Le procédé de préparation du composé de formule I est caractérisé en ce que l'on condense l'anhydride 4- nitro-homophtalique de formule II avec la 3,4-di- méthoxy-phénéthyl-amine, obtient le 2-carboxy-4-nitro- N[P-(3',4'-diméthoxy-phényl)-éthyl]-phénylacétamide de formule III, dont on transforme la fonction acide en ester méthylique de formule IV, soumet ce 2-carbo méthoxy-4-nitro-N-[3-(3',4'-diméthoxy-phényl)
-éthyl]- phénylacétamide à une réduction sélective de sa fonc tion ester à l'aide d'un borohydrure alcalin en présence d'un sel de lithium, traite le 2-hydroxyméthyl-4-nitro-N- [3-3',4'-diméthoxy-phényl)-éthyl]-phénylacétamide résul tant de formule V, par l'oxychlorure de phosphore, puis par un borohydrure alcalin, et obtient la 2,3-diméthoxy- 10-nitro-berbine de formule VI, que l'on réduit en com posé de formule I à l'aide de zinc ou de fer en milieu acide. Les réactions ci-dessus sont illustrées sur le dessin annexé.
Les différents stades du procédé de l'invention peu vent être effectués comme suit a) la condensation de l'anhydride 4-nitro-homo- phtalique de formule II avec la 3,4-diméthoxy-phénéthyl- amine est effectuée dans un hydrocarbure aromatique, tel que benzène ou toluène, au reflux du solvant; b) on transforme le 2-carboxy-4-nitro-N-[3-(3',4'-di- méthoxyphényl)-éthyl]-phénylacétamide de formule III en ester méthylique à l'aide du diazométhane ;
c) on transforme le 2-carboxy-4-nitro-N-[3-(3',4'- diméthoxyphényl)-éthyl]-phénylacétamide de formule III en ester méthylique IV à l'aide de méthanol en présence d'un catalyseur acide, comme l'acide sulfurique ; d) la réduction sélective de la fonction ester du 2 carbo-méthoxy-4-nitro-N-[3-(3',4'-diméthoxy-phényl)- éthyl]-phénylacétamide de formule IV est effectuée à l'aide de borohydrure de potassium en présence de chlorure de lithium, on opère dans le tétra-hydrofuran ;
e) la cyclisation du 2-hydroxyméthyl-4-nitro-N-[3- (3',4'-diméthoxy-phényl)-éthyl]-phénylacétamide de for mule V est effectuée par l'oxychlorure de phosphore dans un solvant organique, à la température du reflux et le traitement subséquent par le borohydrure de po tassium dans l'éthanol, aux environs de la température ambiante ; f) la réduction de la 2,3-diméthoxy-10-nitro-berbine de formule VI par le zinc est effectuée dans l'acide acétique au reflux.
L'exemple suivant illustre l'invention. <I>Exemple</I> Préparation de la 2,3-diméthoxy-10-amino-berbine de formule I Stade A : 2-carboxy-4-nitro-N-[3-(3',4'-diméthoxyphé- nyl)-éthyl]-phényl-acétamide de formule III.
On introduit 4,2 g d'anhydride 4-nitro-homophtalique de formule II, [produit décrit par Whitmore et Coll., J.A.C.S., 66, p. 1237 (1944)], dans 170 cm3 de benzène et chauffe au reflux jusqu'à dissolution.
On ajoute ensuite en l'espace de vingt minutes une solution de 4 g d'homovératrylamine (ou 3,4-diméthoxy- phénéthyl-amine) dans 10 cm3 de benzène et maintient le reflux pendant une heure après l'addition.
On refroidit, essore, lave au benzène, sèche à 801, C et obtient après recristallisation dans le méthanol, 5,46 g soit un rendement de 68 %, de 2-carboxy-4-nitro-N- [3-(3',4'-diméthoxy-phényl)-éthyl]-phénylacétamide de formule III, F. = 1620 C.
Le produit se présente sous forme d'aiguilles inco lores, insolubles dans l'eau, l'éther et le benzène, peu solubles dans l'alcool, solubles dans les alcalis dilués aqueux, l'acétone et le chloroforme.

On détruit l'excès de diazométhane par addition de 0,2 cm3 d'acide acétique, distille le chlorure de mé thylène et cristallise le résidu dans l'acétate d'éthyle.
On glace, essore, lave au mélange acétate d'éthyle- éther (4 : 6), sèche à 80o C et obtient 11,4 g de 2-carbo méthoxy-4-nitro-N-[3',4'-diméthoxy-phényl)-éthyl]-phé- nylacétamide de formule IV, F. = 126o C.
Le produit se présente sous forme d'aiguilles, in solubles dans l'eau, les alcalis dilués aqueux, l'éther, solubles dans l'alcool, l'acétone et le chloroforme.

On refroidit ensuite, glace et essore. On recristallise le produit obtenu dans 14 volumes d'acétate d'éthyle et obtient le 2-carbométhoxy-4-nitro-N[3-3',4'-dimétho- oxy-phényl)-éthyl]-phénylacétamide de formule IV, sous une deuxième forme cristalline, F. étant alors égal à 1380.
Stade C : 2-hydroxyméthyl-4-nitro-N-[3-3',4'-dimétho- oxy-phényl)-éthyl]-phénylacétamide de formule V.
On met en suspension, à la température ambiante, 0,3 g de borohydrure de potassium et 0,25 g de chlorure de lithium dans 10 cm3 de tétrahydrofuran anhydre, puis ajoute 0,5 g de 2-carbométhoxy-4-nitro-N-[3-(3', 4'-diméthoxy-phényl)-éthyl]-phénylacétamide de formule IV et agite pendant deux heures.
On verse ensuite sur la glace, extrait au chlorure de méthylène, récupère la phase organique qu'on lave à l'acide chlorhydrique N, puis à l'eau.
On sèche, élimine le chlorure de méthylène par dis tillation puis cristallise le résidu dans 5 cm3 du mé lange acétate d'éthyle-éther (4: 6) et obtient 0,23 g de 2-hydroxy-méthyl-4-nitro-N-[3-(3',4'-diméthoxy-phényl)- éthyl]-phénylacétamide de formule VII, F. = 1250 C ou 146-148 C suivant la forme cristalline obtenue.
Le produit se présente sous forme de cristaux cou leur brique.

Stade D : 2,3-diméthoxy-10-nitro-berbine de formule VI. <I>a) Cyclisation</I> On introduit une partie de 2-hydroxyméthyl-4-nitro- N-[3-(3',4'-diméthoxy-phényl)-éthyl]-phényl-acétamide de formule VII, dans un mélange composé de 20 volu mes de benzène et 2 volumes d'oxychlorure de phos phore et chauffe au reflux le mélange réactionnel pen dant deux heures.
On verse ensuite sur la glace, laisse en contact pen dant une heure et extrait au chlorure de méthylène.
On recueille la phase organique, lave à l'eau, sèche, distille à sec sous vide et obtient une résine.
<I>b) Réduction</I> On dissout la résine obtenue dans 10 volumes d'al cool à 85%, ajoute une partie en poids, par rapport à la résine, de borohydrure de potassium et agite le mé lange réactionnel à la température ambiante pendant trois heures.
On dilue par addition de 50 parties d'eau et extrait au chlorure de méthylène. On recueille la phase orga nique, lave à l'eau, sèche, distille à sec sous vide et obtient la 2,3-diméthoxy-10-nitro-berbine de formule VIII, qui est utilisée telle quelle pour le stade suivant de la synthèse.
Ce composé n'est pas décrit dans la littérature. Stade E : 2,3-diméthoxy-10-amino-berbine de formule I.
On introduit une partie de 2,3-diméthoxy-10-nitro- berbine de formule VI dans 10 volumes d'acide acétique, ajoute 3 parties de poudre de zinc et chauffe au reflux le mélange réactionnel pendant un quart d'heure. On refroidit ensuite à température ambiante, essore, lave à l'acide acétique, dilue le filtrat avec 50 parties d'eau et neutralise par addition d'ammoniaque.
On extrait au chlorure de méthylène, recueille la phase organique, lave à l'eau, sèche et distille à sec sous vide.
On cristallise le résidu dans 10 volumes du mélange acétate d'éthyle-éther (4 : 6) et obtient la 2,3-diméthoxy- 10-amino-berbine de formule I, F. = 1660 C.
Le produit se présente sous forme de cristaux peu solubles dans l'éther, solubles dans l'alcool, l'acétone et le chloroforme.
