Verfahren zur Herstellung von 20-Monoenolacylaten von 11,20-Diketosteroiden Die Erfindung betrifft ein Verfahren zur Herstel lung von 20-Monoenolacylaten von 11,20-Diketo- steroiden durch Umsetzung der Steroide mit einem Carbonsäureanhydrid in Gegenwart eines organi schen Lösungsmittels und einer Säure.
Insbesondere betrifft die Erfindung ein Verfahren zur Herstellung von 20-Monoenolacylaten von 11,20- Diketosteroiden durch Umsetzung der Steroide mit einem Carbonsäureanhydrid in Gegenwart eines organischen Lösungsmittels und einer gegebenenfalls substituierten Benzolsulfonsäure, welche eine bis drei Nitrogruppen am Benzolkern aufweist.
Eine Anzahl wichtiger Corticosteroide ist in 17a- Stellung durch eine Hydroxylgruppe substituiert. Bei der Synthese der fraglichen Steroide wird diese Gruppe im allgemeinen in. 11,20-Diketosteroide ein geführt, welche ihrerseits als Zwischenprodukte beim Abbau von Gallensäuren oder bestimmten Sapogeni- nen anfallen.
Kritchevsky et a1., J. Am. Chem. Soc., 74, 483 (1956) beschrieben eine derartige Umwandlung, bei der das 3a-Hydroxy-11,20-diketopregnan in das 11,20-Dienolacetat mittels Essigsäureanhydrid um gewandelt wird, worauf man dieses mit Perbenzoe- säure zur 11,20 - Oxydoverbindung oxydiert und schliesslich diese Verbindung zum gewünschten 3a,17a-Dihydroxy-11,20-diketopregnan verseift.
Die gleichen Forscher haben auch festgestellt, dass die Doppelbindung zwischen den Kohlenstoffatomen 9 und 11 im 11,20-Dienolacetat durch die Persäure nicht angegriffen wird und infolgedessen die Carbo- nylgruppe in Stellung 11 sich bei der Verseifung wieder zurückbildet.
Es wurde jedoch festgestellt, dass bei der Oxyda tion der 11,20-Dienolacylate von Steroiden der Allo- Reihe (A/B trans) die Doppelbindung zwischen den Kohlenstoffatomen 9 und 11 oxydiert wird und hier bei eine Mischung der 9,11-Oxydo-, 17,20-Oxydo- und der 9,11-17,20-Dioxydoverbindung entsteht.
Um nun das von Kritchevsky et a1. beschriebene Verfahren auf die Umwandlung von 11,20-Diketo- steroiden der Allo-Reihe übertragen zu können, ist es wesentlich, dass man in dieser Reihe von einem 20-Monoenolacylat ausgeht.
Nach dem in der holländischen Patentschrift Nr. 88994 beschriebenen Verfahren lässt sich die Acylierung eines 20-Monoenols von 11,20-Diketo- steroiden durch Acylierung des Steroids in Gegen wart von Perchlorsäure durchführen.
Ein Nachteil bei diesem Verfahren ist der, dass infolge der oxydierenden Eigenschaften der verwen deten Perchlorsäure die Pregnanseitenketten der 20-Ketosteroide abgebaut wird, wodurch die Aus beute bei diesem Verfahren ungünstig beeinflusst wird.
Es wurde nun überraschenderweise festgestellt, dass man ein 20-Monoenolacylat von 11,20-Diketo- steroiden herstellen kann, wenn man die Acylierungs- reaktion in Gegenwart einer gegebenenfalls substi tuierten Benzolsulfonsäure durchführt, welche min destens eine Nitrogruppe am Benzolkern trägt.
Die Verwendung dieser Säuren besitzt nicht nur den Vorteil, dass sowohl in der normalen (A/B cis) als auch in der Allo-Reihe ausschliesslich 20-Mono- enolacylate erhalten werden, sondern dass auch nach dem erfindungsgemässen Verfahren in der normalen Reihe höhere Ausbeuten an dem gewünschten 17- Hydroxysteroid erhalten werden als nach dem ur sprünglichen Verfahren über die Stufe des 11,20-Di- enolacylates.
Beim Vergleich mit der Verwendung von Per chlorsäure erzielt man nach dem erfindungsgemässen Verfahren höhere Ausbeuten sowohl in der normalen als in der Allo-Reihe, was zum Teil auf der Tat sache beruht, dass die Benzolsulfonsäuren keine oxydierenden Eigenschaften aufweisen und dem entsprechend kein Abbau der Pregnanseitenkette eintritt.
Unter einer gegebenenfalls substituierten Benzol- sulfonsäure ist die Benzolsulfonsäure als solche sowie Benzolsulfonsäuren verstanden, die neben einer oder mehreren Nitrogruppen eine oder mehrere andere Substituenten tragen, z. B. eine Hydroxylgruppe, ein Halogenatom, eine An-iinogruppe, eine Carboxyl- oder eine Alkylgruppe, z.
B. eine Methylgruppe. Zu diesen Säuren zählen auch solche, bei denen der Benzolring mit einem andern aromatischen Ring kondensiert ist. Als Beispiele seien erwähnt: Mono- nitrobenzolsulfonsäuren, wie o-Nitrobenzolsulfonsäure, 2,4-Dinitrobenzolsulfonsäure, 2,4,6-Trinitrobenzolsulfonsäure, 2,6-Dinitro-4-methyl-benzolsulfonsäure und 2,
4-Dinitronaphthalinsülfonsäure. Insbesondere geben Benzolsulfonsäure und Toluol- sulfonsäure mit 2 oder 3 Nitrogruppen hohe Ausbeu ten am gewünschten 20-Monoenolacylat.
Die Acylierungsreaktion gemäss der Erfindung wird im allgemeinen durch Auflösen des 11,20-Di- ketosteroids in einem geeigneten organischen Lö sungsmittel und Zusatz eines Gemisches eines Car- bonsäureanhydrids und einer Benzolsulfonsäure durchgeführt.
Die Reaktion lässt sich mit jedem Carbonsäure- anhydrid vornehmen. Vorzugsweise wird ein alipha- tisches Carbonsäureanhydrid einer Carbonsäure mit 2 bis 6 C-Atomen verwendet. Als Lösungsmittel finden vorzugsweise haloge- nierte Kohlenwasserstoffe, wie 1,2-Dichloräthan oder Tetrachlorkohlenstoff, Verwendung, jedoch sind auch andere Lösungsmittel, wie Äther und Dioxan, brauchbar.
Die Reaktion kann bei verschiedenen Arbeits temperaturen, je nach dem Lösungsmittel und der Dauer der Reaktion, durchgeführt werden. Im all gemeinen arbeitet man bei Temperaturen zwischen -20 und 80 C.
Die Menge an Benzolsulfonsäure beträgt im allgemeinen 0,1 bis 15 Gew.O/a des Steroids.
Die Ausbeute lässt sich durch Abtrennen des er haltenen 20-Enolacylats in an sich bekannter Weise, z. B. durch Extraktion und Kristallisation, bestim men, doch wird im allgemeinen die Ausbeute vom 17-Hydroxy-20-ketosteroid bestimmt, welches durch Oxydation des Reaktionsproduktes mit einer Per säure zum entsprechenden 17,20-Epoxyd und an schliessende Verseifung dieser Verbindungen anfällt. <I>Beispiel 1</I> 5 g 2,4-Dinitrobenzolsulfonsäure werden zu einer Lösung von 100 g 3a-Acetoxy-11,20-diketopregnan in 350 ml Tetrachlorkohlenstoff und 150 ml Essig säureanhydrid gegeben.
Die Reaktionsmischung wird 16 Stunden lang bei Zimmertemperatur stehengelas sen, anschliessend in Wasser gegossen, die Tetra chlorkohlenstoffschicht abgetrennt, mit Wasser ge waschen und schliesslich unter vermindertem Druck auf 175 ml eingeengt.
Auf Grund papierchromatographischer Analyse einer Probe dieser Tetrachlorkohlenstofflösung fiel das dl 7 (=o)-3a,20-Diacetoxy-11-ketopregnen in einer Ausbeute von 980/9 der Theorie an.
Anschliessend werden 635 ml einer 0,85 molaren Lösung von Monoperphthalsäure in Äthylacetat der Tetrachlorkohlenstofflösung zugegeben und dieses Gemisch über Nacht bei Zimmertemperatur stehen gelassen. Das Reaktionsgemisch wird anschliessend mit 1n Natronlauge, dann mit Wasser neutral ge waschen und schliesslich zur Trockne eingedampft. Der Rückstand wird in 1200 ml Äthanol gelöst und danach mit einer Lösung von 80 g Natronlauge in 500 ml Wasser versetzt. Die Mischung wird unter Stickstoff bei 25 C 40 Minuten lang verseift und dann mit Eisessig neutralisiert.
Nach dem Abdestil- lieren des Alkohols unter vermindertem Druck wird der Rückstand in 1200 ml Wasser gegossen. Es fällt das 3 ,17a-Dihydroxy-11,20-diketopregnan aus. Die Fällung wird abfiltriert. Ausbeute 91 g, 970/9 der Theorie; Fp. 197-200 C. Nach Umkristallisation aus Aceton werden 86,2g Reinsubstanz vom Fp. 201 bis 203 C erhalten.
Durch Ersatz des oben erwähnten Essigsäure anhydrids mit einer äquivalenten Menge an Propion- säureanhydrid oder Buttersäureanhydrid erhält man in praktischer gleicher Ausbeute das 3a,17a-Di- hydroxy-11,20-diketopregnan. <I>Beispiel 2</I> 2 g 2,4-Dinitrobenzolsulfonsäure werden zu einer Lösung von 20 g 3ss-Acetoxy-11,20-diketoallopregnan in 70 ml Tetrachlorkohlenstoff und 30 ml Essig säureanhydrid gegeben. Das Reaktionsgemisch wird bei 0 C 18 Stunden lang stehengelassen.
Hierauf wird das Gemisch, wie in Beispiel 1 beschrieben, aufgearbeitet, wobei man eine Lösung des -117( o)- 3ss,20-Diacetoxy-ll-ketoallopregnens in Tetrachlor- kohlenstoff erhält. Auf Grund der papierchromato- graphischen Analyse einer Probe dieser Lösung wurde das gewünschte Enolacetat in 970/aiger Aus beute erhalten.
Nach dem im Beispiel 1 beschriebenen Verfahren wird die so erhaltene Verbindung in das 3ss,17a- Dihydroxy-11,20-diketoallopregnan in einer Aus beute von 18,2 g umgewandelt. Nach Umkristallisa tion aus Aceton werden 17,1 g der reinen Substanz erhalten. [all, _ + 60 C (Dioxan).
<I>Beispiel 3</I> 2 g 2,4,6-Trinitrobenzolsulfonsäure werden zu einer Lösung von 100 g 3a-Acetoxy-11,20-diketo- pregnan in 350 ml Tetrachlorkohlenstoff in 150 ml Essigsäureanhydrid gegeben. Das Reaktionsgemisch wird bei Zimmertemperatur 150 Minuten lang stehen gelassen und anschliessend in Wasser gegossen. Die Tetrachlorkohlenstoffschicht wird abgetrennt, mit Wasser gewaschen und dann unter vermindertem Druck auf ein Volumen von 175 ml eingeengt.
Die Ausbeute an 417(2o) - 3a , 20 -Diacetoxy - 11-keto- pregnen, papierchromatographisch bestimmt, belief sich auf 98 % der Theorie.
Anschliessend wurden 635 ml einer 0,85 molaren Lösung von Monoperphthalsäure in Äthylacetat der Tetrachlorkohlenstofflösung zugegeben und das Ge misch über Nacht bei Zimmertemperatur stehen gelassen. Das Reaktionsgemisch wird mit 1n Natron lauge, dann mit Wasser neutral gewaschen und an schliessend zur Trockene eingedampft. Der Rück stand wird in 1,2 1 Äthanol gelöst, hierauf eine Lö sung von 80 g Ätznatron in 500 ml Wasser zugege ben und das Gemisch 40 Minuten lang unter Stick stoff bei 25a C verseift und mit Eisessig neutralisiert.
Nach dem Abdestillieren des Alkohols unter vermin- dertem Druck wird die Mischung in 1200 ml Wasser gegossen. Es fällt das 3a,17a-Dihydroxy-11,20-di- ketopregnan aus.
Diese Fällung wird abfiltriert. Aus- beute 91 g, 97% der Theorie, Fp. 197-200 C. Nach Umkristallisation aus Aceton fielen 86,2 g Bein substanz vom Fp. 201-203 C an.
<I>Beispiel 4</I> 0,2 g 2,4,6-Trinitrobenzolsulfonsäure werden zu einer Lösung von 20 g 3ss-Acetoxy-11,20-diketo- allopregnan in 70 ml Tetrachlorkohlenstoff und 30 ml Essigsäureanhydrid gegeben und das Reaktions gemisch vier Stunden lang bei 0 C stehengelassen. Die Mischung wird, wie in Beispiel 1 beschrieben, aufgearbeitet, und man erhält eine Lösung von j17(20)-3ss,20-Diacetoxy-11-ketoallopregnen in Tetra chlorkohlenstoff.
Die Ausbeute an Enolacetat belief sich, papierchromatographisch bestimmt, auf 97,5% der Theorie.
Nach dem im Beispiel 3 beschriebenen Verfahren wird die so erhaltene Verbindung in das 3ss,17a-Di- hydroxy-11,20-diketoallopregnan in einer Ausbeute von 18,2 g übergeführt. Nach Umkristallisation aus Aceton werden 17,1g Beinsubstanz mit einem Dreh wert von [all, = + 60 C in Dioxan erhalten.
In analoger Weise wird das 3ss,17a-Dihydroxy- 11,20-diketoallopregnan, ausgehend von 3ss-Acetoxy- 11,20-diketoallopregnan, durch Ersatz von Essig säureanhydrid durch eine äquivalente Menge an Pro- pionsäureanhydrid gewonnen.
<I>Beispiel 5</I> 1,5 g o-Nitrobenzolsulfonsäure werden zu einer Lösung von 10 g 3ss - Acetoxy - 11,20 - diketoallo- pregnan in 40 ml 1,2-Dichloräthan und 7,5 ml Pro- pionsäureanhydrid gegeben. Die Mischung wird bei 50 C 15 Stunden lang reagieren gelassen und an schliessend in Wasser gegossen. Das Gemisch wird, wie in Beispiel 1 beschrieben, aufgearbeitet, und man erhält 3,8,17a - Dihydroxy - 11,20 - diketoallo- pregnan.
<I>Beispiel 6</I> 2 g 2,4 Dinitrobenzolsulfonsäure werden zu einer Lösung von 50 g 3a-Acetoxy-11,20-diketopregnan in 375 ml 1,2-Dichloräthan und 125 ml Essigsäure anhydrid gegeben. Anschliessend wird das Gemisch bei 50a C 6 Stunden lang stehengelassen und dann, wie in Beispiel 1 beschrieben, weiter zum 3a,17a- Dihydroxy-11,20-diketopregnan aufgearbeitet.
<I>Beispiel 7</I> 10 g 3a -Acetoxy-11,20 -diketo -16a - methyl- pregnan, in 35 ml Tetrachlorkohlenstoff und 15 ml Essigsäureanhydrid gelöst, werden mit 1 g 2,4-Dini- trobenzolsulfonsäure versetzt. Das Gemisch wird 30 Stunden bei 25 C reagieren gelassen und dann, wie in Beispiel 1 beschrieben, weiter verarbeitet. Man erhält das 3a,17a-Dihydroxy-11,20-diketo-16a-me- thylpregnan.
<I>Beispiel 8</I> 1 g 2,4-Dinitrobenzolsulfonsäure wird einer Lö sung von 10 g 3,0-Acetoxy-11,20-diketoallopregnan in 35 ml 1,2-Dichloräthan und 5 ml Essigsäure anhydrid zugegeben. Das Gemisch wird zur Reaktion 6 Stunden bei 25 C stehengelassen, dann in Wasser gegossen und schliesslich, wie in Beispiel 1 beschrie ben, weiter verarbeitet. Die Ausbeute ist die gleiche wie in Beispiel 2.
<I>Beispiel 9</I> Zu einer Lösung von 50 g 3a-Acetoxy-11,20-di- ketopregnan in 375 ml 1,2-Dichloräthan und 125 ml Essigsäureanhydrid werden 2 g 2,6-Dinitro-4-methyl- benzolsulfonsäure gegeben und das Gemisch zur Reaktion 8 Stunden bei 50e C stehengelassen. Hierauf wird das Reaktionsgemisch, wie in Beispiel 1 be schrieben, zum 3 a,17a - Dihydroxy - 11,20 - diketo- pregnan weiter verarbeitet.
<I>Beispiel 10</I> 0,3 g 2,4,6-Dinitrobenzolsulfonsäure werden zu einer Lösung von 10 g 3f Acetoxy-11,20-diketoallo- pregnan in 35 ml 1,2-Dichloräthan und 5 ml Essig säureanhydrid gegeben und das Gemisch eine Stunde bei 350C reagieren gelassen. Hierauf wird die Mi schung in Wasser gegossen und, wie in Beispiel 1 beschrieben, weiter verarbeitet. Man erhält die gleiche Ausbeute wie in Beispiel 4.
<I>Beispiel 11</I> Eine Lösung von 50 g 3a-Acetoxy-11,20-diketo- pregnan, in 375 ml 1,2-Dichloräthan und 125 ml Essigsäureanhydrid gelöst, wird mit 0,25 g 2,4,6-Tri- nitrobenzolsulfonsäure versetzt und das Gemisch 150 Minuten lang bei 25 C reagieren gelassen. Hierauf wird das Reaktionsgemisch, wie in Beispiel 3 beschrieben, zum 3a,17a Dihydroxy-11,20-diketo- pregnan weiter verarbeitet.
<I>Beispiel 12</I> 0,3 g 2,4,6-Trinitrobenzolsulfonsäure werden zu einer Lösung von 10 g 3a-Acetoxy-11,20-diketo-16a- methylpregnan in 35 ml Tetrachlorkohlenstoff und 15 ml Essigsäureanhydrid gegeben. Das Reaktions gemisch wird 4,5 Stunden lang bei 25 C stehen gelassen und danach, wie in Beispiel 3 beschrieben, zum 3a,17a-Dihydroxy-11,20-diketo-16a-methyl- pregnan weiter verarbeitet.
<I>Beispiel 13</I> Eine Lösung von 20 g 3,B-Acetoxy-11,20-diketo- allopregnan in 75 ml Tetrachlorkohlenstoff und 30 ml Essigsäureanhydrid werden mit 5 g 2,4-Dini- tronaphthalinsulfonsäure versetzt und das Gemisch bei Zimmertemperatur 20 Stunden lang stehengelas sen. Wie in Beispiel 1 beschrieben, wird das Reak tionsgemisch weiter verarbeitet, und man erhält das 313,17a - Dihydroxy -11,20 - diketoallopregnan mit einem Drehwert von [ab = + 60 C (Dioxan).
<I>Beispiel 14</I> Eine Lösung von 15 g 3f-Hydroxy-11,20-diketo- allopregnan in 65 ml Tetrachlorkohlenstoff und 55 ml Essigsäureanhydrid wird mit 1 g 2,6-Dinitro-4-me- thyl-benzolsulfonsäure versetzt und 12 Stunden lang bei 50 C reagieren gelassen. Das Reaktionsgemisch wird, wie in Beispiel 1 beschrieben, weiter verarbei tet, und man erhält das 3ss,17cc-Dihydroxy-11,20- diketoallopregnan.

A disadvantage of this process is that, due to the oxidizing properties of the perchloric acid used, the pregnancy side chains of the 20-keto steroids are broken down, which has an adverse effect on the yield in this process.
It has now surprisingly been found that a 20-monoenol acylate can be prepared from 11,20-diketo steroids if the acylation reaction is carried out in the presence of an optionally substituted benzenesulfonic acid which has at least one nitro group on the benzene nucleus.
The use of these acids not only has the advantage that only 20-mono-enol acylates are obtained both in the normal (A / B cis) and in the allo series, but also that higher yields are obtained in the normal series in the process according to the invention are obtained on the desired 17-hydroxysteroid than by the ur nal process via the stage of 11,20-dienolacylates.

The acylation reaction according to the invention is generally carried out by dissolving the 11,20-di-keto steroid in a suitable organic solvent and adding a mixture of a carboxylic acid anhydride and a benzenesulfonic acid.
The reaction can be carried out with any carboxylic acid anhydride. An aliphatic carboxylic acid anhydride of a carboxylic acid having 2 to 6 carbon atoms is preferably used. Halogenated hydrocarbons, such as 1,2-dichloroethane or carbon tetrachloride, are preferably used as solvents, but other solvents such as ether and dioxane can also be used.
The reaction can be carried out at different working temperatures, depending on the solvent and the duration of the reaction. In general, you work at temperatures between -20 and 80 C.
The amount of benzenesulfonic acid is generally 0.1 to 15% by weight of the steroid.
The yield can be determined by separating off the 20-enol acylate he has obtained in a manner known per se, for. On the basis of paper chromatographic analysis of a sample of this carbon tetrachloride solution, the dl 7 (= o) -3a, 20-diacetoxy-11-ketopregnene was obtained in a yield of 980/9 of theory.
Then 635 ml of a 0.85 molar solution of monoperphthalic acid in ethyl acetate are added to the carbon tetrachloride solution and this mixture is left to stand overnight at room temperature. The reaction mixture is then washed neutral with 1N sodium hydroxide solution, then with water and finally evaporated to dryness. The residue is dissolved in 1200 ml of ethanol and then a solution of 80 g of sodium hydroxide solution in 500 ml of water is added. After the alcohol has been distilled off under reduced pressure, the residue is poured into 1200 ml of water. The 3,17a-dihydroxy-11,20-diketopregnane precipitates. The precipitate is filtered off. By replacing the above-mentioned acetic anhydride with an equivalent amount of propionic anhydride or butyric anhydride, 3a, 17a-dihydroxy-11,20-diketopregnane is obtained in practically the same yield. <I> Example 2 </I> 2 g of 2,4-dinitrobenzenesulfonic acid are added to a solution of 20 g of 3ss-acetoxy-11,20-diketoallopregnan in 70 ml of carbon tetrachloride and 30 ml of acetic anhydride. The reaction mixture is left to stand at 0 C for 18 hours.
The mixture is then worked up as described in Example 1, a solution of -117 (o) -3ss, 20-diacetoxy-II-ketoallopregnens in carbon tetrachloride being obtained. On the basis of the paper chromatographic analysis of a sample of this solution, the desired enol acetate was obtained in a yield of 970%.
According to the method described in Example 1, the compound thus obtained is converted into the 3ss, 17a-dihydroxy-11,20-diketoallopregnan in a yield of 18.2 g. After recrystallization from acetone, 17.1 g of the pure substance are obtained. [all, _ + 60 C (dioxane).
<I> Example 3 </I> 2 g of 2,4,6-trinitrobenzenesulfonic acid are added to a solution of 100 g of 3a-acetoxy-11,20-diketopregnane in 350 ml of carbon tetrachloride in 150 ml of acetic anhydride. <I> Example 7 </I> 10 g 3a -acetoxy-11,20 -diketo -16a-methyl-pregnane, dissolved in 35 ml carbon tetrachloride and 15 ml acetic anhydride, are mixed with 1 g 2,4-dinitrobenzenesulfonic acid.
