Gegenstand der Erfindung ist ein Verfahren zur Herstellung von 9,10-Dihydro-9, 10-äthano-anthrazenen der Formel

worin R den Methyl- oder Ethylrest darstellt, und ihrer Salze.
Die neuen Verbindungen besitzen wertvolle pharmakologische Eigenschaften. So zeigen sie eine zentralhemmende Wirkung, die durch einen Antagonismus gegenüber psychomotorischen Stoffen, wie z. B. Mescalin, sowie durch eine Hemmung der spinalen Reflex übertragung gekennzeichnet ist, sowie eine histaminolytische Wirkung und können daher als psychotrope, beruhigende Medikamente in der Human- und Veterinärmedizin Verwendung finden. Ihre psychotrope Wirkung ist ausgeprägter und von längerer Dauer als diejenige bekannter Athano-anthrazene. Weiter können die neuen Verbindungen als Ausgangs- oder Zwischenprodukte für die Herstellung anderer wertvoller Verbindungen dienen.
Besonders wertvoll bezüglich seiner pharmakologischen Eigenschaften ist das 2-Chlor-9-diäthylaminomethyl-9 1 0-dihydro-
9,10-äthano-(1,2)-anthrazen der Formel

Das erfindungsgemässe Verfahren zur Herstellung der neuen Verbindungen ist dadurch gekennzeichnet, dass man Verbindungen der Formel

oder Salze davon mit reaktionsfähigen Estern von Alkoholen der Formel R-OH umsetzt.
Ein reaktionsfähiger Ester ist vor allem ein Ester mit einer starken organischen oder anorganischen Säure, wie vor allem einer Halogenwasserstoffsäure, wie Chlor-, Brom- oder Jodwasserstoffsäure, oder einer Sulfonsäure, wie einer Arylsulfonsäure, z. B. der p-Toluolsulfonsäure.
Die Erfindung betrifft auch diejenigen Ausführungsformen des Verfahrens, bei denen ein Ausgangsstoff in Form eines unter den Reaktionsbedingungen gebildeten rohen Reaktionsgemisches eingesetzt wird. So kann man z. B. die Ausgangsstoffe in Form eines unter den Reaktionsbedingungen durch Umsetzen einer Verbindung der Formel

mit einem reaktionsfähigen Ester von Methanol oder Athanol erhältlichen rohen Reaktionsgemisches einsetzen.
Die genannten Reaktionen können in üblicher Weise in An- oder Abwesenheit von Verdünnungs-, Kondensations- und/oder katalytischen Mitteln, bei erniedrigter, gewöhnlicher oder erhöhter Temperatur, gegebenenfalls im geschlossenen Gefäss durchgeführt werden.
Die Ausgangsstoffe können nach an sich bekannten Verfahren gewonnen werden.
Die neuen Verbindungen können je nach den Reak tiousbedingungen und Ausgangsstoffen in freier Form oder in Form ihrer Salze erhalten werden. Die Salze der neuen Verbindungen können in an sich bekannter Weise in die freien Verbindungen übergeführt werden, z. B. Säureadditionssalze durch Reaktion mit einem basischen Mittel. Anderseits können gegebenenfalls erhaltene freie Basen mit anorganischen oder organischen Säuren Salze bilden. Zur Herstellung von Säureadditionssalzen werden insbesondere therapeutisch verwendbare Säuren verwendet, z. B.
Halogenwasserstoffsäuren, beispielsweise Salzsäure oder Bromwasserstoffsäure, Perchlorsäure, Salpetersäure oder Thiocyansäure, Schwefel- oder Phosphorsäuren, oder organische Säuren, wie Ameisensäure, Essigsäure, Propionsäure, Glykolsäure, Milchsäure, Brenztraubensäure, Oxalsäure, Malonsäure, Bernsteinsäure, Maleinsäure, Fumarsäure, Äpfelsäure, Weinsäure, Zitronensäure, Ascorbinsäure, Hydroxymaleinsäure, Dihydroxymaleinsäure, Benzoesäure, Phenylessigsäure, 4-Amino-benzoesäure, 4-Hydroxy-benzoesäure, Anthranilsäure, Zimtsäure, Mandelsäure, Salicylsäure, 4-Amino-salicylsäure, 2-Phenoxybenzoesäure, 2-Acetoxy-benzoesäure, Methansulfonsäure, Athansulfonsäure, Hydroxyäthansulfons äure, Benzolsulfonsäure, p-Toluol-sulfonsäure, Naphthalinsulfonsäure oder Sulfanilsäure, oder Methionin, Tryptophan, Lysin oder Arginin.
Dabei können Mono- oder Polysalze vorliegen.
Die neuen Verbindungen können als Heilmittel in Form von pharmazeutischen Präparaten verwendet werden, welche diese Verbindungen zusammen mit pharmazeutischen, organischen oder anorganischen, festen oder flüssigen Trägerstoffen, die für enterale, z.B. orale, oder parenterale Gabe geeignet sind, enthalten.
Die neuen Verbindungen können auch in der Tiermedizin oder bei der Aufzucht und Ernährung von Tieren in Form von Futtermitteln oder von Zusatzmitteln für Tierfutter verwendet werden.
Die Temperaturen sind in den folgenden Beispielen in Celsiusgraden angegeben.
Beispiel I
Zu einer Lösung von 14 g 2-Chlor-9-methylaminomethyl-9 ,1 0-dihydro-
9,1 0-äthano-(1 ,2)-anthrazen in 150 ml Benzol gibt man 1,2 g Natriumhydrid und erwärmt während einer Stunde unter Rühren auf 700.
Anschliessend werden 7 g Dimethylsulfat zugegeben und weitere 2 Stunden bei 700 gerührt. Das Reaktionsgemisch dampft man im Vakuum ein und versetzt den Rückstand mit 100 ml 2n Natronlauge. Die ausgeschiedene Base wird mit Äther extrahiert. Nach dem Trocknen und Eindampfen des Lösungsmittels verbleibt das

dessen Hydrochlorid bei 244-2450 schmilzt.
Das als Ausgangsmaterial verwendete 2-Chlor-9-methylaminomethyl-9, 1 0-dihydro
9,10-äthano-(1,2)-anthrazen kann folgendermassen hergestellt werden:
Eine Mischung von 42 g 2-Chlor-anthrazen, 54 g N-Methyl-formamid, 60 g Phosphoroxychlorid und 40 cm3 Dichlorbenzol wird unter Rühren während
2 Stunden auf 90-950 erwärmt. Anschliessend gibt man eine Lösung von 280 g Natriumacetat (krist.) in 500 cm3 Wasser zu. Es fällt ein Niederschlag aus, den man abfiltriert und mit Äthanol auskocht. Die heisse Äthanollösung wird filtriert. Beim Abkühlen des Filtrates scheidet sich der 2-Chlor-anthrazen-(9)-aldehyd in gelben Nadeln aus, die nach Umkristallisation aus Eisessig bei 148-1500 schmelzen.
Eine Lösung von 14,0 g 2-Chlor-anthrazen-(9)aldehyd in 200 cm3 Dimethylformamid sättigt man mit Äthylen und erhitzt sie anschliessend im Autoklav 24 Stunden auf 1700. Das Dimethylformamid wird hierauf im Vakuum abdestilliert. Es verbleibt ein zähes Ö1, das in 100 cm3 Athanol gelöst und nach Zugabe von 10 g Monomethylamin im Bombenrohr während 4 Stunden auf 900 erwärmt wird. Die Lösung dampft man anschliessend zur Trockne ein. Der Rückstand wird in 200 cm3 Äthanol gelöst und nach Zugabe von 2 g Raney-Nickel bei 5 atü hydriert. Nach dem Abfiltrieren des Katalysators wird das Reaktionsgemisch im Vakuum zur Trockne eingedampft. Den Rückstand kocht man mit 100 cm3 2n Salzsäure aus und trennt die salzsaure Lösung ab. Beim Abkühlen derselben scheidet sich ein Ö1 aus, das nach einigem Stehen kristallisiert.
Man erhält das 2-Chlor-9 -methylamino-methyl-9, 1 0-dihydro-
9,1 0-äthano-(1 ,2)-anthrazen-hydrochlorid in weissen Kristallen, die nach dem Kristallisieren aus Wasser bei 223-2260 schmelzen.
Beispiel 2
Auf analoge Weise wie im Beispiel 1 beschrieben kann man das 2-Chlor-9-diäthylaminomethyl-9, 1 0-dihydro-
9,10-äthano-(1,2)-anthrazen herstellen, dessen Hydrochlorid bei 2310 schmilzt.
