Abstract
本发明公开了一种可以在口腔中以高溶出速率溶出其中所含有的辅酶Q10的咀嚼组合物。本发明还公开了一种用于改进辅酶Q10在口腔中溶出性能的方法,特别是用于以较高效率溶出咀嚼组合物中所含有的辅酶Q10到口腔中的方法。辅酶Q10被包裹在环糊精中,所述环糊精包合产物用作辅酶Q10成分。优选地,所述包合产物中所含的辅酶Q10是少量的,优选40wt%或更少,更优选20wt%。

技术领域
本发明涉及含有辅酶Q10的咀嚼组合物,更具体而言,本发明涉及含有在口腔中溶出性能改进的辅酶Q10的咀嚼组合物。本发明还涉及用于改进辅酶Q10的溶出性能的方法,更具体而言,还涉及用于改进来自含有辅酶Q10之咀嚼组合物中辅酶Q10在口腔中的溶出性能的方法。
背景技术
辅酶Q10是用于产生生物能的基本成分,其多见于真核细胞的线粒体中,在人体中作为产生ATP(三磷酸腺苷)的不可缺少的成分。
还已知辅酶Q10在体内起到优良的抗氧化剂的作用,预计其能有效地用于预防可能与体内活性氧物质相关的疾病,例如所谓的生活方式相关疾病,比如心肌梗塞、高血压、心绞痛和癌症。此外,还认为其能有效地用于预防脑病(比如阿尔茨海默病、帕金森病和抑郁),牙龈病,肌营养不良以及多种其它疾病,其还能有效地用于抗衰老以及由于其促进代谢的作用而用于抑制肥胖症。
身体所必需的部分辅酶Q10是通过内源性合成来提供的,另外是从食物中吸收的。然而已知内源性合成的辅酶Q10的量随年龄而减少,辅酶Q10仅能少量从食物中吸收。因此,体内提供的辅酶Q10的总量随年龄的增长而减少。因此,为了补充内源性合成的量的目的,当今普遍以例如食品和补充剂的形式外源性吸收辅酶Q10。
辅酶Q10是脂溶性物质。因此,目前可商购的口服剂型的辅酶Q10通常是通过将辅酶Q10晶体粉末分散或溶解在油中、或者将所述晶体粉末装入胶囊中而进行配制的。
还已知当加入到食品中时,辅酶Q10由于其它添加剂的影响而变得不稳定,尤其是当与维生素(特别是维生素E)共存时,辅酶Q10变得极不稳定(参见专利文献1)。由于维生素是药物、准药品(医薬部外品)、食物和饲料的常用成分,因此为了将辅酶Q10添加到制备物或制剂中,需要采取一些措施将其稳定在这些产品中。
用于稳定辅酶Q10的方法公开于专利文献1和2中,其包括将辅酶Q10包裹在环糊精中。更具体而言,专利文献1教导了将辅酶Q10包裹在环糊精中使得甚至在维生素和其它添加剂的存在下能稳定辅酶Q10,而不降低与辅酶Q10一起使用的维生素和其它成分的稳定性。此外,专利文献2教导了将辅酶Q10包裹在环糊精中而赋予了辅酶Q10抗物理或化学作用(尤其是抗紫外线作用),使得环糊精包合物可用作化妆品制剂的成分。因此,环糊精是在药学、化妆品和食品领域中广泛地用作敏感性分子的保护性包衣的物质。

发明内容
本发明待解决的问题
本发明的一个目的在于提供了一种含有在口腔中溶出性能改进的辅酶Q10的咀嚼组合物。本发明的另一个目的在于提供了一种用于改进辅酶Q10溶出性能的方法,更具体而言,提供了一种用于改进来自含有辅酶Q10之咀嚼组合物中的辅酶Q10在口腔中溶出性能的方法。
解决问题的方法
本发明人为解决上述问题而进行了广泛的研究,发现当辅酶Q10被包裹在环糊精中时,其在口腔中的溶出性能得到改进。本发明人还发现通过降低(而不是增加)添加到环糊精中的辅酶Q10的比例可显著提高辅酶Q10的溶出性能,并且确定利用含有适当比例的辅酶Q10的环糊精包合物可制备含有在口腔中具有良好溶出性能的辅酶Q10的咀嚼组合物(比如口香糖)。因此,基于这些发现而完成了本发明。
下面阐明包含在本发明中的实施方案:

(I-1):含有辅酶Q10包合物的咀嚼组合物,其中辅酶Q10被包裹在环糊精中。

发明效果
[11]本发明提供了一种含有具有良好溶出性能的辅酶Q10的辅酶Q10包合物,更具体而言,提供了一种含有适量辅酶Q10并且表现出改进的溶出性能的辅酶Q10包合物。利用所述辅酶Q10包合物能够制备可在口腔中有效释放大量辅酶Q10的咀嚼组合物(比如口香糖)。根据本发明,利用大量的含有低辅酶Q10含量的辅酶Q10包合物(而不是利用含有高辅酶Q10含量的辅酶Q10包合物)使得能够更多地吸收辅酶Q10,即使在两种情形下最终咀嚼组合物中辅酶Q10的含量相同。

[12]本发明涉及含有辅酶Q10的咀嚼组合物。本发明的咀嚼组合物是在口腔中被咀嚼的组合物,或通过在口腔中进行咀嚼而被吸收的组合物。这些组合物的具体实例包括食品比如口香糖,泡泡糖和其它胶(包括粘性胶、板胶和糖包衣胶),粘合胶,牛轧糖等,药物(比如咀嚼剂、咀嚼片、颗粒、锭剂、口含片、牙膏等,以及准药品比如牙膏等。所述咀嚼组合物优选是口香糖。所述咀嚼组合物还优选被糖包衣。
[13]本发明的咀嚼组合物包含以使得辅酶Q10被包裹在环糊精中的形式(以辅酶Q10包合物的形式)的辅酶Q10。所述包合物中辅酶Q10的比例可适当地选自1至80wt%的范围,上限优选为40wt%,更优选为20wt%,还更优选为10wt%;下限优选为1wt%,更优选为2wt%,还更优选为5wt%。更具体而言,所述范围优选为1至40wt%,更优选为2至20wt%,还更优选为2至10wt%。
[14]用于辅酶Q10包合物的环糊精是包合物的主分子,其由在环结构中通过α-1,4-键连接的6、7、8或更多个葡萄糖残基组成。通常已知的是α-、β-、γ-和δ-环糊精。虽然α-、β-、γ-和δ-环糊精以及其衍生物均可用于本发明中,但β-和γ-环糊精是优选的。γ-环糊精是更优选的。
用于本发明的辅酶Q10是泛醌,其中2,3-二甲氧基-5-甲基-6-聚异戊二烯基-1,4-苯醌的类异戊二烯侧链数为10。辅酶Q10是脂溶性物质,其还称为泛癸利酮、CoQ10或辅酶UQ10。用于本发明的辅酶Q10可以是任何来源的或者可以任何方法进行制备,只要其可用作药物或者可作为食物食用。例如,可通过专利文献特开昭54-122795号公报和特开昭55-19006号公报中公开的方法制备辅酶Q10。
可利用多种已知的方法将辅酶Q10包裹在环糊精中。这些已知方法的实例包括在水中将辅酶Q10和环糊精混捏、混合并研磨成粉、冷冻干燥、使用饱和溶液、以及共沉淀(参见例如WO 01/054730和Journal ActaPoloniae Pharmaceutica 52(5s),pp.379-386,1995)。
在制备辅酶Q10包合物的过程中,除辅酶Q10以外,其它成分也可包裹在环糊精中。这些成分的实例包括调味剂(比如l-薄荷醇、dl-薄荷醇、橘子精油等),维生素(比如维生素A、维生素D、维生素E、维生素K等),矿物质(比如钙、钾、镁等),酵母及其提取物,植物提取物(比如蒜提取物、绿茶提取物及其它提取物),以及功能性原料(比如类胡萝卜素、α-硫辛酸、甾醇、异黄酮、胶原、肽、氨基酸、L-肉碱、γ-氨基丁酸、牛磺酸、葡萄糖醛酸盐、大豆异黄酮、卵磷脂、透明质酸、硫酸软骨素、饱和或不饱和脂肪酸及其酯、植物纤维等)。
由此制备的辅酶Q10包合物在相对低含量的辅酶Q10(而不需要含有高含量的辅酶Q10)的情形下表现出辅酶Q10的显著改进的溶出性能。辅酶Q10含量的上限优选为40wt%,更优选为20wt%,还更优选为10wt%;下限优选为1wt%,更优选为2wt%,还更优选为5wt%。更具体而言,所述范围优选为1至40wt%,更优选为2至20wt%,还更优选为2至10wt%。
尽管在所述辅酶Q10包合物中辅酶Q10与环糊精的比例不限于上述条件,但其通常为1至80重量份/100重量份环糊精。优选2至40重量份,更优选5至20重量份。
所述咀嚼组合物可包含如上所述的辅酶Q10包合物形式的辅酶Q10。所述咀嚼组合物还可包含常规成分(取决于咀嚼组合物的类型和形状),并可通过常规方法进行制备。虽然所述咀嚼组合物中辅酶Q10包合物的比例不受限制,但其可适当地选自0.08至25wt%/100wt%咀嚼组合物的通常范围。所述范围优选为1至10wt%,更优选为2至5wt%。当以辅酶Q10含量进行计算时,所述咀嚼组合物中辅酶Q10包合物的含量优选为0.01至10wt%/100wt%咀嚼组合物,更优选为0.1至4wt%,还更优选为0.2至2wt%。

胶基可包含通常被包含在口香糖中的成分。这些成分的实例包括被认为是胶基食品添加剂的各种成分及其混合物;更具体而言,包含漆蜡、酯胶、榄香树脂、地蜡、红没药树脂、巴西棕榈蜡、小烛树蜡、愈创木树脂、银菊胶、古塔胶、杜仲胶、甘油脂肪酸酯、香脂、树脂、橡胶、橡胶再混合物、米糠蜡、醋酸乙烯酯树脂、甘蔗蜡、山达脂树脂、紫胶蜡、节路顿胶、蔗糖脂肪酸酯、香豆果树胶、失水山梨醇脂肪酸酯、香豆胶乳、滑石、碳酸钙、达玛树脂、糖胶树胶、托帕匡斯麻风树胶、卡斯德拉胶、解聚天然橡胶、尼日尔胶、乳香、石蜡、脂肪酸丙二醇酯、粉状纸浆、粉末碎屑、委内瑞拉糖胶树胶、安息香胶、霍霍巴蜡、聚异丁烯、聚丁烯、微晶蜡、乳香、巧克力铁线子、巴拉塔树胶、蜂蜡、没药树脂、日本蜡、褐煤蜡、羊毛脂、磷酸三钙、磷酸氢钙、牛奶、山榄胶、松香等。当胶基存在时,所述胶基的含量通常是咀嚼组合物的10至35wt%,优选20至30wt%。
甜味剂的实例包括单糖、二糖、寡糖、糖醇和高甜度甜味剂。优选的甜味剂的实例包括糖类(比如蔗糖、果糖、液体糖、葡萄糖、寡糖等)、高甜度甜味剂(比如阿斯帕坦、三氯蔗糖、AK糖、奇异果甜蛋白、甜叶菊、阿力甜、纽甜、木糖醇、糖精盐、甘草甜素等)。
着色剂的实例包括β-胡萝卜素、类胡萝卜素色素、红辣椒色素、胭脂树提取物、茜草色素、橙子色素、可可色素、栀子色素、叶绿素、紫草色素、赤藓红、酒石黄、洋葱色素、番茄色素、万寿菊色素、叶黄素、焦糖色素、叶绿素铜、葡萄皮色素、核黄素、核黄素5’-磷酸钠等。

软化剂的实例包括甘油、山梨醇、丙二醇等。
香料的实例包括薄荷醇、dl-薄荷醇、薄荷酮、香草醛、乙基香草醛、肉桂酸、胡椒醛、d-龙脑、麦芽醇、乙基麦芽醇、樟脑、氨基苯甲酸甲酯、肉桂酸甲酯、肉桂醇、N-甲基氨基苯甲酸甲酯、甲基β-萘基酮、苧烯、里哪醇、异硫氰酸烯丙酯等。
本发明的咀嚼组合物优选是口香糖,更优选是糖包衣胶。可通过例如Robert Hendrickson在Gum Base and Gum Products Technology和the Great American Chewing Gum Book(CAFOSA GUM S.A.,1974)中所述的方法和步骤制备口香糖。可在制备过程中的任何阶段添加辅酶Q10包合物,但是优选在接近制备结束时添加以防止辅酶Q10由于加热而被降解或破坏。
此外,在本发明的一个优选实施方案中,利用糖对所述咀嚼组合物进行包衣,所述糖衣料包含辅酶Q10包合物。咀嚼组合物的具体实例包括食品(比如糖包衣胶、粘合胶、牛轧糖等,其中利用糖对芯成分进行包衣),药物和准药品(比如糖包衣咀嚼剂、咀嚼片、颗粒、锭剂、口含片等,其中利用糖对芯成分进行包衣)。可根据常用的包衣技术利用含有辅酶Q10包合物的糖衣料对芯成分进行包衣来制备这些糖包衣产品。含有辅酶Q10包合物的糖衣料成分的实例包括但不限于糖类(比如精制白糖、葡萄糖、果糖、蔗糖、乳糖、麦芽糖、海藻糖等),糖醇(比如赤藻糖醇、麦芽糖醇、甘露醇、山梨醇、木糖醇等),以及多糖(比如阿拉伯胶、普鲁兰多糖、羟丙基纤维素等)。将这些糖之一或其混合物溶解在水中得到糖衣料溶液,然后利用包衣锅或自动糖包衣机通过反复包衣和干燥将所述糖衣料溶液施用于各种芯成分上,从而制得所期望的糖包衣产品。
糖衣料中辅酶Q10包合物的比例可以是使得在最终咀嚼组合物(糖包衣咀嚼组合物)中辅酶Q10包合物的含量为0.01至25wt%、优选1至10wt%、更优选2至5wt%的比例。可根据糖衣料和所述糖包衣咀嚼组合物的芯成分之间的重量比适当地设定所述含量。例如,在糖包衣咀嚼组合物含有50重量份糖衣料/100重量份芯成分的情形下,在100wt%糖衣料中辅酶Q10包合物的含量是1.5至75wt%,优选3至30wt%,更优选6至15wt%。
下述试验实施例中将显示,本发明的咀嚼组合物包含辅酶Q10包合物形式的辅酶Q10,其中所述辅酶Q10包合物含有低含量的辅酶Q10,所述辅酶Q10的含量为40wt%或更少,优选20wt%或更少,更优选10wt%或更少,从而显著提高辅酶Q10在口腔中的溶出效率。换句话说,不是利用少量的含有高辅酶Q10含量的辅酶Q10包合物(其中辅酶Q10的含量为40wt%以上),而是利用大量的含有低辅酶Q10含量的辅酶Q10包合物也能够更多地吸收辅酶Q10,即使在最终咀嚼组合物中辅酶Q10的含量在两种情形下是相同的。
因此,本发明的咀嚼组合物可用于含有辅酶Q10的食品,使得所述食品在咀嚼过程中辅酶Q10能被有效吸收(例如可被糖包衣的口香糖、弹性软糖和牛轧糖),含有辅酶Q10的药物和准药品(比如可被糖包衣的咀嚼剂、咀嚼片、颗粒、锭剂、口含片以及牙膏)。
(2)用于改进辅酶Q
10
溶出性能的方法
本发明还涉及用于改进辅酶Q10溶出性能的方法。
可通过将辅酶Q10包裹在环糊精中来实施所述方法。添加到环糊精中的辅酶Q10的比例通常可选自1至80wt%的范围;上限优选为40wt%,更优选为20wt%,还更优选为10wt%;下限优选为1wt%,更优选为2wt%,还更优选为5wt%。更具体而言,所述范围优选为1至40wt%,更优选为2至20wt%,还更优选为2至10wt%。加入到环糊精中的辅酶Q10的量越少(即将其从40wt%降低到30wt%,从30wt%降低到20wt%,从20wt%降低到10wt%),辅酶Q10的溶出性能就变得更好。基于这一发现实现了本发明。
虽然α-、β-、γ-和δ-环糊精中任何种类都可用于本发明中,但β-和γ-环糊精是优选的。γ-环糊精是更优选的。
在辅酶Q10包合物中辅酶Q10与环糊精的比例不限于上述情况,而是可为1至80重量份辅酶Q10/100重量份环糊精。所述比例优选为2至40重量份/100重量份环糊精、更优选5至20重量份/100重量份环糊精。
本发明的方法改进了辅酶Q10在口腔中的溶出性能。因此,所述方法尤其适于制备在口腔中进行咀嚼的组合物,或通过咀嚼它们来消费的组合物(即可咀嚼的组合物),例如食品(比如口香糖、弹性糖和牛轧糖等),药物和准药品(比如咀嚼剂、咀嚼片、颗粒、锭剂、口含片、牙膏等)。因此,本发明提供了一种用于改进含有辅酶Q10的咀嚼组合物的辅酶Q10在口腔中的溶出性能的方法。
所述方法包括利用含有被包裹在环糊精中的辅酶Q10(辅酶Q10包合物)作为辅酶Q10成分的组合物。如前所述,添加到环糊精中的辅酶Q10的量通常可选自1至80wt%的范围;上限优选为40wt%,更优选为20wt%,还更优选为10wt%;下限优选为1wt%,更优选为2wt%,还更优选为5wt%。更具体而言,所述范围优选为1至40wt%,更优选为2至20wt%,还更优选为2至10wt%。
在辅酶Q10包合物中辅酶Q10与环糊精的比例也如上所述,从而是1至80重量份/100重量份环糊精,优选为2至40重量份/100重量份环糊精、更优选5至20重量份/100重量份环糊精。
在本发明的方法中,除了辅酶Q10包合物以外,还可将适用于咀嚼组合物的类型和形状的常用成分加入到所述咀嚼组合物中,只要辅酶Q10包合物被用作辅酶Q10成分。虽然所述咀嚼组合物中辅酶Q10包合物的比例不受限制,但其可适当地选自常规范围0.08至25wt%/100wt%咀嚼组合物。所述范围优选为1至10wt%,更优选为2至5wt%。优选对上述范围的比例进行调整以使得辅酶Q10的含量通常为0.01至10wt%/100wt%咀嚼组合物,优选为0.1至4wt%/100wt%咀嚼组合物,更优选为0.2至2wt%/100wt%咀嚼组合物。
此外,在本发明的方法中,可利用含有辅酶Q10包合物的糖衣料包裹所述咀嚼组合物。用于制备这些糖衣料和糖包衣咀嚼组合物的方法以及所述糖衣料中辅酶Q10包合物的含量都与上述部分(1)中的相同,部分(1)中的描述同样可用于此处。

(1-1)样品1的制备
在3000份水中,通过加热溶解270份γ-环糊精并维持在70℃。通过加热熔融30份辅酶Q10并将其加入到上述所得的3270份的溶液中,在70℃搅拌24小时。喷雾干燥所得的溶液,得到290份的粉末(样品1,辅酶Q10:γ-环糊精=10:90(以重量计))。
(1-2)样品2的制备
重复制备(1-1)中的相同步骤,但使用240份γ-环糊精、3000份水和60份辅酶Q10,从而得到290份粉末(样品2,辅酶Q10:γ-环糊精=20:80(以重量计))。
(1-3)样品3的制备
重复制备(1-1)中的相同步骤,但使用180份γ-环糊精、3000份水和40份辅酶Q10,从而得到290份粉末(样品3,辅酶Q10:γ-环糊精=40:60(以重量计))。
(1-4)样品4的制备
在700份水中,通过加热溶解90份阿拉伯胶和90份糊精并维持在70℃。向880份的所得溶液中加入通过加热熔融的120份辅酶Q10,在70℃搅拌1小时。利用均质器以400kg/cm2乳化所得的溶液,喷雾干燥得到290份粉末(样品4,辅酶Q10:阿拉伯胶:糊精=40:30:30(以重量计))。
(1-5)样品5的制备
辅酶Q10晶体粉末(由San-Ei Gen F.F.I.,Inc制备)用作样品5的辅酶Q10。
(2)粘性胶的制备
利用上述所得的样品1-5中的一种,根据下述配方混合原料,使得100g所得粘性胶中辅酶Q10的含量为48mg,将每种混合物制模以形成粘性胶。具体步骤如下:首先在70℃捏合胶基与糖、玉米糖浆和甘油,将混合物与柠檬油No.2451*(调味剂)和SanYellowNo.3*(着色剂)相混合。然后将样品1、2、3、4或5中的一种加入到所得混合物中。冷却至室温后,对每种原料进行轧辊和制模以制备咀嚼胶(粘性胶)。用于下述配方的胶基购自Lotte,Co.,Ltd(胶基LT1)。

(3)辅酶含量的分析
(3-1)胶中辅酶Q10含量的分析
取由此制备的每种类型的粘性胶4g(精确量),置于具塞离心管中。向所述管中加入10ml 20%(v/v)四氢呋喃水溶液。然后将所述管装配Dimroth冷凝器,在于82℃的热浴中回流所述溶液30分钟(同时偶尔通过摇动进行混合)。然后将回流的溶液冷却至室温,加入15ml乙醚,之后通过摇动萃取10分钟。将萃取物在4000rpm下离心3分钟,然后将上层(乙醚层)倒入50ml的容量瓶中,将10ml乙醚加入到剩余的底层中,之后将混合物摇动10分钟并离心(萃取操作)。该萃取操作总计重复3次,将每次的上层(乙醚层)倒入上述容量瓶中,最后,用乙醚将所述容量瓶定容至50ml。向3毫升该溶液中加入无水乙醇至50ml。然后用0.45-μm PTFE滤器过滤所述溶液,回收的滤液用作分析样品。在下述条件下对每个样品进行HPLC以检测胶中辅酶Q10的含量(胶中CoQ10的含量)。表2中所示的结果证明每种粘性胶中辅酶Q10的含量都是0.048wt%。
(3-2)释放到唾液中的辅酶Q10的分析
取上述制备的每种类型的胶1g(精确量)(对于4个个体而言,总计4g),然后由4名对象以下述条件咀嚼1g每种所述胶。由4名对象咀嚼过的胶被分别置于具塞离心管中。然后根据咀嚼之前的相同步骤(见上述(3-1))反复萃取,每种回收滤液用作HPLC分析的样品以检测被咀嚼过的胶中辅酶Q10的含量(被咀嚼过的胶中CoQ10的含量)。

结果显示,通过利用含有辅酶Q10的样品1-4而提高了辅酶Q10向唾液中的释放,相比而言,0%的辅酶Q10从利用样品5(纯辅酶Q10)制备的胶中释放到唾液中。此外,样品1-4的结果显示,通过将辅酶Q10包裹在环糊精中(样品1-3)提高了辅酶Q10向唾液中的释放。另外,样品1-3的对比表明辅酶Q10的含量越小,辅酶Q10向唾液中的释放就越多。前述结果表明,为使得大量辅酶Q10从胶中进行有效释放,优选使用具有低含量辅酶Q10的大量的辅酶Q10包合物,而不是使用具有高含量辅酶Q10的辅酶Q10包合物。
试验实施例2:糖包衣胶的制备
制备了糖包衣胶,其各含有包含在糖衣料中的辅酶Q10的样品,评价了辅酶Q10从这些胶中向唾液中的释放。更具体而言,利用试验实施例1中所述的样品1、2、4或5或表4中所示组合物的样品6(作为含有辅酶Q10的样品)制备糖衣料。利用这些糖衣料制备糖包衣片胶。
(1)含有辅酶Q

的样品(样品6)的制备
根据表4中所示的配方制备含有辅酶Q10的样品(样品6)。更具体而言,通过加热将90份阿拉伯胶和150份糊精溶解在700份水中,并维持在70℃。通过加热将60份辅酶Q10熔融并加入到880份上述所得溶液中,在70℃搅拌1小时。利用均质器以400kg/cm2乳化所得的溶液,喷雾干燥得到290份粉末(样品6,辅酶Q10:阿拉伯胶:糊精=20:30:50(以重量计))。

(2)糖包衣胶的制备
以下述方法制备了糖包衣胶(1.5g/片),其均含有在糖衣料中的辅酶Q10,使得在100g所述糖包衣胶中辅酶Q10的含量为20mg。
(2-1)胶芯的制备
在70℃捏合胶基(商品名:胶基LT2,由Lotte,Co.,Ltd.制备)与木糖醇、玉米糖浆和甘油,加入柠檬油No.2451*(香料)之后,将混合物冷却至室温并进行制模成片剂形状。
胶芯的配方(wt%)
(2-2)糖衣料的制备
将麦芽糖醇和阿拉伯胶加入到水中并通过加热溶解。将混合物冷却至室温后,加入样品1、2、4、5或6中之一并均匀混合。
糖衣料的配方
利用包衣锅将由此得到的糖溶液各自喷雾干燥到胶芯上并干燥。糖包衣层的比例是50重量份/100重量份胶芯。

取上述制备的每种类型的糖包衣胶6g(精确量)(4块的总重量),置于具塞离心管中。进行根据试验实施例1中的相同步骤以制备用于HPLC分析的样品滤液。在与试验实施例1相同的条件下对每种样品进行HPLC以测定糖包衣胶中辅酶Q10的含量(糖包衣胶中CoQ10的含量)。表5中所示的结果证明每种类型的糖包衣胶中辅酶Q10的含量均为0.02wt%。
(3-2)释放到唾液中的辅酶Q
10
的分析
取上述制备的每种类型的糖包衣胶6g(精确量)(4块的总重量),然后由4名对象在与试验实施例1中相同的条件下咀嚼每种所述这些胶中的一块。将4名对象咀嚼过的胶分别置于具塞离心管中。其后重复与试验实施例1中相同的步骤,所得滤液用作HPLC分析的样品以检测被咀嚼过的胶中辅酶Q10的含量(被咀嚼过的胶中CoQ10的含量)。在相同条件下对样品进行HPLC以测定被咀嚼过的胶中辅酶Q10的含量,然后根据试验实施例1评价辅酶Q10从这些胶中向唾液中的释放。

结果显示,利用样品1和2制备的胶分别释放约100%和约95%的辅酶Q10,这比分别从利用样品4、5和6制备的胶中的释放显著高约50%、30%和50%。该结果表明,将辅酶Q10(尤其是低含量的辅酶Q10)包裹在环糊精中提高了辅酶Q10的溶出性能。
实施例1-3
利用试验实施例中制备的含有辅酶Q10的样品1-5,以下述方法制备糖包衣胶。
(1)糖包衣胶的制备
利用含有辅酶Q10的样品1,根据下述配方制备在糖衣料中含有辅酶Q10的糖包衣胶(1.5g/片),使得100g糖包衣胶中辅酶Q10的含量为20mg。
(1-1)胶芯的制备
在70℃捏合胶基(商品名:胶基LT2,由Lotte,Co.,Ltd.制备)与玉米糖浆和甘油,加入柠檬油No.2451*(香料)之后,将混合物冷却至室温并制模成片形。
胶芯的配方(wt%)
(1-2)糖衣料的制备
将麦芽糖醇和阿拉伯胶加入到水中并通过加热溶解。将混合物冷却至室温后,加入样品1、4或5中之一并均匀混合。
糖衣料的配方 (wt%)
利用包衣锅将由此得到的糖溶液各自喷雾干燥到胶芯上并干燥。糖衣料层的比例是50重量份/100重量份胶芯。

如表7所示,包含含有最低辅酶Q10含量的样品1的糖包衣胶表现出最高的辅酶Q10溶出性能。此外,除了将糖或糖醇加入到胶基LT2(由Lotte,Co.,Ltd.制备)中以外,以与实施例1、2或3中相同的方法制备样品,同样测量了这些样品中辅酶Q10的释放。结果表明,与上述实验一样,不管是否使用糖或糖醇,包含含有最低辅酶Q10含量的样品1的糖包衣胶都表现出最高的辅酶Q10溶出性能。
Claims (22)
1.一种包含辅酶Q10包合物的咀嚼组合物,其中辅酶Q10被包裹在环糊精中。
2.根据权利要求1的咀嚼组合物,其中所述辅酶Q10包合物包含1-40wt%的辅酶Q10。
3.根据权利要求1的咀嚼组合物,其中所述辅酶Q10的含量是1-80重量份/100重量份环糊精。
4.根据权利要求1的咀嚼组合物,其中所述辅酶Q10的含量是0.08-25wt%/100wt%咀嚼组合物。
5.根据权利要求1的咀嚼组合物,其中所述辅酶Q10包合物的含量是使得100wt%咀嚼组合物中辅酶Q10的比例是0.01-10wt%的含量。
6.根据权利要求1的咀嚼组合物,其中所述环糊精是γ-环糊精。
7.根据权利要求1的咀嚼组合物,其还包含胶基。
8.根据权利要求7的咀嚼组合物,其是口香糖。
9.根据权利要求1的咀嚼组合物,其中所述咀嚼组合物被含有辅酶Q10包合物的糖衣料包裹。
10.一种用于提高辅酶Q10溶出性能的方法,包括将辅酶Q10包裹在环糊精中。
11.一种用于提高含有辅酶Q10的咀嚼组合物中辅酶Q10溶出性能的方法,所述方法包括向所述组合物中加入辅酶Q10包合物,其中辅酶Q10被包裹在环糊精中作为辅酶Q10成分。
12.根据权利要求11的方法,其中所述辅酶Q10包合物包含1-40wt%的辅酶Q10。
13.根据权利要求11的方法,其中所述辅酶Q10包合物中辅酶Q10的含量是1-80重量份/100重量份环糊精。
14.根据权利要求11的方法,其中所述辅酶Q10包合物的含量是0.08-25wt%/100wt%咀嚼组合物。
15.根据权利要求11的方法,包括向所述组合物中加入下述量的所述辅酶Q10包合物,所述量使得辅酶Q10的含量是0.01-10wt%/100wt%咀嚼组合物。
16.根据权利要求11的方法,其中所述环糊精是γ-环糊精。
17.根据权利要求11的方法,其中所述咀嚼组合物还包含胶基。
18.根据权利要求17的方法,其中所述咀嚼组合物是口香糖。
19.根据权利要求11的方法,其中所述咀嚼组合物被含有辅酶Q10包合物的糖衣料包裹。
20.根据权利要求11的方法,其用于提高含有辅酶Q10的咀嚼组合物中辅酶Q10在口腔中的溶出性能。
21.辅酶Q10包合物在制备含有辅酶Q10的咀嚼组合物中的用途,其中辅酶Q10被包裹在环糊精中。
22.根据权利要求21的用途,其中所述咀嚼组合物是口香糖。
