Verfahren zur Herstellung neuer Nitrothiazolverbindungen
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von Nitrothiazolverbindungen der Formel

worin R1 und R2 niedere Alkylreste oder insbesondere Wasserstoffatome und R eine gegebenenfalls substituierte Aminogruppe bedeuten, oder ihrer Salze.
Als niedere Alkylgruppen kommen beispielsweise Methyl-, Äthyl-, Propyl-, Isopropyl-, gerade oder verzweigte in beliebiger Stellung verbundene Butyl-, Pentyloder Hexylgruppen in Betracht.
Die Aminogruppe R ist vorzugsweise mono- oder disubstituiert; insbesondere ist sie eine tertiäre Aminogruppe. Als Substituenten kommen insbesondere Kohlenwasserstoffreste aliphatischen Charakters, die auch durch Heteroatome, wie Stickstoff-, Sauerstoff- oder Schwefelatome, unterbrochen sein und/oder durch Hydroxylgruppen oder Halogenatome, wie Chlor oder Brom substituiert sein können, in Betracht, wie Alkyl-, Hydroxyalkyl-, Halogenalkyl-, Alkenyl-, Cycloalkyl-, Cycloalkyl-alkyl-, Cycloalkenyl-, Cycloalkenylalkyl-, Aralkyl- oder Alkylenreste oder Oxa-, Aza- oder Thiaalkylenreste oder Niederalkoxy-niederalkylreste. Als Alkylreste sind insbesondere niedere Alkylreste, z.
B. die oben genannten, als Alkenylreste insbesondere niedere Alkenylreste, wie Allyl- oder Methallylreste, als Cycloalkylreste insbesondere Cyclopentyl-, Cyclohexyloder Cycloheptylreste, als Cycloalkenylreste insbesondere Cyclopentenyl- oder Cyclohexenylreste zu nennen.
Aralkylreste sind insbesondere Phenylniederalkylreste, die auch durch niedere Alkylgruppen, niedere Alkoxygruppen, z. B. Methoxy- oder Äthoxyreste, Halogenatome, wie Chlor- oder Bromatome, und/oder Trifluormethylgruppen substituiert sein können, wie Benzyl-, a-Phenyläthyl- oder ss-Phenyläthylreste. Alkylenreste, in denen die Alkylenkette durch Heteroatome unterbrochen sein kann, wie Aza-, Thia- oder Oxa-alkylenreste sind z. B.
Butylen-(1,4)-, Pentylen-(1,5)-, Hexylen-(1,5)-, Hexylen-(1, 6)-, Heptylen-(2,6)-, 3 -Oxa- oder 3-Aza pentylen-(1,5)-, 3-Phenyl-3-aza-pentylen-(1,5)-, 3-Nie deralkyl-3-aza-pentylen-( 1,5)-, wie 3-Methyl- oder 3 Athyl-3-aza-pentylen-(1 5)-, 3-Aza-hexylen-(1,6)-, 4 Methyl-4-aza-heptylen-(2, 6)- oder 3 -Thia-pentylen-( 1,5)- reste. Ein Niederalkoxy-niederalkylrest ist besonders der ss-Methoxyäthylrest. Hydroxyalkyl- oder Halogenalkylreste sind insbesondere niedere Hydroxyalkyl- oder Halogenalkylreste, wie ss-Hydroxyäthyl-, ss-Chloräthyloder y-Brompropylreste.
Die Aminogruppe R ist vorzugsweise eine niedere Mono- oder Dialkylaminogruppe oder vor allem eine Pyrrolidino-, Piperidino-, Hexamethylenimino-, Heptamethylenimino-, Morpholino- oder Thiamorpholinogruppe oder eine Piperazinogruppe, wie die N-Methyl piperazinogruppe.
Die neuen Verbindungen besitzen wertvolle pharmakologische, insbesondere antiparasitäre und antibakterielle Eigenschaften. Sie zeigen vor allem eine Wirkung gegen Protozoen und Würmer und sind, z. B. am infizierten Tier, beispielsweise an Mäusen, gegen grampositive und besonders gegen gram-negative Bakterien, z. B. Salmonella typhi oder Coli-Bazillen, wie Esch. coli, wirksam. Insbesondere wirken die neuen Verbindungen, wie sich z. B. bei Versuchen an Hamstern zeigt, gegen Trichomonaden und Amoeben, vor allem auch gegen Amoeben in der Leber, sowie z. B. an Mäusen und Schafen, gegen Schistosomen. Ferner besitzen sie eine Wirkung gegen Coccidien und gegen den Erreger von Enterohepatitis (Blackhead). Die neuen Verbindungen sind entsprechend als antiparasitäre und antibakterielle Mittel nützlich. Sie können auch als Harndesinfizienten verwendet werden.
Insbesondere eignen sie sich zur Behandlung der durch die genannten Erreger verursachten Erkrankungen. Die neuen Verbindungen sind aber auch wertvolle Zwischenprodukte für die Herstellung anderer nützlicher Stoffe.
Besonders hervorzuheben sind Verbindungen der Formel

Das erfindungsgemässe Verfahren zur Herstellung der neuen Verbindungen ist dadurch gekennzeichnet, dass man Verbindungen der Formel

worin X' eine freie oder eine reaktionsfähig abgewandelte Oxygruppe, wie eine reaktionsfähig veresterte oder verätherte Oxygruppe, vor allem ein Halogenatom, insbesondere ein Chlor- oder Bromatom bedeutet, steht, wobei eine Reaktionskomponente gegebenenfalls in Form eines Salzes vorliegt.
Die erfindungsgemässe Reaktion kann in üblicher Weise erfolgen, vorzugsweise in Anwesenheit von Lösungs- und/oder Verdünnungsmitteln, gegebenenfalls in Anwesenheit von Kondensationsmitteln, wie basischen Kondensationsmitteln, bei tiefer, gewöhnlicher oder erhöhter Temperatur im offenen oder im geschlossenen Gefäss unter Druck.
Verbindungen, die auch basische Gruppen enthalten, erhält man je nach der Arbeitsweise in freier Form oder in Form ihrer Salze.
Aus den Basen können therapeutisch verwendbare Salze mit Säuren gebildet werden, z. B. von therapeutisch verwendbaren Säuren, wie der Halogenwasserstoffsäuren, Schwefelsäuren, Phosphorsäuren, Salpetersäure, Perchlorsäure; aliphatischer, alicyclischer, aromatischer oder heterocyclischer Carbon- oder Sulfonsäuren, wie Ameisen-, Essig-, Propion-, Oxal-, Bernstein-, Glykol-, Milch-, Äpfel-, Wein-, Zitronen-, Ascorbin-, Oxymalein-, Dioxymalein- oder Brenztrauben- säure; Phenylessig-, Benzoe-, p-Aminobenzoe-, Anthranil-, p-Oxybenzoe-, Embon-, Salicyl- oder p-Aminosalicylsäure; Methansulfon-, Athansulfon-, Oxyäthansulfon-, Äthylensulfonsäure; Toluolsulfon-, Naphtalinsulfonsäuren oder Sulfanilsäure;
Methionin, Tryptophan, Lysin oder Arginin Anderseits lassen sich auch von sauren Verbindungen mit Basen, wie mit Metallhydroxyden, Salze, z. B. Metallsalze, wie Alkali oder Erdalkall- salze herstellen. Erhaltene Salze lassen sich in die freien Verbindungen umwandeln.
Bei Verbindungen, die basische Gruppen enthalten, lassen sich die Salze auch zur Reinigung der freien Verbindungen verwenden. Infolge der engen Beziehungen zwischen solchen Verbindungen in freier Form und in Form ihrer Salze sind im vorausgegangenen und nachfolgend unter den freien Verbindungen sinn- und zweckgemäss gegebenenfalls auch die entsprechenden Salze zu verstehen.
Die Erfindung betrifft auch diejenigen Ausführungsformen des Verfahrens, bei denen eine Reaktionskomponente gegebenenfalls in Form eines Salzes vorliegt.
Für die erfindungsgemässen Reaktionen werden vornehmlich solche Ausgangs stoffe verwendet, die die oben erwähnten bevorzugten Verbindungen ergeben.
Die Ausgangsstoffe sind bekannt oder können nach an sich bekannten Methoden gewonnen werden.

worin X' die angegebene Bedeutung hat und X" eine reaktionsfähig abgewandelte, wie eine reaktionsfähig veresterte oder verätherte Oxygruppe, vor allem ein Halogenatom, insbesondere ein Chlor- oder Bromatom bedeutet, umsetzt. Die Umsetzung erfolgt in üblicher Weise. Der oben genannte Ausgangsstoff der Formel V braucht dabei nicht isoliert zu werden, sondern man kann diesen direkt ohne Isolierung nach dem erfindungsgemässen Verfahren in das Oxamid überführen.
Die neuen Verbindungen können z. B. in Form pharmazeutischer Präparate, Verwendung finden, welche sie in Mischung mit einem für die enterale, parenterale oder topicale Applikation geeigneten pharmazeutischen organischen oder anorganischen, festen oder flüssigen Trägermaterial enthalten.
Die neuen Verbindungen können auch in der Tiermedizin, z. B. in einer der oben genannten Formen oder in Form von Futtermitteln oder von Zusatzmitteln für Tierlutter verwendet werden.
In den folgenden Beispielen sind die Temperaturen in Celsiusgraden angegeben.
Beispiel 1
Zu einer Lösung von 6,35 g Oxalylchlorid in 150 ml absolutem Tetrahydrofuran werden bei einer Temperatur von 200 innert 30 Minuten 7,25 g 5-Nitro-2-amino- thiazol, gelöst in 100 ml absolutem Tetrahydrofuran, eingetropft. Man rührt eine halbe Stunde bei Raumtemperatur weiter und tropft dann eine Lösung von 3,7 g n-Butylamin in 20 mi absolutem Tetrahydrofuran dazu. Anschliessend wird während 1 Stunde unter Rühren zum Sieden erhitzt. Nach dem Abkühlen wird der ausgefallene Niederschlag abgenutscht und mit Wasser ausgewaschen.
Durch Umkristallisation aus viel Äthanol erhält man das Nt-(n-Butyl)-N2-(5-nitro-2-thiazolyl)- oxamid der Formel

Beispiel 2
Zu einer Lösung von 6,35 g Oxalylchlorid in 150 ml absolutem Tetrahydrofuran werden bei einer Temperatur von 200 innert 30 Minuten 7,25 g 5-Nitro-2-aminothiazol, gelöst in 100 ml absolutem Tetrahydrofuran, eingetropft. Man rührt eine halbe Stunde bei Raumtemperatur weiter und tropft dann innert 10 Minuten bei Raumtemperatur eine Lösung von 6,05 g Methylbenzylamin in 20 ml absolutem Tetrahydrofuran dazu.
Anschliessend erhitzt man während 1 Stunde zum Sieden.
Nach dem Erkalten wird das ausgefallene Salz abgenutscht und die Tetrahydrofuranlösung in 1,5 ml Wasser eingegossen und der Niederschlag nach dem Erstarren abgenutscht. Man kristallisiert das erhaltene Produkt unter Abtrennen des unlöslichen Anteils aus Äthanol um und erhält so das Nl-Methyl-Nt-benzyl-N2- (5-nitro-2-thiazolyl)-oxamid der Formel

Beispiel 3
Zu einer Lösung von 6,35 g Oxalylchlorid in 100 ml absolutem Tetrahydrofuran werden bei einer Temperatur von 200 innert 30 Minuten 7,25 g 5-Nitro-2-aminothiazol, gelöst in 100 ml absolutem Tetrahydrofuran, eingetropft. Man lässt eine halbe Stunde bei Raumtemperatur weiterrühren und tropft dann innert 10 Minuten bei Raumtemperatur eine Lösung von 6,5 g 2,6-Dimethyl-thiomorpholin in 20 ml absolutem Tetrahydrofuran dazu. Anschliessend erhitzt man während 1 Stunde unter Rühren zum Sieden. Die Reaktionslösung wird nach dem Abkühlen in 2 1 Wasser gegossen und der ausgefallene Niederschlag abgenutscht. Man löst das erhaltene Produkt in viel heissem Äthanol und trennt den unlöslichen Anteil ab.
Aus dem Äthanol erhält man das N1,N1-(3-Thia-2,4-dimethyl-1,5-pentylen) N2-(5-m.tro-2qhiazolyl)-oxamid der Formel

Beispiel 4
Zu einer Lösung von 6,35 g Oxalylchlorid in 100 ml absolutem Tetrahydrofuran werden bei einer Temperatur von 200 innert 30 Minuten 7,25 g 5-Nitro-2-aminothiazol, gelöst in 100 ml absolutem Tetrahydrofuran, eingetropft. Man lässt dann eine halbe Stunde bei Raumtemperatur weiterrühren und tropft hierauf innert 10 Minuten bei Raumtemperatur eine Lösung von 4,75 g Morpholin in 20 ml absolutem Tetrahydrofuran dazu.
Anschliessend wird während einer Stunde unter Rühren zum Sieden erhitzt. Das ausgefallene Reaktionsprodukt wird nach dem Abkühlen abgenutscht und mit Wasser verrieben. Man kocht mit sehr viel Athanol aus und trennt das unlösliche Produkt ab. Aus dem Äthanol kristallisiert das N1,N"-(3-Oxa-l ,5-pentylen)-N2-(5- nitro-2-thiazolyl)-oxamid der Formel

Das erhaltene Produkt wird unter Abtrennen des unlöslichen Anteils aus sehr viel Äthanol umkristalli siert. Man erhält so das N1,N1-(3-Phenyl-3-aza-1 5- pentylen) -N2-(5 -nitro-2-thiazolyl) -oxamid der Formel

Beispiel 6
Zu einer Lösung von 6,35 g Oxalylchlorid in 100 ml absolutem Tetrahydrofuran gibt man bei 160 innert 30 Minuten 7,25 g 5-Nitro-2-aminothiazol, gelöst in 100 ml absolutem Tetrahydrofuran. Anschliessend wird während 30 Minuten bei Raumtemperatur weitergerührt und dann eine Lösung von 4 g wasserfreiem ss-Chlor äthylamin, welches unmittelbar vor der Umsetzung unter gutem Kühlen aus dem Hydrochlorid hergestellt wurde, in 20 ml absolutem Tetrahydrofuran innert 10 Minuten zugetropft. Man erwärmt hierauf innert einer Stunde allmählich bis zum Sieden und kocht dann während einer halben Stunde unter Rühren am Rückfluss.
Die entstandene Trübung wird abfiltriert und das klare Filtrat auf 0 abgekühlt, wobei das Nt-Chloräthyl)- N--(5-nitro-2-thiazolyl)-oxamid der Formel

The new connections can e.g. B. in the form of pharmaceutical preparations, which contain them mixed with a pharmaceutical, organic or inorganic, solid or liquid carrier material suitable for enteral, parenteral or topical application.
The new compounds can also be used in veterinary medicine, e.g. B. can be used in one of the above forms or in the form of feed or additives for animal feed.
In the following examples the temperatures are given in degrees Celsius.
example 1
7.25 g of 5-nitro-2-aminothiazole, dissolved in 100 ml of absolute tetrahydrofuran, are added dropwise to a solution of 6.35 g of oxalyl chloride in 150 ml of absolute tetrahydrofuran at a temperature of 200 within 30 minutes. Stirring is continued for half an hour at room temperature and a solution of 3.7 g of n-butylamine in 20 ml of absolute tetrahydrofuran is then added dropwise. The mixture is then heated to boiling for 1 hour while stirring. After cooling, the deposited precipitate is filtered off with suction and washed out with water.
Recrystallization from a large amount of ethanol gives the Nt- (n-butyl) -N2- (5-nitro-2-thiazolyl) oxamide of the formula

Example 2
7.25 g of 5-nitro-2-aminothiazole, dissolved in 100 ml of absolute tetrahydrofuran, are added dropwise to a solution of 6.35 g of oxalyl chloride in 150 ml of absolute tetrahydrofuran at a temperature of 200 within 30 minutes. The mixture is stirred for a further half an hour at room temperature and then a solution of 6.05 g of methylbenzylamine in 20 ml of absolute tetrahydrofuran is added dropwise over a period of 10 minutes at room temperature.
The mixture is then heated to the boil for 1 hour.
After cooling, the precipitated salt is suction filtered and the tetrahydrofuran solution is poured into 1.5 ml of water and the precipitate is suction filtered after it has solidified. The product obtained is recrystallized from ethanol while separating off the insoluble fraction and thus the Nl-methyl-Nt-benzyl-N2- (5-nitro-2-thiazolyl) oxamide of the formula is obtained

Example 3
7.25 g of 5-nitro-2-aminothiazole, dissolved in 100 ml of absolute tetrahydrofuran, are added dropwise to a solution of 6.35 g of oxalyl chloride in 100 ml of absolute tetrahydrofuran at a temperature of 200 within 30 minutes. The mixture is allowed to stir for a further half an hour at room temperature and then a solution of 6.5 g of 2,6-dimethyl-thiomorpholine in 20 ml of absolute tetrahydrofuran is added dropwise at room temperature over a period of 10 minutes. The mixture is then heated to the boil for 1 hour while stirring. After cooling, the reaction solution is poured into 2 l of water and the precipitate which has separated out is filtered off with suction. The product obtained is dissolved in a lot of hot ethanol and the insoluble fraction is separated off.
The N1, N1- (3-thia-2,4-dimethyl-1,5-pentylene) N2- (5-m.tro-2qhiazolyl) oxamide of the formula is obtained from the ethanol

Example 4
7.25 g of 5-nitro-2-aminothiazole, dissolved in 100 ml of absolute tetrahydrofuran, are added dropwise to a solution of 6.35 g of oxalyl chloride in 100 ml of absolute tetrahydrofuran at a temperature of 200 within 30 minutes. The mixture is then left to stir for a further half an hour at room temperature and a solution of 4.75 g of morpholine in 20 ml of absolute tetrahydrofuran is then added dropwise at room temperature over a period of 10 minutes.
The mixture is then heated to boiling with stirring for one hour. The precipitated reaction product is filtered off with suction after cooling and triturated with water. It is boiled with a great deal of ethanol and the insoluble product is separated off. The N1, N "- (3-oxa-1,5-pentylene) -N2- (5-nitro-2-thiazolyl) -oxamide of the formula crystallizes from the ethanol

Example 5

The product obtained is recrystallized from a large amount of ethanol while separating off the insoluble fraction. The N1, N1- (3-phenyl-3-aza-1 5-pentylene) -N2- (5-nitro-2-thiazolyl) oxamide of the formula is obtained in this way

Example 6
7.25 g of 5-nitro-2-aminothiazole, dissolved in 100 ml of absolute tetrahydrofuran, are added over a period of 30 minutes to a solution of 6.35 g of oxalyl chloride in 100 ml of absolute tetrahydrofuran. The mixture is then stirred for a further 30 minutes at room temperature and then a solution of 4 g of anhydrous β-chlorine ethylamine, which was prepared from the hydrochloride immediately before the reaction with good cooling, in 20 ml of absolute tetrahydrofuran is added dropwise within 10 minutes. The mixture is then gradually heated to boiling over the course of an hour and then refluxed for half an hour while stirring.
The resulting turbidity is filtered off and the clear filtrate is cooled to 0, the Nt-chloroethyl) - N - (5-nitro-2-thiazolyl) oxamide of the formula
