Procédé de préparation de dérivés du succinimide La présente invention a pour objet un procédé de préparation de dérivés du succinimide contenant un noyau dibenzocycloheptadiène ou -triène, les deux étant reliés l'un à l'autre de manière à former un système cyclique spiro;
ces nouveaux composés sont le spiro [dibenzo [a,d] [1,4] cycloheptadiène-5, cc-succinimide] et le spiro [dibenzo [a,e] cycloheptatriène-5, a-succinimide] et peuvent être représentés par la formule:

Ces nouveaux dérivés du succinimide se distinguent par une forte action anticonvulsive associée à une faible toxicité.
Ainsi, administrés par voie orale à des animaux d'expérience, ils inhibent très efficacement les effets de l'électrochoc. Les doses auxquelles ils exercent cet effet sont bien inférieures à celles provoquant des mani festations toxiques. Ces composés ont pour propriété surprenante et inattendue une action s'opposant très nettement aux effets convulsivants du métrazole. Ceci est surprenant car suivant Miller et Long [J. Amer, Chem. Soc., 73, 4895 (1951)], l' a, a-diphényl-succinimide. qui est un composé assez voisin, est pratiquement dépourvu d'activité antimétrazolique.
Grâce à ces propriétés biologiques, ces nouveaux dérivés du succinimide sont utiles comme agents théra- peutiques pour le traitement de divers états convulsifs. Ils peuvent être administrés aux patients par voie orale sous des formes pharmaceutiques telles que comprimés ou capsules, contenant l'ingrédient actif en quantité de 100 à 400 mg avec des excipients, par exemple du lactose, de l'amidon, du stéarate de magnésium, etc, en dose quotidienne totale de 100 à 1000 mg.
Suivant l'invention, on prépare tout d'abord le 5-carboxyméthyl-5-cyano-dibenzo [a,d] cycloheptadiène ou le dibenzo [a,e] cycloheptatriène correspondant par condensation du 5-cyano-dibenzo [a,d] [1,4] cyclohepta- diène ou du 5-cyano-dibenzo [a,e] cycloheptatriène avec un a-halo-acétate d'alcoyle inférieur (XCH2C00-alcoyle)
sous l'influence d'un agent de condensation alcalin (M+) et dans un solvant organique inerte. Pratiquement, il est préférable d'utiliser le chloroacétate d'éthyle comme ester. Des agents de condensation alcalins utilisables sont les amidures ou les alcoylates inférieurs de métaux alcalins, comme par exemple l'amidure de sodium ou de lithium ou l'éthylate de sodium, mais, grâce à sa forte réactivité et sa facilité de maniement, la dispersion d'hydrure de sodium dans une huile minérale, que l'on trouve dans le commerce, est préférée.
Comme solvant pour cette réaction, le toluène s'est avéré très satisfaisant, et la réaction peut être réalisée complètement en opérant au point d'ébullition de ce solvant. Comme autres solvants utilisables, on peut citer les hydrocarbures analogues, comme par exemple le benzène et le xylène, ou les alcanols inférieurs.
Les produits immédiats de la réaction, c'est-à-dire les esters éthyliques du 5-carboxyméthyl-5-cyano-dibenzo [a,d] [1,4] cycloheptadiène ou du dibenzo [a,e] cyclo- heptatriène correspondant, sont obtenus sous forme d'huiles que l'on hydrolyse en conditions ménagées, obtenant ainsi le 5-carboxyméthyl-5-cyano-dibenzo [a,d] [l,4] cycloheptadiène ou le dibenzo [a,e] cycloheptatriène correspondant,
par exemple par action d'un hydroxyde de métal alcalin (MOH) dissous dans de l'éthanol aqueux. Le produit, qui est soluble dans les alcalis, peut être facilement séparé des impuretés neutres présentes dans le mélange initial.
Enfin, on traite le 5-carboxyméthyl-5-cyano-dibenzo [a,d] [1,4] cycloheptadiène ou le dibenzo [a,e] cyclo- heptatriène correspondant avec de l'acide chlorhydrique concentré bouillant. Dans des conditions similaires, C. A. Miller et L. M.
Long (10c. cit) ont obtenu de l'acide oc, oc-diphényl-succinique à partir de l'acide P-cyano-P, P-diphényl-propionique au lieu du succinimide corres pondant, c'est-à-dire que le groupe cyano a été hydrolysé en groupe carboxyle.
Contrairement à toute- attente, le traitement du 5- carboxyméthyl-5-cyano-dibenzo [a,d] [1,4] cyclohepta- diène ou du dibenzo [a,e] cycloheptatriène correspondant par l'acide chlorhydrique ne conduit pas à l'acide suc cinique correspondant comme produit principal. En lieu et place, le succinimide désiré est obtenu directement à l'état brut.
Il est facile de séparer ce dernier des sous- produits indésirables en suspendant le produit de réaction brut dans une solution de bicarbonate de sodium diluée, dans laquelle le produit désiré est insoluble. Par recristal- lisation de cette matière insoluble dans un solvant approprié, on obtient le succinimide purifié.
Le 5-cyano-dibenzo [a,d] [1,4] cycloheptadiène de départ peut être préparé facilement par l'action du cyanure d'argent sur le 5-chloro-dibenzo [a,d] [1,4] cycloheptadiène dans de l'acétonitrile ou du benzène anhydre bouillant, comme décrit dans l'article de Martin A. Davis et collab., publié dans Journal of Medicinal Chemistry, Vol. 6, p.<B>251</B> (1963).
Le dibenzo [a,e] cyclo- heptatriène correspondant peut être préparé de manière semblable comme décrit par M. A. Davis et collab. dans J. Med. Chem., Vol. 7, p. 88 (1964).

<I>Exemple</I> A. On ajoute 2,44 g (0,055 mole) d'une dispersion huileuse d'hydrure de sodium à 53,8 % à 50 ml de toluène sec, puis on ajoute 11,0 g (0,05 mole) de 5-cyano- dibenzo [a,d] [1,4] cycloheptadiène dans 50 ml de toluène. On chauffe le mélange à reflux pendant 2 h sous agitation pour compléter la formation de sel de sodium.
On refroidit le mélange à la température ordinaire, on lui ajoute goutte à goutte une solution de 9,2 g (0,075 mole) de chloracétate d'éthyle dans 20 ml de toluène et on le chauffe à nouveau à reflux pendant 21/2 h.
On filtre le mélange au moyen de Celite (terre de diatomées) et on lave le filtrat à l'eau, on le sèche et on l'évapore, obtenant ainsi 13,3 g d'une huile que l'on n'a pas pu solidifier. Ensuite, on chauffe cette huile à reflux pendant 15 min avec 8,0 g d'hydroxyde de potassium dans de l'éthanol aqueux à 50 %. On chasse l'alcool sous vide et, après dilution à l'eau, on extrait la couche aqueuse à l'éther et on l'acidule. Il se forme un précipité, que l'on recristallise dans de l'éthanol aqueux,
obtenant ainsi le 5-carboxy- méthyl-5-cyano-dibenzo [a,d] [1,4] cycloheptadiène; p.f. 173-174 C.
B. On chauffe à reflux pendant 1 h une solution de 55,0 g (0,25 mole) de 5-cyano-dibenzo [a,d] [1,4] cyclo- heptadiène et d'éthylate de sodium [formé à partir de 8,1 g (0,35 atome-gramme) de sodium] dans 400 ml d'éthanol absolu; une couleur verte se développe. On refroidit la solution à la température ordinaire et on lui ajoute goutte à goutte 58,5 g (0,35 mole) de bromacétate d'éthyle. Ensuite, on chauffe le mélange à reflux pendant 3 h, on le filtre et on l'évapore.
On hydrolyse l'ester ainsi obtenu en le chauffant à reflux pendant 15 min avec 20 g d'hydroxyde de potassium dans 300 ml d'un mélange éthanol-eau 1:1. On chasse la majeure partie de l'éthanol sous vide, on dilue la solution avec de l'eau et on l'extrait à l'éther. Par acidification de la couche alcaline, on obtient le cyano-acide, p.f. 171-174 C. Un échantillon analytique a présenté un p.± de 173-175 C (après recristallisation dans de l'éthanol aqueux ou de l'acétate d'éthyle).
L'analyse a confirmé la formule brute Cl$H1sN02.

De cette manière, on obtient le nouveau composé chimique désiré, à savoir le spiro [dibenzo [a,d] [1,4] cycloheptadiène-5, a-succini- mide], sous forme de courtes aiguilles blanches; p.f. 218-219 C.
L'analyse a confirmé la formule brute Cl$H1bN02.

Process for preparing succinimide derivatives The present invention relates to a process for preparing succinimide derivatives containing a dibenzocycloheptadiene or -triene ring, the two being linked to each other so as to form a spiro ring system;
these new compounds are spiro [dibenzo [a, d] [1,4] cycloheptadiene-5, cc-succinimide] and spiro [dibenzo [a, e] cycloheptatriene-5, a-succinimide] and may be represented by the formula:

These new succinimide derivatives are distinguished by a strong anticonvulsant action associated with low toxicity.
Thus, administered orally to experimental animals, they very effectively inhibit the effects of electroshock. The doses at which they exert this effect are much lower than those causing toxic manifestations. These compounds have the surprising and unexpected property of an action very clearly opposed to the convulsant effects of metrazole. This is surprising because according to Miller and Long [J. Amer, Chem. Soc., 73, 4895 (1951)], α, α-diphenyl-succinimide. which is a fairly related compound, is practically devoid of antimetrazole activity.
Owing to these biological properties, these new succinimide derivatives are useful as therapeutic agents for the treatment of various convulsive states. They can be administered to patients orally in pharmaceutical forms such as tablets or capsules, containing the active ingredient in an amount of 100 to 400 mg with excipients, for example lactose, starch, magnesium stearate, etc., in a total daily dose of 100 to 1000 mg.
According to the invention, the 5-carboxymethyl-5-cyano-dibenzo [a, d] cycloheptadiene or the corresponding dibenzo [a, e] cycloheptatriene is first of all prepared by condensation of 5-cyano-dibenzo [a, d] [1,4] cycloheptadiene or 5-cyano-dibenzo [a, e] cycloheptatriene with lower alkyl α-halo-acetate (XCH2C00-alkyl)
under the influence of an alkaline condensing agent (M +) and in an inert organic solvent. Practically, it is preferable to use ethyl chloroacetate as the ester. Alkaline condensing agents which can be used are amides or lower alkylates of alkali metals, such as, for example, sodium or lithium amide or sodium ethoxide, but, thanks to its high reactivity and ease of handling, the dispersion of sodium hydride in mineral oil, which is commercially available, is preferred.
As the solvent for this reaction, toluene has been found to be very satisfactory, and the reaction can be carried out completely by operating at the boiling point of this solvent. As other solvents which can be used, mention may be made of analogous hydrocarbons, such as, for example, benzene and xylene, or lower alkanols.
The immediate products of the reaction, that is to say the ethyl esters of 5-carboxymethyl-5-cyano-dibenzo [a, d] [1,4] cycloheptadiene or of the corresponding dibenzo [a, e] cycloheptatriene , are obtained in the form of oils which are hydrolyzed under controlled conditions, thus obtaining 5-carboxymethyl-5-cyano-dibenzo [a, d] [l, 4] cycloheptadiene or the corresponding dibenzo [a, e] cycloheptatriene ,
for example by the action of an alkali metal hydroxide (MOH) dissolved in aqueous ethanol. The product, which is soluble in alkalis, can be easily separated from the neutral impurities present in the initial mixture.
Finally, the 5-carboxymethyl-5-cyano-dibenzo [a, d] [1,4] cycloheptadiene or the corresponding dibenzo [a, e] cycloheptatriene is treated with boiling concentrated hydrochloric acid. Under similar conditions, C. A. Miller and L. M.
Long (10c. Cit) obtained α, α-diphenyl-succinic acid from P-cyano-P, P-diphenyl-propionic acid instead of the corresponding succinimide, i.e. that the cyano group has been hydrolyzed to a carboxyl group.
Contrary to all- expectations, the treatment of 5-carboxymethyl-5-cyano-dibenzo [a, d] [1,4] cycloheptadiene or of the corresponding dibenzo [a, e] cycloheptatriene with hydrochloric acid does not lead to the corresponding kinic acid juice as the main product. Instead, the desired succinimide is obtained directly in the crude state.
The latter is easily separated from unwanted by-products by suspending the crude reaction product in dilute sodium bicarbonate solution, in which the desired product is insoluble. By recrystallizing this insoluble material from a suitable solvent, the purified succinimide is obtained.
The starting 5-cyano-dibenzo [a, d] [1,4] cycloheptadiene can be easily prepared by the action of silver cyanide on 5-chloro-dibenzo [a, d] [1,4] cycloheptadiene in acetonitrile or boiling dry benzene, as described in the article by Martin A. Davis et al., published in Journal of Medicinal Chemistry, Vol. 6, p. <B> 251 </B> (1963).
The corresponding dibenzo [a, e] cycloheptatriene can be prepared in a similar manner as described by M. A. Davis et al. in J. Med. Chem., Vol. 7, p. 88 (1964).

<I> Example </I> A. 2.44 g (0.055 mole) of an oily dispersion of sodium hydride at 53.8% is added to 50 ml of dry toluene, then 11.0 g ( 0.05 mol) of 5-cyano-dibenzo [a, d] [1,4] cycloheptadiene in 50 ml of toluene. The mixture is heated at reflux for 2 h with stirring to complete the formation of the sodium salt.
The mixture is cooled to room temperature, a solution of 9.2 g (0.075 mole) of ethyl chloroacetate in 20 ml of toluene is added dropwise thereto and again heated under reflux for 21/2 h.
The mixture is filtered through Celite (diatomaceous earth) and the filtrate is washed with water, dried and evaporated, thus obtaining 13.3 g of an oil which is not available. could solidify. Then, this oil is refluxed for 15 min with 8.0 g of potassium hydroxide in 50% aqueous ethanol. The alcohol is removed in vacuo and, after dilution with water, the aqueous layer is extracted with ether and acidified. A precipitate forms, which is recrystallized from aqueous ethanol,
thus obtaining 5-carboxymethyl-5-cyano-dibenzo [a, d] [1,4] cycloheptadiene; m.p. 173-174 C.
B. A solution of 55.0 g (0.25 mol) of 5-cyano-dibenzo [a, d] [1,4] cycloheptadiene and sodium ethoxide [formed in from 8.1 g (0.35 gram atom) of sodium] in 400 ml of absolute ethanol; a green color develops. The solution was cooled to room temperature and 58.5 g (0.35 mole) of ethyl bromoacetate was added dropwise thereto. Then, the mixture is heated under reflux for 3 h, filtered and evaporated.
The ester thus obtained is hydrolyzed by heating it under reflux for 15 min with 20 g of potassium hydroxide in 300 ml of a 1: 1 ethanol-water mixture. Most of the ethanol is removed in vacuo, the solution is diluted with water and extracted with ether. By acidification of the alkaline layer, the cyanoacid is obtained, m.p. 171-174 C. An analytical sample showed a mp of 173-175 C (after recrystallization from aqueous ethanol or ethyl acetate).
Analysis confirmed the gross formula Cl $ H1sN02.
