约50%至约99.99%或约90%至约99%的美容上可接受的水性溶剂或有机溶剂)。合适的有机溶剂的例子包括:丙二醇、聚乙二醇(200-600)、聚丙二醇(425-2025)、甘油、1,2,4-丁三醇、山梨醇酯、1,2,6-己三醇、乙醇以及它们的混合物。
可用于本主题发明的组合物可配制成包含润肤剂的溶液。此类组合物优选包含约2%至约50%的一种或多种润肤剂。如本文所用,“润肤剂”是指用于例如通过预防水分经表皮从皮肤流失来预防或缓解干燥的材料。润肤剂的例子包括但不限于植物油、矿物油、脂族酯等。
洗剂可由此类溶液制成。洗剂通常包含约1%至约20%(例如,约5%至约10%)的一种或多种润肤剂和约50%至约90%(例如,约60%至约80%)的水分。
可由溶液配制的另一种产品是霜剂。霜剂通常包含约5%至约50%(例如,约10%至约20%)的一种或多种润肤剂和约45%至约85%(例如,约50%至约75%)的水分。
尽管优选的是本发明的组合物包括水,但是作为另一种选择,组合物可以是无水的或者是不含水但含有机和/或硅树脂溶剂、油、类脂和蜡的膏剂。膏剂可包含简单的动物或植物油基质或半固体碳氢化合物。油膏剂可含有约2%至约10%的一种或多种润肤剂和约0.1%至约2%的一种或多种增稠剂。
组合物可以配制为乳状液。如果局部用载体是乳状液,则约1%至约10%(如,约2%至约5%)的局部用载体包含乳化剂。乳化剂可以为非离子型、阴离子型或阳离子型。合适的乳化剂的例子包括个人护理和化妆品配方领域中通常鉴定为合适乳化剂的那些。
洗剂和霜膏可配制成乳状液。通常此类乳液包含0.5%至约5%的乳化剂。这种霜剂通常含有约1%至约20%(如约5%至约10%)的一种或多种润肤剂;约20%至约80%(如30%至约70%)的水;以及约1%至约10%(如约2%至约5%)的一种或多种乳化剂。
水包油和油包水型单乳液护肤制剂,例如洗剂和软膏是化妆品领域所熟知的,并且可用于本主题发明。多相乳状液组合物(例如水包油包水型或油包水包油型)也可用于本主题发明。通常,此类单相乳状液或多相乳状液含有水分、润肤剂和乳化剂作为其基本成分。
本发明的组合物还可配制成凝胶(例如,使用合适的胶凝剂制成的水性凝胶、醇类凝胶、醇/水型凝胶或油性凝胶)。用于水性凝胶和/或醇类凝胶的适当胶凝剂包括(但不限于)天然树胶、丙烯酸和丙烯酸酯聚合物和共聚物以及纤维素衍生物(例如羟甲基纤维素和羟丙基纤维素)。用于油(例如矿物油)的适当胶凝剂包括(但不限于)氢化丁烯/乙烯/苯乙烯共聚物和氢化乙烯/丙烯/苯乙烯共聚物。此类凝胶通常包含介于约0.1重量%和5重量%之间的此类胶凝剂。
本发明的组合物也可配制成固体制剂(例如蜡基棒、条皂组合物、粉末或含有粉末的擦拭物)。
除了前述的组分外,用于本主题发明的组合物还可包含各种各样的其他油溶性物质和/或水溶性物质,这些物质通常用于组合物中,按本领域既定的水平施用于皮肤和头发。
其他美容活性剂
在一个实施例中,组合物还包含另一种美容活性剂。如本文所用,“美容活性剂”是对皮肤或毛发具有美容或治疗效果的化合物(如合成化合物或从天然来源或天然提取物分离的化合物),包括、但不限于抗痤疮剂、控油剂、抗微生物剂、其他抗炎剂、抗霉菌剂、抗寄生虫剂、外用止痛剂、防晒剂、光防护剂、抗氧化剂、角质层分离剂、表面活性剂、保湿剂、营养剂、维生素、能量增强剂、防汗剂、收敛剂、除臭剂、固化剂、抗硬角质剂和用于毛发和/或皮肤调理的试剂。
在一个实施例中,所述试剂选自(但不限于)羟基酸、过氧化苯甲酰、D-泛醇、甲氧基肉桂酸辛酯、二氧化钛、水杨酸辛酯、胡莫柳酯、阿伏苯宗、类胡萝卜素、胺(如neutrol)、诸如视黄醇和视黄基棕榈酸酯之类的类视色素、神经酰胺、多不饱和脂肪酸、必需脂肪酸、酶、酶抑制剂、矿物质、诸如雌激素之类的激素、诸如氢化可的松之类的类固醇、2-二甲基氨乙醇、诸如氯化铜之类的铜盐、诸如Cu:Gly-His-Lys之类的含铜肽、辅酶Q10、肽、诸如脯氨酸之类的氨基酸、维生素、乳糖酸、乙酰基-辅酶A、烟酸、核黄素、硫胺素、核糖、诸如NADH和FADH2之类的电子转运蛋白和诸如芦荟、甘菊、麦片之类的其他植物提取物以及它们的衍生物和混合物。美容活性剂通常将以组合物的约0.001重量%至约20重量%(如,约0.005%至约10%,例如约0.01%至约5%)的量存在于本发明的组合物中。
维生素的例子包括但不限于维生素A、维生素B(例如维生素B3、维生素B5和维生素B12)、维生素C、维生素K和维生素E的不同形式(如α、β、γ或Δ生育酚)或它们的混合物以及它们的衍生物。
羟基酸的例子包括,但不限于乙醇酸、乳酸、苹果酸、水杨酸、柠檬酸和酒石酸。
抗氧化剂的例子包括(但不限于)水溶性抗氧化剂,例如巯基化合物和它们的衍生物(如,焦亚硫酸钠和N-乙酰基-半胱氨酸)、硫辛酸和二氢硫辛酸、白藜芦醇、乳铁蛋白以及抗坏血酸和抗坏血酸衍生物(如,抗坏血酸棕榈酸酯和抗坏血酸多肽)。适用于本发明的组合物中的油溶性抗氧化剂包括但不限于丁基化羟基甲苯、类视色素(如,视黄醇和视黄基棕榈酸酯)、生育酚类(如,生育酚乙酯)、生育三烯酚类和泛醌。包含适用于本发明的组合物中的抗氧化剂的天然提取物包括但不限于:包含黄酮类和异黄酮类以及它们的衍生物(如,染料木黄酮和二联玉米蛋白)的提取物、包含白藜芦醇的提取物等。此类天然提取物的例子包括葡萄籽、绿茶、松树皮和蜂胶。
其他材料
各种其他材料也可以存在于组合物中,如本领域所公知的。这些物质包括湿润剂、pH调节剂、螯合剂(如EDTA)、芳香剂和防腐剂(如对羟苯甲酸酯)。
水或醇溶性染料可能也适合用于本发明的组合物。适用于本发明组合物的染料的例子包括焦糖、胭脂红、荧光素衍生物、甲氧补骨脂素、三甲补骨脂内酯、偶氮染料、蒽醌染料、蓝薁、愈创薁、1,4-二甲基薁(chamuzulene)、赤藓红、孟加拉玫瑰红、根皮红、cyanosin、daphinin、曙红G、cosin 10B、酸性红51、红染料4、红染料40、蓝染料1和黄染料5或它们的混合物。
当使用时,基于组合物的总重量,组合物中染料的量可在约0.0001至约0.1、优选约0.0025至约0.025重量百分比之间变化。
包含本发明此类组合物的组合物、制剂和产品可以利用本领域普通技术人员所知的方法来制备。
使用方法
可将本发明的组合物局部施用至需要处理如上所述的一种或多种皮肤老化迹象的哺乳动物皮肤。在一个实施例中,可将组合物施用至需要处理细纹和皱纹和/或弹性损失的皮肤。可根据合适的处理方案,如每月、每周、每隔一天、每天、每天两次等,将组合物施用至需要这种处理的皮肤。
在某些实施例中,本发明的组合物还可用于处理与皮肤相关的其他需要。例如,本发明的组合物可用于处理发炎后色素沉着过度、用于减小毛孔大小、痤疮处理、用于减少皮脂产生以及用于减轻疤痕。在某些其他实施例中,可将本发明的组合物与机械或物理表皮剥脱处理(例如微晶磨皮处理)同时施用或在该处理的数小时内施用,或与化学剥脱剂或角质层分离剂例如水杨酸同时施用。在某些其他实施例中,可将本发明的组合物施用至粘膜或其它组织例如阴道组织、口腔组织或眼组织。在某些其他实施例中,可将本发明的组合物施用至轻度伤口或术后部位以促进愈合、施用至昆虫叮咬处、施用至毒藤皮肤病症或相似皮肤状况,或通常用于减轻发痒。在某些其他实施例中,施用本发明的组合物以减轻皮肤刺激。所述刺激可具有因护肤和美容产品中的成分(例如类视色素及其衍生物、过氧苯甲酰、α-羟基酸及其衍生物、水杨酸、表面活性剂、天然植物提取物、防晒活性物质、脲以及防腐剂等)导致的外部起源。所述刺激可具有其他外部起源,例如太阳、风或剃须。所述刺激也可由内部疾病状况导致,所述内部疾病状况例如痤疮、红斑痤疮、特应性皮炎以及其他疾病状态。
据信,本领域的技术人员可根据本文的描述最大限度地利用本发明。以下具体实施例应被解释为仅仅是说明性的,而不以任何方式限制本公开内容的其余部分。以下非限制性实例进一步说明了本发明。
实例I
对如下测试样品进行了上述“NFκB抑制测试”:Bay 11-7082测试样品(Sigma-Aldrich,St.Louis,MO)、四氢姜黄素CG(Sabinsa Corporation,Piscataway,NJ)以及多种浓度的4-己基间苯二酚。结果在表1中示出,其中对测试样品和对照样品记录NF-κB报告基因激活(发光,L)。另外记录NF-κB抑制百分比。

Bay 11-7082和四氢姜黄素CG显示出强的NF-κB抑制。出乎意料的是,4-己基间苯二酚也导致NFκB激活显著减少。更出乎意料的是,4-己基间苯二酚即使在低浓度时也显示出显著的NF-κB抑制。
实例II
对各自浓度为10μg/ml的一系列取代的间苯二酚进行了上述“NFκB抑制测试”。结果在表2中示出。

从表2中的数据可以看出,仅含无苯基官能团的取代基的取代间苯二酚、仅含无酮官能团的取代基的取代间苯二酚以及包含具有5至11个碳原子的取代基的取代间苯二酚提供较好的NFκB抑制。

通过如下方法制备该组合物。将Synovea HR称重并溶解于HYDROLITE 5中,加入去离子水以形成A相。将油质体与Finsolv TN混合以形成B相。在持续混合下将B相非常缓慢地加入至A相。继续混合15分钟直至形成均一的乳剂。在持续混合下将ARISTOFLEX快速加入至上述乳剂中以获得稠的、平滑且均匀的制剂。
实例V
对多种试剂执行IL-1α检测分析法,以研究促炎介质IL-1α细胞因子在人上皮细胞中的释放。
按下文所述来执行IL-1α检测分析法。KB细胞得自ATCC(ATCC#CCL-17,Manassas,VA)并且以5000个细胞/孔的密度铺板在96孔的组织培养处理板中的添加10%胎牛血清(Invitrogen Corp.,San Diego,CA)的达尔伯克改良伊格尔培养基(DMEM)内。在48小时之后,利用下文所述的指定浓度的试剂来处理细胞24小时,然后收集上清液并且利用市售套件(Millipore Corp.,Billerica,MA)来分析IL-1α细胞因子的释放。结果在下表5中示出。

结果表明,诸如4-己雷琐辛和四氢姜黄素之类的NF-κB抑制剂尽管能够抑制NF-κB,但可增加细胞因子IL-1α的释放。
实例VI
执行如实例5中所述的其他IL-1α检测分析法。为了易于比较,将4-己雷琐辛、根皮素以及它们的组合的IL-1α的报告增加值相对4-己雷琐辛(20μg/mL,比较例2)进行归一化。将四氢姜黄素、甘菊以及它们的组合的IL-1α的报告增加值相对四氢姜黄素(20μg/mL,比较例3)进行归一化。结果在下表6中示出。

实例VII

这些结果表明,NF-κB抑制剂和具有高于约70μg/ml的IC50的抗炎化合物的组合不可与NF-κB抑制剂和具有约70μg/ml或更低的IC50的抗炎化合物的组合一样显著地降低细胞因子IL-1α的产生。上述数据表明,NF-κB抑制剂与具有高于约70μg/ml的IC50的抗炎化合物的组合使得所产生的细胞因子IL-1α减少低于35%。
应该指出的是,在比较例9-14中,大豆未溶于测试培养基中而是分散的。
实例VIII
使用实例5中所述的方法对一系列组合物执行IL-8检测分析法,不同的是分析的细胞因子为IL-8。对各个样品计算IL-8的减少百分比。结果在表8中示出。对于含有NF-κB抑制剂和抗炎化合物的组合的样品,报告出IL-8的减少百分比(相对于对应剂量的抗炎化合物的IL-8的减少百分比)。

结果表明,与任一单独化合物的性能相比,NF-κB抑制剂与具有约70μg/ml或更低的IC50的抗炎化合物的组合极大地降低了IL-8的表达。
实例IX
执行如实例8所述的另一个IL-8检测分析法,不同的是改变了NF-κB抑制剂与具有约70μg/ml或更低的IC50的抗炎化合物的比率。结果在下表9中示出。

这些结果表明,在多个重量比的整个范围上(对于发明实例Ex.13、14和15分别为10∶1、1∶1和1∶10的重量比)可观察到NF-κB抑制剂与具有约70μg/ml或更低的IC50的抗炎化合物的组合的性能的显著改善。
总体而言,本发明的抗炎共混物使得炎性细胞因子IL-1和IL-8均减少,同时还通常提供NFκB抑制。

抗炎化合物,所述抗炎化合物并非为NFκB抑制剂并且具有约70μg/ml或更低的IC50。

3.根据权利要求1所述的组合物,其中所述抗炎化合物具有约40μg/ml或更低的IC50。
