Verfahren zur Herstellung von basisch substituierten Buttersäureaniliden
Es ist bereits bekannt, dass man durch Hydrierung von Ketocarbonsäureamiden in Gegenwart vonAminen Aminocarbonsäureamide erhält (vgl. beispielsweise die niederländische Patentschrift Nr. 57807). Weiterhin sind im Journ. Am. Chem. Soc. 71 (1946), Seite 1906,/3-Amino-buttersäureanilide, deren Anilidrest durch Halogenatome substituiert ist, als Zwischenprodukte für die Herstellung von Chinolin-Derivaten beschrieben ; beispielsweise wurde durch Anlagerung von p-Chloranilin an Crotonsäure - (p-Chlor-anilino)- buttersäure bzw. das entsprechende p-Chlor-anilid hergestellt.
Es wurde nun gefunden, dass man basisch substi tuierte Buttersäureanilide erhält, die bei geringer Toxizität gute antipyretische, antiphlogistische sowie analgetische Eigenschaften aufweisen, wenn man Acetessigsäure-chlor-anilide der Formel

worin R für Wasserstoff oder einen Alkylrest mit höchstens 4 Kohlenstoffatomen steht, vorzugsweise in einstufiger Reaktion, mit Ammoniak oder Aminen und reduzierenden Mitteln behandelt.
Als Ausgangsstoffe seien beispielsweise genannt : Acetessigsaure-o-,-m-oder-p-chlor-anilid, Acetessigsäure-N-methyl-o-,-m-oder p-chlor-anilid, Acet essigsäure-N-äthyl-o-,-m-oder-p-chlor-anilid, Acet essigsäure-N-propyl-o-,-m-oder-p-chlor-anilid, Acetessigsäure-N-isopropyl-o-,-m-oder-p-chlor-anilid, Acetessigsäulre-N-butyl-o-,-m-oder-p-chlor-anilid, Acetessigsäure-N-isobutyl-o-bzw.-m-oder-p-chlor- anilid, Acetessigsäure-N-sek.-butyl-o-,-m-oder p-chlor- anilid. Die Herstellung der Ausgangsverbindungen kann beispielsweise nach der im Journ. Am. Chem.
Soc. 70 (1948), Seite 2403, angegebenen Vorschrift vorgenommen werden ; sie kann aber auch durch Umsetzung der entsprechenden Chlor-anilin-Derivate mit Diketen erfolgen.
Als Amine können beispielsweise herangezogen werden : Methylamin, Dimethylamin, Äthylamin, Di äthylamin, Methyläthylamin, Propylamin, Butylamin, Butylmethylamin, Isobutylamin, sek.-Butylamin, Benzylamin und Benzylmethylamin. Anstelle dieser Verbindungen können jedoch auch gesättigte heterocyclische stickstoffhaltige Verbindungen, wie Piperidin, Pyrrolidin oder Morpholin eingesetzt werden.
Die Umsetzung der Anilide mit Ammoniak oder Aminen erfolgt vorteilhaft gleichzeitig mit der Reduktion. Man kann das Verfahren jedoch auch in zwei Stufen mit oder ohne Isolierung intermediär entstehender Anlagerungsprodukte durchführen.
Wird Ammoniak als Umsetzungskomponente verwendet, so wird zweckmässig eine mit Ammoniakgas in der Kalte gesättigte AlkohoHösung mit der Lösung des entsprechenden Acetessigsäure-chlor-anilids in dem gleichen Alkohol umgesetzt und reduziert.
Die Reduktion kann man mit nascierendem Wasserstoff, beispielsweise mit Aluminiumamalgam, in Gegenwart eines Wasserstoff abspaltenden Lösungsmittels, vornehmen. Als Lösungsmittel kommen insbesondere niedrigmolekulare aliphatische Alkohole, gegebenenfalls mit Wasser gemischt, in Frage. Zweckmässig arbeitet man bei mässig erhöhter Temperatur, vorzugsweise beim Siedepunkt des verwendleten Lösungsmittels.
Mit besonderem Vorteil verwendet man als Re duktionsmethode die katalytische Hydrierung, wobei als Katalysatoren vorzugsweise Metalle der 8. Gruppe des periodischen Systems, beispielsweise Nickel, zweckmässig in Form von Raney-Katalysatoren, verwendet werden. Man arbeitet auch in diesem Falle zwockmässig bei mässig erhöhten Temperaturen von etwa 50 bis 100"C, vorzugsweise bei etwa 60 C, um bei der Hydrierung der Anlagerungsverbindung des Amins an das Acetessigsäure-chlor-anilid das Chloratom des Anilidrestes nicht abzuspalten.
Auch bei der katalytischen Hydrierung ist es angebracht, in Gegenwart von Lösungsmitteln, insbesondere von niedrigmolekularen aliphatischen Alkoholen, die ge gebenenfalls mit Wasser verdünnt sein können, zu arbeiten.
Die Verfahrenserzeugnisse stellen wertvolle Arzneimittel dar, die eine gute Wasserlöslichkeit besitzen.
Beispielsweise ist das ss-Methylamino-buttersäure-p- chlor-anilid-hydrochlorid hinsichtlich seiner pharma kologischen Eigenschaften dem bekannten N-Acetyl-pphenetidin erheblich überlegen ; ausserdemlöst sich ein Teil A-Methylamino-buttersäure-p-chlor-anilid-hydro- chlorid bei Zimmertemperatur in 6 Teilen Wasser, wohingegen die bekannte Verbindung praktisch wasserunlöslich ist. Der mit der guten Wasserlöslichkeit verbundene Vorteil ist insbesondere für die parenterale Applikation der Verfahrenserzeugnisse von Bedeutung.
Weiterhin zeichnet sich beispielsweise das /-Athylamino-buttersäure-p-chlor-anilid-hydrochlorid bei geringer Toxizität durch gute antiphlogistische Eigenschaften aus, wie aus nachstehenden pharmakologischen Daten ersichtlich ist. Die Dos. let. betrug bei i. v. Injektion an der Maus 125 mg/kg. Bei s. c.
Injektion lag sie ebenfalls bei 125 mg/kg. Oral appliziert waren dagegen 1,25 g/kg tödlich. Bei Ratten betrug die Dos. let. bei i. v. Injektion 90 mg/kg, bei s. c. Injektion 400 mg/kg und bei oraler Gabe 2 g/kg.
Bei der durch Injektion von Siliciumdioxyd an der Rattenpfote erzeugten Entzündung waren 100 mg/kg s. c. stark und langdauernd wirksam ; in einem Versuch fanden sich z. B. folgende Schwellungswerte : Bei den Kontrollen war 3 Stunden nach der Injektion die Pfote um durchschnittlich 31 /o geschwollen, nach 6 Stunden um 63"/o und nach 24 Stunden um 70 /o. Eine andere Gruppe von Ratten erhielt einmalig vor dem Versuch 100 mg/kg ss-Sithylamino- buttersäure-p-chlor-anilid-hydrochlorid s. c. Die Werte lagen nach 3 Stunden nur 8 O/o ilber dem Ausgangswert, nach 6 Stunden um 29 /o und nach 24 Stunden um 50 /o.
Wurde Ratten ein Stück Rohwatte subkutan eingepflanzt, so konnte das entstehende Bindegewebsgranulom durch fl-Athylamino-buttersdure-p- chlor-anilid-hydrochlorid gedämpft werden. Bei 7maliger Injektion von 80 mg/kg ss-Äthylamino- buttersäure-p-chlor-anilid-hydrochlorid i. m. in 9 Tagen wurde z. B. die Bindegewebsneubildung um 40,5 /o gegeniiber den Kontrolltieren vermindert. Die allergische Konjunktivitis konnte bei Kaninchen durch prophylaktische Injektion von 50 mg/kg ss-Sithylamino-buttersäure-p-chlor-anilid-hydrochlo- rid s. c. weitgehend gehemmt werden.
Die Verfahrenserzeugnisse lassen sich mit Hilfe von anorganischen und organischen Säuren in übli- cher Weise in entsprechende Säureadditionssalze überführen. Als anorganische Säuren sind beispielsweise Halogenwasserstoffsäuren, wie Chlorwasser stoffsäure, Schwefelstoffsäure, Phosphorsäure, Amidosulfonsäure geeignet. Als organische Säuren kommen beispielsweise in Frage : Essigsäure, Propionsäure, Milchsäure, Malonsäure, Bernsteinsäure, Apfelsäure, Weinsäure, Maleinsäure, Fumarsäure, Zitro nensäure, Benzoesäure, Salicylsäure, Oxyäthansulfon- säure, Acetursäure und Athylendiamintetraessigsäure.
Beispiel 1 a) 53 gAcetessigsäure-p-chlor-anilid und 500 cm3 Methanol werden zusammen mit 20g 40 feiger wässriger Methylaminlösung bei 60-65 C in Gegenwart von Raney-Nickel hydriert. Nach Aufnahme der berechneten Menge Wasserstoff wird vom Katalysator abfiltriert und das Filtrat unter vermindertem Druck eingeengt. Der ölige Rückstand wird in 2n-Salzsäure gelöst, mit Kohle filtriert und das klare Filtrat mit überschüssiger 2n-Natronlauge versetzt. Nach Extrahieren mit Ather wird die Atherlösung getrocknet und das Lösungsmittel entfernt. Es werden 49,5 g eines hellgelben, dicken Sirups erhalten.
Das in üblicher Weise hergestellte Hydrochlorid des erhaltenen-Methylamino-buttersäure-p- chlor-anilids schmilzt bei 200-201 C nach Reinigung durch Lösen in Alkohol unter Ätherzugabe bis zur beginnenden Trübung. b) Die gleiche Verbindung kann auch durch Reduktion mit Aluminiumamalgam erhalten werden, indem 30g Acetessigsäure-p-chlor-anilid mit 15g amalgamierten Aluminiumspänen und 30 g wässrigem 40 feigem Methylamin, gelöst in 150 cm3 Athylalkohol und 10 cm3 Wasser, 4 Stunden auf dem Dampfbad erwärmt werden. Nach Zugabe von 100 cm3 Athylalkohol wird filtriert und wie vorstehend beschrieben, aufgearbeitet.
Es werden 29 g Hydrochlorid des 4-Methylamino-buttersäure-p-chlor- anilids vom Schmelzpunkt 200-201 C erhalten.
Beispiel 2
53 g Acetessigsäure-p-chlor-anilid werden entsprechend der in Beispiel la angegebenen Vorschrift zusammen mit 20 g Butylamin hydriert. Nach Filtration und Abdestillieren des Lösungsmittels wird der Rückstand mit verdünnter Salzsäure und anschliessend mit Methylenchlorid behandelt, wobei das zunächst ölig abgeschiedene Hydrochlorid in die Methylenchloridschicht übergeht. Nach Trennen der beiden Schichten, Trocknen der Methylenchloridlösung und Abdestillieren des Lösungsmittels wird ein fester Rückstand erhalten, der nach Verreiben mit Aceton abgesaugt wird. Nach Umkristallisieren aus Athylalkohol wird das Hydrochlorid des ss-Butyl- amino-buttersäure-p-chlor-anilids vom Schmelzpunkt 206 C erhalten.
Beispiel 3
53 g Acetessigsäure-p-chlor-anilid werden zusammen mit 100 g Methanol, welches in der Kälte mit Ammoniakgas gesättigt wurde und 500 cm3 Methanol, entsprechend der in Beispiel la angegebenen Vorschrift, hydriert und aufgearbeitet. Der Schmelzpunkt des ss-Amino-buttersäure-p-chlor- anilids liegt bei 76-77 C (nach Umkristallisieren aus Cyclohexan und Benzol).
Beispiel 4 a) 56,5 g Acetessigsäure-N-methyl-p-chlor-anilid (hergestellt aus N-Methyl-p-chlor-anilin und Diketen) werden zusammen mit 30 g 30 /0igem wässrigem Methylamin und 500 cm3 Methanol entsprechend Beispiel la hydriert. Nach Filtration wird eingeengt und der Rückstand mit 10 /oiger Essigsäure behandelt. Das hierin nicht gelöste 61 wird ausgeäthert. Die essigsaure Lösung wird mit Natronlauge alkalisch gemacht. Nach Ausäthern, Trocknen und Verdampfen des Lösungsmittels wird ss-Methylamino- buttersäure-N-methyl-p-chlor-anilid als hellbraunes 01 erhalten.
Analyse : Ber. Chlor 14,75 /Ï Gef. 14,591/o
Stickstoff 11,65 /o 11, 45 /o b) 56,5 g Acetessigsäure-N-methyl-p-chlor-anilid, 30g amalgamierte Aluminiumspäne, 30g 30 /oiges wässriges Methylamin und 150 cm3 Athylalkohol werden 4 Stunden auf dem Dampfbad erhitzt. Die Aufarbeitung erfolgt wie vorstehend unter a) beschrieben.
Beispiel 5
56,5 g Acetessigsäure-N-methyl-p-chlor-anilid, 20 g Butylamin, 15 g amalgamierte Aluminiumspäne, 150 cm3 Athylalkohol und 30cm3 Wasser werden 4 Stunden auf dem Dampfbad erhitzt. Die Aufarbeitung erfolgt wie in Beispiel 1 b beschrieben. Es werden 32 g yS-Butylamino-buttersäure-N-methyl-p- chlor-anilid als öl erhalten.
Analyse : Ber. Chlor 12,55 /o Gef. 12,14"/.

Beispiel 6
102,5 g Acetessigsäure-N-äthyl-p-chlor-anilid werden mit 97 g 30 /oigerwässrigerMethylaminlösung entsprechend Beispiel la hydriert. Die Aufarbeitung erfolgt entsprechend Beispiel 4a. -Methylamino- buttersäure-N-äthyl-p-chlor-anilid wird als hellbraunes öl erhalten.

Beispiel 7
53 g Acetessigsäure-p-chlor-anilid und 40 g 34"/oige alkoholische Athylaminlösung werden in 500 cm3 Alkohol in Gegenwart von Raney-Nickel bei 60 C unter Druck hydriert. Die Aufarbeitung erfolgt wie in Beispiel la beschrieben. Es wird -Athylamino- buttersäure-p-chlor-anilid-hydrochlorid vom Schmelzpunkt 178 C erhalten.
Beispiel 8
42,3 g Acetessigsäure-p-chlor-anilid und 16 g sek.
Butylamin werden in 100 cm3 Alkohol gelöst. Diese
Lösung wird mit 15 g Aluminiumamalgam und 20 cm3
Wasser 4 Stunden unter Rückfluss erhitzt. Nach Absau gen und Waschen mit Alkohol wird das FiltratimVa kuum eingeengt, der Rückstand in 21 mit Salzsäure angesäuertem Wasser (congosauer) gelöst und fil triert. Das Filtrat wird unter vermindertem Druck eingeengt. Der feste Rückstand wird mit Aceton di geriert. Nach Umkristallisieren aus wenig Wasser wird das Hydrochlorid des 4-(Butyl-2-amino)-butter- säure-p-chlor-anilidsvom SchmeLzpunkt 206207 C erhalten.
Beispiel 9
40 g Acetessigsäure-p-chlor-anilid, 200 cm3 Al kohol, 40 cm3 Wasser und 20 g n-Propylamin werden mit 20 g Aluminiumamalgam 3 bis 4 Stunden unter
Rückfluss erhitzt. Die Aufarbeitung erfolgt entspre chend der in Beispiel 8 angegebenen Vorschrift be schrieben. Nach Umsetzen mit verdünnter Salzsäure und Eiskühlung kristallisieren 44 g, B-n-Propylamino- buttersäure-p-chlor-anilid-hydrochlorid aus.
Schmelzpunkt 199-200 C nach Umkristallisie- ren aus absolutem Alkohol.
Beispiel 10
40 g Acetessigsäure-p-chlor-anilid, 20 g 1-Amino n-hexan, 200 cm3 Athylalkohol und 40 cm3 Wasser werden mit 20 g Aluminiumamalgam 3 bis 4 Stunden unter Rückfluss gekocht. Nach Filtrieren und Einengen des Filtrats zur Trockene werden 54 g eines hellen
Oles erhalten, die in Ather gelöst und mit alkoho lischer Salzsäure versetzt werden. Der erhaltene
Kristallbrei wird abgesaugt ; nach Lösen in Äthyl alkohol wird mit Ather versetzt. Es werden 35 g p-n-
Hexylamino-buttersäure-p-chlor-anilid-hydrochlorid vom Schmelzpunkt 208 C erhalten.
Beispiel 11
40 g Acetessigsäure-p-chlor-anilid, 25 g Isobutyl amin, 200 cm3 Athylalkohol und 50 cm3 Wasser werden mit 25 g Aluminiumamalgam 6 Stunden unter
Rückfluss gekocht. Die Aufarbeitung erfolgt entspre chend der in Beispiel 10 angegebenen Vorschrift.
Es werden 24 g ss-Isobutylamino-buttersäure-p-chlor- anilid-hydrochlorid vom Schmelzpunkt 173-174 C erhalten.
Beispiel 12
40 g Acetessigsäure-p-chlor-anilid, 25 g Cyclo hexylamin, 250 cm3 Athylalkohol und 50 cm3 Wasser werden mit 25 g Aluminiumamalgam 6 Stunden unter
Rückfluss gekocht. Die Aufarbeitung erfolgt entsprechend der in Beispiel 9 angegebenen Vorschrift. Es werden 32 g S-Cyclohexylamino-buttersäure-p-chlor- anilid-hydrochlorid vom Schmelzpunkt 222-223 C erhalten.
Beispiel 13
53 g Acetessigsäure-o-chlor-anilid, 20 g n-Butyl amin, 200 cm3 Athylalkohol und 40 cm3 Wasser werden mit 20 g Aluminiumamalgam 4 Stunden unter Rückfluss gekocht. Die Aufarbeifung erfolgt entsprechend der in Beispiel 10 angegebenen Vorschrift.
Nach Lösen des erhaltenen Hydrochlorids in Wasser, Ausäthem und Einengen der filtrierten wässrigen Lösung wird das Hydrochlorid in Athylalkohol gelöst und durch Versetzen mit Äther zur Kristallisation gebracht. Es werden 22,7 g ss-n-Butylamino-butter säure-o-chlor-anilid-hydrochlorid vom Schmelzpunkt 149-150 C erhalten.
Beispiel 14
21,5 g Acetessigsäure-p-chlor-anilid, 13,6 g p-Amino-phenyläthylamin, 150 cm3 Athylalkohol und 30 cm3 Wasser werden mit 15 g Aluminiumamalgam 4 Stunden unter Rückfluss gekocht. Die Aufarbeitung erfolgt entsprechend der in Beispiel 10 angegebenen Vorschrift. Das erhaltene Dihydrochlorid (33,6 g) wird in Wasser gelöst, mit Kohle filtriert und mit 2n Natronlauge versetzt. Die zunächst ölige Base kristallisiert bald. Schmelzpunkt 103-104 C.
Nach Versetzen mit der molaren Menge Salzsäure werden 26 g 8-(p-Amino-phenyläthylamino)-butter- säure-p-chlor-anilid-hydrochlorid vom Schmelzpunkt 175-176 C erhalten.
Process for the production of basic substituted butyric anilides

Examples of starting materials include: acetoacetic acid-o-, - m- or -p-chloro-anilide, acetoacetic acid-N-methyl-o-, - m- or p-chloro-anilide, aceto-acetic acid-N-ethyl-o- , -m-or-p-chloro-anilide, acetoacetic acid-N-propyl-o-, - m-or-p-chloro-anilide, acetoacetic acid-N-isopropyl-o-, - m-or-p-chlorine -anilide, acetoacetic acid-N-butyl-o -, - m- or-p-chloro-anilide, acetoacetic acid-N-isobutyl-o- or-m- or-p-chloro-anilide, acetoacetic acid-N-sec. -butyl-o -, - m- or p-chloro anilide. The preparation of the starting compounds can be carried out, for example, according to the method described in Journ. At the. Chem.
Soc. 70 (1948), page 2403, specified procedure can be made; but it can also be done by reacting the corresponding chloro-aniline derivatives with diketene.
The following amines can be used, for example: methylamine, dimethylamine, ethylamine, diethylamine, methylethylamine, propylamine, butylamine, butylmethylamine, isobutylamine, sec-butylamine, benzylamine and benzylmethylamine. Instead of these compounds, however, saturated heterocyclic nitrogen-containing compounds such as piperidine, pyrrolidine or morpholine can also be used.
The reaction of the anilides with ammonia or amines is advantageously carried out simultaneously with the reduction. However, the process can also be carried out in two stages with or without isolation of addition products formed as intermediates.
If ammonia is used as the reaction component, an alcohol solution saturated with ammonia gas in the cold is expediently reacted and reduced with the solution of the corresponding acetoacetic acid-chloro-anilide in the same alcohol.
The reduction can be carried out with nascent hydrogen, for example with aluminum amalgam, in the presence of a hydrogen-releasing solvent. Particularly suitable solvents are low molecular weight aliphatic alcohols, optionally mixed with water. It is expedient to work at a moderately elevated temperature, preferably at the boiling point of the solvent used.
Catalytic hydrogenation is used with particular advantage as the reduction method, preferably metals of group 8 of the periodic table, for example nickel, advantageously in the form of Raney catalysts, being used as catalysts. In this case, too, work is carried out at moderately elevated temperatures of about 50 to 100 ° C., preferably at about 60 ° C., in order not to split off the chlorine atom of the anilide residue during the hydrogenation of the addition compound of the amine to the acetoacetic acid chloroanilide.
In the case of catalytic hydrogenation, too, it is appropriate to work in the presence of solvents, in particular of low molecular weight aliphatic alcohols, which can optionally be diluted with water.
The products of the process represent valuable pharmaceuticals that have good water solubility.
For example, the ß-methylamino-butyric acid-p-chloro-anilide hydrochloride is considerably superior to the known N-acetyl-pphenetidine in terms of its pharmacological properties; In addition, a part of A-methylamino-butyric acid-p-chloro-anilide hydrochloride dissolves in 6 parts of water at room temperature, whereas the known compound is practically insoluble in water. The advantage associated with the good solubility in water is particularly important for the parenteral application of the products of the process.
Furthermore, for example, ethylamino-butyric acid-p-chloro-anilide hydrochloride is characterized by good anti-inflammatory properties with low toxicity, as can be seen from the following pharmacological data. The Dos. let. was at i. v. Injection into the mouse 125 mg / kg. By S. c.

If a piece of raw cotton was implanted subcutaneously in rats, the resulting granuloma of the connective tissue could be dampened by fl-ethylamino-buttersdure-p-chloro-anilide hydrochloride. With 7 injections of 80 mg / kg ss-ethylamino-butyric acid-p-chloro-anilide-hydrochloride i. m. in 9 days z. B. the formation of new connective tissue is reduced by 40.5% compared to the control animals. The allergic conjunctivitis was reduced in rabbits by prophylactic injection of 50 mg / kg ss-sithylamino-butyric acid-p-chloro-anilide-hydrochloride s. c. largely inhibited.
The products of the process can be converted into corresponding acid addition salts in the usual way with the aid of inorganic and organic acids.
