L being a residue which is eliminated during the reaction.
For example, the acid chloride, azide, ester or other derivatives can be used. The reaction is carried out under the usual conditions for converting a carboxylic acid derivative into the corresponding amide or hydrazide.
The products obtainable by the process according to the invention are particularly active anticonvulsants and medullary depressants; this latter property can be demonstrated in particular by the antagonistic action against strychnine [cf., for example, F. M. Berger - Pharmaceutical Reviews, 1, 274 (1949].
They also have fairly high doses of potentiation properties of narcosis. They can be used as non-hypnotic sedative agents, in particular in states of anxiety and nervous tension. They can also have a favorable effect in. the treatment of hypertension precisely because of their properties. sedatives.
The following examples show how the process of the invention can be put into practice; the indicated melting points were determined on the Kofler bench.
Example 1 51 g of ethyl (3-ethylindolyl-2) carboxylate and 120 cm3 of hydrazine hydrate are heated under reflux for 3 hours. Allowed to cool, the -solid is filtered off, washed with 4 times 125 water cure and dried under vacuum. 46.5 g of hydrazide die (3-ethylindolyl-2) carboxylic acid, melting at 183 °, are obtained.
Ethyl (3-ethyl indolyl-2) carboxylate [P. F. = 124.1] is obtained by the action of benzene diazonium chloride on an alcoholic potassium hydroxide solution of ethyl α-acetyl valerianate [Burton Ara. Chem. J. 3, 385 (1881)].
Example 2 A solution of azide d e (3-ethyl indolyl-2) carboxylic acid in ether is prepared by action, on 34 g of "hydrazide of (3-ethyl-2-indolyl) carboxylic acid (obtained as in Example 1) in 750 cm3 of dsoxane,
12 g of sodium nitrite dissolved in 150 cm3 of water then 20 cm3 of normal hydrochloric acid, keeping the temperature at around + 4o, followed by dilution with 1500 cc of water and dissolution of the solid in 300 cm3 of ether. A solution of 150 g of dimethyl-amine in 500 cc of ether is poured into the solution obtained with stirring, while maintaining the temperature at around 0 °.
Stirred for one hour and left to stand overnight at a temperature of 0. The ethereal solution is stirred with 300 cc of saturated aqueous solution of sodium bicarbonate, 300 cc of chloroform is added and the organic layer is separated, which is washed with water and dried over potassium carbonate. The solvent is evaporated off, the residue dissolved in 100 cc of boiling benzene, added 100 cc of hot normal heptane and allowed to crystallize. It is filtered off, washed with heptane and dried under vacuum.
In this way 22.5 g of (3-ethylindolyl-2) carboxylic acid dimethylamide is obtained, melting at 154 (1. Example 3 By operating as in Example 2, from 12 , 2 g of (3-ethyl-indolyl-2) carboxylic acid hydrazide and 50 g of monomethylamine, 8.5 g of (3-ethyl-2-indolyl) carboxylic acid monomethylamide melting at 186 are obtained. , 1. Example 4 In a solution of 13,
1 g of thionyl chloride in 50 cid of toluene is added, at room temperature, in small portions, 18.9 g of (3-ethyl indolyl-2) carboxylic acid. Stirring is continued at 250 for 30 minutes and then slowly heated to a temperature of 760. A clear solution is obtained which is cooled to +5. Then poured drop by drop, while keeping the temperature at +50 by cooling, 27 g of liquid dimethyl-amine.
We let it rest overnight in the cooler. The crystallized product is filtered off, washed first with toluene and then with plenty of water. Dried and thus obtained 16 g of (3-ethylindolyl-2) dimethylcarboxamide, melting at 152-153. By evaporation of the toluene solutions, 4 g of a second jet melting at 150-151 are obtained.
After recrystallization from benzene, 17 g of pure product, melting point 154, are obtained. Example 5 By operating as in Example 3, replacing the monomethylamine with 20 g of ammonia, 5.6 g of melting (3-ethyl-indolyl-2) carboxylic acid amide is obtained at 134, solidifying and then melting at 140.
