La réaction peut être effectuée à partir d'un ester ou d'un halogénure de l'acide phénothiazinyl- 10-carboxylique. Le mode de réalisation préféré consiste à utiliser le chlorure d'acide et à le faire réagir sur un alcool de formule (II).
On peut opérer avec ou sans solvant, de préférence en présence d'un agent de condensation basique tel que par exemple un carbonate ou un hydroxyde alcalin, une amine aliphatique ou aromatique tertiaire ou une base hété- rocyclique tertiaire comme la pyridine. On peut encore faire agir le chlorure d'acide sur un dérivé alcalin d'un alcool de formule (11).
Les composés obtenus suivant le procédé de l'invention possèdent des propriétés pharmacodyna miques intéressantes, en particulier des propriétés spasmolytiques et anesthésiques locales.
Ils, peuvent être utilisés soit directement, soit sous forme de sel contenant un anion pharmaceutiquement acceptable, soit sous forme de sel d'ammonium qua ternaire. Enfin, ils peuvent servir de matières pre mières pour obtenir d'autres produits intéressants. <I>Exemple 1</I> 7,9 g de chlorure de l'acide phénothiazinyl-10- carboxylique, 9,2 g de bis-(diméthylamino)-1,3-pro- panol-2, 30 cm3 de toluène sont chauffés pendant 3 heures.
Après refroidissement, la suspension cris talline obtenue est extraite à l'eau puis à l'acide chlor hydrique dilué. Les solutions aqueuses sont alcalini sées. Le produit qui se sépare est extrait à l'éther. Par évaporation du solvant, on obtient un produit brut qu'on purifie par cristallisation dans l'éther de pétrole. On obtient ainsi 10,5 g de phénothiazinyl- 10'-carboxylate de bis-(diméthylamino)-1,3-propyle-2 fondant (Kofler) à 970.
Le dichlorhydrate correspon dant fond avec décomposition vers 2120 au bloc Kofler.
Le monoiodométhylate obtenu par action d'une molécule d'iodure de méthyle dans l'acétone à reflux fond (Kofler) à 1680. Le bis-(diméthylamino)-1,3-propanol de départ est préparé par action de la diméthylamine sur l'épi- chlorhydrine selon Campbell, J. Org. Chem., 14, 346 (l949).
<I>Exemple 2</I> 11,25 g de chlorure de l'acide phénothiazinyl-10- carboxylique, 10 g de dipipéridino-1,3-propanol-2 et 10 cm3 de toluène sont chauffés à reflux pendant 16 heures. On traite comme précédemment. Par évaporation de l'éther, il reste 19 g d'une huile qui cristallise dans l'isopropanol. On obtient ainsi 8,8 g de phénothiazinyl-10'-carboxylate de dipipéridino- 1,3-propyle-2 fondant (Kofler) à 900.
Le diiodométhylate correspondant obtenu par chauffage à reflux avec de l'iodure de méthyle pré sente, après recristallisation dans l'eau, un point de fusion (Maquenne) de 277,1.
Le dipipéridino-1,3-propanol-2 de départ est obtenu par action de la pipéridine sur l'épichlor- hydrine.
<I>Exemple 3:</I> 13,1 g de chlorure de l'acide phénothiazinyl-10- carboxylique, 8 g de bis-(diméthylaminométhyl)-2,2- éthanol et 10 cm3 de toluène sont chauffés à reflux pendant 17 heures. On traite comme dans l'exemple 1 et on obtient par évaporation de l'éther 9,5 g de phénothiazinyl-10'-carboxylate de bis-(diméthylami- nométhyl)-2,
2-éthyle sous forme d'une huile non distillable et dont le diiodornéthylate obtenu par chauffage à reflux avec de l'iodure de méthyle fond (Maquenne) à 2580.
Le bis- (diméthylaminométhyl)-2,2-éthanol de départ est obtenu selon Mannich et al. B., 65, 384 (1932).
<I>Exemple 4:</I> 12,3 g de bis-(diéthylamino)-1,3-propanol-2 sont dissous dans 10 cm3 de pyridine anhydre. On ajoute 16g de chlorure de l'acide phénothiazinyl-10-car- boxylique et on chauffe 5 heures au bain-marie. Après refroidissement on reprend par de l'eau et du carbonate de potassium et extrait à l'éther. La solu tion éthérée est lavée puis extraite à l'acide chlorhy drique dilué ; par alcalinisation de la solution acide, il se sépare un produit qu'on extrait à l'éther.
On sèche, évapore le solvant en chauffant à la fin vers 1000 sous 1 mm de mercure. On obtient ainsi 6,6 g de phénothiazinyl-10'-carboxylate de bis-(diéthyl- amino)-1,3-propyle-2, huile visqueuse non distillable.
Le bis-(diéthylamino)-1,3-propanol de départ est obtenu par action de la diéthylamine sur l'épichlor- hydrine selon Ingold et Rothstein, Soc., 1931, p. 1672.

The reaction can be carried out from an ester or a halide of phenothiazinyl-10-carboxylic acid. The preferred embodiment consists in using the acid chloride and reacting it with an alcohol of formula (II).
The operation can be carried out with or without a solvent, preferably in the presence of a basic condensing agent such as, for example, a carbonate or an alkali metal hydroxide, a tertiary aliphatic or aromatic amine or a tertiary heterocyclic base such as pyridine. It is also possible to cause the acid chloride to act on an alkaline derivative of an alcohol of formula (11).
The compounds obtained according to the process of the invention possess interesting pharmacodynamic properties, in particular spasmolytic and local anesthetic properties.
They can be used either directly or in the form of a salt containing a pharmaceutically acceptable anion, or in the form of a quaternary ammonium salt. Finally, they can be used as raw materials to obtain other interesting products. <I> Example 1 </I> 7.9 g of phenothiazinyl-10-carboxylic acid chloride, 9.2 g of bis- (dimethylamino) -1,3-propanol-2, 30 cm3 of toluene are heated for 3 hours.
After cooling, the crystalline suspension obtained is extracted with water and then with dilute hydrochloric acid. Aqueous solutions are alkalinized. The product which separates is extracted with ether. By evaporation of the solvent, a crude product is obtained which is purified by crystallization from petroleum ether. In this way 10.5 g of bis- (dimethylamino) -1,3-propyl-2 phenothiazinyl-10'-carboxylate, melting (Kofler) at 970, is obtained.
The corresponding dihydrochloride melts with decomposition around 2120 at the Kofler block.
The monoiodomethoxide obtained by the action of a methyl iodide molecule in acetone at reflux (Kofler) at 1680. The starting bis- (dimethylamino) -1,3-propanol is prepared by the action of dimethylamine on l epichlorohydrin according to Campbell, J. Org. Chem., 14, 346 (1949).
<I> Example 2 </I> 11.25 g of phenothiazinyl-10-carboxylic acid chloride, 10 g of dipiperidino-1,3-propanol-2 and 10 cm3 of toluene are heated at reflux for 16 hours. We treat as before. The corresponding diiodomethoxide obtained by heating under reflux with methyl iodide exhibits, after recrystallization from water, a melting point (Maquenne) of 277.1.
The starting dipiperidino-1,3-propanol-2 is obtained by the action of piperidine on epichlorhydrin.
<I> Example 3: </I> 13.1 g of phenothiazinyl-10- carboxylic acid chloride, 8 g of bis- (dimethylaminomethyl) -2.2- ethanol and 10 cm3 of toluene are heated at reflux for 17 hours. The mixture is treated as in Example 1 and 9.5 g of bis- (dimethylaminomethyl) -2 phenothiazinyl-10′-carboxylate are obtained by evaporation of the ether,
2-ethyl in the form of a non-distillable oil, the diiodornethylate of which obtained by heating under reflux with methyl iodide melts (Maquenne) at 2580.
The starting bis- (dimethylaminomethyl) -2,2-ethanol is obtained according to Mannich et al. B., 65, 384 (1932).
<I> Example 4: </I> 12.3 g of bis- (diethylamino) -1,3-propanol-2 are dissolved in 10 cm3 of anhydrous pyridine. 16 g of phenothiazinyl-10-carboxylic acid chloride are added and the mixture is heated for 5 hours in a water bath. After cooling, the residue is taken up in water and potassium carbonate and extracted with ether. The ethereal solution is washed and then extracted with dilute hydrochloric acid; on alkalization of the acid solution, a product separates which is extracted with ether.
It is dried and the solvent is evaporated off by heating at the end to around 1000 under 1 mm of mercury. 6.6 g of bis- (diethylamino) -1,3-propyl-2 phenothiazinyl-10'-carboxylate, a viscous non-distillable oil, are thus obtained.
The starting bis- (diethylamino) -1,3-propanol is obtained by the action of diethylamine on epichlorhydrin according to Ingold and Rothstein, Soc., 1931, p. 1672.
