Verfahren zur Herstellung eines 6,7-Dihydro-5H-dibenz[c,e]azepins. Die vorliegende Erfindung betrifft die Herstellung von 6,7-Dihydro-5H-dibenz[c,e]- azepin und seinen Derivaten. Diese neuen Verbindungen sind therapeutisch verwendbar. Vor allem besitzen sie adrenolytische Eigen schaften, die die physiologische Wirkung des Epinephrins hemmen oder umkehren.
Gemäss der vorliegenden Erfindung wer den die erwähnten Verbindungen durch Um setzung des o,o'-bis-(Brommethyl)-biphenyls mit Ammoniak oder einem primären oder sekundären Amin hergestellt.
Geeignete primäre Amine sind zum Bei spiel die gesättigten oder ungesättigten Alkyl- amine, die Arylamine, die Cycloalkylamine, die Aralkylamine, die Oxyalkylamine oder die Dialkylaminoalkylamine. Diese primären Amine führen zur Bildung der in 6-Stellung mit dem entsprechenden Radikal substituier ten 6,7-Dihydro-5H-dibenz[c,
e]azepine. Die gebildeten Produkte können ihrerseits mit Säuren in ihre Salze oder mit quaternisieren- den Mitteln in ihre quaternären Salze umge wandelt werden. Wenn man als Ausgangsmaterial ein se kundäres Amin verwendet, erhält man die entsprechenden quaternären Salze des 6, 7-Di- hydro-5H-dibenz [c,e] azepins.
Das o,o' -bis - (Brommethyl) - biphenyl ist eine bekannte Verbindung, die von Kenner und Mitarbeitern im Journal of Chemical Society , London, Band 99 [1911], Seite 2108, beschrieben worden ist.
Gegenstand des vorliegenden Patentes ist nun ein Verfahren zur Herstellung von 6,7-Di- hydro-5H-dibenz [c,e] azepin, das dadurch ge kennzeichnet ist, dass man o,o'-bis-(Brom- methyl)-biphenyl mit. Ammoniak reagieren lässt.
Die Reaktion erfolgt nach folgendem Schema

<I>Beispiel:</I> 5,5 g o,o'-bis-(Brommethyl)-biphenyl wer den in 50 cm3 Äther gelöst. Man vermischt diese Lösung bei 20 C mit 50 cm3 10 % igem met.hanolischem Ammoniak.
Es erfolgt eine leichte Erhöhung der Temperatur, und nach ungefähr einer Stunde beginnen lange Pris- men auszukristallisieren. Nach dem Stehen über Nacht werden diese Kristalle, die aus dem Nebenprodukt 6(7H),6'(7'H)-Spiro-bi- (5H-dibenz[c,e]azepinium)bromid bestehen, abfiltriert. Die _Mutterlauge, die zur Haupt sache das gebildete 6,7-Dihydro-5H-dibenz- [c,e]azepin-hydrobromid enthält, wird zur Trockne eingeengt, worauf der kristallisierte Rückstand mit Äther und verdünntem Ammo niak geschüttelt wird,
wobei die letzten Reste des Nebenproduktes in die wässerige Lösung gehen. Die aus dem Hydrobromid freigesetzte Base bleibt in der Ätherlösung. Diese wird abgetrennt und getrocknet.
Zwecks Reinigung und Herstellung des I3y drobromids kann die Ätherlösung mit alko holischer Bromwasserstoffsäure neutralisiert werden. Das kristallisierte Hydrobromid wird abfiltriert und aus Alkohol umkristallisiert, worauf es bei 283-284 C schmilzt. 3 g des so erhaltenen Hydrobromids des 6,7-Dihydro- 5H-dibenz[c,e]azepins werden in 50 em3 Was- ser gelöst.
Ala.n setzt Ammoniak im Überschuss (etwa 20 cm3) zu, wobei die Base in Form eines Öls freigesetzt wird. Letzteres wird mit Äther extrahiert, und die Ätherlösung wird über Kaliumearbonat getrocknet. Der Äther wird auf dem Wasserbad entfernt. Das zu- riickbleibende viskose 01 wird im Vakuum de stilliert. Das gewonnene 6,7-Dihydro-5H-di- benz[c,e]azepin ist eine farblose zähe Flüssig keit vom Siedepunkt 185-l.90 C/13 mm.
