Description

生产活性成分的固体分散体的方法
本发明涉及一种生产活性成分的固体分散体的方法,其包括将活性成分和基质形成剂供入挤出机并形成均匀的挤出物。
一段时间以来,生产包括固溶液产物在内的固体药物剂型的连续方法是已知的且包括通过注塑或挤出,随后成型而将含有活性成分的聚合物粘合剂熔体转化为所需的药物剂型(例如参见EP-A-240 904,EP-A-240 906和EP-A-337 256)。当活性成分具有低熔点和/或在熔融的聚合物粘合剂中的高溶解度时,在方法中获得令人满意的结果。具有低熔点的活性成分在接触聚合物粘合剂熔体时液化,且液化的活性成分可轻易地分散在聚合物粘合剂熔体中。或者,在熔融的聚合物粘合剂中具有高溶解度的活性成分轻易地溶解在聚合物粘合剂熔体中。
当活性成分具有高熔点和/或在熔融的聚合物粘合剂中的有限溶解时,出现了问题。活性成分的充分分散可能需要挤出机机筒的高温、较长的混合时间和/或高剪切,以产生活性成分与聚合物粘合剂熔体的充分混合。尤其是当使用对剪切和温度敏感的活性成分时,这可导致局部过热和对产品的损害。必需的挤出机机筒高温的另一缺点为高的能量成本。
此外,EP 0 580 860 B2描述了一种生产溶解在聚合物中的药物的固体分散体的方法,其中使用配有浆叶工具或捏合段的双螺杆挤出机。这类捏合段例如由以螺旋阶梯方式偏移放置的盘式凸轮组成。挤压物质通过盘形凸轮和挤出机外罩间窄的锥形间隙。因此在通过挤出机的过程中,使材料经受高的局部剪切力,这可导致活性成分和/或聚合物的过度降解。剪切还可引起挤出设备的过度磨损。
本发明的目的为提供一种生产活性成分在基质形成剂,尤其是在聚合物中的固体分散体的方法,其避免了高温或高的局部剪切力的必要性。
本发明的另一目的为提供一种生产活性成分在基质形成剂,尤其是在聚合物中的固体分散体的方法,其中使活性成分和/或基质形成剂和/或辅助物质的降解最小化。
本发明提供了一种生产活性成分的固体分散体的方法,其包括将活性成分和基质形成剂供入挤出机并形成均匀的挤出物。挤出机包括至少两个旋转轴,其中每个轴带有多个沿轴向一个在另一个之后放置的加工元件。加工元件限定如下区:(i)进料和输送区,(ii)至少一个混合区,以及(iii)排出区。进料和输送区位于最远的上游,接近挤出机的加料斗,所述至少一个混合区位于进料和输送区的下游,且排出区位于最远的下游,接近挤出机的排出开口。此处所用的术语“下游”指其中将材料在挤出机中输送的方向。
加工元件可分开形成。它们可一个在另一个之后成串排列在挤出机的轴上。然而,加工元件还可整体形成。在该情况下,元件的表面结构形成了所述加工元件。
根据本发明,限定混合区的加工元件包括源自螺杆型元件的混合元件。“源自螺杆型元件”的混合元件意欲指其基本形状为螺杆元件形状,但已经以使得其除了输送效果外,还显示配混或混合效果的方式而改变的元件。所述螺杆型元件为强制进料(或“右旋”)的螺杆元件。应理解的是通过本发明混合元件所显示的混合模式主要为分布的,而不是分散的混合。
目前为止,浆叶工具或捏合段通常用于捏合和塑化的药物混合物中。这些捏合段由在外周方向上角度互相偏移的凸轮盘组成。凸轮盘具有垂直于挤出机中总的输送方向的邻接面。而这些捏合段提供了有效的捏合和均化,在凸轮盘边缘处产生高的局部剪切。应理解的是,该局部剪切对活性成分或其它组分有害。
优选的混合元件不具有法线与总的输送方向平行且相对的平面表面区域。混合元件尤其可不具有垂直于总的输送方向的面。
本发明使用的混合元件不具有垂直于总的输送方向的邻接面。通常而言,本发明所用混合元件具有在螺杆型元件的螺纹中形成的凹口。这类混合元件是本身已知的且例如描述在WO 2004/009326 A1、US 5,318,358和US6,106,142中。
优选的混合元件具有多个通过将凹槽转变为螺杆型元件而形成的同心环部分。为此,混合元件具有连续的螺纹,其仅被用环部分转变的凹槽所间隔。
令人惊讶的是,已经发现,与使用浆叶工具或捏合段的常规方法相比,这些混合元件使得能够充分混合或均化且较少降解活性成分或形成其它成分。此外,可选择较低温度的挤出机机筒而仍获得同样质量的挤出物。额外惊人地发现,本发明混合元件提供了较好的自洁效果。该自洁效果防止挤出材料的残留物经延长时间而仍保留在挤出机内。
挤出机包括至少两个轴向平行的轴且在优选实施方案中为双螺杆挤出机。轴可为同步旋转的或反向旋转的,但优选为同步旋转的。挤出机可包括超过两个,例如至多六个轴。配置在相邻轴上的加工元件接近相互啮合。
进料和输送区以及排出区允许供入挤出机的材料从进料端至挤出机排出端的光滑通过。用于进料和输送区或排出区的加工元件通常呈蜗杆元件,即特征在于基本连续的螺纹元件的形式。
在优选实施方案中,加工元件额外包括至少一个背压元件。通常而言,背压元件位于混合区的下游。背压元件用于产生足够的背压以允许所需程度的混合和/或均化。设计背压元件以装载在挤出机中输送的材料。相对于输送元件,它们可源自具有降低的螺距的螺杆型元件。或者,它们可源自反向螺纹的螺杆,以使它们在与挤出机的总的输送方向相反的方向上输送材料。背压元件可与混合元件分开形成或与混合元件整体形成。
根据本发明的有利方面,加工元件限定如下区:
(i)进料和输送区,
(ii)位于进料和输送区下游的第一混合区,和
(iii)位于第一混合区下游的中间输送区,
(iv)位于中间输送区下游的第二混合区,和
(v)排出区。
优选,加工元件额外包括位于第二混合区下游且与之邻近的背压元件。
合适选择进料和输送区的长度,以使当供入挤出机的材料进入(第一)混合区时,材料经受显著软化或接近熔融。进料和输送区优选对应于轴全长的约20%至约40%。排出区优选对应于轴全长的约15%至约30%。
根据本发明的有利方面,使用双螺杆挤出机。其具有至少两个平行的同步旋转轴。在混合区中,轴配有互相啮合的混合元件。混合元件面受对应于混合元件的外螺杆直径、螺杆内径和至多中心距离的圆弧的限制。轴受平行于轴的挤出机外罩的圆形段引导。
有利的是,混合元件包括在首先引起迫使物质通过挤出机外罩间的环形间隙的蓄压的环部分和具有剪切作用和延伸的环部分之间的螺杆部分;然后压力又降低。混合元件上的剪切间隙通道的循环顺序、蓄压、剪切间隙通道等在物质上造成限定应力和因此均匀的应力,而尤其没有过度压迫活性成分。
混合元件的环部分间的螺杆部分可具有相同的螺距。然而,这些螺杆部分的螺距也可以是不同的。根据本发明的有利实施方案,在每个轴上的至少一个混合元件的螺杆部分具有部分正螺纹和部分反向螺纹。
环部分和挤出机外罩的凹的圆形段之间的环形和/或剪切间隙可具有不同高度,以产生活性成分在基质形成剂中的充分混合效果。为此,环部分可仅对应于螺杆轴心径。环形间隙还可具有10-90%的螺杆螺纹深度的高度。此外,环部分的直径可大约对应于两个相邻轴的中心距离。
在物质通过环形或剪切间隙的过程中压迫物质之前,必须通过螺杆部分将其运输一定输送距离,以建立所需压力。为此,位于两个邻近环部分之间的螺杆部分通常具有至少1/10,优选至少1/5的螺杆直径的长度。所转变的环部分的凹槽优选具有例如1/2或更少的螺纹深度的深度。转变的凹槽的侧面的角度例如可为30-90度。倾斜凹槽尤其优选以与轴心线成约60度的角度转变。
通过切削螺杆顶部和侧面,混合元件可配有其它部分。因此,具有显著中性输送作用的混合区尤其可通过切削提供。
在环形间隙之后,螺纹可持续在相同的螺距角(pitch angle)。即除了在环部分区域中的转变的间隔以外,混合元件的螺杆部分可形成连续螺纹。
环部分允许获得额外的分散表面。此外,如果将环部分间的螺杆部分以相同的旋转方向逐渐相互角度偏移地,例如以螺纹角度的一半角度偏移地放置,则可获得分散表面的显著增大。角度偏移螺杆部分形成以逐步方式角度偏移的面作为额外的分散表面。
根据本发明的一个实施方案,用在双螺杆挤出机的轴上的混合元件描述在作为参考在此引入的WO 2004/009326 A1中。WO 2004/009326 A1的图2和5显示了本发明所用的优选混合元件。在下文中参考附图描述了其它实例。
本发明方法制备的固体分散体含有一种或多种活性成分和任选添加剂。添加剂可用于对固体分散体赋予所需性能或便于其制备。尽管可在方法的任何合适段内将活性成分和添加剂掺入挤出混合物中,还可优选将部分或全部活性成分或添加剂与基质形成剂和/或其它组分分开地引入挤出机中。
为此,在本发明方法的实施方案中,将至少部分基质形成剂供入挤出机的加料斗且通过位于混合区上游或混合区中的挤出机机筒的开口将至少一种选自如下的组分引入挤出机中:
(i)基质形成剂的剩余物,
(ii)活性成分,
(iii)添加剂,和
(iv)其组合。
优选在位于或接近进料和输送区和混合区的接点的位置将所述至少一种组分引入挤出机中。组分可为固体,例如粉状的,但优选为液体或液化的。
所述至少一种组分最优选包括可药用的表面活性剂。
将供入挤出机的物质熔融以均化熔体并将活性成分有效地分散或溶解在聚合物中。“熔融”指转变为其中一种组分可均匀地嵌入另一种组分的液态或橡胶态。熔融通常包括加热至聚合物的软化点以上。通常而言,最大熔融温度为70-250℃,优选80-180℃,最优选100-140℃。
加热挤出机外罩,以由供入挤出机的物质形成熔体。应理解的是工作温度还要通过挤出机种类或所用挤出机内的构造的种类决定。在挤出机中熔融、混合和溶解组分的部分能量可由加热元件提供,而挤出机中材料的摩擦和剪切还可为混合物提供显著量的能量并有助于形成组分的均匀熔体。
为获得活性成分的均匀分布和充分分散,将含有活性成分的熔体保存在熔体挤出机的加热机筒内足够长时间。
根据本发明的另一方面,挤出机机筒包括数个加热区。优选将第一混合元件上游的机筒部分维持在比第一混合元件下游的机筒部分低的温度。已经发现该温度分布导致均匀、光滑和透明的挤出物,该挤出物尤其受对活性成分太高的温度所损害。
在根据本发明生产的挤出物中,将一种或多种活性成分均匀分散在整个聚合物中。这包括在聚合物相中具有通常直径小于1μm的活性成分小颗粒的体系。这些体系不含有任何显著量的呈其晶体或微晶态的活性成分,这通过热分析法(DSC)或X-射线衍射分析(WAXS)证明。通常而言,至少98重量%活性成分总量以无定形态存在。
当挤出物在化学或物理上均一或整体均匀或由一个相组成(如热力学所定义)时,分散体被称为“固溶液”。活性成分的固溶液优选为物理体系。
聚合物不含有显著量的挥发性溶剂。术语“挥发性溶剂”意欲包括水和任何在环境温度下为液体且挥发性比水高的化合物。通常而言,基质含有小于25重量%,优选小于6重量%,最优选小于3重量%挥发性溶剂。
优选的由本发明方法形成的挤出物包含如下物质:
约8-99.9重量%(优选40-85重量%,最优选50-70重量%)基质形成剂(或这类基质形成剂的任何组合),
约0.1-49重量%(优选1-30重量%)活性成分或活性成分的组合,
0-25重量%(优选2-15重量%)至少一种可药用的表面活性剂,和
0-25重量%(优选0-15重量%)添加剂。
基质形成剂可为任何能够从液态如熔融态沉降或凝胶以形成连续基质的试剂。当然可使用基质形成剂的混合物。
有用的基质形成剂选自多元醇(即糖醇、糖醇衍生物或麦芽糖糊精)、蜡和类脂。
合适的糖醇包括甘露糖醇、山梨糖醇、木糖醇;糖醇衍生物包括异麦芽糖或氢化的浓帕拉金糖(palatinose)(如DE-A 10262005中所述);其它的基质形成剂为麦芽糖糊精。
基质形成剂优选包括可药用的聚合物或可药用聚合物的混合物。通常而言,可药用聚合物为水溶性的或至少为水分散性的。
通常而言,本发明中所用的可药用聚合物的Tg为至少约+10℃,优选至少约+25℃,最优选约40℃至180℃。“Tg”指玻璃化转变温度。测定有机聚合物Tg值的方法描述在John Wiley & Sons,Inc.,在1992年出版的L.H.Sperling的"Introduction to Physical Polymer Science"第二版中。Tg值可以计算为源自组成聚合物的每个单独的单体i的均聚物的Tg值的加权和,即Tg=∑Wi Xi,其中W为有机聚合物中单体i的重量百分数,X为源自单体i的均聚物的Tg值。均聚物的Tg值在J.Brandrup和E.H.Immergut编辑,John Wiley & Sons,Inc.在1975年出版的"Polymer Handbook"第二版中说明。
具有上述Tg的可药用聚合物允许制备机械稳定且在正常温度范围内对温度足够稳定的固体分散体,以使所述固体分散体可用作不需进一步加工的剂型或可仅用少量压片助剂压片。剂型例如为片、胶囊、植入物、薄膜、泡沫、栓剂。

熔体中可包含各种添加剂,例如流动调节剂如胶态二氧化硅;润滑剂、填料、崩解剂或增塑剂、稳定剂或防腐剂。
可使用各种其它添加剂,例如染料如偶氮染料,有机或无机颜料如铁氧化物或二氧化钛,或天然源的染料;稳定剂如抗氧化剂、光稳定剂、自由基捕捉剂和抗微生物侵袭的稳定剂。
可在方法的任何合适阶段将这些添加剂掺入活性成分和聚合物的混合物中。然而,为了方便处理,方便的是使这类添加剂包含在供入挤出机的基质形成剂和活性成分的粉状混合物中。
从挤出机排出的挤出物从浆糊状到粘性。在使挤出物固化之前,可使挤出物直接成型为实际上所需的任何形状。方便的是,挤出物成型可通过具有两个反向旋转的滚筒的压延机进行,其中滚筒表面上具有相互匹配的凹陷(depression)。宽范围的片剂可通过使用具有不同形式凹陷的滚筒获得。如果滚筒表面上不具有凹陷,则可获得薄膜。或者,通过注塑将挤出物模塑成所需形状。或者,在固化之前(热切)或之后(冷切)使挤出物经受型材挤出并切成片。
另外,如果挤出物含有推进剂,例如气体如二氧化碳,或挥发性化合物如低分子量的烃,或可热降解为气体的化合物,则可形成泡沫。使推进剂在较高的压力条件下溶解在挤出机内的挤出物中,且当挤出物从挤出机模具中出现时,压力突然释放。因此,推进剂的溶解能力降低和/或推进剂汽化,从而形成泡沫。
任选将所得固体分散体产物研磨成颗粒。然后可压实颗粒。压实指其中使包含颗粒的粉状物质在高压下凝聚,以获得孔隙度低的压块如片的方法。压缩粉状物质通常在压片机中,更具体而言在两个移动冲头之间的钢模中完成。
优选将至少一种选自流动调节剂、崩解剂、填充剂(填料)和润滑剂的添加剂用于压实颗粒。崩解剂促进压块在胃中的快速崩解并使释放的颗粒相互分离。合适的崩解剂为交联聚合物如交联的聚乙烯基吡咯烷酮和交联的羧甲基纤维素钠。合适的填充剂(也称作“填料”)选自乳糖、磷酸氢钙、微晶纤维素

润滑剂优选用于压实颗粒。合适的润滑剂选自聚乙二醇(例如具有1000-6000的Mw)、硬脂酸镁和硬脂酸钙、富马酸硬脂酰钠等。

由于显示在图1和2中的挤出机通常是相似的,挤出机的通常配置参考图2描述。
挤出机是本身已知的。已经将其用于生产活性成分在基质形成剂中的固体分散体。挤出机包括在纵向上被分为数个区的外罩或机筒1。在挤出机的上游侧,配置开口8用于供入活性成分和基质形成剂的粉末P。通常而言,将加料斗放置在该开口上,以使粉末P可容易地供入挤出机的机筒1。在挤出机的输送方向X,即开口8的下游,配有用于定量给料另一组分L如表面活性剂的另一开口9。这里将表面活性剂以液体或液化形式泵入或以固体形式定量给料至机筒1的内部。甚至在另一下游,配置另一开口10以将气体G从机筒1的内部吸至机筒1的外部。机筒1在输送方向X上以模具为结束,其中取出分散体。
此外,将挤出机的机筒1分成三个加热区H1、H2和H3。可控制机筒在这些加热区H1、H2和H3中的温度,以控制活性成分和基质形成剂的分散体的熔融。
在挤出机的机筒1中,排列了两个平行轴2,其中一个显示在图1和2的剖视图中。轴2优选反向旋转。轴2配有在轴向上一个在另一个之后放置的加工元件。将加工元件排列在挤出机机筒1内,以使加工元件的径向最外围部分邻近机筒1的内壁。在加工元件的最外围部分和机筒1的内壁之间仅形成非常小的间隙。由于图1和2仅为显示挤出机在纵向上的不同区的示意图,具有加工元件的轴2和挤出机机筒1以相互分开显示。
将具有加工元件的轴2分为数个区。上游最远处的区为进料和输送区A。该区A的上游侧邻近用于将粉末P供入机筒1的开口8。在区A的下游侧,配置机筒1的开口9以将表面活性剂供入机筒1的内部。进料和输送区A的加工元件通过螺杆型元件3形成,其形成了具有进料方向X和均匀的螺距的蜗杆。因此,在区A中,将粉末P供入挤出机1并使其沿下游方向X输送。控制挤出机1的加热区H1和H2,以使机筒1内的物质在进料和输送区A的末端开始熔融。
在区A下游配置混合区B。已经发现,混合区B中加工元件的选择是随后挤出物质量的主要因素。这里,显示常规配置的图1的挤出机不同于用于本发明方法的图2的挤出机。
如图1示意图所示,在常规挤出机中,轴2配有所谓的浆叶工具或由盘式凸轮组成的捏合段4。
如图2示意图所示,在本发明实施方案中所用的挤出机中,轴2配有特定的混合元件11,在下文中参考图3-5更详细地描述所述混合元件。
在混合区B的下游侧,形成中间输送区C。中间区C的加工元件为用于进料和输送区A的相同螺杆型元件3。因此,中间输送区C仅将来自混合区B的熔体输送至下一区。
在中间输送区C的下游,配置第二混合区D。显示在图1中的用于常规挤出机的该第二混合区D的加工元件又不同于显示在图2中的用于本发明挤出机的加工元件。中间输送区C和第二混合区D是任选的。
如图1示意图所示,在常规挤出机中,轴2配有浆叶工具或捏合段5和6。在捏合段6的下游侧,放置背压元件7。背压元件7用于产生足够的背压,以允许所需程度的混合和/或均化。其将材料积聚入混合区B和D。背压元件7源自具有反向螺距的螺杆型元件,这使得其以与挤出机的总的输送方向X相反的方向输送熔体。
应提及的是,用于图1显示的常规挤出机的背压元件7对应于用于图2显示的本发明挤出机的背压元件14。然而,对图1显示的挤出机的配置而言,使用这种背压元件7本身并不已知。背压元件7已经用于常规挤出机中,因此其中使用常规挤出机的方法的结果与其中使用本发明挤出机的方法的结果相当。
如图2示意图所示,在本发明挤出机的第二混合区D中,轴配有特定的混合元件12、13,在下文中参考图3-5对其又更详细地描述。混合元件12、13可与第一混合区B的混合元件11相同。然而,在图2所示的实施方案中,将混合元件分为部分12和13,其中部分12具有强制进料方向且部分13具有负进料方向或反向螺纹。
在混合元件12、13的下游,配置背压元件14,其对应于上述背压元件7。
应注意的是,捏合段4的长度对应于混合元件11的长度,捏合段5、6的长度对应于混合元件12、13的长度。
在第二混合区D的下游,配置排出区E。本发明挤出机的轴2以及常规挤出机的轴2配有螺杆型元件3,其与A和C区所用元件相同。在排出区E中,仅将熔体供给挤出机的模具。
在实践中通过开口8将聚合物和基质形成剂供至机筒1的内部。通过螺杆元件3将基质形成剂和活性成分输送至混合元件11。将加热区H1和H2加热至使得聚合物和基质形成剂恰好在混合元件11前开始熔融的温度。这里同样通过开口9将表面活性剂供至机筒1的内部。然后使熔体通过混合元件11并经由中间输送区C的螺杆元件3输送至包括混合元件12、13和随后的背压元件14的第二混合区D。这里,完成了主要混合和熔融作用。随后,通过排出区E的螺杆元件3将均匀的挤出物输送至挤出机的模具。
在下文中,参考图3-5描述可用于混合区B和D的混合元件的实例。
通常而言,显示在图3-5中且可用作两个轴2上的混合元件11-13的混合元件15、20和24具有由三个圆弧组成的横断面23。一个圆弧具有对应于外螺杆直径的直径,另一个圆弧具有对应于螺杆芯直径的直径,又一个圆弧具有其半径对应于两个混合元件中心距离的直径(参见EP-B-0002 131)。
此外,混合元件15、20和24包括钻孔22,所述钻孔具有用于与轴2的凹槽啮合的凸起,因此混合元件15、20和24可与轴2一起旋转。
从图3A和3B可看出,混合元件15具有与轴心线同心且相互间隔一定距离放置的5个环部分16。环部分16通过转变为混合元件15的凹槽获得。凹槽的侧面18与轴心线的角度为约60度。环部分16与挤出机机筒1的内壁之间的环形间隙19的高度约为螺纹深度,即螺杆芯直径和外径差。因此,环部分8的直径对应于螺杆芯直径。
在混合元件15中可形成连续的螺纹,其仅被用环部分16转变的凹槽所间隔。与此相反,环部分16间的混合元件15的螺杆部分还可以在相同旋转方向上逐步相互角度偏移地放置。
在图3A和3B中显示的实施方案中,混合元件15的环部分16间的螺杆区17a、17b、17c、17d具有相同的螺距。在图3A和3B中显示的混合元件15尤其可用作图2所示混合区B中的混合元件11。
混合元件20的另一实施例显示在图4A和4B中。混合元件20不同于环部分16间的螺杆区21a、21b、21c、21d中的混合元件15。螺杆区21a和21b可对应于混合元件15的17a和17b。然而,混合元件20的螺杆区21c和21d不同于混合元件15的螺杆区17c和17d。即,螺杆区21c和21d具有反向螺纹,因此这些区21c和21d以与挤出机的总的输送方向X和螺杆区21a和21b的输送方向相反的方向输送熔体。
如图4A和4B所示,螺杆区21a和21b可与螺杆区21c和21d整体形成。然而,还可配有两个混合元件,其中一个包括螺杆区21a和21b,另一个包括螺杆区21c和21d。混合元件20可对应于图2所示的第二混合区D的混合元件12、13。
混合元件24的另一实施例显示在图5A和5B中。至于螺杆区26a、26b、26c和26d,混合元件24类似于图4A和4B所示的混合元件20。螺杆区26a和26b具有正螺纹,螺杆区26c和26d具有负螺纹或反向螺纹。
此外,混合元件24不同于环部分25和挤出机机筒1之间的环形间隙27中的混合元件20和15。在混合元件24的实施例中,环形间隙27的高度约为螺纹深度的一半,即螺杆芯直径和外径差的一半。因此,环部分8的直径约对应于两个轴相互间的中心距离。与混合元件20和15的环部分16的直径相比,环部分25的较大直径为熔体提供了阻挡层。发现如果混合元件24用作图2所示的第二混合区D中的混合元件12、13,则这种阻挡层是有利的。该阻挡层在挤出机内提供了压实区,其中提高了挤出物对物质供应侧的压力。
下文提供了其中活性成分在聚合物中的相同固体分散体已经通过如下设备生产的实施例:首先是具有图1所示螺杆配置的挤出机,作为对比例;其次,具有图2所示螺杆配置的挤出机。

使活性成分、聚合物和助流剂完全混合并将所得粉末供入双螺杆挤出机(ZSK-40,德国Werner & Pfleiderer制造)。除了输送元件之外,螺杆构造还包括捏合段且显示在图1中。借助液体计量泵将乳化剂供入挤出机中。在紧邻挤出机中材料到达第一捏合段区之前的位置加入乳化剂。在挤出过程中,使液体乳化剂与粉末共混并熔融混合物。对挤出机最后三分之一段中的混合物施用真空。工艺参数详细列在表2中。在挤出步骤之后,在压延机上成形并冷却材料,显示扁豆型挤出物带。

从表3和表5的结果来看,显而易见的是,在带有捏合段的螺杆的挤出过程中的降解比含有混合元件的螺杆运行过程中的降解更明显。
与配制剂1相比,所观察到的配制剂2的较高降解可归因于工艺参数。为使较高乳化剂量均匀混合入粉末共混物中,螺杆速率和挤出温度需要增加(表2和4)。较高的能量输入不仅导致所需的均匀挤出物,而且导致降解增加。因为螺杆速率的增加通常伴随挤出物中空气的夹带,对配制剂2增加真空度。增加的真空度反过来增加了能量输入,从而有助于增强混合。另一后果为较低的产物水含量。

Claims (26)

1.一种生产活性成分的固体分散体的方法,其包括将活性成分和基质形成剂供入挤出机并形成均匀的挤出物,其中所述挤出机包括至少两个旋转轴(2),其中每个轴(2)带有多个沿轴向一个在另一个之后放置的加工元件,加工元件限定如下区:
(i)进料和输送区(A),
(ii)至少一个混合区(B),和
(iii)排出区(E),
其中限定混合区(B)的加工元件包括源自螺杆型元件的混合元件(11、12、13)。
2.根据权利要求1的方法,其中所述混合元件(11、12、13)具有在螺杆型元件的螺纹中形成的凹口。
3.根据权利要求1或2的方法,其中所述混合元件不具有法线与总的输送方向平行且相对的平面表面区域。
4.根据前述权利要求中任一项的方法,其中所述混合元件(11、12、13)不具有垂直于总的输送方向的面。
5.根据前述权利要求中任一项的方法,其中所述混合元件(11、12、13)不具有垂直于总的输送方向的邻接面。
6.根据前述权利要求中任一项的方法,其中所述混合元件(11、12、13)具有多个通过将凹槽转变为螺杆型元件而形成的同心环部分(16;25)。
7.根据前述权利要求中任一项的方法,其中所述加工元件额外包括至少一个背压元件(14)。
8.根据权利要求7的方法,其中所述背压元件(14)位于混合区(D)的下游。
9.根据前述权利要求中任一项的方法,其中所述轴(2)为同步旋转的。
10.根据权利要求1-8中任一项的方法,其中所述轴(2)为反向旋转的。
11.根据前述权利要求中任一项的方法,其中所述加工元件限定如下区:
(i)进料和输送区(A),
(ii)第一混合区(B),
(iii)中间输送区(C),
(iv)第二混合区(D),和
(v)排出区(E)。
12.根据权利要求11的方法,其中所述加工元件额外包括位于下游且邻近第二混合区(D)的背压元件(14)。
13.根据权利要求1的方法,其中将至少部分基质形成剂供入挤出机的加料斗且通过位于混合区(B)上游或混合区(B)中的挤出机机筒(1)的开口(8)将至少一种选自如下的组分引入挤出机中:
(i)基质形成剂的剩余物,
(ii)活性成分,
(iii)添加剂,和
(iv)其组合。
14.根据权利要求13的方法,其中在位于或接近进料和输送区和混合区(B)的接点的位置将所述至少一种组分引入挤出机中。
15.根据权利要求13或14的方法,其中所述至少一种组分为液体或液化的。
16.根据权利要求13-15中任一项的方法,其中所述至少一种组分包括可药用的表面活性剂。
17.根据前述权利要求中任一项的方法,其中将所述活性成分以固溶液态分散在聚合物中。
18.根据前述权利要求中任一项的方法,其中所述基质形成剂包括可药用的聚合物。
19.根据权利要求18的方法,其中所述可药用的聚合物选自如下物质:
N-乙烯基内酰胺的均聚物,
N-乙烯基内酰胺和一种或多种选自含氮单体和含氧单体的共聚单体的共聚物,
纤维素酯和纤维素醚,
高分子量的聚氧化烯,
聚丙烯酸酯和聚甲基丙烯酸酯,
低聚糖和多糖,
聚羟基酸或其混合物。
20.根据前述权利要求中任一项的方法,其中所述基质形成剂包含选自多元醇、蜡和类脂的成员。
21.根据前述权利要求中任一项的方法,其中额外包括将选自如下的至少一种添加剂供入挤出机:流动调节剂;润滑剂、填料、崩解剂、增塑剂、稳定剂或防腐剂。
