Relative compounds are monohaloacetic acids, especially monochloroacetic acid, and their derivatives formed by functional modification of the carboxyl group, in particular their acid halides, anhydrides, esters, amides, anilides and imidoethers. Example 1:
44.2 g of o-amino-N- (p-chloro-benzyl) -aniline are refluxed for 3 hours with 19 cubic meters of 5N hydrochloric acid and 27 g of monochloroacetic acid. After standing for several hours, the aqueous phase above the oil is drawn off and the remaining oil is triturated with acetone, the substance solidifying. It is filtered off with suction and washed with acetone. The crude 1-p-chloro-benzyl-2-chloromethyl-benzimidazole-chlorohydrate is dried in a vacuum desiccator.
The base freed from this with sodium bicarbonate (F. 99 to 101 -from ligroin) is further processed directly as a raw product.
24.7 g of crude 1-p-chloro @ benzyl-2-chloromethyl = benzimidazole are suspended in 60 cubic meters of dry benzene, and 60 g of 32% benzene dimethylamine solution are added while cooling with ice water. It is left to stand in the cold for an hour and then heated to 50 for two hours.
After standing overnight, the benzene is distilled off in order to remove the excess dimethylamine. The residue is taken up in benzene and water and the benzene phase is separated off, which is washed again with water. For cleaning with the monohydrochloride, the benzene solution is extracted fractionally with hydrochloric acid (until it changes from dimethyl yellow to red).
The hydrochloride of> 1, 'ps-chloro-benzyl-2-dimethyl-a.minomethyl-benzimidazoles is obtained by evaporating the aqueous extracts and purifying it by recrystallization from butanol. F. 202.5 to 203.5.
Example 2: 4.8 g of Np-chloro-benzyl-o-phenylenediamine are mixed with 3.1 g of dimethylamino-acetic acid hydrochloride (produced by condensation of dimethylamine with monochloroacetic acid), 8 cm3 of water, 2 cm3 Alcohol,
2 cm3 conc. Hydrochloric acid and 2 cm3 phosphoric acid syrup in a small flask under nitrogen protection in an oil bath with slowly increasing temperature heated to 140 (short riser pipe). The solvents evaporate in the process. Duration two hours. The residue is taken up in hot water and filtered over charcoal. The base is then released with caustic soda and extracted with ether.
For purification, the base is converted into the hydrochloride and this is recrystallized from butanol. The 1-p-chloro-benzyl-2-dimethylaminomethyl-benzimidazole hydrochloride obtained in this way melts at 200 to 202.
Example 3: 8 g of o-amino-Np-chloro-benzyl-aniline and 2.8 g of pyridine are dissolved in dry ether and mixed with an ethereal solution of 3.9 g of chloroacetyl chloride while cooling with ice = Common salt mixture implemented. There are 8 g of N-p-chlorobenzyl-N'-chloroacetyl-o-phenylene .. diam- obtained, which can be processed as a crude product and has an F. of 130 slightly colored.
8 g of crude N-p-chloro-benzyl-N'-chloroacetyl-o-phenylenediamine are heated to the boil with 3.7 g of dimethylamine, dissolved in benzene, and a further 50 cm3 of benzene. The reaction mixture is washed with water in a separating funnel. It is then evaporated and the basic residue is heated to boiling for two hours with 50 cm3 of glacial acetic acid and 5 g of sodium acetate. After cooling, it is diluted with water and the solution is neutralized with sodium hydroxide solution.
The precipitated base is extracted with ether. For the purpose of purification with hydrochloric acid, the crude base is converted into the monohydrochloride of 1-p-chloro-benzyl-2 = dimethylaminomethyl-benzimidazole and this is then crystallized from butanol. F. 202 to 2040.
