Dans cette formule, A représente un radi cal de formule CnH2n, <I>n</I> étant égal à 2 ou 3, R un radical méthyle ou éthyle<B>;</B> le reste phényle est en position 1 ou 3 du noyau (nu mérotation Beilstein).
Ces nouvelles amines sont obtenues, selon l'invention, par action sur la phénylphéno- thiazine convenable d'une halogénoalcoylamine tertiaire de formule<I>X - A -</I> N(R)2. La réaction est effectuée de préférence en présence d'un agent de condensation capable de donner un dérivé métallique avec la phénothiazine. Ces agents appartiennent au groupe des métaux alcalins ou de leurs dérivés (hydroxydes, hydrures, amidures, alcoolates,
métaux alcoyles ou aryles). Dans ce domaine, on citera plus particulièrement l'amidure de sodium, le so- dium métallique, la soude ou la potasse en poudre, l'hydrure de lithium, le tertiobutylate de sodium, le butyllithium, le phényllithium.
On opère de préférence dans un solvant organique, à la température d'ébullition de ce solvant. Il est particulièrement avantageux d'utiliser l'halogénoalcoylamine tertiaire sous forme de base libre en solution, par exemple dans le benzène, le toluène ou le xylène, et de l'ajouter au mélange des autres réactifs dans lequel la phénothiazine peut déjà se pré senter, du moins en partie, sous forme de sel alcalin.
On peut également effectuer la réac tion sans agent de condensation, en introdui sant peu à peu la solution d'halogénoalcoyl- aminë dans la phénothiazine fondue.
La réaction peut également être réalisée avec un sel de l'halogénoalcoylamine tertiaire, mais, dans ce cas, il faut évidemment- mettre une proportion d'agent de condensation plus forte, de façon à neutraliser l'acide du sel de l'halogénoalcoylamine tertiaire.
La phénylphénothiazine utilisée comme ma tière première peut être préparée par chauf fage de la phényl-3-diphénylamine avec du soufre, en présence d'iode, à une température supérieure à 1500 et comprise de préférence entre 1750 et 1900 ;
un tel mode opératoire peut donner théoriquement à la fois la phényl- 1-phénothiazine et la phényl-3-phénothiazine. Les produits obtenus par le procédé selon l'invention présentent des propriétés pharmaco dynamiques intéressantes qui les rendent utili- sables en thérapeutique humaine ou vétérinaire. Ils peuvent, si on le désire, être transformés en sels d'addition par réaction avec des acides tels que l'acide chlorhydrique.
Les exemples suivants montrent comment le procédé de l'invention peut être mis en #uvre.
<I>Exemple 1</I> A une suspension bouillante de 27,5 g de phénylphénothiazine (P. F. = 175-177o) et 6;5 g d'amidure de sodium à 90 % dans 250 cm3 de xylène, on ajoute peu à peu, en vingt minutes, 88 cm2 d'une solution xylénique à 14,
6 fl/o de diméthylamino-1-chloro-2-éthane. On maintient à l'ébullition pendant encore deux heures après la fin d'addition. Après re froidissement, on reprend par 500 cms d'eau et acidifie au méthylorange par 21 cms d'acide chlorhydrique (d = 1,17).
On sépare le xylène, lave la couche aqueuse par 250 cm8 d'éther puis rend cette couche alcaline à la thymolphtaléine avec 25 cm3 de lessive de soude (d= 1,33), pour libérer la base que l'on extrait avec deux fois 150 cm3 d'éther. On sèche cet extrait éthéré sur sulfate de sodium, chasse l'éther et rectifie.
On obtient ainsi 30,2 g de (diméthylamino- 2'-éthyl)-10-phényl-phénothiazine qui distille à 243-2500 sous 1 mm. On en fait le chlor hydrate par action d'une solution d'acide chlorhydrique dans l'éther sur la base dissoute dans l'acétone. Ce chlorhydrate fond à 145- 146o.
<I>Exempte 2</I> -En opérant de la même manière qu'à l'exemple 1, mais à partir de 27,5 g de phényl- phénothiazine P. F. = 175-177o et de 63 cm3 d'une solution xylénique à 23'% de diméthyl- amino-1-chloro-3-propane, on obtient la (di- méthylamino - 3'-propyl) -10 -phényl-ph6nothia- zine correspondante,

F. = 176-177 et de 48 cm3 de solution xylénique à 19,6% de dim6thylamino-1- chloro-2-propane, on obtient un mélange des bases (diméthylamino-2'-propyl-1')-10-phényl- phénothiazine et (diméthylamino-l'-propyl-2')- 10-phényl-phénothiazine correspondantes, Eb.0,7 = 235-240o.

These novel amines are obtained, according to the invention, by action on the suitable phenylphenothiazine of a tertiary haloalkylamine of formula <I> X - A - </I> N (R) 2. The reaction is preferably carried out in the presence of a condensing agent capable of giving a metal derivative with the phenothiazine. These agents belong to the group of alkali metals or their derivatives (hydroxides, hydrides, amides, alcoholates,
alkyl or aryl metals). In this field, mention will more particularly be made of sodium amide, metallic sodium, powdered sodium hydroxide or potassium hydroxide, lithium hydride, sodium tert-butoxide, butyllithium and phenyllithium.
The operation is preferably carried out in an organic solvent, at the boiling point of this solvent. It is particularly advantageous to use the tertiary haloalkyllamine in the form of the free base in solution, for example in benzene, toluene or xylene, and to add it to the mixture of other reagents in which the phenothiazine can already be present. , at least in part, as an alkali salt.
The reaction can also be carried out without a condensing agent, gradually introducing the haloalkylamine solution into the molten phenothiazine.
The reaction can also be carried out with a salt of the tertiary haloalkyllamine, but, in this case, it is obviously necessary to put a higher proportion of condensing agent, so as to neutralize the acid of the salt of the tertiary haloalkyllamine. .
The phenylphenothiazine used as raw material can be prepared by heating the phenyl-3-diphenylamine with sulfur, in the presence of iodine, at a temperature above 1500 and preferably between 1750 and 1900;
such a procedure can theoretically yield both phenyl-1-phenothiazine and phenyl-3-phenothiazine. The products obtained by the process according to the invention exhibit interesting pharmacodynamic properties which make them usable in human or veterinary therapy. They can, if desired, be converted to addition salts by reaction with acids such as hydrochloric acid.
The following examples show how the process of the invention can be carried out.
<I> Example 1 </I> To a boiling suspension of 27.5 g of phenylphenothiazine (PF = 175-177o) and 6; 5 g of 90% sodium amide in 250 cm3 of xylene, is added little to little, in twenty minutes, 88 cm2 of a xylenic solution at 14,

The xylene is separated, the aqueous layer is washed with 250 cm8 of ether and then made alkaline with thymolphthalein with 25 cm3 of sodium hydroxide solution (d = 1.33), to release the base which is extracted with twice 150 cm3 of ether. This ethereal extract is dried over sodium sulfate, the ether is driven off and rectified.
In this way 30.2 g of (dimethylamino-2'-ethyl) -10-phenyl-phenothiazine are obtained, which distils at 243-2500 under 1 mm. It is made the hydrochloride by the action of a solution of hydrochloric acid in ether on the base dissolved in acetone. This hydrochloride melts at 145-146o.
