La présente invention a pour objet un procédé de préparation de nouveaux dérivés de la 7H-indolizino [5,6,7ij] isoquinoléine de formule générale:

Dans la formule générale (I),
R représente un atome d'hydrogène ou d'halogène, de préférence un atome de chlore, ou un radical alcoyloxyle dont la partie alcoyle contient 1 à 4 atomes de carbone et Rt représente un radical alcoyle contenant 1 à 4 atomes de carbone (substitué par un radical carboxy, alcoyloxycarbonyle dont la partie alcoyle contient 1 à 7 atomes de carbone ou carbamoyle) ou un radical phénylalcoyle (dont la partie alcoyle, qui contient 1 à 4 atomes de carbone, est substituée par un radical carboxy, alcoyloxycarbonyle dont la partie alcoyle contient 1 à 4 atomes de carbone ou carbamoyle).
Selon l'invention, les nouveaux produits de formule générale (I) peuvent être préparés par action d'un ester réactif de formule générale:
Rt-X (II) dans laquelle Rt est défini comme ci-dessus et X représente un reste d'ester réactif tel qu'un atome d'halogène, sur un sel d'ammonium quaternaire, tel que le sel de tétraéthylammonium, de l'oxime de formule générale:

Généralement, le sel de tétraéthylammonium de l'oxime de formule générale (III) est préparé in situ par action de l'hydroxyde de tétraéthylammonium sur l'oxime de formule générale (III) en opérant dans un solvant organique tel qu'un alcool, comme le méthanol ou l'éthanol, ou dans un mélange hydroalcoolique.
La condensâtion de l'ester réactif de formule générale (II) sur le sel de tétraéthylammonium de l'oxime de formule générale (III) s'effectue dans un solvant organique tel qu'un alcool, comme l'éthanol ou le méthanol, un ester, comme l'acétate d'éthyle, ou un amide, comme le diméthylformamide, et éventuellement en présence d'eau à une température comprise entre 0 et 300C.
L'oxime de formule générale (III) peut être obtenue par action de l'hydroxylamine sur une 7-indolizino [5,6,7-ij] isoquinoléinone de formule générale:

Généralement, la réaction s'effectue dans un solvant organique tel que la pyridine éventuellement en présence d'autres solvants tels que les alcools, de préférence l'éthanol, I'eau ou un mélange de ces solvants, à la température d'ébullition du mélange réactionnel. Il est particulièrement avantageux d'utiliser l'hydroxylamine sous forme d'un sel tel que le chlorhydrate.
La 7H-indolizino [5,6,7-ij] isoquinoléinone-7 de formule générale (IV) peut être obtenue par hydrolyse alcaline d'une imine de formule générale:

L'isoquinoléine de formule générale (X) peut être préparée soit par la méthode de Y. Ahmad et D. J. Hey, J. Chem. Soc., 3882 (1961) soit par la méthode de R. Manske et M. Kulka,
Can. J. Research, 27B, 169 (1949).
Les nouveaux produits obtenus selon les procédés de la présente invention peuvent éventuellement être purifiés par des méthodes physiques telles que la cristallisation ou la chromatographie.
Les produits de formule générale (I) dans laquelle Rt représente un radical alcoyle ou phénylalcoyle dans lesquels la portion alcoyle contient 1 à 4 atomes de carbone et porte un substituant alcoyloxycarbonyle dont la portion alcoyle est définie comme ci-avant peuvent être ensuite utilisés pour l'obtention des produits correspondants dans la formule des quels le radical alcoyle ou phénylalcoyle est substitué par un radical carboxy: on les soumet à une saponification, généralement en milieu alcalin en présence de soude ou de potasse et à une température comprise entre 0 et 300C.
Les nouveaux produits de formule générale (I) dans laquelle Rt représente un radical alcoyle substitué par un radical carboxy ou un radical phénylalcoyle dont la partie alcoyle est substituée par un radical carboxy peuvent être transformés en sels métalliques ou en sels d'addition avec les bases azotées par application de méthodes connues en soi. Ainsi, ces sels peuvent être préparés par action d'une base alcaline ou alcalinoterreuse, de l'ammoniaque ou d'une amine sur les produits de formule générale (I) dans un solvant approprié tel qu'un alcool, un éther, une cétone, l'eau; le sel formé précipite éventuellement après concentration de sa solution et il est séparé par filtration ou décantation.
Les nouveaux dérivés de la 7H-indolizino [5,6,7-ij] isoquinoléine selon la présente invention et leurs sels, lorsqu'ils existent, présentent des propriétés chimiothérapeutiques remarquables. Ils sont particulièrement actifs comme antibilharziens, anthelminthiques et antimicrobiens.
Chez la souris infestée par Schistosoma mansoni, les produits se montrent actifs à des doses comprises entre 250 et 1000 mg/kg par jour par voie orale et entre 125 et 250 mg/kg par voie sous-cutanée.
Chez le singe (Macaca mulatta [var. rhésus]) infesté à Schisosoma mansoni, l'activité des produits est particulièrement bonne à des doses comprises entre 30 et 80 mg/kg par voie orale.
L'activité anthelminthique se manifeste essentiellement sur les oxyures et les cestodes.
In vitro, les produits selon l'invention sont actifs sur les germes gram-positifs à des concentrations comprises entre 10 et 100 Ftg/ml.
D'un intérêt tout particulier sont les produits de formule générale:

dans laquelle Rt représente un radical alcoyle contenant 1 à 4 atomes de carbone substitué par un radical carboxy ou alcoyloxycarbonyle dont la partie alcoyle contient 1 à 7 atomes de carbone, ainsi que leurs sels.
Les exemples suivants, donnés à titre non limitatif, montrent comment l'invention peut être mise en pratique.
Exemple 1
A une suspension de 20 g d'hydroxyimino-7 7H-indolizino [5,6,7-ij] isoquinoléine dans 400 cm3 d'éthanol, on ajoute 75,8 cm3 d'une solution aqueuse à 17,5% d'hydroxyde de tétraéthylammonium. On élimine l'éthanol par évaporation à 400C sous pression réduite et on reprend le concentrat brun, huileux, par 240 cm3 de diméthylformamide. A la solution brune, agitée et refroidie vers 10oC, on ajoute, en 10 minutes, 15,03 g de bromacétate d'éthyle puis on maintient à une température voisine de 200C durant 16 heures. On ajoute alors 1440 cm3 d'eau glacée puis extrait le précipité avec, au total, un litre d'acétate d'éthyle. On lave la solution organique avec, au total, 600 cm3 d'eau distillée puis on la sèche sur du sulfate de magnésium anhydre.
On évapore le solvant sous pression réduite et on obtient ainsi 26,2 g d'éthoxycarbonylméthoxyimino-7 7H-indolizino [5,6,7-ij] isoquinoléine brute sous la forme d'un solide partiellement cristallisé.
Après purification par chromatographie sur une colonne de silicagel, on obtient 14,3 g d'éthoxycarbonylméthoxyimino-7 7H-indolizino [5,6,7-ijj isoquinoléine fondant à 1340C.
L'hydroxyimino-7 7H-indolizino [5,6,7-ijj isoquinoléine peut être préparée de la façon suivante:
On maintient à l'ébullition, sous agitation, une solution de 28 g de 7H-indolizino [5,6,7-ijj isoquinoléinone-7 et de 28 g de chlorhydrate d'hydroxylamine dans 560 cm3 de pyridine. La solution brune ainsi obtenue est versée dans 3,92 litres d'eau.
Après 3 heures d'agitation, les cristaux jaune-moutarde obtenus sont essorés, puis lavés abondamment avec de l'eau.
Après séchage, on obtient 27,2 g d'hydroxyimino-7 7H-indolizino [5,6,7-ij] isoquinoléine fondant à 2540C avec décomposition. Après recristallisation dans un mélange eau-pyridine (6-5 en volumes) le produit pur fond à 2580C avec décomposition.
La 7H-indolizino [5,6,7-ij] isoquinoléinone-7 utilisée comme produit de départ peut être préparée de la façon suivante:
On chauffe pendant 4 heures à reflux 110 g d'imino-7 7H-indolizino [5,6,7-ij] isoquinoléine et 11 cm3 de solution normale de soude dans 4,4 litres d'éthanol et 4,4 litres d'eau.
Après refroidissement, on obtient 108,1 g de 7H-indolizino [5,6,7-ij] isoquinoléinone-7 sous la forme d'aiguilles jaunes fondant à 2640C.
L'imino-7 7H-indolizino [5,6,7-ij] isoquinoléine peut être préparée de la façon suivante:
Une solution de 279 g de cyano-1 (pyrrolyl-1)-8 isoquinoléine dans 7 litres d'éther et 7 litres de chloroforme pur saturée d'acide chlorhydrique anhydre est maintenue à une température comprise entre 0 et 5cC durant 72 heures. Après extraction par 15 litres d'eau glacée puis alcalinisation de la solution acide rouge-sang ainsi obtenue, on obtient 223,4 g d'imino-7 7H-indolizino [5,6,7-ijj isoquinoléine sous la forme d'aiguilles jaune-vert fondant à 201oC avec décomposition.
La cyano-1 (pyrrolyl-1)-8 isoquinoléine peut être préparée de la façon suivante:
On maintient pendant 16 heures à une température voisine de 200C un mélange de 446,2 g de cyano-l (pyrrolyl-1)8 p.toluènesulfonyl-2 dihydro-1,2 isoquinoléine et de 68,7 g d'une suspension à 50% d'hydrure de sodium dans de l'huile minérale, dans 2232 cm3 de diméthylformamide. On obtlent ainsi 261 g de cyano-1 (pyrrolyl-1)-8 isoquinoléine brute fondant à 1780C.
La cyano-1 (pyrrolyl-1 )-8 p-toluènesulfonyl-2 dihydro1,2 isoquinoléine peut être préparée de la façon suivante:
293 g de (pyrrolyl-1)-8 isoquinoléine et 295 g de cyanure de potassium dissous dans un mélange de 880 cm3 de chlorure de méthylène et de 880 cm3 d'eau sont agités à une température comprise entre 0 et SOC. Après addition en 45 heures d'une solution de 575 g de p.toluènesulfochlorure dans 880 cm3 de chlorure de méthylène, décantation puis lavage de la solution organique avec de l'eau, on obtient un résidu solide brun.
Ce produit brut fournit, après recristallisation dans 750 cm3 d'acétonitrile, 407 g de cyano-1 (pyrrolyl-1)-8 p.toluènesulfonyl-2 dihydro-1,2 isoquinoléine fondant à 160OC.
Les eaux-mères, après lavage de la lessive de soude, fournissent encore 39,3 g de cyano-1 (pyrrolyl-1)-8 p.toluènesulfonyl-2 dohydro-1,2 isoquinoléine fondant à 160DC.
La (pyrrolyl-1)-8 isoquinoléine peut être préparée de la façon suivante:
A une solution bouillante de 245 g d'amino-8 isoquinoléine dans 1,25 litres d'acide acétique, on ajoute en 6 minutes 300 g de diéthoxy-2,5 tétrahydrofuranne. Après alcanisation par 2,2 litres de lessive de soude, on obtient 327 g d'un solide brun fondant à 900C. Ce produit brut est dissous dans un mélange de 1,55 litre d'acétate d'éthyle et de 1,55 litre de cyclohexane.
Après filtration sur une colonne contenant 3,3 kg d'alumine alcaline puis évaporation des solvants, on obtient 294 g de (pyrrolyl- 1)-S isoquinoléine fondant à 91 OC.
L'amino-8 isoquinoléine peut être préparée selon Y. Ahmad et D. J. Hey, J. Chem. Soc., 3882 (1961).
Exemple 2
En opérant comme à l'exemple 1 mais à partir de 23,5 g d'hydroxyimino-7 7H-indolizino [5,6,7-iji isoquinoléine, 90 cm3 de solution aqueuse à 17,5% d'hydroxyde de tétraéthylammonium et 23,4 g de bromo-2 isobutyrate de tertiobutyle dans 500 cm3 de diméthylformamide, on obtient 35 g de [(t-butoxycarbonyl-1 méthyl-1 éthoxy) imino]-7 7H-indolizino [5,6,7-ij] isoquinoléine. Après recristallisation dans 50 cm3 d'acétonitrile, on obtient 28,9 b de produit fon dant à 158t5C.
Exemple 3
A une suspension agitée de 16,2 g de chloro-3 hydroxy imino-7 7H-indolizino [5,6,7-ij] isoquinoléine dans 210 cm3 de diméthylformamide, on ajoute 50,5 cm3 d'une solution aqueuse à 17,5% d'hydroxyde de tétraéthylammonium. On refroidit vers 10oC la solution brune obtenue, puis ajoute en une seule fois 23,16 g de bromacétate d'éthyle. On abandonne le mélange réactionnel à une température voisine de 200C durant 4 heures puis ajoute 1,8 litre d'eau distillée. On sépare par filtration le précipité vert sombre apparu puis on le lava abondamment à l'eau. On reprend le produit brut humide par 2 litres de chlorure de méthylène puis l'on sèche la solution obtenue sur du sulfate de magnésium anhydre.
Après élimination du solvant sous pression réduite, on obtient 16 g de chloro-3 éthoxycarbonyl-méthoxyimino-7 7H-indolizino [5,6,7-ijj isoquinoléine brute fondant à 172OC. Après recristallisation dans 720 cm3 d'acétate d'éthyle, on obtient 11,36 g de produit pur fondant à 179OC.
Exemple 4
En opérant comme à l'exemple 3 mais à partir de 15 g d'hydroxyimino-7 7H-indolizino [5 ,6,7-ijj isoquinoléine, de 180 cm3 de diméthylformamide, de 55 cm3 de solution aqueuse à 17,5% d'hydroxyde de tétraéthylammonium et de 12,7 g de bromo-2 butyrate d'éthyle, on obtient 17,7 g d' (éthoxycarbonyl- 1 propoxyimino)-7 7H-indolizino [5,6,7-ij] isoquinoléine brute fondant à 148OC.
Après recristallisation dans 20 parties (v/p) d'acétate d'éthyle, le produit pur fond à 148OC.
Exemple S
En opérant comme à l'exemple 3 mais.à partir de 17 g d'hydroxyimino-7 7H-indolizino [5,6,7-ij] isoquinoléine, de 170 cm3 de diméthylformamide, de 63,8 cm3 de solution aqueuse à 17,5% d'hydroxyde de tétraéthylammonium et de 13,72 g de bromo-2 propionate d'éthyle, on obtient 17,55 g d'(éthoxycarbonyl-1 éthoxyimino)-7 7H-indolizino [5,6,7-ij] isoquinoléine brute fondant à 150OC. Après recristallisation dans 28 parties (v/p) d'un mélange d'acétate d'éthyle et de cyclohexane (1-1 en volumes), le produit pur fond à 155oC.
Exemple 6
En opérant comme à l'exemple 3 mais à partir de 20 g d'hydroxyimino-7 7H-indolizino [5 ,6,7-ij] isoquinoléine, de 255 cm3 de diméthylformamide, de 75,3 cm3 de solution aqueuse à 17,5% d'hydroxyde de tétraéthylammonium et de 23 g de bromo-2 phényl-3 propionate d'éthyle, on obtient 13,37 g d' (éthoxycarbonyl- 1 phényl-2 éthoxyimino)-7 7H-indolizino [5,6,7-ij] isoquinoléine brute fondant à 110oC.
Après recristallisation dans 3 parties (v/p) d'un mélange d'acétate d'éthyle et d'oxyde d'isopropyle (1-1 en volumes) le produit pur fond à 115"C.
Exemple 7
En opérant comme à l'exemple 3 mais à partir de 17 g d'hydroxyimino-7 7H-indolizino [5,6,7-ij] isoquinoléine, de 170 cm3 de diméthylformamide, de 63,8 cm3 de solution aqueuse à 17,5% d'hydroxyde de tétraéthylammonium et de 15,85 g de bromo-2 valérate d'éthyle, on obtient 17,5 g d'(éthoxycarbonyl-l butoxyimino)-7 7H-indolizino [5,6,7-ij] isoquinoléine brute fondant à 110OC. Après recristallisation dans 4 parties (v/p) d'un mélange d'oxyde d'isopropyle et d'acétate d'éthyle (9-1 en volumes), le produit pur fond à 110oC.
Exemple 8
En opérant comme à l'exemple 3 mais à partir de 15 g d'hydroxyimino-7 7H-indolizino [5,6,7-ij] isoquinoléine, de 180 cm3 de diméthylformamide, de 55 cm3 de solution aqueuse à 17,5% d'hydroxyde de tétraéthylammonium et de 12,7 g de bromo-2 isobutyrate d'éthyle, on obtient 18,77 g d'(éthoxycarbonyl-1 méthyl-1 éthoxyimino)-7 7H-indolizino [5,6,7-ij] isoquinoléine brute fondant entre 150 et 1550C. Après recristallisation dans un mélange d'acétate d'éthyle et de cyclohexane (1-1 en volumes), le produit pur fond à 154oC.
En opérant de la même manière avec les matières premières correspondantes on peut préparer:

Exemple 12 (a-carboxybenzyloxyimino)-7 7H-indolizino [5 ,6,7-ij] isoquinoléine, fondant à 200OC avec décomposition.
Exemple 13
Isopropyloxycarbonylméthoxyimino-7 7H-indolizino [5,6,7-ijj isoquinoléine qui, solvatée par l'acétate d'éthyle, fond à 700C.

Exemple 23
A une suspension agitée de 0,356 g de chloro-3 éthoxycarbonylméthoxyimino-7 7H-indolizino [5 ,6,7-ijj isoquinoléine dans 15 cm3 d'éthanol, on ajoute 1 cm3 de solution aqueuse normale de soude, puis 35 cm3 d'eau distillée. Après une heure d'agitation à une température voisine de 20OC, on obtient une solution vert-foncé que l'on neutralise par addition de 1 cm3 de solution aqueuse normale d'acide chlorhydrique.
On sépare par filtration le précipité apparu et le lave abondamment à l'eau puis le sèche sous pression réduite. On obtient ainsi 0,28 g de chloro-3 carboxyméthoxyimino-7 7Hindolizino [5,6,7-ijl isoquinoléine, fondant à 202oC avec décomposition.
Exemple 24
En opérant comme à l'exemple 23 mais à partir de 8,24 g d'(éthoxycarbonyl-1 propoxyimino)-7 7H-indolizino [5,6,7-ij] isoquinoléine, de 82,4 cm3 d'eau et de 25 cm3 de solution aqueuse normale de soude, on obtient 6,72 g de (car boxy- 1 propoxyimino)-7 7H-indolizino [5 ,6,7-ij] isoquinoléine, fondant à 1 80OC avec décomposition.
Exemple 25
En opérant comme à l'exemple 23 mais à partir de 18,77 g d' (éthoxycarbonyl- 1 méthyl-1 éthoxyimino)-7 7H-indolizino [5,6,7-ijj isoquinoléine brute, de 282 cm3 d'éthanol et de 55 cm3 de solution aqueuse normale de soude, on obtient 14,07 g du sel de sodium de la (carboxy-1 méthyl-1 éthoxyimino)-7 7H-indolizino [5,6,7-ijj isoquinoléine qui cristallise dans le milieu d'hydrolyse.
A une solution filtrée de 15,43 g de ce sel de sodium dans 308 cm3 d'eau distillée, on ajoute 154 cmi d'éthanol puis on acidifie par addition de 44.9 cm3 de solution aqueuse normale d'acide chlorhydrique.
L'acide qui précipite est séparé par filtration, lavé successivement avec un mélange éthanol-eau (66,34 en volumes), de l'éthanol absolu puis de l'éther. Après séchage sous pression réduite, on obtient 14,21 g de (carboxy-1 méthyl-1 éthoxyimino)-7 7H-indolizino [5,6,7-ij] isoquinoléine pure fondant à 2200C avec décomposition.
Exemple 26
En opérant comme à l'exemple 23, mais à partir de 16,48 g d'(éthoxycarbonyl-1 butoxyimino)-7 7H-indolizino [5,6,7-iji isoquinoléine, de 330 cm3 d'éthanol et de 50 cm3 de solution aqueuse normale de soude, on obtient 15,2 g du sel de sodium de (carboxy-1 butoxyimino)-7 7H-indolizino [5,6,7-ij] isoquinoléine.
A la solution filtrée de ce sel dans 304 cm3 d'eau distillée, on ajoute 152 cm3 d'éthanol puis on acidifie par addition de 42,5 cm3 de solution aqueuse normale d'acide chlorhydrique.
L'acide qui précipite est séparé par filtration, lavé successivement avec un mélange éthanol-eau (66-34 en volumes), de l'éthanol absolu puis de l'éther. Après séchage sous pression réduite, on obtient 12,42 g de (carboxy-1 butoxyimino)-7 7Hindolizino [5,6,7-ijj isoquinoléine pure fondant à 219oC avec décomposition.

In mice infested with Schistosoma mansoni, the products are active at doses of between 250 and 1000 mg / kg per day orally and between 125 and 250 mg / kg subcutaneously.
In the monkey (Macaca mulatta [var. Rhesus]) infested with Schisosoma mansoni, the activity of the products is particularly good at doses of between 30 and 80 mg / kg orally.
The anthelmintic activity manifests itself mainly on pinworms and cestodes.
In vitro, the products according to the invention are active on gram-positive bacteria at concentrations of between 10 and 100 Ftg / ml.

The following examples, given without limitation, show how the invention can be put into practice.
Example 1
To a suspension of 20 g of hydroxyimino-7 7H-indolizino [5,6,7-ij] isoquinoline in 400 cm3 of ethanol, is added 75.8 cm3 of a 17.5% aqueous solution of hydroxide of tetraethylammonium. The ethanol is removed by evaporation at 400C under reduced pressure and the brown, oily concentrate is taken up in 240 cm3 of dimethylformamide. The 7-hydroxyimino-7H-indolizino [5,6,7-ijj isoquinoline can be prepared as follows:
A solution of 28 g of 7H-indolizino [5,6,7-ijj-isoquinolinone-7 and 28 g of hydroxylamine hydrochloride in 560 cm3 of pyridine is kept at the boil, with stirring. The brown solution thus obtained is poured into 3.92 liters of water.
After 3 hours of stirring, the yellow-mustard crystals obtained are drained, then washed thoroughly with water.

The 7H-indolizino [5,6,7-ij] isoquinolinone-7 used as starting material can be prepared as follows:
110 g of imino-7 7H-indolizino [5,6,7-ij] isoquinoline and 11 cm3 of normal sodium hydroxide solution in 4.4 liters of ethanol and 4.4 liters of sodium hydroxide are heated for 4 hours at reflux. water.
After cooling, 108.1 g of 7H-indolizino [5,6,7-ij] isoquinolinone-7 are obtained in the form of yellow needles melting at 2640C.
Imino-7 7H-indolizino [5,6,7-ij] isoquinoline can be prepared as follows:
A solution of 279 g of cyano-1 (1-pyrrolyl) -8 isoquinoline in 7 liters of ether and 7 liters of pure chloroform saturated with anhydrous hydrochloric acid is maintained at a temperature between 0 and 5 ° C. for 72 hours. After extraction with 15 liters of ice-cold water and then alkalinization of the blood-red acid solution thus obtained, 223.4 g of imino-7 7H-indolizino [5,6,7-ijj isoquinoline in the form of needles are obtained. yellow-green melting at 201oC with decomposition.
Cyano-1 (1-pyrrolyl) -8 isoquinoline can be prepared as follows:
A mixture of 446.2 g of cyano-l (pyrrolyl-1) 8 p.toluenesulfonyl-2-dihydro-1,2-isoquinoline and 68.7 g of a suspension is maintained for 16 hours at a temperature of 200 ° C. 50% sodium hydride in mineral oil, in 2232 cm3 of dimethylformamide. In this way 261 g of crude cyano-1 (1-pyrrolyl) -8 isoquinoline is obtained, melting at 1780C.
Cyano-1 (pyrrolyl-1) -8 p-toluenesulfonyl-2 dihydro1,2 isoquinoline can be prepared as follows:
293 g of (1-pyrrolyl) -8 isoquinoline and 295 g of potassium cyanide dissolved in a mixture of 880 cm3 of methylene chloride and 880 cm3 of water are stirred at a temperature between 0 and SOC. After addition over 45 hours of a solution of 575 g of p.toluenesulfochloride in 880 cm3 of methylene chloride, decantation and then washing of the organic solution with water, a brown solid residue is obtained.
This crude product provides, after recrystallization from 750 cm3 of acetonitrile, 407 g of cyano-1 (pyrrolyl-1) -8 p.toluenesulfonyl-2-dihydro-1,2 isoquinoline, melting point 160OC.
The mother liquors, after washing the sodium hydroxide solution, still provide 39.3 g of cyano-1 (1-pyrrolyl) -8 p.toluenesulfonyl-2-dohydro-1,2 isoquinoline, melting at 160 ° C.
The (1-pyrrolyl) -8 isoquinoline can be prepared as follows:
300 g of 2,5-diethoxy tetrahydrofuran are added in 6 minutes to a boiling solution of 245 g of 8-amino isoquinoline in 1.25 liters of acetic acid. After alkanization with 2.2 liters of sodium hydroxide solution, 327 g of a brown solid are obtained, melting at 900C. This crude product is dissolved in a mixture of 1.55 liters of ethyl acetate and 1.55 liters of cyclohexane.
After filtration through a column containing 3.3 kg of alkaline alumina then evaporation of the solvents, 294 g of (pyrrolyl-1) -S isoquinoline, melting point at 91 ° C., are obtained.
The 8-amino isoquinoline can be prepared according to Y. Ahmad and D. J. Hey, J. Chem. Soc., 3882 (1961).
Example 2
By operating as in Example 1 but starting from 23.5 g of hydroxyimino-7 7H-indolizino [5,6,7-iji isoquinoline, 90 cm3 of 17.5% aqueous solution of tetraethylammonium hydroxide and 23.4 g of 2-bromo-butyl isobutyrate in 500 cm3 of dimethylformamide, 35 g of [(t-butoxycarbonyl-1 methyl-1 ethoxy) imino] -7 7H-indolizino [5,6,7-ij] are obtained. isoquinoline. Example 3
To a stirred suspension of 16.2 g of 3-chloro-3-hydroxyimino-7 7H-indolizino [5,6,7-ij] isoquinoline in 210 cm3 of dimethylformamide is added 50.5 cm3 of an aqueous solution at 17, 5% tetraethylammonium hydroxide. The brown solution obtained is cooled to around 10 ° C., then 23.16 g of ethyl bromoacetate is added all at once.
