Die Erfindung betrifft ein neues Verfahren zur Herstellung von Diphenylalkylaminen, die sich durch günstige physiologische Wirkungen, insbesondere auf das Herz und den Kreislauf auszeichnen.
Es wurde ein Verfahren zur Herstellung von Diphenylalkylaminen der Formel I gefunden,

Für das Verfahren gemäss der vorliegenden Erfindung können die folgenden Phenyl-alkyl-Verbindungen eingesetzt werden : 1-Phenyl-l-hydroxy-3- (1-phenyl-propyl-(2)-amino- propan, 1-p-Methoxy-phenyl-1-hydroxy-3- (l-phenyl-propyl- (2-amino)-propan, lm-Chlorphenyl-1-hydroxy-3-(1-phenyl-propyl- (2)-amino)-propan, 1-o-Tolyl-l-hydroxy-3- (1-phenyl-propyl-(2)-amino)- propan, sowie
1-Phenyl-1-hydroxy-3-(1-m-methoxy-phenyl-pro- pyl- (2)-amino)-propan,
1-Phenyl-1-hydroxy-3- (1-phenyl-butyl- (2)-amino)- propan, 1-Phenyl-1-hydroxy-3- (2-phenyl-äthylamino)-pro- pan, 1-Phenyl-1-hydroxy-3- (1-phenyl-propyl-(2)-methyl- amino)-propan,
und weiterhin
1-Phenyl-1-hydroxy-propylamino- (3),

Als Reaktionspartner der vorstehend genannten Verbindungen seien genannt :
Benzol, Toluol, Chlorbenzol, Methoxybenzol, Athoxybenzol und Isopropylbenzol.
Die vorliegende Arylierungsreaktion wird z. B. in n einem geeigneten Lösungsmittel durchgeführt. Als solche kommen neben den im vorstehenden Absatz genannten Reaktionspartnem, die in diesem Falle im tSberschuss zuzusetzen sind, Nitrobenzol oder Chlorkohlenwasserstoffe, wie Tetrachlorkohlenstoff oder Tetrachloräthan, in Betracht.
Die Reaktion wird bei Temperaturen zwischen 50 und 200 C, vorzugsweise zwischen 60 und 140 C durchgeführt. Insbesondere arbeitet man bei der Siedetemperatur des verwendeten Lösungsmittels.
Zur Durchführung des Verfahrens gemäss der Erfindung wird das betreffende 1-Phenyl-l-hydroxy-alkylamin zunächst mit einer Lewis-Säure wie Aluminiumchlorid in einem geeigneten Lösungsmittel, z. B. Benzol, mit Hilfe eines geeigneten Halogenierungsmittels, vorzugsweise einem Säurechlorid des Schwefels wie Thionylchlorid, in das entsprechende Chlorid überführt ; die Hydrochloride der so erhaltenen 1-Phenyl1-chlor-Verbindungen sind im allgemeinen gut kristalli sierbar. Die so erhaltenen 1-Phenyl-l-chlor-alkylamine werden in einem der genannten Lösungsmittel mit dem gewünschten Reaktionspartner und dem Friedel-Crafts Katalysator bei erhöhter Temperatur zu Diphenyl-alkyl-aminen kondensiert. Diese beiden Verfahrensschritte können als Eintopfverfahren ausgeführt werden.
Nach einer weiteren vorteilhaften Ausführungsart des Verfahrens können die 1-Phenyl-l-hydroxy-alkyl- amin-Verbindungen in Gegenwart von Friedel-Crafts- Katalysatoren in den genannten Lösungsmitteln direkt mit den genannten Reaktionspartnern umgesetzt werden. Die Reaktion kann in gleicher Weise ausgehend von den genannten l-Phenyl-1, 2-ungestättigten Alkyl amin-Verbindungen durchgeführt werden.
Die Verfahrenserzeugnisse können als basische Verbindungen mit Hilfe von physiologisch verträglichen anorganischen oder organischen Säuren in die entsprechenden Salze übergeführt werden. Als anorganische Säuren kommen beispielsweise in Betracht : Halogenwasserstoffsäuren wie Chlorwasserstoffsäure, Schwefelsäure, Phosphorsäure und Amidosulfonsäure.
Als organische Säuren seien beispielsweise genannt Essigsäure, Propionsäure, Milchsäure, Glykolsäure, Gluconsäure, Maleinsäure, Bernsteinsäure, Weinsäure, Salicylsäure und Zitronensäure.
Die Verfahrenserzeugnisse können als solche oder in Form ihrer Salze, gegebenenfalls auch in Mischung mit pharmazeutisch üblichen Trägerstoffen, parental oder oral appliziert werden. Im Falle der oralen Applikation kommen als Darreichungsformen vorzugsweise Tabletten oder Dragees in Frage, zu denen die Verfahrenserzeugnisse als Wirkstoffe mit den üblichen Trägerstoffen, wie Milchzucker, Stärke, Tragant und Magnesiumstearat verarbeitet werden.
Die bisher bekannten Verfahren zur Herstellung der hier genannten Stoffe waren schwierig und zeitraubend. Ein Teil der für sie notwendigen Ausgangsmaterialien war schwer zugänglich. Hier eröffnet das vorliegende neue Verfahren einen Zugang zu der ge nannten Stoffgruppe, der in einem für industrielle Verwertung erforderlichen Masstab durchgeführt werden kann. Die nach diesem Verfahren erzielten Ausbeuten liegen zwischen 80 ouzo und 95"/o.
Beispiel 1
15,1 g 1-Phenyl-1-hydroxy-propylamin- (3) werden mit 14,5 g Phenylaceton in 50 ml Benzol 30 Minuten lang gekocht und das entstandene Wasser mit Benzol abdestilliert. Der ölige Rückstand wird in 30 ml Methanol und 5 ml Wasser aufgenommen. In diese Lösung werden portionsweise 1,5 g Natriumborhydrid eingetragen. Dabei erwärmt sich das Reaktionsgemisch auf 40-50 C. Man erhitzt anschliessend noch 30 Minuten auf dem Wasserbade und destilliert die Lösungsmittel ab. Der ölige Rückstand wird mit Äther extrahiert und der Ätherextrakt mit alkoholischer Chlorwasserstoffsäure bis zum Auftreten einer Trübung versetzt.
Es kristallisieren 25 g 1-Phenyl-1-hydroxy-3-(1-phenyl- propyl- (2)-amino)-propan-hydrochlorid vom Schmelzpunkt 144-146 C.
Z5 g der so erhaltenen Verbindung werden portionsweise in eine Lösung von 40 ml Thionylchlorid in 80 ml Benzol bei Zimmertemperatur eingetragen. Es setzt eine starke Chlorwasserstoff-und Schwefeldioxydentwicklung ein. Nach einiger Zeit kristallisiert das Hydrochlorid des 1-Phenyl-1-chlor-3-(1-phenyl-propyl- (2)-amino)-propan. Die Abscheidung wird durch Zugabe von Sither vervollständigt. Man erhält 24 g der Verbindung vom Schmelzpunkt 138-144 C. 10 g dieses Hydrochlorids werden in 40 ml Benzol suspendiert und 8 g wasserfreies Aluminiumchlorid portionsweise eingetragen, wobei die Temperatur bei etwa 50 C liegen soll. Anschliessend wird 30 Minuten lang unter Rückfluss erhitzt. Nach Erkalten wird das Reaktionsgemisch in eine Mischung von 20 ml konz.
Salzsäure, 10 ml Wasser und 100 g Eis gegossen. Nach mehrstündigem Stehen und Zugabe von Ather scheidet sich das Hydrochlorid des 1, 1-Diphenyl-3- (1-phenyl-pro pyl- (2)-amino)-propan in fast farblosen Kristallen aus.
Beispiel 2
Die Rohausbeute beträgt 11,2 g vom Schmelzpunkt, 186-188 C, (aus wässrigem Methanol 190-192 C).
In einem Gemisch von 16 ml Thionylchlorid und 30 ml Benzol werden 15 g 1-Phenyl-l-hydroxy-prop- ylamin- (3) portionsweise eingetragen. Anschliessend wird noch 20 Minuten lang auf dem Wasserbade unter Rückfluss erhitzt. Nach mehrstündigem Stehen wird die Kristallisation durch Zugabe von Ather vervoll ständigt. Man erhält 18,5 g 1-Phenyl-1-chlor-propyl- amin- (3)-hydrochlorid vom Schmelzpunkt 110-112 C.
10 g des so erhaltenen Hydrochlorids werden in 40 ml Benzol suspendiert und 12 g wasserfreies Aluminiumchlorid portionsweise eingetragen. Nach 30 Minuten Erwärmen auf dem Wasserbade wird das Reaktionsge- misch in ein Gemisch aus Salzsäure, Wasser und Eis gegossen. Es kristallisiert das Hydrochlorid des 1, 1-Diphenyl-propylamin- (3) vom Schmelzpunkt 206-209 C. Die Ausbeute beträgt 12 g. Nach Umkristallisieren aus Wasser schmilzt das Produkt bei 217-218 C.
Beispiel 3
10 g 1-Phenyl-1-hydroxy-2-morpholino-äthan werden in 30 ml Benzol gelöst. Dann werden portionsweise 15 g wasserfreies Aluminiumchlorid so zugesetzt, dass das Reaktionsgemisch nicht siedet. Danach wird 30 Minuten lang auf dem Wasserbad erhitzt. Nach dem Abkühlen wird das Reaktionsgemisch in eine Mischung aus Eis, Wasser und konz. Salzsäure gegossen. Nach einigen Minuten kristallisiert das 1,1-Diphe nyl-2-morpholino-äthan-hydrochlorid in einer Ausbeute von 14,6 g.
Nach Umkristallisieren aus Isopropanol/Ather schmilzt die Verbindung bei 211-213 C.
Beispiel 4
In eine Lösung von 5 g 1-Phenyl-1-hydroxy-2-ben- zylaminoäthan in 20 ml Toluol werden 8 g wasserfreies Aluminiumchlorid so eingetragen, dass das Toluol nicht siedet. Danach wird das Reaktionsgemisch 30 Minuten lang zum Sieden erhitzt und nach dem Abkühlen in eine Mischung aus Eis, Wasser und konz.
Salzsäure gegossen. Es kristallisiert das 1-Phenyl-1-p- tolyl-2-benzylaminoäthan-hydrochlorid in einer Ausbeute von 92 /o. Durch Umkristallisieren aus Isopropan/Äther erhält man farblose Nadeln vom Schmelzpunkt 203-205 C.
Beispiel 5
12 g Styroloxyd werden mit 13,5 g 1-Phenyl-prop ylamdn- (2) vermischt und mehrere Stunden auf 100-120 C erhitzt. Man erhält 1-Phenyl-1-hydroxy- 2- (1-penyl-propyl-(2)-am ! ino)-äthan als fast farbloses 01. Eine Lösung dieses Öles in 80 ml Benzol wird por tionsweise mit 20 g wasserfreiem Aluminiumchlorid versetzt. Die Mischung wird 30 Minuten lang unter Rückfluss auf dem Wasserbad zum Sieden erhitzt und nach dem Abkühlen in eine Mischung von Eis, Wasser und Salzsäure eingegossen. Das Hydrochlorid des 1,1 Diphenyl-2- (1-phenyl-propyl- (2)-amino)-äthan scheidet sich als 01 ab. Die freie Base wird durch Zusatz von Natronlauge und Extrahieren mit Ather isoliert.
Nach Zusatz einer alkoholischen Lösung von Maleinsäure kristallisiert das Maleinat vom Schmelzpunkt 168 bis 170 C in einer Ausbeute von 88 /o.
Beispiel 6
Eine Lösung von 14 g 1-Phenyl-1-hydroxy-2-ami- no-äthan und 13,7 g Phenylaceton in 40 ml Methanol wird mit 4 ml Wasser und anschliessend portionsweise mit 1,5 g Natriumborhydrid versetzt. Nach einstündi- gem Stehen bei Zimmertemperatur wird das Reaktionsgemisch im Vakuum eingedampft. Der ölige Rückstand d wird in 40 ml Toluol aufgenommen, portionsweise mit 20 g wasserfreiem Aluminiumchlorid versetzt und wie in Beispiel 5 beschrieben weiterbehandelt. Man erhält l-Phenyl-1-p-tolyl-2- (l-phenyl-propyl-(2)-amino)-äthan als farbloses 01. Nach Zugabe von Maleinsäure erhält man das Maleinat, Schmelzpunkt 166-168 C.
Beispiel 7
Eine Suspension von 29 g l-Phenyl-2-cinnamyl- amino-propan-hydrochlorid vom Schmelzpunkt 237 bis 240 (hergestellt durch Kondensation von Zimtaldehyd mit 1-Phenyl-propylamin- (2) und Reduktion der so erhaltenen Schiff-Base mit Natriumborhydnid) in 100 ml Benzol wird portionsweise mit 30 g wasserfreiem Aluminiumchlorid versetzt und anschliessend 45 Minuten lang zum Sieden erhitzt. Nach dem Erkalten wird das Reaktionsgemisch wie in Beispiel 1 beschrieben in eine Mischung aus Eis und Salzsäure gegossen.
Nach Zugabe von Ather wird das Kristallisat abfiltriert, zur Abtrennung von anorganischen Salzen in Methanol aufgenommen und durch Zugabe von Wasser ausgefüllt. Man erhält 26,5 g 1, l-Diphenyl-3-(l- phenyl-propyl- (2)-amino)-propan-hydrochlorid, das nach dem Umkristallisieren aus Isopropanol bei 190 bis 192 C schmilzt.

The present arylation reaction is carried out e.g. B. carried out in a suitable solvent. The 1-phenyl-1-chloroalkylamines thus obtained are condensed in one of the solvents mentioned with the desired reactant and the Friedel-Crafts catalyst at elevated temperature to give diphenylalkylamines. These two process steps can be carried out as a one-pot process.
According to a further advantageous embodiment of the process, the 1-phenyl-1-hydroxy-alkylamine compounds can be reacted directly with the reactants mentioned in the presence of Friedel-Crafts catalysts in the solvents mentioned. The reaction can be carried out in the same way starting from the l-phenyl-1,2-unsaturated alkyl amine compounds mentioned.
The products of the process can be converted into the corresponding salts as basic compounds with the aid of physiologically compatible inorganic or organic acids. Inorganic acids that can be used are, for example: hydrohalic acids such as hydrochloric acid, sulfuric acid, phosphoric acid and sulfamic acid.
Examples of organic acids are acetic acid, propionic acid, lactic acid, glycolic acid, gluconic acid, maleic acid, succinic acid, tartaric acid, salicylic acid and citric acid.
The products of the process can be administered parentally or orally as such or in the form of their salts, if appropriate also as a mixture with conventional pharmaceutical carriers. In the case of oral administration, the preferred dosage forms are tablets or dragees, into which the process products are processed as active ingredients with the usual carrier substances, such as lactose, starch, tragacanth and magnesium stearate.
The previously known methods for producing the substances mentioned here were difficult and time-consuming. Some of the raw materials they needed were difficult to access. Here the present new process opens up access to the group of substances mentioned, which can be carried out on a scale required for industrial use. The yields achieved by this process are between 80 ouzo and 95 "/ o.

25 g of 1-phenyl-1-hydroxy-3- (1-phenyl-propyl- (2) -amino) -propane hydrochloride with a melting point of 144-146 ° C. crystallize.
Z5 g of the compound thus obtained are introduced in portions into a solution of 40 ml of thionyl chloride in 80 ml of benzene at room temperature. A strong evolution of hydrogen chloride and sulfur dioxide begins. After some time, the hydrochloride of 1-phenyl-1-chloro-3- (1-phenyl-propyl- (2) -amino) -propane crystallizes. The mixture is then refluxed for 30 minutes. After cooling, the reaction mixture is concentrated in a mixture of 20 ml.
Poured hydrochloric acid, 10 ml of water and 100 g of ice. After standing for several hours and adding ether, the hydrochloride of 1,1-diphenyl-3- (1-phenyl-propyl- (2) -amino) -propane separates out in almost colorless crystals.
Example 2
The crude yield is 11.2 g from melting point, 186-188 ° C, (from aqueous methanol 190-192 ° C).
15 g of 1-phenyl-1-hydroxypropylamine- (3) are added in portions to a mixture of 16 ml of thionyl chloride and 30 ml of benzene. The mixture is then refluxed on the water bath for a further 20 minutes. 10 g of the hydrochloride thus obtained are suspended in 40 ml of benzene and 12 g of anhydrous aluminum chloride are added in portions. After 30 minutes of heating in the water bath, the reaction mixture is poured into a mixture of hydrochloric acid, water and ice. The hydrochloride of 1,1-Diphenyl-propylamin- (3) crystallizes with a melting point of 206-209 C. The yield is 12 g. After recrystallization from water, the product melts at 217-218 C.
Example 3
10 g of 1-phenyl-1-hydroxy-2-morpholino-ethane are dissolved in 30 ml of benzene. Then 15 g of anhydrous aluminum chloride are added in portions so that the reaction mixture does not boil. It is then heated on the water bath for 30 minutes. After cooling, the reaction mixture is poured into a mixture of ice, water and conc. Poured hydrochloric acid. After a few minutes, the 1,1-Diphe nyl-2-morpholino-ethane hydrochloride crystallizes in a yield of 14.6 g.
After recrystallization from isopropanol / ether, the compound melts at 211-213 C.
Example 4
8 g of anhydrous aluminum chloride are introduced into a solution of 5 g of 1-phenyl-1-hydroxy-2-benzylaminoethane in 20 ml of toluene in such a way that the toluene does not boil. The reaction mixture is then heated to the boil for 30 minutes and, after cooling, poured into a mixture of ice, water and conc.
Poured hydrochloric acid. The 1-phenyl-1-p-tolyl-2-benzylaminoethane hydrochloride crystallizes in a yield of 92%. Recrystallization from isopropane / ether gives colorless needles with a melting point of 203-205 C.
Example 5
12 g of styrene oxide are mixed with 13.5 g of 1-phenyl-prop ylamdn- (2) and heated to 100-120 ° C. for several hours. 1-Phenyl-1-hydroxy- 2- (1-penyl-propyl- (2) -am! Ino) -ethane is obtained as an almost colorless oil. A solution of this oil in 80 ml of benzene is mixed with 20 g of anhydrous aluminum chloride by the portion offset. The mixture is heated to boiling under reflux on a water bath for 30 minutes and, after cooling, poured into a mixture of ice, water and hydrochloric acid. The hydrochloride of 1,1 diphenyl-2- (1-phenyl-propyl- (2) -amino) -ethane separates out as 01. The free base is isolated by adding sodium hydroxide solution and extracting with ether.
After adding an alcoholic solution of maleic acid, the maleate crystallizes from a melting point of 168 to 170 ° C. in a yield of 88%.
Example 6
A solution of 14 g of 1-phenyl-1-hydroxy-2-amino-ethane and 13.7 g of phenylacetone in 40 ml of methanol is mixed with 4 ml of water and then in portions with 1.5 g of sodium borohydride. After standing for one hour at room temperature, the reaction mixture is evaporated in vacuo. The oily residue d is taken up in 40 ml of toluene, treated in portions with 20 g of anhydrous aluminum chloride and treated further as described in Example 5. L-phenyl-1-p-tolyl-2- (l-phenyl-propyl- (2) -amino) -ethane is obtained as colorless oil. After adding maleic acid, the maleate is obtained, melting point 166-168 C.
Example 7
A suspension of 29 g of l-phenyl-2-cinnamylamino-propane hydrochloride with a melting point of 237 to 240 (produced by condensation of cinnamaldehyde with 1-phenylpropylamine- (2) and reduction of the Schiff base thus obtained with sodium borohydnide) 30 g of anhydrous aluminum chloride are added in portions to 100 ml of benzene and the mixture is then heated to the boil for 45 minutes. After cooling, the reaction mixture is poured into a mixture of ice and hydrochloric acid as described in Example 1.
