Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Herstellung von Hydrophenanthrylcar bonsäuren mit der nachstehenden Struktur des Ringes C

wobei n die Zahl 2 oder 3 bedeutet, mit basischen Mitteln behandelt. Diese Behandlung, vorzugsweise mit Alkalien, wie Natriumhydroxyd Kaliumhydroxyd, wird vorteilhaft bei erhöhter Temperatur und unter Druck, in Gegenwart von Lösungsmitteln, wie Alkoholen, durchgeführt.
Die als Ausgangsstoffe verwendeten Steroidlactame können weiter substituiert sein, z. B. durch freie oder funktionell abgewandelte, d. h. durch veresterte oder verätherte bzw. enolisierte oder ketalisierte Oxyoder Oxogruppen, insbesondere in 3-Stellung. Ferner können sie Doppelbindungen aufweisen, z. B. ausgehend vom Kohlenstoffatom 5, d. h. in 4,5- oder 5,6-Stellung. Zu ihrer Herstellung kann man entsprechend substituierte 11, 17-Dioxo- bzw. 11, 17a-Dioxo- testan-bzw.-a-Androstanverbindungen in die 17bzw. 17a-Monoxime überführen und die letzteren einer Beckmannschen Umlagerung unterwerfen.
Die Verfahrensprodukte sollen als Zwischenprodukte dienen. So können sie z. B. zur Herstellung von 18-oxigenierten Pregnanen Verwendung finden.
In den nachfolgenden Beispielen besteht zwischen Gewichtsteil und Volumteil die gleiche Beziehung wie zwischen Gramm und Kubikzentimeter, Die Temperaturen sind in Celsiusgraden angegeben.
Beispiel 1 1 Gewichtsteil 3a-Acetoxy-11, 17-diketo-17a-aza D-homo-testan wird in 150 Volumteilen einer 10prozentigen alkoholischen Kalilauge 5 Stunden im Druckgefäss auf 150 erhitzt. Anschliessend wird die Lösung mit 2-n. Salzsäure angesäuert (PIr 3-4) und im Vakuum weitgehend eingeengt. Nach dem Vermischen mit Natriumsulfat wird das Reaktionsprodukt mit Chloroform extrahiert. Der Chloroformextrakt liefert 0,95 Gewichtsteile eines Rohprodukts, das im UV eine charakteristische Absorption bei 238 m, (log E = 4,05) aufweist.
Die Auftrennung dieses Rohproduktes in Säuren und Neutralkörper liefert 0,9 Gewichtsteile der reinen d2-2, 13-Dimethyl-4-keto-7a-oxy- dodecahydro-phenanthryl- (1)-propionsäure, die im UV bei 238 m, (log e = 4,05) ein Absorptionsmaximum aufweist. Die Säure ist nur schwierig in kristallisierter Form zu erhalten. Nach dem Trocknen im Hochvakuum schmilzt die amorphe Masse bei 80 bis 95 .
Durch Veresterung der obigen Säure mit ätheri- scher Diazomethanlösung erhält man den A2-2, 13- Dimethyl-4-keto-7a-oxy-dodecahydrophenanthryl- (l) propionsäuremethylester.
Der obige Ausgangsstoff lässt sich wie folgt gewinnen : 1 Gewichtsteil 3a-Acetoxy-11, 17-diketo-testan vom F. = 61 wird in 20 Volumteilen einer Lösung von Hydroxylamin-acetat in Äthanol (bereitet durch kurzes Aufkochen von 1 Gewichtsteil Hydroxylaminhydrochlorid, 2 Gewichtsteilen Natriumacetat und 20 Volumteilen Äthanol und anschliessende Filtration) 2 Stunden zum Sieden erhitzt. Anschliessend wird die Lösung auf die Hälfte ihres Volumens eingedampft, mit 50 Volumteilen Wasser versetzt, worauf sich das Oxim vorerst amorph, dann in kristallisierter Form ausscheidet. Der Niederschlag wird abgenutscht, mit Wasser-Methanol 2 : 1 gewaschen und durch Abdampfen mit Benzol im Vakuum getrocknet.
Ausbeute : 1, 03 Gewichtsteile eines Rohprodukts vom F. = 188 .
Aus Aceton-Hexan erhält man das 3a-Acetoxy-11, 17diketo-testan-17-monoxim in gut ausgebildeten Nadeln vom F. = 194 .
1 Gewichtsteil 3a-Acetoxy-11, 17-diketo-testan
17-monoxim werden in 40 Volumteilen absolutem Pyridin gelöst, mit 1 Gewichtsteil p-Acetamino-ben zolsulfonsäure-chlorid versetzt und 2 Stunden auf 70 erwärmt. Dann wird das Reaktionsgemisch mit 120 Volumteilen Wasser versetzt und weitere 2 Stunden auf dem Wasserbad erwärmt. Durch Zugabe von Wasser wird das Gesamtvolumen auf 240 Volumteile gebracht und unter Kühlung mit 2-n. Schwefelsäure angesäuert. Die Extraktion mit Äther-Chloroform (3 : 1) liefert 1 Gewichtsteil eines kristallisierten Rohprodukts, das bei 292 schmilzt.
Durch Umkristallisieren des 3a-Acetoxy-11, 17-diketo-17a-aza-D-homotestans aus Methanol erhält man Nadeln, die bei 311 schmelzen. Das IR-Absorptionsspektrum der Verbindung zeigt in dem Gebiet von etwa 1700 cm-1 charakteristische Banden bei 1733,1706,1678,1631 und 1603 cl-1.
Beispiel 2
2 Gewichtsteile Lactam des Aaa-l#- (#'-Carboxy- äthyl)-2a-amino-2ss, 4bss-dimethyl-4-oxo-7, 7-äthylen- dioxy-l, 2,3,4,4aa, 4b, 5,6,7,8,10,1 Oass-dodecahydrophenanthrens in 50 Volumteilen 18 /oiger butanolischer Natronlauge werden nach Zugabe von 5 Volumteilen Wasser 4 Stunden unter Stickstoff am Rückfluss erhitzt. Nach dem Verdünnen der Reaktionslösung mit 300 Volumteilen Wasser werden die Neutralteile mit einem Gemisch von Äther-Chloroform (3 : 1) extrahiert. Die wässrige Schicht stellt man mit 10 /oiger Phosphorsäure auf ein pH von 6,5 ein und extrahiert mit Chloroform.
Der neutral gewaschene Extrakt hin terlässt nach dem Eindampfen 1,57 Gewichtsteile saure Anteile, aus welchen sich das reine Z12S ; 8^-lss- (ss'-carboxyäthyl)-2, 4bss-dimethyl-4-oxo-7, 7-äthylen- dioxy-l, 4,4aa, 4b, 5,6,7,8,10,10ass-decahydrophenanthren durch Kristallisation aus Aceton-Pentan gewinnen lässt. Die Verbindung zeigt im UV das für ein a, ss-ungesättigtes Keton charakteristische Maximum bei 236 m, logos = 4,07. IR-Banden bei 1709 cm-1 (Carboxyl) und bei 1650 cm-1 (ass-ungesättigtes Keton).
Der Ausgangsstoff kann folgendermassen hergestellt werden :
15 Gewichtsteile ex 4-3, 11,17-Trioxo-androsten vom F. 223 werden zusammen mit 0,24 Gewichtsteilen p-Toluolsulfosäure-monohydrat und 250 Volumteilen reinem, sorgfältig fraktioniertem 2-Methyl 2-äthyl-1, 3-dioxolan in einem Destillationskolben, versehen mit einer Rasching-Ring-Kolonne, so zum Sieden erhitzt, dass innerhalb von 51/2 Stunden 150 Volumteile Destillat übergehen. Die kalte Lösung ver dünnt man mit Benzol und wäscht die organische Phase dreimal mit gesättigter Natriumhydrogencarbo nat-Lösung und dreimal mit Wasser.
Nach dem Verdampfen des getrockneten Lösungsmittels können 11,2 Gewichtsteile reines 3-3, 3-Athylendioxy-11, 17dioxo-androsten durch Umkristallisation aus Benzol Hexan gewonnen werden. Im UV zeigt diese Verbindung keine charakteristische Absorption in der 240m, u-Region. IR-Banden bei 1749 cm-1 (5-Ring-Keton) und bei 1715 cm-t (6-Ring-Keton).
10 Gewichtsteile Hydroxylaminhydrochlorid und 20 Gewichtsteile wasserfreies Natriumacetat werden in 100 Volumteilen Äthanol aufgekocht, die Lösung heiss vom gebildeten Natriumchlorid abfiltriert und mit 100 Volumteilen heissem Äthanol nachgewaschen.
In die klare Lösung gibt man 10 Gewichtsteile/)"- 3,3-j2ithylendioxy 11, 17-dioxo-androsten und kocht 2 Stunden unter Rückfluss. Anschliessend wird im Vakuum auf die Hälfte eingeengt und das Oxim durch sorgfältiges Zutropfen von Wasser als kristalline Masse ausgefällt. Nach dreimaligem Umkristallisieren aus Aceton-Hexan werden 8,69 Gewichtsteile reines Oxim erhalten.
Die erhaltene Verbindung lässt sich im Ringe D wie folgt umlagern :
10 Gewichtsteile 17-Oxim des S 3-3, 3-Athylen- dioxy-11, 17-dioxo-androstens werden in 250 Gewichtsteilen kollidinfreiem Pyridin gelöst und nach Zugabe von 7,50 Gewichtsteilen p-Acetamino-benzolsulfochlorid während 2 Tagen bei Zimmertemperatur stehengelassen. Das mit 800 Volumteilen verdünnte Reaktionsgemisch bewahrt man 2 weitere Tage bei 20 auf. Die Lösung wird mit Schwefelsäure (1 : 1) angesäuert und durch Extraktion mit Chloroform in üblicher Weise aufgearbeitet.
Das rohe Lactam (9,87 Gewichtsteile) liefert nach Umkristallisation und Entfärbung mit Aktivkohle das reine Lactam des j 8a-I #- (ss'-carboxyäthyl)-2a-amino-2ss, 4bss-dimethyl-4-oxo- 7,7-äthylen-dioxy-1,2,3,4,4aa, 4bop, 5,6,7,8,10,10aSdodecahydrophenanthrens. IR-Banden bei 3185 cm-t ( > NH) ; bei 1678 und 1631 cm-1 (6-Ring-Lactam) und bei 1706 cm-1 (6-Ring-Keton).

where n is the number 2 or 3, treated with basic agents. This treatment, preferably with alkalis such as sodium hydroxide and potassium hydroxide, is advantageously carried out at elevated temperature and under pressure, in the presence of solvents such as alcohols.
The steroid lactams used as starting materials can be further substituted, e.g. B. by free or functionally modified, d. H. by esterified or etherified or enolized or ketalized oxy or oxo groups, especially in the 3-position. They can also have double bonds, e.g. B. starting from carbon atom 5, d. H. in 4,5 or 5,6 position. For their preparation, correspondingly substituted 11, 17-dioxo or 11, 17a-dioxotestane or -a-androstane compounds can be converted into the 17 or. Transfer 17a-monoxime and subject the latter to a Beckmann rearrangement.
The process products are intended to serve as intermediate products. So you can z. B. for the production of 18-oxygenated pregnanes use.

Example 1 1 part by weight of 3a-acetoxy-11,17-diketo-17a-aza D-homo-testane is heated to 150 in 150 parts by volume of a 10 percent alcoholic potassium hydroxide solution in a pressure vessel for 5 hours. Then the solution with 2-n. Hydrochloric acid acidified (PIr 3-4) and largely concentrated in vacuo. After mixing with sodium sulfate, the reaction product is extracted with chloroform. The chloroform extract provides 0.95 parts by weight of a crude product which has a characteristic UV absorption at 238 m (log E = 4.05).
The separation of this crude product into acids and neutral bodies yields 0.9 parts by weight of the pure d2-2, 13-dimethyl-4-keto-7a-oxydodecahydro-phenanthryl- (1) -propionic acid, which in the UV at 238 m, (log e = 4.05) has an absorption maximum. The acid is difficult to obtain in crystallized form. After drying in a high vacuum, the amorphous mass melts at 80 to 95.
Esterification of the above acid with an ethereal diazomethane solution gives the A2-2, 13-dimethyl-4-keto-7a-oxy-dodecahydrophenanthryl- (1) propionic acid methyl ester.
The above starting material can be obtained as follows: 1 part by weight of 3a-acetoxy-11, 17-diketo-testan from F. = 61 is added to 20 parts by volume of a solution of hydroxylamine acetate in ethanol (prepared by briefly boiling 1 part by weight of hydroxylamine hydrochloride, 2 Parts by weight of sodium acetate and 20 parts by volume of ethanol and subsequent filtration) heated to boiling for 2 hours. The solution is then evaporated to half its volume, mixed with 50 parts by volume of water, whereupon the oxime is initially amorphous and then precipitated in crystallized form. The precipitate is filtered off with suction, washed with water-methanol 2: 1 and dried by evaporation with benzene in vacuo.
Yield: 1.03 parts by weight of a crude product from F. = 188.
The 3a-acetoxy-11, 17diketo-testane-17-monoxime is obtained from acetone-hexane in well-formed needles of F. = 194.
1 part by weight of 3a-acetoxy-11,17-diketo-testane
17-monoxime is dissolved in 40 parts by volume of absolute pyridine, 1 part by weight of p-acetaminobenzene sulfonic acid chloride is added and the mixture is heated to 70 for 2 hours. Then the reaction mixture is mixed with 120 parts by volume of water and heated on the water bath for a further 2 hours. By adding water, the total volume is brought to 240 parts by volume and, while cooling, with 2-n. Acidified sulfuric acid. Extraction with ether-chloroform (3: 1) gives 1 part by weight of a crystallized crude product which melts at 292.
By recrystallizing the 3a-acetoxy-11, 17-diketo-17a-aza-D-homotestane from methanol, needles which melt at 311 are obtained. The IR absorption spectrum of the compound shows characteristic bands in the region of about 1700 cm-1 at 1733, 1706, 1678, 1631 and 1603 cl-1.
Example 2
2 parts by weight of lactam des Aaa-l # - (# '- carboxy-ethyl) -2a-amino-2ss, 4bss-dimethyl-4-oxo-7, 7-ethylene-dioxy-l, 2,3,4,4aa, 4b, 5,6,7,8,10,1 Oass-dodecahydrophenanthrens in 50 parts by volume of 18% butanolic sodium hydroxide solution are refluxed under nitrogen for 4 hours after addition of 5 parts by volume of water. After diluting the reaction solution with 300 parts by volume of water, the neutral parts are extracted with a mixture of ether-chloroform (3: 1). The aqueous layer is adjusted to pH 6.5 with 10% phosphoric acid and extracted with chloroform.
The neutral washed extract leaves 1.57 parts by weight of acidic components after evaporation, from which the pure Z12S; 8 ^ -lss- (ss'-carboxyethyl) -2, 4bss-dimethyl-4-oxo-7, 7-ethylene-dioxy-l, 4,4aa, 4b, 5,6,7,8,10,10ass- decahydrophenanthrene can be obtained by crystallization from acetone-pentane. In the UV, the compound shows the maximum characteristic of an α, ß-unsaturated ketone at 236 m, logos = 4.07. IR bands at 1709 cm-1 (carboxyl) and at 1650 cm-1 (ass-unsaturated ketone).
The starting material can be produced as follows:
15 parts by weight of ex 4-3, 11,17-trioxo-androstene from F. 223 together with 0.24 parts by weight of p-toluenesulfonic acid monohydrate and 250 parts by volume of pure, carefully fractionated 2-methyl-2-ethyl-1, 3-dioxolane in a distillation flask equipped with a Rasching ring column, heated to boiling in such a way that 150 parts by volume of distillate pass over within 51/2 hours. The cold solution is diluted ver with benzene and the organic phase is washed three times with saturated sodium hydrogen carbonate solution and three times with water.
After the dried solvent has evaporated, 11.2 parts by weight of pure 3-3, 3-ethylenedioxy-11, 17dioxo-androstene can be obtained by recrystallization from benzene-hexane. In the UV this compound shows no characteristic absorption in the 240m, u region. IR bands at 1749 cm-1 (5-ring ketone) and at 1715 cm-t (6-ring ketone).
10 parts by weight of hydroxylamine hydrochloride and 20 parts by weight of anhydrous sodium acetate are boiled in 100 parts by volume of ethanol, the hot solution is filtered off from the sodium chloride formed and washed with 100 parts by volume of hot ethanol.
10 parts by weight of /) "-3,3-ethylenedioxy 11,17-dioxo-androstene are added to the clear solution and refluxed for 2 hours. Subsequently, it is concentrated to half in vacuo and the oxime is obtained as a crystalline mass by carefully adding dropwise water After three recrystallizations from acetone-hexane, 8.69 parts by weight of pure oxime are obtained.
The connection obtained can be rearranged in ring D as follows:
