Procédé de préparation de nouvelles phénothiazines
La présente invention a pour objet un procédé de préparation de nouvelles phénothiazines de formule:

dans laquelle X représente un atome d'hydrogène ou d'halogène ou un radical alcoyle, alcoyloxyle ou acyle renfermant de 1 à 4 atomes de carbone, cyano, méthylthio, méthanesulfonyle, diméthylsulfamido, ou trifluorométhyle, R représente un atome d'hydrogène ou un radical méthyle, et R1 un radical alcoyle renfermant de 1 à 4 atomes de carbone.
Le procédé selon l'invention est caractérisé en ce que l'on condense un ester réactif de formule:

dans laquelle Y représente un reste d'ester réactif tel qu'un atome d'halogène ou un reste d'ester sulfurique ou sulfonique, sur une phénothiazine de formule:

ou en ce que l'on condense un ester réactif de formule:

Ce procédé est de préférence conduit au sein d'un diluant organique liquide, inerte dans les conditions opératoires, tel qu'un hydrocarbure aromatique, de préférence benzène ou toluène, ou un hydrocarbure chloré, tel que le chloroforme, en présence d'un agent de condensation du groupe des métaux alcalins ou des amines tertiaires. On opère de préférence à la température d'ébullition du solvant.
Les nouvelles phénothiazines obtenues par le procédé selon l'invention présentent des propriétés pharmacodynamiques, en particulier sédatives, analgésiques et antiémétiques, très importantes ; leur toxicité est faible et elles ne présentent pratiquement pas d'actions secondaires indésirables.
Pour l'emploi thérapeutiquoa ces nouvelles phénothiazines sont utilisées de préférence sous forme de base ou de sels renfermant des anions thérapeutiquement acceptables tels que chlorhydrates, sulfates, tartrates, maléates, fumarates, méthanesulfonates ou éthanedisulfonates.
Exemple 1
On dissout en agitant et chauffant à reflux 113 g de diméthylsulfamido-3 phénothiazine dans 1100 cm3de toluène. On ajoute à chaud en 5 minutes 175 cm d'une solution toluénique renfermant 63,2 g de diméthylphénylcarbinolate de sodium. On ajoute ensuite en 15 minutes 360 ces d'une solution-toluénique contenant 96,2 g de (chloro-3'-propyl)-1-(méthanesulfonyl)-4-pipérazine. On poursuit le chauffage à reflux et l'agitation pendant 5 heures. On lave la solution froide avec 600 cm3 d'eau. On agite la solution toluénique avec 500 cm3 d'acide chlorhydrique normal. Le chlorhydrate précipite en glu. On traite ce chlorhydrate avec 250 cm3 de chloroforme et 120cm3 d'une solution saturée de carbonate de potassium.
Après distillation du chloroforme, on obtient 190 g de diméthylsulfamido-3-[(méthanesulfo- nyl-4"-pipérazino)-3'-propyl]-10-phénothiazine qui, après purification par chromatographie sur alumine et recristallisation dans le toluène, fond à 1700.
Le produit de cet exemple peut être transformé comme suit en dérivé d'ammonium quaternaire.
On chauffe à reflux pendant 5 heures, 5,1 g de diméthylsulfamido-3-(méthanesulfonyl-4"-pipéra zino)-3'-propyl-10-phénothiazine avec 75 cm3 d'acétone et 5 cm3 d'iodure de méthyle. Après distillation du solvant, on recristallise dans 450 cm3 d'éthanol.
On obtient 6 g d'iodométhylate de diméthylsulfamido3-[(méthanesulfonyl-4"-pipérazino)-3'-propyl]-10phénothiazine, fondant vers 216-218O avec décomposition.
Exemple 2
En opérant comme dans l'exemple 1, à partir de 22,4 g de cyano-3-phénothiazine, on obtient 14,1 g de cyano-3-[(méthanesulfonyl-4"-pipérazino)-3'-pro- pyl]10-phénothiazine, fondant à 166-1680.

Process for the preparation of new phenothiazines
The subject of the present invention is a process for the preparation of novel phenothiazines of formula:

in which X represents a hydrogen or halogen atom or an alkyl, alkyloxyl or acyl radical containing from 1 to 4 carbon atoms, cyano, methylthio, methanesulfonyl, dimethylsulfamido, or trifluoromethyl, R represents a hydrogen atom or a methyl radical, and R1 an alkyl radical containing 1 to 4 carbon atoms.
The process according to the invention is characterized in that a reactive ester of formula is condensed:

in which Y represents a residue of a reactive ester such as a halogen atom or a residue of a sulfuric or sulphonic ester, on a phenothiazine of formula:

This process is preferably carried out in a liquid organic diluent, inert under the operating conditions, such as an aromatic hydrocarbon, preferably benzene or toluene, or a chlorinated hydrocarbon, such as chloroform, in the presence of an agent. condensation of the group of alkali metals or tertiary amines. The operation is preferably carried out at the boiling point of the solvent.
The novel phenothiazines obtained by the process according to the invention exhibit very significant pharmacodynamic properties, in particular sedatives, analgesics and antiemetics; their toxicity is low and they show practically no undesirable side actions.
For therapeutic use, these new phenothiazines are preferably used in the form of a base or of salts containing therapeutically acceptable anions such as hydrochlorides, sulfates, tartrates, maleates, fumarates, methanesulfonates or ethanedisulfonates.
Example 1
113 g of 3-dimethylsulfamido-phenothiazine are dissolved with stirring and heating under reflux in 1100 cm3 of toluene. Is added hot over 5 minutes 175 cm of a toluene solution containing 63.2 g of sodium dimethylphenylcarbinolate. 360 ces of a toluene solution containing 96.2 g of (chloro-3'-propyl) -1- (methanesulfonyl) -4-piperazine are then added over 15 minutes. Heating under reflux and stirring are continued for 5 hours. The cold solution is washed with 600 cm3 of water. The toluene solution is stirred with 500 cm3 of normal hydrochloric acid. The hydrochloride precipitates as a glue. This hydrochloride is treated with 250 cm3 of chloroform and 120cm3 of a saturated solution of potassium carbonate.
After distillation of the chloroform, 190 g of 3-dimethylsulfamido - [(methanesulfonyl-4 "-piperazino) -3'-propyl] -10-phenothiazine are obtained which, after purification by chromatography on alumina and recrystallization from toluene, melts at 1700.
The product of this example can be converted to the quaternary ammonium derivative as follows.
Is refluxed for 5 hours, 5.1 g of dimethylsulfamido-3- (methanesulfonyl-4 "-pipera zino) -3'-propyl-10-phenothiazine with 75 cm3 of acetone and 5 cm3 of methyl iodide. After distillation of the solvent, recrystallized from 450 cm3 of ethanol.
6 g of dimethylsulfamido3 - [(methanesulfonyl-4 "-piperazino) -3'-propyl] -10phenothiazine iodomethylate are obtained, melting at about 216-218O with decomposition.
Example 2
By operating as in Example 1, from 22.4 g of cyano-3-phenothiazine, 14.1 g of cyano-3 - [(methanesulfonyl-4 "-piperazino) -3'-propyl is obtained. ] 10-Phenothiazine, melting at 166-1680.
