Das Stammpatent Nr. 258296 betrifft ein Verfahren zur Herstellung von neuen 5, 6- Dihydro-6-oxo- pyrido[2, 3-b] [1, 4]benzoxazepinen der allgemeinen Formel I :

Es wurde nun gefunden, dass sich die neuen 5, 6-Dihydro-6-oxo-pyrido[2, 3-b] [l, 4]benzoxazepine (nach der in den Chemical Abstracts verwendeten Nomenklatur werden die Verbindungen besser als
Pyrido[2,3-b] [1,4]benzoxazepin-6(5H)-one bezeichnet) der Formel I, in der die Reste R. und R2 die ange- gebenen Bedeutungen besitzen, herstellen lassen, wenn man direKt von der entsprechenden Salicylsäure der Formel III ausgeht, in der die Reste Ri und R2 die für die Formel I angegebenen Bedeutungen haben und Rg ein Wasserstoffatom oder eine Acetylgruppe bedeutet.
Die Salicylsäure der Formel III kommt dabei mit einem 2-Halogen-3-amino-pyridin der Formel II unter Zugabe von Phosphortrihalogeniden oder Thionylhalogeniden, vorzugsweise in einem Lösungsmittel wie Toluol, Dioxan oder Chlorbenzol und bei-Temperaturen bis zum Siedepunkt dieser Lösungsmittel zur Umsetzung.
Die Umsetzung führt zu einem Zwischenprodukt der Formel IV, welches nicht notwendigerweise isoliert zu werden braucht. Eine Isolierung lässt sich jedoch leicht, z. B. durch Ansäuern mit Kohlen- säure erreichen. Dieses Zwischenprodukt wird auf die im Stammpatent angegebene Weise, also durch
Erhitzen auf Temperaturen oberhalb 1500 C, vorzugsweise in Gegenwart einer starken Base und in
Gegenwart oder in Abwesenheit eines inerten hochsiedenden Lösungsmittels, ringgeschlossen.
Die Verbindungen der Formel I, worin R und/oder R2 eine Aminogruppe bedeuten, können auch durch
Entacylierung der entsprechenden Acylaminoverbindungen erhalten werden. Dies erfolgt nach üblichen
Methoden, beispielsweise durch Umsetzung der Acylaminoverbindungen mit verdünnten Mineral- säuren.
Die Verbindungen der Formel I sind Ausgangsstoffe für die Herstellung von Heilmitteln, beispiels- weise für solche mit antipyretischer, antiphlogistischer, analgetischer, sedativer, antikonvulsiver, histamino- lytischer und reserpinantagonistischer Wirksamkeit ; sie weisen aber auch selbst wertvolle therapeutische
Eigenschaften auf, insbesondere sind sie stark antipyretisch wirksam.
Die nachstehenden Beispiele dienen zur näheren Erläuterung der Erfindung :
Beispiel 1: Pyrido[2,3-b] [1,4]benzoxazepin-6(5H)-on. a) 25, 8 g (0, 2 Mol) 2-Chlor-3-aminopyridin und 33, 2 g (0, 24 Mol) Salicylsäure werden in 120 ml absolutem Toluol in der Wärme gelöst. Zu der 1000 C warmen Lösung werden 31 g (0, 26 Mol) Thionyl- chlorid, gelöst in 20 ml absolutem Toluol, unter Rühren zugetropft. Anschliessend wird 3 h unter Rück- fluss erhitzt. Nach dem Abkühlen fügt man 16 g Ätznatron, gelöst in 120 ml Wasser, zu und trennt die dabei entstehenden zwei Phasen. Die wässerig-alkalische Phase wird mit Aktivkohle filtriert und mit
Wasser auf l, 51 verdünnt.
Durch Einleiten von Kohlensäure in dieser Lösung fällt das 2-Chlor-3- (2'-hy- droxy-benzoylamino)-pyridin aus, welches mit Wasser gewaschen wird. Schmelzpunkt : 193-195 C.
Ausbeute : 86% der Theorie.

Stickstoff langsam auf 235 C erhitzt. Das Reaktionsgemisch wird noch 30 min bei dieser Temperatur gelassen, daraufhin heiss filtriert und der Filterrückstand, der im wesentlichen aus Kaliumchlorid besteht, mit heissem Chinolin ausgewaschen. Die Chinolinlösung wird auf etwa die Hälfte eingeengt. Beim Abkühlen kristallisiert das Pyrido(2,3-b] [1,4]benzoxazepin-6(5H)-on aus. Ausbeute 75% der Theorie. Nach dem Umkristallisieren aus Dimethylformamid/Wasser zeigt diese Substanz einen Schmelzpunkt von 280 bis 2820 C.

Unter kräftigem Rühren erhitzt man 30 min unter Rück- nuss, lässt abkühlen und trennt die Phasen. Die Toluolphase wird mit verdünnter Natronlauge extrahiert. Die vereinigten wässerig-alkalischen Phasen werden über Aktivkohle filtriert und durch Einleiten von CO2-Gas wird im Filtrat das 2-Chlor-3- (2'-hydroxy-3'-methyl-benzoylamino)-pyridin ausgefällt. Es schmilzt nach dem Umkristallisieren aus Isopropanol und anschliessend aus Cyclohexan bei 140-142 C.
'b)'Die nach Beispiel 2 ä) erhaltene Verbindung wird nach der Methode des Beispiels 1 b) cyclisiert.
Es entsteht dabei das 10-Methyl-pyrido[2,3-b] [1,4]benzoxazepin-6(5H)-on, welches, aus Toluol umkristallisiert, einen Schmelzpunkt von 217 bis 219 C zeigt.'
Analyse von CigHioNOz (226, 2) :
Berechnet : C 69, 01% H 4, 46% N l2, 38%
Gefunden : C 68, 70% H 4, 51% N l2, 30%.
Beispiel 3: 9-Methyl-pyrido[2,3-b] [1,4]benzoxazepin-6(5H)-on. a) Auf die in Beispiel 1 a) beschriebene Weise erhält man aus 2-Chlor-3-aminopyridin und m-Kresotinsäure das 2-Chlor-3-(2'-hydroxy-4'-methyl-benzoylamino)-pyridin vom Schmelzpunkt 203-205 C : aus Isopropanol umkristallisiert). b) Die nach Beispiel 3 a erhaltene Verbindung wird nach der im Beispiel 1 b beschriebenen Weise cyclisiert. Es entsteht dabei das 9-Methyl-pyrido[2,3-b] [1,4]benzoxazepin-5(5H)-on vom Schmelzpunkt 260#261 C (aus Isopropanol umkristallisiert).

Gefunden : C 69, 00% H ze N l2, 45% Beispiel 4 : S-Methyl-pyrido [2, 3-b] [1, 4] benzoxazepin-6 (5H)-on. a) Aus 2-Chlor-3-aminopyridin und p-Kr@sotinsäure erhält man nach der im Beispiel 1a beschriebe- nen Methode das 2-Chlor-3-(2'-hydroxy-5'-methyl-benzoylamino)-pyridin vom Schmelzpunkt 163 bis 165 C (aus Isopropanol umkristallisiert). b) Die nach Beispiel 4 a erhaltene Verbindung wird nach der'im Beispiel 1 b beschriebenen Weise cyclisiert, wobei als Lösungsmittel an Stelle von Chinolin Tetraäthylenglykoldimethyläther verwendet wurde.
Das dabei erhaltene 8-Methyl-pyrido-[2,3-b] [1,4]benzoxazepin-6(5H)-on zeigte einen Schmelz- punkt von 250 bis 251 C (aus Isopropanol umkristallisiert).
Analyse von C13H10N2O2 (226,2):
Berechnet : C 69, 01% H 4, 46% N l2, 38%
Gefunden : C 69, 10% H 4, 62% N 12, 40%
Beispiel 5 : 8-tert. Butyl-pyrido[2,3-b] [1,4]-benzoxazepin-6(5H)-on. a) Aus 2-Chlor-3-aminopyridin und 2-Hydroxy-5-tert. butyl-benzoesäure erhält man nach der Methode des Beispiels 1 a) das 2-Chlor-3- (2'-hydroxy-5-tert. butyl-benzoylamino)-pyridin vom Schmelzpunkt 154#156 C (aus Benzin umkristallisiert). b) Die nach Beispiel 5 a erhaltene Verbindung wird nach der in Beispiel 1 b beschriebenen Weise cyclisiert. Das dabei erhaltene Produkt zeigt einen Schmelzpunkt von 197 bis 199 C (aus Isopropanol umkristallisiert).
Analyse von C16H16N2O2 (268,3):
Berechnet : C 71, 62% H 6, 01% N 10, 45%
Gefunden : C 71, 30% H 5, 85% N 10, 46%
Beispiel6 :9-Methoxy-pyrido[2,3-b][1,4]benzoxazepin-6(5H)-on. a) Aus 2-Chlor-3-aminopyridin und 2-Hydroxy-4-methoxy-benzoesäure und nach der im Beispiel 1 a beschriebenen Weise erhält man das 2-Chlor-3-(2'-hydroxy-4'-methoxy-benzoylamino)-pyridin vom Schmelzpunkt 193,5 C (aus Acetonitril). b) Die nach Beispiel 6 a erhaltene Verbindung wird mit Kaliumäthylat nach der im Beispiel 1 b beschriebenen Weise cyclisiert. Die dabei erhaltene Substanz zeigt einen Schmelzpunkt von 313 C (aus Dimethylformamid umkristallisiert).

Berechnet : C 64, 40% H 4, l6% N 11, 56%
Gefunden : C 64, 30% H 4, 42% N 11,70%

1 g 8-Acetamino-pyrido[2,3-b] [1,4]benzoxazepin-6(5H)-on (hergestellt nach Beispiel 9) werden in ? ml 10%iger Salzsäure gelöst und 45 min bei 80 C gerührt. Nach dem Abkühlen wird filtriert, das

Filtrat wird mit NaOH auf den pH-Wert 8 eingestellt. Die hiebei ausgefallene Aminoverbindung wird abgetrennt und aus wässerigem Dimethylformamid umkristallisiert.
F. = 285-287 C.

Beispiel 11 : 9-Amino-pyrido [2, 3-b] [l, 4] benzoxazepin-6 (5H)-on.
46, 0 g (0, 3 Mol) 4-Aminosalicylsäure und 39, 0 g (0, 3 Mol) 2-Chlor-3-aminopyridin werden mit 300 mI absolutem Dioxan verrührt und mit 15, 0 g (0, 11 Mol) Phosphortrichlorid versetzt. Das Gemisch wird unter Rühren 1 h bei 100 C gehalten. Nach dem Abkühlen wird das ausgefallene 2-Chlor-3- (2'-hy- droxy-4'-amino-benzoylamino)-pyridin abgetrennt, in verdünnter NaOH gelöst und mit COg-Gas wieder ausgefällt. Diese Substanz wird dann nach der im Beispiel 1 b beschriebenen Weise cyclisiert. F.-328 C unter Zersetzung (aus wässerigem Pyridin).
