d'un ester réactif de formule générale Y-A-Z (3) dans laquelle A et Z sont définis comme précédem ment et Y représente un atome d'halogène, un reste d'ester sulfurique, par exemple alcoyloxysulfonyl- oxyle,
ou un reste d'ester sulfonique, en particulier alcanesulfonyloxyle, benzènesulfonyloxyle ou p. to- luènesulfonyloxyle.
La réaction peut être effectuée avec ou sans solvant en présence ou non d'un agent de conden sation. I1 est avantageux d'opérer dans un solvant du groupe des hydrocarbures aromatiques (par exem ple toluène ou xylène), des éthers (par exemple l'éther éthylique) ou des. amides tertiaires {par exem ple le diméthylformamide) en présence d'un agent de condensation de :
préférence du groupe des mé taux alcalins et de leurs. dérivés (tels que par exem ple, hydrures, amidures, hydroxydes, alcoolates, mé taux alcoyles ou aryles) et plus particulièrement le sodium et le potassium métalliques, l'amidure de so dium, l'hydroxyde de sodium ou de potassium en poudre, l'hydrure de lithium ou de sodium, le tertio- butylate de sodium, le butyllithium,
le phényllithium ou le phénylsodium. On opère de préférence à la température d'ébullition du solvant. Il est particuliè rement avantageux d'utiliser l'ester réactif Y - A - Z sous forme de base libre en solution par exemple dans le benzène, le toluène ou le xylène et de l'ajou ter au mélange des autres réactifs dans lequel la dihydro-6,11 dibenzoï [b,e] thiazépine [1,4] utilisée peut déjà se présenter, du moins en partie, sous forme de sel alcalin.
La réaction peut également être réalisée avec un sel de l'ester Y - A - Z, mais dans ce cas, il faut évidemment mettre une proportion d'agent de condensation plus, forte de façon à neu traliser l'acide du sel utilisé. 2. - Action d'un dérivé de formule générale

sur un dérivé de formule générale H-Z (5) A, Y et Z étant définis comme précédemment.
Il est avantageux d'effectuer cette réaction dans un solvant tel qu'un hydrocarbure aromatique ou un alcool et d'utiliser comme agent de condensation un excès du composé H - Z.
Les nouveaux dérivés hétérocycliques de formule (1) peuvent être éventuellement purifiés par des mé thodes physiques (telles que distillation, cristallisa tion, chromatographie) ou chimiques (telles que for mation d'un sel, cristallisation, puis décomposition de ce sel en milieu alcalin). Dans ce dernier cas, la na ture de l'anion du sel est indifférente, la seule con dition étant que le sel soit bien défini et aisément cristallisable.
Les nouveaux dérivés hétérocycliques de formule (1) peuvent être transformés en sels d'addition avec les acides et en dérivés ammonium quaternaire.
Les sels d'addition peuvent être obtenus par ac tion des nouveaux dérivés sur des acides dans des solvants appropriés. Comme solvants organiques, on utilise par exemple des alcools, des éthers, des céto nes ou des solvants chlorés ; comme solvant minéral, on utilise avec avantage l'eau. Le sel formé précipite après concentration éventuelle de sa solution et est séparé par filtration ou décantation. Les dérivés am monium quaternaire peuvent être obtenus par action des nouveaux dérivés sur des esters, éventuellement dans un solvant organique, à la température ordinaire ou plus rapidement par léger chauffage.
Les nouveaux dérivés selon l'invention présentent des propriétés pharmacodynamiques intéressantes ce sont des antidépresseurs remarquables. De plus, certains se sont montrés particulièrement actifs comme spasmolytiques. Il s'agit principalement des composés dans lesquels la chaine - A - Z représente un groupe diéthylaminoéthyle, pipéridino-éthyle, ou diéthylamino-2 prropyle.
Les exemples suivants montrent comment l'in vention peut être mise en pratique <I>Exemple 1</I> A une solution bouillante de 10,66 g de dihydro- 6,11 dibenzo [b,e] thiazépine [1,4] dans 120 em3 de xylène anhydre, on ajoute en une fois 2,4 g d'amidure de sodium.
Après 30 minutes de chauf fage au reflux, on ajoute goutte à goutte, en 10 mi nutes, une solution de 7,3 g de diméthylamino-1 chloro-3 propane dans 30 cm3 de xylène anhydre. On poursuit ensuite le chauffage à reflux pendant 3 heures.
Après refroidissement, on ajoute avec précau tion 100 cm3 d'eau et sépare ensuite la couche xylé- nique par décantation. On l'épuise en utilisant au total 100 cm! d'acide chlorhydrique normal. On alca linise la phase aqueuse acide par addition de 12 cm3 de lessive de soude (d = 1,33) et extrait la base li bérée en utilisant au total 100 cm\; d'éther. On sèche la solution éthérée sur sulfate de sodium et la con centre à sec au bain-marie. L'huile résiduelle est sé chée sous vide de 0,5 mm de mercure environ.
On dissout cette huile pesant 15 g dans 300 cm@@ de cyclohexane et on filtre la solution à travers une colonne de 200 g d'alumine spéciale pour chromato graphie. On élue ensuite avec 250 cm-; de cyclo- hexane puis 800 cmg de mélange cyclohexane-ben- zène (1 : 1). On évapore les filtrats sous une pres sion de 20 mm de mercure environ.
On obtient ainsi 2,2 g de (diméthylamino-3 pro- pyl)-11 dihydro-6,11 dibenzo [b,e] thiazépine [1,4]. Le picrate, préparé dans l'éthanol puis recristallisé dans le méthanol, est une poudre cristalline orangée fondant à 126-127 C.
L'oxalate, préparé dans l'acétone et recristallisé dans l'éthan.ol, est une poudre cristalline blanche fondant à 110-111C.
La dihydro-6,11 dibenzo [b,e] thiazépine [1,4J utilisée comme produit de départ a été préparée par cyclisation de l'(amino-2 phényl) (bromo-2 benzyl) sulfure selon le brevet français N,- <B>1176115.</B> <I>Exemple 2</I> A une solution de 10,66 g de dihydro-6,11 di- benzo, [b,e] thiazépine [1,4] dans.
50 cm*# de diméthyl- formamide on ajoute goutte à goutte, en agitant et sous atmosphère d'azote, une suspension de 1,44 g d'hydrure de sodium dans 8,3 g d'huile Bayol 85 et 20 cm2 de toluène. L'addition terminée, on agite encore pendant 24 heures, puis ajoute rapidement une solution de 9,97g de diméthylamino-1 méthyl- sulfonyloxy-3 propane dans 80 cm', de toluène. On poursuit l'agitation pendant 24 heures.
On verse alors le mélange réactionnel dans 500 cm" d'eau et sépare la couche toluénique par décantation. On ex trait la couche aqueuse en utilisant au total 300 em:@ d'éther. Les solutions toluéniques et éthérées sont réunies et épuisées en utilisant au total 80 cm:, d'acide chlorhydrique normal. On alcalinise la phase aqueuse acide par addition de 9 cm" de lessive de soude (d = 1,33) et extrait la base libérée en utilisant au total 150 cm-" d'éther. On sèche la solution éthérée sur sulfate de sodium et la concentre à sec au bain marie.
L'huile résiduelle, pesant 12,9 g est dissoute dans 250 cm3 de cyclohexane et la solution obtenue est filtrée à travers une colonne de 250 g d'alumine spéciale pour chromatographie. On élue ensuite avec 1250 cm3 de mélange cyclohexane-benzène (1 : 1). On évapore le filtrat sous une pression de 20 mm de mercure environ.
On obtient ainsi 8,07 g de (diméthylamino-3 propyl)-11 dihydro-6,11 dibenzo [b,e] thiazépine [1,4] dont le picrate est une poudre cristalline oran gée fondant à 126-127 C.
<I>Exemple 3</I> A une solution de 5,33 g de dihydro-6,11 di- benzo [b,e] thiazépine [1,4] dans 25 cm- de dimé- thylformamide on ajoute goutte à goutte, en agitant et sous atmosphère d'azote, une suspension de 0,72 g d'hydrure de sodium dans 4,1 g d'huile Bayol 85 y> et 14 cm', de toluène. L'addition terminée, on pour suit l'agitation pendant encore 24 heures.
On ajoute alors rapidement une solution de 3,35 g de dimé- thylamino-1 chloro-3 propane dans 40 cm3 de to luène. On poursuit l'agitation pendant 24 heures.
La base, isolée comme à l'exemple 2, est purifiée par distillation sous un vide de 0.,2 mm de mercure. On obtient ainsi 4 g de (diméthylamino-3 propyl)-11 dihydro-6,11 dibenzo [b,e] thiazépine [1,4] bouil lant à 170-175 C sous 0,2 mm de mercure.
Le fumarate, préparé et recristallisé dans l'étha- nol, est une poudre cristalline blanche fondant à 150" C.
<I>Exemple 4</I> En opérant comme dans l'exemple 3, mais à partir de 10,66 g de dihydro-6,11 dibenzo [b,e] thia- zépine [1,4] et de 7,46 g de diéthylamino-1 chloro-2 éthane, on obtient après distillation sous vide, 14 g de (diéthylamino-2 éthyl)-11 dihydro-6,11 dibenzo [b,e] thiazépine [1,4] sous forme d'une huile bouil lant à 170 C sous 0,2 mm de mercure.
Le fumarate, préparé dans l'éthanol, est une pou dre cristalline blanche fondant à 162-163 C. <I>Exemple S</I> En opérant comme dans l'exemple 3, mais à partir de<B>10,66</B> g de dihydro-6,11 dibenzo [b,e] thia- zépine [1,4] et de 9,72 g de (néthyl-4 pipérazino)-1 chloro-3 propane, on obtient 9 g d'une base brute que l'on dissout dans 200 cm- de cyclohexane. On filtre la solution ainsi obtenue à travers une colonne de 200g d'alumine spéciale pour chromatographie.
On élue ensuite avec 700 cm- de benzène puis 100 cm' de mélange benzène-acétate d'éthyle (9 : 1), 100 cm-3 de mélange benzène-acétate d'éthyle (4 : 1), 100 cm-1 de mélange benzène-acétate d'éthyle (1 : 1) et enfin 500 cm-; d'acétate d'éthyle pur. On évapore les filtrats réunis sous une pression de 20 mm de mercure environ.
Le résidu est ensuite recristallisé deux fois dans l'hexane. On obtient ainsi 4 g de [(méthyl-4 pipérazino)-3 propyll-11 dihydro-6,11 dibenzo [b,e] thiazépine [1,4] sous forme d'une pou dre cristalline blanche fondant à 91o C.
<I>Exemple 6</I> En opérant comme dans l'exemple 3, mais à par tir de 21,30 g de dihydro-6,11 dibenzo [b,e] thiazé- pine [1,4] et de 16,24g de pipéridino-1 chloro-2 éthane, on obtient 17,5g d'une base brute que l'on dissout dans 350 cm3 de cyclohexane. On filtre la solution ainsi obtenue à travers une colonne de 400 g d'alumine spéciale pour chromatographie.
On élue ensuite successivement avec 250 cm3 de mélange cy- clohexane-benzène (9 : 1), 250 em3 de mélange cy- clohexane-benzène (4: 1), 1000 c<I>e</I> de mélange cy- clohexane-benzène (1 : 1) et enfin 750 em3 de ben zène pur. On évapore les filtrats réunis sous une pression de 20 mm de mercure.
On obtient ainsi 13,8 g de (pipéridino-2 éthyl)-11 dihydro-6,11 di- benzo [b,e] thiazépine [1,4] sous la forme d'une huile jaune.
Le fumarate, préparé puis recristallisé dans l'étha nol, est une poudre blanche fondant à 2200 C. <I>Exemple 7</I> En opérant comme dans l'exemple 3, mais à par tir de 21,3g de dihydro-6,11 dibenzo [b,e] thiazé- pine [1,4] et de 16,4g de diéthylamino-1 chloro-2 propane, on obtient, après distillation sous vide, 25,5g de (diéthylamino-2 propyl)-11 dihydro-6,
11 dibenzo [b,e] thiazépine [1,4] sous forme d'une huile orangée bouillant à 170-1900 C sous 0,3 mm de mercure.
L'oxalate, préparé dans l'acétone et recristallisé dans l'éthanol, est une poudre blanche fondant à 171-172 C.
<I>Exemple 8</I> En opérant comme dans l'exemple 3, mais à par tir de 10,66 g de dihydro-6,11 dibenzo [b,e] thiazé- pine [1,4] et de 7,35 g de pyrrolidino-1 chloro-2 éthane, on obtient 4,09 d'une base brute que l'on dissout dans 80 cm3 de benzène. On filtre la solu tion ainsi obtenue à travers une colonne de<B>80g</B> d'alumine spéciale pour chromatographie. On élue avec 200 cm3 de benzène et évapore le filtrat sous une pression de 20 mm de mercure environ.
On ob tient ainsi 2,5 g de (pyrrolidino-2 éthyl)-11 dihydro- 6,11 dibenzo [b,e] thiazépine [1,4] sous la forme d'une huile orangée.
Le fumarate, préparé et recristallisé dans l'étha nol, est une poudre cristalline blanche fondant à 204,, C.

The new heterocyclic derivatives of formula (1) can optionally be purified by physical (such as distillation, crystallization, chromatography) or chemical methods (such as formation of a salt, crystallization, then decomposition of this salt in an alkaline medium. ). In the latter case, the nature of the anion of the salt is irrelevant, the only condition being that the salt is well defined and easily crystallizable.
The new heterocyclic derivatives of formula (1) can be converted into addition salts with acids and into quaternary ammonium derivatives.
The addition salts can be obtained by ac tion of the new derivatives with acids in suitable solvents. As organic solvents, for example alcohols, ethers, ketones or chlorinated solvents are used; as inorganic solvent, water is advantageously used. The salt formed precipitates after possible concentration of its solution and is separated by filtration or decantation. The quaternary ammonium derivatives can be obtained by the action of the new derivatives on esters, optionally in an organic solvent, at room temperature or more rapidly by slight heating.
The new derivatives according to the invention exhibit interesting pharmacodynamic properties; they are remarkable antidepressants. In addition, some have been shown to be particularly active as spasmolytics. After 30 minutes of heating under reflux, a solution of 7.3 g of 1-dimethylamino-3-chloropropane in 30 cm3 of anhydrous xylene is added dropwise over 10 minutes. Heating is then continued under reflux for 3 hours.
After cooling, 100 cm 3 of water are carefully added and the xylene layer is then separated by decantation. It is exhausted by using a total of 100 cm! normal hydrochloric acid. The acidic aqueous phase is alkalized by adding 12 cm3 of sodium hydroxide solution (d = 1.33) and the liberated base is extracted using a total of 100 cm \; ether. The ethereal solution is dried over sodium sulphate and con centrated to dryness in a water bath. The residual oil is dried under vacuum of approximately 0.5 mm of mercury.
This oil weighing 15 g is dissolved in 300 cm 3 of cyclohexane and the solution is filtered through a 200 g column of special alumina for chromatography. Then eluted with 250 cm 3; of cyclohexane then 800 cmg of cyclohexane-benzene mixture (1: 1). The filtrates are evaporated under a pressure of about 20 mm of mercury.
2.2 g of (3-dimethylamino propyl) -11 dihydro-6,11 dibenzo [b, e] thiazepine [1,4] are thus obtained. The picrate, prepared in ethanol and then recrystallized in methanol, is an orange crystalline powder melting at 126-127 C.
The oxalate, prepared in acetone and recrystallized from ethanol, is a white crystalline powder melting at 110-111C.
The 6,11-dihydro-dibenzo [b, e] thiazepine [1,4J used as starting material was prepared by cyclization of (2-aminophenyl) (2-bromo-benzyl) sulfide according to the French patent N, - <B> 1176115. </B> <I> Example 2 </I> Has a solution of 10.66 g of 6.11-dihydro-di-benzo, [b, e] thiazepine [1,4] in.

<I> Example 3 </I> To a solution of 5.33 g of 6.11-dihydro-di-benzo [b, e] thiazepine [1,4] in 25 cm- of dimethylformamide is added dropwise , with stirring and under a nitrogen atmosphere, a suspension of 0.72 g of sodium hydride in 4.1 g of Bayol oil 85 y> and 14 cm ', of toluene. <I> Example 4 </I> By operating as in Example 3, but starting from 10.66 g of 6.11-dihydro-dibenzo [b, e] thiazepine [1,4] and 7, 46 g of 1-diethylamino-2-chloroethane, after distillation under vacuum, 14 g of (2-diethylamino-ethyl) -11-dihydro-6,11 dibenzo [b, e] thiazepine [1,4] are obtained in the form of an oil boiling at 170 C under 0.2 mm of mercury.

The residue is then recrystallized twice from hexane. 4 g of [(4-methyl piperazino) -3 propyll-11 dihydro-6,11 dibenzo [b, e] thiazepine [1,4] are thus obtained in the form of a white crystalline powder, melting at 91o C.
<I> Example 6 </I> By operating as in Example 3, but by shooting 21.30 g of 6.11-dihydro-dibenzo [b, e] thiazepine [1,4] and 16 , 24g of 1-piperidino-2-chloroethane, 17.5g of a crude base is obtained which is dissolved in 350 cm3 of cyclohexane. In this way 13.8 g of (2-piperidinoethyl) -11 dihydro-6.11 dibenzo [b, e] thiazepine [1,4] are obtained in the form of a yellow oil.
The fumarate, prepared and then recrystallized from ethanol, is a white powder melting at 2200 C. <I> Example 7 </I> By operating as in Example 3, but by shooting 21.3 g of dihydro- 6.11 dibenzo [b, e] thiazepine [1,4] and 16.4 g of 1-diethylamino-2-chloropropane, after vacuum distillation, 25.5 g of (2-diethylamino propyl) - are obtained. 11 dihydro-6,
11 dibenzo [b, e] thiazepine [1,4] in the form of an orange oil boiling at 170-1900 C under 0.3 mm of mercury.
The oxalate, prepared in acetone and recrystallized from ethanol, is a white powder melting at 171-172 C.
<I> Example 8 </I> By operating as in Example 3, but starting from 10.66 g of 6,11-dihydro-dibenzo [b, e] thiazepine [1,4] and 7 , 35 g of 1-pyrrolidino-2-chloroethane, 4.09 of a crude base is obtained which is dissolved in 80 cm3 of benzene. The solution thus obtained is filtered through a column of <B> 80g </B> alumina special for chromatography. Eluted with 200 cm3 of benzene and the filtrate evaporated under a pressure of about 20 mm of mercury.
2.5 g of (2-pyrrolidinoethyl) -11 dihydro-6,11 dibenzo [b, e] thiazepine [1,4] are thus obtained in the form of an orange oil.
The fumarate, prepared and recrystallized from ethanol, is a white crystalline powder, melting at 204, C.
