Verfahren zur Darstellung von Abkömmlingen der 4-Arylpiperidin-4-carbonsäuren
Gegenstmld des Patents 679281 ist u. a.
.ein Verfahren zur Darstellung von, in i-Stel-
lung durch leinen Kohlenwasserstoffrest substi-
tuierten 4-Arylpip@eridin-4-carboinsäuren und
ihren Abkömmlingen, die durch Umsietzung
von Arylacetonitrilen mit tertiären, zwei
ß-Halogenalkylreste,enthaltonden Aminen mit-
tels geeigneter Kondensationsmittel zunächst
als Nitrile gewonnen werden. Die Herstel-
lung von derartigen, in i-Stellung nicht `sub-
stitulerten Verbinduingen ist indessen insofeM
mit gewissen Schwierigkeiten verbuinden, als
die als Ausgangsstoffe erforderlichen Di-
(ß-chloralkyl)-amine mit sekundärem Stick-
stoff als freie Basen äußerst ,unbeständig sind.
Es wurde nun gefunden, daß sich Acyldi-
(ß-halogenalkyl)-arnide, insbesondere Aryl-
sulfondi-(ß-halogehalkyl)-amide, in einfacher
Weise mit Arylacetonitrilen unter Verwendung
eines die Abspaltung von Halogenwasserstoff
bewirkenden Mittels zu 'i-Acyl-, besolnders
x -Arylsulfon - 4, - arylpiperidin- 4-;.carboln,säurei
nitrilen umsetzten lassen. Diese können durch
Versteifung rund Veresterung leicht in thera-
pleutisch wertvolle Ester der 4-Arylpiperidin-
4-carbonsäunen übergeführt werden.
Beispiele
i. p-Toluolsulfoudi-(ß-oxyäthyl)-amid, das
bei der Umsietzung von Diäthalnolamin und
Natriumcarbonat, gelöst in Wasser, mit
p-Toluolsulfochlorid in guter Ausbeute @ent-
s'teht und ,aus Wasser in Blättchen vom
F. i o i bis i o2' kristallisiert, wird durch Ein-
wirkung von überschüssigem Thionylchlorid,
zuletzt bei t3o°, in glatter Umsetzung ün
p-Toluolslulfondi-(ß-chloräthyl)-ainid Übierge-
führt, farblose Kristalle (aus Methanol) vom
F. 48 bis 49°, unlöslich in Wasser, leicht
löslich in Äther, Benzol und Aceton. Eine
Lösung von 296 Gewichtsteilen p-Toluol-
sulfondi-(ß-chloräthyl),amid und 117 g Beinzyl-
cyanid in 750 Gewichtsteilein Toluol wird
unter Rühren mit insgesamt 8o Gewichts-
teilen fein gepulvertem Natriumamid in An-
teilen -von etwa je 1/8 der Gesamtmenge versetzt. Man hält die
Temperatur des Gemisches durch Kühlung auf etwa 4o bis 45°. Wenn alles eingetragen
ist und keine Umsetzungswärme mehr auftritt, erwärmt m `@ langsam bis zum Sieden
:des Toluols kocht i Stunde unter Rückfiuß. Nach dein Erkalten wird Wasser zur Lösung
des Natriumchlorids zugesetzt und das Toluol mit Wasserdampf abdestilliert. Nach
dem Erkalten -wird der zähflüssige braune Rückstand von der wäßrigen Salzlösung
getrennt und in Methylalkohol durch Erwärmen gelöst. Es scheiden sich farblose Kristalle
ab, die nach dein Abkühlen abgesaugt, mit Methylalkohol gewaschen werden und dann
für die weitere Verarbeitung genügend rein sind. F. Zoo bis 2oi°. Das so -erhaltene
i-p-Toluolsulfo;n-¢-phenylpiperidiln - 4 - carbonsäurenitril ist unlöslich in Wasser;
Natronlauge, Salzsäure, sehr wenig löslich in Methylalkohol; umkristallisierbar
aus Benzol, Essigester, Eisessig und sehr leicht löslich in Chloroform. In gleicher
Weise läßt sich aus Diäthanolamin und B.enzolsulfochlorid das i-Benzolsulfo,ndi-(ß-oxyäthyl)-amid
und daraus mit Thionylchlorid. das i-Benzolsulfondi(-ß-clüoräthyl)-,amid herstellen,
das bei 4 mm um 200° siedet und zu einer kristallinischen Mässe vom F. 34° erstarrt.
Dieses gibt, mit B,enzylcyauid in Toluol .in Gegenwart von Natriumamid umgesetzt,
das i-BenzoisulfOn-q.-phenylpip@eridin-4-carbonsäureanitril, farblose Kristalle

Beide Nitrile können, wenn man sie mit dem il/3faclien ihres Gewichtes
an 75,D/oiger Schwefelsäure vorsichtig auf 14o bis i 5o' erhitzt und diese Temperatur
während i Stunde beibehält, in glatter Umsetzung zur 4-Phenylpiperidin-4-carbo@usäüre
verseift werden. Zwecks überführung in -den Äthylester braucht man die Säure aus
der schwefelsauren Lösung gar -nicht "erst abzuscheiden, sondern kann durch diese
bei etwa i i o° so lange Äthylalkohol leiten, bis das jeweilige Destillat wasserfrei
wird. Der so entstandene 4-Phenylpiperidin-4-Carbonsäureäthylester wird durch Zugabe
von verdünnter Natronlauge runter Kühlung abgeschieden, in Äther aufgenommen und
über Kaliumcarbonat getrocknet. Er siedet unter 3 mm Druck bei i 5o' als farbloses
Öl über und erstarrt völlig zu einer farblosen, an der Luft CO2 anziehenden Kristallmasse.
Erstarrungspunkt 33°. Hydrochlorid, aus Essigester kristallisiert, F.133 bis 134°.
Pikrat, aus Methyl alkohol kristallisiert, F.157 bis 158'. z. B,enzöesäuredi-(ß-chlorätliyl)-amid
wird durch Umsetzung von Di-(ß-chloräthyl)-aminhydrochlorid; Benzoylchlorid und
Natronlauge, Aufnehmen .des Umsetzungsgutes in .'.,.: Äther, Trocknen über Kaliumcarbonat
und Verdampfen des Äthers dargestellt. Es ist schwach bräunliche Flüssigkeit, die,
in keiner Menge destilliert, unter 2 mm Druck bei 182° übergeht, dabei ;aber zum
Teil eine Umlegierung erfährt. In eine Mischung von 246 Gewichtsteilen Benzo,esäuredi-(ß-chloräthyl)-amid,
117 Gewichtsteilen Benzylcyanid und 6oo Gewichtsteilen Toluol werden unter Rühren.
und pessiender Kühlung 8o Gewichtsteile pulverisiertes Natriumamid in Anteilen von,
etwa je 1/1o der Menge eingetragen, wobei man die Temperatur des Gemisches zwischen
35 und 40° hält. Darauf erhitzt man langsam bis iooa und verrührt das Gemisch bei
dieser Temperätur noch i bis 2 Stunden. Nach Odem Erkalten wird die Toluollösungg
mit Wasser ausgeschüttelt und das Toluol unter vermindertern. Drück abdestilliert.
Der Rückstand bildet sein zähes; Junkelbraunes Öl, aus de reines i-Benzoyl-4-phenylpiperidin-4-carbonsäurenitril
nicht auf einfache Weise zu gewinnen ist. Er wird mit der il/2fachen Gewichtsmenge
70%iger Schwefelsäure 2 Stunden unter Rückfluß gekocht. Alsdann leitet man durch
das Gemisch bei zio° so langte Äthylalkohol, bis: das jeweilige Destillat wasserfrei
ist. Nun gießt man ,das Gemisch unter Kühlung in Wasser, -entfernt die nicht in
Lösung gegangenen nichtbasischen Anteile, wie den Bien#2oesäuneäthylester, durch
Ausäthern und fällt aus der sauren, wäßrigein Lösung .durch Übersättigen mit Natronlauge
die basischen Bestandteile. Nach Aufnehmen in Äther und Trocknen über Kalium carbonat
wird der 4-Ph@enylpipieridin-4-earbo!nsäureäthylester durch mehrmaliges Fraktionieren
im Vakuum gereinigt.
