EMI1.1

A ist zweckmässigerweise Schwefel.

The invention relates to a process for the preparation of certain novel pyridylthioureas which have a beneficial effect on the rate of gastric secretion.
In British Patent No. 1,267,433, a general class of compounds of the general formula
EMI1.1
where Ai is hydrogen or methyl, A2 is hydrogen, methyl, bromine or chlorine, As is hydrogen or methyl, Ai is hydrogen, methyl, bromine or chlorine and A5 is hydrogen or benzoyl, described as suitable intermediates in the preparation of certain pharmaceutically active imidazolines. The compounds of formula (1) were not believed to have any pharmacological activity. It has now been found that certain thioureas of the formula (1) and related thioureas have advantageous pharmacological properties and, in particular, reduce the rate of gastric acid secretion and the occurrence of gastric ulcers.
The invention thus provides a process for the preparation of new pyridylureas of the general formula

wherein Rg and R4 have the above meaning, is implemented.
When a compound of the formula (da.) Is employed, the reaction is conveniently carried out in an inert solvent at a temperature that is not extreme. Suitable solvents are, for example, chloroform, ethanol, dioxane, dimethylformamide, benzene or ethyl acetate. The reaction can

reaching. Ambient temperatures are preferred.
If a compound of the formula (mb) is used, the reaction is usually carried out in ethanolic solution at ambient temperature or under reflux.
Most conveniently, R5 is methyl or benzyl. A is conveniently sulfur.
This process variant usually does not give particularly good yields when both R1 and R2 are halogen.
If a compound of the formula (Dia) is used, the precursor of this compound is an isothiocyanate, which is produced by thermal decomposition of the corresponding unsubstituted thiourea of the general formula
EMI2.6
wherein R1 and R2 have the above meanings, can be prepared in a conventional manner, for example by heating in chlorobenzene.
Such thioureas of the formula (V) can be prepared according to the general process described in British Patent No. 1, 267, 433, for example by debenzoying an N-benzoylthiourea of the formula
EMI2.7
wherein R1 and R2 have the above meaning, which has been formed by reacting benzoyl isothiocyanate with a suitably ring-substituted 2-aminopyridine.
A second advantageous process for the preparation of such isothlocyanates consists in the implementation

Toluene.
In the case of compounds of formula (inb), the precursor can be prepared from the corresponding isothiocyanate by reaction with a compound H-AR5. The preparation of such isothiocyanates has already been described above.
If desired, the compound of the formula (II) can also be prepared directly from an unsubstituted thiourea of the formula (V) by heating this compound in the presence of an amine HNRgR. Such a reaction generally takes place in a high boiling solvent such as chlorine

benzene, or in a pressure vessel if the amine HNR3R4 has a boiling point below about 1100C.
Presumably the reaction takes place via the isothiocyanate of the formula (Dia).
According to a particularly preferred process variant, a compound of the formula (IIIa) is reacted with methylamine, 1- (3,5-dichloro-2-pyridyl) -3-methyl-2-thiourea being obtained.
The following example is intended to explain the invention in more detail without, however, being restricted thereto.

5.5 g (55%) of the desired isothiocyanate in the form of pale yellow needles, m.p. 45 to 460C. b) 1- (3,5-dichloro-2-pyridyl) -3-methyl-2-thiourea: A solution of 5.13 g of 3,5-dichloro-2-pyridyl isothiocyanate in 20 ml of benzene became a solution of
Added methylamine in ethanol (5 ml, 33%) and stirred the mixture at ambient temperature for 18 hours.
30ml petroleum ether (60 to 800C) were added and the precipitate obtained was filtered off and made from benzene
EMI3.2
Recrystallized petroleum ether - 2-thiourea were obtained, mp 105-1090C.
