Die Erfindung betrifft ein Verfahren zur Herstellung neuer Verbindungen der Formel I, worin n eine ganze Zahl von 0 bis 2 bedeutet, R1 für eine gegebenenfalls durch Fluor, Chlor, Brom, Methoxy oder niederes Alkyl monosubstituierte Phenylgruppe steht und R2 Wasserstoff oder Methyl bedeutet, und ihrer Säureadditionssalze.
Erfindungsgemäss gelangt man zu den neuen Verbindungen der Formel I und ihren Säureadditionssalzen, indem man Verbindungen der Formel II, worin R, und n obige Bedeutung besitzen und X für Chlor oder Brom steht, mit Verbindungen der Formel III, worin R2 obige Bedeutung besitzt, umsetzt und gewünschtenfalls die erhaltenen Verbindungen der Formel I in ihre Säureadditionssalze überführt.
Falls R1 einen durch eine niedere Alkylgruppe substituierten Phenylrest darstellt, so besteht diese vorzugsweise aus 1 bis 4 Kohlenstoffatomen.
Vorzugsweise wird das erfindungsgemässe Verfahren mit einem Überschuss der Verbindung der Formel III, beispielsweise 5-10 Mol einer Verbindung der Formel III, bezogen auf ein Mol einer Verbindung der Formel II oder in Gegenwart eines anderen basischen Mittels, welches die bei der Umsetzung entstehende Säure zu binden vermag, z.B. eines tertiären Amines oder eines Alkalimetall- oder Erdalkalimetallhydroxides oder -karbonates durchgeführt. Die Reaktion kann beispielsweise in einem unter den Reaktionsbedingungen inerten Lösungsmittel, z.B. einem niederen Alkohol, wie Methanol oder Athanol, oder einem cyclischen Äther, wie Dioxan oder Tetrahydrofuran, bei Temperaturen zwischen ca. 20 und ca.
150", vorzugsweise bei Temperaturen zwischen 80 und 120 bzw. bei Siedetemperatur des Reaktionsgemisches ausgeführt werden, und die Reaktionszeit kann zwischen 1 und 20 Stunden betragen. Gegebenenfalls kann auch ein Überschuss der Verbindung der Formel III als Lösungsmittel dienen.
Die nach dem vorstehend beschriebenen Verfahren hergestellten Hydrazinverbindungen können nach bekannten Methoden isoliert und gereinigt werden. Aus den freien Basen lassen sich in üblicher Weise Säureadditionssalze herstellen und umgekehrt.
Die Ausgangsverbindungen können beispielsweise folgendermassen hergestellt werden:
Die Verbindungen der Formel II können erhalten werden, indem man Verbindungen der Formel IIa, worin X obige Bedeutung besitzt, mit einer Verbindung der Formel IV, worin Rls n und X obige Bedeutung besitzen, in einem unter den Reaktionsbedingungen inerten Lösungsmittel, z.B. einem chlorierten Kohlenwasserstoff, wie Chloroform oder Äthylenchlorid, in Gegenwart eines säurebindenden Mittels, z.B. eines Alkalimetallkarbonates oder einer tertiären Base, wie Tri äthylamin, umsetzt.
Die Verbindungen der Formel IIa können erhalten werden, indem man die Verbindung der Formel Va mit einem geeigneten Chlorierungs- bzw. Bromierungsmittel, z.B. mit Phosphoroxychlorid, Phosphortri- oder -pentachlorid oder Phosphoroxybromid, erhitzt, vorzugsweise auf Temperaturen bis zu ca.
100 bzw. auf Siedetemperatur des Reaktionsgemisches.
Die Verbindung der Formel Va wird z.B. erhalten, indem man in Verbindungen der Formel Vb, worin Rl niederes Alkoxy bedeutet, die Carbalkoxygruppe sauer, z.B. durch mehrstündiges Erhitzen in saurem Medium, beispielsweise in Salzsäure oder alkalisch, z.B. durch Erhitzen mit einem Alkalimetallhydroxid, wie Kaliumhydroxid, in einem höhersiedenden Alkanol, wie n-Butanol, abspaltet.
Verbindungen der Formel Vb können z.B. erhalten werden, indem man Verbindungen der Formel VI, worin Rl obige Bedeutung besitzt, in einem unter den Reaktionsbedingungen inerten Lösungsmittel, z.B. einem niederen Alkohol, wie Äthanol, in Gegenwart von mindestens äquivalenten Mengen Eisessig mit Hydrazinhydrat oder einem Salz des Hydrazins gegebenenfalls unter Inertgasatmosphäre durch Erhitzen auf Temperaturen zwischen 70 bis 110", vorzugsweise auf Siedetemperatur des Reaktionsgemisches. cyclisiert und die gebildeten Verbindungen der Formel VII, worin Rl obige Bedeutung besitzt, oxydiert, vorzugsweise mit Brom unter Verwendung eines halogenierten Kohlenwasserstoffes, wie Chloroform oder Äthylenchlorid. als Lösungsmittel.
Verbindungen der Formel VI können beispielsweise erhalten werden, indem man Verbindungen der Formel VIII, worin Rl obige Bedeutung besitzt. mit einem sekundären Amin, vorzugsweise einem acyclischen Amin, wie z.B. Pyrrolidin, Morpholin oder Piperidin, vorzugsweise in einem unter den Reaktionsbedingungen inerten organischen Lösungsmittel, z.B.
einem aromatischen Kohlenwasserstoff, wie Benzol, gegebenenfalls unter Zusatz eines Katalysators, z.B. p-Toluolsulfonsäure oder eines Molekularsiebes, bei erhöhter Temperatur, vorzugsweise bei Siedetemperatur des Reaktionsgemisches, zu einem Enamin der Formel IX, worin Rl obige Bedeutung besitzt und Y für eine sekundäre Aminogruppe steht, umsetzt, dieses mit Bromessigsäurealkylester in Gegenwart eines unter den Reaktionsbedingungen inerten organischen Lösungsmittels, z.B. eines aromatischen Kohlenwasserstoffes, wie Benzol.
eines Halogenkohlenwasserstoffes, wie Chloroform oder eines Äthers oder Dimethylformamid bei Raumtemperatur versetzt, das Reaktionsgemisch mehrere Stunden erhitzt, vorzugsweise auf Siedetemperatur. und in dem erhaltenen Reaktionsprodukt der Formel X, worin Rl und Y obige Bedeutung besitzen, die Enamingruppierung durch Erhitzen mit Wasser, gegebenenfalls unter Zugabe von verdünnter Lauge, Ammoniaklösung oder verdünnter Mineralsäure wieder spaltet.
Die Verbindungen der Formel I und ihre pharmakologisch verträglichen Säureadditionssalze sind in der Literatur bisher noch nicht beschrieben worden. Sie zeichnen sich durch interessante pharmakodynamische Eigenschaften aus und können daher als Heilmittel verwendet werden.
Wie sich bei Versuchen an der hypertonen Grollmann-Ratte zeigt. besitzen die Substanzen antihypertensive Wirkungen.
Die zu verwendenden Dosen variieren naturgemäss je nach Art der Substanz, der Administration und des zu behandelnden Zustandes. Im allgemeinen werden jedoch bei Testtieren befriedigende Resultate mit einer Dosis von 0,1 bis 10 mg/kg Körpergewicht erhalten: diese Dosis kann nötigenfalls in 2 bis 3 Anteilen oder auch als Retardform verabreicht werden. Für grössere Säugetiere liegt die Tagesdosis bei etwa 5 bis 500 mg.
Für orale Applikationen enthalten die Teildosen etwa 2 bis 250 mg der Verbindungen der Formel I neben festen oder flüssigen Trägersubstanzen.
Aufgrund ihrer blutdrucksenkenden Wirkung können die Substanzen in der Hochdrucktherapie Anwendung finden.
Als Heilmittel können die Verbindungen der Formel I bzw.
ihre physiologischsverträglichen Säureadditionssalze allein oder in geeigneter Arzneiform mit pharmakologisch indifferenten Hilfsstoffen verabreicht werden.
Soweit die Herstellung der Ausgangsverbindungen nicht beschrieben wird. sind diese bekannt oder nach an sich bekannten Verfahren bzw. analog zu den hier beschriebenen oder analog zu an sich bekannten Verfahren herstellbar.
In den nachfolgenden Beispielen, die die Erfindung näher erläutern, ihren Umfang aber in keiner Weise einschränken sollen, erfolgen alle Temperaturangaben in Celsiusgraden.
Beispiel 1 6-Benzoyl-3-hydrazino-5.6,7,8-telrahydro pyrido[4.3-cipyridazin
Eine Aufschlämmung von 1.6 g 6-Benzoyl-3-chloro 5,6,7,8-tetrahydropyrido[4,3-c]pyridazin in 80 ml Hydrazinhydrat wird bei einer Badtemperatur von 110o während 1 Stunde unter gelindem Kochen am Rückfluss gerührt. Nach ca. 15 Minuten Reaktionszeit erhält man eine vollständige Lösung.
Der Ansatz wird mit Eis gekühlt und die ausgefallene Substanz abfiltriert und mit wenig absolutem Äthanol nachgewaschen.
Das Rohprodukt.wird in 60 ml Dimethylformamid gelöst, filtriert und das Filtrat mit 60 ml absolutem Äthanol versetzt, worauf die Verbindung auskristallisiert. Man filtriert die im Titel genannte analysenreine Substanz ab. Sie hat nach dem Trocknen einen Smp. von 220-223" (Zersetzung).
Das als Ausgangsmaterial benötigte 6-Benzoyl-3 chloro-5,6,7,8 -tetrahydropyrido [4,3-c]pyridazin kann folgendermassen hergestellt werden: a) Eine Mischung von 342,6 g 1-Carbäthoxy-4-piperidon und 214,0 g Pyrrolidin in 800 ml Benzol werden während
2 Stunden unter Sieden am Rückfluss erhitzt. Das wäh rend dieser Zeit entstandene Wasser wird azeotrop aus der Reaktionsmischung kontinuierlich entfernt. Der
Ansatz wird zum orangen Öl eingeengt und dieses im
Hochvakuum destilliert. Als Hauptfraktion erhält man bei Kp 144-150"/0,03 mm den analysenreinen 1,2,3,6
Tetrahydro-4-pyrrolidinylpyridin- 1 -carbonsäureäthyl ester.
b) Eine Mischung von 1460 g 1,2,3,6-Tetrahydro-4 pyrrolidinylpyridin- l-carbonsäure äthylester und 1250 g
Bromessigsäureäthylester in 6,0 1 Benzol wird während
20 Stunden unter Rühren am Rückfluss erhitzt. Der
Ansatz wird mit 2,01 Wasser versetzt und erneut 2 Stun den unter Kochen am Rückfluss erhitzt. Nach dem
Abkühlen auf Zimmertemperatur setzt man 400 ml 10 %eigen Ammoniak zu und trennt die Benzolphase ab.
Diese wird mit Natriumsulfat getrocknet und nach dem
Filtrieren zum Öl eingeengt. Die Hauptfraktion, der 1 Carbäthoxy-4-piperidon-3 -essigsäureäthylester, siedet bei einem Kp 145-170"/0,05 mm.
c) Eine Mischung von 514 g 1-Carbäthoxy-4-piperidon
3-essigsäureäthylester und 100 g Hydrazinhydrat in 1,5 1
95 %igem Äthanol und 200 ml Eisessig erhitzt man in einem Stickstoffstrom während 4 Stunden unter Rühren am Rückfluss. Danach wird der Ansatz vollständig ein geengt. Der kristallisierte Rückstand wird aus 1,0 1
95 %igem Äthanol umkristallisiert und ergibt den 2,3,4,4a,5,6,7,8-0ctahydro-3-oxo-6- pyrido[4,3 -c]pyridazincarbonsäureäthylester,
Smp. 163-166" (Zersetzung).
d) 450,5 g 2,3,4,4a,5,6,7,8-Qctahydro-3-oxo-6- pyrido[4,3-c]pyridazincarbonsäureäthylester werden in
2,0 1 Chloroform gelöst und die Lösung auf Rückfluss temperatur erhitzt. In diese kochende Lösung gibt man innerhalb von 2l/2 Stunden 320 g Brom in 500 ml Chlo roform. Der Ansatz wird nach der Bromzugabe noch eine weitere Stunde unter Rückfluss gerührt. Nach dem
Abkühlen auf Raumtemperatur wird der Ansatz mit 1 kg
Eis/Wasser versetzt und der Chloroformteil abgetrennt.
Der saure wässrige Teil wird mit weiteren 500 ml Chloro form ausgezogen. Die Chloroformteile werden der Reihe nach mit 300 ml Wasser gewaschen, über Natriumsulfat getrocknet und im Vakuum eingeengt. Der halbkristalline
Rückstand wird mit 250 ml absolutem Äthanol umkri stallisiert und ergibt das analysenreine 6-Carbäthoxy-5,6,7,8-tetrahydro-
3(2H)pyrido[4,3-c]pyridazinon,
Smp. 165-1680 (Zersetzung).
e) Eine Lösung von 223,2 g 6-Carbäthoxy-5,6,7,8 tetrahydro-3(2H)pyrido[4,3-c]pyridazinon in 1,0 1 kon- zentrierter Salzsäure wird während 22 Stunden unter
Kochen am Rückfluss gerührt. Der Ansatz wird im
Vakuum vollständig eingeengt, der kristalline Rückstand in Methanol aufgenommen und abfiltriert. Das rohe
Hydrochlorid des 5,6,7,8-Tetrahydro-3(2H)pyrido[4,3 -c]pyridazinon,
Smp. 307-3100 (Zersetzung), wird in 0,75 1 Methanol aufgeschlämmt und 0,4 1 Triäthylamin langsam zugege ben. Die violette Aufschlämmung wird nach 15 Minuten
Rühren auf 30O gekühlt, abfiltriert und mit wenig Metha nol/Äther nachgewaschen.
Man erhält die Rohbase vom
Fp. 216-2190 (Zersetzung). 25,0 g Rohbase werden aus
300 ml Methanol, dem man 10 ml konzentrierten Ammo niak und 40 ml Wasser beimischt, unter Zusatz von wenig
Kohle um kristallisiert und ergeben das analysenreine 5,6,7,8-Tetrahydro-3 (2H)pyrido[4,3-c]pyridazinon vom Smp. 223-2250 (Zersetzung).
f) Eine Lösung von 30,3 g 5,6,7,8-Tetrahydro
3(2H)pyrido[4,3-c]pyridazinon in 250 ml Phosphoroxy chlorid wird während 2 Stunden unter Kochen am Rück fluss gerührt. Der Ansatz wird im Vakuum vollständig eingeengt und zum kalten Rückstand werden 500 g Eis/
Wasser gegeben. Die saure, erneut gekühlte Lösung wird in ein Gemisch aus 500 ml Chloroform und 250 ml kon zentriertem Ammoniak langsam hineingerührt. Die wäss rige Phase wird mit vier weiteren Portionen zu je 250 ml
Chloroform, total 1,5 1 Chloroform extrahiert, diese
Chloroformteile über Natriumsulfat getrocknet und im
Vakuum eingeengt. Man erhält die rohe, unstabile 3-Chlor-5,6,7,8-tetrahydropyrido[4,3-c]pyridazin-Base, die zur Weiterverarbeitung in das Maleinat übergeführt wird. Dazu löst man 24,8 g Base in 150 ml Methanol und gibt 17,5 g Maleinsäure zu.
Nach dem Aufkochen, Filtrie ren, Abkühlen erhält man das rohe Maleinat der Verbin dung vom Smp. 156-1590 (Zersetzung). Eine Probe davon wird aus Methanol unter Zusatz von wenig Kohle umkristallisiert und ergibt die analysenreine Verbindung vom Smp. 162-1640 (Zersetzung).
g) In eine Aufschlämmung von 25,6 g 3-Chlor-5,6,7,8 tetrahydropyrido[4,3-c]pyridazin-maleinat in 250 ml Äthylenchlorid und 21,8 g Triäthylamin tropft man wäh rend 18 Minuten unter Rühren bei Zimmertemperatur eine Mischung von 12,6 g Benzoylchlorid in 100 ml Äthylenchlorid. Der Ansatz wird während 14 Stunden bei der gleichen Temperatur weitergerührt, mit 200 ml Was ser versetzt und die organische Phase abgetrennt. Sie wird über Natriumsulfat getrocknet, filtriert und im Vakuum zu einem Öl eingeengt. Beim Versetzen dieses Öles mit Äther/Dimethoxyäthan erhält man das rohe
6-Benzoyl-3-chlor-5,6,7,8 tetrahydropyrido[4,3 -c]pyridazin.
Dieses wird aus absolutem Äthanol unter Zusatz von wenig Kohle um kristallisiert und ergibt die analysenreine
Verbindung vom Smp. 125-1270 (Zersetzung).
Beispiel 2 Bis[3-Hydrazino-5,6,7,8-tetrahydro-6-(o-toluoyl)- pyrido[4,3 -c]pyridazin]-trisfumarat
Eine Aufschlämmung von 13,0 g 3-Chlor-5,6,7,8-tetra hydro-6-(o-toluoyl)pyrido[4,3-c]pyridÅazin in 100 ml Hydrazin hydrat wird bei einer Badtemperatur von 110o während 2 Stunden unter gelindem Kochen am Rückfluss gerührt. Nach ca. 15 Minuten Reaktionszeit erhält man eine vollständige Lösung. Die rote Reaktionslösung wird im Vakuum zu einem dunkelbraunen Öl eingeengt, dieses in 300 ml Chloroform aufgenommen, über Natriumsulfat getrocknet, filtriert und erneut eingeengt.
12,0 g des Öls werden mit 21,0 g Fumarsäure in 100 ml Äthanol auf dem Wasserbad erhitzt. Nach gutem Kühlen wird das ausgefallene Hydrazinfumarat abfiltriert und verworfen.
Aus der Mutterlauge kristallisiert nach einigem Stehen das rohe Fumarat des 3-Hydrazino-5,6,7,8-tetrahydro-6- (o-toluoyl)pyrido[4,3-c]pyridazins.
Nach zweimaligem Umkristallisieren aus 95 %igem Äthanol erhält man die analysenreine Titelverbindung vom Smp. 98 100o (Zersetzung).
Das als Ausgangsmaterial benötigte 3-Chlor-5,6,7,8 tetrahydro-6-o-toluoylpyrido[4,3-c]pyridazin kann folgendermassen hergestellt werden:
In eine Aufschlämmung von 28,5 g 3-Chlor-5,6,7,8 tetrahydropyrido[4,3-c]pyridazin-maleinat (Herstellung siehe Beispiel 1, Stufen a bis f) in 250 ml Äthylenchlorid und 21,8 g Triäthylamin tropft man während 18 Minuten unter Rühren bei Zimmertemperatur eine Mischung von 16,9 g o-Toluoylchlorid in 100 ml Äthylenchlorid. Der Ansatz wird während 24 Stunden bei der gleichen Temperatur weitergerührt und in üblicher Weise aufgearbeitet. Das erhaltene 3-Chlor-5,6,7,8-tetrahydro-6-o- toluoylpyrido[4,3-c]pyridazin schmilzt nach Umkristallisation aus 95 %igem Äthanol/Äther bei 123 bis 1260 (Zersetzung).
Beispiel 3 6-Benzoyl-5 ,6,7,8-tetrahydro-3-(1-methylhydrazino)- pyrido[4,3 -cjpyridazin-bisgentisinat
Eine Aufschlämmung von 13,7 g 6-Benzoyl-3-chlor 56,7, 8-tetrahydropyrido [4,3-c]pyridazin (Herstellung siehe Beispiel 1, Stufen a bis g) in 40 ml Methylhydrazin wird bei einer Badtemperatur von 50O während 6'/4 Stunden gerührt.
Nach einer Viertelstunde entsteht eine vollständige Lösung.
Die Reaktionslösung wird im Vakuum zum Öl eingeengt.
Dieses wird in 200 ml Chloroform gelöst, über Natriumsulfat getrocknet, filtriert und erneut zum Öl eingeengt. Dieses wird mit 12,0 g Gentisinsäure in 100 ml Methanol kurz auf dem Wasserbad aufgekocht, die Lösung auf etwa 2/3 eingeengt und das ausgefallene Material nach dem Kühlen mit Eis abfiltriert.
Nach dem Umkristallisieren aus absolutem Äthanol erhält man die analysenreine im Titel genannte Verbindung vom Smp.
166 bis 1680 (Zersetzung).
Beispiel 4 Bis[6-(o-Chlorbenzoyl)-3 -hydrazino-5,6,7,8tetrahydropyridot4,3 -c]pyridazin]-trisfumarat
Eine Aufschlämmung von 7,7 g 3-Chlor-6-(o-chlorben zoyl)-5,6,7,8-tetrahydropyrido[4,3-c]pyridazin in 30 ml Hydrazinhydrat wird bei einer Badtemperatur von 800 während 20 Stunden gerührt. Nach I l/4 Stunden gibt man zur Reaktion 25 ml Isopropanol, worauf eine vollständige Lösung entsteht. Die Reaktionslösung wird nach Beendigung der Reaktion zum dunkelbraunen Schaum eingeengt. Dieser wird in 200 ml Chloroform aufgenommen und mit 25 ml Wasser ausgeschüttelt. Die organische Phase wird abgetrennt, über Natriumsulfat getrocknet, filtriert und im Vakuum zum hellgelben Schaum eingeengt.
Dieser wird in 50 ml 95%igem Äthanol gelöst, mit 5,8 g Fumarsäure versetzt und auf dem Wasserbad kurz aufgekocht.
Das kristalline Fumarat wird abfiltriert und aus 20 ml 95%igem Äthanol umkristallisiert. Man erhält dabei die analysenreine im Titel genannte Verbindung vom Smp. 144 bis 1160 (Zersetzung).
Das als Ausgangsmaterial benötigte 3-Chlor-6-(o-chlor benzoyl)-5,6,7,8-tetrahydropyrido[4,3-cjpyridazin kann analog Beispiel 1g aus 28,5 g 3-Chlor-5,6,7,8-tetra- hydropyrido[4,3-c]pyridazin-maleinat und 16,9 g - o-Chlorbenzoylchlorid erhalten werden. Smp. nach Umkristallisation aus 95 cXcigem Äthanol 97 bis 99o (Zersetzung).
Beispiel 5 3-Hydrazino-5,6,7,8-tetrahydro-6-(3 -phenylpropionyl)pyrido [4,3-cjpyridazin
Eine Aufschlämmung von 8,5 g 3-Chlor-5,6,7,8-tetra hydro-6-(3-phenylpropionyl)pyrido[4,3-c]pyridazin und 80 ml Hydrazinhydrat wird bei einer Badtemperatur von 800 während 4 Stunden gerührt. Nach 3l/2 Stunden erhält man eine vollständige Lösung, die nach Beendigung der Reaktion mit Eis gekühlt wird. Die ausgefallene Substanz wird abfiltriert und aus 95 O/cigem Äthanol umkristallisiert. Das so erhaltene analysenreine, im Titel genannte Produkt hat den Smp. 162 bis 1650 (Zersetzung).
Das als Ausgangsmaterial benötigte 3-Chlor-5,6,7,8- tetrahydro-6-(3-phenylpropionyl)-pyrido[4,3-c]pyridazin kann analog Beispiel 1g aus 28,6 g 3-Chlor-5,6,7,8 tetrahydropyrido[4,3-cjpyridazin-maleinat und 31,5 g 3-Phenylpropionylchlorid erhalten werden. Smp. nach Umkristallisation aus 956Scigem Äthanol 134 bis 137".
Beispiel 6 Bis[6-(p-tert.Butylbenzoyl)-3-hydrazino-5,6,7,8- tetrahydropyrido[4,3-c]pyridazin]-trisfumarat
Eine AufSchlämm ung von 12,0 g 6-(p-tert.Butylbenzoyl) 3-chlor-5,6,7,8-tetrahydropyrido[4,3-c]pyridazin in 75 ml
Hydrazinhydrat wird bei einer Badtemperatur von 100o während 4 Stunden gerührt. Der Ansatz wird mit Eis gekühlt und das kristalline Material abfiltriert. Das Rohprodukt wird zusammen mit 7,2 g Fumarsäure in 50 ml Methanol kochend gelöst und ergibt das analysenreine, im Titel genannte Produkt vom Smp. 115 bis 1180 (Zersetzung).
Das als Ausgangsmaterial benötigte 6-(p-tert.Butyl benzoyl)-3-chlor-5,6,7,8-tetrahydropyrido[4,3-c]pyridazin kann analog Beispiel 1g aus 34,2 g 3-Chlor-5,6,7,8- tetrahydropyrido[4,3-c]pyridazin-maleinat und 25,0 g tert.Butylbenzoylchlorid erhalten werden. Smp. nach Umkristallisa tion aus 95Sigem Äthanol 157 bis 1600 (Zersetzung).
Beispiel 7 3-Hydrazino-5,6,7,8-tetrahydro-6 (p-toluoyl)pyrido[4,3-c]pyridazin
Eine Aufschlämmung von 12,0 g 3-Chlor-5,6,7,8-tetrahydro-6-(p-toluoyl)pyrido [4,3-c]pyridazin und 50 ml Hydrazinhydrat wird bei einer Badtemperatur von 100o während 2 Stunden gerührt. Nach Beendigung der Reaktion wird gekühlt und das ungelöste Material abfiltriert. Nach einmaligem Umkristallisieren des Rohproduktes aus einer Mischung von 100 ml 956Scigem Äthanol und 50 ml Methanol erhält man die analysenreine. im Titel genannte Verbindung mit dem Smp. 207 bis 210O (Zersetzung).
Das als Ausgangsmaterial benötigte 3-Chlor-5,6,7,8 tetrahydro-6- (p-toluoyl)pyrido[4,3-cjpyridazin kann analog Beispiel 2 aus 28,5 g 3-Chlor-5,6,7,8-tetrahydropyrido[4,3-c]pyridazin-maleinat und 18,2 g p-Toluoylchlorid hergestellt werden, Smp. 152-1530 (Zersetzung, aus absolutem Athanol).
Beispiel 8 6-(m-Chlorbenzoyl)-3-hydrazino-5,6,7,8tetrahydropyrido[4,3 -c]pyridazin-fumarat
Eine anfängliche Aufschlämmung von 10,8 g 3-Chlor-6 (m-chlorbenzoyl)-5,6,7,8-tetrahydropyrido[4, 3-cj- pyridazin und 50 ml Hydrazinhydrat wird bei einer Badtemperatur von 100o während total 4 Stunden gerührt. Nach 2stündiger Reaktionszeit gibt man zur Aufschlämmung 25 ml Isopropanol, nach 4 Stunden ist eine vollständige Lösung entstanden. Der Ansatz wird im Vakuum zum kristallinen Rückstand eingeengt. Dieser wird zwischen 150 ml Chloroform und 30 ml Wasser verteilt. Der organische Teil wird mit wenig Wasser nachgewaschen, über Natriumsulfat getrocknet, filtriert und im Vakuum zur Rohbase eingeengt.
Diese wird mit Fumarsäure in absolutem Äthanol in das kristalline, im
Titel genannte Fumarat überführt, Smp. 178 bis 1800 (Zersetzung).
Das als Ausgangsmaterial benötigte 6-(m-Chlorbenzoyl) 3-chlor-5,6,7,8-tetrahydropyrido[4,3-c]pyridazin wird analog Beispiel 1g aus 28,5 g 3-Chlor-5,6,7,8-tetrahydropy rido[4,3-c]pyridazin-maleinat und 19,3 g m-Chlorbenzoyl chlorid erhalten, Smp. 194 bis 1970 (Zersetzung, aus Metha nol).
Beispiel 9
6-(p-Brombenzoyl)-3-hydrazino-5,6,7,8 tetrahydropyrido[4,3-c]pyridazin
Eine Aufschlämmung von 10,2 g 6-(p-Brombenzoyl)-3 chlor- 5,6,7,8-tetrahydropyrido[4,3-c]pyridazin und 55 ml Hydrazinhydrat wird bei einer Badtemperatur von 100o während 2 Stunden gerührt. Es entsteht nie eine vollständige
Lösung. Die Reaktionsmischung wird mit Eis gekühlt und das ungelöste Material abfiltriert. Das Rohprodukt wird aus einem
Gemisch von 75 ml absolutem Äthanol und 25 ml Methanol umkristallisiert und ergibt die analysenreine, im Titel genannte
Verbindung vom Smp. 192 bis 1950 (Zersetzung).
Das als Ausgangsmaterial benötigte 6-(p-Brombenzoyl) 3-chlor-5,6,7,8-tetrahydropyrido[4,3-c]pyridazin kann analog Beispiel ig aus 14,3 g 3-Chlor-5,6,7,8-tetrahydropy rido[4,3-c]pyridazin-maleinat und 13,2 g p-Brombenzoyl chlorid erhalten werden, Smp. nach Umkristallisation aus absolutem Äthanol 164 bis 1670 (Zersetzung).
Beispiel 10 3 -Hydrazino-5,6,7,8-tetrahydro-6-(p-methoxybenzoyl)-pyrido- [4,3-c]pyridazin
Eine Aufschlämmung von 10,5 g 3-Chlor-5,6,7,8-tetra hydro-6-(p-methoxybenzoyl)-pyrido[4,3-c]pyridazin in 100 ml Hydrazinhydrat wird bei einer Badtemperatur von 100o während total 1 Stunde gerührt. Nach 50 Minuten Reaktionszeit entsteht eine vollständige Lösung, die mit Eis gekühlt wird. Das ausgefallene Reaktionsprodukt wird abfiltriert und einmal aus 15 ml 95%igem Äthanol umkristallisiert. Man erhält die analysenreine, im Titel genannte Verbindung vom Smp. 169 bis 1720 (Zersetzung).
Das als Ausgangsmaterial benötigte 3-Chlor-5,6,7,8 tetrahydro-6-(p-methoxybenzoyl)pyridol[4,3-cjpyridazin kann analog Beispiel 1g aus 28,5 g 3-Chlor-5,6,7,8 tetrahydropyrido [4,3-c]pyridazin-maleinat und 17,1 g p-Methoxybenzoylchlorid erhalten werden, Smp. nach Umkristallisation aus 95%igem Äthanol 163 bis 1640.
Beispiel 11 6-(o-Fluorbenzoyl)-3-hydrazino-5,6,7,8- tetrahydropyrido[4,3 -c]pyridazin
Eine Aufschlämmung von 11,6 g 3-Chlor-6-(o-fluor benzoyl)-5,6,7,8-tetrahydropyrido[4,3 -cjpyridazin in 80 ml Hydrazinhydrat wird bei einer Badtemperatur von 100o während 3'/2 Stunden gerührt. Der Ansatz wird mit Eis gekühlt und das feste Material abfiltriert. Nach einmaligem Umkristallisieren des Rohproduktes aus einem Lösungsmittel gemisch aus 100 ml 95%igem Äthanol und 100 ml Methanol erhält man die analysenreine, im Titel genannte Verbindung vom Smp. 210 bis 2130 (Zersetzung).
Das als Ausgangsmaterial benötigte 3-Chlor-6 (o-fluorbenzoyl)-5,6,7 ,8-tetrahydropyrido [4,3-c]- pyridazin kann analog Beispiel 1g aus 28,6 g 3-Chlor 5,6 ,7,8-tetrahydropyrido[4,3-c]pyridazin-maleinat und
17,4 g o-Fluorbenzoylchlorid erhalten werden. Smp. nach Kristallisation und Umkristallisation aus 95 %igem Äthanol
144 bis 1460 (Zersetzung).
Beispiel 12 3-Hydrazino-5,6,7,8-tetrahydro-6- (o-methylphenacetyl)pyrido[4,3 -c]pyridazin
Eine anfängliche Aufschlämmung von 10,0 g 3-Chlor 5 ,6,7,8-tetrahydro-6-(o-methylphenacetyl)pyrido- [4,3-c]pyridazin in 35 ml Hydrazinhydrat wird bei einer Badtemperatur von 100o während 30 Minuten gerührt. Während dieser Zeit entsteht eine vollständige Lösung. Die Reaktionslösung wird langsam abkühlen gelassen und das während die ser Zeit ausgefallene Reaktionsprodukt abfiltriert. Nach einmaligem Umkristallisieren aus absolutem Äthanol erhält man die analysenreine, im Titel genannte Verbindung mit dem
Smp. 155 bis 1570 (Zersetzung).
Das als Ausgangsmaterial benötigte 3-Chlor-5,6,7,8tetrahydro-6 -(o-methylphenacetyl)pyrido[4,3-c]pyridazin kann analog Beispiel 1g aus 28,6 g 3-Chlor 5,6,7,8-tetrahydropyrido[4,3-c]pyridazin-maleinat und 20,2 g o-Methylphenacetylchlorid erhalten werden. Smp.
nach Kristallisation und Umkristallisation aus 95 %igem.Atha- nol 125 bis 1270 (Zersetzung).
Beispiel 13 Bis[3-hydrazino-5,6,7,8-tetrahydro-6 phenacetylpyrido[4,3 -c]pyridazin] -trisfumarat
Eine Aufschlämmung von 9,7 g 3-Chlor-5,6,7,8-tetra hydro-6-phenacetylpyrido[4,3-c]pyridazin in 35 ml Hydrazinhydrat und 35 ml Isopropylalkohol wird während 1 Stunde 30 Minuten bei einer Badtemperatur von 100o unter Sieden am Rückfluss gerührt. Der Ansatz wird im Vakuum zu einem roten Öl eingeengt und dieses zwischen Chloroform und Wasser verteilt. Die Chloroformphase wird zur rohen Base eingeengt und diese mit äthanolischer Fumarsäure in die Titelverbindung mit dem Smp. 107 bis 109o (Zersetzung, aus absolutem Äthanol) überführt.
Das als Ausgangsprodukt benötigte 3-Chlor-5,6,7,8 tetrahydro-6-phenacetylpyrido[4,3-c]pyridazin wird analog Beispiel 1g aus 57,0 g 3-Chlor-5,6,7,8-tetrahydropyrido[4,3-c]pyridazin-maleinat und 46,6 g Phenylessigsäurechlorid hergestellt. Smp. 156 bis 1570 (Zersetzung, aus absolutem Äthanol).
Beispiel 14 6- [3 -(4-Chlorphenyl)propionyl] -3-hydrazino 5,6,7,8-tetrahydropyrido[4,3-c]pyridazin
Eine Aufschlämmung von 11,0 g 3-Chlor-6-[3-(4-chlor phenyl)propionyl]-5,6,7,8-tetrahydropyrido[4,3-c]- pyridazin in 175 ml Hydrazinhydrat wird in einem er Ölbad von 100o während 4 Stunden intensiv gerührt. Nach einer Reaktionszeit von 3'/2 Stunden erhält man eine vollständige Lösung.
Nach Kühlen mit Eis fällt das rohe Reaktionsprodukt aus. Es wird abfiltriert und die Titelverbindung aus absolutem Äthanol umkristallisiert, Smp. 150 bis 1530 (Zersetzung).
Das als Ausgangsmaterial benötigte 3-Chlor-6-[3 (4-chlorphenyl)propionyl]-5 ,6,7,8-tetrahydropyrido- [4,3-c]pyridazin kann folgendermassen hergestellt werden:
In eine Lösung von 28,6 g 3-Chlor-5,6,7,8-tetrahydro pyrido[4,3-c]pyridazin-maleinat in 150 ml Äthylenchlorid und 22,2 g Triäthylamin tropft man während 20 Minuten unter Rühren bei Zimmertemperatur eine Mischung von 20,3 g 4 Chlorphenylpropionsäurechlorid in 100 ml Äthylenchlorid.
Der Ansatz wird während 24 Stunden bei Zimmertemperatur weitergerührt, mit 100 ml 5%iger wässriger Natronlauge versetzt, weitere 2 Stunden gerührt und die ausgefallene Zwischenschicht abfiltriert. Die organische Phase wird abgetrennt, über Natriumsulfat getrocknet, filtriert und im Vakuum eingeengt. Die kristalline Zwischenschicht und der Rückstand der organischen Phase werden vereinigt und aus absolutem Alkohol umkristallisiert. Man erhält das analysenreine 3-Chlor-6-{3-(4-chlorphenyl)propionyl]- 5,6,7,8-tetrahydropyridol43-c]pyridazins Smp. 162 bis 164O (Zersetzung).

The 3-chloro-5,6,7,8 tetrahydro-6-o-toluoylpyrido [4,3-c] pyridazine required as starting material can be prepared as follows:
In a slurry of 28.5 g of 3-chloro-5,6,7,8 tetrahydropyrido [4,3-c] pyridazine maleate (preparation see Example 1, steps a to f) in 250 ml of ethylene chloride and 21.8 g Triethylamine is added dropwise to a mixture of 16.9 g of o-toluoyl chloride in 100 ml of ethylene chloride at room temperature over 18 minutes while stirring. The batch is stirred for a further 24 hours at the same temperature and worked up in the usual way. The 3-chloro-5,6,7,8-tetrahydro-6-o-toluoylpyrido [4,3-c] pyridazine obtained melts after recrystallization from 95% ethanol / ether at 123 to 1260 (decomposition).
Example 3 6-Benzoyl-5,6,7,8-tetrahydro-3- (1-methylhydrazino) pyrido [4,3-cjpyridazine bisgentisinate
A slurry of 13.7 g of 6-benzoyl-3-chloro 56.7, 8-tetrahydropyrido [4,3-c] pyridazine (preparation see Example 1, steps a to g) in 40 ml of methylhydrazine is at a bath temperature of 50O stirred for 6 '/ 4 hours.
A complete solution is obtained after a quarter of an hour.
The reaction solution is concentrated to an oil in vacuo.
This is dissolved in 200 ml of chloroform, dried over sodium sulfate, filtered and again concentrated to an oil. This is briefly boiled with 12.0 g of gentisic acid in 100 ml of methanol on a water bath, the solution is concentrated to about 2/3 and the precipitated material is filtered off after cooling with ice.
After recrystallization from absolute ethanol, the analytically pure compound mentioned in the title of mp.
166 to 1680 (decomposition).
Example 4 Bis [6- (o-chlorobenzoyl) -3 -hydrazino-5,6,7,8-tetrahydropyridote4,3 -c] pyridazine] trisfumarate
A slurry of 7.7 g of 3-chloro-6- (o-chlorobenzoyl) -5,6,7,8-tetrahydropyrido [4,3-c] pyridazine in 30 ml of hydrazine hydrate is at a bath temperature of 800 for 20 hours touched. After 11/4 hours, 25 ml of isopropanol are added to the reaction, after which a complete solution is formed. After the reaction has ended, the reaction solution is concentrated to give a dark brown foam. This is taken up in 200 ml of chloroform and extracted with 25 ml of water. The organic phase is separated off, dried over sodium sulfate, filtered and concentrated in vacuo to give a pale yellow foam.
This is dissolved in 50 ml of 95% ethanol, treated with 5.8 g of fumaric acid and briefly boiled on a water bath.
The crystalline fumarate is filtered off and recrystallized from 20 ml of 95% ethanol. The analytically pure compound mentioned in the title and having a melting point of 144 to 1160 (decomposition) is obtained.
The 3-chloro-6- (o-chlorobenzoyl) -5,6,7,8-tetrahydropyrido [4,3-cjpyridazine required as starting material can be obtained analogously to Example 1g from 28.5 g of 3-chloro-5,6,7 , 8-tetrahydropyrido [4,3-c] pyridazine maleate and 16.9 g - o-chlorobenzoyl chloride can be obtained. After recrystallization from 95 cXcigem ethanol 97 to 99o (decomposition).
Example 5 3-Hydrazino-5,6,7,8-tetrahydro-6- (3-phenylpropionyl) pyrido [4,3-cjpyridazine
A slurry of 8.5 g of 3-chloro-5,6,7,8-tetra hydro-6- (3-phenylpropionyl) pyrido [4,3-c] pyridazine and 80 ml of hydrazine hydrate is at a bath temperature of 800 for 4 Stirred for hours. After 31/2 hours a complete solution is obtained, which is cooled with ice after the reaction has ended. The precipitated substance is filtered off and recrystallized from 95% ethanol. The analytically pure product named in the title obtained in this way has a melting point of 162 to 1650 (decomposition).
The 3-chloro-5,6,7,8-tetrahydro-6- (3-phenylpropionyl) -pyrido [4,3-c] pyridazine required as starting material can be prepared analogously to Example 1g from 28.6 g of 3-chloro-5, 6,7,8 tetrahydropyrido [4,3-pyridazine maleate and 31.5 g of 3-phenylpropionyl chloride are obtained. After recrystallization from 956 Scigem ethanol 134 to 137 ".
Example 6 Bis [6- (p-tert-butylbenzoyl) -3-hydrazino-5,6,7,8-tetrahydropyrido [4,3-c] pyridazine] trisfumarate
A slurry of 12.0 g of 6- (p-tert-butylbenzoyl) 3-chloro-5,6,7,8-tetrahydropyrido [4,3-c] pyridazine in 75 ml
Hydrazine hydrate is stirred at a bath temperature of 100o for 4 hours. The batch is cooled with ice and the crystalline material is filtered off. The crude product is dissolved at the boil together with 7.2 g of fumaric acid in 50 ml of methanol and gives the analytically pure product named in the title with a melting point of 115 to 1180 (decomposition).
The 6- (p-tert-butylbenzoyl) -3-chloro-5,6,7,8-tetrahydropyrido [4,3-c] pyridazine required as starting material can be obtained analogously to Example 1g from 34.2 g of 3-chloro-5 , 6,7,8-tetrahydropyrido [4,3-c] pyridazine maleate and 25.0 g of tert-butylbenzoyl chloride are obtained. After recrystallization from 95% ethanol 157 to 1600 (decomposition).
Example 7 3-Hydrazino-5,6,7,8-tetrahydro-6 (p -toluoyl) pyrido [4,3-c] pyridazine
A slurry of 12.0 g of 3-chloro-5,6,7,8-tetrahydro-6- (p-toluoyl) pyrido [4,3-c] pyridazine and 50 ml of hydrazine hydrate is at a bath temperature of 100 ° for 2 hours touched. When the reaction has ended, the mixture is cooled and the undissolved material is filtered off. After the crude product has been recrystallized once from a mixture of 100 ml of 95% ethanol and 50 ml of methanol, the analytically pure product is obtained. Example 8 6- (m-Chlorobenzoyl) -3-hydrazino-5,6,7,8-tetrahydropyrido [4,3 -c] pyridazine fumarate
An initial slurry of 10.8 g of 3-chloro-6 (m-chlorobenzoyl) -5,6,7,8-tetrahydropyrido [4,3-cj-pyridazine and 50 ml of hydrazine hydrate is at a bath temperature of 100 ° for a total of 4 hours touched. After a reaction time of 2 hours, 25 ml of isopropanol are added to the suspension, and after 4 hours a complete solution has formed. The batch is concentrated to a crystalline residue in vacuo. A slurry of 10.2 g of 6- (p-bromobenzoyl) -3 chloro-5,6,7,8-tetrahydropyrido [4,3-c] pyridazine and 55 ml of hydrazine hydrate is stirred at a bath temperature of 100 ° for 2 hours. There is never a complete one

Example 10 3 -Hydrazino-5,6,7,8-tetrahydro-6- (p-methoxybenzoyl) -pyrido- [4,3-c] pyridazine
A slurry of 10.5 g of 3-chloro-5,6,7,8-tetra hydro-6- (p-methoxybenzoyl) -pyrido [4,3-c] pyridazine in 100 ml of hydrazine hydrate is at a bath temperature of 100o during stirred for a total of 1 hour. After a reaction time of 50 minutes, a complete solution is formed, which is cooled with ice. The precipitated reaction product is filtered off and recrystallized once from 15 ml of 95% strength ethanol. The analytically pure compound mentioned in the title and having a melting point of 169 to 1720 (decomposition) is obtained.
The 3-chloro-5,6,7,8 tetrahydro-6- (p-methoxybenzoyl) pyridol [4,3-cjpyridazine required as starting material can be prepared analogously to Example 1g from 28.5 g 3-chloro-5,6,7, 8 tetrahydropyrido [4,3-c] pyridazine maleate and 17.1 g of p-methoxybenzoyl chloride are obtained, mp 163 to 1640 after recrystallization from 95% ethanol.
Example 11 6- (o-Fluorobenzoyl) -3-hydrazino-5,6,7,8-tetrahydropyrido [4,3 -c] pyridazine
A slurry of 11.6 g of 3-chloro-6- (o-fluoro benzoyl) -5,6,7,8-tetrahydropyrido [4,3-cjpyridazine in 80 ml of hydrazine hydrate is at a bath temperature of 100o for 3 '/ 2 Stirred for hours. The batch is cooled with ice and the solid material is filtered off. After the crude product has been recrystallized once from a solvent mixture of 100 ml of 95% strength ethanol and 100 ml of methanol, the analytically pure compound mentioned in the title with a melting point of 210 to 2130 (decomposition) is obtained.
The 3-chloro-6 (o-fluorobenzoyl) -5,6,7,8-tetrahydropyrido [4,3-c] - pyridazine required as starting material can be obtained analogously to Example 1g from 28.6 g of 3-chloro 5,6,7 , 8-tetrahydropyrido [4,3-c] pyridazine maleate and
17.4 g of o-fluorobenzoyl chloride are obtained. After crystallization and recrystallization from 95% ethanol
144 to 1460 (decomposition).
Example 12 3-hydrazino-5,6,7,8-tetrahydro-6- (o-methylphenacetyl) pyrido [4,3 -c] pyridazine
An initial slurry of 10.0 g of 3-chloro 5, 6,7,8-tetrahydro-6- (o-methylphenacetyl) pyrido- [4,3-c] pyridazine in 35 ml of hydrazine hydrate is at a bath temperature of 100o for 30 Minutes stirred. During this time a complete solution is created. The reaction solution is allowed to cool slowly and the reaction product which has precipitated out during this time is filtered off. After recrystallizing once from absolute ethanol, the analytically pure compound mentioned in the title with the
M.p. 155 to 1570 (decomposition).
The 3-chloro-5,6,7,8tetrahydro-6 - (o-methylphenacetyl) pyrido [4,3-c] pyridazine required as starting material can be prepared analogously to Example 1g from 28.6 g of 3-chloro 5,6,7, 8-tetrahydropyrido [4,3-c] pyridazine maleate and 20.2 g of o-methylphenacetyl chloride are obtained. M.p.
after crystallization and recrystallization from 95% ethanol 125 to 1270 (decomposition).
Example 13 Bis [3-hydrazino-5,6,7,8-tetrahydro-6 phenacetylpyrido [4,3 -c] pyridazine] trisfumarate
A slurry of 9.7 g of 3-chloro-5,6,7,8-tetrahydro-6-phenacetylpyrido [4,3-c] pyridazine in 35 ml of hydrazine hydrate and 35 ml of isopropyl alcohol is added for 1 hour 30 minutes at a bath temperature stirred from 100o under reflux. The batch is concentrated in vacuo to a red oil and this is partitioned between chloroform and water. The chloroform phase is concentrated to the crude base and this is converted with ethanolic fumaric acid into the title compound with a melting point of 107-109 (decomposition, from absolute ethanol).
The 3-chloro-5,6,7,8 tetrahydro-6-phenacetylpyrido [4,3-c] pyridazine required as the starting product is prepared analogously to Example 1g from 57.0 g of 3-chloro-5,6,7,8-tetrahydropyrido [4,3-c] pyridazine maleate and 46.6 g of phenylacetic acid chloride. M.p. 156 to 1570 (decomposition, from absolute ethanol).
Example 14 6- [3 - (4-Chlorophenyl) propionyl] -3-hydrazino 5,6,7,8-tetrahydropyrido [4,3-c] pyridazine
A slurry of 11.0 g of 3-chloro-6- [3- (4-chlorophenyl) propionyl] -5,6,7,8-tetrahydropyrido [4,3-c] - pyridazine in 175 ml of hydrazine hydrate is in a he 100o oil bath was intensively stirred for 4 hours. A complete solution is obtained after a reaction time of 3½ hours.
After cooling with ice, the crude reaction product precipitates. It is filtered off and the title compound is recrystallized from absolute ethanol, m.p. 150 to 1530 (decomposition).
The 3-chloro-6- [3 (4-chlorophenyl) propionyl] -5,6,7,8-tetrahydropyrido- [4,3-c] pyridazine required as starting material can be prepared as follows:
A solution of 28.6 g of 3-chloro-5,6,7,8-tetrahydro pyrido [4,3-c] pyridazine maleate in 150 ml of ethylene chloride and 22.2 g of triethylamine is added dropwise over the course of 20 minutes while stirring Room temperature a mixture of 20.3 g of 4 chlorophenylpropionic acid chloride in 100 ml of ethylene chloride.
The batch is stirred for a further 24 hours at room temperature, 100 ml of 5% aqueous sodium hydroxide solution are added, the mixture is stirred for a further 2 hours and the precipitated intermediate layer is filtered off. The organic phase is separated off, dried over sodium sulfate, filtered and concentrated in vacuo. The crystalline intermediate layer and the residue of the organic phase are combined and recrystallized from absolute alcohol. The analytically pure 3-chloro-6- {3- (4-chlorophenyl) propionyl] -5,6,7,8-tetrahydropyridol43-c] pyridazine melting point 162 to 1640 (decomposition) is obtained.
