Erfindungsgemäss gelangt man zu den Verbindungen der Formel I, indem man eine Verbindung der Formel II, worin X, Y und R2 obige Bedeutung besitzen und Rs für Halogen steht, mit einer Verbindung der Formel III, worin Rl obige Bedeutung besitzt, umsetzt.
Das erfindungsgemässe Verfahren kann beispielsweise in einem unter den Reaktionsbedingungen inerten Lösungsmittel, z. B. in einem aromatischen Kohlenwasserstoff wie Benzol, vorzugsweise bei erhöhter Temperatur, z. B. bei Siedetemperatur des Reaktionsgemisches und unter Wasserausschluss durchgeführt werden. Aus dem Reaktionsgemisch kann das Endprodukt nach an sich bekannten Methoden isoliert und gegebenenfalls gereinigt werden.
Die Verbindungen der Formel I können in ihre Säureadditionssalze überführt werden und umgekehrt.
Die durch R2, R3 oder R4 dargestellten, bzw. darin enthaltenen niederen Alkylgruppen besitzen vorzugsweise 1 bis 4, insbesondere 1 bis 2 Kohlenstoffatome. Die durch Rl symbolisierten niederen Piperazinylgruppen können vorzugsweise am zweiten Stickstoffatom in Position 4 des Ringes substituiert sein, und zwar z. B. durch eine niedere Alkyl-, eine niedere Hydroxyalkyl- oder eine niedere(nieder Alkoxy)alkylgruppe, wobei gegebenenfalls auch eine Disubstitution mit gleichzeitiger Quartärnisierung des Stickstoffs möglich ist.
Die Verbindungen der Formel I besitzen bei geringer Toxizität interessante pharmakodynamische Eigenschaften und können daher als Heilmittel verwendet werden. Sie entfalten eine Hemmwirkung gegen Bakterien, wie sich durch Untersuchungen in vitro mit dem Agarplattentest unter Verwendung verschiedener Bakterienstämme zeigen lässt. Diese Hemmwirkung wurde ab einer Konzentration von ca. 1 bis 50 Fg/ml festgestellt. Daher können die erfindungsgemässen Verbindungen als Hemmstoffe des Bakterienwachstums verwendet werden.
Als Heilmittel können die Verbindungen der Formel I allein oder in geeigneten Arzneiformen gemeinsam mit anorganischen oder organischen, pharmakologisch indifferenten Hilfsstoffen verabreicht werden. Beispielsweise werden sie als Bestandteil von Kapseln oder Tabletten eingesetzt, wobei die Konzentration in der zubereiteten Form ca. 50 bis 500 mg Wirkstoff pro Einheit der Dosisform betragen kann.
Weiters entfalten die Verbindungen der Formel I auch eine abtötende Wirkung gegen Trichomonaden und Amöben, wie sich durch in vitro Untersuchungen mit dem Reihenverdünnungstest wie auch in vivo Versuchen in der Maus zeigen lässt.
Die im CACH-Medium (Müller et al., Angew. Parasit. 11, 170-176 (1970) geprüfte Trichomonadenwirksamkeit liegt im Bereich von 1-2 gFlg/ml. Im TTY-SB-Medium (Diamond L. S., J. Parasitol. 54, 715-719 (1968) erwiesen sich die Substanzen im Konzentrationsbereich von 12-25 llg/ml amöbizid. Die trichomonazide Wirksamkeit wurde in der Maus mit einer Dosis ab ca. 10 mg/kg Körpergewicht bei oraler oder parenteraler Applikation nachgewiesen.
Die Substanzen erscheinen zur chemotherapeutischen Behandlung der verschiedenen Formen der Trichomoniasis und Amöbiasis geeignet, wobei ihre systemische Wirkung von besonderem Vorteil angesehen wird.
Als Heilmittel können die Verbindungen der Formel I allein oder in geeigneten Arzneiformen gemeinsam mit anorganischen oder anorganischen, pharmakologisch indifferenten Hilfsstoffen verabreicht werden. Beispielsweise werden sie als Bestandteil von Kapseln, Tabletten oder einer Injektionslösung eingesetzt, wobei die Kapseln oder Tabletten z. B. 10 bis 500 mg Wirkstoff pro Kapsel oder Tablette enthalten können.
Die als Ausgangsprodukte benötigten Verbindungen der Formel II können erhalten werden, indem man eine Verbindung der Formel IV, worin X und Y obige Bedeutung besitzen und R6 für eine niedere Alkylgruppe steht, mit einer Verbindung der Formel V, worin R2 obige Bedeutung besitzt, zu Verbindungen der Formel VI, worin X, Y und R2 obige Bedeutung besitzen, umsetzt und diese anschliessend halogeniert.
Beispielsweise kann das Verfahren in einem unter den Reaktionsbedingungen inerten Lösungsmittel, z. B. in einem niederen Alkohol wie Methanol bei Raumtemperatur durchgeführt werden. Die Halogenierung wird vorzugsweise mit Phosphorpentachlorid als Halogenierungsmittel vorzugsweise bei erhöhter Temperatur, z. B. bei Siedetemperatur des Reaktionsgemisches und unter Wasserausschluss ausgeführt.
Soweit die Herstellung der Ausgangsprodukte nicht beschrieben wird, sind diese bekannt oder nach an sich bekannten Verfahren bzw. analog zu den hier beschriebenen oder analog zu an sich bekannten Verfahren herstellbar.
In den nachfolgenden Beispielen, die die Erfindung näher erläutern, ihren Umfang aber in keiner Weise einschränken sollen, erfolgen alle Temperaturangaben in Celsiusgraden.
Beispiel 1 2-(5-Nitro-2-thiazolyl)-4-(4-N-methyl- piperazinyl)chinazolin
2,6 g 4-Chlor-2-(5-nitro-2-thiazolyl)chinazolin in 80 ml absolutem Benzol mit 1,8 g 4-N-Methylpiperazin 3 Stunden unter Rückfluss und Feuchtigkeitsausschluss gekocht. Dabei geht das Ausgangsmaterial zunächst in Lösung und ein Teil der Substanz fällt als Dihydrochlorid aus. Die Mutterlauge wird bei 40ob/1 Torr eingedampft, der ölige Rückstand in Chloroform aufgenommen und unter Zusatz von ätherischer Salzsäure weiteres Dihydrochlorid ausgefällt.

Beispiel 2
2-(5 -Nitro-2-thiazolyl)-4-(4-N-dimethyl) piperazinylium)chinazolin-jodid
Man verfährt wie in Beispiel 1 beschrieben, nimmt 2,2 g der erhaltenen freien Base in 50 ml Chloroform und 10 ml Acetonitril auf, versetzt mit 6 ml Methylenjodid und lässt über Nacht stehen. Man saugt vom gebildeten Kristallisat ab und wäscht mit Chloroform nach.

Das als Ausgangsprodukt benötigte 4-Chlor-2-(5-nitro2-thiazolyl)chinazolin kann folgendermassen erhalten werden: a) 2-(5-Nitro-2-thiazolyl)chinazolon-(4)
4,9 g 5-Nitro-2-thiazolcarbiminoäthyläther werden mit 5,08 g 2-Aminobenzoesäure in 50 ml Methanol 80 Stunden bei Zimmertemperatur stehen gelassen. Man saugt dann von bereits chromatographisch reinem Kristallisat ab und dampft die Mutterlauge auf ca. 15 ml ein. Nach kurzem Stehen ist noch eine zweite kristalline Fraktion abtrennbar. Man vereinigt, kocht in wenig Methanol auf, filtriert und wäscht mit Äther nach.

b) 4-Chlor-2-(5 -nitro-2-thiazolyl)chinazolin:
4 g 2-(5-Nitro-2-thiazolyl)chinazolon-(4) werden in 170 ml Phosphoroxychlorid suspendiert, 3,4 g Phosphorpentachlorid zugesetzt und unter Wasserausschluss 2 Stunden unter Rückfluss erhitzt. Es wird anschliessend bei ca. 1 Torr in Vakuum konzentriert, der Rückstand in Wasser suspendiert und mit Methylenchlorid mehrfach extrahiert. Man wäscht die Lösung mit 5 %iger wässriger Natriumhydrogenkarbonatlösung bis zur neutralen Reaktion, trocknet über Magnesiumsulfat und dampft im Vakuum ein. Nach dem Umfällen aus Methylenchlorid/Äther wird die Titelsubstanz mit einem Smp. von 2112120 erhalten.

According to the invention, the compounds of the formula I are obtained by reacting a compound of the formula II in which X, Y and R2 have the above meaning and Rs is halogen with a compound of the formula III in which Rl has the above meaning.
The inventive method can, for example, in a solvent inert under the reaction conditions, eg. B. in an aromatic hydrocarbon such as benzene, preferably at an elevated temperature, e.g. B. be carried out at the boiling point of the reaction mixture and with exclusion of water. The end product can be isolated from the reaction mixture by methods known per se and, if necessary, purified.
The compounds of the formula I can be converted into their acid addition salts and vice versa.
The lower alkyl groups represented by R2, R3 or R4 or contained therein preferably have 1 to 4, in particular 1 to 2, carbon atoms. The lower piperazinyl groups symbolized by Rl can preferably be substituted on the second nitrogen atom in position 4 of the ring, namely e.g. B. by a lower alkyl, a lower hydroxyalkyl or a lower (lower alkoxy) alkyl group, where appropriate disubstitution with simultaneous quaternization of the nitrogen is also possible.
The compounds of the formula I have interesting pharmacodynamic properties with low toxicity and can therefore be used as medicaments. They develop an inhibitory effect against bacteria, as can be shown by studies in vitro with the agar plate test using different bacterial strains. This inhibitory effect was found from a concentration of about 1 to 50 μg / ml. Therefore, the compounds of the present invention can be used as bacterial growth inhibitors.
As medicaments, the compounds of the formula I can be administered alone or in suitable pharmaceutical forms together with inorganic or organic, pharmacologically indifferent auxiliaries. For example, they are used as a component of capsules or tablets, the concentration in the prepared form being approximately 50 to 500 mg of active ingredient per unit of the dosage form.
Furthermore, the compounds of the formula I also develop a killing effect against trichomonads and amoebas, as can be shown by in vitro studies with the serial dilution test and in vivo tests in the mouse.
The trichomonad activity tested in CACH medium (Müller et al., Angew. Parasit. 11, 170-176 (1970) is in the range of 1-2 gFlg / ml. In TTY-SB medium (Diamond LS, J. Parasitol. 54, 715-719 (1968), the substances were found to be amebicidal in the concentration range of 12-25 μg / ml. The trichomonacidal activity was demonstrated in the mouse with a dose of about 10 mg / kg body weight with oral or parenteral administration.
The substances appear to be suitable for the chemotherapeutic treatment of the various forms of trichomoniasis and amebiasis, their systemic action being considered to be particularly advantageous.

The compounds of the formula II required as starting materials can be obtained by combining a compound of the formula IV, in which X and Y have the above meaning and R6 is a lower alkyl group, with a compound of the formula V, in which R2 has the above meaning, to form compounds of the formula VI, in which X, Y and R2 have the above meaning, reacted and then halogenated.
For example, the process can be carried out in a solvent which is inert under the reaction conditions, e.g. B. be carried out in a lower alcohol such as methanol at room temperature. The halogenation is preferably carried out with phosphorus pentachloride as the halogenating agent, preferably at an elevated temperature, e.g. B. carried out at the boiling point of the reaction mixture and with exclusion of water.
If the production of the starting products is not described, these are known or can be produced by processes known per se or analogously to those described here or analogously to processes known per se.
In the following examples, which explain the invention in greater detail but are not intended to limit its scope in any way, all temperatures are given in degrees Celsius.
Example 1 2- (5-Nitro-2-thiazolyl) -4- (4-N-methyl-piperazinyl) quinazoline
2.6 g of 4-chloro-2- (5-nitro-2-thiazolyl) quinazoline in 80 ml of absolute benzene with 1.8 g of 4-N-methylpiperazine were refluxed with exclusion of moisture for 3 hours. The starting material initially goes into solution and part of the substance precipitates as dihydrochloride. The mother liquor is evaporated at 40ob / 1 Torr, the oily residue is taken up in chloroform and further dihydrochloride is precipitated with the addition of ethereal hydrochloric acid.

Example 2
2- (5 -Nitro-2-thiazolyl) -4- (4-N-dimethyl) piperazinylium) quinazoline iodide
The procedure described in Example 1 is followed, 2.2 g of the free base obtained are taken up in 50 ml of chloroform and 10 ml of acetonitrile, 6 ml of methylene iodide are added and the mixture is left to stand overnight. The crystals formed are filtered off with suction and washed with chloroform.

4.9 g of 5-nitro-2-thiazolecarbiminoethyl ether are left to stand for 80 hours at room temperature with 5.08 g of 2-aminobenzoic acid in 50 ml of methanol. Crystals that are already chromatographically pure are then filtered off with suction and the mother liquor is evaporated to approx. 15 ml. After standing for a short time, a second crystalline fraction can be separated off. They are combined, boiled up in a little methanol, filtered and washed with ether.

b) 4-chloro-2- (5-nitro-2-thiazolyl) quinazoline:
4 g of 2- (5-nitro-2-thiazolyl) quinazolon- (4) are suspended in 170 ml of phosphorus oxychloride, 3.4 g of phosphorus pentachloride are added and the mixture is heated under reflux with exclusion of water for 2 hours. It is then concentrated in vacuo at about 1 torr, the residue is suspended in water and extracted several times with methylene chloride. The solution is washed with 5% strength aqueous sodium hydrogen carbonate solution until the reaction is neutral, dried over magnesium sulfate and evaporated in vacuo. After reprecipitation from methylene chloride / ether, the title substance with a melting point of 2112120 is obtained.
