Verfahren zur Herstellung von neuen Thiophen-Derivaten
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von neuen 4H-Benzo[4,5]cyclohepta[1,2-b]thiophen-Derivaten der allgemeinen Formel I

und ihren Säureadditionssalzen, worin Z die Gru, ppe -CH= CH-oder-CH2-CHr, eines von Ri und R2 ein Wasserstoffatom, das andere ein Wasserstoff-oder Halogenatom und R3 ein Wasserstoffatom oder die Methylgruppe bedeutet.
ErNndungsgemäss gelangt man zu den neuen Verbindungen der allgemeinen Formel I und ihren Säure- additionssalzen, indem man ein 4H-Benzo[4,5]cyclohepta[l,2-b]thiophen-Derivat der Formel II

reduziert und die erhaltene Verbindung gew nschtenfalls in ihre SÏureadditionssalze berf hrt.
Ausgangssubstanzen der Formel II sind 4H-Benzo [4,5]cyclohepta[l,2-b]thiophen- bzw. ihre 9,10-Dihydroderivate, die in 4-Stellung den 3-Methylaminopropyliden- oder den 3-Methylamino-2-methyl-propyliden Rest enthalten, ferner die entsprechenden, in 6- oder 7 Sbellung durch Chlor, Brom oder Jod substituierten Verbindungen.
Das erfindungsgemässe Verfahren kann vorteilhafterweise so ausgeführt werden, dass man die Lösung einer Verbindung der Formel II, wie beispielsweise 4- (3'- Methylamino-propyliden)-9, 10-dihydro-4H-benzo [4, 5]cyclohepta [l,2-b]thiophen-hydrochlorid,in Eisessig mit rotem Phosphor und Jodwasserstoffsäure kurze Zeit er hitzt. Nach Filtration des Reaktionsgemisches und Ein dampfen des Filtrates wird der Rückstand in Gegenwart von Alkalien, z. B. 20proz. Natronlauge, in einem organischen Lösungsmittel, vorzugsweise Methylen- chlorid, aufgenommen.
Aus der organischen Phase wird das Jod mittels Natriumthiosulfat entfernt und das gewünschte Endprodukt dann in bekannter Weise isoliert und vorzugsweise durch ¯berf hrung in ein geeignetes Salz gereNinigt.
B, evorzugte Salze sind beispielsweise die Hydro- chloride, Hydrobromide, Phosphate, Sulfate, Acetatc, Malonate, Fumarate, Maleinate, Tartrate, Malate, Hexahydrobenzoate, Benzolsulfonate oder p-Toluolsulfonate.
Die neuen Verbindungen der allgemenen Formel I sowie ihre Salze mit pharmazeutisch verträglichen orga- nischen oder anorganischen Säuren zeichnen sich insbe- sondere durch antidepressive Wirkung au, s, was sich z. B. durch einen Antagonismus gegenüber zentralen und peripheren Reserpinwirkungen äussert.
Die neuen Verbindungen sollen deshalb in der Therapie als Antidepressiva. Verwendung finden.
Ausgangsprodukte der Formel II sind neu und werden z. B. hergestellt, indem man ein 4H-Benzo [4, 5]cyclohepta [1, 2-b]thiophen-4-on der Formel III

4H-Benzo [4, 5] cyclohepta [1, 2-b] thiophen-4-on-Deri- vate der Formel III sind zum Teil neu und können nach folgendem Verfahren hergestellt werdien : Man konden- siert 2-Thenyl-diäthylphosphonat isn einem geeigneten wasserfreien organischen Lösungsmittel und in Gegenwart eines alkalischen Kondensationsmittels mit einer gegebenenfalls in 4-oder 5-Stellung durch Halogen- atome substituierten o-Phthalaldehydsäure, reduzieat die so erhaltene o-[2-Thienyl - (2') - vinyl] - benzoesÏure respektive ihre Halogen-Denivatfe zur r o-[2-Thienyl-(2') äthyl]-benzoesäure respektive ihren Derivaten und unterwirft diese einem intramolekularen Ringschluss, wobei 9,
10-Dihydro - 4H-benzo [4, 5] cyclohepta [1, 2-b] thiophen4-on respektive seine in 6-oder 7-Stellung durch Halogen substituierten Derivate erhalten werden.
Als Reduktionsmittel kann beispielsweise Natriumamalgam in wässerigem Alkohol, als Kondensationsmittel f r den Ringschluss Polyphosphorsäure verwendet werden.
Die Dehydrierung in 9, 10-Stellung kann zweckmä- ssig, falls erwünscht, wie folgt ausgeführt werden :
Das oben erhaltene 9, 10-Dihydro-4H-benzo [4, 5] cyclohepta [1, 2-b] thiophen-4-on odar seine in 6-oder 7-Stel) lung durch Halogen substituierten Derivabe wer- den mit N-Bromsuccinimid in abs. Tetrachlorkohlenstoff und in Gegenwart katalytischer Mengen Dibenzoylperoxyd erhitzt und das entstandene Reaktionsprodukt an schliessend mit einem Trialkylamin erhitzt.
In den nachfolgenden Beispielen, welche die Ausführung des Verfahrens erläutern, den Umfang der Er findung aber in keiner Weise einschränken sollen, erfolgen alle Temperaturangaben in Celsiusgraden und sind unkorrigiert.
Beispiel 1
4-(3'-Methylamino-propyl)-9,10-dihydro
4H-benzo [4, 5] cyclohepta [1, 2-b] thiophen
Die L¯sung von 12,0 g 4-(3'-Methylamino-propyliden) - 9,10-dihydro-4H-benzo[4,5]cyclohepta[l,2-b]thiophen-hydrochlorid in 240 cm3 Eisessig wird mit
12, 5 g rotem Phosphor und 64 cm3 56proz. Jodwasser stoffsäure versetzt und das Gemisch während Minuben am Rückfluss erhitzt. Anschliessend wird das Reak- tionsgemisch filtriert und das Filtrat unter verminder- tem Druck eingedampft. Der ölige Rückstand wird in 20proz.Natronlaugeundin Methylenchlorid gelöst, die organische Phase abgetrennt und der wässerige Teil noch zweimal mit Methylenchlorid extrahiert.
Die vereinigten Methylenchloridextrakte werden zweimal mit 5proz.
Natriumthiosulfatl¯sung, dann noch zweimal mit Wasser gewaschen, über Kaliumcarbonat getrocknet und unter vermindertem Druck eingedampft. Die Lösung des R ckstands in 20 cm3 Athanol versetzt man darauf mit der berechneten Menge äthanolischer Salzsäure. Nach einiger Zeit kristallisiert das Hydrochlorid aus. Es wird aus ¯thanol umkristallisiert. Smp. 210,5-212¯.
Das als Ausgangssubstanz verwendete 4-(3'-Methylamino-propyliden) - 9,10 - dihydro-4H-benzo[4,5]cyclohepta [1, 2-b] thiophen-hydrochlonid wird'wie folgt hergestellt : a) Zu einer Lösung von 117 g Thenyl-diäthyl-phos- phat (Sdp. 120-124¯/0,06 mm Hg) in 200 cm3 frisch destilliertem Dimethylformamid gibt man 30 g pulverisiertes, gut getrocknetes Natriummethylat, wobei sich die Lösung auf 45-50 erwärmt. Dann stellt man den Kolben in ein Eisbad und lϯt eine Lösung von 80 g o-Phthalaldehydsäure in 200 cm3 Dimethylformamid so zutropfen, da¯ die Temperatur zwischen 35-40¯ bleibt, und rührt dann noch 30-60 Minuten bei Raumtempera- tur weiter.
Die Reaktionslösung wird nun unter gutem Kühlen mit 1600 cm3 Wasser (Tempelratur 10-15 ) versetzt, wobei sich ein rotes 01 abscheidet. Man stellt dann mit Kaliumcarbonat alkalisch, wobei sich das Öl wieder l¯st schüttelt die rotbraune Lösung dreimal mit Benzol aus und stellt die wässenige Lösung vorsichtig bei 10-15 mit Salzsäure auf pH 4. Nach einigen Stunden im Kühlschrank wird die ausgefallene Säure abfiltriert, getrocknet und aus Benzol umkristallisiert. Smp. der o-[2-Thienyl-(2')-vinyl]-benzoesäure 133-135 . Die Mutterlauge wird dreimal mit) Methylenchlorid ausge- schüttelt, die organische Phase ber Natriumsulfat getrocknet und bei 15 mm Hg eingedampft.
Der Rückstand wird aus Benzol kristallisiert, wobei noch eine weitere Portion Säure vom Smp. 133-135¯ erhalten wird. b) 7, 5 g Natrium werden unter wasserfreiem Toluol geschmolzen, worauf man unter öfterem Schütteln 375 g reines Quecksilber so zutropft, dass das Toluol siedet.
Dann erhitzt man das Gemisch unter Rühren auf 120 bas 140¯ und k hlt, sobald alles Toluol abdestilliert ist, auf 50 ab. Das homogene Amalgam übergiessb man n nun mit einer Lösung von 50 g o-[2-Thienyl-(2')-vinyl] benzoasäure in 150 cm3 95proz. ¯thanol und sch ttelt die Mischung wÏhrend 30 Minute. Darauf trennt man das Quecksilber ab, wäscht es zweimal mit Athanol und verdünnt die vereinigten äthanolischen Losungen mit
1200 cm3 Wasser. Die Lösung wird durch hochgerei- nigte Diatomeenerde filtriert, mit Salzsäure angesäuert und auf 5 abgekiihlt. Nach einigen Stunden filtriert man die ausgefallene o-[2-Thienyl-(2')-Ïthyl]-benzoesÏure ab und kristallisiert sie aus Chloroform/Hexan.
Smp. 110-111¯. c) 59 cm3 84proz. Phosphorsäure und 86 g Phosphorpentoxyd werden zuerst bei 125-130¯ wÏhrend 30 Minuten gerührt. Dann werden bei dieser Temperatur 20 g pulverisierte o- [2-Thienyl- (2)-äthyl]-benzoesäure während 30 Minuten eingetragen. Man r hrt das Reak tionsgemisch noch 2 Stunden bei 125-130 , giesst es in
1000 cm3 Wasser, filtriert die Lösung durch hochgerei- nigte Diatomeenerde und extrahiert sie dreimal mit Methylenchlorid.
Die organische Phase wird mit 2n Natriumcarbonatlösung gewaschen, über Magnesiumsul- fat getrocknet, das L¯sungsmittel abgedampft und der Rückstand im Hochvakuum destilliert, wobei das 9, 10
Dihydro-4H-benzo[4,5]cyclohepta[1,2-b]thiophen-4-on bei 125-140 /0, 05 mm Hg als grünes Öl übergeht, n 24 D = 1, 6559. d) 2, 2 g mit Jod aktiviertes Magnesium werden mit wenig Tetrahydrofuran überschichtet und mit einigen Tropfen Sithyienbromid versetzt.
Nach Einsetzen der Reaktion lässt man eine Lösung von 10, 8 g 3-Dimethyl- ammopropylchlorid in 20 cm3 Tetrahydrofuran so zutropfen, dass daN Lösungsmittelsiedet, und erhitzt anschliessend noch 2 Stunden zum Sieden. Man läl3t nun innerhalb 10 Minuten eine Lösung von 8, 2 g 9, 10-Di hydro-4H-benzo [4, 5] cyclohepta [1, 2-b] thiophen-4-on in 30 cm3 Tetrahydrofuran zutropfen und erhitzt unter Rühren noch weitere 10 Minuten zum Sieden.
Nach Abk hlen wird das Reaktionsgemisch in eine Lösung von 30 g Ammoniumchlorid in 200 cm3 Wasser gegossen, 200 cm3 Methylenchlorid zugesetzt und das Ganze durch hochgereinigte Diatomeenerde filtriert. Nach dem Abtrennen der organischen Phase wird der wässe rige Teil noch zweimal mit Methylenchlorid ausgeschüt- telt, die vereinigten Methylenchloridlosungen mit Wasser gewaschen, über Magnesiumsulfat getrocknet und bei
15 mm Hg eingedampft. Der ¯lige Rückstand wird aus ¯ther/PetrolÏther kristallisiert.
Das 4-3'-Dimethylaminopropyl)-9, 10-dihydro-4H-benzo[4,5]cyclohepta[l,2-b] thiophen-4-ol schmilzt bei 101 . e) 8g 4- (3'-Dimethylaminopropyl)-9, 10-dihydro-4H- benzo [4, 5] cyclohepta [1, 2-b]thiophen-4-ol, 80 cm3 Eisessig und 32 cm3 konz. SalzsÏure werden 30 Minuten unter Rückfluss erhitzt, das Reaktionsgemisch bei 15 mm Hg eingedampft und der Rückstand mit ¯thanol/¯ther (1 : 1) verrieben. Das ausgefallene Hydrochlorid wird abfiltriert und aus ¯thanol/¯ther umkristallisiert. Das 4-(3'-Dimethylamino-propyliden)-9, 10-dihydro-4H-benzo [4, 5] cyclohepta [l, 2-b] thiophen-hydroohlorid schmilzt bei 222-224 (Zers.).
f) Zu einer Lösung von 6, 5 g ChlorameisensÏure äthylester in 25 cm3 abs. Benzol lϯt man innert 30 Minuten bei Raumtemperatur eine Lösung von 5, 7 g 4-(3'-Dimethylamino-propyliden)-9, 10-dihydro-4H-benzo [4, 5] cyclohepta [1, 2-b]thiophen in 25 cm3 abs. Benzol zutropfen. Anschliessend erhitzt man das Gemisch unter Rühren noch 2 Stunden zum Sieden, wäscht es nach dem Abkühlen dreimal mit 2n Salzsäure, dann noch zweimal mit Wasser und trocknet die Benzollösung über Magne siumsulfat.
Nach Eindampfen des Lösungsmittels erhält man das reine 4-(3-Mefhyl-3-äthoxycarbonylamino-' propyliden)-9, 10-dihydro-4H-benzo [4, 5] cyclohepta, [1, 2-b]thiophen. n 25 D= 1, 5770. g) Man erhittzt eine Lösung von 6, 2 g 4- (3'-Methyl- 3'-Ïthoxycarbonylamino-propyliden)-9, 10-dihydro-4Hbenzo [4, 5] cyclohepta [1, 2-b] thiophen und 5 g Kaliumhy droxyd in 55 cm3 n-Butanol unter Rühren während 6 Stunden in einer StickstoffatmosphÏre zum Sieden. Anschliessend dampft man das Lösungsmittel ein, löst den Rückstand bei 60¯ in einem Gemisch von 65 cm3 2n Schwefelsäure und 55 cm ? Wasser und kühlt die Lösung ab.
Die saure Lösung wird mit 55 cm3 Hexan ausge- schüttelt, wobei drei. Schichten erhalten werden. Man trennt die zwei unteren Schichten ab und wäscht die obere Hexanschicht nochmals mit 20 cm3 n Schwefel- säure. Die vereinigten wässerigen L¯sungen werden mit Natronlauge stark alkalisch gestellt und dreimal mit Äther extrahiertt. Die vereinigten, mit Wasser gewasche- nen und über Magnesiumsulfat ge'brockneten ätherischen Extrakte werden anschliessend eingedampft. Der erhaltene Rückstand wird darauf in Isopropanol gelöst und die entstandene Lösung mit einer isopropanolischen Chlorwasserstofflösung versetzt.
Nach einigen Stunden filtriert man das 4-(3'-Methylamino-propyliden)-9, 10 dihydro-4H-benzo [4, 5] cyclohepta [1, 2-b] thiophen-hydro- chlorid ab und kristallisiert es aus Äthanol/Isopropanol um. Smp. 236, 5-238, 5 (Zers.).
Beispiel 2 4- (3-Methylamino-2-methyl-propyl)-9, 10-dihydro-
4H-benzo [4, 5] cyclohepta[1, 2-b] thiophen
Das Gemisch von 0, 7 g 4- (3-Methylamino-2-methyl- propyliden)-9, 10-dihydro-4H-benzo [4, 5] cyclohepta- [1, 2-b] thiophen, 1, 0 g rotem Phosphor, 5, 6 cm3 56proz.
Jodwasserstoffsäuref und 20 cm Bisessig wird 10 Minu- ten unter Rückfluss erhitzt, sodann heiss filtriert und das Filtrat unter vermindertem Druck eingedampft. Darauf versetzt man den Rückstand mit einem Gemisch von 20 cm3 20proz. Natronlauge und 20 cm3 Methylen- chlorid, trennt die organische Phase ab, schüttelt den wässerigen Teil noch zweimal mit Methylenchlorid aus und wÏscht die vereinigten Methylenchloridextrakte zweimal mit 5proz. Natriumthiosulfatl¯sung und zweimal mit Wasser nach. Nach Trocknen über Natriumsulfat dampft man das Lösungsmittel unter vermindertem Druck ab.
Die Lösung des Rückstandes in 10 cm3 Methanol versetzt man sodann mit einer Lösung der berechneten Menge Oxalsäure in Athanol. Das ausgefallene Oxalat wird abfiltriert und aus Methanol/Äthanol umkristallt- siert. Das Hydrogen-oxalat schmilzt bei 178-180 (Zers.).
Das als Ausgangssubstanz verwendete 4- (3-Methyl- amino-2-methyl-propyliden)-9, 10-dihydro-4H-benzo [4, 5]- cyclohepta [l, 2-b] thiophen wird wie folgt hergestellt : a) Zu einer Lösung von 2, 0 g Chlorameisensäure- äthylester in 15 cm3 abs. Benzol lϯt man während 15 Minuten eine Lösung von 1, 8 g 4-(3-Dimethylamino-2methyl-propyliden)-9, 10-dihydro-4H-benzo [4, 5] cyclohepta [1, 2-b] thiophen in 15 cm3 abs. Benzol bei Raum temperatur zutropfen. Anschliessend erhitzt man das Reaktionsgemisch unter Rühren noch 2 Stunden zum Sieden, wäscht es nach dem Abkühlen dreimal mit ln Salzsäure, dann noch zweimal mit Wasser und trocknet die Benzollösung über Natriumsulfat.
Nach dem Ver dampfen des Lösungsmittel erhält man das reine 4- (3- Methyl-3-Ïthoxycarbonylamino-2-methyl-propyliden)9, 10-dihydro-4H-benzo [4, 5] cyclohepta [1, 2-b] thiophen. n 25 D =1, 5700. b) Man erhitzt die Lösung von 1, 7 g 4- (3-Methyl-3- äthoxycarbonylamino-2-methyl-propyliden)-9, 10-dihydro- 4H-Benzo [4, 5] cyclohepta [1, 2-b] thiophen, 1, 5 g Kaliumhydroxyd und 15 cm3 n-Butanol unter Rühren während 5 Stunden in einer Stickstoffatmosphäre zum Sieden.
Nach dem Abkühlen gibt man 20 cm3 Toluol zu und wäscht fünfmal mit Wasser nach. Die organische Lösung wird darauf dreimal mit 2n Weinsäure ausgeschüttelt, die weinsaure Lösung unter Kühlen mit Natronlauge alkalisch gestellt und mit Methylenchlomd extrahiert.
Der Methylenchloridextrakt wird anschliessend mit Was- ser gewaschen, über Natriumsulfat getrocknet und das Lösungsmittel verdampft.
Hydrochlorid :
Eine Lösung der rohen Base in Aceton versetzt man mit der berechneten Menge acetonischer Salzsäure, filtriert nach einiger Zeit das ausgefallene Hydrochlorid ab und kristallisiert es aus Aceton um. Das Hydro- chlorid schmilzt bei i 194-195, 5 (Zers.).

Starting substances of the formula II are 4H-benzo [4,5] cyclohepta [l, 2-b] thiophene or their 9,10-dihydro derivatives, which in the 4-position are 3-methylaminopropylidene or 3-methylamino-2- contain methyl-propylidene radical, as well as the corresponding compounds substituted in 6- or 7-position by chlorine, bromine or iodine.
The inventive method can advantageously be carried out so that the solution of a compound of the formula II, such as 4- (3'-methylamino-propylidene) -9, 10-dihydro-4H-benzo [4, 5] cyclohepta [l, 2-b] thiophene hydrochloride, heated in glacial acetic acid with red phosphorus and hydriodic acid for a short time. After filtering the reaction mixture and evaporating the filtrate, the residue is in the presence of alkalis, eg. B. 20% Sodium hydroxide solution, added in an organic solvent, preferably methylene chloride.
The iodine is removed from the organic phase using sodium thiosulphate and the desired end product is then isolated in a known manner and preferably purified by converting it into a suitable salt.
Preferred salts are, for example, the hydrochlorides, hydrobromides, phosphates, sulfates, acetate, malonates, fumarates, maleinates, tartrates, malates, hexahydrobenzoates, benzenesulfonates or p-toluenesulfonates.
The new compounds of the general formula I and their salts with pharmaceutically acceptable organic or inorganic acids are distinguished, in particular, by an antidepressant effect, B. expressed by an antagonism to central and peripheral reserpine effects.
The new compounds are therefore intended to be used in therapy as antidepressants. Find use.

The 9, 10-dihydro-4H-benzo [4, 5] cyclohepta [1, 2-b] thiophen-4-one obtained above or its derivative substituted in the 6- or 7-position by halogen are denoted by N -Bromosuccinimide in abs. Carbon tetrachloride and heated in the presence of catalytic amounts of dibenzoyl peroxide and the resulting reaction product is then heated with a trialkylamine.
In the following examples, which explain the implementation of the method, but are not intended to limit the scope of the invention in any way, all temperatures are given in degrees Celsius and are uncorrected.
example 1
4- (3'-methylamino-propyl) -9,10-dihydro
4H-benzo [4, 5] cyclohepta [1, 2-b] thiophene
The solution of 12.0 g of 4- (3'-methylamino-propylidene) - 9,10-dihydro-4H-benzo [4,5] cyclohepta [1,2-b] thiophene hydrochloride in 240 cm3 of glacial acetic acid becomes With
12.5 g red phosphorus and 64 cm3 56% Hydroiodic acid is added and the mixture is refluxed for minuben. The reaction mixture is then filtered and the filtrate is evaporated under reduced pressure. The oily residue is dissolved in 20% sodium hydroxide solution and in methylene chloride, the organic phase is separated off and the aqueous part is extracted twice more with methylene chloride.
The combined methylene chloride extracts are twice with 5proz.
Sodium thiosulfate solution, then washed twice more with water, dried over potassium carbonate and evaporated under reduced pressure. The solution of the residue in 20 cm3 of ethanol is then mixed with the calculated amount of ethanolic hydrochloric acid. After a while, the hydrochloride crystallizes out. It is recrystallized from ethanol. M.p. 210.5-212¯.
The 4- (3'-methylamino-propylidene) - 9,10 - dihydro-4H-benzo [4,5] cyclohepta [1,2-b] thiophene hydrochloride used as the starting substance is prepared as follows: a) To one A solution of 117 g of thenyl diethyl phosphate (bp. 120-124¯ / 0.06 mm Hg) in 200 cm3 of freshly distilled dimethylformamide is added to 30 g of powdered, well-dried sodium methylate, the solution warming to 45-50 . The flask is then placed in an ice bath and a solution of 80 g of o-phthalaldehyde acid in 200 cm3 of dimethylformamide is added dropwise so that the temperature remains between 35-40¯, and the mixture is then stirred for another 30-60 minutes at room temperature continue.
The reaction solution is then mixed with 1600 cm3 of water (temple temperature 10-15) with thorough cooling, a red oil separating out. It is then made alkaline with potassium carbonate, which dissolves the oil again, shakes the red-brown solution three times with benzene and carefully adjusts the aqueous solution to pH 4 at 10-15 with hydrochloric acid. After a few hours in the refrigerator, the precipitated acid is filtered off, dried and recrystallized from benzene. Mp of o- [2-thienyl- (2 ') -vinyl] -benzoic acid 133-135. The mother liquor is shaken out three times with) methylene chloride, the organic phase is dried over sodium sulfate and evaporated at 15 mm Hg.
The residue is crystallized from benzene, a further portion of acid having a melting point of 133-135¯ being obtained. b) 7.5 g of sodium are melted under anhydrous toluene, whereupon 375 g of pure mercury are added dropwise with frequent shaking so that the toluene boils.
The mixture is then heated to 120 bas 140¯ with stirring and cooled to 50 as soon as all of the toluene has distilled off. The homogeneous amalgam is then poured over with a solution of 50 g of o- [2-thienyl- (2 ') - vinyl] benzoic acid in 150 cm3 95%. Ethanol and shake the mixture for 30 minutes. The mercury is then separated off, washed twice with ethanol and the combined ethanolic solutions are diluted with it
1200 cm3 of water. The solution is filtered through highly purified diatomaceous earth, acidified with hydrochloric acid and cooled to 5. After a few hours, the precipitated o- [2-thienyl- (2 ') - ethyl] benzoic acid is filtered off and crystallized from chloroform / hexane.
M.p. 110-111¯. c) 59 cm3 84% Phosphoric acid and 86 g of phosphorus pentoxide are first stirred at 125-130¯ for 30 minutes. Then 20 g of pulverized o- [2-thienyl- (2) -ethyl] -benzoic acid are introduced over 30 minutes at this temperature. The reaction mixture is stirred for a further 2 hours at 125-130, it is poured into
1000 cm3 of water, filter the solution through highly purified diatomaceous earth and extract it three times with methylene chloride.
The organic phase is washed with 2N sodium carbonate solution, dried over magnesium sulfate, the solvent is evaporated and the residue is distilled in a high vacuum, the 9.10
Dihydro-4H-benzo [4,5] cyclohepta [1,2-b] thiophen-4-one turns as a green oil at 125-140 / 0.05 mm Hg, n 24 D = 1.6559. D) 2, 2 g magnesium activated with iodine are covered with a little tetrahydrofuran and mixed with a few drops of sithyiene bromide.
After the reaction has started, a solution of 10.8 g of 3-dimethyl ammopropyl chloride in 20 cm3 of tetrahydrofuran is added dropwise so that the solvent boils, and the mixture is then heated to the boil for a further 2 hours. A solution of 8.2 g of 9, 10-dihydro-4H-benzo [4, 5] cyclohepta [1, 2-b] thiophen-4-one in 30 cm3 of tetrahydrofuran is then added dropwise over the course of 10 minutes and the mixture is heated with stirring another 10 minutes to simmer.
After cooling, the reaction mixture is poured into a solution of 30 g of ammonium chloride in 200 cm3 of water, 200 cm3 of methylene chloride is added and the whole is filtered through highly purified diatomaceous earth. After the organic phase has been separated off, the aqueous part is extracted twice more with methylene chloride, and the combined methylene chloride solutions are washed with water, dried over magnesium sulfate and added
15 mm Hg evaporated. The oily residue is crystallized from ether / petroleum ether.
The 4-3'-dimethylaminopropyl) -9, 10-dihydro-4H-benzo [4,5] cyclohepta [1,2-b] thiophen-4-ol melts at 101. e) 8g 4- (3'-dimethylaminopropyl) -9, 10-dihydro-4H-benzo [4, 5] cyclohepta [1, 2-b] thiophen-4-ol, 80 cm3 glacial acetic acid and 32 cm3 conc. Hydrochloric acid is refluxed for 30 minutes, the reaction mixture is evaporated at 15 mm Hg and the residue is triturated with ethanol / ether (1: 1). The precipitated hydrochloride is filtered off and recrystallized from ¯thanol / ¯ther. The 4- (3'-dimethylamino-propylidene) -9,10-dihydro-4H-benzo [4,5] cyclohepta [1,2-b] thiophene hydrochloride melts at 222-224 (dec.).
f) To a solution of 6.5 g of ethyl chloroformate in 25 cm3 of abs. Benzene is left within 30 minutes at room temperature to a solution of 5.7 g of 4- (3'-dimethylamino-propylidene) -9, 10-dihydro-4H-benzo [4, 5] cyclohepta [1, 2-b] thiophene in 25 cm3 abs. Add benzene dropwise. The mixture is then heated to boiling for a further 2 hours with stirring, washed three times with 2N hydrochloric acid and then twice with water after cooling, and the benzene solution is dried over magnesium sulfate.
After evaporation of the solvent, the pure 4- (3-methyl-3-ethoxycarbonylamino- 'propylidene) -9, 10-dihydro-4H-benzo [4,5] cyclohepta, [1,2-b] thiophene is obtained. n 25 D = 1.5770. g) A solution of 6.2 g of 4- (3'-methyl-3'-Ïthoxycarbonylamino-propylidene) -9, 10-dihydro-4Hbenzo [4, 5] cyclohepta [1 , 2-b] thiophene and 5 g of potassium hydroxide in 55 cm3 of n-butanol with stirring for 6 hours in a nitrogen atmosphere to boiling. The solvent is then evaporated and the residue is dissolved at 60¯ in a mixture of 65 cm3 of 2N sulfuric acid and 55 cm? Water and cool the solution.
The acidic solution is shaken out with 55 cm3 of hexane, with three. Layers are obtained. The two lower layers are separated off and the upper hexane layer is washed again with 20 cm3 N sulfuric acid. The combined aqueous solutions are made strongly alkaline with sodium hydroxide solution and extracted three times with ether. The combined essential extracts washed with water and dried over magnesium sulphate are then evaporated. The residue obtained is then dissolved in isopropanol and an isopropanolic hydrogen chloride solution is added to the resulting solution.
After a few hours, the 4- (3'-methylamino-propylidene) -9, 10 dihydro-4H-benzo [4, 5] cyclohepta [1, 2-b] thiophene hydrochloride is filtered off and it is crystallized from ethanol / Isopropanol. M.p. 236, 5-238, 5 (dec.).
Example 2 4- (3-methylamino-2-methyl-propyl) -9, 10-dihydro-
4H-benzo [4, 5] cyclohepta [1, 2-b] thiophene
The mixture of 0.7 g of 4- (3-methylamino-2-methyl-propylidene) -9,10-dihydro-4H-benzo [4,5] cyclohepta- [1,2-b] thiophene, 1.0 g red phosphorus, 5.6 cm3 56%
Hydroiodic acid and 20 cm acetic acid are heated under reflux for 10 minutes, then filtered while hot and the filtrate is evaporated under reduced pressure. The residue is then mixed with a mixture of 20 cm3 20%. Sodium hydroxide solution and 20 cm3 of methylene chloride, the organic phase is separated off, the aqueous part is extracted twice more with methylene chloride and the combined methylene chloride extracts are washed twice with 5%. Sodium thiosulfate solution and twice with water. After drying over sodium sulfate, the solvent is evaporated off under reduced pressure.
The solution of the residue in 10 cm3 of methanol is then mixed with a solution of the calculated amount of oxalic acid in ethanol. The precipitated oxalate is filtered off and recrystallized from methanol / ethanol. The hydrogen oxalate melts at 178-180 (dec.).
The 4- (3-methyl-amino-2-methyl-propylidene) -9, 10-dihydro-4H-benzo [4, 5] - cyclohepta [l, 2-b] thiophene used as the starting material is prepared as follows: a ) To a solution of 2.0 g of ethyl chloroformate in 15 cm3 of abs. Benzene is left with a solution of 1.8 g of 4- (3-dimethylamino-2methyl-propylidene) -9, 10-dihydro-4H-benzo [4, 5] cyclohepta [1, 2-b] thiophene for 15 minutes in 15 cm3 abs. Add benzene dropwise at room temperature. The reaction mixture is then heated to boiling for a further 2 hours with stirring, washed three times with 1N hydrochloric acid and then twice with water after cooling, and the benzene solution is dried over sodium sulphate.
After evaporation of the solvent, the pure 4- (3-methyl-3-Ïthoxycarbonylamino-2-methyl-propylidene) 9, 10-dihydro-4H-benzo [4, 5] cyclohepta [1, 2-b] thiophene is obtained . n 25 D = 1.5700. b) The solution of 1.7 g of 4- (3-methyl-3-ethoxycarbonylamino-2-methyl-propylidene) -9, 10-dihydro-4H-benzo [4.5 ] cyclohepta [1, 2-b] thiophene, 1.5 g of potassium hydroxide and 15 cm3 of n-butanol with stirring for 5 hours in a nitrogen atmosphere to boiling.
After cooling, 20 cm3 of toluene are added and washed five times with water. The organic solution is then extracted three times with 2N tartaric acid, the tartaric acid solution is made alkaline with sodium hydroxide solution while cooling and extracted with methylene chloride.
The methylene chloride extract is then washed with water, dried over sodium sulfate and the solvent is evaporated.
Hydrochloride:
The calculated amount of acetone hydrochloric acid is added to a solution of the crude base in acetone, and after a while the precipitated hydrochloride is filtered off and recrystallized from acetone. The hydrochloride melts at i 194-195.5 (decomp.).
