Als Substituenten der sekundären oder tertiären Aminogruppe kommen vor allem niedere Kohlenwasser stoffreste in Frage, die auch durch Heteroatome, wie Sauerstoff, Schwefel oder Stickstoff, unterbrochen und/ oder durch freie Hydroxy-, Amino- oder Mercapto- gruppen oder Halogenatome, wie Fluor, Chlor, Brom oder Jod, substituiert sein können.
Als niedere Kohlen wasserstoffreste sind vor allem zu nennen: niedere Alkyl- oder Alkenylreste, wie Methyl-, Äthyl-, Propyl-, Isopropylreste, gerade oder verzweigte, in beliebiger Stelle verbundene Butyl-, Pentyl-, Hexyl- oder Heptylreste, Allyl- oder Methallylreste,
unsubstituierte oder alkyl-substituierte Cycloalkyl- oder Cycloalkenylreste, wie Cyclopentyl-, Cyclohexyl-, Cycloheptyl-, Cyclopentenyl-, Cyclohexenylreste, unsubstituierte oder alkyl-substituierte Cycloalkyl- oder -alkenyl-alkylreste, wie Cyclopentyl- oder Cyclohexenylmethyl-,
-äthyl- oder -propylreste, Aralkyl- oder Aralkenyl-, wie Phenylmethyl-, äthyl-, -vinyl- oder propylreste, oder Aryl-, insbesondere Phenylreste, oder Alkylen- oder Alkenylenreste, wie z. B.
B,utylen-(1,4), Pentylen-(1,5), 1, 5-Dimethylenpentylen-(1, 5), Hexylen-(1,6), Hexylen-(1,5).
Durch Heteroatome unterbrochene Reste dieser Art sind z. B.
Alkoxyalkyl- oder Oxa-cycloalkylalkylreste, wie Methoxyäthyl, Äthoxyäthyl, Propoxyäthyl, Butoxyäthyl, Methoxypropyl, Methoxyäthoxyäthyl, Tetrahydrofurylmethyl, Methylmercaptoäthyl, Oxa-, Aza- oder Thiaalkylen- oder -alkenylenreste, wie 3-Aza-, Oxa- oder Thiapentylen-(1,5),
3-Aza-hexylen-(1,6), 1,5-Dimethyl-3-aza-pentylen-(1,5), 3-Methyl-3-aza-pentylen-(1,5) oder 3-Hydroxyäthyl-3-aza-pentylen-(1,5).
Die Aminogruppe ist vor allem eine Mono- oder Di-niederalkylaminogruppe, wie die Methylamino-, Äthylamino-, n-Butylamino-, Dimethylamino-, Diäthylamino-, Dipropylamino-, N-Methyl-N-äthylaminogruppe, eine N-Niederalkyl-N-cycloalkylaminogr-uppe, wie die N-Methyl-N-cyclopentyl- oder -cyclohexylgruppe, eine Pyrrolidino-, Piperidino-,
Morpholino- oder Thio-morpholinogruppe, wie die Pyrrolidino-, Piperidino-, Morpholino-, Piperazino-, N-Methyl-, N-Athyl- oder N-f-Hydroxyäthyl-piperazinogruppe. Die neuen Verbindungen besitzen wertvolle pharma kologische Eigenschaften. So zeigen sie eine zentralhem mende Wirkung, die durch einen Antagonismus gegen über psychomotorischen Stoffen, wie z.
B. Mescalin, sowie durch eine Hemmung der spinalen Reflexüber tragung gekennzeichnet ist, sowie eine histaminolytische Wirkung und können daher als psychotrope, beruhigende Medikamente in der Human- und Veterinärmedizin Ver wendung finden. Sie eignen sich aber auch als Zusätze zu Tierfutter, da sie eine bessere Nahrungsverwertung bewirken. Weiter können die neuen Verbindungen als Ausgangs- oder Zwischenprodukte für die Herstellung anderer wertvoller Verbindungen dienen.
Besonders wertvoll sind die Verbindungen der For mel I, worin die Aminogruppe eine Mono- oder Di-niederalkyl- oder -cycloalkyl-amino-, Pyrrolidino-, Piperidino-, Morpholino-, Piperazino-, N-Methylpiperazino-, N-Äthylpiperazino- oder N-f-Hydroxyäthyl-piperazinognuppe darstellt, und vor allem die Methylamino-, Äthylamino-, n-Butylamino-,
Dimethylamino- oder Diäthylaminognuppe bedeutet. Ganz besonders wertvoll sind die folgenden Ver bindungen: 9-y-Dimethylamino-propyl-10-chlor-9,10-dihydro- 9,10-äthano-(1,2)-anthracen, 2-Chlor-9-y-dimethylaminopropyl-9,10-dihydro- 9,10-äthano-(1,2)-anthracen, 3-Chlor-9-y-dimethylamino-propyl-9,10-dihydro- 9,10-äthano-(1,2)-anthracen rund 3-Chlor-9-(y-monomethyl-aminopropyl)-9,10- dihydro-9,10-äthano-(1,2)-anthracen, und ihre Salze.
Das erfindungsgemässe Verfahren zur Herstellung der neuen Verbindungen ist dadurch gekennzeichnet, dass man Verbindungen der Formel

die in mindestens einer der Stellungen 1-8 und 10 ein Halogenatom tragen, und worin X eine reaktionsfähig veresterte Hydroxylgruppe bedeutet, mit einem primären oder sekundären Amin umsetzt.
Eine reaktionsfähig ver- esterte Hydroxylgruppe ist vor allem eine mit einer Halogenwasserstoffsäure, wie Chlor-, Brom- oder Jod- wasserstoffsäure, oder einer Arylsulfonsäure, z. B. der p-Toluolsulfonsäure, veresterte Hydroxylgruppe.
In erhaltene sekundäre Aminogruppen können in üblicher Weise noch weitere Stickstoffsubstibuenten ein- geführt werden, z. B. durch Behandlung mit reaktions fähigen Estern, z.
B. den oben genannten, der entspre chenden Alkohole oder nach der Methode der reduk- tiven Alkylierung unter Verwendung der entsprechen den Carbonylverbindungen oder durch Acylierung mit Carbonsäuren und Reduktion der erhaltenen N-Acyl- verbindungen.
Ein Ausgangsstoff kann auch in Form eines Salzes verwendet werden.
Die genannten Reaktionen können in üblicher Weise in An- oder Abwesenheit von Verdünnungs-, Konden- sations- und/oder katalytischen Mitteln, bei erniedrigter, gewöhnlicher oder erhöhter Temperatur, gegebenenfalls im geschlossenen Gefäss durchgeführt werden.
Die Ausgangsstoffe sind bekannt oder können nach an sich bekannten Verfahren gewonnen werden.
Die neuen Verbindungen können je nach den Reak tionsbedingungen :und -Ausgangsstoffen in freier Form oder in Form ihrer Salze erhalten werden. Die Salze der neuen Verbindungen können in an sich bekannter Weise in die freien Verbindungen übergeführt werden, z. B. Säureadditionssalze durch Reaktion mit einem basischen Mittel. Anderseits können gegebenenfalls erhaltene freie Basen mit anorganischen oder organischen Säuren Salze bilden. Zur Herstellung von Säureadditionssalzen wer den insbesondere therapeutisch verwendbare Säuren ver wendet, z.
B. Halogenwasserstoffsäuren, beispielsweise Salzsäure oder Bromwasserstoffsäure, Perchlorsäure, Salpetersäure oder Thiocyansäure, Schwefel- oder Phosphorsäuren, oder organische Säuren, wie Ameisensäure, Essigsäure, Propionsäure, Glykolsäure, Milchsäure, Brenztraubensäure, Oxalsäure,- Malonsäure, Bernsteinsäure, Maleinsäure, Fumarsäure,
Äpfelsäure, Weinsäure, Zitronensäure, Ascorbinsäure, Hydroxymaleinsäure, Dihydroxymaleinsäure, Benzoesäure, Phenylessigsäure, 4-Amino benzoesäure, 4-Hydroxy-benzoesäure, Anthranilsäure, Zimtsäure, Mandelsäure, Salicylsäure, 4-Amino-salicylsäure, 2-Phenoxybenzoesäure, 2-Acetoxy-benzoesäure, Methansulfonsäure, Äthansulfonsäure,
Hydroxyäthansulfonsäure, Benzolsulfonsäure, p-Toluol-sulfonsäure, Naphthalinsulfonsäure oder Sulfanilsäure, oder Methionin, Tryptophan, Lysin oder Arginin. Dabei können Mono- oder Polysalze vorliegen.
Die neuen Verbindungen können als Heilmittel in Form von pharmazeutischen Präparaten verwendet wer den, welche diese Verbindungen zusammen mit pharma zeutischen, organischen oder anorganischen, festen oder flüssigen Trägerstoffen, die für enterale, z. B. orale, oder parenterale Gabe geeignet sind, enthalten.
Die neuen Verbindungen können auch in der Tier medizin, z. B. in einer der oben genannten Formen, oder bei der Aufzucht und Ernährung von Tieren in Form von Futtermitteln oder von Zusatzmitteln für Tierfutter verwendet werden. Dabei werden z. B. die üblichen Streck- und Verdünnungsmittel bzw. Futtermit tel angewendet.
Im folgenden Beispiel sind die Temperaturen in Celsiusgraden angegeben.
<I>Beispiel</I> 10,0 g 9-y-Chlorpropyl-10-chlor-9,10-dihydro-9,10- äthano-(1,2)-anthracen werden mit 5 g Dimethylamin in 50 ml Alkohol in Autoklaven während 10 Stunden auf 100 erhitzt. Hierauf dampft man in Vakuum ein, löst den Rückstand in Äther und extrahiert mit 2n Salz säure. Die saure Lösung wird durch Zugabe von lOn Natronlauge alkalisch gestellt, wonach die ausgeschie dene Base mit Äther extrahiert wird.
Nach dem Trock nen und Eindampfen des Äthers verbleibt das 9-y Di- methylaminopropyl-10-chlor-9,10-dihydro -9,10-äthano- (1,2)-anthracen der Formel

das als Hydrochlorid bei 225-226 schmilzt.
Das als Ausgangsmaterial verwendete 9-y-Chlorpro- pyl- 10-chlor-9,10-dihydro-9,10-äthano-(1,2)-anthracen wird wie folgt erhalten: Zu einer Suspension von 5 g Lithiuma1uminrumhy- drid in 200 ml absolutem Tetrahydrofuran tropft man eine Lösung von ss-[10-Chlor-9,10-dihydro-9,10-äthano- (1,2) - 9 - anthryl] - propionsäure in 150 ml absolutem Tetrahydrofuran und rührt 4 Stunden bei 60 .
Dann wird auf Zimmertemperatur abgekühlt und man gibt vorsichtig 20 ml Wasser zu. Der ausgeschiedene Nieder schlag wird filtriert. Nach dem Eindampfen des Filtrates verbleibt das rohe 9-y-Hydroxypropyl - 9,10 - dihydro- 9,10-äthano-(1,2)-anthracen, das mit 100 ml Thionyl- chlorid während 2 Stunden unter Rückfluss gekocht wird.
Hierauf dampft man im Vakuum zur Trockne ein. Es verbleibt das 9-y-Chlorpropyl -10 - chlor-9,10- dihydro-9,10-äthano-(1,2)-anthracen, das nach Sublima tion bei 105-110 schmilzt.
Auf analoge Weise kann man die folgenden Ver bindungen herstellen: a) 2-Chlor-9-y-dimethylaminopropyl-9,10-dihydra-9,10- äthano-(1,2)-anthracen-hydrochlorid, Smp. 171 bis 173 (nach Umkristallisation aus Isopropanol), b) 3-Chlor - 9 - y - dimethylaminopropyl-9,10-dihydro- 9,10=äthano,(1,2)-anthracen maleat, Smp. 154-156 (nach Umkristallisation aus Äthanol-Äther), c) 3 - Chlor - 9 - (y-monomethyl-aminopropyl)-9,10-di- hydro-9,
