mit einem entsprechenden Säureanhydrid,
Säure chlorid oder Mischungen derselben umsetzt.
Das Verfahren wird beispielsweise so ausgeführt, dass 5,7-Dihalogen-8-oxychinaldine mit einem Säure anhydrid in Gegenwart eines katalytischen Zusatzes, z. B. konz. Schwefelsäure oder Pyridin, auf höhere Temperatur, vorzugsweise 130-160', erhitzt werden. Die Reaktionslösung wird sodann direkt auf Eis gegossen oder zuerst das überschüssige Acylierungs- mittel teilweise abdestilliert und erst dann auf Eis gegossen. Nach dem Abnutschen und Nachwaschen mit Wasser erhält man die entsprechenden Ester.
Ist das für die Acylierung der 5,7-Dihalogen-8-oxy- chinaldine verwendete Acylierungsmittel ein Säure chlorid oder eine Mischung von Säurechlorid und Säureanhydrid, dann gestaltet sich der Umsatz so, dass die 5,7-Dihalogen-8-oxychinaldine mit dem Acylhalogenid zusammen in einem inerten organi schen Lösungsmittel, z. B. Benzol, Toluol, Xylol und Pyridin, gelöst werden.
Die Lösung der Reak tionspartner wird bei Zimmertemperatur undjoder bei erhöhter Temperatur, z. B. in einem Dampfbad, stehengelassen; schliesslich werden die gebildeten Ester nach bekannten Methoden isoliert und gerei nigt.
Die nach dem vorliegenden Verfahren hergestell ten, bisher unbekannten Ester der 5,7-Dihalogen-8- oxychinaldine sind feste Basen. Sie sind schlechter wasserlöslich als die 5,7-Dihalogen-8-oxychinaldine und werden aus diesem Grunde schlechter als diese resorbiert. Sie zeichnen sich durch therapeutisch ver wertbare pharmakologische Eigenschaften aus, indem sie eine erhöhte Wirksamkeit gegen pathogene Keime und auch andere Krankheitserreger aufweisen.
Die neuen Verbindungen sollen in der Therapie als Ober flächen-Antiseptika verwendet werden, wobei ihre schlechte Wasserlöslichkeit resp. Resorbierbarkeit eine besonders gute System-Verträglichkeit gewähr leistet. Sie dienen ferner als Zwischenprodukte zur Herstellung von Medikamenten. In den nachfolgenden Beispielen sind die Schmelzpunkte in Celsiusgraden und unkorrigiert angegeben.
<I>Beispiel 1</I> 5,7-Dichlor-8-acetoxychinaldin Eine Mischung von 20,0 g 5,7-Dichlor-8-oxy- chinaldin (Smp. 110112 , hergestellt nach dem USA-Patent Nr. 2 411<B>670</B> der Firma Geigy) und 75,0 g Essigsäureanhydrid mit 8 Tropfen konz. Schwefelsäure werden während 3 Stunden in einem Ölbad von 160' erhitzt.
Nach dem Abkühlen wird der gesamte Ansatz auf 500g Eis gegossen, und nach vollständigem Schmelzen des Eises wird das ausgefallene Material abgenutscht und dann mit Eis wasser nachgewaschen, bis das Waschwasser nicht mehr lackmussauer reagiert. Nach dem Trocknen des Rohproduktes in einem Vakuumtrockenschrank bei 8011 bis zur Gewichtskonstanz wird dieses aus Leichtbenzin umkristallisiert, und man erhält das analysenreine 5,7-Dichlor-8-acetoxychinaldin mit dem konstanten Smp. 103,5-105 .
<I>Beispiel 2</I> 5,7-Dibrom-8-acetoxychinaldin a) Eine Mischung von 40 g 5,7-Dibrom-8-oxy- chinaldin (Smp. 125-126 , hergestellt nach dem USA-Patent Nr. 2 411 670 der Firma Geigy) und <B>100,0</B> g Essigsäureanhydrid mit 8 Tropfen konz. Schwefelsäure werden während 3 Stunden in einem Ölbad von 160 erhitzt.
Nach dem Abkühlen wird der gesamte Ansatz auf 500 g Eis gegossen, und nach vollständigem Schmelzen des Eises wird das ausgefallene Material abgenutscht und dann mit Eis wasser nachgewaschen, bis das Waschwasser nicht mehr lackmussauer reagiert. Nach dem Trocknen des Rohproduktes in einem Vakuumtrockenschrank bei 70b bis zur Gewichtskonstanz wird dieses aus Tetrachlorkohlenstoff umkristallisiert, und man er hält das analysenreine 5,7-Dibrom-8-acetoxychinaldin mit dem konstanten Smp. 125-127b.
b) Zu einer Lösung von 47,5 g 5,7-Dibrom-8- oxychinaldin (Smp. 125-126b, hergestellt nach dem USA-Patent Nr. 2 411<B>670</B> der Firma Geigy) in 100 cm3 Pyridin werden unter Kühlen in Eis portio- nenweise 17,7 g Acetylchlorid (Sdp. 51-52b) unter Umschwenken hinzugegeben. Nachdem der Ansatz bei Zimmertemperatur 3 Stunden stehengelassen wurde, wird im Wasserstrahlvakuum bei 400 zur Trockne eingeengt.
Der Rückstand wird mit 100 cm3 Eiswasser angeteigt, abgenutscht und mit Eiswasser nachgewaschen, bis das Waschwasser nicht mehr lackmussauer reagiert. Nach dem Trocknen des Roh produktes in einem Vakuumtrockenschrank bei 809 bis zur Gewichtskonstanz wird dieses aus Tetrachlor- kohlenstoff umkristallisiert, und man erhält das ana lysenreine 5,
7-Dibrom-8-acetoxychinaldin mit dem konstanten Smp. 125-127b. <I>Beispiel 3</I> 5-Chlor-7-brom-8-acetoxychinaldin Eine Mischung von 40 g 5-Chlor-7-brom-8-oxy- chinaldin (Smp. 113-114b, hergestellt aus 5-Chlor-8- oxychinaldin nach dem Schweizer Patent Nr. 235224 der Firma Geigy) und 135g Essigsäureanhydrid mit 4 Tropfen konz. Schwefelsäure wird während 3 Stun den in einem Ölbad von<B>1600</B> erhitzt.
Nach dem Abkühlen wird der gesamte Ansatz auf 500 g Eis gegossen, und nach vollständigem Schmelzen des Eises wird das ausgefallene Material abgenutscht und dann mit Eiswasser nachgewaschen, bis das Waschwasser nicht mehr lackmussauer reagiert. Nach dem Trocknen des Rohproduktes in einem Vakuumtrockenschrank bei 809 bis zur Gewichts konstanz wird dieses aus Leichtbenzin umkristalli siert, und man erhält das analysenreine 5-Chlor-7- brom-8-acetoxychinaldin mit dem konstanten Smp. 121-122 .
<I>Beispiel 4</I> 5,7-Dibrom-8-benzoxychinaldin Zu einer Lösung von 47,5 g 5,7-Dibrom-8-oxy- chinaldin (Smp. 125-126 , hergestellt nach dem USA-Patent Nr. 2 411<B>670</B> der Firma Geigy) in 100 cm3 Pyridin werden unter Kühlen in Eis portio- nenweise 31,6 g Benzoylchlorid (Sdp. 194-196e) unter Umschwenken hinzugegeben.
Nachdem der Ansatz bei Zimmertemperatur 3 Stunden stehen gelassen wurde, wird im Wasserstrahlvakuum bei 40b zur Trockne eingeengt. Der Rückstand wird mit 100 cm3 Eiswasser angeteigt, dann 200 cm 3 Chloro form zugesetzt und der Chloroformteil zweimal mit je 100 cm3, total 200 cm3 einer l0 0h igen wässe rigen Natriumbicarbonatlösung ausgeschüttelt.
Der zweimal mit je 25 cm3, total 50 cm 3 Wasser nach gewaschene Chloroformteil wird über Natriumsulfat getrocknet, im Wasserstrahlvakuum eingeengt, worauf man das rohe 5,7-Dibrom-8-benzoxychinaldin vom Smp. 124-130b erhält. Nach wiederholtem Um kristallisieren der Substanz aus Benzol,Leichtbenzin erhält man das analysenreine 5,7-Dibrom-8-benzoxy- chinaldin mit dem konstanten Smp. 130-132b.
<I>Beispiel 5</I> 5,7-Dibrom-8-benzoxychinaldin Zu einer Suspension von 63,4 g 5,7-Dibrom-8- oxychinaldin (Smp. 125-1266), 15,8 g Pyridin und 300 cm3 Tetrachlorkohlenstoff werden bei Raum temperatur 28,1 g Benzoylchlorid unter Rühren hinzugetropft. Der Ansatz wird hierauf bei einer Innentemperatur von<B>50,1</B> während 61j. Stunden ge rührt.
Nach dem Abkühlen auf Raumtemperatur wird das gebildete Pyridinhydrochlorid abgenutscht, mit 100 cm3 Benzol nachgewaschen und das Filtrat im Vakuum zur Trockne eingeengt. Das rohe 5,7- Dibrom-8-benzoxychinaldin hat einen Smp. von 120 bis 12611. Nach wiederholtem Umkristallisieren der Substanz aus Tetrachlorkohlenstoff erhält man das analysenreine 5,
7-Dibrom-8-benzoxychinaldin mit dem konstanten Smp. 130-132 . <I>Beispiel 6</I> 5,7-Dijod-8-acetoxychinaldin Zu einer Lösung von 20,5 g 5,7-Dijod-8-oxy- chinaldin (Smp. 144-145b) in 20 cm3 Pyridin wer den 50,0 g Acetanhydrid hinzugegeben und der An satz bei einer Ölbadtemperatur von 1609 5 Stunden unter Rückfluss erhitzt. Dann wird im Vakuum bei 60 zur Trockne eingeengt.
Der Rückstand wird mit 50 cm3 Eiswasser angerieben, dann 200 cm3 Chloro form zugesetzt und der Chloroformteil zweimal mit je 100 cm3 einer 5'1hIgen wässerigen Natriumbi- carbonatlösung ausgeschüttelt. Der zweimal mit je 50 cm3 Wasser nachgewaschene Chloroformteil wird über Natriumsulfat getrocknet, im Vakuum eingeengt, worauf man das rohe ölige 5,7-Dijod-8-acetoxy- chinaldin erhält.
Nach wiederholtem Umkristallisie- ren der Substanz aus Leichtbenzin erhält man das analysenreine 5,7-Dijod-8-acetoxychinaldin mit dem konstanten Smp. 120-122 .
<I>Beispiel 7</I> 5,7-Dichlor-8-benzoxychinaldin Zu einer Lösung von 34,2 g 5,7-Dichlor-8-oxy- chinaldin (Smp. 111-112 ) in 100 cm3 Pyridin wer den unter Kühlen mit Eis portionenweise 32,7 g Benzoylchlorid unter Umschwenken hinzugegeben. Nachdem der Ansatz bei Zimmertemperatur 4 Stun den stehengelassen wurde, wird im Vakuum bei 40 zur Trockne eingeengt.
Der Rückstand wird mit 100 cm3 Eiswasser angerieben, dann 250 cm3 Chloroform zugesetzt und der Chloroformteil zwei mal mit je 100 cm3 einer 511/eigen wässerigen Na- triumbicarbonatlösung ausgeschüttelt. Der zweimal mit je 25 cm- Wasser nachgewaschene Chloroform teil wird über Natriumsulfat getrocknet und im Va kuum eingeengt, worauf man das rohe 5,7-Dichlor- 8-benzoxychinaldin vom Smp. 118-121 erhält.
Nach wiederholtem Umkristallisieren der Substanz aus Leichtbenzin erhält man das analysenreine 5,7-Di- chlor - 8 - benzoxychinaldin mit dem konstanten Sm_p. 121-1231.
<I>Beispiel 8</I> 5,7-Dibrom-8-(p-chlorbenzoxy)-chinaldin Zu einer Suspension von 31,7 g 5,7-Dibrom-8- oxychinaldin (Smp. 125-126a), 8 cm-' Pyridin und 160 cm3 Äthylenchlorid werden bei Raumtemperatur 19,2 g p-Chlorbenzoylchlorid (Sdp. 97-100 112 mm Hg) unter Rühren hinzugetropft. Der Ansatz wird hierauf bei einer Badtemperatur von 50 während 4 Stunden gerührt.
Nach dem Abkühlen auf Raum temperatur wird das gebildete Pyridinhydrochlorid abgenutscht, mit 100 cm-' Benzol nachgewaschen und das Filtrat im Vakuum zur Trockne eingeengt. Das rohe 5,7-Dibrom-8-(p-chlorbenzoxy)-chinaldin hat einen Smp. von 140-145 . Nach wiederholtem Umkristallisieren der Substanz aus Tetrachlorkohlen- stoff, dann aus Benzol erhält man das analysenreine 5,7-Dibrom-8-(p-chlorbenzoxy)-chinaldin mit dem konstanten Smp. 148-150 .
<I>Beispiel 9</I> 5,7-Dibrom-8-(p-nitrobenzoxy)-chinaldin Eine Suspension von 31,7 g 5,7-Dibrom-8-oxy- chinaldin (Smp. 125-126 ), 8 cm3 Pyridin, 350 cm3 Äthylenchlorid und 18,6 g p-Nitrobenzoyl-chlorid (Smp. 72 ) wird 4 Stunden bei einer Badtemperatur von 50 gerührt.
Nach dem Stehen über Nacht wird anderntags das gebildete Pyridinhydrochlorid ab- genutscht, mit 100 cm3 Benzol nachgewaschen und das Filtrat im Vakuum zur Trockne eingeengt. Das rohe 5,7-Dibrom-8-(p-nitrobenzoxy)-chinaldin hat einen Smp. von 156-160 .
Nach wiederholtem Um kristallisieren der Substanz aus Tetrachlorkohlen- stoff erhält man das analysenreine 5,7-Dibrom-8- (p - nitrobenzoxy) - chinaldin mit dem konstanten Smp. 160,5-162<B>0</B>.
<I>Beispiel 10</I> 5-Chlor-7-brom-8-benzoxychinaldin Zu einer Suspension von 40,8 g 5-Chlor-7-brom- 8-oxychinaldin (Smp. 113-114 ), 11,8 g Pyridin und 500 cm3 Tetrachlorkohlenstoff werden bei Raum temperatur 22,1g Benzoylchlorid (Sdp. 194-196 ) unter Rühren hinzugetropft. Der Ansatz wird hierauf bei einer Innentemperatur von 50 während 4 Stun den gerührt.
Nach dem Abkühlen auf Raumtempe ratur wird das gebildete Pyridinhydrochlorid abge- nutscht, mit 100 ein- Benzol nachgewaschen und das Filtrat im Wasserstrahlvakuum zur Trockne ein geengt. Das rohe 5-Chlor-7-brom-8-benzoxychinaldin hat einen Smp. von 121-123 . Nach Umkristallisie- ren der Substanz aus Leichtbenzin erhält man das analysenreine 5 - Chlor - 7 - brom-8-benzoxychinaldin mit dem konstanten Smp. 122-124 .
<I>Beispiel 11</I> 5-Chlor-7-brom-8-(p-nitrobenzoxy)-chinaldin Eine Suspension von 40,8 g 5-Chlor-7-brom-8- oxychinaldin (Smp. 113-114 ), 12 cm3 Pyridin, 1000 cm3 Tetrachlorkohlenstoff und 29,3 g p-Nitro- benzoylchlorid (Smp. 72 ) wird 4 Stunden bei. einer Innentemperatur von 50 gerührt.
Nach dem Ab kühlen auf Zimmertemperatur wird das gebildete Pyridinhydrochlorid abgenutscht, mit 100 cm3 Benzol nachgewaschen und das Filtrat im Wasserstrahl vakuum zur Trockne eingeengt. Das rohe 5-Chlor-7- brom-8-(p-nitrobenzoxy)-chinaldin hat einen Smp. von 148-153 . Nach wiederholtem Umkristallisieren der Substanz aus Tetrachlorkohlenstoff erhält man das analysenreine 5-Chlor-7-brom-8-(p-nitrobenzoxy)- chinaldin mit dem konstanten Smp. 155-157 .
<I>Beispiel 12</I> 5,7-Dichlor-8-(p-chlorbenzoxy)-chinaldin Zu einer Suspension von 22,8 g 5,7 Dichlor-8- oxychitialdin (Smp. 111-112 ), 8,0 g Pyridin und 150 cm3 Benzol werden bei Raumtemperatur 19,3 g p-Chlorbenzoylchlorid (Sdp. 97-100 112 mm Hg) in 50 cm3 Benzol unter Rühren hinzugetropft. Der An satz wird hierauf bei einer Badtemperatur von 50 während 4 Stunden gerührt.
Nach dem Abkühlen auf Raumtemperatur wird das gebildete Pyridin- hydrochlorid abgenutscht, mit 100 cm3 Benzol nach gewaschen und das Filtrat im Wasserstrahlvakuum zur Trockne eingeengt. Das rohe 5,7-Dichlor-8-(p- chlorbenzoxy)-chinaldin hat einen Smp. von 139 bis l41 . Nach dem Umkristallisieren der Substanz aus Tetrachlorkohlenstoff erhält man das analysenreine 5,7-Dichlor-8-(p-chlorbenzoxy)-chinaldin mit dem konstanten Smp. 139-141 .
<I>Beispiel 13</I> 5,7-Dichlor-8-(p-nitrobenzoxy)-chinaldin Eine Suspension von 12,8 g 5,7-Dichlor-8-oxy- chinaldin (Smp. 111-112 ), 4,5 g Pyridin, 150 cm- Äthylenchlorid und 10,9 g p - Nitrobenzoylchlorid (Smp. 721) wird 4 Stunden bei einer Badtemperatur von 501 gerührt. Nach dem Abkühlen auf Zimmer temperatur wird das gebildete Pyridinhydrochlorid abgenutscht,
mit 100 em3 Benzol nachgewaschen und das Filtrat im Wasserstrahlvakuum zur Trockne eingeengt. Das rohe 5,7-Dichlor-8-(p-nitrobenzoxy)- chinaldin hat einen Smp. von 158-162 . Nach wie derholtem Umkristallisieren der Substanz aus Tetra chlorkohlenstoff erhält man das analysenreine 5,7- Dichlor-8-(p-nitrobenzoxy)-chinaldin mit dem kon stanten Sinp. 163-1641.
<I>Beispiel 14</I> 3-Methyl-5,7-dibrom-8-acetoxychinaldin a) 3-Methyl-5,7-dibrom-8-hydroxychinaldin. Eine Lösung von 10,0 g 3-Methyl-8-hydroxy- chinaldin (Smp. 91-921, hergestellt durch Kondensa tion von o-Aminophenol mit a-Methylcrotonaldehyd) in 60 cm3 85-0/u#iger Ameisensäure wird auf etwa 01 gekühlt und unter Rühren innerhalb von 5 Stunden mit 24 g Brom versetzt.
Man verdünnt das Reak tionsgemisch mit 120 cm3 Wasser, fügt 120 cm3 einer 204/digen Natriumbisulfitlösung zu und rührt eine weitere Stunde. Das Produkt wird abgenutscht, mit etwa 2 1 Wasser gewaschen, auf Tontellern ge trocknet und aus abs. Alkohol kristallisiert. Durch nochmaliges Umkristallisieren aus demselben Lö sungsmittel erhält man analysenreines 3-Methyl-5,7- dibrom-8-hydroxychinaldin vom Smp. 148-1491.
b) 3-Methyl-5,7-dibrom-8-acetoxychinaldin.
Ein Gemisch von 3,0 g 3-Methyl-5,7-dibrom-8- hydrochinaldin und 25 cm?, Essigsäureanhydrid mit 3 Tropfen konz. Schwefelsäure wird während 3 Stun den im Ölbad unter Rückfluss erhitzt. überschüssiges Essigsäureanhydrid wird unter vermindertem Druck abdestilliert, der Rückstand in 50 cm3 heissem Ligroin gelöst, filtriert und kristallisiert.
Das so erhaltene Rohprodukt kristallisiert man aus Isopropanol um und erhält das analysenreine 3-Methyl-5,7-dibrom- 8-acetoxychinaldin vom Smp. 134,5-1351. <I>Beispiel 15</I> 3-Methyl-5,7-dibrom-8-benzoyloxychinaldin Zu einer Lösung von 1,0 g 3-Methyl-5,7-dibrom,- 8-hydroxychinaldin in 2,0 cm3 Pyridin werden unter Kühlung im Eisbad 0,64 g Benzoylchlorid gegeben. Man lässt 17 Stunden bei Raumtemperatur stehen und dampft im Vakuum zur Trockne ein.
Der Ein dampfrückstand wird mit 5 cm3 Eiswasser versetzt und mit Chloroform ausgezogen. Den mit Natrium- hydrogencarbonatlösung gewaschenen Extrakt trock net man mit Natriumsulfat und dampft im Vakuum ein.
Das aus 30 cm3 kristallisierte 3-Methyl-5,7-di- brom-8-benzoyloxychinaldin ist analysenrein und schmilzt bei 168,5-169,5 . <I>Beispiel 16</I> 3-Methyl-5,7-dibrom-8-(p-chlorbenzoxy)-chinaldin Man löst 1,0 g 3-Methyl-5,7-dibrom-8-hydroxy- chinaldin (siehe Beispiel 14a) in 2,0 em3 Pyridin und versetzt unter Eiskühlung mit 0,795 g p-Chlor- benzoylchlorid. Nach 16stündigem Stehen bei Raum temperatur wird im Vakuum zur Trockne einge dampft,
der Rückstand mit 5 cm33 Eiswasser versetzt und mit 15 cm3 Chloroform ausgezogen. Den mit Natriumhydrogencarbonat gewaschenen und über Natriumsulfat getrockneten Extrakt dampft man ein und kristallisiert das Rohprodukt unter Verwendung von Norit aus Aceton. Nach mehrmaligem Umkri- stallisieren aus obigem Lösungsmittel schmilzt das analysenreine 3 - Methyl - 5,7 - dibrom - 8 - (p-chlor- benzoxy)-chinaldin konstant bei 172-1731.
<I>Beispiel 17</I> 3-Methyl-5,7-dibrom-8-(p-nitrobenzoxy)-chinaldin Zu einer Lösung von 1,0 g 3-Methyl-5,7-dibrom- 8-hydroxychinaldin (siehe Beispiel 14a) in 2,0 cm-' Pyridin werden unter Eiskühlung 0,84 g p-Nitro- benzoylchlorid gegeben und das Reaktionsgemisch 15 Stunden stehengelassen. Man dampft im Vakuum zur Trockne ein, versetzt den Rückstand mit 5 cm Eiswasser und extrahiert mit 20 cm3 Chloroform. Der mit Natriumhydrogencarbonat gewaschene Aus zug wird über Natriumsulfat getrocknet und einge dampft.
Man kristallisiert das Produkt aus 85 cm?, Essigester und erhält das analysenreine 3-Methyl- 5,7-dibrom-8-(p-nitrobenzoxy)-chinaldin in analysen reiner Form. Smp. 193-193,51.
<I>Beispiel 18</I> 5-Chlor-7-brom-8-(p-chlorbenzoxy)-chinaldin Zu einer Suspension von 27,2 g 5-Chlor-7-brom- 8-oxychinaldin (Smp. 113-1141), 10,1 g Triäthyl- amin und 250 ein- Benzol werden bei Raumtempe ratur 19,2 g p-Chlorbenzoylchlorid (Sdp. 97-1001/ 12 mm Hg) hinzugetropft. Der Ansatz wird hierauf bei einer Badtemperatur von 501 während 4 Stunden gerührt.
Das gebildete Triäthylaminhydrochlorid wird abgenutscht, mit 100 cm- Benzol nachgewa schen und das Filtrat im Vakuum zur Trockne ein geengt. Das rohe 5-Chlor-7-brom-8-(p-chlorbenzoxy)- chinaldin hat einen Smp. von 135-1401. Nach zwei maligem Umkristallisieren der Substanz aus Tetra chlorkohlenstoff erhält man das analysenreine 5-Chlor - 7 - brom - 8 - (p-chlorbenzoxy)-chinaldin mit dem konstanten Smp. 144-1461.

Das ungelöste Material wird abgenutscht, mit 60 cm?, Tetrachlorkohlenstoff nachgewaschen und das Filtrat im Vakuum zur Trockne eingeengt. Der Rückstand wird in 50 cm3 Chloroform aufge nommen, einmal mit 20 cma kalter 5 /oiger wässeri ger Natriumbicarbonatlösung und zweimal mit je 20 cm3 Wasser ausgezogen. Die wässerigen Teile werden mit zweimal je 20 cm3 Chloroform nach gewaschen und dann verworfen. Die vereinigten Chloroformteile werden über Natriumsulfat getrock net und im Vakuum zur Trockne eingeengt.
Nach dreimaligem Umkristallisieren des Rohproduktes aus Hexan erhält man das analysenreine 5,7-Dijod-8- benzoxychinaldin mit dem konstanten Smp. 132 bis 134 .

The process is carried out, for example, so that 5,7-dihalo-8-oxychinaldine with an acid anhydride in the presence of a catalytic additive, e.g. B. conc. Sulfuric acid or pyridine, heated to a higher temperature, preferably 130-160 '. The reaction solution is then poured directly onto ice or the excess acylating agent is first partially distilled off and only then poured onto ice. After filtering off with suction and washing with water, the corresponding esters are obtained.
If the acylating agent used for the acylation of the 5,7-dihalo-8-oxyquinaldines is an acid chloride or a mixture of acid chloride and acid anhydride, then the conversion is so that the 5,7-dihalo-8-oxychinaldine with the Acyl halide together in an inert organic solvent, e.g. B. benzene, toluene, xylene and pyridine are dissolved.
The solution of the reactants is at room temperature andjoder at elevated temperature, e.g. B. in a steam bath; Finally, the esters formed are isolated and purified by known methods.
The previously unknown esters of 5,7-dihalo-8-oxychinaldines are made by the present process and are solid bases. They are less soluble in water than the 5,7-dihalo-8-oxychinaldines and are therefore more poorly absorbed than these. They are characterized by therapeutically usable pharmacological properties in that they have increased effectiveness against pathogenic germs and other pathogens.
The new compounds are to be used in therapy as surface antiseptics, their poor water solubility, respectively. Resorbability ensures particularly good system compatibility. They also serve as intermediate products in the manufacture of drugs. In the following examples, the melting points are given in degrees Celsius and without correction.
<I> Example 1 </I> 5,7-dichloro-8-acetoxyquinaldine A mixture of 20.0 g of 5,7-dichloro-8-oxyquinaldine (melting point 110112, prepared according to the USA patent no. 2 411 <B> 670 </B> from Geigy) and 75.0 g of acetic anhydride with 8 drops of conc. Sulfuric acid are heated in a 160 'oil bath for 3 hours.
After cooling, the entire batch is poured onto 500g of ice, and after the ice has completely melted, the precipitated material is suction filtered and then washed with ice water until the wash water no longer reacts to the lackmussauer. After the crude product has been dried in a vacuum drying cabinet at 8011 to constant weight, it is recrystallized from light gasoline, and analytically pure 5,7-dichloro-8-acetoxyquinaldine with a constant melting point of 103.5-105 is obtained.
<I> Example 2 </I> 5,7-dibromo-8-acetoxyquinaldine a) A mixture of 40 g of 5,7-dibromo-8-oxyquininaldine (melting point 125-126, prepared according to the USA patent no 2 411 670 from Geigy) and <B> 100.0 </B> g acetic anhydride with 8 drops of conc. Sulfuric acid are heated in a 160 oil bath for 3 hours.
After cooling, the entire batch is poured onto 500 g of ice, and after the ice has completely melted, the precipitated material is suction filtered and then washed with ice water until the wash water no longer reacts lackmussauer. After the crude product has been dried in a vacuum drying cabinet at 70b to constant weight, it is recrystallized from carbon tetrachloride and the analytically pure 5,7-dibromo-8-acetoxyquinaldine with a constant melting point of 125-127b is kept.
b) To a solution of 47.5 g of 5,7-dibromo-8-oxychinaldine (melting point 125-126b, produced according to the USA patent No. 2,411 670 from Geigy) in 100 cm3 of pyridine are added in portions to 17.7 g of acetyl chloride (boiling point 51-52b) while being cooled in ice. After the batch has been left to stand at room temperature for 3 hours, it is concentrated to dryness at 400 in a water jet vacuum.
The residue is made into a paste with 100 cm3 of ice water, suction filtered and washed with ice water until the wash water no longer reacts to the lackmussauer. After the raw product has been dried in a vacuum drying cabinet at 809 to constant weight, it is recrystallized from carbon tetrachloride, and the analytically pure 5 is obtained,
7-dibromo-8-acetoxyquinaldine with constant m.p. 125-127b. <I> Example 3 </I> 5-chloro-7-bromo-8-acetoxyquinaldine A mixture of 40 g of 5-chloro-7-bromo-8-oxyquininaldine (m.p. 113-114b, prepared from 5-chloro -8- oxychinaldin according to the Swiss patent No. 235224 from Geigy) and 135g acetic anhydride with 4 drops of conc. Sulfuric acid is heated for 3 hours in an oil bath of <B> 1600 </B>.
After cooling, the entire batch is poured onto 500 g of ice, and after the ice has completely melted, the precipitated material is suction filtered and then washed with ice water until the wash water no longer reacts to the lackmussauer. After the crude product has been dried in a vacuum drying cabinet at 809 to constant weight, it is recrystallized from light gasoline, and the analytically pure 5-chloro-7-bromo-8-acetoxyquinaldine with a constant melting point of 121-122 is obtained.
<I> Example 4 </I> 5,7-dibromo-8-benzoxyquinaldine To a solution of 47.5 g of 5,7-dibromo-8-oxyquininaldine (melting point 125-126, prepared according to the USA patent No. 2 411 670 from Geigy) in 100 cm 3 of pyridine are added in portions with cooling in ice, 31.6 g of benzoyl chloride (boiling point 194-196e) while swirling.
After the batch has been left to stand at room temperature for 3 hours, it is concentrated to dryness in a water-jet vacuum at 40b. The residue is made into a paste with 100 cm3 of ice water, then 200 cm 3 of chloroform are added and the chloroform part is shaken out twice with 100 cm3 each, totaling 200 cm3 of a 10 hour aqueous sodium bicarbonate solution.
The chloroform part, washed twice with 25 cm 3 each, total 50 cm 3 of water, is dried over sodium sulfate and concentrated in a water-jet vacuum, whereupon the crude 5,7-dibromo-8-benzoxyquinaldine of melting point 124-130b is obtained. After repeated recrystallization of the substance from benzene and light gasoline, analytically pure 5,7-dibromo-8-benzoxyquinaldine with a constant melting point of 130-132b is obtained.
<I> Example 5 </I> 5,7-Dibromo-8-benzoxyquinaldine To a suspension of 63.4 g of 5,7-dibromo-8-oxychinaldine (melting point 125-1266), 15.8 g of pyridine and 300 cm3 carbon tetrachloride are added dropwise at room temperature with 28.1 g benzoyl chloride while stirring. The approach is then carried out at an internal temperature of <B> 50.1 </B> for 61 years. Hours stirred.
After cooling to room temperature, the pyridine hydrochloride formed is filtered off with suction, washed with 100 cm3 of benzene and the filtrate is concentrated to dryness in vacuo. It is then concentrated to dryness in vacuo at 60.
The residue is rubbed with 50 cm3 of ice water, then 200 cm3 of chloroform are added and the chloroform part is shaken out twice with 100 cm3 of a 5% aqueous sodium bicarbonate solution each time. The chloroform part, washed twice with 50 cm3 of water each time, is dried over sodium sulfate and concentrated in vacuo, whereupon the crude, oily 5,7-diiodo-8-acetoxyquinaldine is obtained.
After repeated recrystallization of the substance from light gasoline, analytically pure 5,7-diiodo-8-acetoxyquinaldine with a constant melting point of 120-122 is obtained.
<I> Example 7 </I> 5,7-dichloro-8-benzoxyquinaldine To a solution of 34.2 g of 5,7-dichloro-8-oxyquinaldine (melting point 111-112) in 100 cm3 of pyridine while cooling with ice, 32.7 g of benzoyl chloride were added in portions while swirling. After the batch has been left to stand for 4 hours at room temperature, it is concentrated to dryness in vacuo at 40.
The residue is rubbed with 100 cm3 of ice water, then 250 cm3 of chloroform are added and the chloroform part is shaken out twice with 100 cm3 of a 511 / own aqueous sodium bicarbonate solution each time. The chloroform part, washed twice with 25 cm water each time, is dried over sodium sulphate and concentrated in vacuo, whereupon the crude 5,7-dichloro-8-benzoxyquinaldine of melting point 118-121 is obtained.
After repeated recrystallization of the substance from light gasoline, the analytically pure 5,7-dichloro-8-benzoxyquinaldine with the constant Sm_p is obtained. 121-1231.
<I> Example 8 </I> 5,7-dibromo-8- (p-chlorobenzoxy) -quinaldine To a suspension of 31.7 g of 5,7-dibromo-8-oxychinaldine (melting point 125-126a), 8 cm- 'pyridine and 160 cm3 of ethylene chloride are added dropwise at room temperature 19.2 g of p-chlorobenzoyl chloride (bp. 97-100 112 mm Hg) with stirring. The batch is then stirred at a bath temperature of 50 for 4 hours.
After cooling to room temperature, the pyridine hydrochloride formed is filtered off with suction, washed with 100 cm- 'benzene and the filtrate is concentrated to dryness in vacuo. The crude 5,7-dibromo-8- (p-chlorobenzoxy) quinaldine has a melting point of 140-145. After repeated recrystallization of the substance from carbon tetrachloride and then from benzene, the analytically pure 5,7-dibromo-8- (p-chlorobenzoxy) quinaldine with a constant melting point of 148-150 is obtained.
<I> Example 9 </I> 5,7-dibromo-8- (p-nitrobenzoxy) quinaldine A suspension of 31.7 g of 5,7-dibromo-8-oxyquininaldine (melting point 125-126), 8 cm3 of pyridine, 350 cm3 of ethylene chloride and 18.6 g of p-nitrobenzoyl chloride (melting point 72) are stirred at a bath temperature of 50 for 4 hours.
After standing overnight, the pyridine hydrochloride formed is filtered off with suction the next day, washed with 100 cm3 of benzene and the filtrate is concentrated to dryness in vacuo. The crude 5,7-dibromo-8- (p-nitrobenzoxy) quinaldine has a m.p. 156-160.
After repeated recrystallization of the substance from carbon tetrachloride, the analytically pure 5,7-dibromo-8- (p-nitrobenzoxy) -quinaldine with the constant melting point 160.5-162 is obtained.
<I> Example 10 </I> 5-chloro-7-bromo-8-benzoxyquinaldine To a suspension of 40.8 g of 5-chloro-7-bromo-8-oxychinaldine (m.p. 113-114), 11.8 g of pyridine and 500 cm3 of carbon tetrachloride are added dropwise at room temperature, 22.1 g of benzoyl chloride (boiling point 194-196) with stirring. The batch is then stirred at an internal temperature of 50 for 4 hours.
After cooling to room temperature, the pyridine hydrochloride formed is filtered off with suction, washed with 100% benzene and the filtrate is concentrated to dryness in a water jet vacuum. The crude 5-chloro-7-bromo-8-benzoxyquinaldine has a melting point of 121-123. After recrystallization of the substance from light gasoline, the analytically pure 5-chloro-7-bromo-8-benzoxyquinaldine with the constant melting point 122-124 is obtained.
<I> Example 11 </I> 5-Chloro-7-bromo-8- (p-nitrobenzoxy) -quinaldine A suspension of 40.8 g of 5-chloro-7-bromo-8-oxychinaldine (m.p. 113-114 ), 12 cm3 of pyridine, 1000 cm3 of carbon tetrachloride and 29.3 g of p-nitrobenzoyl chloride (m.p. 72) for 4 hours at. an internal temperature of 50 stirred.
After cooling to room temperature, the pyridine hydrochloride formed is filtered off with suction, washed with 100 cm3 of benzene and the filtrate is evaporated to dryness in a water jet. The crude 5-chloro-7-bromo-8- (p-nitrobenzoxy) quinaldine has a melting point of 148-153. After repeated recrystallization of the substance from carbon tetrachloride, analytically pure 5-chloro-7-bromo-8- (p-nitrobenzoxy) quinaldine with a constant melting point of 155-157 is obtained.
<I> Example 12 </I> 5,7-dichloro-8- (p-chlorobenzoxy) quinaldine To a suspension of 22.8 g of 5.7 dichloro-8-oxychitialdine (melting point 111-112), 8, 0 g of pyridine and 150 cm3 of benzene are added dropwise at room temperature to 19.3 g of p-chlorobenzoyl chloride (boiling point 97-100 112 mm Hg) in 50 cm3 of benzene with stirring. The batch is then stirred at a bath temperature of 50 for 4 hours.
After cooling to room temperature, the pyridine hydrochloride formed is filtered off with suction, washed with 100 cm3 of benzene and the filtrate is concentrated to dryness in a water-jet vacuum. The crude 5,7-dichloro-8- (p-chlorobenzoxy) quinaldine has a melting point of 139 to 141. After recrystallization of the substance from carbon tetrachloride, analytically pure 5,7-dichloro-8- (p-chlorobenzoxy) quinaldine with a constant melting point of 139-141 is obtained.
<I> Example 13 </I> 5,7-dichloro-8- (p-nitrobenzoxy) quinaldine A suspension of 12.8 g of 5,7-dichloro-8-oxyquininaldine (melting point 111-112), 4.5 g of pyridine, 150 cm of ethylene chloride and 10.9 g of p-nitrobenzoyl chloride (melting point 721) are stirred at a bath temperature of 501 for 4 hours. After cooling to room temperature, the pyridine hydrochloride formed is filtered off with suction,
Washed with 100 cubic meters of benzene and the filtrate concentrated to dryness in a water-jet vacuum. The crude 5,7-dichloro-8- (p-nitrobenzoxy) quinaldine has a melting point of 158-162. <I> Example 14 </I> 3-Methyl-5,7-dibromo-8-acetoxyquinaldine a) 3-Methyl-5,7-dibromo-8-hydroxyquinaldine. A solution of 10.0 g of 3-methyl-8-hydroxyquinaldine (melting point 91-921, produced by condensation of o-aminophenol with a-methylcrotonaldehyde) in 60 cm3 of 85-0 / u # strength formic acid is increased to about 01 cooled and mixed with 24 g of bromine within 5 hours while stirring.

b) 3-methyl-5,7-dibromo-8-acetoxyquinaldine.
A mixture of 3.0 g of 3-methyl-5,7-dibromo-8-hydroquinaldine and 25 cm ?, acetic anhydride with 3 drops of conc. Sulfuric acid is refluxed in an oil bath for 3 hours. Excess acetic anhydride is distilled off under reduced pressure, the residue is dissolved in 50 cm3 of hot ligroin, filtered and crystallized.
The crude product thus obtained is recrystallized from isopropanol and the analytically pure 3-methyl-5,7-dibromo-8-acetoxyquinaldine with a melting point of 134.5-1351 is obtained. <I> Example 15 </I> 3-Methyl-5,7-dibromo-8-benzoyloxyquinaldine To a solution of 1.0 g of 3-methyl-5,7-dibromo, -8-hydroxyquinaldine in 2.0 cm3 of pyridine 0.64 g of benzoyl chloride are added with cooling in an ice bath. The mixture is left to stand at room temperature for 17 hours and evaporated to dryness in a vacuum.
The steam residue is mixed with 5 cm3 of ice water and extracted with chloroform. The extract, washed with sodium hydrogen carbonate solution, is dried with sodium sulfate and evaporated in vacuo.
The 3-methyl-5,7-di-bromo-8-benzoyloxyquinaldine crystallized from 30 cm3 is analytically pure and melts at 168.5-169.5. <I> Example 16 </I> 3-Methyl-5,7-dibromo-8- (p-chlorobenzoxy) -quinaldine Dissolve 1.0 g of 3-methyl-5,7-dibromo-8-hydroxyquinaldine ( see Example 14a) in 2.0 cubic meters of pyridine and treated with 0.795 g of p-chlorobenzoyl chloride while cooling with ice. After 16 hours of standing at room temperature, it is evaporated to dryness in a vacuum,
the residue was mixed with 5 cm33 of ice water and extracted with 15 cm3 of chloroform. <I> Example 17 </I> 3-Methyl-5,7-dibromo-8- (p-nitrobenzoxy) -quinaldine To a solution of 1.0 g of 3-methyl-5,7-dibromo-8-hydroxyquinaldine ( see Example 14a) in 2.0 cm- 'pyridine, 0.84 g of p-nitrobenzoyl chloride are added with ice cooling and the reaction mixture is left to stand for 15 hours. It is evaporated to dryness in vacuo, the residue is mixed with 5 cm of ice water and extracted with 20 cm3 of chloroform. The train washed with sodium hydrogen carbonate is dried over sodium sulfate and evaporated.

<I> Example 18 </I> 5-chloro-7-bromo-8- (p-chlorobenzoxy) quinaldine To a suspension of 27.2 g of 5-chloro-7-bromo-8-oxychinaldine (melting point 113- 1141), 10.1 g of triethylamine and 250 one-benzene are added dropwise at room temperature, 19.2 g of p-chlorobenzoyl chloride (boiling point 97-1001 / 12 mm Hg). The batch is then stirred at a bath temperature of 501 for 4 hours.
The triethylamine hydrochloride formed is filtered off with suction, washed with 100 cm benzene and the filtrate is concentrated to dryness in vacuo. The crude 5-chloro-7-bromo-8- (p-chlorobenzoxy) quinaldine has a melting point of 135-1401. After the substance has been recrystallized twice from carbon tetrachloride, the analytically pure 5-chloro-7-bromo-8- (p-chlorobenzoxy) quinaldine with a constant melting point of 144-1461 is obtained.
<I> Example 19 </I> 5,7-Diiodo-8-benzoxyquinaldine To a suspension of 6.0 g of 5,7-Diiodo-8-oxyquininaldine (melting point 144-145) and 2.46 g of sodium - bicarbonate in 300 cms of carbon tetrachloride are added dropwise 2.25 g of benzoyl chloride (boiling point 194 to 196). The batch is then stirred at an internal temperature of 50 for 6 hours.
The undissolved material is filtered off with suction, washed with 60 cm ?, carbon tetrachloride and the filtrate is concentrated to dryness in vacuo. The residue is taken up in 50 cm3 of chloroform, extracted once with 20 cm3 of cold 5% aqueous sodium bicarbonate solution and twice with 20 cm3 of water each time. The aqueous parts are washed with twice 20 cm3 of chloroform and then discarded. The combined chloroform parts are dried over sodium sulfate and concentrated to dryness in vacuo.
After three recrystallization of the crude product from hexane, analytically pure 5,7-diiodo-8-benzoxyquinaldine with a constant melting point of 132 to 134 is obtained.
