in welcher R einen niederen gesättigten oder un,esätt.igten Alkylrest, insbesondere den lIe- thylrest, bedeutet, beschrieben.
Es wurde nun gefunden, dass substituierte Tropasäure - N - (y-pieolyl) -amide der obigen Formel, worin R. einen Alkylrest mit 3 oder -1 Kohlenstoffatomen bedeutet, besonders kräf tige spasmolytisehe Eigenschaften zeigen.
Die neuen Verbindungen werden erfin dungsgemäss durch Kondensation eines Alkyl- (;,-picolyl)-amins, dessen Alkylrest 3 oder 4 Kohlenstoffatome enthält, mit. 'Tropasäure- ehlorid oder mit Aeetyltropasä.ureehlorid und gegebenenfalls naehfolgende Abspaltung der Aeetylgruppe hergestellt. Die erhaltenen Ver bindungen bilden mit Säuren ,Salze, zum Bei spiel Hydrochloride, Hydrobromide usw.
Die als Ausgangsmaterial benötigten Al kyl - y - picolylamine können beispielsweise dureh Umsetzung von 4-Halogenmethyl-pyri- din mit. einem entsprechenden Alkylamin oder dureh Umsetzung von Xi-(p-Toluolsulfonyl)-4- aminomethyl-pyridin mit einem entsprechen- den Alkylhalogenid oder dem entsprechenden Sulfat und nachfolgende Entacetylierung ge wonnen werden.
<I>Beispiel 1</I> Eine Lösung von 82 Gewichtsteilen y-Chlormethyl-pyridin-hydrochlorid in 80 Ge wichtsteilen Wasser wird unter Rühren in eine Mischung von 180 Gewichtsteilen Iso- propylamin und 50 ,Gewichtsteilen Wasser bei einer Temperatur von 5 bis 10 C eingetropft. Dann wird noch eine Stunde bei 20 C und eine Stunde bei 3,0 bis 3-5 C weitergerührt, gekühlt. und in der Kälte mit festem Kalium hydroxyd gesättigt.
Durch Extraktion mit Äther und nachfolgende Destillation wird Iso- propyl-(y-picolyl)-amin bei 120 C/2!0 mm er halten. Das Dihydrochlorid hat einen Schmelz punkt von 1,88 bis 190 C.
Zu einer llisehung von 41,2 Gewichts teilen Isopropyl-(y-picolyl)-amin und<B>27,5</B> Ge wichtsteilen trockenem Pyridin in 150 Ge- wichtsteil-en trockenem Chloroform wird rohes, aus 46 Gewichtsteilen Tropasäure erhaltenes Acetyltropasäixrechlorid unter Rühren und Eiswasserkühlung nach und. nach zugegeben. Zum Schluss wird noch 1 Stunde bei 25 C gerührt. Dann wird die Chloroformlösung mit 2.00 Gewichtsteilen Äther verdünnt und mit. 3n - Salzsäure geschüttelt.
Die schwach kongosaure Lösung wird 1 Stunde auf dem Dampfbad erwärmt wobei die Acetylgruppe des Reaktionsproduktes abgespalten wird. Es wird durch Kohle filtriert. und mit über schüssigem, konzentriertem Ammoniak ver setzt. Das ausgefallene Kondensationsprodukt wird in Chloroform aufgenommen, die Lösung getrocknet und das Chloroform destilliert.
Durch Umlösen des Rückstandes in Essigester erhält man das Tropasä1,ire-N-isopropyl-N- (y-picolyl)-amid als farblose Kristalle vom Schmelzpunkt 110-111p C. <I>Beispiel 2</I> Eine Lösung von 50 Gewichtsteilen y- Chlormethyl-pyridin-hydrochlorid in 50 Ge- wiehtsteilen Wasser wird unter Rühren in.

Die ölige Schicht wird abgetrennt, über Kal.ium- hydroxy d getrocknet und' destilliert, wobei das Biityl-(y-picolyl)-amin bei 139-140 C/ 23 mm übergeht.. Das Dihydrochlorid schmilzt bei 186-188 C.
Zu einer Mischung von 26,8 Gewichtstei len. Bntyl-(y-pieolyl)-amin und 17 Gewichts teilen trockenem Pyridin in 12O Gewichtstei len trockenem Chloroform wird. rohes, aus 28 Tropasäure erhaltenes Acetyltropasäiirechlorid unter Rühren und Eiswasserkühlung langsam zugegeben. Zum Sehluss wird noch 1 Stunde bei 25 C gerührt.
Dann wird die Chloroformlösung mit 200 Ge- wiehtsteilen Äther verdünnt und mit 3n-Salz- säure geschüttelt. Die schwach kongosaure Lö sung wird 1 Stunde auf dem Dampfbad er wärmt, um die Acetylgruppe des Reaktions produktes abzuspalten, durch Kohle filtriert Lind mit. überschüssigem, konzentriertem Am moniak versetzt. Das abgeschiedene Öl wird in Chloroform aufgenommen, die Lösung ge trocknet und das Chloroform destilliert.
Das Tropasäiire-.1;-butyl-(y-picolyl)-amid wird als Hydrochlorid isoliert und gereinigt. Schmelz punkt 127-129 C. <I>Beispiel 3</I> Eine Lösung von 50 Gewichtsteilen y-Chlor- methylpyridin-hydroehlorid in 50 Gewichts teilen Wasser wird in eine Mischung von 1'20 Gewichtsteilen n-Propylamin und 30 Ge wichtsteilen Wasser bei einer Temperatur von 5 biss 10 C eingetropft.
Dann wird noch 1 Stunde bei 2'5 C und 1 Stunde bei 3'5p C weitergerührt, gekühlt und in der Kälte mit Kaliumhydroxyd gesättigt. Durch Extraktion m *t Äther und nachfolgende Destillation wird das Propyl-(y-picolyl)-amin bei 115-116 C% 10 mm erhalten. Das Dihydrochlorid schmilzt. bei 186-188 C.
Zu einer Mischung von 28,1 Gewichts teilen Propyl-(y-picolyl)-amin und 19 Ge- wichtsteilen trockenem lr'y ridin in 120 CTe- wichtsteil'en trockenem Chloroform wird rohes, aus 32 Gewichtsteilen Tropasäure erhaltenes Acetyltropa-sällrechlorid unter Rühren und Eiswasserkühlung nach und nach zugegeben. Zum Schluss wird noch 2 Stunden bei 25 C gerührt.
Dann wird die Chloroformlösung mit 200 Gewichtsteilen Äther verdünnt und mit 3n-Salzsäure geschüttelt. Die schwach kongosaure Lösung wird 1 Stunde auf dem Dampfbad erwärmt, wobei die Acetylgruppe des Kondensationsproduktes abgespalten wird.
