Die Erfindung betrifft ein Verfahren zur Herstellung neuer Verbindungen der Formel I, worin n eine ganze Zahl von 0 bis 2 bedeutet, R1 für eine gegebenenfalls durch Fluor, Chlor, Brom, Methoxy oder niederes Alkyl monosubstituierte Phenylgruppe steht, R2 Wasserstoff oder Methyl bedeutet und R3 und R4 je für niederes Alkyl stehen oder zusammen mit dem Kohlenstoffatom, an das sie gebunden sind, einen Cycloalkylidenrest mit 5 bis 8 Kohlenstoffatomen bilden, und ihrer Säureadditionssalze.
Erfindungsgemäss gelangt man zu den neuen Verbindungen der Formel I und ihren Säureadditionssalzen, indem man Verbindungen der Formel Ia, worin Ri, R2 und n obige Bedeutung besitzen, mit Verbindungen der Formel XI, worin R3 und R4 obige Bedeutung besitzen, umsetzt und die erhaltenen Verbindungen der Formel I gewünschtenfalls in ihre Säureadditionssalze überführt.
Falls R1 einen durch eine niedere Alkylgruppe substituierten Phenylrest darstellt, so besteht diese vorzugsweise aus 1 bis 4 Kohlenstoffatomen. Die durch R3 und R4 symbolisierten Alkylgruppen enthalten vorzugsweise 1 bis 4 Kohlenstoffatome und stellen insbesondere Methylgruppen dar.
Beispielsweise geht man so vor, dass man eine Verbindung der Formel Ia mit einer Verbindung der Formel XI, gegebenenfalls unter Zusatz von einem unter den Reaktionsbedingungen inerten, vorzugsweise polaren organischen Lösungsmittel wie einem niederen Alkohol, z.B. Methanol, Äthanol oder Isopropanol oder einem offenkettigen oder cyclischen Äther, z.B. Diäthylenglykoldimethyläther, Tetrahydrofuran oder Dioxan, nötigenfalls unter Erwärmen reagieren lässt, indem man das Reaktionsgemisch '/2bis 20 Stunden bei Temperaturen von ca. 06 bis Siedetemperatur hält und anschliessend zur Trockne eindampft oder das Rohprodukt direkt oder nach weitgehendem Einengen der Lösung auskristallisieren lässt. Die erhaltenen Verbindungen der Formel I können nach an sich bekannten Methoden isoliert und gereinigt werden.
Die freien Basen lassen sich in üblicher Weise in ihre Säureadditionssalze überführen und umgekehrt.
Die Ausgangsverbindungen können beispielsweise wie folgt hergestellt werden:
Verbindungen der Formel Ia können erhalten werden, indem man Verbindungen der Formel IIa, worin Rl und n obige Bedeutung besitzen und X für Chlor, Brom, die Mercapto- oder eine - SRs-Gruppe, worin Rs Benzyl oder niederes Alkyl bedeutet, steht, mit Verbindungen der Formel III, worin R2 obige Bedeutung besitzt, umsetzt.
Vorzugsweise verwendet man einen Überschuss der Verbindung der Formel III, beispielsweise 5 bis 10 Mol einer Verbindung der Formel III, bezogen auf 1 Mol einer Verbindung der Formel IIa, oder man arbeitet in Gegenwart eines anderen basischen Mittels, welches allfällige bei der Umsetzung entstehende Säure zu binden vermag, z.B. eines tertiären Amines oder eines Alkalimetall- oder Erdalkalimetallhydroxydes oder -karbonates umsetzt. Die Reaktion kann beispielsweise in Gegenwart von einem unter den Reaktionsbedingungen inerten. vorzugsweise polaren organischen Lösungsmittel, z.B.
einem niederen Alkohol wie Methanol, Äthanol oder Isopropanol oder einem offenkettigen oder cyclischen Äther wie Dioxan, Diäthylenglykoldimethyläther oder Tetrahydrofuran ausgeführt werden. Gegebenenfalls kann auch ein berschuss der Verbindung der Formel III als Lösungsmittel dienen.
Falls Verbindungen der Formel IIb, worin X' für Chlor, Brom oder die Mercaptogruppe steht und R1 und n obige Bedeutung besitzen, eingesetzt werden, kann die Umsetzung beispielsweise durch Erhitzen der Verbindungen der Formel IIb in Hydrazinhydrat oder Methylhydrazin, gegebenenfalls unter Zusatz eines der oben genannten Lösungsmittel und/oder basischen Kondensationsmittel bei Normaldruck bei Temperaturen zwischen ca. 20 und ca. 150% vorzugsweise bei Temperaturen zwischen 80 und 120 bzw. bei Siedetemperatur des Reaktionsgemisches. erfolgen und die Reaktionszeit kann zwischen 1 und 20 Stunden betragen.
Falls eine Verbindung der Formel IIc. worin R1. Rs und n obige Bedeutung besitzen, eingesetzt wird. wird die Umsetzung mit vorzugsweise einem Überschuss Hydrazinhydrat oder Methylhydrazin gegebenen falis unter Zusatz eines der oben genannten Lösungsmittel und'oder basischen Kondensationsmittel im Autoklaven bei Temperaturen zwischen 80 und 150 durchgeführt.
Die Verbindungen der Formel IId. worin Rl und n obige Bedeutung besitzen und X" für Chlor oder Brom steht, können erhalten werden, indem man Verbindungen der Formel IIe, worin X" obige Bedeutung besitzt, mit einer Verbindung der Formel IV. worin Rl. n und Xll obige Bedeutung besitzen.
in einem unter den Reaktionsbedingungen inerten Lösungsmittel, z.B. einem chlorierten Kohlenwasserstoff, wie Chloroform oder Äthylenchlorid. in Gegenwart eines säurebindenden Mittels, z.B. eines Alkalimetallkarbonates oder einer tertiären Base wie Triäthylamin. umsetzt.
Die Verbindungen der Formel IIe können erhalten werden, indem man die Verbindung der Formel Va mit einem geeigneten Chlorierungs- bzw. Bromierungsmittel. z.B. mit Phosphor oxychlorid, Phosphortri- oder -pentachlorid oder Phosphoroxybromid, erhitzt, vorzugsweise auf Temperaturen bis zu ca.
100 bzw. auf Siedetemperatur des Reaktionsgemisches.
Verbindungen der Formel lif, worin R1 und n obige Bedeutung besitzen. können z.B. erhalten werden, indem man Verbindungen der Formel IId mit Thioharnstoff oder gegebenenfalls auch Natriumsulfid umsetzt. Die Reaktion erfolgt vorzugsweise in einem polaren organischen Lösungsmittel, z.B.
einem niederen Alkohol, wie Athanol oder Dimethylformamid, bei Temperaturen zwischen ca. 30 und ca. 100 und kann zwischen ca. '. und 4 Stunden dauern.
Verbindungen der Formel IIc können beispielsweise erhalten werden, indem man Verbindungen der Formel IIf mit Verbindungen der Formel XII. worin Rs und Xll obige Bedeutung besitzen, in Gegenwart eines säurebindenden Mittels, beispielsweise eines Erdalkali- oder Alkalikarbonates oder -hydroxides, wie z.B. Kaliumkarbonat. umsetzt. Die Umsetzung wird vorzugsweise in einem polaren organischen Lösungsmittel, welches ein gutes Lösungsvermögen für die Verbindungen der Formel IIf besitzt, beispielsweise in Dimethylformamid.

Die Verbindung der Formel Va wird z.B. erhalten, indem man in Verbindungen der Formel Vb, worin Rll niederes Alkoxy bedeutet, die Carbalkoxygruppe sauer, z.B. durch mehrstündiges Erhitzen in saurem Medium. beispielsweise in Salzsäure, oder alkalisch. z.B. durch Erhitzen mit einem Alkalimetallhydroxid. wie Kaliumhydroxid, in einem höhersiedenden Alkanol, wie n-Butanol. abspaltet.
Verbindungen der Formel V. worin Rl für niederes Alkoxy oder für eine Rl(CH2)n-gruppe, worin Rl und n obige Bedeutung besitzen, steht, können z.B. erhalten werden, indem man
Verbindungen der Formel VI, worin R' obige Bedeutung besitzt, in einem unter den Reaktionsbedingungen inerten Lösungsmittel. z.B. einem niederen Alkohol, wie Methanol, in Gegenwart von mindestens äquivalenten Mengen Eisessig mit
Hydrazinhydrat oder einem Salz des Hydrazins gegebenenfalls unter Inertgasatmosphäre durch Erhitzen auf Temperaturen zwischen 70 bis 110C. vorzugsweise auf Siedetemperatur des Reaktionsgemisches.
cyclisiert und die gebildeten Verbindungen der Formel VII, worin R, obige Bedeutung besitzt, oxydiert, vorzugsweise mit Brom unter Verwendung eines halogenierten Kohlenwasserstoffes. wie Chloroform oder Äthylenchlorid, als Lösungsmittel.
Verbindungen der Formel VI können beispielsweise erhalten werden, indem man Verbindungen der Formel VIII, worin Rl obige Bedeutung besitzt, mit einem sekundären Amin, vorzugsweise einem cyclischen Amin, wie z.B. Pyrrolidin, Morpholin oder Piperidin, vorzugsweise in einem unter den Reaktionsbedingungen inerten organischen Lösungsmittel, z.B.
einem aromatischen Kohlenwasserstoff, wie Benzol, gegebenenfalls unter Zusatz einer Katalysators, z.B. p-Toluolsulfonsäure oder eines Molekularsiebes, bei erhöhter Temperatur, vorzugsweise bei Siedetemperatur des Reaktionsgemisches, zu einem Enamin der Formel IX, worin Rl obige Bedeutung besitzt und Y für eine sekundäre Aminogruppe steht, umsetzt, dieses mit Bromessigsäurealkylester in Gegenwart eines unter den Reaktionsbedingungen inerten organischen Lösungsmittels, z.B.
eines aromatischen Kohlenwasserstoffes, wie Benzol, eines Halogenkohlenwasserstoffes, wie Chloroform, oder eines Äthers oder Dimethylformamid bei Raumtemperatur versetzt, das Reaktionsgemisch mehrere Stunden erhitzt, vorzugsweise auf Siedetemperatur, und in dem erhaltenen Reaktionsprodukt der Formel X, worin Rl und Y obige Bedeutung besitzen, die Enamingruppierung durch Erhitzen mit Wasser, gegebenenfalls unter Zugabe von verdünnter Lauge, Ammoniaklösung oder verdünnter Mineralsäure wieder spaltet.
Die Verbindungen der Formel I und ihre pharmakologisch verträglichen Säureadditionssalze sind in der Literatur bisher noch nicht beschrieben worden. Sie zeichnen sich durch interessante pharmakodynamische Eigenschaften aus und können daher als Heilmittel verwendet werden.
Wie sich bei Versuchen an der hypertonen Grollmann Ratte zeigt, besitzen die Substanzen antihypertensive Wirkungen.
Die zu verwendenden Dosen variieren naturgemäss je nach Art der Substanz, der Administration und des zu behandelnden Zustandes. Im allgemeinen werden jedoch bei Testtieren befriedigende Resultate mit einer Dosis von 0,1 bis 10 mg/kg Körpergewicht erhalten; diese Dosis kann nötigenfalls in 2 bis 3 Anteilen oder auch als Retardform verabreicht werden. Für grössere Säugetiere liegt die Tagesdosis bei etwa 5 bis 500 mg.
Für orale Applikationen enthalten die Teildosen etwa 2 bis 250 mg der Verbindungen der Formel I neben festen oder flüssigen Trägersubstanzen.
Aufgrund ihrer blutdrucksenkenden Wirkung können die Substanzen in der Hochdrucktherapie Anwendung finden.
Als Heilmittel können die Verbindungen der Formel I bzw.
ihre physiologisch verträglichen Säureadditionssalze allein oder in geeigneter Arzneiform mit pharmakologisch indifferenten Hilfsstoffen verabreicht werden.
Soweit die Herstellung der Ausgangsverbindungen nicht beschrieben wird, sind diese bekannt oder nach an sich bekannten Verfahren bzw. analog zu den hier beschriebenen oder analog zu an sich bekannten Verfahren herstellbar.
In den nachfolgenden Beispielen, die die Erfindung näher erläutern, ihren Umfang aber in keiner Weise einschränken sollen, erfolgen alle Temperaturangaben in Celsiusgraden.
Beispiel 1
6-Benzoyl-3 -isopropylidenhydrazino-5 ,6,7,8 tetrahydropyrido[4,3 -c]pyridazin
Eine Lösung von 0,5 g 6-Benzoyl-3-hydrazino-5,6,7,8tetrahydropyrido[4,3-c]pyridazin in 15 ml absolutem Aceton, dem 4 Tropfen Eisessig zugesetzt sind, wird 1 Stunde auf dem Wasserbad erhitzt. Die Lösung wird im Vakuum zur Trockne eingeengt und das verbleibende Öl mit Äther kristallisiert. Die rohe Verbindung wird aus absolutem Äthanol um kristallisiert und ergibt die analysenreine, im Titel genannte Verbindung vom Smp. 188-190o (Zersetzung).
Das als Ausgangssubstanz benötigte 6-Benzoyl-3-hydrazino 5,6,7 ,8-tetrahydropyrido[4,3 -cjpyridazin kann folgendermassen hergestellt werden: a) Eine Mischung von 342,6 g 1-Carbäthoxy-4-piperidon und 214,0 g Pyrrolidin in 800 ml Benzol werden während
2 Stunden unter Sieden am Rückfluss erhitzt. Das wäh rend dieser Zeit entstandene Wasser wird azeotrop aus der Reaktionsmischung kontinuierlich entfernt. Der
Ansatz wird zum orangen Öl eingeengt und dieses im
Hochvakuum destilliert. Als Hauptfraktion erhält man bei Kp 144-150q0,03 mm den analysenreinen 1,2,3,6 Tetrahydro-4-pyrrolidinylpydin- 1 -carbonsäureäthyl- ester.
b) Eine Mischung von 1460 g 1,2,3,6-Tetrahydro-4-pyrroli dinylpyridin- 1 -carbonsäureäthylester und 1250 g Brom essigsäureäthylester in 6,0 1 Benzol wird während 20
Stunden unter Rühren am Rückfluss erhitzt. Der Ansatz wird mit 2,0 1 Wasser versetzt und erneut 2 Stunden unter Kochen am Rückfluss erhitzt. Nach dem Abkühlen auf Zimmertemperatur setzt man 400 ml 10%gen
Ammoniak zu und trennt die Benzolphase ab. Diese wird mit Natriumsulfat getrocknet und nach dem Filtrieren zum Öl eingeengt. Die Hauptfraktion, der 1-Carbäthoxy
4-piperidon-3-essigsäureäthylester, siedet bei einem Kp.
145-170'/0,05 mm.
c) Eine Mischung von 514 g 1-Carbäthoxy-4-piperidon-3 essigsäureäthylester und 100 g Hydrazinhydrat in 1,5 1
95 %igem Äthanol und 200 ml Eis essig erhitzt man in einem Stickstoffstrom während 4 Stunden unter Rühren am Rückfluss. Danach wird der Ansatz vollständig ein geengt. Der kristallisierte Rückstand wird aus 1,0 1
95 %igem Äthanol umkristallisiert und ergibt den 2,3,4,4a,5,6,7,8-Octahydro-3-oxo-6- pyrido[4,3 -c]pyridazincarbonsäureäthylester vom Smp. 163-166 (Zersetzung).
d) 450,5 g 2,3,4,4a,5 ,6,7,8-0ctahydro-3-oxo-6-pyrido- [4,3 -c]pyridazincarbonsäureäthylester werden in 2,0 1
Chloroform gelöst und die Lösung auf Rückflusstempera tur erhitzt. In diese kochende Lösung gibt man innerhalb von 2'/2 Stunden 320 g Brom in 500 ml Chloroform. Der
Ansatz wird nach der Bromzugabe noch eine weitere
Stunde unter Rückfluss gerührt. Nach dem Abkühlen auf
Raumtemperatur wird mit 1 kg Eis/Wasser versetzt und der Chloroformteil abgetrennt. Der saure wässrige Teil wird mit weiteren 500 ml Chloroform ausgezogen. Die
Chloroformteile werden der Reihe nach mit 300 ml
Wasser gewaschen, über Natriumsulfat getrocknet und im
Vakuum eingeengt.
Der halbkristalline Rückstand wird mit 250 ml absolutem Äthanol umkristallisiert und ergibt das analysenreine 6-Carbäthoxy-5,6,7,8-tetrahydro-
3 (2H)pyrido[4,3 -c]pyridazinon,
Smp. 165-168 (Zersetzung).
e) Eine Lösung von 223,2 g 6-Carbäthoxy-5,6,7,8-tetra hydro-3(2H)pyrido[4,3-c]pyridazinon in 1,0 1 konzen- trierter Salzsäure wird während 22 Stunden unter Kochen am Rückfluss gerührt. Der Ansatz wird im Vakuum vollständig eingeengt, der kristalline Rückstand in Metha nol aufgenommen und abfiltriert. Das rohe Hydrochlorid des 5,6,7,8-Tetrahydro-3(2H)pyrido[4, 3-c]pyridazinon,
Smp. 3073100 (Zersetzung), wird in 0,75 1 Methanol aufgeschlämmt und 0,4 1 Triäthylamin langsam zugege ben. Die violette Aufschlämmung wird nach 15 Minuten
Rühren auf 30O gekühlt, abfiltriert und mit wenig Metha nol/Äther nachgewaschen.
Man erhält die Rohbase vom
Smp. 216-2190 (Zersetzung). 25,0 g Rohbase werden aus
300 ml Methanol, dem man 10 ml konzentrierten Ammo niak und 40 ml Wasser beimischt, unter Zusatz von wenig
Kohle umkristallisiert und ergeben das analysenreine 5,6,7,8-Tetrahydro-3(2H)pyrido[4,3-c]pyridazinon vom Smp. 223-2250 (Zersetzung).
f) Eine Lösung von 30,3 g 5,6,7,8-Tetrahydro-3(2H)pyrido [4,3-cjpyridazinon in 250 ml Phosphoroxychlorid wird während 2 Stunden unter Kochen am Rückfluss gerührt.
Der Ansatz wird im Vakuum vollständig eingeengt und zum kalten Rückstand werden 500 g Eis/Wasser gegeben.
Die saure, erneut gekühlte Lösung wird in ein Gemisch aus 500 ml Chloroform und 250 ml konzentriertem
Ammoniak langsam hineingerührt. Die wässrige
Phase wird mit vier weiteren Portionen zu je 250 ml
Chloroform, total 1,5 l Chloroform, extrahiert, diese
Chloroformteile über Natriumsulfat getrocknet und im
Vakuum eingeengt. Man erhält die rohe, unstabile 3 Chlor-5,6,7,8-tetrahydropyrido[4,3-c]pyridazin-Base, die zur Weiterverarbeitung in das Maleinat übergeführt wird.
Dazu löst man 24,8 g Base in 150 ml Methanol und gibt
17,5 g Maleinsäure zu. Nach dem Aufkochen, Filtrieren,
Abkühlen erhält man das rohe Maleinat der Verbindung vom Smp. 156-1590 (Zersetzung). Eine Probe davon wird aus Methanol unter Zusatz von wenig Kohle umkristalli siert und ergibt die analysenreine Verbindung vom Smp.
162-1640 (Zersetzung).
g) In eine Aufschlämmung von 25,6 g 3-Chlor-5,6,7,8-tetra hydropyrido[4,3-c]pyridazin-maleinat in 250 ml Äthylen chlorid und 21,8 g Triäthylamin tropft man während 18
Minuten unter Rühren bei Zimmertemperatur eine
Mischung von 12,6 g Benzoylchlorid in 100 ml Äthylen chlorid. Der Ansatz wird während 14 Stunden bei der gleichen Temperatur weitergerührt, mit 200 ml Wasser versetzt und die organische Phase abgetrennt. Sie wird über Natriumsulfat getrocknet, filtriert und im Vakuum zu einem Öl eingeengt. Beim Versetzen dieses Öles mit ÄtherlDimethoxyäthan erhält man das rohe 6-Benzoyl 3-chlor-5 ,6,7,8-tetrahydropyrido[4,3-c]pyridazin. Dieses wird aus absolutem Alkohol unter Zusatz von wenig
Kohle umkristallisiert und ergibt die analysenreine Ver bindung vom Smp. 125-127 (Zersetzung).
h) Eine Aufschlämmung von 21,6 g 6-Benzoyl-3-chloro-5,6, 7,8-tetrahydropyrido[4,3-c]pyridazin in 80 ml Hydrazin hydrat wird bei einer Badtemperatur von 110O während 1
Stunde unter gelindem Kochen am Rückfluss gerührt. Nach ca. 15 Minuten Reaktionszeit erhält man eine vollständige Lösung. Der Ansatz wird mit Eis gekühlt und die ausgefallene Substanz abfiltriert und mit wenig absolutem Äthanol nachgewaschen. Das Rohpro dukt wird in 60 ml Dimethylformamid gelöst, filtriert und das Filtrat mit 60 ml absolutem Äthanol versetzt, worauf die Verbindung auskristallisiert. Man filtriert das analy senreine
6-Benzoyl-3-hydrazino-5,6,7,8 tetrahydropyrido[4,3-c]pyridazin ab. Es hat einen Smp. von 220-2230 (Zersetzung).
Beispiel 2 6-Benzoyl-3 -(2-butylidenhydrazino)-5,6,7.8- tetrahydropyrido[3.3-c]pyridazin
Eine Aufschlämmung von 13,5 g 6-Benzoyl-3-hydrazino 5,6,7,8-tetrahydropyrido[1,3-c]pyridazin in 100 ml Methyl äthylketon wird während 4 Stunden unter Rühren am Rückfluss erhitzt. Während dieser Reaktionszeit beobachtet man eine deutliche Umfällung. Nach gutem Kühlen mit Eis wird das feste Material abfiltriert. Nach einmaligem Umkristallisieren aus 95 %igem Äthanol erhält man die analysenreine im Titel genannte Substanz vom Smp. 191-193 (Zersetzung).
Beispiel 3
6-Benzoyl-3-cyclohexylidenhydrazino-
5,6'7,8-tetrahydropyrido[4,3-cjpyridazin
Eine Aufschlämmung von 8,3 g 6-Benzoyl-3-hydrazino 5 ,6,7,8-tetrahydropyrido[4 3 -cjpyridazin in 20 ml Cyclohexan wird bei einer Badtemperatur von 180 während 2 Stunden unter Rückfluss gekocht. Während des Aufheizens entsteht eine vollständige Lösung. Der Ansatz wird im Vakuum zum halbkristallinen Rückstand eingeengt, mit 100 ml Äther versetzt und das kristalline Reaktionsprodukt abfiltriert. Nach einmaligem Umkristallisieren aus Acetonitril erhält man die analysenreine, im Titel genannte Verbindung vom Smp. 188 191' (Zersetzung).
Beispiel 4 3-Isopropylidenhydrazino-6-phenacetyl-
5,6*7,8-tetrahydropyrido[4,3-c]pyridazin
9,6 g rohes öliges 3-Hydrazino-6-phenacetyl-5,6,7.8- tetrahydropyrido[4.3-c]pyridazin werden mit 20 ml Aceton 15 Minuten auf dem Wasserbad erhitzt. Nach ca. 5 Minuten beginnt aus der Reaktionslösung festes Material auszufallen.
Dieses wird nach dem Kühlen abfiltriert und ergibt nach dem Umkristallisieren aus Methanol unter Zusatz von wenig Kohle die analysenreine Titelverbindung vom Smp. 1951960 (Zersetzung).
Das Ausgangsmaterial kann analog Beispiel 1. Stufen g und h. hergestellt werden: a) 3-Chlor-6-phenacetyl-5.k.',9-tetrahydropyrido[4.3-c]- pyridazin: aus 57,0 g 3-Chlor-5*6.7..9-tetr2h .dropyrido- [4,3-c]pyridazin-maleinat und 46.6 g Phenylessigsäure- chlorid, Smp. 156-157 (Zersetzung. aus absolutem Äthanol).
b) 3-Hvdrazino-6-phenacetv!-5.fi.78-tetrahydropyrido- [4,3-c]pyridazin: aus 19.5 g 3-Chlor-6-phenacetyl-
5,6,7,8 tetrahydropyrido[4,3-cjpyridizin in 70 ml Hydrazinhy drat und 70 ml Isopropanol. Badtemperatur von 100',
Reaktionszeit 5 Stunden. Das nach Aufarbeitung erhal tene Rohprodukt wird direkt weiterverarbeitet.
Beispiel 5
6-(p-Chlorbenzoyl)-3 -isopropylidenhydrazino 5,6,7,8-tetrahydropyrido[4,3-c]pyridazin

Zur Aufarbeitung wird mit 100 ml 3 %iger wässriger
Natronlauge versetzt und nach weiteren 2 Stunden die organische Phase abgetrennt, getrocknet, und im Vakuum eingeengt. Smp. 170-1720 (Zersetzung, aus absolutem Äthanol).
b) 6-(p-Chlorbenzoyl) -3-hydrazino-5,6,7,8-tetrahydro- pyrido [4,3-c]pyridazin: aus 15,4 g 3-chlor-6-(p-chlorbenzoyl)-
5,6,7,8-tetrahydropyrido[4,3-c]pyridazin in 50 ml Hydra zinhydrat und 40 ml Dioxan. Reaktionszeit 9l/2 Stunden bei einer Badtemperatur von 800 und 7 Stunden bei

Beispiel 6 3-Cyclohexylidenhydrazino-5,6,7,8 - tetrahydro-6-(p-toluoyl)pyrido[4,3 -c]pyridazin
Eine anfängliche Aufschlämmung von 1,0 g 3-Hydrazino5,6,7,8 tetrahydro-6-(p-toluoyl)pyrido[4,3-cjpyridazin in 10 ml Cyclohexanon wird in einem Ölbad von 180 während 1 Stunde unter Rückfluss erhitzt. Die entstandene Reaktionslösung wird im Vakuum vollständig eingeengt und das kristalline Rohprodukt aus 10 ml 95 O/oigem Äthanol umkristallisiert. b) 3-Hydrazino-5,6,7,8-tetrahydro-6-(p-toluoyl)pyrido- [4,3-c]pyridazin: hergestellt analog Beispiel Ih aus 12,0 g 3-Chlor-5,6,7,8-tetrahydro-6-(p-toluoyl)pyridor4,3-clpy- ridazin und 50 ml Hydrazinhydrat;

An initial slurry of 1.0 g of 3-hydrazino5,6,7,8 tetrahydro-6- (p-toluoyl) pyrido [4,3-cjpyridazine in 10 ml of cyclohexanone is refluxed in a 180 oil bath for 1 hour. b) 3-hydrazino-5,6,7,8-tetrahydro-6- (p-toluoyl) pyrido- [4,3-c] pyridazine: prepared analogously to Example Ih from 12.0 g of 3-chloro-5,6 , 7,8-tetrahydro-6- (p-toluoyl) pyridor4,3-clpyridazine and 50 ml of hydrazine hydrate;

Example 7 6- (o-Fluorobenzoyl) -5,6,7,8-tetrahydro-3-isopropylidenehydrazinopyrido [4,3-c] pyridazine
An initial slurry of 1.0 g of 6- (o-fluorobenzoyl) 3-hydrazino-5,6,7,8-tetrahydropyrido [4,3-c] pyridazine in 10 ml of acetone is refluxed on the water bath for 30 minutes . The gradually formed reaction solution is concentrated in vacuo and the crystalline crude product is recrystallized once from 15 ml of absolute ethanol. The analytically pure compound named in the title has a smp.
