Description
技术领域
本发明涉及分子标记技术,具体涉及一种多花黄精微卫星分子遗传标记。
背景技术
微卫星DNA又称作短串连重复(Short Tandem Repeats,STRs)、简单重复序列(Simple Sequence Repeat,SSRs)、简单序列长度多态性(SSLP)。是指基因组中以1~6个核苷酸为单位串联组成的核苷酸序列。根据重复单元的构成,微卫星DNA序列被分为3种类型:单一型,复合型(compound)和间断型(interrupted)。例如:
单一型:ATATATATATATATATATATATAT
复合型:ATATATCACACACACACACAC
间断型:ATATATCA ATATATCA ATATATA
微卫星DNA作为一种分子标记具有以下特点:广泛分布于真核生物的基因组中;核心序列的重复次数在个体间呈高度变异性,多态性信息容量高:呈共显性,遵循孟德尔法则遗传;选择中性等等。
基于以上特点,微卫星DNA被广泛用于植物遗传图谱的构建、基因定位、品种鉴定和种质保存、数量性状基因的分析、辅助育种、构建指纹图谱、遗传多样性及物种进化与亲缘关系等方面的研究。
多花黄精为百合科黄精属多年生草本植物,是我国特有植物,药用其根茎。2005年版《中国药典》中以黄精,多花黄精和滇黄精的干燥根茎作为黄精药材入药。黄精的使用历史至今己逾两千年,是著名的食疗补益中药。其肉质根状茎具有补气养阴、健脾润肺、益肾生津等功效,被视为传统的中药材,并誉有“血气双补之土”之称。另外多花黄精还具有食用和观赏价值。
近年来,人们对黄精及其黄精属植物的研究不断深入。现代药理研究证实,其化学成分主要有:黄精多糖、粘液质、淀粉、甾体皂苷、蒽醌类化合物、生物碱、强心苷、木脂素、维生素和多种对人体有用的氨基酸等化合物。目前有研究表明黄精多糖能增强机体的免疫、延缓衰老、抗菌、抗病毒和降脂、降糖作用。李循等研究发现黄精多糖能活化巨噬细胞、活化淋巴细胞、提高NK细胞和LAK细胞的活性、促进有丝分裂作用、增强网状内皮系统、促进细胞因子分泌、增强红细胞免疫等作用而提高宿主免疫功能。而张庭亭通过研究小鼠发现PSP可提高7月龄小鼠全血中SOD以及GSH-Px的活性,降低小鼠心、肝、脑组织中LF和MDA含量;说明黄精多糖在一定程度上有抗衰老作用的。辜红梅等通过研究发现黄精多糖能显著提高病毒感染的Vero细胞的活力,对细胞有保护作用。黄精甾体皂苷具有去痰止咳之功效,还具有抗炎、抗肿瘤、抗真菌等作用。
中药上的黄精包括黄精,多花黄精,滇黄精。黄精属植物还有其他的一些药用植物,如玉竹,长梗黄精等,但它们与黄精在药理和药效上是存在差异的,有些不能混用。如黄精和玉竹在临床上就是不能混用的,入药时需将它们区分开来。
黄精的分子标记研究起步很晚,目前报道的只是RAPD分子标记,ISSR分子标记(周晔,RAPD标记法鉴定中药黄精及长梗黄精的研究;周晔,ISSR法鉴定中药黄精和卷叶黄精),微卫星标记尚未见报道。但RAPD分子标记稳定性,重复性差,ISSR分子标记通常为显性标记,不能区分纯和基因型和杂合基因型。
发明内容
本发明解决的技术问题:分离克隆多花黄精的微卫星DNA分子标记,建立多花黄精微卫星DNA技术体系并用这些分子标记研究多花黄精遗传多样性,黄精属植物种间和居群间的变异和进化关系以及进行多花黄精品种鉴定。
实现上述发明的技术方案:包括多花黄精微卫星(CT)n富集文库的构建;含微卫星序列阳性克隆的筛选及测序,得到55个含有微卫星重复序列的克隆;筛选得到14个多态性高的微卫星分子标记Pc1、Pc2、Pc3、Pc4、Pc5、Pc6、Pc7、Pc8、Pc9、Pc10、Pc11、Pc12、Pc13、Pc14;Pc1为480个核苷酸,Pc2为550个核苷酸,Pc3为310个核苷酸,Pc4为338个核苷酸,Pc5为630个核苷酸,Pc6为767个核苷酸,Pc7为435个核苷酸,Pc8为584个核苷酸,Pc9为399个核苷酸,Pc10为595个核苷酸,Pc11为677个核苷酸,Pc12为512个核苷酸,Pc13为488个核苷酸,Pc14为473个核苷酸。

具体实施方式
实施例1:
1、多花黄精微卫星DNA富集文库的构建
1.1基因组的提取与酶切:
用Doyle J介绍的CTAB法(Doyle J.DNA protocols for plants-CTAB totalDNA isolation[A].In Hewitt GM,Johnston A(eds.),Molecular Techniquesin Taxonomy[M].Berlin:Springer,1991,283-293)提取基因组,用限制性内切酶Sau3AI(购自TaKaRa公司)酶切基因组,酶切产物在1%琼脂糖凝胶上电泳,紫外光下切下300-900bp大小的DNA片段并用胶回收试剂盒(购自上海生工)纯化回收的酶切产物。
1.2加接头:
将纯化回收的酶切产物的两端加入接头:
LA(5’-GGCCAGAGACCCCAAGCTTCG-3’)
LB(5’-PO4-GATCCGAAGCTTGGGGTCTCTGGCC-3’),
在T4连接酶(购自Promega公司)作用下16℃水浴连接过夜。
1.3PCR检测接头连接:
以LA为引物,以1.2工序制的连接产物为模板进行PCR,(50μl反应体系如下:25μl TaqGreen(购自Promega公司),5μl引物LA,5μl连接产物,加水H2O补至50μl。PCR反应程序如下:94℃预变性5分钟,94℃变性50秒,56℃退火一分钟,72℃延伸2分钟,变性至退火三个步骤重复25次,72℃充分延伸10分钟。PCR产物用纯化试剂盒(购于Axygen公司)进行纯化。
1.4磁珠富集:
将1.3所制的纯化产物与杂交探针(CT)15在70℃下杂交一个半小时。将杂交液加入已平衡好的磁珠,室温放置30分钟。将离心管依次用6×SSC,2×SSC,1×SSC于60℃洗两次,最后用0.1×SSC室温洗一次。加入TE缓冲液,在PCR机上95℃变性15分钟,收集上清。
1.5PCR富集:
以磁珠富集产物为模板,LA为引物进行PCR扩增,25μl反应体系如下:
TaqGreen 12.5μl,LA 1.5μl,富集后的DNA2.5μl,加水补至25μl。PCR反应程序如下:94℃预变性3分钟,94℃变性35秒,60℃退火35秒,72℃延伸1分钟,变性至退火三个步骤重复25次,72℃充分延伸12分钟。PCR产物用纯化试剂盒进行纯化。
1.6感受态细胞的制备:
将1mL过夜培养的E.coli DH5α培养液接于50mL液体LB培养基中,37℃振荡培养2小时至对数生长期(OD600≈0.3-0.6)时取出。将菌液于4℃离心10分钟。弃上清,加入1mL 0.1mol/L预冷的Cacl2重新悬浮细胞,用枪头轻轻打匀。4℃离心10分钟。弃上清,加入预冷的甘油/CaCl2混合物重新悬浮菌体,用枪头打匀,-80℃保存。
1.7倒平板:
将LB固体培养基完全融化,冷却至50℃左右,配成LB/Amp/X-Gal/IPTG平板培养基(比例为:LB固体培养基100mL;Amp青霉素100μl;20%异丙基-β-D-硫代半乳糖苷(IPTG)7μl;2%5-溴-4-氯-3-吲哚-β-D-半乳糖苷(X-Gal)40μl),混合均匀后倒入一只培养皿里,待培养基凝固后放入37℃烘箱,晾干水蒸气。
1.8连接转化:
取1.5工序所制的PCR富集后的纯化产物加入pMD19-T载体(购自TaKaRa公司)于16℃水浴连接2小时,制成连接产物。
取出感受态细胞,冰上复苏至液态,加入上述连接产物,混匀。冰浴30分钟后放入42℃水浴锅,热激90秒,立即放入冰中冰浴2分钟,加入新鲜的LB培养基,37℃振荡培养1-1.5小时。取70μl培养液涂布于LB/Amp/X-gal/IPTG培养基上,37℃倒置培养过夜。
2.阳性克隆的筛选:
将平板上的白色单菌落分别放入事先装有LB/Amp培养基的EP管中。37℃振荡培养至菌液浑浊。15μl PCR反应体系包括:Taq酶0.075U(购自TaKaRa公司),10×Buffer1.5μl,dNTP0.2μM,mg2+1.5μM,引物0.4μM(LA,primer-c(CT15)),水。反应程序:94℃预变性3分钟,94℃变性50秒,56℃退火35秒,72℃延伸1分钟,变性至延伸三个步骤重复30次。最后72℃充分延伸12分钟,1.5%琼脂糖凝胶电泳检测。
3.阳性克隆测序及引物设计:阳性克隆送生物公司测序,共得到84条含有微卫星DNA的序列。根据微卫星DNA两侧的序列,利用软件primerprimer5设计引物,如表1所示。
实施例2:
2.1筛选有多态性的引物:
用1.1的CTAB法提取长梗黄精基因组。用设计的引物(表1)扩增基因组。反应程序:94℃预变性4分钟,94℃变性45秒,退火30秒(退火温度见表1),72℃延伸35秒,变性至退火三个步骤重复35次。最后72℃充分延伸8分钟。1.5%琼脂糖凝胶电泳检测。PCR产物用PAGE电泳进行检测,共得到14个有多态性的微卫星标记;Pc1为480个核苷酸,Pc2为550个核苷酸,Pc3为310个核苷酸,Pc4为338个核苷酸,Pc5为630个核苷酸,Pc6为767个核苷酸,Pc7为435个核苷酸,Pc8为584个核苷酸,Pc9为399个核苷酸,Pc10为595个核苷酸,Pc11为677个核苷酸,Pc12为512个核苷酸,Pc13为488个核苷酸,Pc14为473个核苷酸。
2.2结果分析:
使用Cervus3.0软件计算期望杂合度和观察杂合度。使用Genepop3.4软件计算哈代-温伯格以及连锁不平衡的值,以此来描述14个多花黄精微卫星DNA多态位点的特征。
由附图1-14可看出,本发明的14个微卫星序列的长度在供试的23个多花黄精中都出现的多样性,由此可见,本发明的这14个微卫星标记能用于研究多花黄精的遗传多样性,黄精属植物种间和居群间的变异和进化关系及多花黄精品种的鉴定,具有重复性好的特点,是一种可靠有效的分子标记。
表1中F表示5’端引物,R表示.3’端引物。
