Procédé de préparation de nouvelles phénothiazines
La présente invention a pour objet un procédé de préparation de nouvelles phénothiazines de formule:

dans laquelle X représente un atome d'hydrogène ou d'halogène ou un radical alcoyle, alcoyloxyle ou acyle renfermant de 1 à 4 atomes de carbone, cyano, mé thylthio, méthanesulfonyle, diméthylsulfamido ou trifluorométhyle, R représente un atome d'hydrogène ou un radical méthyle, et Rj un radical alcoyle renfermant de 1 à 4 atomes de carbone.
Le procédé selon l'invention est caractérisé en ce que l'on décompose par la chaleur un composé de formule:

Cette décomposition peut être effectuée par chauffage à une température supérieure à 100o C et de préférence entre 150 et 2500 C. Il n'y a pas d'avantage à opérer à des températures plus élevées, les produits réactionnels se trouvant alors généralement plus colorés. On peut chauffer le produit seul, sans diluant, ou dans un solvant organique inerte à point d'ébullition élevé tel que diphényle, oxyde de diphényle, quinoléine, bases faibles ou o. dichlorobenzène.
Les phénothiazines de départ de la formule Il peuvent être obtenues par les méthodes connues, par exemple: action d'un halogénure (ou d'un ester) d'acide phénothiazinyl-10 carboxylique sur un composé de formule générale:

Les nouvelles phénothiazines obtenues par le procédé selon l'invention présentent des propriétés pharmacodynamiques, en particulier sédatives, analgésiques et antiémétiques, très importantes; leur toxicité est faible et elles ne présentent pratiquement pas d'actions secondaires indésirables.
Pour l'emploi thérapeutique, ces nouvelles phé- nothiazines sont utilisées de préférence sous forme de base ou de sels renfermant des anions thérapeutique- ment acceptables tels que chlorhydrates, sulfates, tartrates, maléates, fumarates, méthanesulfonates ou éthanedisulfonates.
Exemple 1
On chauffe pendant 1 heure vers 2300 sous 1,5 mm de mercure 1 g de chloro-3 [(méthanesulfo nyl-4" pipérazino)- 3' propoxycarbonyl] - 10 phénothiazine. On recristallise dans l'éthanol et on obtient 0,4 g de chloro-3 [(méthanesulfonyl-4" pipérazino)-3' propyl]-l0 phénothiazine, fondant à 1380.
On prépare la chloro-3 [(méthanesulfonyl-4" pi pérazino)3' propoxycarbonyl]-l0 phénothiazine en chauffant à reflux pendant 18 heures 29,6 g de chlorure de l'acide chloro-3 phénothiazinyl-l0 carboxylique avec 25 g de méthanesulfonyl-l (hydroxy-3' propyl)-4 pipérazine dans la butanone avec 1,4 g de carbonate de potassium. On concentre sous vide, reprend par 50 cm3 d'une solution saturée de carbonate de potassium et 100 cm3 de chloroforme. On extrait la base avec 75 cm8 d'acide chlorhydrique à 100/0. On libère la base par le carbonate de potassium et extrait au chloroforme. On obtient 21,5 g de chloro-3 [(mé thanesulfonyl-4" pipérazino)-3' propoxycarbonyl] - 10 phénothiazine qui, par recristallisation dans l'éthanol, fond à 1300.
Exemple 2
En opérant comme dans l'exemple 1, à partir de 1 g de diméthylsulfamino-3 [(méthanesulfonyl-4" pi pérazino)3' propoxycarbonyl]-l0 phénothiazine, on obtient 0, 4 g de diméthylsulfamido-3 [(méthanesul fonyl-4" pipérazino)-3' propyl]-l0 phénothiazine qui, par recristallisation dans l'acétate d'éthyle, fond à 165-1660.
Cette base peut être transformée en dérivé d'ammonium quaternaire comme suit:
On chauffe à reflux pendant 5 heures, 5,1 g de diméthylsulfamido-3 [(méthanesulfonyl-4" pipéra- zino)-3' propyl]-10 phénothiazine avec 75 cm3 d'acétone et 5 cm3 d'iodure de méthyle. Après distillation du solvant, on recristallise dans 450cm3 d'éthanol.
On obtient 6 g d'iodométhylate de diméthylsulfamido3 [(méthanesulfonyl-4" pipérazino)- 3' propyl]- 10 phénothiazine, fondant vers 216-2180 avec décomposition.
On prépare la diméthylsulfamido-3 [(méthanesul fonyl-4" pipérazino)-3' propoxycarbonyl]-l0 phénothiazine de départ par chauffage à reflux pendant 4 heures de 9 g de chlorure de l'acide diméthylsulfamido-3 phénothiazinyl-l0 carboxylique avec 9 g de méthanesulfonyl-l (hydroxy-3' propyl)-4 pipérazine dans le toluène. Par le traitement habituel et recristallisation dans l'éthanol, on obtient 8,3 g de diméthyl-sulfamido-3 [(méthanesulfonyl-4" pipérazino)-3' propoxycarbonyl]-10 phénothiazine, fondant à 1700.
Exemple 3
On chauffe pendant 1 heure vers 2000, sous 1,5 mm de mercure, en agitant, 54 g de cyano-3 [(méthanesulfonyl-4" pipérazino)-3' propyloxycarbo ny]]-10 phénothiazine avec 5,4 g de poudre de verre.
On chauffe vers 800 en agitant avec 500 cm9 de toluène et 150 cm3 d'acide chlorhydrique normal. On obtient par concentration du toluène décanté, 6,5 g de cyano-3 phénothiazine récupérée. On traite la solution aqueuse acide et le chlorhydrate huileux précipite avec 20 cm5 de lessive de soude d = 1,33 et on agite à une température de 500 avec 250 cm3 de benzène. La solution benzénique décantée et refroidie donne 28 g de cyano-3 [(méthanesulfonyl-4" pipéra- zino)-3' propyl] - 10 phénothiazine, fondant après recristallisation dans le benzène à 166-1680.
On obtient la cyano-3 [(méthanesulfonyl-4" pipé razino)-3' propyloxycarbonyl] - 10 phénothiazine en coulant une solution de 177,6 g d'hydroxy-3' propyl)-l méthanesulfonyl-4 pipérazine dans 150 cm3 de chloroforme dans une solution tiède de 114,6 g de chlorure de cyano-3 phénothiazinyl- 10 carbonyle dans 500 cm3 de toluène. On distille le chloroforme en une heure puis on chauffe à reflux pendant 7 heures. On traite avec 400 cm3 d'eau et ajoute 50 cm8 de lessive de soude d = 1,33. On décante et lave les eaux au benzène. On agite la couche organique avec 500cm3 d'acide chlorhydrique normal.
On décante, ajoute à la solution aqueuse 75 cm5 de lessive de soude d = 1,33 et extrait la base avec 500 cm3 et deux fois 250 cm5 d'acétate d'éthyle. Par concentration on obtient 140 g de cyano-3 [(méthanesulfonyl 4" pipérazino)-3' propyloxycarbonyl]-l0 phénothiazine qui, après recristallisation dans l'acétate d'éthyle, fond à 162-1630.
En procédant de manière analogue aux exemples ci-dessus, on peut préparer les phénothiazines suivantes: [(méthanesulfonyl-4" pipérazino)-3' propyl] - 10 phénothiazine, p.f. 120-1220; méthyl-3 [(méthanesulfonyl-4" pipérazino)-3' pro- pyl]-10 phénothiazine, p.f. 121-1230; éthyl-3 [(méthanesulfonyl-4" pipérazino)-3' pro- pyl]-l0 phénothiazine, p.f. 131-1330; méthoxy-3 [(méthanesulfonyl-4" pipérazino)-3' pro- pyl]-l0 phénothiazine, p.f. 1180;

This decomposition can be carried out by heating at a temperature above 100 ° C. and preferably between 150 and 2500 C. The new phenothiazines obtained by the process according to the invention exhibit very significant pharmacodynamic properties, in particular sedative, analgesic and antiemetics; their toxicity is low and they show practically no undesirable side actions.
For therapeutic use, these novel phenothiazines are preferably used in the form of a base or of salts containing therapeutically acceptable anions such as hydrochlorides, sulfates, tartrates, maleates, fumarates, methanesulfonates or ethanedisulfonates.
Example 1
1 g of 3-chloro [(methanesulfonyl-4 "piperazino) - 3 'propoxycarbonyl] - phenothiazine is heated for 1 hour at around 2300 under 1.5 mm of mercury.
