发明简述
本发明提供了包含分离的α-半乳糖苷酶的清洁组合物。在一些特别优选的实施方案中,所述的分离α-半乳糖苷酶包含与来自里氏木霉(Trichoderma reesei)的α-半乳糖苷酶相关的氨基酸序列。本发明也提供在清洁应用中使用这种α-半乳糖苷酶的方法。在一些实施方案中,所述清洁组合物还包含至少一种表面活性剂。在一些优选的实施方案中,所述清洁组合物具有至少约pH 5.0的工作pH。本发明也提供使用本发明的清洁组合物清洁物体的方法。
在一些实施方案中,该α-半乳糖苷酶具有与里氏木霉α-半乳糖苷酶至少约70%、至少约80%、至少约90%、至少约95%或至少约98%同一性的氨基酸序列。在其他一些实施方案中,该α-半乳糖苷酶与里氏木霉α-半乳糖苷酶免疫学交叉反应。
在一些实施方案中,本发明的清洁组合物是固体(例如,粉剂或片剂),而在其他实施方案中,它们是液体、凝胶剂、泡沫剂或其他形式。在一些优选的实施方案中,将所述清洁组合物配制为衣物洗涤剂、盘碟洗涤剂或洗衣添加剂。因而在一些优选的实施方案中,所述清洁组合物还包含用于降解非淀粉食物多糖的至少一种额外酶,所述的额外酶包括,但不限于以下酶,如半纤维素酶、甘露聚糖酶、果胶酶或木聚糖酶。在又一些优选的实施方案中,所述清洁组合物还包含用于降解其他污渍成分的至少一种额外酶,所述的额外酶包括,但不限以下酶,如蛋白酶、淀粉酶、纤维素酶、脂肪酶、角质酶或氧化还原酶。实际上,与本发明的α-半乳糖苷酶组合使用的合适酶包括,但不限于半纤维素酶、过氧化物酶、蛋白酶、纤维素酶、木聚糖酶、脂肪酶、磷脂酶、酯酶、角质酶、果胶酶、角蛋白酶、还原酶、氧化酶、酚氧化酶、脂肪加氧酶、木质素酶、支链淀粉酶、鞣酸酶、戊聚糖酶、黑素酶(malanase)、β-葡聚糖酶、阿拉伯糖苷酶、透明质酸酶、软骨素酶、漆酶和淀粉酶或其混合物。在一些实施方案中,使用酶的组合(即“混合物”),所述的组合包括与α-半乳糖苷酶联合的常规应用的酶,如蛋白酶、脂肪酶、角质酶和/或纤维素酶。

本发明提供包含分离的α-半乳糖苷酶的清洁组合物。在一些特别优选的实施方案中,所述的分离α-半乳糖苷酶包含与来自里氏木霉(Trichoderma reesei)的α-半乳糖苷酶相关的氨基酸序列。本发明也提供在清洁应用中使用所述α-半乳糖苷酶的方法。

在一些实施方案中,除信号序列之外,该融合蛋白还含有“载体蛋白”,其中所述的载体蛋白是里氏木霉宿主细胞内源且被高水平分泌的蛋白质的一部分。合适的载体蛋白包括,但不限于里氏木霉甘露聚糖酶I(Man5A或MANI)、里氏木霉纤维二糖水解酶II(Cel6A或CBHII){参见例如Paloheimo等,Appl.Environ.Microbiol.,69:7073-7082[2003])或里氏木霉纤维二糖水解酶I(CBHI)的那些载体蛋白。在一些实施方案中,该载体蛋白是包括CBHI核心区和部分CBHI接头区的截短里氏木霉CBHl蛋白。在一些实施方案中,本发明包括编码融合蛋白的核酸,其中所述的融合蛋白从氨基端至羧基端含有有效连接的信号序列、载体蛋白和α-半乳糖苷酶。
在一些实施方案中,该α-半乳糖苷酶的编码序列进行密码子优化,以便在所用宿主细胞中表达该α-半乳糖苷酶。由于列出众多宿主细胞(包括里氏木霉)中每个密码子用法的密码子选择表是本领域已知的(参见例如Nakamura等,NuclAcids Res.,28:292[2000])或可容易衍生,故而容易地设计此类核酸以产生待表达的α-半乳糖苷酶的氨基酸序列。
除了编码序列外,在一些实施方案中,该核酸还包含对于在宿主细胞中表达所述α-半乳糖苷酶必需的其他元件。例如,在一些实施方案中,该核酸含有用于转录编码序列的启动子和转录终止子。示例性启动子包括,但不限于里氏木霉cbh1、cbh2、egl1、egl2、eg5、xln1和xln2启动子或其杂合体或截短形式。例如,在一些实施方案中,该启动子是里氏木霉cbh1启动子。合适的终止子包括,但不限于里氏木霉cbh1、cbh2、egl1、egl2、eg5、xln1和xln2终止子和众多其他终止子,包括例如来自黑曲霉(A.niger)或泡盛曲霉(A.awamori)葡糖淀粉酶基因(参见前文Nunberg等[1984]和前文Boel等[1984])、构巢曲霉(Aspergillus nidulans)邻氨基苯甲酸合酶基因、米曲霉(Aspergillus oryzae)TAKA淀粉酶基因或构巢曲霉trpC(Punt等,Gene 56:117-124[1987])的终止子。在一些实施方案中,启动子和/或终止子相对于木霉宿主细胞是天然的,而在其他实施方案中,它们是非内源性的。
在一些实施方案中,里氏木霉宿主细胞用于表达所述α-半乳糖苷酶。在一些优选的实施方案中,遗传地修饰里氏木霉细胞以减少该细胞内源的分泌性蛋白质表达。在一些实施方案中,该细胞含有已经被缺失或失活的一个或多个天然基因,尤其编码分泌性蛋白质的基因。例如,在一些实施方案中,缺失或失活一个或多个蛋白酶编码基因(例如天冬氨酰蛋白酶编码基因;参见Berka等,Gene 86:153-162[1990]和美国专利号6,509,171)或纤维素酶编码基因。在一些实施方案中,木霉宿主细胞是在cbh1、cbh2和egl1与egl2基因中含有失活性缺失的里氏木霉宿主细胞,如WO 05/001036中描述。在一些实施方案中,上述核酸存在于木霉宿主细胞的核基因组内,而在其他实施方案中,它存在于该木霉宿主细胞内复制的质粒中。
意图使用众多合适技术(例如电穿孔、细胞核显微注射、转导、转染[例如脂质转染法介导和DEAE-Dextrin介导的转染]、与磷酸钙DNA沉淀物孵育、DNA包被的微粒的高速轰击和原生质体融合)的任一项技术将所述核酸导入木霉宿主细胞。常见转化技术是本领域已知的(见例如美国专利号6,022,725;美国专利号6,103,490;美国专利号6,268,328和美国公开专利申请20060041113、20060040353、20060040353及20050208623,全部文献通过引用的方式并入本文)。在一些实施方案中,制备用于转化的木霉包括从真菌菌丝体制备原生质体。(参见Campbell等,Curr.Genet.16:53-56[1989])。在一些实施方案中,菌丝体从萌发的营养性孢子获得。
在一些实施方案中,一旦α-半乳糖苷酶被分泌至培养基,则使用本领域已知的任一便利方法(例如通过沉淀法、离心法、亲和法、过滤法和任何其他方法)回收该α-半乳糖苷酶。例如,使用亲和层析法(Tilbeurgh等,FEBS Lett.,16:215[1984]);离子交换层析法(Goyal等,Biores.Technol.,36:37[1991];Fliess等,Eur.J.Appl.Microbiol.Biotechnol.,17:314[1983];Bhikhabhai等,J.Appl.Biochem.6:336[1984];和Ellouz等,Chromatography 396:307[1987]),包括使用高分辨力材料的离子交换法(Medve等,(J.Chromatography A 808:153[1998];疏水相互作用层析法(Tomaz和Queiroz,J.Chromatography A 865:123[1999];双相分配法(Brumbauer等,(Bioseparation 7:287[1999]);乙醇沉淀法;反相HPLC;在硅胶或阳离子交换树脂(例如DEAE)上层析法;层析聚焦法;SDS-PAGE;硫酸铵沉淀法;或凝胶过滤法(使用例如Sephadex G-75)。在一些实施方案中,该α-半乳糖苷酶不从培养基的其他成分中纯化出来即使用。在这些实施方案的部分中,所述培养基仅进行浓缩并且随后不将所述蛋白质从生长培养基的其他成分中进一步纯化出来即使用,或不作任何其他修饰即使用。
清洁组合物
本发明提供包含上述α-半乳糖苷酶的清洁组合物。在一些实施方案中,该清洁组合物是织物清洁组合物(即衣物洗涤剂)、表面清洁组合物、盘碟清洁组合物或自动洗碗机用洗涤剂组合物。示例性清洁组合物的配方在通过引用方式并入本文的WO0001826中极详细地描述。
在一些实施方案中,主题清洁组合物(例如衣物或盘碟洗涤剂)含有约1%到约80%(例如约5%到约50%)(按重量计)的至少一种表面活性剂(例如非离子表面活性剂、阳离子表面活性剂、阴离子表面活性剂或两性离子表面活性剂或其任意混合物)。示例性表面活性剂包括,但不限于烷基苯磺酸盐(ABS),包括直链烷基苯磺酸盐和直链烷基磺酸钠、烷基苯氧基聚乙氧基乙醇(例如,壬基苯氧基乙氧基化物或壬基酚)、二乙醇胺、三乙醇胺和单乙醇胺。在洗涤剂、尤其在衣物洗涤剂中可以存在的示例性表面活性剂于美国专利号3,664,961、3,919,678、4,222,905和4,239,659中描述。
在一些实施方案中,所述清洁组合物为固体(例如处于粉末或片状形式)或液体形式。在一些额外的实施方案中,所述清洁组合物还包含至少一种缓冲剂(例如碳酸钠、碳酸氢钠)、洗涤剂助洗剂、漂白剂、漂白激活剂、酶、酶稳定剂、增泡剂、抑制剂、防黯剂、防蚀剂、悬污剂、脱污剂、杀菌剂、pH调节剂、非助洗剂碱性源、螯合剂,有机或无机填充剂、溶剂、水溶助长剂、荧光增白剂、染料、香料等。在一些实施方案中,该清洁组合物作为衣物洗涤添加剂在使用前与洗涤剂组合。
在一些实施方案中,主题清洁组合物含有另外的非淀粉食物多糖降解酶(例如半纤维素酶、甘露聚糖酶、果胶酶、木聚糖酶或果胶酸裂合酶)和任选的含有一种或多种额外的酶如蛋白酶,如枯草蛋白酶和/或SSI蛋白质、脂肪酶、淀粉酶、纤维素酶、角质酶、脂肪酶、氧化还原酶等,用于去除其他污渍。
在洗涤清洁组合物中有用的多种其他成分也用于本文提供的组合物中,所述的其他成分包括其他活性成分、载体、水溶助长剂、加工助剂、染料或色素、用于液体制剂的溶剂等。在需要额外起泡作用的一些实施方案中,将增泡剂如C10-C16链烷醇酰胺掺入所述组合物,一般以约1%-约10%的水平。
在一些实施方案中,该洗涤剂组合物包含水和/或其他溶剂作为载体。适于使用低分子量伯醇或仲醇,例如甲醇、乙醇、丙醇和异丙醇。一元醇优选用于增溶表面活性剂,不过也可使用多元醇,如含有约2个-约6个碳原子和约2个-约6个羟基的那些多元醇(例如1,3-丙二醇、乙二醇、甘油和1,2-丙二醇)。在一些实施方案中,该组合物包含约5%-约90%(一般约10%-约50%)的此类载体。
在一些实施方案中,如此配制本文的洗涤剂组合物,从而在含水清洁操作中使用期间,洗涤水具有约5.0-约11.5的pH。因此,制成品通常在这个范围内配制。用于控制pH位于推荐使用水平上的技术包括使用缓冲剂、碱、酸等,并且是本领域技术人员熟知的。在一些实施方案中,该清洁组合物是一种自动餐具洗涤洗涤剂,其具有约pH 9.0-约pH 11.5、约pH9.0-约pH 9.5、约pH 9.5-约pH 10.0、约pH 10.0-约pH 10.5、约pH 10.5-约pH 11.0或约pH 11.0-约pH 11.5的工作pH。在一些其他实施方案中,该清洁组合物是一种液体衣物洗涤剂,其具有约pH 7.5-约pH 8.5、约pH7.5-约pH 8.0或约pH 8.0-约pH 8.5的工作pH。在一些其他实施方案中,该清洁组合物是一种固体衣物洗涤剂,其具有约pH 9.5-约pH 10.5、约pH9.5-约pH 10.0或约pH 10.0-约pH 10.5的工作pH。
本文所述的清洁组合物要求有效量的所述α-半乳糖苷酶。在一些实施方案中,该α-半乳糖苷酶在所述清洁组合物中的工作浓度是约0.01ppm(百万分之,w/v)-约100ppm、约0.01ppm-约0.05ppm、约0.05ppm-约0.1ppm、约0.1ppm-约0.5ppm、约0.5ppm-约1ppm、约1ppm-约5ppm、约5ppm-约10ppm或约10ppm-约100ppm。
多种漂白化合物如过碳酸盐、过硼酸盐等也可在本发明的清洁组合物中使用。在一些实施方案中,这些漂白化合物一般以按重量计约1%-约15%的水平存在。在一些额外的实施方案,此类组合物也含有本领域已知的漂白激活剂(例如四乙酰基乙二胺、壬酰氧基苯磺酸盐等)。使用水平一般是按重量计约1%-约10%。
多种脱污剂,尤其阴离子寡酯型脱污剂、多种螯合剂,尤其氨基膦酸盐和乙二胺二琥珀酸盐、多种粘质污渍去除剂,尤其乙氧基化四亚乙基戊胺、多种分散剂,尤其聚丙烯酸盐和聚天冬氨酸盐、多种增白剂,尤其阴离子增白剂、多种抑泡剂,尤其硅氧烷和仲醇、多种织物柔软剂,尤其蒙脱石等也可在本发明的组合物中以按重量计约1%-约35%的水平使用。标准配方是本领域技术人员熟知的。
酶稳定剂也可在本发明的清洁组合物中使用。此类稳定剂包括,但不限于丙二醇(优选约1%-约10%)、甲酸钠(优选约0.1%-约1%)和甲酸钙(优选约0.1%-约1%)。
在一些实施方案中,硬质表面清洁组合物和织物清洁组合物还包含按重量计约5%-约50%水平的多种助洗剂。常见助洗剂包括1-10微米沸石、多羧酸盐如柠檬酸盐和氧联二琥珀酸盐、层叠硅酸盐、磷酸盐等。其他常用助洗剂列于标准配方中。
其他任选成分包括螯合剂、粘质污渍去除/防再沉积剂、聚合物分散剂、漂白剂、增白剂、抑泡剂、溶剂和美观剂。
本发明的清洁组合物可在合适的清洁方法中使用。在一些实施方案中,该清洁方法包括:使分离的α-半乳糖苷酶与物体(例如织物或盘碟)在适于所述α-半乳糖苷酶活性的条件下接触以清洁该物体,其中所述的α-半乳糖苷酶包含与里氏木霉α-半乳糖苷酶相关的氨基酸序列。根据所用清洁组合物的工作pH,将所述α-半乳糖苷酶与该物体在例如约pH 5-约6.5、约pH 6.5-约7.5、约pH 7.5-约8.5、约pH 9.5-约10.5或约pH 10.0-约11.5的pH下接触。在一些实施方案中,该物体是弄脏的物体并且在一些其他实施方案中,该物体被含有非淀粉食物多糖如半乳甘露聚糖胶(例如,瓜尔胶或利马豆胶等)的食品弄脏。在一些其他实施方案中,该物体被巧克力浆、冰淇淋或色拉调味料弄脏。
本文所述的清洁组合物比不含α-半乳糖苷酶的同等清洁组合物在去除某些污渍(例如因含有半乳甘露聚糖多糖的食品所致的污渍)方面更有效。在一些实施方案中,本发明的清洁组合物与不含α-半乳糖苷酶的其他同等清洁组合物相比,在污渍去除方面更有效。使用基于标准反射计的测定法,例如实施例4中描述的那种测定法,本发明的一些清洁组合物比不合α-半乳糖苷酶的同等清洁组合物多去除至少约20%、至少约40%、至少约60%、至少约80%或至少约90%的污渍和/或使其褪色。

实施例2
里氏木霉细胞的转化
全部载体通过粒子轰击法转移至最初从RL-P37(Sheir-Neiss等,Appl.Microbiol.Biotechnol.,20:46-53[1984];美国专利号4,797,361)衍生的四重缺失(Achb1、Acbh2、Δegl1和Aegl2)里氏木霉菌株(WO 05/001036)或1A52pyr4-菌株中。
制备来自待转化的木霉菌株的孢子混悬液(约5×108个孢子/ml)。将100μl~200μl孢子悬浮液涂布在MM乙酰胺培养基的平板中心上(MM乙酰胺培养基具有以下组成:0.6g/L乙酰胺;1.68g/L CsCl;20g/L葡萄糖;20g/L KH2PO4;0.6g/L CaCl2.2H2O;1ml/L 1000×微量元素溶液;20g/LNoble琼脂;pH 5.5。1000×微量元素溶液含有5.0g/L FeSO4·7H2O、1.6g/LMnSO4·H2O、1.4g/L ZnSO4·7H2O和1.0g/L CoCl2·6H2O)。随后使该孢子混悬液在MM乙酰胺培养基的表面上干燥。
实施例3
酶活性分析
在培养瓶中培育含有agl1、agl2、agl3和man1的载体的细胞培养物并且使用SDS PAGE分析来自每种培养物的上清液。对于每种培养上清液,使用4-硝苯基-α-D-吡喃半乳糖苷作为底物在Mcl1vaine缓冲液中测量α-半乳糖苷酶活性。酶活性测定法使用Sigma方案进行(α-半乳糖苷酶的酶测定法,Sigma产品信息;还参见McCleary,Meth.Enzymol.,160:627-632[1988];和来自Megazyme公司黑曲霉和瓜尔豆的α-半乳糖苷酶技术数据页),如下简述。
首先,0.10ml底物添加至16×125mm玻璃管,所述玻璃管随后在水浴中通过温育至少5分钟加热至想要的温度。随后,将0.10ml稀释的酶以15秒间隔添加至每只管内并涡旋混合酶与底物。该混合物在试验的预定温度(30℃、37℃、40℃、45℃、60℃和75℃)下温育5分钟。为终止反应,以相同的15秒间隔添加3.0ml的2%碳酸钠溶液。使溶液混合并从水浴取出所述管以在410nm下读数。
在这些实验中,将所述酶稀释液在每个pH下(pH2.1、2.5、3、4、5、6、7和8的Mcl1vaine;pH 4.5的0.1M乙酸钠)的适宜缓冲液中制备为10mM。初步试验的酶底物是4-硝苯基-α-D-吡喃半乳糖苷(Sigma,目录号:877,MW:301.25)。在每个试验中包括空白,所述空白包含底物、终止试剂和酶空白(使用对硝基苯酚作为底物参照)。
图2-4中所示的SDS-PAGE凝胶表明对于每种上清液,产生了尺寸大致正确的蛋白质。在所用的分析条件下,AGL1(其具有预测分子量45.7kDa)具有pH 5的最适pH和约60℃的最适温度(图2),AGL2(其具有预测分子量79.5kDa)具有pH 4-5的最适pH和约60℃的最适温度(图3)并且AGL3(其具有预测分子量66.3kDa)具有pH 2-4的最适pH、在pH 4.5下约60℃的最适温度和在pH 2.5下约45℃的最适温度(图4)。
实施例4
木霉α-半乳糖苷酶蛋白清洁活性的圆盘测定分析法
使用以下方法测试AGL1、AGL2和AGL3清洁被巧克力浆、色拉调味料和瓜尔胶色素弄脏的小块布样的能力。
用带有色素的色拉调味料(STC CFT CS-6)、巧克力浆(STC EMPA160)和瓜尔胶色素(STC CFT CS-43)弄脏棉质小块布样(Test Fabrics,Inc.WestPittston,PA,USA)。巧克力冰淇淋圆形物(10cm棉质小块布样上的4cm污渍)从Warwick-Equest Limited,Consett,County Durham,England获得。
用于微量平板测定法的小块布样用配备5/8英寸冲压裁剪机的B型纺织品冲床(textile Punch Press Mode1B)切成15cm圆形物(圆盘)。将单个圆盘置于24孔微量平板(Costar 3526)的各个孔内。将每升含有1.5mlAATCC HDL(标准液体洗涤剂)洗涤剂、50mMHepes缓冲液(pH 7.4)的1ml洗涤溶液添加至各孔。用正压式移液器(positive displacement pipette)添加1~20μg稀释的酶。所述AATCC 2003标准液体洗涤剂含有12%直链烷基磺酸盐、8%脂肪醇乙氧基化物、8%丙二醇、1.2%柠檬酸、4%脂防酸和4%氢氧化钠,余量是水。对照孔不含酶。该微量平板用其塑料盖盖好并在37℃温育,伴以100转/分钟柔和转动。4~16小时后,上清液通过抽吸去除并且每孔用1.5ml Dulbecco′s PBS(pH 7.3)洗涤3次并用1.5ml蒸馏水洗涤3次。每个圆盘从各孔取出并在空气中干燥过夜。圆盘以目视方式检查并且用在标准白色色板(standard white tile)上校正过的Minolta反射计CR-200分析。用数据的标准差百分数计算平均L值,每个对照和试验样品通常进行一式四份。
使用强效洗涤剂(HDD)或自动餐具洗涤机(ADW)洗涤剂的实验利用无磷酸盐的pH 10的0.015%~0.1%AATCC HDD和无磷酸盐的0.015%~0.1%WFK ADW洗涤剂B型进行。无增白剂的AATCC 1993标准参考强效洗涤剂含有18%直链烷基磺酸盐、2%直链脂肪醇乙氧基化物和碳酸钠至100%、25%ZeoliteA、18%碳酸钠、0.5%硅酸钠、22.13%硫酸钠、10%水分(moisture)和共聚物、酶或羧甲基纤维素的空间(6.28%)。无增白剂且无磷酸盐的WFK自动餐具洗涤机洗涤剂B型含有30%无水柠檬酸钠、12%马来酸钠盐、一水合过硼酸钠、2%四乙酰乙二胺、25%二硅酸钠、2%直链脂肪醇乙氧基化物和无水碳酸钠至100%。
在以下实施例和附图中,含有AGL1的蛋白质提取物称作NSP-6,含有AGL2的蛋白质提取物称作NSP-8并且含有AGL3的蛋白质提取物称作NSP-9。
如图5中所示,使用微量平板圆盘法,NSP-6(α-半乳糖苷酶1)、NSP-8(α-半乳糖苷酶2)和NSP-9(α-半乳糖苷酶3)在0.15%AATCC强效液体洗涤剂中对巧克力浆污渍显示优异的清洁作用。NSP-20是一种β-CWDE,是细胞壁降解酶的缩写。图6显示在0.022%的AATCC强效液体洗涤剂(pH 7.4)中α-半乳糖苷酶1对色拉调味料污渍的清洁作用。图7显示低浓度(0.5~1.0ppm)的NSP-8(α-半乳糖苷酶2)在0.15%AATCC强效液体洗涤剂中对瓜尔胶色素技术性污渍产生明显的清洁作用。
实施例5
木霉α-半乳糖苷酶蛋白质清洁活性的涤垢仪分析
涤垢仪研究使用了维持在37℃的7243S型6钵(pot)涤垢仪(U.S.Testing,Co.Inc.Hoboken,N.J.)。搅拌速度设为100转/分钟。将在棉质小块布样上的6个巧克力冰淇淋圆形物添加至1升含有6gpg硬度(稀释自含有1.735M氯化钙和0.67M氯化镁的15000gPg硬度贮存溶液)和50mMHepes缓冲液(pH 7.4)的AATCC HDL洗涤剂。
图8显示α-半乳糖苷酶2(NSP-8)在涤垢仪条件下以1ppm酶在30分钟内清洁了冰淇淋小块布样。
图9显示全部三种α-半乳糖苷酶并且尤其α-半乳糖苷酶2(NSP-8)在微量平板圆盘法中表现了明显的清洁作用(如实施例中所描述,在0.015%自动餐具洗涤机洗涤剂(WFK)(pH 10.5)或在0.015%AATCC固体衣物洗涤剂(pH 10.2)中以20ppm配制时)。
以上实施例证实里氏木霉α-半乳糖苷酶从被色拉调味料、巧克力浆、冰淇淋和瓜尔胶色素污渍弄脏的棉质小块布样上有效去除污渍。对瓜尔胶技术性污渍的活性可以作为清洁作用的基础,因为色拉调味料和冰淇淋经常含有瓜尔胶作为一种成分。测试的α-半乳糖苷酶在远超预期的pH范围内表现良好。

Claims (19)
1.包含分离的α-半乳糖苷酶的清洁组合物,所述α-半乳糖苷酶包含与里氏木霉的α-半乳糖苷酶有至少90%同一性的氨基酸序列。
2.权利要求1的清洁组合物,且还包含至少一种表面活性剂。
3.权利要求1的清洁组合物,其中所述的清洁组合物具有大于pH 5的工作pH。
4.权利要求1的清洁组合物,其中所述的清洁组合物是固体。
5.权利要求1的清洁组合物,其中所述的清洁组合物是液体。
6.权利要求1的清洁组合物,其中所述的清洁组合物包含衣物洗涤剂。
7.权利要求1的清洁组合物,其中所述的清洁组合物包含盘碟洗涤剂。
8.权利要求1的清洁组合物,且还包含一种或多种额外的酶。
9.权利要求8的清洁组合物,其中所述额外的酶选自半纤维素酶、甘露聚糖酶、果胶酶、淀粉酶、木聚糖酶、胶质裂合酶、蛋白酶、纤维素酶、角质酶、脂肪酶和氧化还原酶。
10.权利要求1的清洁组合物,其中所述的分离α-半乳糖苷酶与里氏木霉的α-半乳糖苷酶免疫学交叉反应。
11.一种清洁方法,包括:
将分离的α-半乳糖苷酶与物体在适于所述α-半乳糖苷酶活性的条件下接触以清洁所述物体,其中所述的α-半乳糖苷酶包含与里氏木霉α-半乳糖苷酶有至少90%同一性的氨基酸序列。
12.权利要求11的方法,其中所述的α-半乳糖苷酶与所述物体在大于pH 5的pH下接触。
13.权利要求11的方法,其中所述物体是被弄脏的物体。
14.权利要求13的方法,其中所述被弄脏的物体包含非淀粉食物多糖。
15.权利要求14的方法,其中所述的非淀粉食物多糖是半乳甘露聚糖胶。
16.权利要求14的方法,其中所述的非淀粉食物多糖是瓜尔胶或利马豆胶。
17.权利要求13的方法,其中所述被弄脏的物体被巧克力浆、冰淇淋或色拉调味料弄脏。
18.权利要求11的方法,其中所述的物体是织物。
19.权利要求11的方法,其中所述的物体是餐具。
