Verfahren zur Herstellung eines neuen basischen Äthers. Gegenstand des vorliegenden Patentes ist. ein Verfahren zur Herstellung eines neuen hasisehen Äthers, nämlich des 1-(2'-Methoxy- l)lienoxy )-2-isopropy laminopropans. Es wurde gefunden, dass dieser Äther das vegetative NTer- v ensystem beeinflusst.
Er soll daher als Pharmazeuticum und auch als Zwischenpro dukt zur Herstellung weiterer Derivate Ver wendung finden.
Das den Gegenstand des Patentes bildende Verfahren ist dadurch gekennzeichnet, dass man eine Verbindung der Formel

umsetzt, in welchen beiden Formeln eines der X, ein 'Sauerstoffatom und das andere ein Wasserstoffatom und einen zur reduktiven Alkylierung befähigten stickstoffhaltigen Rest bedeutet.
Man kann erfindungsgemäss beispielsweise 1-(2'-lIethoxyphenoxy)-"-oxopropan mit Iso- propylamin und einem Reduktionsmittel zur Reaktion bringen. Man kann dabei so vor gehen, dass das Isopropylamin in Gegenwart. des andern Ausgangsstoffes aus 2-Nitropropan gebildet wird. Als Reduktionsmittel kann man die für solche Fälle üblichen, wie zum Beispiel Zink und Mineralsäuren, aktiviertes Alumi nium usw., verwenden. Als weitaus ergiebigstes Reduktionsmittel ist aber katalytisch erregter Wasserstoff anzusprechen.
Als Katalysatoren sind die Metalle der Raney-Gruppe, die be kannten Platin-Katalysatoren usw., verwend bar. Wird in Gegenwart von Raney-Katalysa- toren reduziert, so arbeitet man vorzugsweise unter Druck; beim Arbeiten in Gegenwart von Platin-Katalysatoren kann der Wasser stoff ohne Druck oder mit nur leichtem Über druck angewendet werden. Die Reduktion wird vorteilhaft in einem Lösungsmittel, bei spielsweise in niederen Alkoholen durchge führt.
Eine weitere Auführungsform des Verfahrens besteht darin, dass man 1-(2' Methoxyphenoxy)-'2-oxopropan mit Isopropyl- benzylamin zusammen katalytisch reduziert, wobei man zugleich die reduktive Alkylierung und Debenzylierung vornimmt,. Man kann er findungsgemäss auch 1-(2'-Methoxy-phenoxy)- 2-aminopropan mit Aceton und einem Re- duktionsmittel umsetzen.
Dabei kann man das 1-(2'-hlethoxyphenoxy)-2-aminopropan in Ge genwart des andern Ausangsstoffes aus 1- Z 2'-1let.hoxyphenoxy )-?-nitropropan herstellen. Das so erhaltene 1-(?'-hlethoxyplienoxy)-?- isopropylaminopropan siedet unter 14 bis 15 mm bei 150 bis 151 und bildet farblose :adeln, die bei 33 bis 34 schmelzen.
Das Chlorhydrat der Verbindung schmilzt bei 135 bis 137 und das Pi'krat bei 133 bis 134 . Der neue Basisehe Äther soll als Zwischen- produkt und als Pharmazeutikum Verwendung <B>M</B> f indes.
Beispiel <I>1:</I> 120 g 1-(2'-3ietlioxyphenoxy)-2-oxopropan, 190 ema Isopropy lamm und -180 eins Me thanol werden mit einngen Gramm Raney- Niekel im Autoklaven bei \?0 unter einem Wasserstoffdruek von 110 at geschüttelt.
Naeli ?4 Stunden filtriert man den Katalysatoren ab und verdampft das Filtrat im Vakuum. Der Rückstand wird in 330 ems 'n-Salzsäure aufgenommen und die Lösung zwecks Entfer nung von nicht tungesetztem Ausgan ggsketon ausgeäthert. Die salzsaure Lösung wird an schliessend wieder alkalisch gemacht und aus geäthert. Die Äthersehieht wird getrocknet und verdampft.
Der Rückstand wird im Va kuum destilliert. Man erhält so durehsehnitt- lieh 90 bis 100 g 1-(Z'-Methoxyplienoxy)-2- isopropylaminopropan, das. ein -unter 14 bis 15 mm bei 150 bis 151 siedendes farbloses Öl. bildet. Dieses verfestigt sieh nach einiger Zeit zti gut ausgebildeten Nadeln, die bei 33 bis 34 schmelzen. Das Chlorhydrat des neuen Äthers schmilzt bei 13G5 bis 137 und das Pikrat bei 133 bis 134 .
Beispiet <I>2:</I> Es wurden etwa 1 Mol 1-(2'-lletlioxy l)lieil- oxy)-2-oxopropan und 1 Mol 2-Nitropropan unter den in Beispiel 1 angegebenen Bedin- gungen zusammen reduziert und danach auf gearbeitet. Man erhielt 1-(2'-Methoxyplien- oxy)=?-isopropylamin in befriedigender Au- beute.
<I>Beispiel<B>3:</B></I> 0,5 Mol 1-(2'-Methoxyphenoxy)-?-anrinopro- pan und 1 bis 1,5 Mol Aceton werden in Ge- genwart von einigen Urarom Raney-Niekel bei 80 unter einem Wasserstoffdruck von 150 at geschüttelt. Nach dein Aufhören der Wasserstoffaufnalmie filtriert man den Kata lysatoren ab und verdampft zur Trockne.
Der Rückstand wird in @ssi@ester mit ätherischer Salzsäure versetzt und das ausgefallene Chlor hydrat zweimal aus Essigester umkristalli siert. Man erhält so das Chlorhydrat. des 1- ( _' - lletlioxyplien oxz-) - ? - isopropyl aminopro- pans mit dem Sehmelzpinikt 135 bis 137 in guter Reinheit.
