Die vorliegende Erfindung betrifft ein Verfahren zuI Herstellung von neuen 4H-Benzo[4,5]cyclohepta[1 ,2-b]thiophen-Derivaten der allgemeinen Formel I

und ihren Säureadditionssalzen, worin entweder Rl und R2 ein Wasserstoffatom oder eine niedere Alkylgruppe und R5 und R4 je eine niedere Alkylgruppe bedeuten bzw. R3 und R4 zusammen eine Alkylenkette bilden oder R4 für eine niedere Alkylgruppe steht und R3 mit Rt oder mit R2 zusammen eine Di-, Tri- oder Tetramethylengruppe bildet, wobei der andere der Substituenten Rt und R für Wasserstoff steht.
Erfindungsgemäss gelangt man zu den neuen Verbindungen der allgemeinen Formel 1 und ihren Säureadditionssalzen, indem man 4H-Benzo [4,5] cyclohepta- [1 ,2-b]thiophen-4-on der Formel

Das Verfahren wird beispielsweise folgendermassen ausgeführt: Mit Tetrahydrofuran oder Diäthyläther über schichtete Magnesiumspäne werden mit der Lösung eines basisch substituierten Halogenalkyl-Derivates in Tetra hydrofuran oder Diäthyläther versetzt.
Erfindungsgemäss können als basisch substituierte
Halogenalkyl-Derivate verwendet werden: : 3-Dialkyl- amino-propyl-halogenide, wie z. B. 3-Dimethylamino oder 3 -Diäthylamino-propylhalogenide, Pyrrolidino-, Pipendino-, l-Alkyl-pyrrolidyl-(3)-oder l-Alkyl-piperi dyl-(3 )-methylhalogenide, 1-Alkyl-pyrrolidyl-(2)-oder l-Alkyl-piperidyl-(2)-äthylhalogenide, wobei als Halogenide Chloride, Bromide oder Jodide und als Alkylreste Methyl, Äthyl oder Isopropyl in Frage kommen.
Die Bildung der Orignardverbindung wird vorzugsweise durch Zusatz einer kleinen Menge Äthylenbromid, Äthylbromid oder Methyljodid und einer Spur Jod gefördert. Das Reaktionsgemisch wird gegebenenfalls zur Vervollständigung der Reaktion am Rückfluss erhitzt.
Anstelle von Magnesiumspänen kann auch Kupfer
Magnesium-Legierung (nach Gilman) verwendet werden.
Diese Grignardiösung wird darauf bei 0-300 mit dem im gleichen Lösungsmittel gelösten 4H-Benzo[4,5]- cyclohepta ,2-b]thiophen-4-on versetzt, und das Ge misch zweckmässig noch einige Zeit bei Raumtempera tur gerührt bzw. anschliessend noch zum Sieden erhitzt.
Die so erhaltenen Reaktionsprodukte werden auf ge eignete Weise, z. B. durch Behandlung mit eiskalter wäs seriger Ammoniumchloridlösung, zu Verbindungen der allgemeinen Formel IV hydrolysiert, die auf an sich bekannte Weise isoliert werden können.
Aus diesen Verbindungen der allgemeinen Formel IV können dann durch Einwirkung geeigneter wasserab spaltender Mittel die Verbindungen der allgemeinen Formel 1 gewonnen werden. Als wasserabspaltende Mittel können zum Beispiel Mineralsäuren, starke organi sche Säuren, Essigsäureanhydrid, Thionylchlorid oder Phosphoroxychlorid verwendet werden. Die Uberführung der so erhaltenen Verbindungen der allgemeinen FormelI in ihre Säureadditionssalze kann durch Behandlung mit organischen oder anorganischen Säuren in an sich bekannter Weise geschehen. Bevorzugte Salze sind beispielsweise die Hydrochloride, Hydrobromide, Phosphate, Sulfate, Malonate, Fumarate, Maleinate, Tartrate, Malate, Hexahydrobenzoate oder p-Toluolsulfonate.
Die neuen Verbindungen der allgemeinen FormelI sowie ihre Salze mit pharmazeutisch verträglichen organischen oder anorganischen Säuren zeichnen sich durch eine starke neuroleptische und antidepressive Wirkung aus und sollen daher vor allem zur Behandlung neurotischer und psychotischer Störungen Verwendung finden.
Ausserdem können die neuen Verbindungen auch als Zwischenprodukte zur Herstellung von Medikamenten dienen.
Das 4H-Benzo[4,5]cyclohepta[1 ,2-b]thiophen-4-on, eine bisher unbekannte Verbindung, kann durch folgendes Verfahren hergestellt werden: Man kondensiert Thenyl-diäthylphosphonat in einem geeigneten wasserfreien organischen Lösungsmittel und in Gegenwart eines alkalischen Kondensationsmittels mit o-Phthalaldehydsäure, reduziert die so erhaltene o-[2-Thienyl-(2')-vinyl]benzoesäure zur o-[2-Thienyl-(2')-äthyl]benzoesäure und unterwirft diese einem intramolekularen Ringschluss, wobei das 9,1 O-Dihydro-4H-benzo 4,5]cyclohepta[l ,2-b]- thiophen-4-on erhalten wird. Als Reduktionsmittel kann beispielsweise Natriumamalgam in einem wässerigen Alkohol, und als Kondensationsmittel für den Ringschluss Polyphosphorsäure oder Schwefelsäure verwendet werden.
Die Einführung der Doppelbindung in 9,10-Stellung erfolgt z. B. in der Weise, dass man 9,10-Dihydro-4H- benzo[4,5]cyclohepta[1 ,2-b]thiophen-4-on mit Bromsuccinimid in abs. Tetrachlorkohlenstoff und in Gegenwart katalytischer Mengen Benzoylperoxyd erhitzt und anschliessend das durch Diatomeenerde gereinigte und vom Lösungsmittel befreite Reaktionsprodukt mit einem Trialkylamin erhitzt.
In den nachfolgenden Beispielen, welche die Ausführung des Verfahrens erläutern, den Umfang der Erfindung aber in keiner Weise einschränken sollen, erfolgen alle Temperaturangaben in Celsiusgraden und sind unkorrigiert.

Beispiel 1 a) 4-(3'-Dimethylamino-propyl)4H-benzo[4,5]cyclo- hepta ,2-b]thiophen-4-ol
2,2 g mit Jod aktiviertes Magnesium werden mit
15 ccm abs. Tetrahydrofuran überschichtet und mit einigen Tropfen Athylenbromid versetzt. Nach Einsetzen der Reaktion lässt man eine Lösung von 10,8 g 3-Dimethylamino-propylchlorid in 20 ccm abs. Tetrahydrofuran so zutropfen, dass das Lösungsmittel siedet und erhitzt das Gemisch anschliessend noch 1 Stunde zum Sieden. Unter Kühlen Iässt man darauf bei 20 innert 15 Minuten eine Lösung von 8,2 g 4H-Benzo[4,5]cyclo- hepta[1 ,2-b]thiophen-4-on in 30 ccm abs. Tetrahydrofuran zutropfen und erhitzt weitere 30 Minuten zum Sieden.
Nach Abkühlen wird das Reaktionsgemisch in 250 ccm gesättigte wässerige Ammoniumchloridlösung gegossen, 200 ccm Methylenchlorid zugesetzt und das Ganze durch hochgereinigte Diatomeenerde filtriert.
Nach Abtrennen der organischen Phase wird der wässerige Anteil noch zweimal mit Methylenchlorid ausgeschüttelt, die vereinigten Methylenchloridlösungen mit Wasser gewaschen, über Magnesiumsulfat getrocknet und bei 15 mm Hg eingedampft. Der kristalline Rückstand wird aus Äthanol oder l2ithanol/Hexan umknstal- lisiert. Smp. 121-122 . b) 4-(3'-Dimeíhylamino-propyliden)-4H-benzo [4, 5]- cyclohepta[1 2-b] thiophen
Man erhitzt eine Lösung von 4 g 4-(3'-Dimethylamino-propyl)-4H-benzo [4,5] cyclohepta[ 1 ,2-b]thiophen4-ol in 250 ccm Essigsäureanhydrid während 6 Stunden zum Sieden.
Nach Abdestillieren von etwa 200 ccm Lösungsmittel giesst man den Rückstand unter Rühren in 1200 ccm Wasser, filtriert die wässerige Lösung durch hochgereinigte Diatomeenerde, stellt sie mit 200/oiger Natronlauge stark alkalisch und extrahiert die alkalische Lösung dreimal mit Äther. Die vereinigten, mit Wasser gewaschenen und über Magnesiumsulfat getrockneten Ätherextrakte werden sodann eingedampft. Der erhaltene Rückstand wird im Hochvakuum destilliert, wobei das 4-(3'-Dimethylamino-propyliden)-4H-benzo [4, 5]- cyclohepta[l,2-b]thiophen bei 160-165"/0,1 mm Hg übergeht.
Malonat: Eine Lösung von 3,8 g Base in 10 ccm Isopropanol versetzt man mit einer heissen Lösung von 1,4 g Malonsäure in 10 ccm Isopropanol und filtriert nach dem Abkühlen das ausgefallene Salz ab. Das Malonat schmilzt nach Umkristallisieren aus äthanols Isopropanol bei 142-143 (Zers.).
Das als Ausgangsmaterial verwendete 4H-Benzo [4,5]cyclohepta[l ,2-b]thiophen-4-on wird wie folgt hergestellt: Thenyl-diäthyl-phosphonat
133 g Thenyl-chlorid (Sdp. 73-75 /15 mm Hg) werden auf 1500 erhitzt, worauf man unter Rühren bei dieser Temperatur 175 g frisch destilliertes Triäthylphosphit langsam zutropfen lässt und noch weitere 2 Stunden unter Rühren auf 160-170 erhitzt.
Das Reaktionsgemisch wird dann im Hochvakuum destilliert; das Thenyldiäthyl-phosphonat siedet bei 1201240l0,06 mm Hg. o-[2-Thielzyl-(2 ')-äthyliden]-b enzoesäure
Zu einer Lösung von 117 g Thenyl-diäthyl-phosphonat in 200 ccm frisch destilliertem Dimethylformamid gibt man 30 g pulverisiertes, gut getrocknetes Natriummethylat, wobei sich die Lösung auf 45-50 erwärmt. Dann stellt man den Kolben in ein Eisbad und lässt eine Lösung von 80 g o-Phthalaldehydsäure in 200 ccm Dimethylformamid so zutropfen, dass die Temperatur zwischen 35400 bleibt und rührt dann noch 30 bis 60 Minuten bei Raumtemperatur weiter. Die Reaktionslösung wird nun unter gutem Kühlen mit 1600 ccm Wasser (Temperatur 10-15 ) versetzt, wobei sich ein rotes Öl abscheidet.
Man stellt dann mit Kaliumcarbonat alkalisch, wobei sich das Öl wieder löst, schüttelt die rotbraune Lösung dreimal mit Benzol aus und stellt die wässerige Lösung vorsichtig bei 10-15 mit Salzsäure auf pH 4. Nach einigen Stunden im Kühlschrank wird die ausgefallene Säure abfiltriert, getrocknet und aus Benzol umkristallisiert. Smp. der o-[2 Thienyl-(2')-äthyliden]-benzoesäure 133-135 .
Die Mutterlauge wird dreimal mit Methylenchlorid ausgeschüttelt, die organische Phase über Natriumsulfat getrocknet und bei 15 mm Hg eingedampft. Der Rückstand wird aus Benzol kristallisiert, wobei noch eine weitere Portion Säure vom Smp. 133-135 erhalten wird. o-[2-Thienyl-(2')-äthyl]-benzoesäure
7,5 g Natrium werden unter wasserfreiem Toluol geschmolzen, worauf man unter öfterem Schütteln 375 g reines Quecksilber so zutropft, dass das Toluol siedet.
Dann erhitzt man das Gemisch unter Rühren auf 120 bis 140 und kühlt, sobald alles Toluol abdestilliert ist, auf 500 ab. Das homogene Amalgam übergiesst man nun mit einer Lösung von 20 g o-[2-Thienyl-(2') äthyliden]-benzoesäure in 150 ccm 950/oigem Äthanol und schüttelt die Mischung während 30 Minuten. Darauf trennt man das Quecksilber ab, wäscht es zweimal mit Äthanol und verdünnt die vereinigten äthanolischen Lösungen mit 1200 ccm Wasser. Die Lösung wird durch hochgereinigte Diatomeenerde filtriert, mit Salzsäure angesäuert und auf 5 gekühlt. Nach einigen Stunden fil- triert man die ausgefallene Säure ab und kristallisiert sie aus Chloroform/Hexan. Smp. 110-111".
9,1 O-Dihydro-4H-benzo[4,5]cyclohepta[1 ,2-b]- thiophen-4-on
59 ccm 840/oige Phosphorsäure und 86 g Phosphorpentoxyd werden zuerst bei 125-1300 während 30 Minuten gerührt. Dann werden bei dieser Temperatur 20 g pulverisierte o-[2-Thienyl-(2')-äthyl]-benzosäure während 30 Minuten eingetragen. Man rührt das Reaktionsgemisch noch 2 Stunden bei 125-130 , giesst es in
1000 ccm Wasser, filtriert die Lösung durch hochgereinigte Diatomeenerde und extrahiert sie dreimal mit Methylenchlorid. Die organische Phase wird mit 2-n.
Natriumcarbonatlösung gewaschen, über Magnesiumsulfat getrocknet, das Lösungsmittel abgedampft und der Rückstand im Hochvakuum destilliert, wobei das 9,10 Dihydro-4H- benzo [4,5]cyclohepta[1 ,2-b]thiophen-4 - on bei 125-1400/0,05 mm Hg als grünes Öl übergeht. n2D = 1, 6559.
4H-Benzo[4,5]cyclohepta[1 2-b] thiophen-4-on
Zur Einführung der Doppelbindung in 9,10-Stellung verfährt man in der Weise, dass man ein Gemisch von 32,1 g der erhaltenen Dihydroverbindung, 26,7 g N Bromsuccinimid und 0,3 g Benzoylperoxyd in 250 ccm abs. Tetrachlorkohlenstoff während 4 Stunden zum Sieden erhitzt. Nach Abkühlen auf 50 filtriert man das Reaktionsgemisch durch hochgereinigte Diatomeenerde und dampft das Lösungsmittel bei 15 mm Hg ein. Der erhaltene ölige Rückstand wird darauf während 2 Stunden mit 200 ccm Triäthylamin unter Rühren erhitzt.
Nach Verdampfen von unverändertem Triäthylamin wird der Rückstand mit 250 ccm Methylenchlorid versetzt und die erhaltene Lösung dreimal mit 2-n. Salzsäure und zweimal mit Wasser gewaschen. Nach Trocknen der Lösung über Magnesiumsulfat wird das Lösungsmittel unter vermindertem Druck entfernt. Der Rückstand wird darauf im Hochvakuum destilliert, wobei das 4H-Benzo [4,5]cyclohepta[l ,2-b]thiophen-4-on bei 173-1 800/ 0,1 mm Hg als Öl übergeht und beim Erkalten kristallisiert. Smp. 109-110 nach Umkristalllsieren aus Äthanol.
Beispiel 2 4-(2'-[1 "-Methyl-piperidyl-(2 ")l-äthyliden)-4H-benzo- [4,5]cyclohepta[1 ,2-b]thiophen
2,2 g mit Jod aktiviertes Magnesium werden mit 15 ccm Tetrahydrofuran überschichtet und mit einigen Tropfen Äthylenbromid versetzt. Nach Einsetzen der Reaktion lässt man eine Lösung von 12,6 g 2-[1'-Methylpiperidyl-(2')]-äthylchlorid in 15 ccm Tetrahydrofuran so zutropfen, dass das Lösungsmittel siedet und erhitzt anschliessend noch 1t/2 Stunden zum Sieden. Man Iässt nun bei 20 innert 15 Minuten eine Lösung von 8,2 g 4H-Benzo[4,5]cyclohepta[l ,2-b]thiophen-4-on in 20 ccm Tetrahydrofuran zutropfen und erhitzt unter Rühren noch weitere S/4 Stunden auf 400.
Nach Abkühlen wird das Reaktionsgemisch in 250 ccm 200/oige Ammoniumchloridlösung gegossen, mit 200 ccm Methylenchlorid verdünnt und durch hochgereinigte Diatomeenerde filtriert. Nach dem Abtrennen der organischen Phase wird der wässerige Teil noch dreimal mit Methylenchlorid ausgeschüttelt, die vereinigten Methylenchloridextrakte mit Wasser gewaschen, über Natriumsulfat getrocknet und bei 15 mm Hg eingedampft.
Das ölige 4-(2'-[l"-Methyl-piperidyl-(2")1-äthyl 4H-benzo [4, 5] cyclohepta [1, ,2-b]thiophen-4-ol wird dann in 200 ccm Salzsäure (1:1) gelöst, die saure Lösung zweimal mit Äther gewaschen und dann mit Natronlauge unter Kühlen stark alkalisch gestellt. Diese wässerige Lösung wird dreimal mit Methylenchlorid ausgeschüttelt, die Methylenchloridextrakte mit Wasser gewaschen, über Magnesiumsulfat getrocknet und bei 15 mm Hg eingedampft. Der ölige Rückstand wird in Isopropanol gelöst und mit einer isopropanolischen Salicylsäurelösung versetzt. Nach Abkühlen wird das ausgefallene 4-{2'-[1" Methyl-piperidyl- (2")1- äthyliden)-4H-benzo [4,5] cyclo hepta[l ,2-b]thiophen-salicylat abfiltriert und aus Isopropanol umkristallisiert. Smp. 167-168 .

The new compounds of the general formula I and their salts with pharmaceutically acceptable organic or inorganic acids are distinguished by a strong neuroleptic and antidepressant effect and should therefore be used primarily for the treatment of neurotic and psychotic disorders.
The new compounds can also be used as intermediates in the manufacture of drugs.
The 4H-Benzo [4,5] cyclohepta [1,2-b] thiophen-4-one, a previously unknown compound, can be prepared by the following process: Thenyl diethyl phosphonate is condensed in a suitable anhydrous organic solvent and in the presence of a alkaline condensation agent with o-phthalaldehyde acid, reduces the o- [2-thienyl- (2 ') - vinyl] benzoic acid thus obtained to o- [2-thienyl- (2') - ethyl] benzoic acid and subjects this to an intramolecular ring closure, whereby the 9,1 O-dihydro-4H-benzo 4,5] cyclohepta [1,2-b] -thiophen-4-one is obtained. Sodium amalgam in an aqueous alcohol can be used as a reducing agent, for example, and polyphosphoric acid or sulfuric acid as a condensation agent for the ring closure.
The introduction of the double bond in the 9,10-position takes place z. B. in such a way that 9,10-dihydro-4H-benzo [4,5] cyclohepta [1,2-b] thiophen-4-one with bromosuccinimide in abs. Carbon tetrachloride and heated in the presence of catalytic amounts of benzoyl peroxide and then the reaction product purified by diatomaceous earth and freed from solvent is heated with a trialkylamine.
In the following examples, which explain the implementation of the process but are not intended to restrict the scope of the invention in any way, all temperatures are given in degrees Celsius and are uncorrected.

Example 1 a) 4- (3'-Dimethylaminopropyl) 4H-benzo [4,5] cyclohepta, 2-b] thiophen-4-ol
2.2 g of iodine-activated magnesium are added
15 cc abs. Tetrahydrofuran covered and mixed with a few drops of ethylene bromide. After the start of the reaction, a solution of 10.8 g of 3-dimethylaminopropyl chloride in 20 ccm of abs. Add tetrahydrofuran dropwise so that the solvent boils and then heat the mixture to boiling for a further 1 hour. After cooling, the reaction mixture is poured into 250 cc of saturated aqueous ammonium chloride solution, 200 cc of methylene chloride is added and the whole is filtered through highly purified diatomaceous earth.
After the organic phase has been separated off, the aqueous component is extracted twice more with methylene chloride, and the combined methylene chloride solutions are washed with water, dried over magnesium sulfate and evaporated at 15 mm Hg. A solution of 4 g of 4- (3'-dimethylaminopropyl) -4H-benzo [4,5] cyclohepta [1,2-b] thiophen4-ol in 250 cc of acetic anhydride is heated to the boil for 6 hours.
After about 200 cc of solvent have been distilled off, the residue is poured into 1200 cc of water with stirring, the aqueous solution is filtered through highly purified diatomaceous earth, made strongly alkaline with 200% sodium hydroxide solution and the alkaline solution is extracted three times with ether. The combined ether extracts washed with water and dried over magnesium sulphate are then evaporated. The residue obtained is distilled in a high vacuum, the 4- (3'-dimethylamino-propylidene) -4H-benzo [4,5] -cyclohepta [1,2-b] thiophene at 160-165 "/ 0.1 mm Hg transforms.
Malonate: A solution of 3.8 g of base in 10 cc of isopropanol is mixed with a hot solution of 1.4 g of malonic acid in 10 cc of isopropanol and, after cooling, the precipitated salt is filtered off. The malonate melts after recrystallization from ethanol isopropanol at 142-143 (decomp.).
The 4H-Benzo [4,5] cyclohepta [1,2-b] thiophen-4-one used as the starting material is prepared as follows: Thenyl diethyl phosphonate
133 g of thenyl chloride (boiling point 73-75 / 15 mm Hg) are heated to 1500, whereupon 175 g of freshly distilled triethyl phosphite are slowly added dropwise with stirring at this temperature and heated to 160-170 for a further 2 hours with stirring.
The reaction mixture is then distilled in a high vacuum; the thenyl diethyl phosphonate boils at 1201240l0.06 mm Hg. o- [2-Thielzyl- (2 ') -äthyliden] -benzoic acid
To a solution of 117 g of thenyl diethyl phosphonate in 200 cc of freshly distilled dimethylformamide is added 30 g of powdered, well-dried sodium methylate, the solution warming to 45-50. The flask is then placed in an ice bath and a solution of 80 g of o-phthalaldehyde acid in 200 cc of dimethylformamide is added dropwise so that the temperature remains between 35,400 and the mixture is then stirred for a further 30 to 60 minutes at room temperature. The reaction solution is then admixed with 1600 ccm of water (temperature 10-15) with thorough cooling, a red oil separating out.
It is then made alkaline with potassium carbonate, the oil dissolving again, the red-brown solution is extracted three times with benzene and the aqueous solution is carefully adjusted to pH 4 at 10-15 with hydrochloric acid. After a few hours in the refrigerator, the precipitated acid is filtered off and dried and recrystallized from benzene. Mp. Of o- [2 thienyl- (2 ') -ethylidene] -benzoic acid 133-135.
The mother liquor is extracted three times with methylene chloride, the organic phase is dried over sodium sulfate and evaporated at 15 mm Hg. The residue is crystallized from benzene, a further portion of acid having a melting point of 133-135 being obtained. o- [2-Thienyl- (2 ') -ethyl] -benzoic acid
7.5 g of sodium are melted under anhydrous toluene, after which 375 g of pure mercury are added dropwise with frequent shaking so that the toluene boils.
The mixture is then heated to 120 to 140 with stirring and cooled to 500 as soon as all the toluene has distilled off. The homogeneous amalgam is then poured over with a solution of 20 g of o- [2-thienyl- (2 ') ethylidene] -benzoic acid in 150 cc of 950% ethanol and the mixture is shaken for 30 minutes. The mercury is then separated off, washed twice with ethanol and the combined ethanolic solutions are diluted with 1200 cc of water. The solution is filtered through highly purified diatomaceous earth, acidified with hydrochloric acid and cooled to 5. After a few hours, the acid which has precipitated is filtered off and crystallized from chloroform / hexane. M.p. 110-111 ".
9.1 O-Dihydro-4H-benzo [4.5] cyclohepta [1,2-b] -thiophen-4-one
59 cc of 840% phosphoric acid and 86 g of phosphorus pentoxide are first stirred at 125-1300 for 30 minutes. Then 20 g of pulverized o- [2-thienyl- (2 ') -ethyl] -benzoic acid are introduced over 30 minutes at this temperature. The reaction mixture is stirred for a further 2 hours at 125-130, it is poured into
1000 ccm of water, filtered the solution through highly purified diatomaceous earth and extracted it three times with methylene chloride. The organic phase is with 2-n.
Washed sodium carbonate solution, dried over magnesium sulfate, the solvent evaporated and the residue distilled in a high vacuum, the 9,10 dihydro-4H-benzo [4,5] cyclohepta [1,2-b] thiophen-4 - one at 125-1400 / 0.05 mm Hg passes as a green oil. n2D = 1.6559.
4H-Benzo [4,5] cyclohepta [1 2-b] thiophen-4-one
To introduce the double bond in the 9,10 position, the procedure is that a mixture of 32.1 g of the dihydro compound obtained, 26.7 g of N bromosuccinimide and 0.3 g of benzoyl peroxide in 250 ccm of abs. Carbon tetrachloride heated to boiling for 4 hours. After cooling to 50, the reaction mixture is filtered through highly purified diatomaceous earth and the solvent is evaporated at 15 mm Hg. The oily residue obtained is then heated for 2 hours with 200 cc of triethylamine while stirring.
After evaporation of the unchanged triethylamine, 250 cc of methylene chloride are added to the residue and the resulting solution is treated three times with 2-n. Hydrochloric acid and washed twice with water. After drying the solution over magnesium sulfate, the solvent is removed under reduced pressure. The residue is then distilled in a high vacuum, the 4H-benzo [4,5] cyclohepta [1,2-b] thiophen-4-one passing over as an oil at 173-1,800 / 0.1 mm Hg and crystallizing on cooling. M.p. 109-110 after recrystallization from ethanol.
Example 2 4- (2 '- [1 "-Methyl-piperidyl- (2") 1-ethylidene) -4H-benzo- [4,5] cyclohepta [1,2-b] thiophene
2.2 g of magnesium activated with iodine are covered with a layer of 15 cc of tetrahydrofuran and a few drops of ethylene bromide are added. After the reaction has started, a solution of 12.6 g of 2- [1'-methylpiperidyl- (2 ')] ethyl chloride in 15 cc of tetrahydrofuran is added dropwise so that the solvent boils and then heated to boiling for a further 1/2 hour. A solution of 8.2 g of 4H-benzo [4,5] cyclohepta [1,2-b] thiophen-4-one in 20 cc of tetrahydrofuran is then added dropwise over a period of 15 minutes at 20 and a further S / 4 is heated with stirring Hours to 400.
After cooling, the reaction mixture is poured into 250 cc of 200% ammonium chloride solution, diluted with 200 cc of methylene chloride and filtered through highly purified diatomaceous earth. After the organic phase has been separated off, the aqueous part is extracted three times with methylene chloride, and the combined methylene chloride extracts are washed with water, dried over sodium sulphate and evaporated at 15 mm Hg.
The oily 4- (2 '- [l "-Methyl-piperidyl- (2") 1-ethyl 4H-benzo [4,5] cyclohepta [1,2-b] thiophen-4-ol is then dissolved in 200 ccm Hydrochloric acid (1: 1) dissolved, the acidic solution washed twice with ether and then made strongly alkaline with sodium hydroxide solution with cooling.This aqueous solution is extracted three times with methylene chloride, the methylene chloride extracts washed with water, dried over magnesium sulfate and evaporated at 15 mm Hg. The oily residue is dissolved in isopropanol and an isopropanolic salicylic acid solution is added. After cooling, the precipitated 4- {2 '- [1 "methyl-piperidyl- (2") 1- ethylidene) -4H-benzo [4,5] cyclo Hepta [l, 2-b] thiophene salicylate is filtered off and recrystallized from isopropanol.
