Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung neuer heterocyclischer Verbindungen der allgemeinen Formel 1

worin entweder R, und R2 Wasserstoff oder eine niedere Alkylgruppe und R3 und R4 je eine niedere Alkylgruppe bedeuten, bzw. R3 und Ri zusammen die Tetramethylenoder die Pentamethylengruppe bilden, oder R4 für eine niedere Alkylgruppe steht und R3 entweder mit Ri oder mit R2 zusammen eine Di-, Tri-oder Tetramethylengruppe bildet, und ihrer Säureadditionssalze mit organischen und anorganischen Säuren.
Erfindungsgemäss gelangt man zu diesen Verbindungen der allgemeinen Formel I und ihren Salzen, indem man 9, 10-Dihydro-4H-benzo [4, 5] cyclohepta [1, 2-b] thiophen-4-on mit einer Grignard-Verbindung der allgemeinen Formel II,

aus diesen anschliessend Wasser abspaltet und die so erhaltenen Verbindungen der allgemeinen Formel I gegebenenfalls in an sich bekannter Weise in ihre Säureadditionssalze überführt.
Das Verfahren kann vorteilhafterweise so ausgeführt werden, dass man eine Lösung des 9, 10-Dihydra-4H benzo [4, 5] cyclohepta [1, 2-b] thiophen-4-on in einem inerten organischen Lösungsmittel zu einer Lösung einer Verbindung der allgemeinen Formel II in einem organischen Lösungsmittel zutropfen und das Reaktionsge- misch einige Zeit bei Zimmertemperatur stehen lässt, bzw. anschliessend noch zum Sieden erhitzt. Die so erhaltenen Reaktionsprodukte werden auf geeignete Weise, z. B. durch Behandlung mit eiskalter wässriger Ammoniumchloridlösung, zu Verbindungen der allgemeinen Formel III hydrolysiert, die auf an sich bekannte Weise isoliert werden können.
Aus diesen Verbindungen der allgemeinen Formel III können dann durch Einwirkung geeigneter wasserabspaltender Mittel die Verbindungen der allgemeinen Formel I gewonnen werden. Als wasserabspaltende Mittel können zum Beispiel Minerasäuren, starke organische Säuren, Essigsäureanhydrid, Thionylchlorid oder Phosphoroxychlorid verwendet werden. Die Überführung der so erhaltenen Verbindungen der allgemeinen Formel 1 in ihre Säureadditionssalze kann durch Behandlung mit organischen oder anorgani- schen Säuren in an sich bekannter Weise geschehen.
Das 9, 10-Dihydro-4H-benzo [4, 5] cyclohepta [1, 2-b] thiophen-4-on, eine bisher unbekannte Verbindung, kann auf folgendem Weg hergestellt werden : Man kondensiert Thenyl-diäthyl-phosphonat in einem geeigneten wasserfreien organischen Lösungsmittel und in Gegenwart eines alkalischen Kondensationsmittels mit o-Phthalaldehyd- saure ; reduziert die so erhaltene o-[2-Thienyl-(2')-vinyl]- benzoesäure zur o- [2-Thienyl-(2')-äthyl]-benzoesäure und unterwirft diese einem intramolekularen Ringschluss.
Als Reduktionsmittel kann beispielsweise Natriumamalgam in wässrigem Alkohol verwendet werden, als Kon densationsmittel für den Ringschluss Polyphosphorsäure.
Die verfahrensgemäss hergestellten neuen Verbindungen sind bei Raumtemperatur kristallin oder ölig und bilden mit anorganischen oder organischen Säuren beständige, bei Raumtemperatur kristallisierte Salze.
Die Verbindungen selbst sowie ihre Salze mit pharmazeutisch verträglichen organischen oder anorganischen Säuren zeichnen sich durch eine starke neuroleptische und antidepressive Wirkung aus und sollen daher vor allem zur Behandlung neurotischer und psychotischer Störungen Verwendung finden.
In den nachfolgenden Beispielen erfolgen alle Temperaturangaben in Celsiusgraden. Die Schmelzpunkte sind unkorrigiert.
Beispiel 1 4- (3'-Dimethylamino-propyliden)-9, 10-dihydro-4H- benzoF4, 5] cyclohepta [1, 2-b] thiophen-hydrochlorid a) 4- (3'-Dimethylaminopropyl)-9, 10-dihydro-4H benzo [4, 5] cyclohepta [1, 2-b] thiophen-4-ol
2, 2 g mit Jod aktiviertes Magnesium werden mit wenig Tetrahydrofuran überschichtet und mit einigen Tropfen Athylenbromid versetzt. Nach Einsetzen der Reaktion lässt man eine Lösung von 10, 8 g 3-Dimethyl aminopropylchlorid in 20 ml Tetrahydrofuran so zutropfen, dass das Lösungsmittel siedet, und erhitzt anschlies- send noch 2 Std. zum Sieden.
Man lässt nun innert 10 Min. eine Lösung von 8, 2 g 9, 10-Dihydro-4H-benzo [4, 5] cyclohepta [1, 2-b] thiophen-4-on in 30 ml Tetrahydrofuran zutropfen und erhitzt unter Rühren noch weitere
10 Min. zum Sieden.
Nach Abkühlung wird das Reaktionsgemisch in eine Lösung von 30 g Ammoniumchlorid in 200 ml Wasser gegossen, 200 ml Methylenchlorid zugesetzt und das Ganze durch Hyflo abfiltriert. Nach dem Abtrennen der organischen Phase wird der wässrige Teil noch zweimal mit Methylenchlorid ausgeschüttelt, die vereinigten Methylenchloridlösungen werden mit Wasser gewaschen, über Magnesiumsulfat getrocknet und im Vakuum eingedampft.
Der ölige Rückstand wird aus l2ither/Petroläther kristallisiert. Smp. des 4- (3'-Dimethylaminopropyl)-9, 10-di- hydro4H-benz [4, 5] cyclohepta [1, 2-b] thiopehn-4-ol : 101 . b) 4- (3'-Dimethylamino-propyliden)-9, 10-dihydro-4H- benzo [4, 5] cyclohepta [1, 2-b] thiophen-hydrochlorid
8 g 4- (3'-Dimethylaminopropyl)-9, 10-dihydro-4H- benzo [4, 5] cyclohepta [I, 2-b] thiophen-4-ol, 80 ml Eisessig und 32 ml konz. Salzsäure werden 30 Min. unter Rückfluss gekocht, das Reaktionsgemisch im Vakuum eingedampft und der Rückstand mit Athanol/Äther 1 : 1 verrieben.
Das ausgefallene Hydrochlorid wird abfiltriert und aus Äthanol/Äther umkristallisiert. Smp. des 4- (3'- Dimethylamino-propyliden)-9, 10-dihydro-4H-benzo [4, 5] cyclohepta [1, 2-b] thiophen-hydrochlorid : 222-224 (Zers.).
Das als Ausgangsmaterial verwendete 9, 10-Dihydro4H-benzo [4, 5] cycloheptafl, 2-b] thiophen-4-on kann wie folgt erhalten werden : a) Thenyl-diäthyl-phosphonat
133 g Thenyl-chlorid (Kp. 73-75 /15 mm) werden auf 150 erhitzt, worauf man unter Rühren bei dieser Temperatur 175 g frisch destilliertes Triäthylphosphit langsam zutropfen lässt und noch weitere 2 Stunden unter Rühren auf 160-170 erhitzt.
Das Reaktionsge- misch wird dann im Hoch vakuum destilliert ; das Thenyl diäthyl-phosphonat siedet bei 120-124 /0, 06 mm. b) o-[2-Thienyl-(2')-äthylen]-benzoesäure
Zu einer Lösung von 117 g Thenyl-diäthyl-phos- phonat in 200 ml frisch destilliertem Dimethylformamid gibt man 30 g pulverisiertes, gut getrocknetes Natriummethylat, wobei sich die Lösung auf 45-50 erwärmt.
Dann stellt man den Kolben in ein Eisbad und lässt eine Lösung von 80 g o-Phthalaldehydsäure in 200 ml Dimethylformamid so zutropfen, dass die Temperatur zwischen 35-40 bleibt und rührt dann noch /2-1 Std. bei Zimmertemperatur weiter. Die Reaktionslösung wird nun unter gutem Kühlen mit 1600 ml Wasser (Temperatur 10-15 ) versetzt, wobei sich ein rotes Öl abschei- det. Man stellt dann mit Pottasche alkalisch, wobei sich das Ol wieder löst, schüttelt die rotbraune Lösung dreimal mit Benzol aus und stellt die wässrige Lösung vor- sichtig mit Salzsäure auf pH 4.
Nach einigen Stunden im Kühlschrank wird die ausgefallene Säure abfiltriert, getrocknet und aus Benzol umkristallisiert. Smp. der o2-Thienyl- (2')-äthylen]-benzoesäure 133-135 .
Die Mutterlauge wird dreimal mit Methylenchlorid ausgeschüttelt, die organische Phase über Natriumsulfat getrocknet und im Vakuum eingedampft. Der Rückstand wird aus Benzol kristallisiert, wobei noch eine weitere Portion Säure vom Smp. 133-135 erhalten wird. c) o- [2-Thienyl- (2')-äthyl]-benzoesäure
7, 5 g Natrium werden unter wasserfreiem Toluol ge schmolzen, worauf man unter öfterem Schütteln 375 g reines Quecksilber so zutropft, dass das Toluol siedet.
Dann erhitzt man das Gemisch unter Rühren auf 120140 und kühlt, sobald alles Toluol abdestilliert ist, auf 50 ab. Das homogene Amalgam übergiesst man nun mit einer Lösung von 20 g o¯ [2-Thienyl- (2')-äthylen]- benzoesäure in 150 ml 95prozentigem Äthanol und schüttelt die Mischung/2 Std. lang. Darauf trennt man das Quecksilber ab, wäscht es zweimal mit Äthanol und verdünnt die vereinigten äthanolischen Lösungen mit
1200 ml Wasser. Die Lösung wird durch Hyflo filtriert, mit Salzsäure angesäuert und auf 5 abgekiihlt. Nach einigen Stunden filtriert man die ausgefallene Säure ab und kristallisiert sie aus Chloroform/Hexan um. Smp.
110-111 . d) 9, 10-Dihydro-4H-benza [4, 5] cyclohepta [1, 2-b] thiophen- (4)-on
59 ml 84"/oigne Phosphorsäure und 86 g Phosphorpentoxyd werden zuerst bei 125-130 1/2 Std. lang gerührt. Dann werden bei dieser Temperatur 20 g pulverisierte o-[2-Thienyl-(2')-äthyl]-benzoesäure während Va Std. eingetragen. Man rührt das Reaktionsgemisch noch 2 Std. bei 125-130 , giesst es in 1 Liter Wasser, filtriert die Lösung durch Hyflo und extrahiert sie dreimal mit Methylenchlorid.
Die organische Phase wird mit 2n Sodalösung gewaschen, über Magnesiumsulfat getrocknet, das Lösungsmittel abgedampft und der Rückstand im Hochvakuum destilliert, wobei das 9, 10-Dihydro-4H-benzo [4, 5] cyclohepta [1, 2-b]-thiophen- (4)-on bei 125-140 /0, 05 mm als grünes 61 übergeht. nD24 = 1, 6559.
Beispiel 2 4- {2'- [1"-Methyl-pyrrolidyl- (2")]-äthyliden}-9, 10-di hydro-4H-benzo [4, 5] cyclohepta [1, 2-b] thiophen a) 4-{2'-[1"-Methyl-pyrrolidyl-(2")]-äthyl}-9, 10 dihydro-4H-benzo [4, 5] cyclohepta [1, 2-b] thiophen
4-ol
1, 64 g mit Jod aktiviertes Magnesium werden mit 10 ccm Tetrahydrofuran überschichtet und mit einigen Tropfen Äthylenbromid versetzt. Nach Einsetzen der Reaktion lässt man eine Lösung von 9, 6 g 1-Methyl-2 (iS-chloräthyl)-pyrrolidin in 10 ccm Tetrahydrofuran so zutropfen, dass das Lösungsmittel siedet, und erhitzt anschliessend noch 3/4 Std. zum Sieden.
Man lässt nun bei 20 innert 15 Min. eine Lösung von 7, 8 g 9, 10-Dihydro4H-benzo [4, 5] cyclohepta [1, 2-b] thiophen-4-on in 15 ccm Tetrahydrofuran zutropfen und erhitzt unter Rüh- ren noch weitere 15 Min. zum Sieden. Nach Abkühlen wird das Reaktionsgemisch in 200 ccm 20"/oigne Am moniumchloridlösung gegossen, 200 ccm Methylenchlorid zugesetzt, und das ganze durch hochgereinigte Diatomeenerde filtriert. Nach dem Abtrennen der organischen Phase wird der wässerige Teil noch zweimal mit Methylenchlorid ausgeschüttelt, die vereinigten Methylenchloridlösungen mit Wasser gewaschen, über Magnesiumsulfat getrocknet und bei 15 mm Hg eingedampft.
Der Rückstand wird mit wenig Äther verrieben, die ausgefallene Substanz abfiltriert und aus Hexan umkristallisiert. Das 4-2'- [1"-Methyl-pyrrolidyl (2")] äthyl}-9, 10-dihydro-4H-benzo [4, 5] cyclohepta [1, 2-b]thiophen-4-ol schmilzt bei 105-106 . b) 4-{2' < [1"-Methyl-pyrrolidyl-(2")]-äthyliden}-9, 10-di hydro-4H-benzo [4, 5] cyclohepta [1, 2-b] thiophen
3, 27 g 4-f T- [1"-Methyl-pyrrolidyl- (2")]-äthyl)- 9, 10-dihydro-4H-benzo [4, 5] cyclohepta [1, 2-b] thiophen4-ol werden in 10 ccm 2-n. isopropanolischer Salzsäure- lösung gelöst, mit 15 ccm Isopropanol verdünnt und 5 Min.
auf 50 erwännt. Unter Rühren lässt man nun langsam auf Raumtemperatur abkühlen, filtriert die Lösung durch Tierkohle und dampft das Lösungsmittel bei 15 mm Hg ein. Der Rückstand wird in Wasser gelöst, die Lösung mit Natronlauge unter Kühlen alkalisch gestellt und dreimal mit Äther extrahiert. Der Atherextrakt wird mit Wasser gewaschen, über Magnesiumsulfat getrocknet und eingedampft. Der Rückstand wird mit 10 ccm Isopropanol gelöst und mit einer Lösung von 1, 0 g Fumarsäure in 4 ccm Isopropanol versetzt. Nach dem Abkühlen wird das ausgefallene Fumarat abfiltriert und aus Methanonsopropanol umkristallisiert.
Man erhält 4-- (2'- [1"-Methyl-pyrrolidyl- (2") l-äthyliden1-9, 10dihydro-4-benzo [4, 5] cyclohepta [1, 2-b] thiophen-hydro- gen-fumarat vom Smp. 203-205 (Zers.).
Beispiel 3 4- {2'- [l"-Methylpiperidyl- (2")]-äthyliden}-9, 10-di hydro-4H-benzo [4, 5] cyclohepta [1, 2-b] thiophen
1, 2 g mit Jod aktiviertes Magnesium wird mit Tetra hydrofuran überschichtet und mit einigen Tropfen Äthylenbromid versetzt. Nach Einsetzen der Reaktion lässt man eine Lösung von 7, 9 g 2- [1'-Methylpiperidyl- (2')]-äthylchlcrid in 10 ml Tetrahydrofuran so zutropfen, dass das Lösungsmittel siedet, und erhitzt noch 1 Std. zum Sieden.
Man lässt nun innert 15 Min. eine Lösung von 5, 2 g 9, 10-Dihydro-4H-benza [4, 5] cyclohepta [1, 2-b] thiophen-4-on in 10 ml Tetrahydrofuran zutropfen und erhitzt noch unter Rühren weitere 15 Min. zum Sieden. Nach Abkühlen wird das Reaktionsgemisch in eine Lösung 30 g Ammoniumchlorid in 150 ml Wasser gegossen, mit 150 ml Methylenchlorid verdünnt und das Ganze durch Hyflo filtriert. Nach dem Abtrennen der organischen Phase wird die wässrige Phase zweimal mit Methylenchlorid ausgeschüttelt, die vereinigten Methylenchloridlösungen mit Wasser gewaschen, über Pottasche getrocknet und das Lösungsmittel im Vakuum abgedampft.
Der ölige Rückstand wird in l Seiger Salzsäure gelöst, die Lösung zweimal mit Äther gewaschen, unter Kühlung mit Soda alkalisch gestellt und dreimal mit Methylenchlorid ausgeschüttelt. Der Methylenchloridextrakt wird über Pottasche getrocknet, das Lösungs mittel im Vakuum abgedampft und der Rückstand aus Äthanol umkristallisiert. Smp. des 4-f2'- [l"-Methyl- piperidyl- (2")]-äthyliden}-9, 10-dihydro-4H-benzo [4, 5]- cyclohepta [1, 2-b] thiophen : 115-115, 5 .
Beispiel 4 4-(3'-DiäthylaminoWpropyliden)-9, 10-dihydro-4H- benzo [4, 5] cyclohepta [1, 2-b] thiophen a) 4- (3'-Diäthylaminopropyl)-9, 10-dihydro-4H-benzo- 1 [4, 5] cyclohepta [1, 2-b] thiophen-4-ol
1, 64 g mit Jod aktiviertes Magnesium werden mit 10 ml Tetrahydrofuran überschichtet und mit einigen Tropfen Äthylenbromid versetzt. Nach Einsetzen der Reaktion lässt man eine Lösung von 9, 8 g 3-Diäthyl- aminopropylenchlorid in 10 ml Tetrahydrofuran so zutropfen, dass das Lösungsmittel siedet und erhitzt anschliessend noch 1t/2 Std. zum Sieden.
Man lässt nun bei 20 innert 15 Min. eine Lösung von 7, 8 g 9, 10-Dihydro4H-benzo [4, 5] cyclohepta [1, 2-b] thiophen-4-on in 15 ml Tetrahydrofuran zutropfen und erhitzt unter Rühren noch weitere 30 Min. zum Sieden.
Nach Abkühlen wird das Reaktionsgemisch in 200 ml gesättigte Ammoniumchloridlösung gegossen, 200 ml Methylenchlorid zugesetzt und das Ganze durch hochgereinigte Fullererde abfiltriert. Nach dem Abtrennen der organischen Phase wird der wässrige Teil noch zweimal mit Methylenchlorid ausgeschüttelt, die vereinigten Methylenchloridlösungen werden mit Wasser gewaschen, über Magnesiumsulfat getrocknet und im Vakuum eingedampft.
Der Rückstand wird aus Isopropanol umkristallisiert und liefert 4- (3'-Diäthylamino propyl)-9, 10-dihydro-4H-benzo [4, 5] cyclohepta [1, 2-b] thiophen-4-ol vom Smp. 104, 5-105, 5 . b) 4- (3'-Diäthylamino-propyliden)-9, 10-dihydro-4H benzo [4, 5] cyclohepta [1, 2-b] thiophen
6 g 4-(3'-Diäthylaminopropyl)-9, 10-dihydro-4H- benzo [4, 5] cyclohepta [1, 2-b] thiophen-4-ol, 80 ml Eisessig und 32 ml konz. Salzsäure werden 1 Std. unter Rückfluss gekocht, das Reaktionsgemisch wird im Vakuum eingedampft und der Rückstand in Wasser gelöst.
Die wässrige Lösung wird mit Natronlauge stark alkalisch gestellt, dann dreimal mit Äther ausgeschüttelt, das Extrakt mit Magnesiumsulfat getrocknet und das Lösungsmittel eingedampft. Der ölige Rückstand wird in Aceton gelöst, mit einer ätherischen Chlorwasserstoff- lösung versetzt und das ausgefallene Hydrochlorid aus Isopropanol umkristallisiert. Smp. 175, 5-177 .
Beispiel 5 4- (3'-Dimethylamino-2'-methylpropyliden)-9, 10-di hydro-4H-benzo [4, 5] cyclohepta [1, 2-b] thiophen a) 4- (3'-Dimethylamino-2'-methylpropyl)-9, 10-di hydro-4H-benzo [4, 5] cyclohepta [1, 2-b] thiophen-
4-ol Gemäss vorhergehendem Beispiel, aus 1, 64 g Magnesium, 8, 8 g 3-Dimethylamino-2-methyl-propylchlorid und 7, 8 g 9, 10-Dihydro-4H-benzo [4, 5] cyclohepta [1, 2-b]-thiophen-4-on in 35 ml Tetrahydrofuran. Smp.
115-116 nach Umkristallisieren aus Hexan. b) 4- (3'-Dimethylamino-2'-methylpropyliden)-9, 10-di hydro-4H-benzo [4, 5] cyclohepta [1, 2-b] thiophen Gemäss vorhergehendem Beispiel, aus 7, 5 g 4- (3' Dimethylamino-2'-methylpropyl)-9, 10-dihydro-4Hbenzo [4, 5] cyclohepta [1, 2-b]thiophen-4-ol in 80 ml Eisessig und 32 ml konz. Salzsäure, durch einstündiges Erhitzen zum Sieden. Smp. des Hydrochlorides 229-231 (Zers.) nach Umkristallisieren aus Isopropanol.
Beispiel 6 4- [3'- (1"-Pyrrolidyl)-propyliden]-9, 10-dihydro-4H benzo [4, 5] cyclohepta [1, 2-b] thiophen a) 4- [3'- (1"-Pyrrolidyl)-propyl]-9, 10-dihydro-4H- benzo [4, 5] cyclohepta [l, 2-b] thiophen-4-ol Gemäss vorhergehendem Beispiel, aus 2, 13 g Magnesium, 12, 5 g 3- (1'-Pyrrolidyl)-propylchlorid und 10, 3 g 9, 10-Dihydro4H-benzo [4, 5] cyclohepta [1, 2-b]- thiophen-4-on in 50 ml Tetrahydrofuran.
Smp. 132 133 nach Umkristallisieren aus Äthanol 95%. b) 4- [3'- (1"-Pyrrolidyl)-propyliden]-9, 10-dihydro-4H benzo [4, 5] cyclohepta [1, 2-b] thiophen Gemäss vorhergehendem Beispiel, aus 8 g 4- [3'-1"- Pyrrolidyl)-propyl]-9, 10-dihydro-4H-benzo [4, 5] cyclohepta[1,2-b]thiophen-4-ol in 80 ml Eisessig und 32 ml konz. SalzsÏure, durch einst ndiges Erhitzen zum Sieden.
Smp. des Hydrochlorides 215-217 (Zers.) nach Umkristallisieren aus Isopropanol/Äther.
Beispiel 7 4- [3'- (1"-Piperidyl)-propyliden]-9, 10-dihydro-4H benzo [4, 5] cyclohepta [1, 2-b] thiophen a) 4- [3'- (1"-Piperidyl)-propyl]-9, 10-dihydro-4H- benzo [4, 5] cyclohepta [1, 2-b] thiophen-4-ol Gemäss vorhergehendem Beispiel, auf 1, 64 g Magnesium, 10,5 g 3-(1'-Piperidyl)-propylchlorid und 7,8 g 9,10-dihydro-4H-benzo[4,5]cyclohepta[1,2-b]thiophen4-on in 40 ml Tetrahydrofuran.
Smp. 129-130¯ nach Umkristallisieren aus ¯thanol 80 %. b) 4- [3'- (1"-Piperidyl)-propyliden]-9, 10-dihydro-4H benzo [4, 5] cyclohepta [1, 2-b] thiophen Gemäss vorhergehendem Beispiel, aus 8 g 4- [3'- (1"- Piperidyl) -propyl] - 9,10-dihydro-4H-benzo,[4,5] cyclohepta[1,2-b]thiophen-4-ol in 80 ml Eisessig und 32 ml konz. SalzsÏure, durch einst ndiges Erhitzen zum Sieden. Die Base wird im Hochvakuum destilliert ; Sdp.
205-210 /0, 3 mm Hg. Das Destillat wird in Aceton gelöst, die Lösung mit Chlorwasserstoffgas gesättigt und nach 8-10 Std. das auskristallisierte Hydrochlorid abfiltriert ; Smp. 169-170 nach Umkristallisieren aus Äthanol/Isopropanol.

According to the invention, these compounds of the general formula I and their salts are obtained by adding 9, 10-dihydro-4H-benzo [4, 5] cyclohepta [1, 2-b] thiophen-4-one with a Grignard compound of the general Formula II,

The process can advantageously be carried out so that a solution of the 9, 10-dihydra-4H benzo [4, 5] cyclohepta [1, 2-b] thiophen-4-one in an inert organic solvent to a solution of a compound of General formula II in an organic solvent is added dropwise and the reaction mixture is left to stand for some time at room temperature or is then heated to the boil. The reaction products thus obtained are suitably treated, e.g. B. by treatment with ice-cold aqueous ammonium chloride solution, hydrolyzed to compounds of general formula III, which can be isolated in a known manner.
The compounds of general formula I can then be obtained from these compounds of general formula III by the action of suitable water-releasing agents. Mineral acids, strong organic acids, acetic anhydride, thionyl chloride or phosphorus oxychloride, for example, can be used as dehydrating agents. The compounds of general formula 1 thus obtained can be converted into their acid addition salts by treatment with organic or inorganic acids in a manner known per se.
The 9, 10-dihydro-4H-benzo [4, 5] cyclohepta [1, 2-b] thiophen-4-one, a previously unknown compound, can be prepared in the following way: Thenyl diethyl phosphonate is condensed in one suitable anhydrous organic solvent and in the presence of an alkaline condensing agent with o-phthalaldehyde acid; reduces the o- [2-thienyl- (2 ') -vinyl] -benzoic acid obtained in this way to o- [2-thienyl- (2') -ethyl] -benzoic acid and subjects this to an intramolecular ring closure.
Sodium amalgam in aqueous alcohol can be used as a reducing agent, for example, and polyphosphoric acid as a condensation agent for the ring closure.
The new compounds prepared according to the process are crystalline or oily at room temperature and form stable salts with inorganic or organic acids which crystallize at room temperature.
The compounds themselves and their salts with pharmaceutically acceptable organic or inorganic acids are distinguished by a strong neuroleptic and antidepressant effect and should therefore be used primarily for the treatment of neurotic and psychotic disorders.
In the following examples, all temperatures are given in degrees Celsius. The melting points are uncorrected.
Example 1 4- (3'-Dimethylamino-propylidene) -9, 10-dihydro-4H-benzoF4, 5] cyclohepta [1, 2-b] thiophene hydrochloride a) 4- (3'-Dimethylaminopropyl) -9, 10 -dihydro-4H benzo [4,5] cyclohepta [1,2-b] thiophen-4-ol
2.2 g of magnesium activated with iodine are covered with a little tetrahydrofuran and mixed with a few drops of ethylene bromide. After the reaction has started, a solution of 10.8 g of 3-dimethyl aminopropyl chloride in 20 ml of tetrahydrofuran is added dropwise in such a way that the solvent boils, and the mixture is then heated to boiling for a further 2 hours.
A solution of 8.2 g of 9, 10-dihydro-4H-benzo [4, 5] cyclohepta [1, 2-b] thiophen-4-one in 30 ml of tetrahydrofuran is then added dropwise within 10 minutes and the mixture is heated with stirring even more
10 minutes to simmer.
After cooling, the reaction mixture is poured into a solution of 30 g of ammonium chloride in 200 ml of water, 200 ml of methylene chloride are added and the whole is filtered off through Hyflo. After the organic phase has been separated off, the aqueous part is extracted twice more with methylene chloride, the combined methylene chloride solutions are washed with water, dried over magnesium sulphate and evaporated in vacuo.
The oily residue is crystallized from l2ither / petroleum ether. M.p. des 4- (3'-dimethylaminopropyl) -9,10-di-hydro4H-benz [4,5] cyclohepta [1,2-b] thiophen-4-ol: 101. b) 4- (3'-Dimethylamino-propylidene) -9, 10-dihydro-4H-benzo [4, 5] cyclohepta [1, 2-b] thiophene hydrochloride
8 g of 4- (3'-dimethylaminopropyl) -9, 10-dihydro-4H-benzo [4, 5] cyclohepta [I, 2-b] thiophen-4-ol, 80 ml of glacial acetic acid and 32 ml of conc. Hydrochloric acid is refluxed for 30 minutes, the reaction mixture is evaporated in vacuo and the residue is triturated with ethanol / ether 1: 1.
The precipitated hydrochloride is filtered off and recrystallized from ethanol / ether. M.p. des 4- (3'-dimethylamino-propylidene) -9, 10-dihydro-4H-benzo [4,5] cyclohepta [1,2-b] thiophene hydrochloride: 222-224 (dec.).
The 9, 10-dihydro4H-benzo [4, 5] cycloheptafl, 2-b] thiophen-4-one used as starting material can be obtained as follows: a) Thenyl diethyl phosphonate
133 g of thenyl chloride (b.p. 73-75 / 15 mm) are heated to 150, whereupon 175 g of freshly distilled triethyl phosphite are slowly added dropwise with stirring at this temperature and heated to 160-170 for a further 2 hours with stirring.
The reaction mixture is then distilled in a high vacuum; the thenyl diethyl phosphonate boils at 120-124 / 0.06 mm. b) o- [2-thienyl- (2 ') -ethylene] -benzoic acid
To a solution of 117 g of thenyl diethyl phosphonate in 200 ml of freshly distilled dimethylformamide is added 30 g of powdered, well-dried sodium methylate, the solution warming to 45-50.
The flask is then placed in an ice bath and a solution of 80 g of o-phthalaldehyde acid in 200 ml of dimethylformamide is added dropwise so that the temperature remains between 35-40 and the mixture is then stirred for a further 2-1 hours at room temperature. The reaction solution is then admixed with 1600 ml of water (temperature 10-15) with good cooling, a red oil separating out. It is then made alkaline with potash, the oil dissolving again, the red-brown solution is extracted three times with benzene and the aqueous solution is carefully adjusted to pH 4 with hydrochloric acid.
After a few hours in the refrigerator, the precipitated acid is filtered off, dried and recrystallized from benzene. Mp. Of the o2-thienyl- (2 ') -ethylene] -benzoic acid 133-135.
The mother liquor is extracted three times with methylene chloride, the organic phase is dried over sodium sulfate and evaporated in vacuo. The residue is crystallized from benzene, a further portion of acid having a melting point of 133-135 being obtained. c) o- [2-thienyl- (2 ') -ethyl] -benzoic acid
7.5 g of sodium are melted under anhydrous toluene, whereupon 375 g of pure mercury are added dropwise with repeated shaking so that the toluene boils.
The mixture is then heated to 120-140 with stirring and cooled to 50 as soon as all of the toluene has distilled off. The homogeneous amalgam is now poured over with a solution of 20 g of [2-thienyl- (2 ') - ethylene] - benzoic acid in 150 ml of 95 percent ethanol and the mixture is shaken for 2 hours. The mercury is then separated off, washed twice with ethanol and the combined ethanolic solutions are diluted with it
1200 ml of water. The solution is filtered through Hyflo, acidified with hydrochloric acid and cooled to 5. After a few hours, the acid which has precipitated is filtered off and recrystallized from chloroform / hexane. M.p.
110-111. d) 9,10-dihydro-4H-benza [4,5] cyclohepta [1,2-b] thiophen- (4) -one
59 ml of 84% phosphoric acid and 86 g of phosphorus pentoxide are first stirred at 125-130 1/2 hours. Then 20 g of pulverized o- [2-thienyl- (2 ') -ethyl] -benzoic acid are added at this temperature The reaction mixture is stirred for a further 2 hours at 125-130, poured into 1 liter of water, the solution filtered through Hyflo and extracted three times with methylene chloride.
The organic phase is washed with 2N sodium carbonate solution, dried over magnesium sulfate, the solvent is evaporated and the residue is distilled in a high vacuum, the 9, 10-dihydro-4H-benzo [4, 5] cyclohepta [1, 2-b] -thiophene- (4) -on at 125-140 / 0.05 mm transitions as green 61. nD24 = 1.6559.
Example 2 4- {2'- [1 "-Methyl-pyrrolidyl- (2")] -ethylidene} -9, 10-di hydro-4H-benzo [4,5] cyclohepta [1,2-b] thiophene a ) 4- {2 '- [1 "-Methyl-pyrrolidyl- (2")] - ethyl} -9, 10 dihydro-4H-benzo [4,5] cyclohepta [1,2-b] thiophene

1.64 g of magnesium activated with iodine are covered with a layer of 10 ccm of tetrahydrofuran and a few drops of ethylene bromide are added. After the reaction has started, a solution of 9.6 g of 1-methyl-2 (iS-chloroethyl) pyrrolidine in 10 cc of tetrahydrofuran is added dropwise so that the solvent boils, and the mixture is then heated to boiling for a further 3/4 hours.
A solution of 7.8 g of 9, 10-dihydro4H-benzo [4, 5] cyclohepta [1, 2-b] thiophen-4-one in 15 cc of tetrahydrofuran is then added dropwise over the course of 15 minutes and the mixture is heated with stirring - simmer for another 15 minutes. After cooling, the reaction mixture is poured into 200 cc of 20 "/ oigne ammonium chloride solution, 200 cc of methylene chloride is added, and the whole is filtered through highly purified diatomaceous earth. After the organic phase has been separated off, the aqueous part is extracted twice more with methylene chloride and the combined methylene chloride solutions with water washed, dried over magnesium sulfate and evaporated at 15 mm Hg.
The residue is triturated with a little ether, the precipitated substance is filtered off and recrystallized from hexane. Example 3 4- {2'- [l "-Methylpiperidyl- (2")] ethylidene} -9,10-dihydro-4H-benzo [4,5] cyclohepta [1,2-b] thiophene
1.2 g of magnesium activated with iodine is covered with tetrahydrofuran and a few drops of ethylene bromide are added. After the reaction has started, a solution of 7.7 g of 2- [1'-methylpiperidyl- (2 ')] ethyl chloride in 10 ml of tetrahydrofuran is added dropwise so that the solvent boils, and the mixture is heated to boiling for a further 1 hour.
A solution of 5.2 g of 9, 10-dihydro-4H-benza [4, 5] cyclohepta [1, 2-b] thiophen-4-one in 10 ml of tetrahydrofuran is then added dropwise within 15 minutes and the mixture is heated under Stir for a further 15 min.
