Procédé de préparation de nouvelles phénothïazines
La présente invention a pour objet un procédé de préparation de nouvelles phénothiazines de formule :

dans laquelle X représente un atome d'hydrogène ou d'halogène ou un radical alcoyle, alcoyloxyle ou acyle renfermant de 1 à 4 atomes de carbone, cyano, méthylthio, méthanesulfonyle, diméthylsulfamido, ou trifluorométhyle, R représente un atome d'hydrogène ou un radical méthyle, et Ri un radical alcoyle renfermant de 1 à 4 atomes de carbone.
Le procédé selon l'invention est caractérisé en ce que l'on condense sur une pipérazine de formule générale :

un halogénure d'alcanesulfonyle de formule : Hal-SO-Rl dans laquelle Hal désigne un atome d'halogène.
Ce procédé est de préférence conduit au sein d'un diluant organique liquide, inerte dans les conditions opératoires, tel qu'un hydrocarbure aromatique, de préférence benzène ou toluène, ou un hydrocarbure chloré, tel que le chloroforme, en présence d'un agent de condensation du groupe des métaux alcalins ou des amines tertiaires. On opère de préférence à la température d'ébullition du solvant.
Les nouvelles phénothiazines obtenues par le procédé selon l'invention présentent des propriétés pharmacodynamiques, en particulier sédatives, analgésiques et antiémétiques, très importantes ; leur toxicité est faible et elles ne présentent pratiquement pas d'actions secondaires indésirables.
Pour l'emploi thérapeutique, ces nouvelles phéno- thiazines sont utilisées de préférence sous forme de base ou de sels renfermant des anions thérapeutique- ment acceptables tels que chlorhydrates, sulfates, tartrates, maléates, fumarates, méthanesulfonates ou éthanedisulfonates.
Exemple 1
On ajoute goutte à goutte en 5 minutes, en agitant, 3 g de méthanesulfochlorure dans une solution de 8, 8 g de cyano-3 (pipérazino-3'propyl)-10 phéno- thiazine dans 50 cm3 de toluène et 2, 1 g de pyridine anhydre. On chauffe pendant 3 heures au bain-marie.
On traite avec 50 cm3 de soude à 8 /o et 50 cm3 de chloroforme. On décante et épuise la fraction aqueuse avec deux fois 20 cm3 de chloroforme. On lave à l'eau la solution chloroformique, sèche sur carbonate de potassium et concentre sous vide. On obtient 9, 5 g de cyano-3 [(méthanesulfonyl-4"pipérazino)-3'pro- pyl]-10 phénothiazine qui, après recristallisation dans le benzène fond à 166-168 .
Exemple 2
On ajoute en 5 minutes, en agitant, 6 g de méthanesulfochlorure dans une solution de 17, 9 g de chloro-3 (pipérazino-3'propyl)-10 phénothiazine dans 120 cnr de toluène et 4, 2 g de pyridine anhydre.
On laisse une nuit à 20¯ puis on chauffe au bainmarie pendant 2 heures et demie. On ajoute 100 cm de soude à 8"/e et 100 cm de chloroforme. On lave à l'eau, sèche sur carbonate de potassium et concentre sous vide. On obtient 20 g de chloro-3 [(méthane- sulfonyl-4"pipérazino)-3'propyl]-10 phénothiazine qui. après recristallisation dans l'éthanol, fond à 138".
Exemple 3
En opérant comme à l'exemple 2, à partir de 1, 9 g de méthanesulfochlorure et 6, 5 g de diméthyl- sulfamido-3 (pipérazino-3'propyl)-10 phénothiazine, on obtient 7, 3 g de chloro-3 [ (éthanesul- fonyl-4"pipérazino)-3'propyl]-10 phénothiazine,
fondant à 93¯.
Exemple 13
En opérant comme dans l'exemple 4, avec 17, 5 g
de cyano-3 (pipérazino-3'propyl)-10 phénothiazine et 6, 4 g d'éthanesulfochlorure, en recristallisant dans l'acétate d'éthyle, on obtient 13, 5 g de cyano-3 [ (ethanesulfonyl-4" pipérazino)-3'propyl]-10 phéno- thiazine, fondant à 128-1300.
Exemple 14
En opérant comme dans l'exemple 4, avec 6, 5 g de dimëthylsulfamido-3 (pipérazino-3'propyl)-10 phenothiazine et 2 g d'éthanesulfochlorure, en recris tallisant dans le méthanol, on obtient 3, 3 g de diméthylsulfamido-3 [ (6thanesulfonyl-4" pip6razino)-3' propyl]-10 phÚnothiazine, fondant à 119^.
Exemple 15
On ajoute en 15 minutes une solution de 3, 1 g de méthanesulfochlorure dans 5 cm3 de chlorofoune à une solution de 9, 9 g de trifluorométhyl-3 (pipéra- zino-3'propyl)-10 phenothiazine dans 50 cm3 de chloroforme et 2, 1 g de pyridine anhydre. On chauffe à reflux en agitant pendant 2 heures. On laisse re froidir et agite avec 40 cm3 d'une solution saturée de carbonate de potassium. On distille le chloroforme et purifie une solution dans le benzène contenant 25 /n de cyclohexane par chromatographie sur colonne d'alumine.
On obtient 10, 5 g de trifluorométhyl-3 [(méthane- sulfonyl-4"pipérazino)-3'propyll-10 phenothiazine, fondant après recristallisation dans l'éthanol, à 157 .
Exemple 16
En opérant comme dans l'exemple 15, à partir de 12, 5 g de méthylthio-3 (pipérazino-3'méthyl-2' propyl)-10 phénothiazine et 3, 8 g de méthanesulfochlorure, on obtient 8, 6 g de méthylthio-3 [(méthane- sulfonyl-4"pipérazino)-3'méthyl-2'propyl]-10 phe- nothiazine, fondant à 177ç}.
Exemple 17
En opérant comme dans l'exemple 15 à partir de 12, 5 g de méthanesulfonyl-3 (pipérazino-3'méthyl-2' propyl)-10 phÚnothiazine et 3,8 g de mÚthanesulfochlorure, on obtient 11,7 g de mÚthanesulfonyl-3 [(méthanesulfonyl-4"pipérazino)-3'méthyl-2'pro- pyl]-10 phenothiazine qui, après recristallisation dans l'acétate d'éthyle fond à 178".
Exemple 18
En opérant comme dans l'exemple 15, à partir de 10, 8 g de chloro-3 (pipérazino-3'propyl)-10 pheno- thiazine et de 6, 6 g de butanesulfobromure, on obtient après purification par chromatographie et recristallisation dans l'Úthanol, 8, 6 g de chloro-3 [(butane sulfonyl-4"pipérazino)-3'propyl-10 phenothiazine, fondant à 96^.
Exemple 19
En opérant comme dans l'exemple 15, à partir de 13 g de diméthylsulfamido-3 (pipérazino-3'pro- pyl)-10 phenothiazine et de 6, 6 g de butanesulfobromure, on obtient après recristallisation dans l'Úthanol, 11, 6 g de dimÚthylsulfamido-3 [(butanesulfonyl-4" pipérazino)-3'propyl]-10 phénothiazine, fondant à 136-1380.
Exemple 20
En opérant comme dans l'exemple 15, à partir de 10, 8 g de chloro-3 (pipérazino-3'propyl)-10 phé- nothiazine et de 6, 2 g de propanesulfobromure, on obtient, après chromatographie et recristallisation dans l'Úthanol, 7, 5 g de chloro-3 [(propanesulfonyl 4"pipérazino)-3'propyl]-10 phénothiazine, fondant à 112 .
Exemple 21
En opérant comme dans l'exemple 15, à partir de 13 g de diméthylsulfamido-3 (pipérazino-3'pro- pyl)-10 phénothiazine et de 6, 2 g de propanesulfobromure, on obtient après chromatographie et recristallisation dans l'Úthanol, 10, 6 g de diméthylsul famido-3 [(propanesulfonyl-4" pipÚrazino)-3' pro pyl]-10 phénothiazine, fondant à 1340.

This process is preferably carried out in a liquid organic diluent, inert under the operating conditions, such as an aromatic hydrocarbon, preferably benzene or toluene, or a chlorinated hydrocarbon, such as chloroform, in the presence of an agent. condensation of the group of alkali metals or tertiary amines. The novel phenothiazines obtained by the process according to the invention exhibit very significant pharmacodynamic properties, in particular sedatives, analgesics and antiemetics; their toxicity is low and they show practically no undesirable side actions.

Example 1
3 g of methanesulfochloride in a solution of 8.8 g of cyano-3 (piperazino-3′propyl) -10 phenothiazine in 50 cm3 of toluene and 2.1 g are added dropwise over 5 minutes, with stirring. of anhydrous pyridine. It is heated for 3 hours in a water bath.
Treated with 50 cm3 of 8 / o sodium hydroxide and 50 cm3 of chloroform. Decanted and the aqueous fraction exhausted with twice 20 cm3 of chloroform. The chloroform solution is washed with water, dried over potassium carbonate and concentrated in vacuo. 9.5 g of cyano-3 [(methanesulfonyl-4 "piperazino) -3'propyl] -10 phenothiazine are obtained which, after recrystallization from benzene, melts at 166-168.
Example 2
6 g of methanesulfochloride in a solution of 17.9 g of chloro-3 (piperazino-3′propyl) -10 phenothiazine in 120 cm of toluene and 4.2 g of anhydrous pyridine are added over 5 minutes, with stirring.

melting at 93¯.
Example 13
By operating as in Example 4, with 17.5 g
of cyano-3 (piperazino-3′propyl) -10 phenothiazine and 6.4 g of ethanesulfochloride, by recrystallizing from ethyl acetate, 13.5 g of cyano-3 [(ethanesulfonyl-4 "piperazino ) -3'propyl] -10 phenothiazine, mp 128-1300.
Example 14
By operating as in Example 4, with 6.5 g of 3-dimethylsulfamido (piperazino-3′propyl) -10 phenothiazine and 2 g of ethanesulfochloride, by recreating tallizing in methanol, 3.3 g of dimethylsulfamido are obtained. -3 [(6thanesulfonyl-4 "pip6razino) -3 'propyl] -10 phÚnothiazine, melting at 119%.
Example 15
A solution of 3.1 g of methanesulfochloride in 5 cm3 of chloroform is added over 15 minutes to a solution of 9.4 g of trifluoromethyl-3 (piperazino-3'propyl) -10 phenothiazine in 50 cm3 of chloroform and 2 , 1 g of anhydrous pyridine. Heated to reflux with stirring for 2 hours. Allowed to cool and stirred with 40 cm3 of a saturated solution of potassium carbonate. The chloroform is distilled off and a solution in benzene containing 25 / n of cyclohexane is purified by chromatography on an alumina column.

Example 16
By operating as in Example 15, from 12.5 g of 3-methylthio (piperazino-3'methyl-2 'propyl) -10 phenothiazine and 3.8 g of methanesulfochloride, 8.6 g of methylthio are obtained. -3 [(methanesulfonyl-4 "piperazino) -3'methyl-2'propyl] -10 phe-nothiazine, mp 177%.
Example 17
