Verfahren zur Herstellung basischer Thymoldimethylaminoäthyläther
Es wurde gefunden, daß man dadurch zu bisher nicht bekannten, therapeutisch sehr
wertvoIlen Produkten, deren wasserlösliche Salze hervorragende sympathicolytische
Eigenschaften besitzen, gelangt, wenn man in Thymol in der p-Stellung zur Phenolhydroxylgruppe
eine Acetaminogruppe einführt, die Phenolhydroxylgruppe mit der Dimethylaminoäthylgruppe
veräthert, die Acetaminogruppe durch die Hydroxylgruppe ersetzt und letztere, gegebenenfalls
durch Einführung der Methylgruppe, veräthert oder acetyliert. Die genannten Substanzen
können auch dargestellt werden dadurch, daß manThymolabkömmlinge, deren p-Stellung
zur Phenolhydroxylgruppe durch t Halogenatom besetzt ist, in die Dimethylaminoäthyläther
überführt. Es sind basische Phenoläther beschrieben worden, die in o-Stellung zur
Phenolhydroxylgruppe eine Isopropylgruppe und zu dieser in p-Stellung eine Methylgruppe
haben. Diese unterscheiden sich also von den neuen Verbindungen dadurch, daß die
p-Stellung zur Phenolhydroxylgruppe nicht substituiert ist. Weiter sind solche Verbindungen
beschrieben worden, die neben phenolischen Hydroxylgruppen noch ungesättigte Alkylreste
im Molekül enthalten, von denen mindestens einer in o-Stellung zur Phenolhydroxylgruppe
substituiert sein muß und die nicht Abkömmlinge N-acetylierter Aminophenole sind.
Auch bei diesen Verbindungen ist in p-Stellung zur zu veräthernden Hydroxylgruppe
kein Substituent eingeführt.
Während diese bereits bekannten Produkte
eine sekaleähnliche, das heißt eine auf den Uterus gerichtete Wirkung zeigen, fehlt
den neuen Substanzen in vergleichbaren Dosen diese Wirkung ganz. Dafür tritt die
sympathikolytische Wirkung außerordentlich stark in den Vordergrund. Man hat daher
den Vorteil, daß man gerade diese therapeutische Wirkung besonders zur Geltung bringen
kann, ohne die unangenehmen Nebenerscheinungen durch die Wirkung auf den Uterus.Process for the preparation of basic thymoldimethylaminoethyl ether

g Natriumacetat, 107 g Eisessig und 8o g Essigsäureanhvdrid vermischt. Hierbei
wird 4-Acetaminothymol (Plancher, Gazzetta Chimica Italiana, 25, 1I, S.388) erhalten.
Davon werden 156 Gewichtsteile in 6oo ccm Alkohol zu einer Auflösung von 17,6 Gewichtsteilen
Natrium in 6oo ccm Alkohol gegeben und mit 82 g Dimethylaminoäthylchlorid einige
Zeit rückfließend gekocht. Das Reaktionsgut wird mit Wasser versetzt, mit Salzsäure
kongosauer eingestellt und der Alkohol im Vakuum abdestilliert. Die durch Alkalien
frei gemachte Base wird in Äther aufgenommen. Nach Eindampfen der Ätherlösung erhält
man den Dimethylaminoäthyläther des 4-Acetaminothymols als braungelbes Öl, das nach

i,i9) und 400 ccm Wasser werden ioo g der erhaltenen Base eingetragen und i Stunde
unter Rückfluß gekocht. Darauf wird alkalisch gemacht, mit Äther extrahiert und
der Äther abdestilliert. 23,6 g des so erhaltenen 4-Aminothymoxyäthyldimethylamins
in schwefelsaurer Lösung werden bei Temperaturen nicht über o° mit einer Auflösung
von 7,2 g Natriumnitrit in 70 ccm Wasser diazotiert und das Diazotat nach
Zusatz von i g Kupfersulfat zum Sieden erhitzt, bis die eingetretene Gasentwicklung
beendet ist. Darauf wird alkalisch gemacht und Kohlendioxyd eingeleitet. Die Base
fällt erst ölig aus, wird dann bald kristallin. Das so erhaltene 4-Oxythymoxyäthyldimethylamin
bildet ein in Wasser mit neutraler Reaktion leicht lösliches Hydrochlorid vom F.
i74 bis 175,5°. Analyse: o,i46o g = 5,35 ccm n/io AgN 03 = 13 0/, Cl (her. 12,99
Beispiel 2 4-Acetoxythymoxyäthyldimethylaminhydrochlorid 56,8 g Oxythymoxyäthyldimethylamin
werden mit 16o ccm Essigsäureanhydrid und 17,5 ccm Pyridin i Stunde auf dem kochenden
Wasserbad gehalten. Danach wird mit Wasser versetzt, alkalisch gemacht, die Base
mit Äther extrahiert und der Äther abdestilliert. Die Base bildet mit Säuren gut
kristallisierende in Wasser leicht lösliche Salze. Das Hydrochlorid hat den F. 2o8
bis 2io°. 759 Oxythymoxyäthyldimethylamin werden in iooo ccm Äther gelöst und mit einer
Lösung von 13,3 g Diazomethan in 4000 ccm Äther versetzt und bei Zimmertemperatur
2o Stunden stehengelassen, bis die eingetretene Gasentwicklung nachläßt. Darauf
wird bis zum Sieden erhitzt und etwa 5 Stunden rückfließend gekocht. Nach Abdestillieren
des Äthers wird der Rückstand in verdünnter Salzsäure gelöst, die Base durch Alkali
frei gemacht und ausgeäthert. Aus der Ätherlösung wird mit isopropanolischer Salzsäure
das Hydrochlorid gefällt. Nach Umkristallisieren aus Isopropanol erhält man in Wasser
mit neutraler Reaktion leicht lösliche Kristalle vom F. 173 bis 175°. Analyse: 0,1708

von Zoo g Thymol mit Zoo g Sulfurylchlorid wird 4-Chlorthymol (B o c ch i , Gazzetta
Chimica Italiana, 26, 1I, S. 403) erhalten. Eine Lösung von 184,5 Gewichtsteilen
4-Chlorthymol in 500 ccm Alkohol werden zu einer Auflösung von 23 g Natrium
in iooo ccm Alkohol gegeben und mit 11o g Dimethylaminoäthylchlorid einige Zeit
rückfließend gekocht. Das Reaktionsgut wird mit Wasser versetzt, mit Salzsäure kongosauer
eingestellt und der Alkohol im Vakuum abdestilliert. Die durch Alkalien frei gemachte
Base wird in Ätheraufgenommen. Nach Eindampfen derÄtherlösung erhält man den Dimethylaminoäthyläther
des 4-Chlorthymols, dessen Hydrochlorid nach Umkristallisieren aus Isopropanol den
F. 168 bis i7o° zeigt und in Wasser mit schwach saurer Reaktion leicht löslich ist.
