Gegenstand des Hauptpatentes ist ein Verfahren zur Herstellung von Pyrido[4,3-c]pyridazin-Derivaten der Formel A, worin n eine ganze Zahl von 0 bis 2 bedeutet, R für eine gegebenenfalls durch Fluor, Chlor, Brom, Methoxy oder niederes Alkyl monosubstituierte Phenylgruppe steht, R2 Wasserstoff oder Methyl bedeutet und R3 und R4 je für niederes Alkyl stehen oder zusammen mit dem Kohlenstoffatom, an das sie gebunden sind, einen Cycloalkylidenrest mit 5 bis 8 Kohlenstoffatomen bilden.
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung neuer Pyrido[4.3-c]pyridazin-Derivate der Formel I, worin Rl eine niedere Alkyl- oder Phenylalkylgruppe bedeutet, R2 Wasserstoff oder Methyl bedeutet und R3 und R4 je für niederes Alkyl stehen oder zusammen mit dem Kohlenstoffatom. an das sie gebunden sind, einen Cycloalkylidenrest mit 5 bis 8 Kohlenstoffatomen bilden, und ihrer Säureadditionssalze.
Erfindungsgemäss gelangt man zu den neuen [4,3-c]pyridazin-Derivaten der Formel I und ihren Säureadditionssalzen, indem man Verbindungen der Formel Ia, worin Rl und R2 obige Bedeutung besitzen, mit Verbindungen der Formel IV. worin R3 und R4 obige Bedeutung besitzen, umsetzt und die erhaltenen Verbindungen der Formel I gewünschtenfalls in ihre Säureadditionssalze überführt.
Falls R, eine niedere Alkylgruppe bedeutet, so besteht diese vorzugsweise aus 1 bis 4 Kohlenstoffatomen, falls Rl eine Phenylalkylgruppe bedeutet, so enthält deren Alkylkette vorzugsweise 1 bis 4 Kohlenstoffatome. Die durch R3 und R4 symbolisierten Alkylgruppen enthalten vorzugsweise 1 bis 4 Kohlenstoffatome und stellen insbesondere Methylgruppen dar.
Beispielsweise geht man so vor, dass man eine Verbindung der Formel Ia mit einer Verbindung der Formel IV, gegebenenfalls unter Zusatz von einem unter den Reaktionsbedingungen inerten, vorzugsweise polaren organischen Lösungsmittel wie einem niederen Alkohol, z. B. Methanol, Äthanol oder Isopropanol oder einem offenkettigen oder cyclischen Äther, z. B. Diäthylenglykoldimethyläther, Tetrahydrofuran oder Dioxan, nötigenfalls unter Erwärmen reagieren lässt, indem man das Reaktionsgemisch 1/2bis 20 Stunden bei Temperaturen von ca. 09 bis Siedetemperatur hält und anschliessend zur Trockne eindampft oder das Rohprodukt direkt oder nach weitgehendem Einengen der Lösung auskristallisieren lässt.
Die erhaltenen Verbindungen der Formel I können nach an sich bekannten Methoden isoliert und gereinigt werden. Die freien Basen lassen sich in üblicher Weise in ihre Säureadditionssalze überführen und umgekehrt.
Die Ausgangsverbindungen können beispielsweise wie folgt hergestellt werden:
Verbindungen der Formel Ia können erhalten werden, indem man Verbindungen der Formel IIa, worin Rl obige Bedeutung besitzt und X für Chlor, Brom, die Mercapto- oder eine-SRs-Gruppe, worin Rs Benzyl oder niederes Alkyl bedeutet. steht, mit Verbindungen der Formel III, worin R2 obige Bedeutung besitzt, umsetzt.
Vorzugsweise verwendet man einen Überschuss der Verbindung der Formel III, beispielsweise 5 bis 10 Mol einer Verbindung der Formel III bezogen auf 1 Mol einer Verbindung der Formel IIa, oder man arbeitet in Gegenwart eines anderen basischen Mittels, welches allfällige bei der Umsetzung entstehende Säure zu binden vermag, z. B. eines tertiären Amines oder eines Alkalimetall- oder Erdalkalimetallhydroxydes oder -karbonates umsetzt. Die Reaktion kann beispielsweise in Gegenwart von einem unter den Reaktionsbedingungen inerten, vorzugsweise polaren organischen Lösungsmittel, z. B.
einem niederen Alkohol wie Methanol, Äthanol oder Isopropanol oder einem offenkettigen oder cyclischen Äther wie Dioxan, Diäthvlenglykoldimethyläther oder Tetrahydrofuran ausgeführt werden Gegebenenfalls kann auch ein Überschuss der Verbindung der Formel III als Lösungsmittel dienen.
Falls Verbindungen der Formel IIb, worin X' für Chlor, Brom oder die Mercaptogruppe steht und Rl obige Bedeutung besitzt, eingesetzt werden, kann die Umsetzung beispielsweise durch Erhitzen der Verbindungen der Formel IIb in Hydrazinhydrat oder Methylhydrazin, gegebenenfalls unter Zusatz eines der oben genannten Lösungsmittel und/oder basischen Kondensationsmittel bei Normaldruck bei Temperaturen zwischen ca. 20 und ca. 1500, vorzugsweise bei Temperaturen zwischen 80 und 1200 bzw. bei Siedetemperatur des Reaktionsgemisches, erfolgen und die Reaktionszeit kann zwischen 1 und 20 Stunden betragen.
Falls eine Verbindung der Formel IIc, worin Rl und Rs obige Bedeutung besitzen, eingesetzt wird, wird die Umsetzung mit vorzugsweise einem Überschuss Hydrazinhydrat oder Methylhydrazin gegebenenfalls unter Zusatz eines der oben genannten Lösungsmittel und/oder basischen Kondensationsmittel im Autoklaven bei Temperaturen zwischen 80 und 1500 durchgeführt.
Verbindungen der Formel IId, worin Rl obige Bedeutung besitzt und X" für Chlor oder Brom steht, können z. B. erhalten werden, indem man Verbindungen der Formel IIe, worin Rl obige Bedeutung besitzt, mit einem geeigneten Chlorierungs- bzw. Bromierungsmittel, z. B. mit Phosphoroxychlorid, Phosphortri- oder -pentachlorid oder Phosphoroxybromid, erhitzt, vorzugsweise auf Temperaturen bis zu ca. 100 bzw.
auf Siedetemperatur des Reaktionsgemisches.
Verbindungen der Formel IIf, worin Rl obige Bedeutung besitzt und Xlll für Chlor, Brom, die Hydroxy- oder Mercaptogruppe steht, können z. B. erhalten werden, indem man Verbindungen der Formel IIg, worin XXli obige Bedeutung besitzt, mit Verbindungen der Formel V, worin Rl obige Bedeutung besitzt, in einem unter den Reaktionsbedingungen inerten Lösungsmittel, z. B. einem chlorierten Kohlenwasserstoff wie Chloroform oder Äthylenchlorid, in Gegenwart eines säurebindenden Mittels, z.
B. eines Alkalimetallkarbonates oder einer tertiären Base wie Triäthylamin, umsetzt, und im Falle, dass X'11Hydroxy- oder Mercaptogruppen bedeutet, die als Nebenprodukt entstehenden Phenol- resp.Thiophenolester unter schwach alkalischen oder sauren Bedingungen wieder verseift.
Verbindungen der Formel IIh, worin Rl obige Bedeutung besitzt. können z. B. erhalten werden, indem man Verbindungen der Formel IId mit Thioharnstoff oder gegebenenfalls auch Natriumsulfid umsetzt. Die Reaktion erfolgt vorzugsweise in einem polaren organischen Lösungsmittel. z. B. einem niederen Alkohol wie äthanol oder Dimethylformamid, bei Temperaturen zwischen ca. 30 und ca. 100o und kann zwischen ca. 1/2 und 4 Stunden dauern.
Verbindungen der Formel IIc können beispielsweise erhalten werden, indem man Verbindungen der Formel IIh mit Verbindungen der Formel VI, worin Rs und Xl' obige Bedeutung besitzen, in Gegenwart eines säurebindenden Mittels, beispielsweise eines Erdalkali- oder Alkalikarbonates oder -hydroxydes, wie z. B. Kaliumkarbonat, umsetzt. Die Umsetzung wird vorzugsweise in einem polaren organischen Lösungsmittel, welches ein gutes Lösungsvermögen für die Verbindungen der Formel IIh besitzt, beispielsweise in Dimethylformamid, durchgeführt.
Die Verbindung der Formel IIi kann beispielsweise hergestellt werden, indem man in Verbindungen der Formel IIe die RiO-CO-Gruppe sauer, z. B. durch mehrstündiges Erhitzen in saurem Medium, beispielsweise in Salzsäure, oder alkalisch, z. B. durch Erhitzen mit einem Alkalimetallhydroxyd wie Kaliumhydroxyd in einem höhersiedenden Alkanol wie z. B.
n-Butanol, abspaltet.
Verbindungen der Formel IIj, worin Xll obige Bedeutung besitzt, können aus der Verbindung der Formel IIi analog der Überführung von Verbindungen der Formel IIe in Verbindungen der Formel IId, die Verbindung der Formel IIk kann aus Verbindungen der Formel IIj analog der Überführung von Verbindungen der Formel lid in Verbindungen der Formel IIh gewonnen werden.
Verbindungen der Formel IIe können z. B. erhalten werden, indem man Verbindungen der Formel VII, worin Rl obige Bedeutung besitzt, in einem unter den Reaktionsbedingungen inerten Lösungsmittel, z. B. einem niederen Alkohol wie Äthanol in Gegenwart von mindestens äquivalenten Mengen Eisessig mit Hydrazinhydrat oder einem Salz des Hydrazins, gegebenenfalls unter Inertgasatmosphäre durch Erhitzen auf Temperaturen zwischen 70 bis 110o, vorzugsweise auf Siedetemperatur des Reaktionsgemisches, cyclisiert und die gebildeten Verbindungen der Formel VIII, worin Rl obige Bedeutung besitzt, oxydiert, vorzugsweise mit Brom unter Verwendung eines halogenierten Kohlenwasserstoffes wie Chloroform oder Äthylenchlorid als Lösungsmittel.
Verbindungen der Formel VII können erhalten werden, indem man Verbindungen der Formel IX, worin Rl obige Bedeutung besitzt, mit einem sekundären Amin, vorzugsweise einem cyclischen Amin wie z. B. Pyrolidin, Morpholin oder Piperidin, vorzugsweise in einem unter den Reaktionsbedingungen inerten organischen Lösungsmittel, z. B. einem aromatischen Kohlenwasserstoff wie Benzol, zu einem Enamin der Formel X, worin Rl obige Bedeutung besitzt und Y für eine sekundäre Aminogruppe steht, umsetzt, dieses mit Bromessigsäurealkylester in Gegenwart eines unter den Reaktionsbedingungen inerten organischen Lösungsmittels, z.
B. eines aromatischen Kohlenwasserstoffes wie Benzol, eines Halogenkohlenwasserstoffes wie Chloroform oder eines Äthers oder Dimethylformamid bei Raumtemperatur versetzt, das Reaktionsgemisch mehrere Stunden erhitzt, vorzugsweise auf Siedetemperatur, und in dem erhaltenen Reaktionsprodukt der Formel XI, worin R, und Y obige Bedeutung besitzen, die Enamingruppierung durch Erhitzen mit Wasser wieder spaltet.
Die Verbindungen der Formel I und ihre pharmakologisch verträglichen Säureadditionssalze sind in der Literatur bisher noch nicht beschrieben worden. Sie zeichnen sich durch interessante pharmakodynamische Eigenschaften aus und können daher als Heilmittel verwendet werden. Insbesondere besitzen sie antihypertensive Eigenschaften, wie sich bei Tierversuchen an der hypertonen Grollmann-Ratte (Methode nach A. Grollmann, [Proc. Soc. Exptl. Biol. and Med. 57, 104 (1944)] bei Dosen von 1 bis 10 mg/kg zeigt.
Aufgrund ihrer blutdrucksenkenden Wirkung können die Substanzen z. B. in der Hochdrucktherapie Anwendung finden. Die zu verwendenden Dosen variieren naturgemäss je nach Art der Substanz, der Administration und des zu behandelnden Zustandes. Im allgemeinen werden jedoch befriedigende Resultate mit einer Dosis von 0,5 bis 10 mg/kg Körpergewicht erhalten; diese Dosis kann nötigenfalls in 2 bis 4 Anteilen oder auch als Retardform verabreicht werden. Für grössere Säugetiere liegt die Tagesdosis bei etwa 30 bis 500 mg. So enthalten z. B. für orale Applikationen die Teildosen etwa 10 bis 250 mg der Verbindungen der Formel I neben festen oder flüssigen Trägersubstanzen.
Als Heilmittel können die Verbindungen der Formel I bzw.
ihre physiologisch verträglichen Säureadditionssalze allein oder in geeigneter Arzneiform mit pharmakologisch indifferenten Hilfsstoffen verabreicht werden.
Soweit die Herstellung der Ausgangsverbindungen nicht beschrieben wird, sind diese bekannt oder nach an sich bekannten Verfahren bzw. analog zu den hier beschriebenen oder analog zu an sich bekannten.Verfahren herstellbar.
In dem nachfolgenden Beispiel, das die Erfindung näher erläutern, ihren Umfang aber in keiner Weise einschränken soll, erfolgen alle Temperaturangaben in Celsiusgraden.
Beispiel 1 5 ,6,7,8-Tetrahydro-3-isopropylidenhydrazino-6- pyrido[,4,3-c]pyridazincarbonsäureäthylester
Eine Aufschlämmung von 23,7 g 3-Hydrazino-5,6,7,8 tetrahydro-6-pyrido[4,3-c]pyridazincarbonsäure- äthylester in 100 ml Aceton wird 4 Stunden unter Rühren am Rückfluss erhitzt. Während des Aufheizens geht das Ausgangsmaterial vollständig in Lösung. Der Ansatz wird mit Eis gekühlt und das ausgefallene Rohprodukt abfiltriert. Nach einmaligem Umkristallisieren aus Methanol unter Zusatz von wenig Kohle erhält man die im Titel genannte analysenreine Substanz vom Fp. 171-1740 (Zersetzung).
Der als Ausgangssubstanz benötigte 3-Hydrazino 5,6,7,8-tetrahydro-6-pyrido[4,3-c]pyridazincarbon- säureäthylester kann folgendermassen erhalten werden: a) Eine Mischung von 342,6 g 1-Carbäthoxy-4-piperidon und
214,0 g Pyrrolidin in 800 ml Benzol werden während 2
Stunden unter Sieden am Rückfluss erhitzt. Das während dieser Zeit entstandene Wasser wird azeotrop aus der
Reaktionsmischung kontinuierlich entfernt. Der Ansatz wird zum orangen Öl eingeengt und dieses im Hochvakuum destilliert. Als Hauptfraktion erhält man bei Kp 144-1500/
0,03 mm den analysenreinen 1,2,3 ,6-Tetrahydro-4-pyrroli- dinylpyridin- 1 -carbonsäureäthylester.
b) Eine Mischung von 1460 g 1,2,3,6-Tetrahydro-4 pyrrolidinylpyridin- 1 -carbonsäureäthylester und 1250 g
Bromessigsäureäthylester in 6,0 1 Benzol wird während 20
Stunden unter Rühren am Rückfluss erhitzt. Der Ansatz wird mit 2,0 1 Wasser versetzt und erneut 2 Stunderi unter
Kochen am Rückfluss erhitzt. Nach dem Abkühlen auf
Zimmertemperatur setzt man 400 ml 10%gen Ammoniak zu und trennt die Benzolphase ab. Diese wird mit Natrium sulfat getrocknet und nach dem Filtrieren zum Öl ein geengt. Die Hauptfraktion der 1 -Carbäthoxy-4-piperidon-
3-essigsäureäthylester siedet bei einem Kp 145-1700/
0,05 mm.
c) Eine Mischung von 514,0 g 1-Carbäthoxy-4 piperidon-3-essigsäureäthylester und 100 g Hydrazinhydrat in 1,5 1 95 %dem Alkohol und 200 ml Eisessig erhitzt man in einem Stickstoffstrom während 4 Stunden unter Rühren am Rückfluss. Danach wird der Ansatz vollständig ein geengt. Der kristallisierte Rückstand wird aus 1,0 1
95 %igem Äthanol umkristallisiert und ergibt den 2,3,4,4a,5,6,7,8-Octahydro-3-oxo-6-pyrido[4,3-c]pyrida- zincarbonsäureäthylester vom Fp. 1631660 (Zersetzung).
d) 450,5 g 2,3,4,4a,5,6,7,8-Octahydro-3-oxo-6- pyrido[4,3-c]pyridazincarbonsäureäthylester werden in 2,0 1
Chloroform gelöst und die Lösung auf Rückflusstemperatur erhitzt. In diese kochende Lösung gibt man innerhalb von 2l/2 Stunden 320 g Brom in 500 ml Chloroform. Der
Ansatz wird nach der Bromzugabe noch eine weitere
Stunde unter Rückfluss gerührt. Nach dem Abkühlen auf
Raumtemperatur wird der Ansatz mit 1 kg Eis/Wasser versetzt und der Chloroformteil abgetrennt. Der saure wässrige Teil wird mit weiteren 500 ml Chloroform ausge zogen. Die Chloroformteile werden der Reihe nach mit
300 ml Wasser gewaschen, über Natriumsulfat getrocknet und im Vakuum eingeengt.
Der halbkristalline Rückstand wird mit 250 ml absolutem Äthanol umkristallisiert und ergibt das analysenreine 6-Carbäthoxy-5 ,6,7,8-tetrahydro-
3(2H)pyrido[4,3-c]pyridazinon. Fp. 1651680 (Zerset zung).
e) Eine Lösung von 300,0 g 6-Carbäthoxy-5,6,7,8 tetrahydro-3 (2H)pyrido[4,3-c]pyridazinon in 850 ml
Phosphoroxychlorid wird während 1 Stunde am Rückfluss zum Sieden erhitzt. Das Reaktionsgemisch wird vollständig zum harzigen Rückstand eingeengt und dieser wird noch warm auf 4,0 kg Eis/Wasser gegossen. Durch längeres
Rühren erhält man eine vollständige Lösung. die unter
Zugabe weiteren Eises mit ca. 250 ml konzentriertem
Ammoniak bis zu einem pH von 4-5 neutralisiert wird. Der
Ansatz wird mit total 2,5 1 Chloroform extrahiert, diese
Chloroformteile mit 250 ml Wasser nachgewaschen, über
Natriumsulfat getrocknet und im Vakuum zum kristallinen
Rohprodukt eingeengt.
Nach dem Umkristallisieren aus
700 ml absolutem Alkohol erhält man den analysenreinen 3-Chlor-5,6,7,8-tetrahydro-6-pyrido[4,3-c]pyridazincar- bonsäureäthylester vom Fp. 102-1050 (Zersetzung).
f) 113,0 g 3-Chlor-5,6,7,8-tetrahydro-6-pyrido[4,3-c] pyridazincarbonsäureäthylester und 470 ml Hydrazinhydrat werden bei einer Badtemperatur von 110 während 1
Stunde unter Rückfluss gerührt. Der Ansatz wird mit Eis gekühlt und das ausgefallene Rohprodukt abfiltriert, mit wenig Alkohol und anschliessend mit Äther nachgewaschen und getrocknet. Nach dem Umkristallisieren aus absolutem
Alkohol erhält man den analysenreinen 3-Hydrazino 5,6,7,8-tetrahydro-6-pyrido[4,3-c]pyridazincarbonsäure- äthylester vom Fp. 145-147 (Zersetzung).
Beispiel 2 3-(2-Butylidenhydrazino)-5,6,7,8-tetrahydro-
6-pyrido[4,3-c]pyridazincarbonsäureäthylester
Eine Aufschlämmung von 23,7 g 3-Hydrazino-5,6,7,8 tetrahydro-6-pyrido[4,3-c]pyridazincarbonsäureäthylester in 100 ml Methyläthylketon wird 4 Stunden unter Rühren am Rückfluss erhitzt. Während des Aufheizens geht das Ausgangsmaterial vollständig in Lösung. Die Reaktionslösung wird im Vakuum teilweise eingeengt mit Eis gekühlt und das ausgefallene Material abfiltriert. Das Rohprodukt wird einmal aus Methanol um kristallisiert und ergibt die im Titel genannte analysenreine Verbindung vom Smp. 142 bis 146 (Zersetzung).

The compounds of the formula I and their pharmacologically acceptable acid addition salts have not yet been described in the literature.
