Abstract

Description

转化生物质的方法和设备
本发明涉及将生物质,特别是将含木质纤维素的生物质转化为可发酵糖的方法。本发明还涉及适合实施该方法的设备。
将来,可再生材料将起到越来越重要的作用。生物转化方法将是由可再生起始材料(例如生物质)生产有价值产品(例如乙醇)的重要方法。碳水化合物从木质纤维素(生物质的主要成分)中的释放步骤构成了生物质转化过程中的瓶颈。为了使生物催化剂(例如微生物)进入起始材料,有必要将木质纤维素解聚和/或解晶化。
为此目的的公知技术是强酸处理,其见述于例如:US-A-5562777、US-A-5580389、US-A-5597714、US-A-5620877、US-A-5726046、US-A-5782982、US-A-5820687、US-A-6054611和US-A-6239198。
国际申请WO-A-94/23071描述了由含纤维素和半纤维素的材料生产糖的方法。该方法包括用酸将纤维素和半纤维素解晶化并水解,再将水解产物分为糖和酸。
US-A-4427584描述了逐步的方法:用液体或气体三氧化硫将晶型α-纤维素转化为无定形α-纤维素以引起α-纤维素的解晶作用,然后水解经处理的纤维以产生糖。
将纤维素材料转化为可发酵产物的公知技术通常需要机械搅拌装置,以获得纤维素材料与强酸的充分混合。实际上这常常导致复杂化,因为运动机件与强酸的结合会引起腐蚀问题。纤维素材料通常也需要机械预处理以获得所需的颗粒大小(通常直径为10毫米或者更小)和含水量(通常小于10%)。
另外,为了克服使强酸与纤维素材料良好接触方面的困难,公知技术需要将纤维素材料剁碎或者碾碎成不大于10目的颗粒尺寸。使纤维素材料悬浮在热空气流中也需要这种小的颗粒尺寸。
另外,独立的蒸发器是必需的,以产生浓酸。这导致了额外投资费用。
另外,所述公知技术需要相当大量的酸,这些酸随后要使用适量的碱中和。这导致了相当大量的废物流,例如石膏,这需要被处理掉,因而引起花费。
本发明希望克服与现有技术相关的这些问题,以及下文中将显现出来的其它问题。
发现用以下方法可以达到这个目的,在该方法中使生物质填充床或生物质堆接触到酸,并接触作为连续相的气体。在该方法中,生物质被分裂成10毫米或更大的颗粒尺寸。优选地,使生物质顺流与酸接触而逆流与气体接触。可以将生物质与大比表面积的惰性材料(例如塑料鲍尔环)混合,以保证所需的填充生物质的结构长度,并确保气体和液体的良好分布。因而,一方面,本发明涉及将生物质转化为可发酵糖的方法,该方法包括使所述生物质在反应器中与酸接触,同时使气流通过所述反应器,其中所述气体是能吸收(take up)水,从而有效改变反应器中pH值的惰性气体。
使气流通过反应器,可以改变酸溶液中水的蒸发,并且可以有效控制酸的浓度以及pH和含湿量。另外,气流有助于将酸均匀分布在整个反应器中,而不需要机械搅拌装置。所用的酸可以是任何本领域公知的适于此目的的强酸,例如盐酸、磷酸、氢氟酸和硫酸。最优选的是硫酸,因为可以用生物方法将其除去。此外,可以将(气体)三氧化硫加入反应器中存在的水相中,以此形式使用硫酸。
在将生物质装入反应器之后加入酸,例如通过从反应器顶部将其喷雾加入。任选或者作为可选方式,加入SO3。如果需要,可以加入水以获得有理想酸浓度的水相。随后加入气体,优选从反应器底部加入。反应器中所用的酸可以再循环回来进入反应器,这依赖于生物质的解聚程度。气流也被循环。可以调整气体流速以使溶液的pH(可以例如用pH电极装置经常监控)保持在期望水平上。酸的浓度优选被控制在70至75%重量,基于所述反应器中存在的酸(用kg H2SO4/kg无水纤维素材料表示)和水的重量计算。
以此方式,可以在差不多不变的且高的酸浓度下进行解聚。其结果是,木质纤维素转化为粘浆向下流到反应器底部,在这里可被收集。在足够长的时间后,可将包含解聚的纤维素和半纤维素的产物从反应器中移出,并进行后续处理步骤,任选在为除去粗的未转化部分的过筛步骤之后进行后续处理步骤。解聚步骤通常以分批过程进行。也可以半连续方式进行:将生物质加入反应器,同时,气体和酸被循环,直到未转化物质的量是将其从反应器中移走所需的量。
本发明方法的又一个好处是,未转化的物质(其常常由生物质进料中的污染物,例如沙子或者塑料碎片所形成)残留在反应器中,并且在反应完成后可以相对容易地从反应器中移走。
本发明方法将耗酸量减到了最少。此外,甚至可以有效地转化大块生物质(例如大块木头)。因为可以通过本发明方法相对容易地控制酸的浓度和含湿量,所以可以使用的不同类型的进料的含水量可以变化很大,这增加了本方法的通用性。
用来通过反应器的气体原则上可以是任何能吸收足量水的气体。该气体对于酸处理过程是惰性的,也就是说,该气体不妨碍酸处理过程。优选地,这种气体包含CO2(通常大于90%体积,例如大于99%体积),因为可以由随后的发酵步骤获得CO2,在发酵步骤中,经酸处理转化的生物质被转化为例如乙醇。CO2的另一个好处是,它有助于抑制不需要的产物的形成,特别是抑制氧化产物的形成。其它适合的气体包括氮气(N2)和空气。因此,气流优选包括选自以下的组分:CO2、氮气、空气及其混合物。由于反应器中pH低,空气相对于反应器内容物差不多也是惰性的。
出于方法经济的考虑,优选使气流循环。这包括在气流重新加回反应器之前把气流中的水除去。可以使用公知的技术除去水,例如冷却至足够低的温度。发现在ca.0至4℃的温度下气体足够干燥。在约2℃的温度下获得良好的结果。
反应器中所用的酸优选硫酸。可以将浓H2SO4(例如90%重量或者更浓)加入反应器,但是也可通过将三氧化硫气体加入反应器中,在原处形成硫酸,所述三氧化硫气体在溶解于水中时生成硫酸:SO3+H2O→H2SO4。可以使用氧气或空气燃烧硫化合物,由不同方法步骤获得三氧化硫。优选地,这种硫化合物是硫化氢,它可以由硫酸盐还原步骤获得,该硫酸盐还原步骤可以用来除去产物中的硫化合物。
在典型实施方式中,保持通过床的气体速度相对低(例如低于0.5米/秒),以使经过床的压力下降有限(例如限于约50mm H2O)。
反应器中的压力可以为常压或者稍高。
反应器中的温度优选60至100℃,更优选75至85℃,因为这已经提供了适合的反应速度,并同时避免了水的过分蒸发。
酸的量是低的。优选存在的酸量少于2kg酸/kg干物质,更优选1.2至1.4kg H2SO4/kg干物质。用强酸处理生物质的现有技术通常使用的酸量远远多于2kg酸/kg干物质。
本发明方法中酸的浓度优选控制在70至75%重量,基于所述反应器中酸(用kg H2SO4/kg干物质表示)和水的重量计算。
当反应器内容物的停留时间设为10至14小时,优选约12小时,可获得良好的化学转化。
实施本发明方法的设备通常包括非搅拌的分批反应器容器,其可以方便地由混凝土(廉价且结实)构成,其内部具有耐酸衬里,例如TeflonTM衬里或者PVC衬里。该反应器还具有气体入口和气体出口装置,以及加酸装置,特别是一个或多个分布酸溶液的喷嘴。该设备还可以包括在气体进入流进入反应器之前将其加热的加热器。在操作过程中,木质纤维素解聚并形成浆体。该浆体向下移并在反应器底部被收集。实施本发明的设备还可以包括将浆体从反应器中泵出的泵。其还可以包括一个或多个筛网以除去产物流中粗的部分。
从本发明方法获得的产物可以进一步在后续步骤中处理。通常这包括加入水,其作用是将解聚的纤维素和半纤维素水解成低聚糖或者单糖。在该步骤中,可以使木质素沉淀并在过滤步骤中滤除掉。存在于溶液中的碳酸盐和重碳酸盐转化为CO2,该CO2可在进行该步骤的反应器顶部收集。该CO2流可以用来通过将生物质水解成低聚糖或者单糖的反应器。

Claims (18)
