本发明的基础源自于发明人发现:花色素和叶黄素协同作用,以提供提高水平的I型溶胶原,其是I型胶原蛋白的前体。因为I型溶胶原的数量化学计量地反映合成的I型胶原蛋白的数量,因此I型溶胶原的合成被认为是I型胶原蛋白合成的标志。I型胶原蛋白是最丰富的胶原蛋白类型,并且其尤其在皮肤中发现。胶原蛋白的束称为胶原蛋白纤维,其是细胞外基质的主要组分,所述细胞外基质支撑大多数组织并从外部赋予细胞结构。特别地,I型胶原蛋白是皮肤的主要组分,其负责赋予皮肤以强度和弹性。因此,它随年龄的降解导致皱纹和松弛。因此,使用本发明组合物将改善皮肤弹性并减少皱纹和松弛的出现。
术语“其衍生物”在涉及花色素时,意在包括相关花青素衍生物,其实例是葡糖苷,芦丁糖苷和槐糖苷。
术语“可食用的”指的是人可食用的。所述组合物可以是乳液形式,其结合或未结合乳固体。
所述花色素可以选自橙曲菌素(aurantindin),矢车菊色素,花燕草色素,europindin,藤黄菌素(luteolindin),天竺葵色素,锦葵色素,芍药色素,3'-甲花翠素和rosindin。优选所述花色素是矢车菊色素。任选的是,所述花色素的衍化物是花青素。
一般,矢车菊色素本身独立存在或者与一种或多种其它花色素类化合物相结合存在。经常,在存在矢车菊色素时,化合物矢车菊色素苷,越桔色苷,花青素鼠李葡萄糖苷,花青-3-葡糖苷及其衍生物仅以痕量存在,如同天然植物提取物常见的那样。例如,所涉及化合物可以占本发明组合物的矢车菊色素组分的重量的小于1wt%,优选小于0.2wt%,经常小于0.05wt%,更经常小于0.01wt%。优选的是矢车菊色素与其它花色素化合物相结合存在,所述矢车菊色素的存在量为花色素组分的超过50重量%,优选超过75重量%。
包括矢车菊色素的花色素是在许多红色浆果中发现的色素,所述红色浆果包括但不限于覆盆子,黑莓,蓝莓,樱桃,红莓,接骨木果,山楂,罗甘莓,巴西莓(acai berry)和树莓。它们也在其它水果如苹果和李子中发现。在所述水果的皮中发现最高浓度的花色素。
所述叶黄素的衍化物可以是其酯。
所述组合物优选包括一种或多种额外组分,其选自抗氧化剂,调味剂,防腐剂和稳定剂。
本发明的组合物可以呈现任何适合形式,包括例如食品和营养增补剂。口服的组合物包括饮料,条及其它液体和固体形式,如片、丸、胶囊和粉末(其可以含结晶材料)以及涂抹料,人造黄油,膏剂,沙司,装饰料,蛋黄酱,冰淇淋,馅料,糖果和麦片。本发明组合物可以以带有局部组合物的试剂盒的形式出售,所述局部组合物具有选自提高胶原蛋白在皮肤中沉积,减少皱纹的出现和减少松弛的相同皮肤护理益处。因此,这种试剂盒包括本发明的处理老化皮肤的可食用组合物和在上文提到的局部组合物,在使用时,其可以从皮肤“内侧”和“外侧”都起作用,以提供选自提高胶原蛋白在皮肤中的沉积、减少皱纹的出现和减少松弛的所述皮肤护理益处。
在一个实施方案中,本发明组合物优选是水基的,即包含至少50%水,优选至少60%或至少70%w/w水。其可以是液体或冷冻的。因此所述产品具有常规水基产品的感觉,并可以定期消费,作为消费者的日常饮食的一部分。例如,它可以取代通常在早餐时间消费的果汁。本发明组合物优选包装成饮料,例如包装在容器如硬纸盒或上浆纸或纸板、玻璃或塑料制成的瓶中。所述容器优选容积为10-500毫升,如20-100毫升。
在一个备选实施方案中,本发明组合物装在胶囊中,与告知用户建议的服用方法的说明书一起提供。所述胶囊可以由本领域公知的任何适合材料(如明胶)构成。所述胶囊适用于由消费者吞咽,通常每天一到四次服用一个或二个胶囊。
或者,本发明组合物可以包括成复合食品的一个组分,例如所述组合物可以以固体或胶状的形式存在,作为条或类似产品内的馅料或层。所述组合物因此可以被引入各种日常食品,例如在“保健食品”条中,其可以作为其它小吃食品的替代而食用。
除了花色素及其衍生物(它们本身就是抗氧化剂)以外的一种或多种额外抗氧化剂优选存在于本发明组合物中,以防止或减缓所述组合物的天然氧化降解。适合的额外抗氧化剂可以选自但不是排他性地选自以下所列的物质,它们可以独立存在或彼此结合:叔丁基对苯二酚(TBHQ),抗坏血酸酯(例如抗坏血酰棕榈酸酯),抗坏血酸,生育酚,迷迭香提取物,水果浓缩物或提取物,红茶或绿茶提取物,没食子酸丙酯,丁基化羟基苯甲醚(BHA),丁基化羟基甲苯(BHT),柠檬酸或酯,生育三烯酚,多酚,酚化合物,其它类黄酮化合物和氧清除剂。尤其优选的额外抗氧化剂是维生素C和E。这些不仅是有效的抗氧化剂,而且在消费时它们还显示出赋予皮肤益处。额外抗氧化剂的数量可以是在典型的至少6个月的保质期内足以防止所述组合物发生腐败的量。显然,抗氧化剂的数量将取决于所用抗氧化剂的类型和活性。
本发明组合物可以包含调味剂,但是如果通过调味物质如果蔬汁提供所述叶黄素或花色素时,就没必要加入调味剂。适合的调味剂可以是天然的或合成的。可能需要调味剂以使得所述产品消费起来更适口。
所述组合物可以包含乳化剂,更优选食品级磷脂乳化剂。优选所述磷脂乳化剂是卵磷脂。磷脂乳化剂是油溶性的,但是所述卵磷脂可以在乳化前加入任一相中。优选将它加到水相中。任何乳化剂优选在所述组合物中的存在量为至少0.01%,优选0.05-3%,更优选0.1-1%w/w。
本发明组合物可以包含多不饱和脂肪酸,如ω-3脂肪酸(即具有12-26个碳原子的不饱和羧酸)。优选的ω-3脂肪酸选自二十二碳六烯酸(DHA),二十碳五烯酸(EPA)和其混合物。适合的多不饱和脂肪酸也可以选自油酸,亚油酸,γ-亚油酸,亚麻酸,花生四烯酸。所述多不饱和脂肪酸可以作为天然油(如鱼油)的组分存在。
所述组合物也可以包含大豆异黄酮(包含染料木黄酮或黄豆苷原,其呈糖基化的和/或非糖基化的形式),一般的量为0.0001-0.1%w/w。
本发明组合物可以通过制备水相和油相来制备。如果使用乳化剂,优选将其加入水相。然后共混所述油相和水相以形成乳液。在一种优选方法中,所述油在粉末载体材料上以帮助形成乳液。然后可将乳液包装在密封容器,如金属、涂敷纸板(例如Tetra Pak)或塑料容器中。然后优选将所述容器密封而不留下顶部空间或留下排除氧的气体填充(例如氮或二氧化碳)的顶部空间。这进一步有助于防止氧化。或者,所述乳液可以冷冻和包装并以冷冻消费品进行销售。
在本发明的第二方面,提供了一种用于提供选自以下的皮肤益处的化妆方法:提高胶原蛋白在皮肤中的沉积,改善皮肤弹性,减少皱纹的出现和减少松弛,所述方法包括每日给予本发明组合物,每天至少一份,优选至少两份,更优选至少三份,最优选至少四份相同或不同的份额。
在本发明的第三方面,提供了本发明的可食用组合物的用途,用于提供选自以下的皮肤益处:提高胶原蛋白在皮肤中的沉积,改善皮肤弹性,减少皱纹的出现和减少松弛。
在本发明的第四方面,提供了本发明的可食用组合物的用途,用于提供选自以下的皮肤益处:提高胶原蛋白在皮肤中的沉积,改善皮肤弹性,减少皱纹的出现和减少松弛,所述用途包括给予本发明组合物,每天至少一份,优选至少两份,更优选至少三份,最优选至少四份相同或不同的份额(serving)。

图1显示对于包含各μM浓度的叶黄素、矢车菊色素或两者的试验溶液,每μg蛋白质中的I型溶胶原的ng浓度。
具体实施方式
实施例1:叶黄素和矢车菊色素对原发人皮肤成纤维细胞中的I型溶胶原水平的影响
将第一组原发人皮肤的成纤维细胞在Dulbecco改良的Eagle培养基(Dulbecco's modified Eagle medium,DMEM)中培养,所述培养基购自Gibco,并补加10%胎牛血清(FBS)。然后将所述细胞在6-孔组织培养皿中,以在每个孔的2毫升完全培养基(DMEM和FBS的混合物)中播种密度为约5000个细胞/平方厘米,按常规铺板24小时,并在37oC在5%CO2中培养。然后除去所述完全培养基,并在处理前使所述细胞在DMEM和1%FBS中生长24小时。然后对所述细胞进行计数,制粒并对细胞溶胞产物进行针对I型溶胶原-1(PC-1)表达的分析。
制备用于针对I型溶胶原-1(PC-1)表达的分析的第二组细胞,其与上述第一组细胞的不同之处在于,它们用1mM的4-β-佛波醇-12-肉豆蔻酸酯-13-乙酸酯(PMA)(Sigma P8139)进行氧化应激24小时,以使得PMA的最终浓度为1μM。然后,通过分析细胞上清液的乳酸脱氢酶(其因细胞死亡而释放),测定细胞毒性水平。PMA模拟了紫外辐射对皮肤细胞的影响,并降低胶原蛋白合成。然后对所述细胞进行计数,制粒并对细胞溶胞产物进行针对I型溶胶原-1(PC-1)表达的分析。
试验溶液制备在DMEM中,其包含1%FBS和各种μM浓度的叶黄素,矢车菊色素或两者,如图1所示。
购自Takara Bio Incorporated的I型溶胶原C-肽酶免疫分析(EIA)试剂盒使得可以对I型溶胶原C-肽(P1 P)进行定量测定。制备八个P1 P标准品,样品稀释物浓度范围为0-640纳克/毫升。将100微升的抗体-过氧化物酶共轭溶液与20微升细胞溶胞产物(1微克蛋白质)或一个标准品加入两个孔中。将所述孔板密封并在37oC培养3小时,然后用400微升磷酸盐缓冲液(PBS)洗涤四次。然后将各孔中盛上100微升所述试剂盒中提供的底物溶液,并将孔板在室温下培养15分钟。此后,向各孔中加入100微升停止溶液,其也由所述试剂盒提供,并用孔板读取器在450nm测量吸光度。将相对于P1 P浓度的平均吸光度制成标准曲线,并通过回归分析计算最佳拟合的线。由此估算所有试验溶液的未知P1 P浓度。结果显示在图1中,其清楚表明叶黄素和矢车菊色素的结合对升高I型溶胶原水平的协同效应。转化生长因子β(TGF β)用作阳性对照物。介质是二甲基亚砜(DMSO)。
实施例2:包含接骨木果衍生的花青素和叶黄素的胶囊
胶囊包封法是本领域技术人员公知的,一般包括合并125毫克标准化的25%花青素接骨木果提取物(Artemis International Inc.),12.5毫克叶黄素在红花油中的20%悬浮液(购自Kemin Health LC的在红花油中的20%FloraGlo lutein液体),8毫克硬脂酸镁和12毫克二氧化硅,并将混合物包封在羟丙基甲基纤维素(HPMC)胶囊中。
早上服用两粒胶囊,晚上再服用另外两粒胶囊。一般在12周之后可观察到改善皮肤弹性、减少皱纹的出现和减少松弛的可见迹象。
实施例3
合并100毫克卵磷脂,100微升叶黄素在红花油中的20%悬浮液(购自Kemin Health LC的在红花油中的20%FloraGlo lutein液体),4克标准化的25%花青素接骨木果提取物(Artemis International Inc.)和水以补足到100毫升,由此产生一种饮料。
通过首先在常规乳化装置中将卵磷脂共混到水中,然后共混叶黄素和接骨木果提取物,从而制成所述饮料。
Claims (10)
1.一种处理老化皮肤的可食用的组合物,其包含:
(a)2-1000,优选5-750,最优选10-500毫克的花色素及其衍生物;和
(b)1-20,优选2-15,更优选2-10,最优选3-10毫克的叶黄素及其衍生物;
其中所述花色素与叶黄素的重量比为至少1:1,优选至少10:1,更优选至少25:1,更加优选至少50:1,最优选至少100:1;
其中所述组合物排除独立形式或与葡萄糖酸铜和氧化铜相结合形式的铜;
并且还独立排除或与上述铜排除一起排除南天竹或其提取物。
2.根据权利要求1的可食用的组合物,其为乳液形式。
3.根据权利要求1或权利要求2的可食用的组合物,其还包含乳固体。
4.根据在前权利要求的任一项的可食用的组合物,其中所述花色素选自橙曲菌素,矢车菊色素,花燕草色素,europindin,藤黄菌素,天竺葵色素,锦葵色素,芍药色素,3'-甲花翠素和rosindin。
5.根据权利要求4的可食用的组合物,其中所述花色素是矢车菊色素。
6.根据权利要求1-4任一项的可食用的组合物,其中花色素的衍化物是花青素。
7.根据在前权利要求的任一项的可食用的组合物,其中叶黄素的衍化物是其酯。
8.一种用于提供选自以下的皮肤益处的化妆方法:提高胶原蛋白在皮肤中的沉积,改善皮肤弹性,减少皱纹的出现和减少松弛,所述方法包括给予根据权利要求1-7任一项的可食用的组合物,每天至少一份,优选至少两份,更优选至少三份,最优选至少四份相同或不同的份额。
9.根据权利要求1-7任一项的可食用的组合物的用途,用于提供选自以下的皮肤益处:提高胶原蛋白在皮肤中的沉积,改善皮肤弹性,减少皱纹的出现和减少松弛。
10.根据权利要求1-7任一项的可食用的组合物的用途,用于提供选自以下的皮肤益处:提高胶原蛋白在皮肤中的沉积,减少皱纹的出现和减少松弛,所述用途包括给予所述可食用的组合物,每天至少一份,优选至少两份,更优选至少三份,最优选至少四份相同或不同的份额。
