Die Erfindung betrifft ein Verfahren zur Herstellung von neuen, substituierten Benzo-1,3-oxazin- - dionen- (2, 4) der Formel I :

Die Umsetzung mit einem Phenylisocyanat der Formel III bei erhöhten Temperaturen wird vorzugs- weise zwischen 50 und 1000C vorgenommen ; es werden vorzugsweise äquimolare Mengen der Ausgangs- stoffe eingesetzt. Als inerte Lösungsmittel werden vorteilhaft Benzol, Toluol, Chloroform, Äthylenchlorid,
Dioxan usw. benutzt.
Die Umsetzung mit einem Phenylcarbaminsäurechlorid der Formel IV in Gegenwart eines halogen- wasserstoffbindenden Mittels wird zweckmässig bei Zimmertemperatur oder unter Kühlung durchgeführt.
Als halogenwasserstoffbindende Mittel können tertiäre organische Basen, wie Triäthylamin oder Pyridin oder auch anorganische Basen, wie Alkalihydroxyde, Alkalicarbonate usw., verwendet werden ; falls tertiäre organische Basen verwendet werden, können diese auch als Lösungsmittel dienen.
Der Ringschluss der aromatischen o"O-Phenylcarbamoyl-carbonsäure der Formel V zu einer Verbindung der Formel I, der durch Erhitzen auf Temperaturen zwischen 100 und 2000C bewirkt wird, kann z. B. unter Verwendung von Tetralin oder Dekalin als hochsiedendem Lösungsmittel erfolgen. Beim Erhitzen mit wasserabspaltenden Mitteln, vorzugsweise auf Temperaturen zwischen 80 und 100 C, können beispielsweise Thionylchlorid oder Phosphorhalogenide eingesetzt werden. Die letztgenannte Umsetzung wird gegebenenfalls in Anwesenheit eines Lösungsmittels und in einem Überschuss des wasserabspaltenden Mittels durchgeführt.
Die Aufarbeitung des Reaktionsproduktes erfolgt dann in üblicher Weise. Nach Abdestillieren des hochsiedenden Lösungsmittels bzw. des Überschusses des wasserabspaltenden Mittels im Vakuum wird die zurückbleibende Substanz durch Umkristallisieren gereinigt.
Die als Ausgangsstoffe verwendeten Phenylisocyanate der Formel III und die Phenylcarbaminsäurechloride der Formel IV können, soweit sie nicht bekannt sind, aus den entsprechenden Aminen in bekannter Weise hergestellt werden.
Die neuen Verbindungen weisen eine ausgezeichnete antimikrobielle Wirksamkeit auf und sind in dieser Hinsicht konstitutionsmässig nahestehenden Verbindungen wesentlich und in überraschender Weise überlegen. Besonders hervorzuheben ist ihre sehr gute Wirksamkeit gegenüber Staphylokokken und Streptokokken, ihre geringe Toxizität und ihre gute Hautverträglichkeit. Ausserdem weisen sie noch eine gute cestocide (bandwurmabtötende) Wirkung auf.
Das nachstehende Beispiel dient zur näheren Erläuterung der Erfindung.
Beispiel : 3- (4'-Chlorphenyl)-6-chlor-benzo-1, 3-oxazin-dion- (2, 4).
34, 4g 5-Chlorsalicylsäure und 30, 6 g p-Chlorphenylisocyanat wurden in 200 ml Dioxan gelöst, 10 h auf 80-90 C erhitzt und dann zwei Tage bei Zimmertemperatur aufbewahrt. Dann wurde das Lösungsmittel im Vakuum abdestilliert und der Rückstand mit 30g Thionylchlorid l h lang am Rückfluss erhitzt. Nach dem Abdestillieren des überschüssigen Thionylchlorids wurde der Rückstand aus Dioxan/Methanol kristallisiert. Man erhielt 33 g Substanz vom F. = 2150C.

The new compounds have excellent antimicrobial activity and, in this respect, are essential and surprisingly superior to compounds of similar constitution. Particularly noteworthy are their very good effectiveness against staphylococci and streptococci, their low toxicity and their good skin tolerance. They also have a good cestocidal (tapeworm-killing) effect.
The following example serves to explain the invention in more detail.
Example: 3- (4'-Chlorophenyl) -6-chloro-benzo-1, 3-oxazine-dione- (2, 4).
34.4 g of 5-chlorosalicylic acid and 30.6 g of p-chlorophenyl isocyanate were dissolved in 200 ml of dioxane, heated to 80-90 ° C. for 10 hours and then stored at room temperature for two days.
