EMI1.2
stands for a cyclically structured polypeptide residue of the type of peptide residues contained in the ergot peptide alkaloids.
According to the invention, the compounds of the formula I are obtained by allowing a carboxylic acid anhydride to act in the presence of a sufficiently strong tertiary organic base according to a modified Polonovski reaction on compounds of the formula II, in which R1, R2, R and X have the above meanings.
The 6-N-oxides of the formula II can be obtained, for example, by oxidation of compounds of the formula III, in which R1, R2, R and X have the above meanings, with organic peracids in a solvent which is inert under the reaction conditions.
The two-stage conversion starting from compounds of the formula III to give the desired compounds of the formula I is advantageously carried out in a one-pot reaction without isolating the 6-N-oxides. The reaction can be carried out at temperatures between -30 "and room temperature; the reaction time is 30 minutes to 3 hours, depending on the ratio of the reaction components.
As an organic peracid, for example, m-chloroperbenzoic acid can be used; Suitable organic solvents which are inert under the reaction conditions are, for example, chlorinated hydrocarbons, such as, preferably, methylene chloride or chloroform. For the conversion of the 6-N-oxide into compounds of the formula I, acetic anhydride is preferably used in a proportion of 1 to 5 equivalents; propionic anhydride or benzoic anhydride can also be used. Triethylamine or diaza bicydo [2,2,2octane, which are added in an excess of 2 to 10 equivalents; pyridine can also be used as the tertiary organic base.
After the reaction has ended, the compounds of the formula I can be isolated from the reaction mixture in a known manner, for example by extraction and subsequent chromatography.
The introduction of the various substituents in the ring structure of 9,10-dihydrolysergic acid derivatives of the formula IIIa, where X has the above meaning, which is necessary for the preparation of the starting materials of the formula III used, can be carried out, for example, as follows: a) Position 1
Compounds of the formula IIIb in which X has the above meaning and R1 stands for lower alkyl can be obtained by 1-alkylation of compounds of the formula IIIa in liquid ammonia with the addition of 1 equivalent of potassium amide with the corresponding alkyl iodide or bromide.
b) Position 2
The selective introduction of a halogen atom in position 2 of compounds of the formula IIla or IIIb can take place with N, 2,6-trichloro-4-nitro-acetanilide or with N-bromo-succinimide in a dioxane solution. In this way, uniform 2-chloro or 2-bromo derivatives of the formula IIIc, in which Rl and X have the above meanings and Hal stands for chlorine or bromine, are obtained.
The direct methylation in position 2 succeeds, for example, with 2-methoxy-1,3-dithiolane (formula IVa) or with cyclic orthothioesters such as 1,2-bis- (1,3-dithiolan-2-ylthio) ethane (formula IVb ) via a Friedel-Crafts reaction. The reaction is carried out in the presence of titanium tetrachloride in chloroform at about 0 ". The compounds of the formula V, in which Rl and X have the above meanings, are subsequently converted into the compounds of the formula IIId in which Rt and X have the above meaning, desulphurized.
c) Position 2, 13 or 13
The introduction of 2 bromine atoms in positions 2 and 13 can be carried out with a brominating agent which contains elemental bromine, preferably with pyridine hydrobromide perbromide in an organic solvent such as acetic acid or propionic acid in a temperature range of 10 to 80. From the compounds of formula IIIe obtained, in which R1 and X have the above meaning, the bromine atom in position 2 can be removed reductively, for example with zinc dust / hydrochloric acid at elevated temperature in glacial acetic acid, resulting in the compounds of formula IIif in which R1 and X have the above meaning.
The compounds of the formula I prepared according to the invention are used as medicaments because of their interesting properties which have been determined in pharmacological testing. But they also serve as intermediate products in the production of therapeutically valuable compounds.
If the preparation of the starting compounds is not described, they are known or can be prepared by processes known per se or analogously to those described here or analogously to processes known per se.
In the following examples, which explain the invention in greater detail but are not intended to limit its scope in any way, all temperatures are given in degrees Celsius.
For the sake of simplicity, the basic compound represented by formula VI is referred to here as A9b10-ergopeptin.
