Verfahren zur Herstellung von Methylenhydrophenanthrenen
Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Herstellung von 2-Methylen-hydrophenanthrenen, das dadurch gekennzeichnet ist, dass man auf 2-Methyl-hydrophenanthrene welche in 2-Stellung durch eine freie oder substituierte Oxyoder Aminogruppe oder durch eine Ammoniumgruppe substituiert sind, Mittel einwirken lässt, die die genannte Gruppe unter Bildung einer exocyclischen Doppelbindung abspalten.
2-Methyl-hydrophenanthren-2-ole werden vorteilhaft nach Umwandlung in Sulfonsäureester, zum Beispiel Methan-oder p-Toluolsulfonsäureester, mit basischen Verbindungen, wie tertiären Aminen, zum Beispiel Collidin, Pyridin oder Triäthylamin, behandelt oder durch Einwirkung anorganischer Säure- chloride, wie Phosphoroxychlorid oder Thionylchlorid, insbesondere in Gegenwart von Pyridin, dehydratisiert. Besonders einheitlich verläuft die Abspaltung bei Aminen und quaternären Ammoniumbasen unter der Einwirkung von Alkalien, zum Beispiel Alkalihydroxyden, und/oder bei erhöhter Temperatur.
Als Ausgangsstoffe verwendet man besonders im Ringsystem gesättigte oder ungesättigte Verbindungen der folgenden Formel

Die im Ringsystem ungesättigten Verbindungen können eine oder mehrere Doppelbindungen aufweisen, vorzugsweise eine vom Kohlenstoffatom 8a ausgehende Doppelbindung.
Diese Verbindungen ergeben bei der erfindungsgemässen Behandlung im Ringsystem gesättigte oder ungesättigte 2-Methylen-hydrophenanthrene der Formel

welche als Zwischenprodukte zur Herstellung von in 18-Stellung substituierten Steroiden, insbesondere des hochwirksamen Nebennierenrindenhormons Aldosteron, dienen können.
Die Ausgangsstoffe lassen sich nach bekannten Methoden, insbesondere durch Persäureoxydation oder durch Beckmannsche Urnlagerung der Oxime aus 17-Oxo-steroiden bzw. 17e0xo-D-homostero- iden herstellen. Aus den Oximen von 17-Oxosteroiden oder 17a-Oxo-D-homosteroiden erhältman zum Beispiel Lactame der Formel II bzw.

Die Verlängerung der Seitenkette am Kohlenstoffatom 1, zum Beispiel nach der Methode von Arndt und Eistert,
sowie Umwandlungen im Ring A lassen sich sowohl vor als auch nach der Einfüh- rung der Methylendoppelbindung durchführen. So kann zum Beispiel eine z18^-Oxy-Gruppierung in eine d8-7-Oxo-Gruppierung umgewandelt werden und/oder letztere kann in eine ds-7-Athylen-dioxy- Gruppierung übergeführt werden.
Aus der Verbindung V erhält man über das entsprechende Säurechlorid und durch Umsetzung mit Dimethylcadmium das Methylketon IX, welches für die Herstellung 18-oxigenierter Steroide besonders geeignet ist. Aus den den Verbindungen VI und V entsprechenden Nitrilen gelangt man durch Umsetzung mit Methylmagnesiumhalogeniden ebenfalls zu Methylketonen der Formel IX.
Durch Epoxylierung der exocyclischen Doppelbindung und Aufspaltung des entstandenen Epoxyds mit einer Halogenwasserstoffsäure sind Halogenhydrine zum Beispiel der Teilformel

zugänglich, welche, gegebenenfalls vor oder nach Oxydation der Oxymethylgruppe zur Carboxylgruppe und Veresterung, durch intramolekulare Alkylierung zu in 18-Stellung eine Sauerstoffunktion aufweisenden Steroiden cyclisiert werden können. Falls diese Verbindungen in 11-Stellung keine Hydroxylgruppe aufweisen, kann diese zum Beispiel auf mikrobiologischem Wege eingeführt und mit der Carboxylgruppe laktonisiert werden.
Man erhält auf diese Weise zum Beispiel das (18-bll)-Lakton der ?4-3, 20-Dioxo-11/-oxy-pregnen-18-säure, welches ein bekanntes Zwischenprodukt zur Synthese von Aldosteron darstellt.
Beispiel 1
400 mg dsa-1 3- (3'-Carboxy-äthyl)-2a-amino- 2, 4b¯ - dimethyl-1, 2, 3, 4, 4aa, 4b, 5, 6, 7, 8, 10, 1OajB-do- decahydrophenanthren-7¯-ol werden in 10 cm3 Methanol gelöst und durch Zugabe der berechneten Menge methanolischer Kalilauge neutralisiert. Das Lösungsmittel wird im Vakuum abgedampft und das Kaliumsalz im Hochvakuum gut getrocknet. Das in 10 cm3 absolutem Methanol gelöste Kaliumsalz versetzt man mit 50 g Methyljodid und erhitzt das Gemisch 3 Tage am Rückfluss. Während dieser Zeit wird unter ständiger Kontrolle der Wert des PH durch kleine Zugaben von 0, 5 n methanolischer Kalilauge (insgesamt 10 cm3) über 7 gehalten. Am Ende der Reaktion wird die nun schwach saure Lösung im Vakuum zur Trockne eingedampft.
Den gelben, öligen Rückstand nimmt man in Chloroform auf und filtriert vom ungelösten Kaliumjodid ab.
Das rohe quaternäre Ammoniumjodid (655 mg) wird in 10 cm3 Athylenglylcol und 5 cm3 Wasser gelöst und nach Zusatz von 2, 5 g Kaliumhydroxyd 5 Stunden unter Stickstoff auf 150 bis 170 erhitzt.
Das Einsetzen der Reaktion kann durch die Entwicklung von Trimethylamin erkannt werden. Die kalte Lösung wird darauf durch Extraktion mit Chloroform aufgearbeitet. Aus dem sauren Anteil lassen sich 191 mg kristallisiertes dSa-lfl- (#'-Carb- oxy-äthyl)-2-methylen-4b,-methyl-1, 2, 3, 4, 4aa, 4b, 5, 6, 7, 8, 10, 10α¯ - dodecahydrophenanthren -7¯-ol vom F = 184 bis 185 gewinnen. Die Verbindung kristallisiert aus Methylenchlorid-Aceton, [a] 2D =-83 (c = 0, 829 in Athanol).
IR-Absorptionsspektrum (in Nujol) : Banden bei 3300 cm-1 ( > CH-OH), 1692 cm-1 (Carboxyl) und bei 1648 cm-1 und 895 cm-I ( > C=CH2).
Der Methylester des @8a-1¯-(¯'-CarboxyÏthyl)- 2-methylen-4b-methyl-1, 2, 3, 4, 4aa, 4b, 5, 6, 7, 8, 10, 10a¯-dodecahydrophenanthren-7¯-ols schmilzt bei 76 bis 77 , [α] 20 =:-85¯ (c = 0, 80 in Chloroform).
Bei der Verseifung dieses Esters mit 4prozentiger methanolischer Kalilauge kann in quantitativer Ausbeute die Methylensäure vom F. = 185 zurückerhalten werden.
Das aus dem Methylester durch Dehydrierung der Hydroxylgruppe in 7, B-Stellung zur Oxogruppe unter gleichzeitiger Wanderung der Doppelbindung von der 8a, 9- in die 8, 8a-Stellung erhältliche ?8
1¯-(¯'-Carbomethoxy-Ïthyl)-2-methylen-4b¯ -methyl 1, 2, 3, 4, 4aa, 4b, 5, 6, 7, 9, 10, 10a¯ - dodecahydrophenanthren-7-on weist im UV-Absorptionsspektrum ein Maximum bei 240 m, (log E = 4, 2) auf, und die entsprechende freie Säure kristallisiert aus Aceton Isopropyläther in Prismen vom Smp. 166 ; UV-Absorptionsmaximum in Feinsprit bei 242 niu. (log @ = 4, 26).
Beispiel 2
In einem Kolben, versehen mit Vibromischer und einem Rückflusskühler, werden 1, 11 g Natriumsalz des @8a-1¯-(γ'-Carboxypropyl)-2α-amino-2¯,4b¯- dimethyl-7, 7-äthylendioxy-1, 2, 3, 4aaq, 4b, 5, 6, 7, 8, 10, 10a¯-dodecahydrophenanthrens in 50 cm3 absolutem Methanol gelöst und zusammen mit 7 g wasserfreiem Kaliumcarbonat und 100 g Methyljodid 70 Stunden unter Feuchtigkeitsausschluss gekocht. Die Lösungs- mittel werden im Vakuum bei Zimmertemperatur entfernt.
Den weissen, kristallinen Rückstand löst man in einem Gemisch von 20 cm3 Athylenglykol und 10 cm3 Wasser und erhitzt das Gemisch nach Zugabe von 5 g Kaliumhydroxyd in einem offenen Kolben langsam auf 150 , wobei Trimethylamin und Wasser entweichen. Nach 1/2 Stunde wird ein Rückflusskühler aufgesetzt und unter Einleiten von Stickstoff die Badtemperatur weitere 4 Stunden bei 150 bis 170 gehalten. Dann giesst man die Lösung auf Eis, trennt neutrale Anteile mit Chloroform ab und extrahiert die wässerige Phase nach Zusetzen von 2n-Phosphorsäure (bis PH 7-8) mit Chloroform.
Der neutral gewaschene Extrakt hinterlässt nach dem Eindampfen 578 mg kristallines Produkt.
Das reine @8A-1¯- (y-Carboxy-propyl)-2-methylen- 4b¯-methyl-7, 7-äthylendioxy-1, 2, 3, 4, 4aα, 4b, 5, 6, 7, 8, 10, 10ass-dodecahydrophenanthren kristallisiert aus Aceton-Isopropyläther in Nadeln vom F. = 148 bis 149 , [α]20 =-45¯ (c = 0, 935 in Athanol).
Die entsprechende Verbindung mit freier 7-Oxogruppe, das ds-lfl- (y'-Carboxypropyl)-2-methylen- 1, 2, 3, 4, 4aa, 4b, 5, 6, 7, 9, 10, 10a¯- dodecahydrophenan thren-7-on, weist folgende Eigenschaften auf : UV : @max = 242m, u (log e = 4, 17) ; IR-Banden bei 896 ; 1647 cm-i ( > C=CH2) ; 1622 & 1676 cm-1 (α,¯-ungesÏttigtes Keton) ; 1737 cm-i (Esterbande).
Der in diesem Beispiel verwendete Ausgangsstoff lässt sich zum Beispiel wie folgt herstellen :
In eine konzentrierte Lösung von Hydroxylamin-acetat in Athanol werden 1, 63 g?5-3¯-Acetoxy17a-oxo-D-homo-androsten eingetragen. Durch weitere Zugaben von Athanol in der Wärme wird eine klare Lösung erhalten, die 4 Stunden am Rückfluss erhitzt wird. Etwa die Hälfte des Lösungsmittels wird abgedampft und das Oxim durch Zufügen von heissem Wasser ausgefällt. Der kristalline Niederschlag wird auf der Nutsche mit Wasser gewaschen.
Aus Methanol kristallisiert das Oxim (1, 28 g) in gut ausgebildeten Prismen vom F. = 211 bis 213 , =-213 (c = 1, 045 in Chloroform).
4 g Oxim werden in 40 cm3 absolutem collidinfreiem Pyridin unter gelindem Erwärmen gelöst und mit 2 g p-Acetamino-benzolsulfochlorid versetzt.
Die sich dunkel färbende Lösung wird während 2 Tagen bei Zimmertemperatur unter Feuchtigkeitsausschluss stehengelassen. Die Reaktionslösung wird in die Sfache Menge Eiswasser gegossen, 2 weitere Tage bei 20 aufbewahrt, mit verdünnter Schwefelsäure (1 : 1) angesäuert und durch Extraktion mit Chloroform aufgearbeitet. Das gelbe Rohprodukt wird mit Aktivkohle weitgehend entfärbt und aus Methanol-Wasser umkristallisiert. Das in glänzen- den Schuppen anfallende Gemisch (3 g) der zwei isomeren Lactame schmilzt nach dreimaligem Uml¯sen bei 224 bis 225¯, [α]22 = -178¯ (c = 0,957 in Chloroform).
Das reine Lactam des ?8a-1¯-(γ'- Carboxypropyl)-2a-amino-2ss-4bss-dirnethyl-1, 2, 3, 4, 4aa 4b, 5, 6, 7, 8, 10, 10ajss-dodecahydrophenanthren-7jss- ols, das durch alkalische Verseifung des Lactam Gemisches, Auftrennung des Hydrolysates in neutrale und saure Anteile und thermische Recyclisation der sauren Anteile leicht erhalten werden kann, schmilzt bei 261", [a] D =-156". IR-Banden bei 3250 ; 3190 cm-1 (-OH, > NH) ; bei 1632 cm-1 (Lactam).
6, 53 g Lactam des d8a-l- (y'-Carboxypropyl)- 2a-amino-2, 8, 4bp-dimethyl-1, 2, 3, 4, 4aa, 4b, 5, 6, 7, 8, 10, lOa, B-dodecahydrophenanthrens werden in 300 cm3 absolutem Benzol und 250 cm3 absolutem Aceton in der Wärme gelöst und nach Zugabe von 35 g Aluminium-tert.-butylat während 22 Stunden unter Feuchtigkeitsausschluss am Rückfluss gekocht. Das Reaktionsgemisch wird in Eiswasser gegossen und mit 2n-Schwofelsäure angesäuert. Die wässerige Schicht schüttelt man mit Chloroform aus. Die vereinigten organischen Phasen werden mit Wasser neutral gewaschen, getrocknet und eingedampft.
Der kristalline Rückstand wird an 200 g Aluminiumoxyd (Akt. II) adsorbiert. Die Benzol-und Benzol Ather (l : l)-Fraktionen liefern das Lactam des @8-
1¯-(γ'-Carboxy-propyl)-2α-amino-2¯,4b¯- dimethyl- 1, 2, 3, 4, 4aα, 4b, 5, 6, 7, 9, 10, 1 Oass-dodecahydrophenan- thren-7-ons, das nach Umlösen aus Aceton und Sublimation im Hochvakuum bei 180 bei 266 bis 267 schmilzt ; [a] D =-3 (c = 1, 044 in Chloroform).
Aus einer Lösung von 1, 4 g des Keto-lactams vom F. = 266 bis 267 in 40 cm3 absolutem Benzol und 2 cm3 trockenem Äthylenglykol werden letzte Spuren von Wasser durch Abdampfen von etwas Benzol entfernt. Nach Zugabe von 15 mg p-Toluolsulfosäuremonohydrat wird das Reaktionsgemisch 16 Stunden am Rückfluss gekocht unter Abscheidung des dabei gebildeten Wassers. Anschliessend wird die Lösung mit gesättigter Natriumhydrogencarbonat Lösung alkalisch gemacht und durch Extraktion mit Benzol aufgearbeitet. Das rohe, farblose Rohprodukt kristallisiert aus Methanol-Isopropyläther in Nadeln (1, 25 g).
Nach dreimaligem Umkristallisieren schmilzt das reine Lactam des @8a-1¯-(γ-Carboxy- propyl)-2, 4bss-dimethyl-7, 7 - Ïthylendioxy-1, 2, 3, 4, 4aa, 4b, 5, 6, 7, 8, 10, 10aai-dodecahydrophen- anthren bei 266 ; [α]20 = -93¯ (c = 0, 996 in Chloroform). Im W zeigt es keine Absorption.
1, 64 g des ¯thylenketal- lactams vom F. = 266 in 40 cm3 18prozentiger butanolischer Natronlauge werden nach Zugabe von 2 cm3 Wasser 4 Stunden unter Stickstoff am Rückfluss erhitzt. Mit Ather und Chloroform werden neutrale Anteile vom kalten Reaktionsgemisch abgetrennt. Mit In-Salzsäure stellt man unter gutem Durchmischen das PH der eisge- kühlten wässerigen Lösung auf 9 ein. Im Wasserstrahlvakuum wird diese Lösung auf 150 cm3 ein geengt, wobei sich eine dicke Kristallmasse ausscheidet. Das Kristallisat wird abgenutscht und in 150 cm3 0, 05 n Natronlauge gelöst.
Nach dem Filtrieren engt man die klare Lösung auf 60 cm3 ein, fällt das Natriumsalz des d$a-1- (y'-Carboxy- propyl)-2a-amino-2, 4a-dimethyl-7, 7-äthylendioxy- 1, 2, 3, 4, 4aα, 4b, 5, 6, 7, 8, 10, 10a¯-dodecahydrophenanthrens durch Zugabe von Natriumchlorid aus und lässt zur Vervollständigung der Kristallisation im Kühlschrank stehen. Zur Reinigung nimmt man das Natriumsalz in Methanol auf, filtriert von ungelösten Bestandteilen ab, dampft zur Trockne ein und erhält 1, 33 g vom F. = 328 bis 330 ; [a] 20 = -13 (c = 0, 921 in O, ln-Natronlauge).
Beispiel 3
105 g Lactam des ?8a-1¯-(¯'-Carboxy-Ïthyl) 2a-amino-2ss, 4bss-dimethyl-7, 7-äthylendioxy-1, 2, 3, 4, 4aa, 4b, 5, 6, 7, 8, 10, 10ass-dodecahydrophenanthrens werden in einem Gemisch von 2 1 Butanol und 200 cm3 Wasser gelöst und mit 400 g festem Natriumhydroxyd versetzt. Nach gutem Schütteln kocht man das Gemisch während 5 Stunden unter R ckfluss, giesst nach dem Erkalten auf 4 Liter Eiswasser und extrahiert die Suspension kontinuierlich im Kutscher-Steudel-Apparat mit Ather. Die wässerige Suspension wird unter gutem Rühren mit konzentrierter Salzsäure sorgfältig auf pH 10 eingestellt, mit Kochsalz gesättigt und nach einigem Stehen filtriert.
Das so gewonnene Natriumsalz der Aminosäure löst man in 1 Liter heissem Wasser, filtriert von ungelösten Bestandteilen ab und sättigt die Lösung mit Kochsalz, wobei das Natriumsalz des -l- (-Carboxy-äthyl)- 2a-amino-2jss, 4b-dime- thyl-7, 7-äthylendioxy-1, 2, 3, 4, 4aa, 4b, 5, 6, 7, 8, 10, 10α¯- dodecahydrophenanthrens in feinen Nadeln ausfällt.
Das am Niederschlag haftende Wasser wird azeotrop mit Benzol durch Kochen während 12 Stunden am Wasserabscheider abgetrennt. Neben 82, 3 g Natriumsalz der Aminosäure, welches nach dreimaligem Umkristallisieren aus Methanol bei 301 bis 304 schmilzt, können aus dem Atherextrakt 19, 65 g unverändertes Ausgangsmaterial gewonnen werden.
35 g Natriumsalz des ?8a-1¯-(¯'-Carboxy-Ïthyl) 2a-amino-2ss, 4b/3-dimethyl-7, 7-Ïthylendioxy-1, 2, 3, 4, 4aa, 4b, 5, 6, 7, 8, 10, 10ass-dodecahydrophenanthrens vom F. 301 bis 304 werden zusammen mit 400 cm3 absolutem Methanol, 450 g Methyljodid und 100 g wasserfreiem Kaliumcarbonat unter heftigem Mischen mit einem Vibromischer während 2 Tagen unter Rückfluss gekocht. Nach 24 Stunden werden noch weitere 100 g Methyljodid zugesetzt. Das nicht umgesetzte Methyljodid destilliert man ab und filtriert das Reaktionsgemisch durch Watte und wäscht mit absolutem Methanol gut nach. Im Vakuum wird bis zur Trockne eingedampft.
Der Rückstand wird in 600 cm3 Athylenglykol gelöst und mit einer Lösung von 150 g Kaliumhydroxyd in 150 cm3 Wasser versetzt. Unter Stickstoff wird vorerst 1 Stunde auf 135 erhitzt, wobei ein Teil des Wassers zusammen mit Trimethylamin entweicht. Anschliessend setzt man einen Rückflusskühler auf und hält während 31/2 Stunden bei 175 . Das Reaktionsgemisch giesst man nach dem Erkalten auf Eis und entfernt neutrale Anteile mit Methylenchlorid. Die wässerige, stark alkalische Lösung wird unter gutem Rühren mit 5-n. Phosphorsäure auf ein pH von 6, 5 bis 7, 0 eingestellt und mit Methylenchlorid extrahiert.
Aus der gut gewaschenen organischen Phase lassen sich 23 g saure Anteile gewinnen, die nach Kristalli- sation aus Aceton-Isopropyläther 20, 6 g ?8a-1¯-(¯' Carboxy-äthyl)-2-methylen-4bss-methyl-7, 7-äthylen- dioxy-1, 2, 3, 4, 4aa, 4b, 5, 6, 7, 8, 10, 10a¯-dodecahydrophenanthren liefern. Die reine Verbindung schmilzt nach Umkristallisieren bei 121 . [a] 20 =-66 (c = 0, 970 in Chloroform). IR Banden bei 896, 1647 cm-l ( > =CH2) und bei 1708 cmI (COOH).
Das als Ausgangsstoff verwendete Lactam des ?8a-1¯'-(¯'-Carboxy-Ïthyl)-2α-amino - 2¯,4b¯ - dime- thyl-7, 7-äthylendioxy-1, 2, 3, 4, 4aa, 4b, 5, 6, 7, 8, 10, 10a¯dodecahydrophenanthrens lässt sich wie folgt herstellen :
100 g Lactam des ?8a-1¯-(¯'-Carboxy-Ïthyl)-2α- amino-2¯, 4b¯-dimethyl-1, 2, 3, 4, 4aa, 4b, 5, 6, 7, 8, 10, 10a,-dodecahydrophenanthren-7ss-ols werden zusammen mit 7, 5 g wasserfreiem Natriumacetat in 1, 66 Liter Eisessig gelöst. Innerhalb von 30 Minuten wird unter kräftigem Rühren eine Lösung von 52, 9 g Brom (1 Aquivalent) in 750 cm3 Eisessig bei 10 bis 15 zugetropft.
Anschliessend fügt man der schwach gelben Lösung 33, 1 g Chromtrioxyd, in 400 cm3 90prozentiger Essigsäure gelöst, zu und lässt nach gutem Umschütteln 12 Stunden bei Zimmertemperatur stehen. Dberschüssige Chromsäure zersetzt man mit 100 cm3 Methanol. Nach 1 Stunde wird das Gemisch bei 90 unter heftigem Turbinieren mit 309 g Zinkstaub versetzt und während 30 Minuten auf dieser Temperatur gehalten. Das noch warme Reaktionsgemisch filtriert man und wäscht den Zinkstaub mit viel Methylenchlorid. Das Filtrat wird mit 6 Liter Wasser verdünnt und wiederholt mit Methylenchlorid extrahiert, welches man der Reihe nach mit 2-n. Salzsäure, eiskalter 2-n. Natronlauge und Wasser wäscht.

After 1/2 hour a reflux condenser is attached and the bath temperature is maintained at 150 to 170 for a further 4 hours while passing in nitrogen.
