Gegenstand der Erfindung ist ein Verfahren zur Herstellung von 9-Aminoalkyl-9, 1 0-dihydro-9, 10- äthano-(1,2)-anthrazenen der Formel

worin A eine gegebenenfalls mono- oder disubstituierte Aminogruppe und Alk einen Alkylenrest bedeutet, und die gegebenenfalls in einer oder mehreren der Stellungen 1-8 und 10 substituiert sein können, oder ihren Salzen.
In den neuen Verbindungen. kann die Aminogruppe des Amino-alkylrestes unsubstituiert sein.
Vorzugsweise ist sie jedoch mono- oder disubstituiert.
Dabei kommen als Substituenten vor allem niedere Kohlenwasserstoffreste in Frage, die auch durch Heteroatome, wie Sauerstoff, Schwefel oder Stickstoff, unterbrochen und/oder durch freie Hydroxy-, Amino- oder Mercaptogruppen oder Halogenatome, wie Fluor, Chlor, Brom oder Jod, substituiert sein können.
Als Kohlenwasserstoffreste sind vor allem zu nennen: Alkyl- oder Alkenylreste, wie
Methyl-, Äthyl-, Propyl-, Isopropylreste, gerade oder verzweigte, in beliebiger Stelle verbundene
Butyl-, Pentyl-, Hexyl- oder Heptylreste,
Allyl- oder Methallyfreste, unsubstituierte oder alkyl-substituierte Cycloalkyloder Cycloalkenylreste, wie
Cyclopentyl-, Cyclohexyl-, Cycloheptyl-,
Cyclopentenyl-, Cyclohexenylreste, unsubstituierte oder alkyl-substituierte Cycloalkyl oder -alkenyl-alkyireste, wie
Cyclopentyl oder Cyclohexenylmethyl-, -äthyl-. oder -propylreste, Aralkyl- oder Aralkenyl-, wie
Phenylmethyl-, -äthyl-, vinyl- oder -propykeste, oder aromatische Kohlenwasserstoffreste, insbesondere Phenylreste, oder Alkylen- oder Alkenylenreste, wie z. B.
Butylen-(1,4), Pentylen-(1,5),
1 ,5-Dimthyl-pentylen-(1,5),
Hexylen-(1,6), Hexylen-(1,5).
Durch Heteroatome unterbrochene Reste dieser Art sind z. B. Alkoxyalkyl- oder Oxa-cycloalkylalkylreste, wie
Methoxyäthyl, Athoxyäthyl, Propoxyäthyl,
Butoxyäthyl, Methoxypropyl,
Methoxyäthoxyäthyl,
Tetrahydrofurylmethyl, Methylmercaptoäthyl, Oxa-, Aza- oder Thiaalkylen- oder -alkenylenreste, wie
3-Aza-, Oxa- oder Thiapentylen-(t,5),
3-Aza-hexylen-(1,6),
1,5-Dimethyl-3-aza-pentylen-(l 5), 3-Methyl-3-aza-pentylen-(1,5) oder 3-Hydroxyäthyl-3-aza-pentylen-(1 5).
Die Aminogruppe ist vor allem eine Di-niederalkylaminogruppe, wie die
Dimethylamino-, Diäthylamino-, Dipropylamine, N-Methyl-N-äthylaminogruppe, eine N - Niederalkyl - N - cycloalkylaminograppe, wie die
N-Methyl-N-cyclopentyl oder -cyclohexylamino- gruppe, eine Pyrrolidino-, Piperidino-, Morpholino- oder Thia-morpliolinogruppe, wie die Pyrrolidino-Piperidino-, Morpholino-,
Piperazino-, N-Methyl-, N-Äthyl- oder N-15-Hydroxyäthyl-piperazinogruppe.
Die genannten Verbindungen lassen sich quaternisieren.
Als vierter Substituent an einer quaternären Ammoniumgruppe sind vor allem niedere Alkyloder Alkenylreste, wie Methyl-, Äthyl-, Allyl- oder Propylreste, Benzylreste oder Phenoxy-niederalkylreste, wie der Phenoxyäthylrest, zu erwähnen.
Der die genannte Aminogruppe mit dem Anthrazenkern verbindende Alkylenrest ist vor allem ein nie derer, gerader oder verzweigter Alkylenrest, vorzugsweise mit 1-4 Kohlenstoffatomen, wie der
Methylen-, Äthylen (1, 2)-, PropyIen-(1,2)-,
Propylen-(1,3)-, Butylen-(1,2)-, Butylen-(1, 3)-, Butylen-(2'3)- oder Butylen-(1,4)-rest.
Die neuen Verbindungen können noch weiter substituiert sein. So können sie z. B. in den Stellungen 1-S des Anthrazenringes oder an sonstigen aromatischen Ringen niedere Alkyl-, Alkoxy-, Alkenyloxy- oder Alkylendioxy- oder Alkylmercaptogruppen, Halogenatome, wie Fluor, Chlor, Brom oder Jod, oder das Pseudohalogen Trifluormethyl, Alkylsulfonyl-, Alkanoyl-, Nitro- oder Aminogruppen tragen, wobei als Alkylreste z. B. Methyl, Äthyl, Propyl, Isopropyl, Butyl, i-Butyl, tert. Butyl, als Alkoxy-, Alkenyloxy- oder Alkylendioxygruppen die Methoxy-, Äthoxy-, Allyloxy- oder Methylendioxygruppe, als Alkylinercaptograppen die Methyl- oder Äthylmercaptogruppe, und als Alkanoylreste vor allem der Acetyl-, Propionyl- oder Butyrylrest zu nennen sind.
In 10-Stellung können die neuen Verbindungen vor allem einen aliphatischen Kohlenwasserstoffrest wie einen der oben genannten niederen Alkyl- oder Alkenylreste, oder auch ein Halogenatom aufweisen.
Die genannten Verbindungen besitzen wertvolle pharmakologische Eigenschaften. So zeigen sie eine zentralhemmende Wirkung, die durch einen Antagonismus gegenüber psychomotorischen Stoffen, wie z. B. Mezcalin, sowie durch eine Hemmung der spinalen Reflexübertragung gekennzeichnet ist, und können daher als beruhigende und tranquillisierende Medikamente in der Human- und Veterinärmedizin Verwendung finden. Sie eignen sich aber auch als Zusätze zu Tierfutter, da sie eine bessere Nahrungsverwertung bewirken. Weiter können die neuen Verbindungen als Ausgangs- oder Zwischenprodukte für die Herstellung anderer wertvoller Verbindungen dienen.
Besonders wertvoll sind die 9-Amino-alkyl-9,10- dihydro- 9,10-äthano- (1,2) - anthrazene und ihre Salze, in denen der Alkylenrest in 9-Stellung ein Methylen- oder Äthylen-(1,2)-rest ist, und vor allem diejenigen darunter, in denen die Aminogruppe tertiär, vorzugsweise eine Di-niederalkyl oder Cycloalkyl-amino-, Pyrrolidino-, Piperidino-, Morpholino-, Piperazino-, N-Methylpiperazino-, N-Äthylpiperazino- oder N-ss-Hydroxyäthyl-piperazinogruppe ist.
Besonders zu erwähnen sind die Verbindungen der Formeln

und ihre Salze, worin A eine tertiäre Aminogruppe, besonders eine der zuletzt erwähnten Aminogruppen ist, und speziell das
9-Diäthylamino-methyl-,
9-Dimethylamino-methyl- oder
9 -N-Cyciop entyl-N-methyl- amino-methyl-
9,1 0-dihydro-9, l0-äthano-(l ,2)-anthrazen und ihre Salze und das 9-ss-Diäthylamino-äthyl-,
9-p-Dimethylamino-äthyl- oder
9-p-N-Cyclopentyl-N-methylamino-äthyl-
9, lSdihydro-9, 1Gäthano-(1, 2)-anthrazen und ihre Salze.
Das erfindungsgemässe Verfahren besteht darin, dass man 9,1 0-D.ihydro-9, 10-äthano-(1, ,2)-anthrazene, die in 9Stellung eine reaktionsfähig abgewandelte Hydroxyalkylgruppe aufweisen, mit Ammoniak oder mono oder disubstituierten Aminen umsetzt.
Als reaktionsfähig abgewandelte Hydroxylalkylgrup pen sind dabei z. B. reaktionsfähig veresterte Hydroxylalkylreste zu nennen, wobei als Ester vor allem diejenigen der Halogenwasserstoffsäuren, wie Chlor-, Brom- oder Jodwasserstoffsäuren, oder von Arylsulfonsäuren, z. B. p-Toluolsulfonsäuren, zu erwähnen sind.
In erhaltene primäre, sekundäre oder tertiäre Aminogruppen lassen sich in üblicher Weise noch weitere Stickstoffsubstituenten einführen, z. B. durch Behandlung mit reaktionsfähigen Estern, z. B. den obengenannten, der entsprechenden Alkohole oder nach der Methode der reduktiven Alkylierung unter Verwendung der entsprechenden Carbonylverbindungen oder durch Acylierung mit Carbonsäuren und Reduktion der erhaltenen N-Acylverbindungen.
Die genannten Reaktionen können in üblicher Weise durchgeführt werden.
Die Ausgangsstoffe sind bekannt oder können nach an sich bekannten Verfahren gewonnen werden.
Die neuen Verbindungen werden je nach den Reaktionsbedingungen und Ausgangsstoffen in freier Form oder in Form ihrer Salze erhalten. Die Salze der neuen Verbindungen können in an sich bekannter Weise in die freien Verbindungen übergeführt werden, z. B. Säureadditionssalze, durch Reaktion mit einem basischen Mittel. Anderseits können gegebenenfalls erhaltene freie Basen mit anorganischen oder organischen Säuren Salze bilden. Zur Herstellung von Säureadditionssalzen werden insbesondere therapeutisch verwendbare Säuren verwendet, z. B.
Halogenwasserstoffsäuren, beispielsweise
Salzsäure oder Bromwasserstoffsäure,
Perchlorsäure, Salpetersäure oder Thiocyansäure, Schwefel- oder Phosphorsäuren, oder organische Säuren, wie
Ameisensäure, Essigsäure, Propionsäure,
Glykolsäure, Milchsäure, Brenztraubensäure,
Oxalsäure, Malonsäure, Bernsteinsäure,
Maleinsäure, Fumarsäure, Äpfelsäure,
Weinsäure, Zitronensäure, Ascorbinsäure,
Hydroxymaleinsäure, Dihydroxymaleinsäure,
Benzoesäure, Phenylessigsäure,
4-Amino-benzoesäure, 4-Hydroxy-benzoesäure,
Anthranilsäure, Zimtsäure, Mandelsäure,
Salicylsäure, SAmino-salicylsäure,
2-Phenoxy-benzoesäure,
2-Acetoxy-benzoesäure, Methansulfonsäure, Äthansulfonsäure, Hydroxyäthansulfonsäure,
Benzolsulfonsäure, pToluol-sulfonsäure,
Naphthalinsulfonsäure oder Sulfanilsäure, oder
Methionin, Tryptophan, Lysin oder Arginin.
Dabei können Mono oder Polysalze vorliegen.
Quaternäre Ammoniumsalze können auch z. B. durch Einwirkung von frisch gefälltem Silberoxyd auf die Ammoniurnhalogenide, oder Einwirkung von Barytlauge auf die Ammoniumsulfate, oder unter Verwendung von basischen Ionenaustauschern in die Ammoniumhydroxyde übergeführt werden, aus denen durch Umsatz mit Säuren, z. B. den oben angeführten, andere Ammoniumsalze erhalten werden können. Dieser Austausch kann auch direkt, unter Verwendung geeigneter Ionenaustauscher stattfinden.
Die neuen Verbindungen können als Heilmittel in Form von pharmazeutischen Präparaten verwendet werden, welche diese Verbindungen zusammen mit pharmazeutischen, organischen oder anorganischen, festen oder flüssigen Trägerstoffen, die für enterale, z. B. orale, oder parenterale Gabe geeignet sind, enthalten. Für die Bildung derselben kommen solche Stoffe in Frage, die mit den neuen Verbindungen nicht reagieren, wie z. B. Wasser, Gelatine, Milchzucker, Stärke, Magnesiumstearat, Talk, pflanzliche Öle, Benzylalkohole, Gummi, Polyalkylenglykole, Cholesterin oder andere bekannte Arzneimittelträger. Die pharmazeutischen Präparate können z. B. als Tabletten, Dragees, Kapseln oder in flüssiger Form als Lösungen, Suspensionen oder Emulsionen vorliegen.
Gegebenenfalls sind sie sterilisiert und bzw. oder enthalten Hilfsstoffe, wie Konservierungs-, Stabilisierungs-, Netz- oder Emulgiermittel, Salze zur Veränderung des osmotischen Druckes oder Puffer. Sie können auch noch andere therapeutisch wertvolle Stoffe enthalten.
Die neuen Verbindungen können auch in der Tiermedizin, z. B. in einer der obengenannten Formen, oder bei der Aufzucht und Ernährung von Tieren in Form von Futtermitteln oder von Zusatzmitteln für Tierfutter verwendet werden. Dabei werden z. B. die üblichen Streck- und Verdünnungsmittel bzw. Futtermittel angewendet.
Die Temperaturen sind im folgenden Beispiel in Celsiusgraden angegeben.
Beispiel
Zu einer Lösung von 5 g Dimethylamin in 50 cm3 Alkohol gibt man 10 g
9-y-Chlor-propyl-9, 10-dihydro-9, 10-äthano- (1,2)-anthrazen und erwärmt 4 Stunden im Autoklaven auf 1000.
Das Reaktionsgemisch wird anschliessend zur Trockene eingedampft. Den Rückstand versetzt man mit 50 cm3 2n Salzsäure und extrahiert mit Äther.
Der saure Auszug wird hierauf durch Zugabe von Natronlauge alkalisch gestellt und mit Äther extrahiert. Nach dem Trocknen und Eindampfen des Ethers verbleibt das 91-Dimethylaminopropyl-9, 1 O-dihydro- 9,1 0-äthano-( 1 ,2)-anthrazen der Formel

das nach Sublimation bei 88-900 schmilzt.
Das als Ausgangsmaterial verwendete
9-y-Chlor-propyl-9, 10-dihydro-9, 10-äthano- (1 ,2)-anthrazen kann wie folgt hergestellt werden:
Zu einer Suspension von 4 g Lithiumaluminium- hydrid in 150 cm3 Tetrahydrofuran tropft man eine Lösung von 15 g fi-[9,1 0-Dihydro-9, 1 äNano-(1, ,2)-9-anthryl]- propionsäure und rührt 2 Stunden bei 600. Anschliessend wird abgekühlt und vorsichtig 10 cm3 Wasser zugetropft.
Den ausgeschiedenen Niederschlag filtriert man ab und dampft das Filtrat im Vakuum ein. Es verbleibt ein kristalliner Rückstand, den man mit 200 cm3 Thionylchlorid versetzt und 4 Stunden auf 400 erwärmt. Anschliessend wird das überschüssige Thionylchlorid im Vakuum abdestilliert. Es bleibt ein kristalliner Rückstand von
9-y-Chlorpropyl-9, 10-dihydro-9, 10-äthano- (1,2)-anthrazen, das nach Sublimation bei 120-1250 schmilzt.
In ähnlicher Weise, ausgehend von entsprechenden reaktionsfähig veresterten 9-Hydroxy-alkyl-ver- bindungen erhält man: 9-Diäthylamino-methyl-9, 10-dihydro-9,10 äthano-( 1 ,2)-anthrazen vom F. 112-1150 (aus Äthanol),

The 9-amino-alkyl-9,10-dihydro-9,10-ethano- (1,2) -anthracenes and their salts, in which the alkylene radical in the 9-position is a methylene or ethylene (1, 2) radical, and especially those among them in which the amino group is tertiary, preferably a di-lower alkyl or cycloalkyl-amino, pyrrolidino, piperidino, morpholino, piperazino, N-methylpiperazino, N-ethylpiperazino or N-ß-hydroxyethyl piperazino group.

The process according to the invention consists in reacting 9,1 0-D.ihydro-9,10-ethano- (1,2) -anthracenes, which have a reactive modified hydroxyalkyl group in the 9-position, with ammonia or mono- or disubstituted amines.
As reactive modified Hydroxylalkylgrup pen are z. B. reactive esterified hydroxylalkyl radicals, the esters especially those of the hydrohalic acids, such as chlorine, bromine or hydroiodic acids, or of arylsulfonic acids, eg. B. p-toluenesulfonic acids are to be mentioned.
In the primary, secondary or tertiary amino groups obtained, further nitrogen substituents can be introduced in the usual way, eg. B. by treatment with reactive esters, e.g. B. the above, the corresponding alcohols or by the reductive alkylation method using the corresponding carbonyl compounds or by acylation with carboxylic acids and reduction of the N-acyl compounds obtained.
The reactions mentioned can be carried out in a customary manner.
The starting materials are known or can be obtained by processes known per se.
The new compounds are obtained in free form or in the form of their salts, depending on the reaction conditions and starting materials. The salts of the new compounds can be converted into the free compounds in a manner known per se, eg. B. acid addition salts, by reaction with a basic agent. On the other hand, any free bases obtained can form salts with inorganic or organic acids. For the preparation of acid addition salts, therapeutically useful acids are used in particular, e.g. B.

Mono or poly salts can be present.
Quaternary ammonium salts can also be e.g. B. by the action of freshly precipitated silver oxide on the ammonium halides, or the action of barite liquor on the ammonium sulfates, or using basic ion exchangers in the ammonium hydroxides, from which by reaction with acids, eg. B. the above, other ammonium salts can be obtained. This exchange can also take place directly, using suitable ion exchangers.
The new compounds can be used as medicaments in the form of pharmaceutical preparations, which these compounds together with pharmaceutical, organic or inorganic, solid or liquid carriers which are suitable for enteral, e.g. B. oral or parenteral administration are suitable. For the formation of the same substances come into question that do not react with the new compounds, such as. B. water, gelatin, milk sugar, starch, magnesium stearate, talc, vegetable oils, benzyl alcohols, gum, polyalkylene glycols, cholesterol or other known excipients. The pharmaceutical preparations can e.g. B. as tablets, dragees, capsules or in liquid form as solutions, suspensions or emulsions.
If necessary, they are sterilized and / or contain auxiliaries such as preservatives, stabilizers, wetting agents or emulsifiers, salts to change the osmotic pressure or buffers. They can also contain other therapeutically valuable substances.
The new compounds can also be used in veterinary medicine, e.g. B. in one of the forms mentioned above, or in the rearing and feeding of animals in the form of feed or additives for animal feed. Here z. B. the usual extenders and thinners or feed used.
In the following example, the temperatures are given in degrees Celsius.

10 g are added to a solution of 5 g of dimethylamine in 50 cm3 of alcohol
9-y-chloro-propyl-9, 10-dihydro-9, 10-ethano- (1,2) -anthracene and heated to 1000 for 4 hours in an autoclave.
The reaction mixture is then evaporated to dryness. The residue is mixed with 50 cm3 of 2N hydrochloric acid and extracted with ether.
The acidic extract is then made alkaline by adding sodium hydroxide solution and extracted with ether. After drying and evaporation of the ether, the 91-dimethylaminopropyl-9, 10-dihydro-9,1 0-ethano- (1, 2) -anthracene of the formula remains

which melts at 88-900 after sublimation.
That used as the starting material
9-y-chloro-propyl-9, 10-dihydro-9, 10-ethano- (1, 2) -anthracene can be prepared as follows:
A solution of 15 g of fi [9,1 0-dihydro-9,1-nano- (1,2) -9-anthryl] propionic acid is added dropwise to a suspension of 4 g of lithium aluminum hydride in 150 cm3 of tetrahydrofuran and the mixture is stirred 2 hours at 600. It is then cooled and 10 cm3 of water are carefully added dropwise.
The deposited precipitate is filtered off and the filtrate is evaporated in vacuo. A crystalline residue remains, to which 200 cm3 of thionyl chloride are added and the mixture is heated to 400 for 4 hours. The excess thionyl chloride is then distilled off in vacuo. There remains a crystalline residue of
9-y-chloropropyl-9, 10-dihydro-9, 10-ethano- (1,2) -anthracene, which melts at 120-1250 after sublimation.
In a similar way, starting from the corresponding reactive esterified 9-hydroxy-alkyl compounds, one obtains: 9-diethylamino-methyl-9, 10-dihydro-9,10 ethano- (1, 2) -anthracene from F. 112- 1150 (from ethanol),
